Oversulfated chondroitin sulfate (OSCS) was identified as a contaminant in certain heparin preparations as the cause of adverse reactions in patients. OSCS was found to possess both plasma anticoagulant activity and the ability to activate prekallikrein to kallikrein. Differentially sulfated chondroitin sulfates were prepared by synthetic modification of chondroitin sulfate and were compared to the activity of OSCS purified from contaminated heparin. Whilst chondroitin sulfate was found to have minimal anticoagulant activity, increasing sulfation levels produced an anticoagulant response which we directly show for the first time is mediated through heparin cofactor II. However, the tetra-sulfated preparations did not possess any higher anticoagulant activity than several tri-sulfated variants, and also had lower heparin cofactor II mediated activity. Activation of prekallikrein was concentration dependent for all samples, and broadly increased with the degree of sulfation, though the di-sulfated preparation was able to form more kallikrein than some of the tri-sulfated preparations. The ability of the samples to activate the kinin system, as measured by bradykinin, was observed to be through kallikrein generation. These results show that whilst an increase in sulfation of chondroitin sulfate did cause an increase in anticoagulant activity and activation of the kinin system, there may be subtler structural interactions other than sulfation at play given the different responses observed.

BackgroundWall shear stress (WSS) is a stimulus for vessel wall remodeling. Differences in ascending aorta (AAo) hemodynamics have been reported between bicuspid aortic valve (BAV) and tricuspid aortic valve patients with aortic dilatation, but the confounding impact of aortic valve stenosis (AS) is unknown. Methods and ResultsFive hundred seventy‐one subjects underwent 4‐dimensional flow magnetic resonance imaging in the thoracic aorta (210 right‐left BAV cusp fusions, 60 right‐noncoronary BAV cusp fusions, 245 tricuspid aortic valve patients with aortic dilatation, and 56 healthy controls). There were 166 of 515 (32%) patients with AS. WSS atlases were created to quantify group‐specific WSS patterns in the AAo as a function of AS severity. In BAV patients without AS, the different cusp fusion phenotypes resulted in distinct differences in eccentric WSS elevation: right‐left BAV patients exhibited increased WSS by 9% to 34% (P

A lack of quality control tools limits the enforcement of fortification policies. In alignment with the World Health Organization’s ASSURED criteria (affordable, sensitive, specific, user-friendly, rapid and robust, equipment-free, and deliverable), a paper-based assay that interfaces with a smartphone application for the quantification of iron fortificants is presented. The assay is based on the Ferrozine colorimetric method. The reaction started after deposition of the 5 µL aqueous sample and drying. After developing color, pixel intensity values were obtained using a smartphone camera and image processing software or a mobile application, Nu3px. From these values, the actual iron concentration from ferrous sulfate and ferrous fumarate was calculated. The limits of detection, quantification, linearity, range, and errors (systematic and random) were ascertained. The paper-based values from real samples (wheat flour, nixtamalized corn flour, and infant formula) were compared against atomic emission spectroscopy. The comparison of several concentrations of atomic iron between the spectrophotometric and paper-based assays showed a strong positive linear correlation (y = 47.01x + 126.18; R2 = 0.9932). The dynamic range (5.0−100 µg/mL) and limit of detection (3.691 µg/mL) of the paper-based assay are relevant for fortified food matrices. Random and systematic errors were 15.9% and + 8.65 µg/g food, respectively. The concept can be applied to limited-resource settings to measure iron in fortified foods.

We share our self-study research of faculty-building a successful collaborative culture and team teaching experience in a unique Master's program for experienced teachers. As part of self-study and reflective practice, this faculty team shared its collaborative experiences with teachers. This transparency had positive effects on the teachers' perceptions of faculty and on their own teaming experiences. To frame our work, we use the perspective of learning zones adapted from Vygotsky's conception of zone of proximal development. A multi-vocal perspective on the processes of faculty professional development and program development is presented.

