Abstract

Fluorescence lifetime imaging (FLIm) and Raman spectroscopy are two promising methods to support morphological intravascular imaging techniques with chemical contrast. Both approaches are complementary and may also be used in combination with OCT/IVUS to add chemical specificity to these morphologic intravascular imaging modalities. In this contribution, both modalities were simultaneously acquired from two human coronary specimens using a bimodal probe. A previously trained SVM model was used to interpret the fluorescence lifetime data; integrated band intensities displayed in RGB false color images were used to interpret the Raman data. Both modalities demonstrate unique strengths and weaknesses and these will be discussed in comparison to histologic analyses from the two coronary arteries imaged.

Abstract

Absorption, scattering, and fluorescence are processes that increase with electric field intensity. The most prominent way to enhance electric field intensity is to use localized or propagating surface plasmon polaritons (SPPs) based on metallic particles and nanostructures. In addition, several other, much less well-known, photonic structures that increase electric field intensity exist. Interference enhancement provided by thin dielectric coatings on reflective substrates is able to provide electric field intensity enhancement over the whole substrate and not only at certain hotspots, thereby being in particular suitable for the spectroscopy of thin surface layers. The same coatings on high refractive index substrates may be used for interference-enhanced total internal reflection-based spectroscopy in much the same way as Kretschmann or Otto configuration for exciting propagating SPPs. The latter configurations can also be used to launch Bloch surface waves on 1D photonic crystal structures for the enhancement of electric field intensity and thereby absorption, scattering, and fluorescence-based spectroscopies. High refractive index substrates alone can also, when nanostructured, enhance infrared absorption or Raman scattering via Mie-type resonances. As a further method, this review will cover recent developments to employ phonon polaritons in the reststrahlen region.

Abstract

In this contribution we present a dual modality fiber optic probe combining fluorescence lifetime imaging (FLIm) and Raman spectroscopy for in vivo endoscopic applications. The presented multi-spectroscopy probe enables efficient excitation and collection of fluorescence lifetime signals for FLIm in the UV/visible wavelength region, as well as of Raman spectra in the near-IR for simultaneous Raman/FLIm imaging. The probe was characterized in terms of its lateral resolution and distance dependency of the Raman and FLIm signals. In addition, the feasibility of the probe for in vivo FLIm and Raman spectral characterization of tissue was demonstrated. Abstract

In biospectroscopy, suitably annotated and statistically independent samples (e. g. patients, batches, etc.) for classifier training and testing are scarce and costly. Learning curves show the model performance as function of the training sample size and can help to determine the sample size needed to train good classifiers. However, building a good model is actually not enough: the performance must also be proven. We discuss learning curves for typical small sample size situations with 5 - 25 independent samples per class. Although the classification models achieve acceptable performance, the learning curve can be completely masked by the random testing uncertainty due to the equally limited test sample size. In consequence, we determine test sample sizes necessary to achieve reasonable precision in the validation and find that 75 - 100 samples will usually be needed to test a good but not perfect classifier. Such a data set will then allow refined sample size planning on the basis of the achieved performance. We also demonstrate how to calculate necessary sample sizes in order to show the superiority of one classifier over another: this often requires hundreds of statistically independent test samples or is even theoretically impossible. We demonstrate our findings with a data set of ca. 2550 Raman spectra of single cells (five classes: erythrocytes, leukocytes and three tumour cell lines BT-20, MCF-7 and OCI-AML3) as well as by an extensive simulation that allows precise determination of the actual performance of the models in question., Comment: The paper is published in Analytica Chimica Acta (special issue "CAC2012").
