The effect of microsphere fabrication methods on the stability and release kinetics of ovalbumin encapsulated in polyanhydride microspheres was investigated. The polyanhydrides used were poly(sebacic anhydride) (poly(SA)) and a 20:80 random copolymer of poly[1,6-bis(p-carboxyphenoxy)hexane] (poly(CPH)) and poly(SA). Microspheres were fabricated using three double emulsion methods (water/oil/water, water/oil/oil and solid/oil/oil) and cryogenic atomization. The encapsulation efficiency was highest for cryogenic atomization and lowest when the w/o/w technique was used. Microspheres fabricated by the s/o/o method had the largest initial burst of released protein. All the methods resulted in zero-order release of the protein after the burst. The release of ovalbumin from poly(SA) and 20:80 (CPH:SA) microspheres lasted approximately 3 and approximately 6 weeks, respectively. For all fabrication methods the primary structure of released ovalbumin was conserved as determined by gel electrophoresis. The secondary structure of ovalbumin encapsulated in 20:80 (CPH:SA) w/o/w microspheres was not conserved.

When encapsulating proteins in polymer microspheres for sustained drug delivery there are three stages during which the stability of the protein must be maintained: (1) the fabrication of the microspheres, (2) the storage of the microspheres, and (3) the release of the encapsulated protein. This study focuses on the effects of polymer degradation products on the primary, secondary, and tertiary structure of tetanus toxoid, ovalbumin (Ova), and lysozyme after incubation for 0 or 20 days in the presence of ester (lactic acid and glycolic acid) and anhydride (sebacic acid and 1,6-bis(p-carboxyphenoxy)hexane) monomers. The structure and antigenicity or enzymatic activity of each protein in the presence of each monomer was quantified. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis, circular dichroism, and fluorescence spectroscopy were used to assess/evaluate the primary, secondary, and tertiary structures of the proteins, respectively. Enzyme-linked immunosorbent assay was used to measure changes in the antigenicity of tetanus toxoid and Ova and a fluorescence-based assay was used to determine the enzymatic activity of lysozyme. Tetanus toxoid was found to be the most stable in the presence of anhydride monomers, while Ova was most stable in the presence of sebacic acid, and lysozyme was stable when incubated with all of the monomers studied.

Salicylate-based poly(anhydride-esters), collectively referred to as PolyAspirin, hydrolytically degrade into salicylic acid, a nonsteroidal anti-inflammatory drug (NSAID). The variations of the poly(anhydride-ester) investigated in this article are linked by adipic acid, suberic acid, or sebacic acid. To elucidate the erosion mechanism of these polymers, water uptake, mass loss, contact angle, and changes in device thickness were monitored as a function of in vitro degradation time. The polymers examined here appeared to primarily undergo surface erosion. The adipate homopolymer absorbed the most water and therefore swelled more than the other versions of the poly(anhydride-ester). Additionally, the adipate homopolymer eroded most quickly. Increasing the length of the linker moiety decreased the driving force for hydrolysis, which prolonged the lifetime of the polymer sample., ((c) 2005 Wiley Periodicals, Inc.)

We have designed a new synthesis route to create polyanhydrides based on monomers that contain hydrophilic entities within highly hydrophobic backbones. The method results in polyanhydrides that can be easily processed into drug-containing tablets. The synthesis, characterization, and erosion studies of polyanhydride copolymers based on 1,6-bis(p-carboxyphenoxy)hexane (CPH), which is highly hydrophobic, and 1,8-bis(p-carboxyphenoxy)-3,6-dioxaoctane (CPTEG), which has hydrophilic oligomeric ethylene glycol segments in the monomer unit, was performed using a combination of molecular spectroscopy, thermal analysis, gravimetry, and scanning electron microscopy. The studies demonstrate that by increasing the CPH content in the CPTEG:CPH copolymers, the erosion of the system can be tailored from bulk-eroding to surface-eroding mechanism. These systems have promise as protein carriers.

