BACKGROUND Mandarins and mandarin hybrids have excellent flavor and color attributes, making them good candidates for consumption as fresh fruit. When processed into juice, however, they are less palatable, as they develop delayed bitterness when stored for a period of time. In this study the kinetics of delayed bitterness in two citrus mandarin hybrid siblings, 'Ambersweet' and USDA 1-105-106, was explored by sensory and instrumental analyses. In addition to the bitter limonoids, other quality factors (i.e. sugars, acids, pH, soluble solids content ( SSC), titratable acidity ( TA) and the ratio SSC/ TA) were also measured. RESULTS The two citrus hybrid siblings had different chemical profiles, which were perceived by taste panels. USDA 1-105-106 developed delayed bitterness when the juice was stored for more than 4 h, similar to juice from 'Navel' oranges, but 'Ambersweet' did not. Bitterness in 'Ambersweet' was more affected by harvest maturity, as juice from earlier harvest had lower SSC but higher TA and bitter limonoids. CONCLUSION Since juice of USDA 1-105-106 shows delayed bitterness when stored for more than 4 h, this cultivar is not suitable for juice processing. Our finding that siblings can differ in chemical and sensory properties emphasize the importance of post-processing storage studies before releasing cultivars for juice. Published 2015. Background: Although many of the volatile constituents of flavor and aroma in citrus have been identified, the knowledge of molecular mechanisms and regulation of volatile production are very limited. Our aim was to understand mechanisms of flavor volatile production and regulation in mandarin fruit. Result: Fruits of two mandarin hybrids, Temple and Murcott with contrasting volatile and non- volatile profiles, were collected at three developmental stages. A combination of methods, including the isobaric tags for relative and absolute quantification (iTRAQ), quantitative real-time polymerase chain reaction, gas chromatography, and high-performance liquid chromatography, was used to identify proteins, measure gene expression levels, volatiles, sugars, organic acids and carotenoids. Two thirds of differentially expressed proteins were identified in the pathways of glycolysis, citric acid cycle, amino acid, sugar and starch metabolism. An enzyme encoding valencene synthase gene (Cstps1) was more abundant in Temple than in Murcott. Valencene accounted for 9.4% of total volatile content in Temple, whereas no valencene was detected in Murcott fruit. Murcott expression of Cstps1 is severely reduced. Conclusion: We showed that the diversion of valencene and other sesquiterpenes into the terpenoid pathway together with high production of apocarotenoid volatiles might have resulted in the lower concentration of carotenoids in Temple fruit. Thresholds for flavour volatiles have been traditionally calculated in water or air, but they may vary widely in more complex matrices. Thresholds of key aroma compounds of orange juice (OJ) were determined in a deodorized OJ matrix. The three-alternative-forced-choice (3-AFC) method was used (ASTM: E-679). Untrained panelists, 33-58 in number, were presented with deodorized orange juice samples arranged in five rows of three samples, corresponding to five spiking levels, each separated by a factor of 3, with a 3-AFC presentation at each level. The test was repeated at least three times for experienced panelists. Odour thresholds in the orange juice matrix were 15 times (citral, hexanal) to over 200 times ( β-pinene, limonene) higher than published values in water. Retronasal odour thresholds were more consistent with published values, being higher only by 2-60-fold, except for octanal which was higher by 187-fold. Sweet orange (Citrus sinensis) exhibits limited genetic diversity and high susceptibility to Huanglongbing (HLB). Breeding HLB-tolerant orange-like hybrids is in dire need. However, our understanding of the key compounds responsible for orange flavor and their genetic regulation remains elusive. Evaluating 179 juice samples, including oranges, mandarins, Poncirus trifoliata, and hybrids, distinct volatile compositions were found. A random forest model predicted untrained samples with 78% accuracy and identified 26 compounds crucial for orange flavor. Notably, seven esters differentiated orange from mandarin flavor. Cluster analysis showed six esters with shared genetic control. Differential