Graphical abstract, The inositol phosphates, InsP5 and InsP6, have recently been identified as binding partners of fibrinogen, which is critically involved in hemostasis by crosslinking activated platelets at sites of vascular injury. Here, we investigated the putative physiological role of this interaction and found that platelets increase their InsP6 concentration upon stimulation with the PLC-activating agonists thrombin, collagen I and ADP and present a fraction of it at the outer plasma membrane. Cone and plate analysis in whole blood revealed that InsP6 specifically increases platelet aggregate size. This effect is fibrinogen-dependent, since it is inhibited by an antibody that blocks fibrinogen binding to platelets. Furthermore, InsP6 has only an effect on aggregate size of washed platelets when fibrinogen is present, while it has no influence in presence of von Willebrand factor or collagen. By employing blind docking studies we predicted the binding site for InsP6 at the bundle between the γ and β helical subunit of fibrinogen. Since InsP6 is unable to directly activate platelets and it did not exhibit an effect on thrombin formation or fibrin structure, our data indicate that InsP6 might be a hemostatic agent that is produced by platelets upon stimulation with PLC-activating agonists to promote platelet aggregation by supporting crosslinking of fibrinogen and activated platelets.

108. SuperTIGER-2 2018 Flight Payload Recovery and Preliminary Instrument Assessment

- Abstract
The SuperTIGER (Trans-Iron Galactic Element Recorder) experiment was launched for the second time from the Long Duration Balloon (LDB) site near McMurdo Station, Antarctica on December 20, 2018 at 1:36 AM NZDT. The balloon reached a peak altitude of ∼79,300 ft before it began to descend, and the flight had to be terminated after just over six hours aloft. The payload landed at 8:17 AM NZDT approximately 150 miles from McMurdo Station at 75.80 S and 161.68 E. Satellite imagery of the site revealed that it was in a crevasse field, but a Twin Otter reconnaissance flight showed none in the immediate vicinity. A subsequent site survey by a team flown in by helicopter with ground penetrating radar was able to flag a safe zone including helicopter landing sites. The SuperTIGER-2 payload was recovered in two days with two Bell 212 helicopters followed by one day with a Basler, which involved transferring the payload to a safe fixed wing landing site roughly two miles away in three helicopter sling loads. After return to the LDB site the instrument modules were reassembled, rewired and tested prior to their being shipped north. The instrument was found to be in overall good condition, with all testable electronics channels working, and minimal mechanical damage. SuperTIGER-2 could be refurbished to fly again as early as this next Antarctic season.

The CALorimetric Electron Telescope (CALET) is an imaging calorimeter under construction for launch to the ISS in 2014 for a planned 5 year mission. CALET consists of a charge detection module (CHD) with two segmented planes of 1 cm thick plastic scintillator, an imaging calorimeter (IMC) with a total of 3 radiation lengths ( X ∘ ) of tungsten plates read out with 8 planes of interleaved scintillating fibers, and a total absorption calorimeter (TASC) with 27 X ∘ of lead tungstate (PWO) logs. The primary objectives of the instrument are to measure electron energy spectra from 1 GeV to 20 TeV, to detect gamma-rays above 10 GeV, and to measure the energy spectra of nuclei from protons through iron up to 1,000 TeV. In this paper we describe how the geomagnetic field at the 51.6° inclination orbit of the ISS can be used to allow CALET to measure the rare ultra-heavy (UH) cosmic ray (CR) abundances, which provide important clues for the CR source and acceleration mechanism. The CHD scintillator response is relatively insensitive to energy above minimum ionization, and the angle-dependent rigidity as a function of geomagnetic latitude can be exploited to discriminate particles above this energy threshold. Such events require corrections for trajectory in instrument that can be made with only the top 4 layers of the IMC, which allows for considerably greater geometric acceptance than for events that require passage through the TASC for energy determination. Using this approach CALET will be able to measure UH CR relative abundances over its expected mission with superior statistics to previous space instruments.