This chapter reviews recent developments in biodegradable synthetic polymers focusing on tailoring polymer structures to meet material specification for emerging applications such as tissue engineered products and therapies. Major classes and new families of synthetic polymers are discussed with regard to synthesis, properties and biodegradability, and known degradation modes and products are summarized based on studies reported during the past 10-15 years. Polyesters and their copolymers, polyurethanes, polyphosphazenes, polyanhydrides, polycarbonates, polyesteramides and recently developed injectable polymer systems based on polypropylenefumarates, polyurethanes and acrylate/urethane systems are reviewed. Polyesters such as polyglycolides, polylactides and their copolymers still remain as the major class of synthetic biodegradable polymers with products in clinical use. Although various copolymerization methods have addressed needs of different applications, release of acidic degradation products, processing difficulties and limited range of mechanical properties remains as major disadvantages of this family of polymers. Injectable polymers based on urethane and urethane/acrylate have shown great promise in developing delivery systems for tissue engineered products and therapies.

Major progresses in the development of parenteral sustained-release systems have been made in recent years as evidenced by the regulatory approval and market launch of several new products. Both the availability of novel carrier materials and the advances in method of fabrication have contributed to these commercial successes. With the formulation challenges associated with biologics, new delivery systems have also been evolved specifically to address the unmet needs in the parenteral sustained release of proteins. In this review paper, different new carriers systems and preparation methods are discussed with special focus on their applications to biologicals.

In this work, a novel diacid monomer was synthesized in a very convenient scheme. The monomer is derived from naturally occurring products and emits strong fluorescence when polymerized to polyanhydride. The chemical structure of the monomer dCPS is as follows: HOC(O)ArOC(O)(CH2)2C(O)O--Ar--COOH. Copolyanhydrides composed of dCPS and sebacic acid were further prepared by melt copolycondensation, and characterized by IR, NMR, UV-Vis, DSC and fluorometry. The emission wavelength (lambda(em)) of the copolymers could be tuned by the excitation wavelength (lambda(ex)). Fluorescence intensity increased with the increase of dCPS content. The microspheres fabricated from the copolymer with dCPS content as low as 10% could be clearly visualized with fluorescence microscopy. Either blue or green images of the microspheres could be captured with an excitation of UV and visible light. The degradation rate of the copolyanhydrides decreased as the dCPS fraction increased, and the degradation duration could be modulated from several days to more than three months. In addition, it was found that the copolyanhydrides displayed surface degradation characteristics. In view of the advantages of the novel copolyanhydrides, such as easy preparation, unique inherent luminescent properties, and widely adjustable degradation rate, they might be useful for biomedical engineering.

Many drugs that are administered in an unmodified form by conventional systemic routes fail to reach target organs in an effective concentration, or are not effective over a length of time due to a facile metabolism. Various types of targeting delivery systems and devices have been tried over a long period of time to overcome these problems. Targeted delivery or localized drug delivery offers an advantage of reduced body burden and systemic toxicity of the drugs, especially useful for highly toxic drugs like anticancer agents. Local drug delivery via polymer is a simple approach and hypothesized to avoid the above stated problems. Polyanhydrides are a unique class of polymer for drug delivery because some of them demonstrate a near zero order drug release and relatively rapid biodegradation in vivo. Further, the release rate of polyanhydride fabricated device can be altered over a thousand fold by simple changes in the polymer backbone. Hence, these are one of the best-suited polymers for drug delivery, with biodegradability and biocompatibility. The review focuses on the advantages of polyanhydride carriers in localized drug delivery along with their degradability behavior, toxicological profile and role in various disease conditions.

Aromatic diacids were synthesized from chlorinated triethylene or pentaethylene glycols and p-hydroxy benzoic acid. The diacids were acetylated with acetic anhydride to produce homopolymers of polyanhydrides for controlled release applications such as the stabilization of proteins and drugs. Adding ethylene glycol segments into the acid monomer allows tailoring of the hydrophobicity of the polyanhydrides, which in turn dictates the solubility of molecules within the polymer matrix and degradation rate of the polymer. The glycol containing polyanhydrides were characterized by NMR, GPC, DSC and dissolution testing. The release characteristics and mechanism of the new polyanhydrides were evaluated using hydrophilic and hydrophobic dyes.

This report presents the formulation and surface modification of poly(ester-anhydride) copolymers. Two synthetic schemes were used to generate a variety of copolymers with varying compositions and molecular weights. These polymers were formulated into both micro- and nanospheres using a water-in-oil-in-water double emulsion technique. Microspheres were between 3 and 10 microm in diameter; whereas, nanospheres were between 300 and 500 nm. Finally, both micro- and nanospheres were surface modified using cystamine and quantified, following cystamine reduction, with DTNB (Ellman's reagent). Surface labeling was observed at 0.20-35 micromol/g for microspheres and 0.6-100 micromol/g for nanospheres and followed the expected trend of increased labeling with increased polyanhydride content.

