- Abstract
Objectives: The Maraviroc Switch (MARCH) study week 48 data demonstrated that maraviroc, a chemokine receptor-5 (CCR5) inhibitor, was a safe and effective switch for the ritonavir-boosted protease inhibitor (PI/r) component of a two nucleos(t)ide reverse transcriptase inhibitor [N(t)RTI] plus PI/r-based antiretroviral regimen in patients with R5-tropic virus. Here we report the durability of this finding. Method(s): MARCH, an international, multicentre, randomized, 96-week open-label switch study, enrolled HIV-1-infected adults with R5-tropic virus who were stable (> 24 weeks) and virologically suppressed [plasma viral load (pVL) < 50 HIV-1 RNA copies/mL]. Participants were randomized to continue their current PI/r-based regimen (PI/r) or to switch to MVC plus two N(t)RTIs (MVC) (1:2 randomization). The primary endpoint was the difference in the proportion with pVL < 200 copies/mL at 96 weeks. The switch arm was defined as noninferior if the lower limit of the 95% confidence interval (CI) for the difference was < -12% in the intention-to-treat (ITT) population. Safety endpoints (the difference in the mean change from baseline or a comparison of proportions) were analysed as key secondary endpoints. Result(s): Eighty-two (PI/r) and 156 (MVC) participants were randomized and included in the ITT analysis; 71 (87%) and 130 (83%) were in follow-up and on therapy at week 96. At week 96, 89.0% and 90.4% in the PI/r and MVC arms, respectively, had pVL < 50 copies/mL (95% CI -6.6, 10.2). Moreover, in those switching away from PI/r, there were significant reductions in mean total cholesterol (differences 0.31 mmol/L; P = 0.02) and triglycerides (difference 0.44 mmol/L; P < 0.001). Changes in CD4 T-cell count, renal function, and serious and nonserious adverse events were similar in the two arms. Conclusion(s): MVC as a switch for a PI/r is safe and effective at maintaining virological suppression while having significant lipid benefits over 96 weeks.Copyright © 2017 British H

El sistema cromatográficoen fase reversa, con detección espectrofotométricaa 266 nm, está constituido por una columna de octadecilsilano C 18 y una fase móvil conformada por metanolagua. Theophylline is a drug widely known for treating asthma and sorne other chronic respiratory diseases. At present time,the blood levels of drugs can be related with eficacy and side effects. There are around 10 drug products ofprogrammedrelease theophyIline forms, commercially available in Colombia. Most of the problems found in the utilization are caused by the non advisable interchange of theophyIline drug products, without any bioequivalency data for them. This study proposes a specific and validated analytical methodology for theophylline in biological fluid s including blood, which is useful for bioavailability and bioequivalencystudies, rutinary monitoring of theophylline blood levels and in forensic chemistry. This metodology has the advantage. touse a mobile phase les s contaminant and cheaper than others previously used. Besides, it makes easier the cleaning of the equipment and extend the useful life of the column. The chromatographic separation system by HPLC-RP, with spectrophotometric detection at 266 nm, ineludes an octadecylsilane C-18 column, and methanol/water as mobile phase.

110. Abnormalities in Trigeminal Nerve Conduction May be Predictor of Death in Chronic Hemodialysis Patients

Peripheral neuropathy is a well-known complication of chronic kidney disease (CKD). Objective: The aim of this study was to analysis the abnormalities in trigeminal nerve conduction in patients treated with hemodialysis (HD) and to determine the relationship between this parameter and death. Methodology: The cross-sectional study included 20 non-diabetic non-alcoholic patients with CKD stage 5 undergoing HD. Conduction of trigeminal nerve was performed using blink reflex. Results: We found clinical evidence of trigeminal neuropathy in 20% of patients and blink reflex abnormalities in 60%. Blink reflex alterations were associated with dialysis time longer and death after one year of follow up (72 ± 19 months and 46 ± 17 months, p=0.03 and 75 vs. 37.5%, p=0.04, respectively). Conclusion: The trigeminal nerve conduction study is useful to detect the abnormalities in peripheral nerves of the ureamic patients under chronic HD and the late response may be associated with death.