gene expression analysis identified C. sinensis alcohol acyltransferase 1 (CsAAT1) responsible for ester production in orange. Its activity was validated through overexpression assays. Phylogeny revealed the functional allele was inherited from pummelo. A SNP-based DNA marker in the coding region accurately predicted phenotypes. An herbaceous/grassy-like flavor has been reported by Chilean producers of Regina sweet cherry. There are no previous academic reports related to this flavor occurrence. Sweet cherries from five phenological stages were collected from six orchards with high herbaceous flavor incidence spanning Chilean production zones during the 2019/2020 season. Four experienced panelists tasted the fruit to identify the off-flavor incidence and intensity from four phenological stages, and the same cherries were analyzed for volatile compounds. Thirty-nine volatiles were identified and semi-quantified using solid-phase microextraction (SPME) and GC-MS. The highest off-flavor incidence was found at the bright red (stage 3) and mahogany colors (stage 4). No single volatile explained the herbaceous flavor consistently among orchards. However, it appeared that the off-flavor was related to delayed ripening in cherries, with more C6 aldehydes and less esters. Furthermore, rainfall and the elevation of the orchard had a significant effect on the incidence of off-flavor. Preharvest practices that promote fruit ripening along with avoiding early harvests are recommended to reduce the incidence of herbaceous flavor in Regina. [ABSTRACT FROM AUTHOR]

Avocado (Persea americana Mill.) is a high-value fruit that continues to increase in consumer demand. A population of 'Hass'-'Bacon' hybrids was planted at USDA-ARS, Fort Pierce, as part of a study to find selections with good horticultural and postharvest quality traits for Florida. Extensive phenotypic data on quality were collected over 3 years. Ten selections were identified in 2014 and 2015 with promising fruit quality and postharvest shelf life characteristics and were tested in sensory panels using store-bought 'Hass' as the standard. In general, the selections had fruit quality similar to commercial 'Hass'. Avocados that were most liked were described as creamy in texture with buttery and nutty flavor. Only one selection (R7T54 in 2014) and one store-bought control ('Hass' in 2015) were disliked, which was associated with greater firmness at the time of evaluation, likely relating to insufficient postharvest conditioning. Furthermore, CA 'Hass' commercial requirements for minimum dry matter (20.8%) were generally achieved by these selections under Florida conditions, ranging from 18.4% to 25.7%. The effect of controlled-release chlorine dioxide (ClO2) gas on the safety and quality of grapefruit was studied. The experiments were run under controlled chamber systems with inoculated fruit, and in boxed fruit under commercial conditions. For the inoculation test, fruit artificially inoculated with either Escherichia coli or Penicillium digitatum, or naturally inoculated Xanthomonas citri ssp. citri (Xcc) (fruits with citrus canker lesions), were incubated in a chamber containing a dose equivalent to 0-60 mg⋅L-1 of pure ClO2 as an antimicrobial agent. After 24 hours, the microbial population on treated grapefruit was significantly reduced compared with that of control fruit: a dosage of 5 mg⋅L-1 completely inhibit the growth of E. coli and P. digitatum, but a dosage of 60 mg⋅L-1 was needed to completely kill Xcc. For the simulated commercial experiment, fruit were harvested in late Oct. 2015 passed through a commercial packing line, and packed in 29 L citrus boxes. ClO2 packets were attached to the top lids with the following five treatments: fast-release, slow-release, slow/fast-release combination (each containing 14.5 mg⋅L-1 of pure ClO2), double dose fast-release (containing 29 mg⋅L-1 of ClO2), and control. After 6 weeks of storage at 10 °C (to simulate storage and transportation) + 1 week of storage at 20 °C (to simulate retail marketing), the fruit quality was evaluated. The slow-release treatment at standard dose exhibited the best antimicrobial activity, reducing total aerobic bacterial count and yeast/mold count by 0.95 and 0.94 log colony-forming units (cfu)/g of fruit, respectively, and maintained the best visual, sensory, and overall quality. Peach-to-nectarine mutants showed broad pleiotropic effects on fruit