Abstract

National audience; L’objectif de cette étude est d’évaluer l’influence de stimulations microbiennes de poules sur le système de défense innée du blanc d’oeuf en utilisant deux modèles expérimentaux. Le premier est basé sur la comparaison de l’activité antibactérienne des blancs d’oeufs de poules axéniques élevées en conditions stériles (Ax), de poules exemptes d’organismes pathogènes spécifiques (EOPS) et de poules conventionnelles (C). Le second modèle compare des oeufs de poules contrôles ou immunostimulées par injection de lipopolysaccharide bactérien (LPS). Les résultats ont montré que les blancs d’oeufs des groupes C et EOPS ont une activité inhibitrice renforcée vis-à-vis de Staphylococcus aureus comparée à ceux du groupe Ax. De même, une meilleure activité antimicrobienne contre ce pathogène est observée pour les blancs d’oeufs du groupe LPS par rapport à ceux du groupe contrôle. En revanche, l’activité anti-Escherichia coli n’est pas modifiée ni par le degré de contamination du milieu des poules (Ax vs EOPS vs C) ni par l’injection de lipopolysaccharide (LPS vs contrôle). L’analyse de l’expression de gènes codants pour des protéines/peptides antimicrobiens de l’oeuf et les mesures des activités du lysozyme et d’antiprotéases dans le blanc n’ont pas permis d’expliquer les stimulations de l’activité antimicrobienne du blanc d’oeuf. Ces résultats suggèrent que les poules peuvent renforcer modérément l’activité antimicrobienne du blanc d’oeuf en réponse à des stimuli microbiens de leur milieu et anticiper les besoins de l’embryon en termes de protection. Les mécanismes moléculaires à l’origine de cette modulation demeurent néanmoins inconnus à ce jour.

Abstract

L’œuf de poule est le lieu de développement de l’embryon, mais aussi un aliment de base et une source importante de molécules dotées d’activités biologiques largement utilisées en santé humaine et dans les industries agroalimentaires, cosmétiques et pharmaceutiques. L’identification des protéines de l’œuf initialement réalisée par des techniques classiques de biochimie, a connu un essor considérable grâce aux développements d’outils d’analyse à haut débit, la publication de la séquence génomique de la poule et l’essor de la bioinformatique. Dans cette revue, nous décrirons quel est l’apport de la génomique fonctionnelle et comment l’analyse transcriptomique et protéomique constituent des avancées majeures dans la connaissance des protéines de l’œuf. Ces avancées ont permis en quelques années l’identification et la caractérisation dans les différents compartiments de l’œuf de centaines de nouveaux composés aux fonctions riches et variées. Des études expérimentales supplémentaires seront nécessaires pour explorer les fonctions des composés nouvellement identifiés dans l’œuf. Ces nouvelles molécules constituent potentiellement des composés biologiquement actifs aux propriétés bénéfiques pour l’Homme ou l’animal. Certains de ces composés pourront être utilisés comme marqueurs biologiques dont le polymorphisme d’un nucléotide simple (SNP) permettra de développer une Sélection Assistée par Marqueurs (SAM) pour améliorer les défenses naturelles de l’œuf et réduire le risque de toxi-infections alimentaires humaines, The avian egg is a nutritious food and also a major source of biologically active compounds that are beneficial for human health. These biologically active molecules are widely used by pharmaceutical, cosmetic and food industries. Egg proteins were previously studied using classical biochemical techniques. The development of high-throughput techniques, the recent publication of the chicken genome sequence, and the development of new bioinformatics tools are major scientific advances leading to the identification and characterisation of a number of minor egg components, which were not previously identified. We illustrate in this review the most recent developments in egg biochemistry (proteomics) and molecular biology (transcriptomics) using recent data on the characterisation of egg proteins. Further experimental studies have been initiated to explore the functional properties of these novel egg proteins. The newly identified molecules may be a source of useful active compounds beneficial for human and animal health. They may be putative biological markers, the single nucleotide polymorphisms (SNP) of which could be used in marker-assisted genetic selection (MAS), to improve egg defencs and to reduce the risk of food-borne disease outbreaks in humans.

Abstract

Concomitantly with the development of the embryo and extra-embryonic structures, initial chicken egg defenses are progressively altered, which suggests that alternative mechanisms appear to protect the embryo until hatching. To better understand the role of extra-embryonic fluids in the chicken egg defense during incubation, we analyzed the biochemical composition and antibacterial properties of the chicken amniotic and allantoic fluids. As for humans, the chicken amniotic fluid protects the embryo against mechanical shocks and microbial infections. Moreover, at the 12th day of incubation, the transfer of antibacterial molecules from the egg white increases the antibacterial potential of the chicken amniotic fluid, thus protecting the embryo until hatching, and probably after, all along its digestive tract, after oral absorption of the amniotic fluid. Some of these antibacterial proteins are specific to oviparous species and/or birds, which highlights the processes of evolution and adaptation of birds to their terrestrial environment. On the other hand, the antibacterial potential of the allantoic fluid still needs to be explored, but our study demonstrated the presence of active proteases, which could contribute to the digestion and recycling of embryonic metabolic wastes.; Avec le développement concomitant de l’embryon et des structures extra-embryonnaires, les défenses initiales de l’oeuf de poule s’altèrent, ce qui suggère la mise en place de systèmes relais pour protéger l’embryon jusqu’à son éclosion. Pour comprendre le rôle des fluides extra-embryonnaires dans cette fonction, nous avons analysé la composition et les propriétés biochimiques et antibactériennes des fluides amniotique et allantoïque de l’oeuf. Comme chez l’humain, le fluide amniotique chez la poule protège l’embryon contre les agressions physiques et microbiennes. De plus, nous avons démontré que le transfert des molécules antibactériennes du blanc au 12ème jour d’incubation augmente son potentiel antibactérien, assurant ainsi une protection de l’embryon jusqu’à l’éclosion, et probablement après, dans son tractus digestif, après l’ingestion du fluide amniotique. Les spécificités phylogénétiques de certaines des protéines identifiées suggèrent un processus d’adaptation des oiseaux à la flore microbienne terrestre. Pour le fluide allantoïque en revanche, nous n’avons pas pu confirmer son rôle antibactérien, mais nous avons mis en évidence la présence de protéases actives qui pourraient contribuer à la digestion et au recyclage des déchets métaboliques de l’embryon.

Abstract

Natural antimicrobial peptides are present in different compartments (eggshell, egg white, and vitelline membranes) of the hen egg and are expected to be involved in the protection of the embryo during its development and to contribute to the production of pathogen-free eggs. In the present study, we used vitelline membranes from hen (Gallus gallus) eggs as a source of avian β-defensin 11 (AvBD11). A purification scheme using affinity chromatography and reverse-phase chromatography was developed. Purified AvBD11 was analyzed by a combination of mass spectrometry approaches to characterize its primary sequence and structure. A monoisotopic molecular species at [M + H](+) of 9,271.56 Da was obtained, and its N- and C-terminal sequences were determined. We also examined posttranslational modifications and identified the presence of 6 internal disulfide bonds. AvBD11 was found to exhibit antimicrobial activity toward both Gram-positive and Gram-negative bacteria.