Background: One of the factors limiting successful processing of alpaca (Vicugna pacos) semen is the viscosity of seminal plasma. The viscous nature of the collected ejaculate has hindered sperm cryopreservation as well as artificial insemination (AI) under field conditions. Aims: The objective of this investigation was to evaluate recovery, motility, and plasma membrane integrity of alpaca spermatozoa after centrifugation in one of two different solutions at one of three different combinations of speed and time. Methods: A total of 24 ejaculates was recovered from seven reproductively sound Huacaya males using a modified artificial vagina (AV) after training the animals for semen collection. A 2 × 3 factorial treatment arrangement was utilized for this study. Ejaculates were divided into fractions for centrifugation in one of two solutions (Tris extender or PureSperm®80 density gradient solution) at one of three combinations of speed and time (492 × g for 15 min, 1968 × g for 10 min, or 4448 × g for 7 min). The experiment was replicated eight times. Results: Analysis revealed that centrifugation at 4448 × g for 7 min in PureSperm®80 provided a high recovery rate of spermatozoa with the highest sperm motility and functional integrity of plasma membrane post-centrifugation. Conclusion: Results suggest that adoption of this procedure (centrifugation at 4448 × g for 7 min in PureSperm®80) in the initial processing of alpaca ejaculates may enhance subsequent ability to use semen for AI and other assisted reproductive biotechnologies in this species. Determining fatty acid methyl or ethyl esters, in the presence of mono-, di- and tri glycerides, is very important when studying fatty compounds’ methanolysis or ethanolysis, as well as for controlling the quality of petrochemical products. This work presents a useful technique for determining fatty acids methyl or ethyl esters by high temperature gas chromatography in the presence of mono-, di- and triglycerides. Samples were silylated with N, O-bis (trimethylsilyl) tritluoroacetamide (BSTFA) and then passed through a 12 m HT5 column coated with a phenylpolysiloxane-carborane film. Standard methyl and ethyl palmitate, methyl and ethyl oleate, DL-α palmitin, dipalmitin, tripalmitin and triolein solutions were used for calibrating the technique, using tricaprin as internal standard. Retention times and response factors were also determined. The results were employed in following-up palm oil methanolysis and ethanolysis reactions.

113. Tentacle Transcriptome and Venom Proteome of the Pacific Sea Nettle, Chrysaora fuscescens (Cnidaria: Scyphozoa)

- Abstract
Jellyfish venoms are rich sources of toxins designed to capture prey or deter predators, but they can also elicit harmful effects in humans. In this study, an integrated transcriptomic and proteomic approach was used to identify putative toxins and their potential role in the venom of the scyphozoan jellyfish Chrysaora fuscescens. A de novo tentacle transcriptome, containing more than 23,000 contigs, was constructed and used in proteomic analysis of C. fuscescens venom to identify potential toxins. From a total of 163 proteins identified in the venom proteome, 27 were classified as putative toxins and grouped into six protein families: proteinases, venom allergens, C-type lectins, pore-forming toxins, glycoside hydrolases and enzyme inhibitors. Other putative toxins identified in the transcriptome, but not the proteome, included additional proteinases as well as lipases and deoxyribonucleases. Sequence analysis also revealed the presence of ShKT domains in two putative venom proteins from the proteome and an additional 15 from the transcriptome, suggesting potential ion channel blockade or modulatory activities. Comparison of these potential toxins to those from other cnidarians provided insight into their possible roles in C. fuscescens venom and an overview of the diversity of potential toxin families in cnidarian venoms.

The advantages of fatty acid methyl esters as basic oleochemicals over fatty acids, the seventies world energy crisis and the use of those oleochemicals as fuels, have increased research interests on fats and oils trans-esterification. In this document, a review about basic aspects, uses, process variables and problems associated to the production process of fatty acid methyl esters is presented. A global view of recent researches, most of them focused in finding a new catalyst with same activity as the alcohol-soluble hydroxides (NaOH, KOH), and suitable to be used in transforming fats and oils with high levels of free fatty acids and water, avoiding separation problems and reducing process costs, is also discussed.

