Aim: The aim was to determine the occurrence of Rhodococcus equi in equines and their environment in Jammu (R.S. Pura, Katra), molecular characterization and to determine the antibiotic resistance pattern of R. equi. Materials and Methods: A total of 96 nasopharyngeal swab samples were collected from equines. The organism was isolated on Columbia nalidixic acid agar containing 5% sheep blood as well as on sheep blood agar and was later confirmed by cultural characteristics and biochemical tests. Molecular detection of R. equi isolates was done by 16S rRNA gene amplification followed by virulence associated protein A (Vap A) gene amplification. Antibiogram was performed against five antibiotics, viz., amoxicillin, penicillin G, streptomycin, rifampicin, and methicillin. Results: During the study, 9 R. equi isolates were identified on the basis of cultural and biochemical tests. In the polymerase chain reaction based detection, 3 among the 9 rhodococcal isolates were positive for species-specific 16S rRNA gene and revealed amplicon of 450 bp for confirmation of 16S rRNA gene. None of the sample was found positive for Vap A gene. In antibiogram, R. equi isolates were found sensitive for amoxicillin, while some isolates were also found resistant to the most conventional antibiotic penicillin G. Conclusion: From this study, it was concluded that R. equi infection is prevalent in equines in Jammu region of India and the indiscriminate use of the antibiotics is leading toward the development of resistant strains of R. equi.

More than 50% of patients with systemic lupus erythematosus (SLE) have renal involvement at presentation or during their illness. Lupus nephritis (LN) encompasses several patterns of renal disease, including glomerular, tubulointerstitial, and vascular pathologies. The presence and significance of renal vascular lesions (VLs) are often overlooked. VLs in LN are not rare with an incidence of 10%–40% on renal biopsies from various studies and their presence is often labeled as poor prognostic markers. The current treatment protocol for LN is mainly based on the glomerular pathology, and no guidelines/consensus exists for treatment of LN with VLs. We describe the clinical presentation, course, response to therapy, and outcomes in five patients with SLE with histological evidence of renal VLs.

We aim to evaluate the disturbances in mineral metabolism, abnormalities in bone mineral density (BMD), and extraskeletal calcification in newly detected, untreated predialysis stage 4 and 5 chronic kidney disease (CKD) patients at a tertiary care hospital in North India. This is cross-sectional observational study. A total of 95 (68 males, 27 females) newly detected patients underwent clinical evaluation, biochemical assessment [serum calcium, phosphorus, alkaline phosphatase (ALP), albumin, creatinine, intact parathyroid hormone (iPTH), 25- hydroxyvitamin D (25(OH)D)], BMD measurement (at spine, hip, and forearm) by dual-energy X-ray absorptiometry (DXA), lateral abdominal radiograph [for abdominal aortic calcification (AAC)], skeletal survey (to look for any abnormality including fractures), and echocardiography [for any cardiac valvular calcification (CVC)]. Symptoms related to CKD-mineral bone disorder were seen in 33.6% of the study patients. Prevalence of hypocalcemia, hyperphosphatemia, hyperparathyroidism, and hypovitaminosis D was 64.2%, 81.1%, 49.5%, and 89.5%, respectively. CVC was seen in 22.1% of patients on echocardiography, mostly involving the mitral valve. Patients with CVC were more likely to be males and smokers. There was no significant difference in iPTH levels between patients with or without CVC. AAC was seen in 10.5% of patients on lateral abdominal X-ray. Patients with AAC had higher levels of iPTH, phosphorus, and ALP and lower levels of calcium compared to patients without AAC. BMD by DXA showed a low bone mass in 41.05% of our patients and was more prevalent in CKD stage 5. Most of the study patients had hyperparathyroidism and low 25(OH)D levels. Our study shows that newly detected, naïve Indian CKD patients have a high prevalence of disturbances of mineral metabolism including hyperparathyroidism, Vitamin D deficiency, abnormal BMD, and valvular and vascular calcification, even before initiating dialysis.

Aim: To study the prevalence of antimicrobial resistance due to extended spectrum beta-lactamase (ESBL)-producing Escherichia coli in samples collected from the ceca of healthy broilers of poultry sale outlets (PSOs) Jabalpur. Materials and Methods: A total of 400 cecal swab samples were taken randomly from freshly slaughtered poultry of 39 PSOs located at four different zones or areas of Jabalpur and were screened for the presence of ESBL-producing E. coli using standard methods. Further they were characterized phenotypically by standard methods. Results: All the 400 samples were screened for E. coli producing ESBL enzyme. Among the samples positive for E. coli 135 were positive for ESBL E. coli giving an overall prevalence of 33.5%. Conclusion: This study related to the prevalence of ESBL-producing E. coli in healthy broilers in Jabalpur is indicative of antibiotic resistance prevalent in the healthy birds which are used for human consumption as well. It also signifies resistance prevalent against beta-lactam antibiotics including third and fourth generations of cephalosporins.