The Preschool Family Support Initiative is a collaboration among low-income immigrant families, their neighborhood preschool, community agencies and organizations, and University faculty and students. It was designed to create a network of support for preschool children's early learning and development. The Initiative developed in the context of an on-going University/community partnership called the Urban Alternative. This article describes the framework of partnership within which the collaboration developed, the processes involved, and the impact of this collaborative effort from the perspectives of the participants.

Childhood ataxia may be due to multifactorial causes of impairment in the coordination of movement and balance. Acutely presenting ataxia in children may be due to infectious, inflammatory, toxic, ischemic, or traumatic etiology. Intermittent or episodic ataxia in children may be manifestations of migraine, benign positional vertigo, or intermittent metabolic disorders. Nonprogressive childhood ataxia suggests a congenital brain malformation or early prenatal or perinatal brain injury, and progressive childhood ataxia indicates inherited causes or acquired posterior fossa lesions that result in gradual cerebellar dysfunction. CT and MRI of the central nervous system are the usual modalities used in imaging children presenting with ataxia, based on the clinical presentation. This document provides initial imaging guidelines for a child presenting with acute ataxia with or without a history of recent trauma, recurrent ataxia with interval normal neurological examination, chronic progressive ataxia, and chronic nonprogressive ataxia. The ACR Appropriateness Criteria are evidence-based guidelines for specific clinical conditions that are reviewed annually by a multidisciplinary expert panel. The guideline development and revision process support the systematic analysis of the medical literature from peer reviewed journals. Established methodology principles such as Grading of Recommendations Assessment, Development, and Evaluation or GRADE are adapted to evaluate the evidence. The RAND/UCLA Appropriateness Method User Manual provides the methodology to determine the appropriateness of imaging and treatment procedures for specific clinical scenarios. Predators that feed on chemically-defended prey often experience non-lethal effects that result in learned avoidance of the prey species. Some predators are able to consume toxic prey without ill-effect. The Chinese mantid is able to consume cardenolide-containing monarch caterpillars without immediate adverse effects. Although they discard the caterpillars’ gut contents, mantids consume sequestered cardenolides. Although consumption of these cardenolides does not elicit an acute response, there may be long-term costs associated with cardenolide consumption. We tested the hypothesis that consumption of monarch caterpillars will adversely affect adult mantid biomass gain and reproductive condition. We reared mantids from egg to adult and assigned them to one of four toxicity groups that differed in the number of monarch caterpillars offered over a 15-day period. Mantids consumed similar amounts of prey biomass during the experiment. Yet, mantids in the high-toxicity group had a higher conversion efficiency and gained more biomass than mantids in other groups. Mantids in all treatment groups produced similar numbers of eggs. However, mantids in the high-toxicity group produced heavier eggs and devoted a greater portion of their biomass toward egg production than those in the control group. This increase in reproductive condition is probably driven by variation in prey nutritional value and/or the nutritional advantages inherent in eating multiple food types. Our results demonstrate the mantid is able to incorporate ‘toxic’ monarch prey into its diet without acute or chronic ill-effects.

Although studies have addressed the exhaustion of the host's immune response to HCV and its role in treatment, there is little information about the possible contribution of innate immunity to treatment-induced clearance. We hypothesized that because intact myeloid dendritic cell (MDC) pathogen sensing functions are associated with improved HCV-specific CD8+ T cell functionality in some chronically infected patients, it might enhance HCV clearance rate under standard interferon therapy. To investigate this hypothesis, TLR-induced MDC activation and HCV-specific CD8+ T cell response quality were monitored longitudinally at the single-cell level using polychromatic flow cytometry in chronically infected patients undergoing interferon therapy. We correlated the immunological, biochemical and virological data with response to treatment. We demonstrate that the clinical efficacy of interferon-induced viral clearance is influenced by the extent to which HCV inhibits MDC functions before treatment, rather than solely on a breakdown of the extrinsic T cell immunosuppressive environment. Thus, viral inhibition of MDC functions before treatment emerges as a co-determining factor in the clinical efficacy of interferon therapy during chronic HCV infection.