- Abstract
Genes vary in their likelihood to undergo adaptive evolution. The genomic factors that determine adaptability, however, remain poorly understood. Genes function in the context of molecular networks, with some occupying more important positions than others and thus being likely to be under stronger selective pressures. However, how positive selection distributes across the different parts of molecular networks is still not fully understood. Here, we inferred positive selection using comparative genomics and population genetics approaches through the comparison of 10 mammalian and 270 human genomes, respectively. In agreement with previous results, we found that genes with lower network centralities are more likely to evolve under positive selection (as inferred from divergence data). Surprisingly, polymorphism data yield results in the opposite direction than divergence data: Genes with higher centralities are more likely to have been targeted by recent positive selection during recent human evolution. Our results indicate that the relationship between centrality and the impact of adaptive evolution highly depends on the mode of positive selection and/or the evolutionary time-scale.

This paper describes the performance under cyclic oxidation of (Mo,Cr)
3 Si compound with different Cr additions for possible coating application. Cyclic oxidation was carried out at 1,000 °C at different intervals during 250 h. Oxidized surface samples were analysed by scanning electron microscope where epitaxial oxide scales were observed mainly in samples with higher Cr content which may provide protection against surface oxidation. X-ray diffraction studies have shown the Cr2 O3 and SiO2 formation as the main oxide scale; after analyses, it was found that these oxides are responsible for the best oxidation protection, with 36 at.% Cr being the optimal chromium concentration. At lower chromium concentrations, pest reaction occurred in the oxidized samples at times less than 25 h as a result of the formation of the unstable molybdenum oxide. Las matrices hidrofílicas son uno de los sistemas de liberación controlados más empleados a escala mundial. El reconocido éxito global de este tipo de sistemas está ligado a su manufactura por medio de tecnología convencional para la obtención de comprimidos, además de su bajo costo. Estos sistemas han sido objeto de investigación desde hace ya algunas décadas, mediante el uso de técnicas como la creación de imágenes a partir de resonancia magnética, calorimetría de barrido diferencial y la espectroscopia dieléctrica de baja frecuencia, entre otras. Varios autores han estudiado los diferentes estados del agua dentro de una matriz, así como el estado y los cambios en los estados de los materiales en el tiempo, con el fin de estudiar los mecanismos de cesión de los activos a partir de las matrices, así como para buscar modelos matemáticos que describan la evolución de la concentración en el tiempo. En la actualidad se cuenta con modelos matemáticos de gran utilidad que permiten identificar dichos mecanismos a partir de un análisis de los parámetros de los modelos y que conducen finalmente a predecir la velocidad de liberación a partir de estos sistemas. The hydrophilic matrices are one of the most used controlled delivery systems in the world, due to the simple technology and low cost. A number of publications, over the last decades, have reported investigations in regard with the mechanisms of drug release from hydrophilic matrices. Nevertheless, the mechanisms of drug release from these systems continue to be a matter of debate. Differential scanning calorimetry, low frequency dielectric spectroscopy and nuclear magnetic resonance have been used to examine the distribution of water within ether cellulose matrices, as well as to describe the state of the materials inside the device. The objective is to study the release and hydration rate of hydrophilic matrices in order to contribute to the rationalization of the design of these controlled release systems through mathematical modeling and to obtain a better knowledge of the processes that occur during the release of the drug.

El Sistema de Clasificación Biofarmacéutica (SCB), publicado por Gordon Amidon y cols. en 1995, se basa en un sólido fundamento científico para clasificar un fármaco considerando los parámetros de solubilidad y permeabilidad, factores estrechamente relacionados con el proceso de absorción, y plantea como objetivo, la posibilidad de establecer correlaciones in vitro-in vivo que permitan sustituir los ensayos realizados en humanos por ensayos de disolución in vitro, de acuerdo con la clasificación obtenida para el fármaco. Actualmente la aplicación del SCB está enfocada a los estudios de bioequivalencia para demostrar intercambio de medicamentos, presentados en forma sólida de liberación inmediata, de administración peroral y al aseguramiento de que los cambios realizados durante el escalonamiento o en las formulaciones o procesos de éstos, una vez aprobados, no tengan incidencia en su comportamiento in vivo. La proyección de la aplicación del SCB se está dando hacia su implantación como herramienta en el desarrollo de nuevos fármacos, en el diseño y desarrollo de medicamentos y en la posibilidad de bioexenciones para los sistemas de liberación controlada. The Biopharmaceutics Classification System (BCS), published in 1995 by Gordon Amidon and coworkers, is proposed based on recognizing the underlying process that is controlling the drug absorption rate and extent, namely, drug solubility and intestinal membrane permeability, classifying a drug according to this. Its objective is the possibility to set standards for in vitro drug dissolution testing methodology which will correlate with the in vivo process, avoiding human studies. The BCS can be used to classify drugs and set standards for scale-up and post-approval changes to show that the efficacy has not been altered, as well as standards for in vitro/in vivo correlation for bioequivalence studies, for solid oral immediate release products. About this, exists discusses for further potential applications of the BCS in the development of new drugs and drug products and in the possibility of biowaivers for controlled-release products.