Biochronomer (AP Pharma) is a fourth-generation poly(ortho ester) prepared by the condensation of diols and a diketene acetal. The polymer contains a copolymerised latent acid whose concentration controls erosion rate. The polymer has been shown to undergo a surface erosion process and a number of applications have been explored. Among these, the delivery of plasmid DNA for vaccines is currently of most interest. This application takes advantage of the acid-labile nature of the polymer, which leads to rapid polymer hydrolysis and hence rapid release of plasmid DNA once internalised in the acidic environment within the endosomes, and the non-acidic environment within the polymer that conserves plasmid DNA conformation. A low molecular semisolid polymer is now in Phase II clinical trials for the delivery of mepivacaine to control postoperative pain, and in Phase I clinical trials for the systemic delivery of granisetron to control nausea.

p-Carboxyphenoxy propane (CPP) prepolymer consisting of 4 units and sebacic acid (SA) prepolymer consisting of about 10 units were synthesized by reacting CPP and SA in the presence of excess acetic anhydride, respectively. Polyanhydride, poly(CPP-SA) copolymers were copolymerized by a melt polycondensation process with a mixture of CPP and SA prepolymer. Copolymers of average molecular weight up to 110,000 g/mol were achieved. The crystallinity of poly(CPP-SA) copolymers was decreased by the addition of the CPP homopolymer segment to SA homopolymer. Poly(CPP-SA) copolymers gradually degraded for period of 10 days. No large difference of weight loss observed according to molecular weight variation of poly(CPP-SA) copolymers. BCNU release from wafers fabricated by poly(CPP-SA) showed a sustained release pattern with no initial burst and delay of drug release.

Poly(anhydride-esters), based on carboxyphenoxydecanoate (CPD), are biocompatible polymers that hydrolytically degrade. The mechanical properties of the poly(anhydride-esters) can be altered by copolymerization with para-carboxyphenoxyhexane (pCPH). Mechanical properties of three CPD:pCPH compositions (30:70, 40:60, and 50:50) are reported as a function of hydrolytic degradation. The mechanical characteristics evaluated were tensile modulus at 1% strain (E(1%)), tensile strength (sigma(B)), ultimate elongation (epsilon(B)), and toughness (E(r)). The 30:70 CPD:pCPH fibers maintained higher values for tensile modulus at all time points than the two other fiber compositions. In addition, the 30:70 CPD:pCPH fibers maintained lower values for both tensile strength and toughness than the two other fiber compositions. These phenomena resulted from the brittle nature of pCPH, the major component of the 30:70 CPD:pCPH fibers; increasing the pCPH concentration in the polymer lowers both tensile strength and toughness of the polymer by decreasing ductility. With increasing amounts of pCPH, the hydrolytic degradation occurred more slowly, as reflected in the copolymers' improved ability to retain their mechanical properties. Therefore, copolymerization is useful for controlling the mechanical properties of the fibers as well as the polymer degradation rate, which ultimately determines the rate at which physically or chemically encapsulated drugs can be released., (Copyright 2004 Wiley Periodicals, Inc. Sustained delivery of DNA has the potential to enhance long-term gene therapy; however, precise control of a wide range of DNA release profiles may be needed. In this work, multifunctional anhydride monomers were photocrosslinked to produce hydrophobic, highly crosslinked polymer networks that degrade by surface erosion. Surface-eroding polymers can deliver molecules of a wide range of sizes at sustained, steady rates, which is advantageous for DNA delivery, where the high molecular weight may complicate control of the release profiles. When plasmid DNA was released from photocrosslinked polyanhydride matrices, DNA recovery was low (approximately 25%). Electrophoresis indicated that the plasmid DNA was released primarily in the relaxed and supercoiled forms, yet the relative fraction of released DNA in the supercoiled form decreased over time. To improve DNA recovery and reduce the damaging effects of polymer degradation, DNA was pre-encapsulated in alginate microparticles, which served as a temporary coating that quickly dissolved upon microparticle release from the polyanhydride matrix. As photocrosslinked polyanhydrides have highly predictable drug release profiles that depend on the polymer erosion rate and implant geometry and not on the entrapped molecule size, they can serve dual purposes in many biomaterial applications where structural support and drug release would be beneficial., (Copyright 2004 Elsevier B.V.)