Aim: The aim was to determine the occurrence of streptococci in equines in Jammu (R. S. Pura, Katra), characterization of Streptococci equi subsp. equi and Streptococcus equi subsp. zooepidemicus with respect to their virulence traits and to determine antibiotic sensitivity pattern of virulent Streptococcus isolates. Materials and Methods: A total of 96 samples were collected from both clinically affected animals (exhibiting signs of respiratory tract disease) and apparently healthy animals and were sent to laboratory. The organisms were isolated on Columbia nalidixic acid agar containing 5% sheep blood as well as on sheep blood agar and confirmed by cultural characteristics and biochemical tests. Molecular detection of Streptococcus was done directly from cultures using sodA and seM gene-based polymerase chain reaction (PCR). Antibiogram was performed against five antibiotics such as amoxicillin, penicillin G, streptomycin, rifampicin, and methicillin. Results: During this study, a total 40 streptococcal isolates were obtained out of which 2 isolates were of S. equi subsp. equi, 12 isolates were from S. equi subsp. zooepidemicus. In the PCR-based detection, we revealed amplicons of 235 bp and 679 bp for confirmation of sodA and seM gene, respectively. In antibiogram, two isolates of S. equi subsp. equi were found resistant to penicillin G, and all other isolates were found sensitive to amoxicillin and streptomycin. Conclusion: The majority of streptococcal infections was due to S. equi subsp. Zooepidemicus, and thus was recognized as a potential pathogen of diseases of equines besides S. equi subsp. equi.

In recent times, reproductive biotechnologies have emerged and started to replace the conventional techniques. It is noteworthy that for sustained livestock productivity, it is imperative to start using these techniques for facing the increasing challenges for productivity, reproduction and health with impending environment conditions. These recent biotechniques, both in male and female, have revolutionized and opened avenues for studying and manipulating the reproductive process both in vitro and in vivo in various livestock species for improving tis efficiency. This review attempts to highlight pros and cons, on the recent developments in reproductive biotechnologies, both in male and female in livestock species.

Aim: The purpose of this study was to determine the virulence genes and serotype of Shiga toxin producing Escherichia coli (STEC) strains isolated from animals and birds. Materials and Methods: A total of 226 different samples viz., fecal, intestinal content, rectal swab and heart blood were collected from different clinically affected/healthy animals and birds and were streaked on McConkeys’ lactose agar and eosin methylene blue agar for isolation of E. coli, confirmed by staining characteristics and biochemical tests. By polymerase chain reaction (PCR) all the E. coli isolates were screened for certain virulence genes, viz., Shiga toxin 1 (stx1), stx2 and eae and enterohemolytic (Ehly) phenotype was observed in washed sheep blood agar plate. All the isolated E. coli strains were forwarded to the National Salmonella and Escherichia Centre, Central Research Institute, Kasauli (Himachal Pradesh) for serotyping. Results: Out of 226 samples 138 yielded E. coli. All the isolates were screened for molecular detection of different virulent genes, viz. stx1, stx2 and eae, based on which 36 (26.08%) were identified as STEC. Among those STEC isolates, 15 (41.67%), 14 (38.89%), 1 (2.78%) exhibited eae, stx2, stx1 alone, respectively, whereas 4 (11.11%) and 2 (5.56%) carried both stx1 and stx2, stx2 and eae, respectively. Among the STEC isolates 22 were belonged to 15 different sero-groups, viz., O2, O20, O22, O25, O43, O60, O69, O90, O91, O95, O106, O118, O130, O162 and O170 and others were untypable. Ehly phenotype was observed in 10 (27.78%) the STEC isolates. Conclusion: The present study concluded that STEC could be isolated from both clinically affected as well as healthy animals and birds. Regular monitoring of more samples from animal and bird origin is important to identify natural reservoir of STEC to prevent zoonotic infection.