size, taste, and aroma, in addition to hairlessness. In this study, we compared nine fruit attributes and 27 detected volatiles in the peach progenitor, 'Flameprince' (FPP), its two independently discovered peach-to-nectarine mutants (HFN and PFN), and a selected nectarine hybrid (SLN). HFN and PFN differed from FPP in fruit size and taste, but shared high similarities in fruit blush coverage and pattern, pit shape, and flesh and pit color. Compared to FPP, the two mutants were significantly lighter and smaller, but higher in soluble solids content (SSC), titratable acidity (TA), and the SSC/TA ratio. The 27 volatiles varied among the cultivars and harvest dates, in terms of total abundance and detection rate, and could be roughly clustered into six subgroups. Subgroup 1 included five volatiles and two fruit attributes that were low on Aug 6 but dramatically increased on Aug 13. Subgroup 2 had a similar tendency to subgroup 1 but one of the cultivars was an exception. Subgroup 3 included those that had the opposite tendency to subgroup 1 or 2. Volatiles in the three subgroups appeared closely associated with the ripening process. Fruit volatiles and attributes in the other three subgroups generally were inconsistent among the four cultivars on the two dates. Hexanol was the only volatile undetected in the peach, but detected in the three nectarines. Fresh strawberries have short shelf life because of rapid weight loss, softening, and decay. The objective of this research was to study the response of strawberries to ClO when applied to the fruit using a controlled release pad. Four experiments were conducted over 2011 to 2013. Strawberries were packed in perforated commercial clamshells, with or without ClO treatments, and stored at 1 and 6 °C for up to 14 days and at 10 and 20 °C for up to 7 days. The effect of ClO on strawberries was assessed by measuring decay incidence, weight loss, firmness, volatile composition, and stomate activity during storage. Chlorine dioxide treatments induced closing of stomata, markedly slowed weight loss and softening, and reduced decay incidence of strawberry fruit at 10 °C or lower temperatures, but not at 20 °C. Blueberry (Vaccinium spp.) is a specialty crop with expanding production and consumption worldwide. Southern Highbush Blueberries (SHB) require few chilling hours and include early-ripening cultivars which can be harvested in an early market window making the crop more profitable for growers in sub-tropical regions. To expand the marketability of the crop, fruit breeders are seeking to meet the current demand for fruit quality. To this end, this study gathered sensory measurements (firmness, sweetness, sourness, blueberry flavor, green flavor and off-flavors), chemical (pH, titratable acidity and soluble solids concentration), and physical attributes (compression force) on six SHB cultivars, in order to understand how fruit quality traits are affected by environmental conditions such as storage duration, year and harvest date. Differences across environmental conditions for all attributes were observed. Indigocrisp and Keecrisp were the firmest cultivars and maintained firmness over time in storage. Additionally, Indigocrisp was the sweetest and had the most blueberry and green flavors among the cultivars. On the other hand, Arcadia was the softest, as measured by sensory and compression force, but varied with year and harvest. Arcadia and Emerald were the least sweet and most sour of the cultivars, and Endura and Patrecia were intermediate for all sensory attributes. In general, sensory descriptors were well explained by instrumental measurements. Variations across seasons and storage were inconsistent among cultivars, but Indigocrisp was the most stable cultivar for the quality traits evaluated. Agaricus bisporus is the most commonly consumed edible mushroom in the US, but research on its sensory properties is limited. This study characterized aroma and flavor of three raw and roasted A. bisporus mushrooms (white, crimini, and portobello) using quantitative descriptive analysis. Sixteen sensory attributes were chosen and included definitions and reference standards prepared with chemical solutions representing aromas perceived in the samples. All three raw A. bisporus possessed key sensory characters of mushroom, earthy, hay, soybean, potato, and woody aroma and flavors. Raw portobello and crimini had significantly higher (p < 