108. Simulation of an 1857-like Mw 7.9 San Andreas fault earthquake and the response of tall steel moment frame buildings in southern California -A prototype study

Abstract

In 1857, an earthquake of magnitude 7.9 occurred on the San Andreas fault, starting at Parkfield and rupturing in a southeasterly direction for more than 360 km. Such a unilateral rupture produces significant directivity toward the San Fernando and Los Angeles basins. The strong shaking in the basins due to this earthquake would have had significant long-period content (2-8 s), and the objective of this study is to quantify the impact of such an earthquake on two 18-story steel moment frame building models, hypothetically located at 636 sites on a 3.5 km grid in southern California. End-to-end simulations include modeling the source and rupture of a fault at one end, numerically propagating the seismic waves through the earth structure, simulating the damage to engineered structures and estimating the economic impact at the other end using high-performance computing. In this prototype study, we use an inferred finite source model of the magnitude 7.9, 2002 Denali fault earthquake in Alaska, and map it onto the San Andreas fault with the rupture originating at Parkfield and propagating southward over a distance of 290 km. Using the spectral element seismic wave propagation code, SPECFEM3D, we simulate an 1857-like earthquake on the San Andreas fault and compute ground motions at the 636 analysis sites. Using the nonlinear structural analysis program, FRAME3D, we subsequently analyze 3-D structural models of an existing tall steel building designed using the 1982 Uniform Building Code (UBC), as well as one designed according to the 1997 UBC, subjected to the computed ground motion at each of these sites. We summarize the performance of these structural models on contour maps of peak interstory drift. We then perform an economic loss analysis for the two buildings at each site, using the Matlab Damage and Loss Analysis (MDLA) toolbox developed to implement the PEER loss-estimation methodology. The toolbox includes damage prediction and repair cost estimation for structural and non-structural components and allows for the computation of the mean and variance of building repair costs conditional on engineering demand parameters (i.e. inter-story drift ratios and peak floor accelerations). Here, we modify it to treat steel-frame high-rises, including aspects such as mechanical, electrical and plumbing systems, traction elevators, and the possibility of irreparable structural damage. We then generate contour plots of conditional mean losses for the San Fernando and the Los Angeles basins for the pre-Northridge and modern code-designed buildings, allowing for comparison of the economic effects of the updated code for the scenario event. In principle, by simulating multiple seismic events, consistent with the probabilistic seismic hazard for a building site, the same basic approach could be used to quantify the uncertain losses from future earthquakes.

Abstract

Traditional shading design principles guide the vertical and horizontal orientation of fins, louvres and awnings being applied to orthogonal planar façades. Due to doubly curved envelopes characterising many contemporary designs, these rules of thumb are now not always applicable. Operable blinds attempt to regulate the fluctuating luminance of daylight and aid in shading direct sunlight. Mostly they remain closed, as workers are commonly too preoccupied to continually adjust them so a reliance on electrically powered lights remains a preference. To remedy these problems, the idea of what it is to sustainable enclose space is reconsidered through the geometric and kinetic optimisation of a parametric skin, with sunlight responsive modules that regulate interior light levels. This research concludes with an optimised design and also defines some unique metrics to gauge the design’s performance in terms of, the amount of exterior unobstructed view, its ability to shade direct sunlight and, its daylight glare probability.

Abstract

An investigation into the possible causes of childhood cancer has been carried out throughout England, Scotland and Wales over the period 1991-1998. All children known to be suffering from one or other type of the disease over periods of 4-5 years have been included, and control children matched for sex, age and area of residence have been selected at random from population registers. Information about both groups of children (with and without cancer) has been obtained from parental questionnaires, general practitioners' and hospital records, and from measurement of the extent of exposure to radon gas, terrestrial gamma radiation, and electric and magnetic fields. Samples of blood have also been obtained from the affected children and their parents and stored. Altogether 3,838 children with cancer, including 1,736 with leukaemia, and 7,629 unaffected children have been studied. Detailed accounts are given of the nature of the information obtained in sections describing the general methodology of the study, the measurement of exposure to ionizing and non-ionizing radiation, the classification of solid tumours and leukaemias, and the biological material available for genetic analysis.

Contains fulltext : 97633.pdf (publisher's version ) (Closed access), The transient receptor potential channel C6 (TRPC6) is a slit diaphragm-associated protein in podocytes involved in regulating glomerular filter function. Gain-of-function mutations in TRPC6 cause hereditary focal segmental glomerulosclerosis (FSGS), and several human acquired proteinuric diseases show increased glomerular TRPC6 expression. Angiotensin II (AngII) is a key contributor to glomerular disease and may regulate TRPC6 expression in nonrenal cells. We demonstrate that AngII regulates TRPC6 mRNA and protein levels in cultured podocytes and that AngII infusion enhances glomerular TRPC6 expression in vivo. In animal models for human FSGS (doxorubicin nephropathy) and increased renin-angiotensin system activity (Ren2 transgenic rats), glomerular TRPC6 expression was increased in an AngII-dependent manner. TRPC6 expression correlated with glomerular damage markers and glomerulosclerosis. We show that the regulation of TRPC6 expression by AngII and doxorubicin requires TRPC6-mediated Ca(2+) influx and the activation of the Ca(2+)-dependent protein phosphatase calcineurin and its substrate nuclear factor of activated T cells (NFAT). Accordingly, calcineurin inhibition by cyclosporine decreased TRPC6 expression and reduced proteinuria in doxorubicin nephropathy, whereas podocyte-specific inducible expression of a constitutively active NFAT mutant increased TRPC6 expression and induced severe proteinuria. Our findings demonstrate that the deleterious effects of AngII on podocytes and its pathogenic role in glomerular disease involve enhanced TRPC6 expression via a calcineurin/NFAT positive feedback signaling pathway.