Cu electroplating on an ultrathin Co seed has been developed for superconformal filling of advanced interconnects, in an acidic CuSO4 electrolyte containing plating additives, i.e., halide, suppressor, accelerator, and leveler. A suppressor-halide adlayer is found to play a bifunctional role in both suppressing Cu growth and inhibiting Co dissolution. Corrosion inhibition is attributed to adsorption of hydrophobic suppressor molecules on a halide-terminated Co surface that blocks water from interacting with Co, thereby retarding the formation of Co(OH)+, a corrosion immediate with which hydronium from the electrolyte would react to form soluble Co2+. With enhanced suppression, Co loss is mainly confined to the removal of native Co oxides in acid. Correspondingly, galvanic Cu deposition forms a monolayer shortly after immersion at open-circuit potential, becoming self-limited with growth of a second layer over the next 20 s as dynamic surface processes make more underlying Co available for the displacement reaction. Growth of the first Cu layer is controlled by the receding of native oxides in an exponential-decay manner. Native Co oxides, if not removed, promote Cu protrusions in electroplating. Crohn disease is an inflammatory condition of the gastrointestinal tract with episodes of exacerbation and remission occurring in children, adolescents, and adults. Crohn disease diagnosis and treatment depend upon a combination of clinical, laboratory, endoscopic, histological, and imaging findings. Appropriate use of imaging provides critical information in the settings of diagnosis, assessment of acute symptoms, disease surveillance, and therapy monitoring. Four variants are discussed. The first variant discusses the initial imaging for suspected Crohn disease before established diagnosis. The second variant pertains to appropriateness of imaging modalities during suspected acute exacerbation. The third variant is a substantial discussion of recommendations related to disease surveillance and monitoring of Crohn disease. Finally, panel recommendations and discussion of perianal fistulizing disease imaging completes the document. Imaging plays an integral role in the evaluation of suspected musculoskeletal infections in children, not only in the accurate identification of infection such as osteomyelitis or septic arthritis, but also in guiding management. Various diagnostic modalities serve different purposes in the assessment of suspected pediatric musculoskeletal infections. The purpose of this document is to provide imaging guidance in the most frequently encountered clinical scenarios in which osteomyelitis and/or septic arthritis are suspected, outside of the axial skeleton. On 16 November 2006, a female juvenile megamouth shark, Megachasma pelagios, was caught off the coast of Mexico in the Pacific Ocean, near Sebastián Vizcaíno Bay. This specimen, that has informally been referred to as “Megamouth No. 38”, measured 2265 mm in total length. It represents the third smallest female recorded for this taxon and the first report of M. pelagios off the Pacific coast of Mexico. The specimen is formally described here with basic anatomical data, and some aspects of this megamouth shark are compared with those of previously reported M. pelagios specimens. The occurrence near Sebastián Vizcaíno Bay is significant because it further emphasizes the high elasmobranch diversity in the area that should be regarded as a key region for shark conservation.

Arbuckle-Simpson aquifer in southern Oklahoma, USA, is a major source of water for industrial and municipal use. It is also a major source for spring-fed streams in the area. As part of an ongoing study to map and characterize the Arbuckle-Simpson aquifer, an electrical resistivity tomography (ERT) study and electrical resistivity sounding studies were conducted in Johnston County, Oklahoma, USA, to map the subsurface of a small area of the carbonate aquifer. The main aim of the study was to obtain constraints on the location of near surface faults and fractures and how they control groundwater flow in the study area. The interpreted resistivity section along an N-S profile indicates that the water table in the region is deepening to the south and probably bounded in the north by a south dipping fault. Inverse modeling of 2D electrical resistivity tomography (ERT) data acquired at two adjacent locations within the study area indicate shallow, fractured Arbuckle group rocks saturated with water adjacent to dry rocks. From electrical resistivity mapping results, it is inferred that the Mill Creek block in the Arbuckle-Simpson aquifer is an isolated system, interacting with the northern segment of a silicate-based aquifer through dissolution faults and fractures.