First results of a cosmic-ray electron + positron spectrum, from 10 GeV to 3 TeV, is presented based upon observations with the CALET instrument on the ISS starting in October, 2015. Nearly a half million electron + positron events are included in the analysis. CALET is an all-calorimetric instrument with total vertical thickness of 30 $X_0$ and a fine imaging capability designed to achieve a large proton rejection and excellent energy resolution well into the TeV energy region. The observed energy spectrum over 30 GeV can be fit with a single power law with a spectral index of -3.152 $\pm$ 0.016 (stat.+ syst.). Possible structure observed above 100 GeV requires further investigation with increased statistics and refined data analysis., 7 pages, 6 figures, published 1 November 2017 in Physical Review Letters

No new antimicrobials have been approved for the treatment of mastitis since 2006; it would be beneficial to perform controlled field trials comparing 2 existing treatments. Non-inferiority trials are valuable in this regard, as they do not require a negative control as in a randomized, controlled FDA drug trial. If a therapy with similar efficacy is available, decision making criteria can include convenience, lower costs, less side effects, improved delivery systems, and better integration into current protocols. The purpose was to perform a non-inferiority comparison of 2 intramammary treatments for clinical mastitis (CM). - Abstract
The SuperTIGER (Trans-Iron Galactic Element Recorder) instrument was launched from Williams Field, Antarctica on December 8, 2012 and flew for 55 days at a mean altitude of 125,000 feet on a long-duration balloon flight. SuperTIGER measured the relative abundances of Galactic cosmic-ray nuclei with high statistical precision and well resolved individual element peaks in the charge range Z=10-40. In addition, SuperTIGER made exploratory measurements of the relative abundances up to Z=56. Although the statistics are low for charges greater than Z=40, we will show how the relative abundances of charges Z=40-56 compare to those reported by HEAO3. A second flight, SuperTIGER-II, is planned for December 2017. As SuperTIGER-II will fly during solar minimum, we estimate a ~50% increase in the particles collected per unit time as compared to SuperTIGER-I. With the combined data sets of SuperTIGER-I and II we will improve statistics in the Z=30-40 range and measure individual elemental abundances up to Z=56.

- Abstract
SuperTIGER (Trans-Iron Galactic Element Recorder) is a large-area balloon-borne instrument built to measure the galactic cosmic-ray abundances of elements from Z=10 (Ne) through Z=56 (Ba) at energies from 0.8 to ~10 GeV/nuc. SuperTIGER successfully flew around Antarctica for a record-breaking 55 days, from December 8, 2012 to February 1, 2013. In this paper, we present results of an analysis of the data taken during the flight for Fe (Z=26). We report on energy calibrations and instrumental and atmospheric corrections to obtain cosmic ray intensities vs. energy, and we will compare selected SuperTIGER galactic cosmic ray Fe spectrum with those from ACE/SIS during the time of the SuperTIGER flight.

- Abstract
First results of a cosmic-ray electron + positron spectrum, from 10 GeV to 3 TeV, is presented based upon observations with the CALET instrument on the ISS starting in October, 2015. Nearly a half million electron + positron events are included in the analysis. CALET is an all-calorimetric instrument with total vertical thickness of 30 $X_0$ and a fine imaging capability designed to achieve a large proton rejection and excellent energy resolution well into the TeV energy region. The observed energy spectrum over 30 GeV can be fit with a single power law with a spectral index of -3.152 $\pm$ 0.016 (stat.+ syst.). - Abstract
In August 2015, the CALorimetric Electron Telescope (CALET), designed for long exposure observations of high energy cosmic rays, docked with the International Space Station (ISS) and shortly thereafter began tocollect data. CALET will measure the cosmic ray electron spectrum over the energy range of 1 GeV to 20 TeV with a very high resolution of 2% above 100 GeV, based on a dedicated instrument incorporating an exceptionally thick 30 radiation-length calorimeter with both total absorption and imaging (TASC and IMC) units. Each TASC readout channel must be carefully calibrated over the extremely wide dynamic range of CALET that spans six orders of magnitude in order to obtain a degree of calibration accuracy matching the resolution of energy measurements. These calibrations consist of calculating the conversion factors between ADC units and energy deposits, ensuring linearity over each gain range, and providing a seamless transition between neighboring gain ranges. This paper describes these calibration methods in detail, along with the resulting data and associated accuracies. The results presented in this paper show that a sufficient accuracy was achieved for the calibrations of each channel in order to obtain a suitable resolution over the entire dynamic range of the electron spectrum measurement., Comment: 13 pages, 11 figures, published online 4 March 2017