The rice−wheat cropping system being the backbone of food security in South-Asia has resulted in soil health deterioration, declining water table, and air pollution affecting livability index of the region. The effect of rice residue retention (RRR), irrigation levels and foliar application of K on wheat grain yield (GY), water use efficiency (WUE) and profitability was tested over three years. RRR increased wheat GY (5224 kg ha−1), above-ground biomass (AGBM = 11.9 t ha−1), tillers per square meter (TPM = 469) and grains per meter square (GrPMS = 13,917) significantly. Relative water content (RWC = 93.8) and WUE (2.45 k gm−3) were also increased significantly by RRR. Consequently, profitability (Net return = 624.4 $ and Benefit to cost (B:C) ratio) was enhanced. Foliar application of K enhanced GY (5151 kg ha−1), AGBM (12 t ha−1), RWC (94.1), SPAD (52.2), WUE (2.40 kg m−3), net returns (625.2 $) and BC ratio (1.62) significantly. RRR increased GY (15.66%) and WUE (17.39%) with additional revenue of 151 $ with only one irrigation at the CRI stage (ICS). RRR adopted over 10% of the area can earn 187 million-US$ annually. RRR if adopted over existing practice on a large area would reduce environmental degradation with an enhanced income to small and marginal farmers.

A study was conducted on the combining ability analysis in a 11×11 half-diallel cross of garden pea (Pisum sativum L.). F1 hybrids (55) along with 11 parental lines were grown in randomized block design with three replications. The mean square due to GCA and SCA were highly significant for all the characters indicating the importance of additive and non-additive genetic components for the characters under study. Further, σ2s was more than σ2g for all the characters suggested the importance of non-additive variance for the improvement of these characters. The parents P2 (GP-17), P5 (GP-473) and P7 (VRP-7) were observed to be good combiners for most characters under study including yield per plant. The crosses P6 × P10, P2 × P8 and P8 × P10 were most promising combinations for total yield per plant. The results indicated the importance of recombinant breeding for effective utilization of non-additive genetic variance in garden pea.

The present study was undertaken to investigate the effect of -tocopherol on in-vitro maturation of vitrified bovine oocytes. The cumulus-oocyte-complexes (COCs) recovered from follicles (3–5 mm diameter) by aspiration and slicing techniques were equally divided into 5 (five) groups consisting 66 COCs in each. The vitrified bovine oocytes were sub-grouped into control and treatment group. The vitrified- and non-vitrified- control group of oocytes were in-vitro matured in TCM-199 media without supplementation of vitamin E. In three vitrified treatment groups, the oocytes were in-vitro cultured in TCM-199 media supplemented with vitamin E (-tocopherol) @ 50 μM, 100 μM and 200 μM, respectively. The mean percentage of cumulus cell expansion and polar body formation in non-vitrified control and vitrified control groups were 85.18±2.57 and 65.38±1.83, and 51.67±1.94 and 30.26±0.16, respectively. The rate of cumulus cell expansion and polar body formation in the oocytes of three vitrified treatment groups media supplemented with vitamin E @ 50 μM, 100 μM and 200 μM were 56.39±3.49 and 34.62±1.83, 69.95±3.20 and 56.23±1.61, and 54.44±4.73 and 31.62±4.50% respectively. Mean percentage of both cumulus cell expansion and polar body formation in the non-vitrified control group were significantly higher than that in the vitrified control group. In the treatment group supplemented with vitamin E @ 100 μM, both cumulus cells expansion and polar body formation were significantly higher than that in media supplemented with 50 μM and 200 μM vitamin E as well as in vitrified control group.

Diarrhea due to Escherichia coli is one of the major economic concerns of bovine industry in the first few weeks after birth. The present study was carried out for characterization of virulence traits of Escherichia coli associated with diarrhea in calves less than 3 months of age and their antimicrobial profiling. Escherichia coli isolates (700) were recovered in this study, which belonged to 25 different serogroups out of which O2 and O6 were most predominant. The isolates were screened for 7 virulence genes, viz. sta, stx1, stx2, lt, f5, f41 and eae. Out of 700 isolates, 65 (9.2%) carried virulence genes either alone (f5, eae, f41, sta, stx1 and stx2) or in different combinations (f41/sta, sta/lt, stx1/eae and f5/stx2). Based on the presence of virulence factors, these isolates were classified as Shiga-toxin producing E. coli (27), enterotoxigenic E. coli (20), enteropathogenic E. coli (15) and 3 were unclassified. These 65 isolates were resistant to commonly used antibacterial agents like amoxicillin, ceftriaxone, cephalexin, cloxacillin, enrofloxacin and gentamicin. Multidrug resistance was also observed in 84.6% of them.This study reveals that PCR based detection of E. coli virotypes can be used in diagnosis and epidemiological studies. Also increasing multidrug resistance for most of the commonly used antibiotics is a matter of concern.