Abstract

Steroid-resistant nephrotic syndrome (SRNS) is a frequent cause of end-stage renal failure. Identification of single-gene causes of SRNS has generated some insights into its pathogenesis; however, additional genes and disease mechanisms remain obscure, and SRNS continues to be treatment refractory. Here we have identified 6 different mutations in coenzyme Q10 biosynthesis monooxygenase 6 (COQ6) in 13 individuals from 7 families by homozygosity mapping. Each mutation was linked to early-onset SRNS with sensorineural deafness. The deleterious effects of these human COQ6 mutations were validated by their lack of complementation in coq6-deficient yeast. Furthermore, knockdown of Coq6 in podocyte cell lines and coq6 in zebrafish embryos caused apoptosis that was partially reversed by coenzyme Q10 treatment. In rats, COQ6 was located within cell processes and the Golgi apparatus of renal glomerular podocytes and in stria vascularis cells of the inner ear, consistent with an oto-renal disease phenotype. These data suggest that coenzyme Q10-related forms of SRNS and hearing loss can be molecularly identified and potentially treated.

Abstract

Recently, there has been increasing interest in simulating all aspects of the seismic risk problem, from the source mechanism to the propagation of seismic waves to nonlinear time-history analysis of structural response and finally to building damage and repair costs. This study presents a framework for performing truly “end-to-end” simulation. A recent region-wide study of tall steel-frame building response to a M_w 7.9 scenario earthquake on the southern portion of the San Andreas Fault is extended to consider economic losses. In that study a source mechanism model and a velocity model, in conjunction with a finite-element model of Southern California, were used to calculate ground motions at 636 sites throughout the San Fernando and Los Angeles basins. At each site, time history analyses of a nonlinear deteriorating structural model of an 18-story steel moment-resisting frame building were performed, using both a pre-Northridge earthquake design (with welds at the moment-resisting connections that are susceptible to fracture) and a modern code (UBC 1997) design. This work uses the simulation results to estimate losses by applying the MDLA (Matlab Damage and Loss Analysis) toolbox, developed to implement the PEER loss-estimation methodology. Abstract

The safety of building occupants during and immediately after disasters, such as a major earthquake, is highly dependent on the way in which people interact with the damaged physical environment. While there are extensive studies on evacuation from undamaged structures and on structural behavior under seismic and other hazards, research on the influence of building damage on human evacuation behavior is limited. This study presents a framework by which models for buildings and human behavior can be coupled to analyze the dynamic influences of building damage on the evacuation process. The framework combines nonlinear dynamic finite-element modeling of structures, probabilistic modeling of damage, and agent-based modeling of human occupants to investigate the behavior of people as they interact with each other and with their dynamically-deteriorating environment as they attempt to evacuate the building. A case study is presented for a typical three-story commercial office building subjected to the ground motions of the 1994 Northridge, California earthquake. By using exit flow rates and other measures related to evacuation time histories as the outcomes of interest, it is shown how the proposed framework can be used as a tool to enhance building design and to develop recommendations for improved evacuation strategies. Abstract

Common bacterial blight (CBB) caused by Xanthomonas campestris pv. phaseoli reduces common bean (Phaseolus vulgaris L. ) yield and quality worldwide. Genetic resistance provides effective disease control; however, a high level of resistance is difficult to attain and does not exist in pinto bean, the most important dry bean market class in North America. Our objective was to determine if a backcross breeding approach with the aid of molecular markers linked to quantitative trait loci (QTL) for resistance to CBB in a donor parent could be used to attain higher levels of resistance to CBB in pinto bean. QTL conditioning CBB resistance from the donor parent XAN 159 were introgressed into the recurrent parent ‘Chase’ using classical backcross breeding and intermittent marker-assisted selection. ‘Chase’ pinto bean is moderately resistant and the breeding line XAN 159 is highly resistant to Xanthomonas campestris. Marker assays confirmed the presence of independent QTL from GN no. 1 Sel 27 and XAN 159 in advanced backcross-derived pinto bean lines with improved CBB resistance. Agronomic characteristics of ‘Chase’ were fully recovered in the backcross-derived lines. An important QTL for CBB resistance from XAN 159 on linkage group B6 was not introgressed because tight linkage between this QTL and the dominant V allele that causes an unacceptable black-mottled seed coat color pattern in pinto bean could not be broken.