Die bakterielle Kontamination des Ejakulates kann die Spermienqualität und damit auch die männliche Fertilität negativ beeinflussen. Über das Ejakulat-Keimspektrum bei infertilen Patienten gibt es in der Literatur unterschiedliche Angaben. Wir untersuchten daher die primären Ejakulatkulturen infertiler Patienten, die unsere andrologische Sprechstunde aufsuchten. Bei 160 Patienten wurden retrospektiv die primären Ejakulatkulturen ausgewertet, wobei eine Keimzahl über 103 Keime/ml als positive Kultur (laut WHO 1993) gewertet wurde. Eine positive Ejakulatkultur (Keimzahl 103) wurde in 52 % (n = 83), eine bakterielle Kontamination mit einer Keimzahl von unter 103 in 41 % (n = 65) und sterile Kulturen lediglich bei 7 % (n = 12) der Patienten nachgewiesen. Von den Patienten mit einer positiven Ejakulatkultur hatten 36 % (n = 30) Mischkulturen. Die häufigsten Erreger waren Ureaplasmen/Mycoplasmen mit 26 % (n = 22) und Enterococcus faecalis mit 19 % (n = 16), wobei Ureaplasmen/Mykoplasmen in 17 % (n = 14) mit anderen Keimen kombiniert waren. In 8 % (n = 7) waren Staphylokokken (Staph. aureus, Staph. epidermidis) nachweisbar, in 5 % (n = 4) Escherichia coli. Eine positive Ejakulatkultur ist bei infertilen Patienten häufig zu finden. Aufgrund der negativen Auswirkung der bakteriellen Kontamination auf die Spermatozoenqualität sollte im Rahmen der Primärdiagnostik immer eine Ejakulatkultur erhoben werden, um eine testgerechte antibiotische Therapie zu ermöglichen.

The CALorimetric Electron Telescope (CALET) is an imaging calorimeter under construction for launch to the ISS in 2014 for a planned 5 year mission. CALET consists of a charge detection module (CHD) with two segmented planes of 1 cm thick plastic scintillator, an imaging calorimeter (IMC) with a total of 3 radiation lengths ( X ∘ ) of tungsten plates read out with 8 planes of interleaved scintillating fibers, and a total absorption calorimeter (TASC) with 27 X ∘ of lead tungstate (PWO) logs. The primary objectives of the experiment are to measure the electron e - + e + energy spectra from 1 GeV to 20 TeV, to detect gamma-rays above 10 GeV, and to measure the energy spectra of nuclei from protons through iron up to 1000 TeV. In this paper we describe how the geomagnetic field at the 51.6 ° inclination orbit of the ISS can be used to allow CALET to measure the distinct electron and positron fluxes. The positron fraction has been seen to rise above ∼ 10 GeV by previous experiments (HEAT, AMS-01), and more recently to continue to increase to higher energies ( ∼ 80 GeV for PAMELA, ∼ 200 GeV for Fermi and ∼ 350 GeV with the best statistics for AMS-02). Utilizing the geomagnetic cutoff, CALET will be able to distinguish electrons and positrons in the ∼ 3 –20 GeV energy range where the positron fraction turns upward to complement existing high statistics measurements.