Bioelectrical impedance analysis (BIA) is a method for the assessment of nutritional status. We studied the effect of graft function on nutritional status in postrenal 45 transplant patients with borderline to good allograft function using BIA. We conducted a prospective study (between November 2013 and January 2015) to identify "foot at risk" in the diabetic renal transplant patients at a Tertiary Care Hospital in India. Patients (151) were divided into three groups: diabetic transplant recipients (n = 42), new onset diabetes after transplantation (NODAT) (n = 59), and controls (nondiabetic renal transplant recipients) (n = 50). Foot neuropathy and vasculopathy were assessed by standard methods. Patients with "at risk" feet were given foot care advice. Of the 151 patients, 144 patients were male and seven were female with a male:female ratio of 20:1. Peripheral neuropathy was present among 42.9% of diabetic transplant patients, 35.6% of NODAT patients, and 12% of control patients. Vasculopathy was present in 28.6% of diabetic transplant patients, 23.7% of NODAT patients, and 2% of control patients. On multivariate analysis, patient′s age, mean time interval since transplantation, and HbA1c levels were significantly associated with neuropathy, whereas the duration of diabetes and vibration perception threshold was associated with vasculopathy. After undergoing renal transplantation, a significant number of diabetic and NODAT patients has their "feet at risk" who in future may develop full-blown lesions of the diabetic foot. Proper advice to patients and information to the treating doctor helps to mitigate the risk.

Two index cases of living-related donor renal allografts patients developed C4d-negative rejection. Both cases had negative cytotoxic crossmatch and negative flow crossmatch before transplantation. The serum creatinine levels were tabulated. Both cases experienced augmented anti-T cell therapy (intravenous methyl prednisolone) at the time of rejection, which failed to improve renal function. Meantime, our HLA lab identified circulating anti-class I and/or II HLA antibodies towards donor mismatched antigens by Luminex multiplex bead array. Additional therapy included high-dose IVIg and plasma exchange. The renal function improved significantly. Furthermore, the donor-specific antibody strength decreased after combined plasmapheresis and IVIG therapy. These cases highlight the importance of donor-specific antibody detection by sensitive solid phase assays in the context of C4d-negative ABMR.

Aim: This study was carried out to investigate the effect of incorporation of different level of walnut cake in concentrate mixture on in vitro dry matter degradation in order to determine its level of supplementation in ruminant ration. Materials and Methods: Walnut cake was used @ 0, 10, 15, 20, 25 and 30% level to formulate an iso-nitrogenous concentrate mixtures and designated as T1, T2, T3, T4, T5 and T6 respectively. The different formulae of concentrate mixtures were used for in vitro gas production studies using goat rumen liquor with wheat straw in 40:60 ratio. There is a tremendous growth of the use of the component based software engineering (CBSE) approach for the development of software systems. The selection of the best suited COTS components which fulfils the necessary requirement for the development of software(s) has become a major challenge for the software developers. The complexity of the optimal selection problem increases with an increase in alternative potential COTS components and the corresponding selection criteria. In this research paper, the problem of ranking and selection of Data Base Management Systems (DBMS) components is modeled as a multi-criteria decision making problem. A ‘Fuzzy Distance Based Approach (FDBA)’ method is proposed for the optimal ranking and selection of DBMS COTS components of an e-payment system based on 14 selection criteria grouped under three major categories i.e. ‘Vendor Capabilities’, ‘Business Issues’ and ‘Cost’. The results of this method are compared with other Analytical Hierarchy Process (AHP) which is termed as a typical multi-criteria decision making approach. The proposed methodology is explained with an illustrated example.

Aim: The aim was to assess the effect of multi nutrient block (MNB) supplementation with and without tanniferous leaf meal mixture on feed intake, hematological profile, immune response, and body weight changes of goats that were experimentally infected with Haemonchus contortus. Materials and Methods: Total 12 adult male goats of similar age and body weight (26.49±0.87) were allocated in 3 groups in completely randomized design. MNB supplemented in first two groups i.e. in T1 (no infection) and T2 (H. contortus infection @ 1500 L3/goat) group, while, MNB-condensed tannin (CT) supplemented in T3 (H. contortus infection @ 1500 L3/goat + CT source). All goats were fed concentrate mixture @ 100 g/day/goat, ad lib wheat straw and MNB or MNB-CT to meet their requirement for maintenance. Body weights were recorded and blood and fecal samples were collected at 0 day and thereafter at 15 days intervals for a period of 75 days for the assessment of body weight changes, hematological profile and H. contortus loads.