Actualmente, los computadores personales permiten aplicar metodologías de planificación, reduciendo sofisticados problemas en simples estructuras de datos, con apoyo visual de gráficos y tablas, lo que conlleva a un mejor control, tanto de la producción como de los costos. El estudio de caso presentado corresponde al tipo de estas empresas, productora de madera aserrada de pino radiata, en la cual se aplicó una planificación agregada (PA). Se utilizaron datos del último año de producción, y se evaluaron cuatro estrategias productivas relacionadas con la fuerza laboral, el nivel de inventario, de producción y de demanda. Los resultados llevan a concluir que el método propuesto es recomendable para empresas del tamaño estudiado, ya que se puede aplicar fácilmente y permite una adecuada gestión. Con la elección empresarial elegida, fue posible reducir los costos de producción, asegurando políticas laborales y se manejó adecuadamente el nivel de inventario; se redujo en un 1,92% el costo por pulgada producida, ahorrando la empresa más de 6 millones de pesos anuales. The planning of the production in small-scale industries is a usual problem. Since most of the time, engineers are not provided with the time or the appropriate hardware to do a good management of the production. At the time, personal computers allow to apply methodologies of planning, reducing sophisticated problems in simple structures of data, with visual support of graphs and sheet work, which it bears to a better control, both in the production as it is costs. The study case presented corresponds to the type of these industries, producer of saw wood radiate pine, in which an aggregate production (AP) was applied. There was used information from last year´s production, and there were evaluated four productive strategies related to the labor force, the inventory level, production and demand. The results lead to conclude that the proposed method is advisable for companies of the studied size, since it possible to apply easily and allows an appropriate management. With the managerial elected election, it was possible to reduce the costs production, assuring political labor and an appropriately inventory level; the cost diminished in 1.92% for produced inch, saving the company more than 6 million Chilean pesos annual.

134. Comparación del dolor secundario a lipoaspiración tradicional versus lipolisis láser: Estudio prospectivo

La lipoaspiración, técnica tradicional usada para el tratamiento de las lipodistrofias con cicatrices mínimas, ha sido a través de los años mejorada con los avances anestésicos, farmacológicos y de instrumental. A su vez han aparecido otras técnicas que han publicitado ventajas respecto al tiempo de recuperación, sintomatología y resultados estéticos. Una de estas técnicas es la laserlipolisis a la que se atribuye menos dolor, menos equimosis y rápido postoperatorio. No se encuentran trabajos que acrediten estas ventajas, por lo que decidimos estudiar si existen ventajas de laserlipolisis en comparación con la lipoaspiración tradicional respecto del dolor, síntoma importante a considerar en el postoperatorio. Para esto se diseñó un trabajo prospectivo randomizado con una muestra de 60 pacientes a los cuales se aplicaron las dos técnicas en diferentes áreas y al mismo paciente. Se evaluó el dolor con escala numérica analógica a las 4 horas, 48 horas y 5 días de postoperatorio. Los resultados mostraron que la laserlipolisis presenta menos dolor en los muslos medido a las 48 horas y 5 días de la intervención. No hay diferencias entre estas dos técnicas en las otras zonas medidas como abdomen, áreas pretrocantéreas ("alforjas"), zona lumbar y pectoral. Considerando todos los pacientes sin diferenciar la técnica usada, las "alforjas" a las 4 horas duelen menos que el abdomen y a los 5 días duelen menos los muslos que el abdomen. No encontramos relación entre volumen aspirado e intensidad del dolor. The traditional suction-assisted lipoplasty, is a well established and commonly performed technique used for the treatment of lipodystrophias, which has undergone improvements with advances in anesthetics, pharmacology and introduction of new instruments. New techniques have appeared, promising advantages in recovery periods, symptomathology and aesthetic results when compared to the traditional suction-assisted lipoplasty. One of these newly introduced techniques is the laser-assisted lipoplasty (laserlipolysis), which claims less pain, less bruising and faster postoperative recovery. No evidence has been published to support these advantages when comparing laser-assisted lipoplasty to the traditional method, specifically when refering to pain, an important symptom to be taken into consideration in the postoperative period. For this reason we designed a prospective, randomized clinical study comprising 60 patients to which the two techniques were applied to different body areas. Pain was assessed using the numeric analog scale at 4 and 48 hours, and at 5 days after surgery. Evidence showed that laserlipolysis results in less pain of the thighs measured at 48 hours and 5 days after surgery. No difference in pain was found between both techniques on the other body areas evaluated (abdomen, trochanteric bulge, lumbar and pectoral areas). Assesing all patients, independent of the technique used, trochanteric bulge at 4 hours was found to be less painful than the abdominal area, and at 5 day evaluation the thighs were less painful than the abdominal area. No relationshipwas found between pain intensity and lipoaspirated volume.

Se analiza la competitividad en los principales mercados de las exportaciones chilenas de madera aserrada de pino radiata en el período 1994-2004 (Contreras, 2006). Se aplica la metodología "Competitive Análisis of Nation" (CAN), creada por la Comisión Económica para América Latina y el Caribe (CEPAL), que permite obtener una visión dinámica de las situaciones de competitividad del producto en los mercados estudiados. Los resultados se compararon con un estudio similar del periodo 1987 a 1997, realizado por Fuenzalida (1999). El análisis de la competitividad en el periodo 1994-2004 muestra que el producto tiene una posición competitiva de Estrella Naciente, en los mercados de China, España, Estados Unidos. y Marruecos; de Estrella Menguante en los mercados de México y Canadá; de Oportunidad Perdida en Japón, Arabia Saudita y Reino Unido; y de Retroceso, en Corea del Sur. Por otro lado, comparando los dos periodos en estudio, se encontró que los mercados de Estados Unidos, Corea del Sur, Japón, Reino Unido y Arabia Saudita, que se repiten en ambos periodos, disminuyeron su posición competitiva del primer periodo al segundo. Considerando que en el ámbito mundial el diseño de nuevos fármacos involucra un costo muy alto, tanto en tiempo como en dinero, la preocupación en los años recientes se ha centrado en diseñar y desarrollar medicamentos que controlen la liberación del activo, de manera que se les pueda dar un valor agregado a moléculas ya existentes, lo que significa una menor inversión en términos de costos. En este sentido, para la presente revisión se seleccionó el sistema de liberación osmótica, uno de los medicamentos de liberación controlada con más participación en el mercado en el mundo. En este escrito se muestran los aspectos fisicoquímicos relacionados con su formulación, las clases de sistemas osmóticos de administración oral existentes y sus aplicaciones, así como el estado del arte en este contexto. Hacia el futuro, estos sistemas osmóticos parecen ser muy promisorios, debido a sus ventajas y gran mercado potencial. In recent years, the worldwide, the attention has been focused on the development of new drug products with controlled delivery. The reason for this new tendency is relatively low cost and time required to development this kind of product as compared to a new drug. In this sense, the osmotic systems, controlled delivery drug products, have been chosen for the present review to illustrate their physicochemical principles that govern their formulation. Different types of oral osmotic systems and their applications are also discussed. Finally, some studies related to this subject were reviewed. Because of their advantages and strong market potential, the future of osmotic systems seems so good.

From the preoccupation by the increase of the depression frequency and the diminution of the age of the people who suffer this type of problem, as well as some indicators that accompanies it and that is within the concept of psycho-social misalignment, arise the necessity to study these phenomena in the scholastic population. Objectives: demostrate the interdependent relation of the variable adjustment psycho-social and depression in students of the city of Lima, Peru, in function to the type of school (state and private) and sort. Design: Descriptive comparative and correlational. Materials and methods: The sample was constituted by 480 students of state and particular educative centers, whose ages oscillate between the 12 and 17 years. For the evaluation of the variable psychosocial adjustment, the Inventory of Adaptation of Conduct of M. Victoria de la Cruz and Agustín Cordero was used, and to evaluate the variable depression, it was used the Questionnaire of Depression for children and adolescents of M. Lang and M. Tisher. The statistical analysis derived in the first place, in an analysis of validity and trustworthiness of the tests, and secondly, a inferencial analysis for the contrast of the raised hypotheses was made. Results: One has been that the two used tests are highly valid and reliable., A partir de la preocupación por el aumento de la frecuencia de depresión y la disminución de la edad de las personas que padecen este tipo de problema, así como algunos indicadores que lo acompañan y que se encuentra dentro del concepto de desajuste psicosocial, surge la necesidad de estudiar dichos fenómenos en la población escolar. Objetivos: Demostrar la relación interdependiente de las variables ajuste psicosocial y depresión en escolares del nivel secundario de Lima Metropolitana, en función al tipo de colegio (estatal y particular) y género. Diseño: Es una investigación de tipo básica y de diseño descriptivo comparativo y correlacional. Material y método: La muestra estuvo constituida por 480 escolares de centros educativos estatales y particulares, cuyas edades oscilan entre los 12 y 17 años. Para la evaluación de la variable ajuste psicosocial, se utilizó el Inventario de Adaptación de Conducta de M. Victoria de la Cruz y Agustín Cordero, y para evaluar la variable depresión, se utilizó el Cuestionario de Depresión para niños y adolescentes de M. Lang y M. Tisher. El análisis estadístico derivó en primer lugar, en un análisis de validez y confiabilidad de las pruebas, y en segundo lugar, se realizó un análisis inferencial para la contrastación de las hipótesis planteadas. Resultados: Se ha encontrado que las dos pruebas utilizadas son altamente válidas y confiables.

139. Análisis del efecto del ph en la cinética de liberación de la teofilina en un medicamento de liberación programada: parte i. microgránulos encapsulados recubiertos con goma laca

En los medicamentos de liberación modificada de administración peroral, es importante garantizar que la liberación del principio activo, sea independiente del pH, para evitar variaciones significativas en los niveles sanguíneos, que puedan conducir a fallas terapéuticas, o a manifestaciones de toxicidad. Uno de los medicamentos que más se comercializa en formas de liberación modificada es la teofilina, existiendo en el mercado más de una docena de marcas de diversos sistemas de entrega como tabletas, cápsulas con pellets o con microgránulos y en varias concentraciones como 100, 125, 200 y 300 mg. En este trabajo se presentan los resultados de un producto comercializado como microgránulos encapsulados con 125 y 300 mg de teofilina. Se evaluó la liberación de la teofilina a 8 valores de pH que cubren las condiciones del tracto gastrointestinal. Se encontró que la liberación de la teofilina se ajusta a lo establecido en la USP 25 para los ensayos 2 y 4, pero tanto la cinética como la velocidad de liberación son altamente dependientes del pH. Para valores de pH por debajo de 6.4 el proceso de liberación se ajusta a un modelo de primer orden cinético con una vida media de disolución que va de 161 min. hasta 247 min. a medida que el pH se acidifica. Para un pH de 6.4 a 8.0 la disolución de la teofilina se ajusta a un modelo de orden cero y la vida media de disolución disminuye de 92 a 27 minutos, en la medida que el pH se vuelve más alcalino. In the modified release per oral medicaments, is important to assure the independence of drug deliverance and pH, in order to avoid significative variations in blood levels that can cause therapeutics lacks or toxic effects. One of the most commercialized medicaments in controlled delivery forms is the theophylline, being in the market more than one dozen of registered trademarks, many delivery systems such as tablets, capsules with pellets or microgranules and in different concentrations: 100, 125, 200 and 300 mg. This written work shows the results of a product marketing as capsulated microgranules with 125 and 300 mg of theophylline. We evaluated the delivery of theophylline at 8 pH values, which cover the conditions inside the gastrointestinal tract. We found that theophylline delivery is within the USP 25 requirements for 2 and 4 assays, but the kinetics and the delivery rate are highly dependent on pH. For pH values lower than 6.4 the delivery process follows a first order kinetics model and a dissolution shelf life between 161 min. and 247 min. as the pH is acidified. For pH values from 6.4 to 8.0 theophylline dissolution follows zero order kinetics and the half-life dissolution decreases from 92 min. to 27 min. as the pH becomes more alkaline.

140. Estudio de bioequivalencia in vitro de cuatro productos de amoxicilina del mercado colombiano

Los estudios de bioequivalencia permiten demostrar si el principio activo tiene el mismo desempeño farmacocinético en el medicamento genérico que en el innovador, para así poderlos considerar intercambiables y garantizar que la eficacia clínica y seguridad del innovador se aplica al genérico. La amoxicilina pertenece a la clase I en el Sistema de Clasificación Biofarmacéutica y por lo tanto se justifica la realización de estudios de bioequivalencia por medio de perfiles de disolución. En esta investigación se efectuó un estudio de bioequivalencia in vitro de 4 productos de amoxicilina trihidrato del mercado colombiano en cápsulas de 500 mg. Se concluye que existe bioequivalencia entre 2 productos del estudio, por medio de perfiles de disolución in vitro. Sin embargo, los análisis de control de calidad de algunos de estos medicamentos permitieron detectar lotes de amoxicilina fuera de los estándares establecidos. Bioequivalence studies let to assume that pharmacokinetic processes of an active principle are the same in generic drugs as in their correspondent innovator drugs, they are interchangeable, and both clinical efficacy and safety of the innovator can be applied to the generic drug. In agreement with Biopharmaceutics Classification System, amoxycillin belongs to first class and therefore the execution of bioequivalence studies by dissolution profiles has been justified. An in vitro bioequivalence study of four commercially available amoxycillin trihydrate (500 mg capsules) preparations from Colombia was carried out in this research. By in vitro dissolution profiles, it was concluded from this study that 2 preparations are bioequivalent. However, the quality control analyses of some drugs permitted to detect sub-standard amoxycillin batches.

141. Preformulación de tabletas de teofilina al 65% del tipo matriz hidrofílica y evaluación de la cinética de liberación modificada del fármaco

- Abstract
Fueron elaborados 6 lotes de tabletas a partir de teofilina, Methocel E4MCRÃ’ y Ludipress LCEÃ’, se mantuvo constante la proporción del fármaco y se modificó la composición del excipiente, para estudiar el efecto de la formulación sobre la liberación, la que se evaluó a ocho pHs. El efecto del Methocel E4MCRÃ’ en la formulación 2 fue desintegrante, para la 3 este efecto se presentó en una menor magnitud y fue contrarrestado por la formación del gel a causa de la hidratación del Methocel E4MCRÃ’. En las formulaciones 4, 5 y 6, los perfiles de disolución se caracterizaron por la aparición de dos fases con el mismo orden cinético, donde la primera fase corresponde a la disolución rápida de la teofilina ubicada sobre la superficie de la tableta, y la segunda a una liberación más uniforme por la difusión de la teofilina a través del gel formado. Los perfiles para todas las formulaciones fueron comparados con los perfiles de dos productos del mercado Colombiano administrados para 12 horas, siendo la formulación 4 la que presentó las características deseadas. Controlled Release (CR) tablets were prepared using the following: anhydrous theophylline (Parke Davis) Methocel E4MCR® (supplied by Colorcon Sucursal de Colombia, Dow Chemical Company), Ludipress LCEÃ’ (supplied by BASF Fine Chemicals) and Magnesium Stearate. There were studied the CR tablets at eight pHs. The Methocel E4MCRÃ’ effect in the lowest Methocel E4MCRÃ’ formulation was disintegrant. 4, 5 and 6 formulations dissolution profiles were characterized by an initial burst effect with a greater amount of drug released, followed by a more-uniform release of drug. In the initial phase, the drug release was possibly due to the dissolution of free drug on the surface of the matrix and erosion. In the second phase, a linear relationship of the natural logarithm of remanent percentage drug to time was observed from such formulations, suggesting a diffusion-controlled mechanism of drug release. There were compared the dissolutions profiles of all the formulations with two formulations already marketed, it was found that 4 formulation showed the desired dissolution profile.