0.05) flavor intensities in mushroom, earthy, dark meat, woody, and cabbage flavors, umami and bitterness taste, than white. When roasted, sensory profiles significantly (p < 0.05) shifted to dark meat, roasted, and fried notes, and portobello showed the highest intensity in dark meat flavor. Conversely, hay, woody, and earthy notes decreased in all roasted mushrooms. The results contribute to the growing body of research on mushroom sensory properties and potential use in savory products. (P. trifoliata), a relative of the genus Citrus , in their pedigree to produce hybrids that are cold- and disease-resistant. Initial hybrids of Citrus × P. trifoliata typically produce fruit that have unacceptable flavor, but more advanced hybrids with greater proportions of Citrus in their pedigrees have various degrees of off-flavor and taste. This study reports the volatile constituents, as well as nonvolatile limonoids and flavonoids, in the juice of six Citrus hybrids containing P. trifoliata in their pedigrees, compared to two hybrids derived solely from Citrus. Secondary metabolite profiles reflected the complex genetic backgrounds of those hybrids. Hybrids 6_23−20 and US 119 had juice composition most similar to that of P. trifoliata , with more volatiles, and in particular, more esters and sesquiterpenes with higher amounts in total. Furthermore, hesperidin concentration in US 119 was low (10 μg g−1) in comparison with other hybrids (125−326 μg g−1) while concentrations of narirutin and isosakuranetin-7-O-rutinoside were unusually high (135−211 μg g−1 and 304−182 μg g−1, respectively) in comparison with the other hybrids (5−60 μg g−1 and 10−75 μg g−1, respectively), which is in agreement with the composition of pure P. trifoliata. Juice of P. trifoliata derived hybrids 5−18-24, 5−18-31, 1−76-100, and 1−77-105 presented secondary metabolite compositions closer to those of pure Citrus hybrids. Based on volatiles, limonoids, and flavonoid profiles, we conclude that it is possible to generate advanced Citrus hybrids including P. trifoliata in their pedigrees that approach the quality of common Citrus types. Background: Valencene is a major sesquiterpene in citrus oil and biosynthesized by valencene synthase (Cstps1; EC: 4.2.3.73) from the 15-carbon substrate farnesyl diphosphate. It is abundant in juice of some mandarins (e.g. Citrus reticulata Blanco cv. Fortune), however, it is undetectable in others (e.g. C. reticulata Blanco cv. Murcott), We have discovered that the Murcott mandarin Cstps1 gene expression is severely reduced. A previous genetic mapping study using an F1 population of Fortune × Murcott found that the segregation of valencene production in fruit exhibited a Mendelian inheritance ratio of 1:1. There was only one dominant locus associated with valencene content detected on the mandarin genetic map. The goal of this study was to understand the molecular mechanism underlying the valencene deficiency observed in some citrus hybrids. Results: There was a clear relationship between presence or absence of the valencene synthase gene (Cstps1) expression, and presence or absence of valencene among randomly selected mandarin hybrids. Cloning the coding regions of Cstps1 from Fortune and Murcott mandarin, and aligning with previous reported Valencia orange Cstps1 sequence, showed that they both exhibited extremely high similarity with the known Cstps1. By further cloning and analyzing the promoter region of Cstps1 from Valencia, Fortune and Murcott, a 12-nucleotide deletion at approximately − 270 bp from the Cstps1 coding region was only found in Murcott. Three binary vectors, designated as p1380-FortP-GUSin, p1380-MurcP-GUSin and p1380-MurcP(+ 12)-GUSin, were developed for promoter activity analysis. Transient over-expression of Fortune Cstps1 promoter in sweet orange showed notable GUS activity, but the Murcott Cstps1 promoter did not. In addition, by re-inserting the 12-nucleotide fragment, the activity of the Murcott Cstps1 promoter was mostly recovered. Conclusion: The deficiency of valencene production in some mandarins is probably due to a 12-nucleotide deletion in the promoter region of the Cstps1, which could be a crucial switch of Cstps1 transcription. Two of the major postharvest diseases impacting grapefruit shelf life and marketability in the state of Florida (USA) are stem-end rot (SER) caused by Lasiodiplodia theobromae and green mold (GM) caused by Penicillium digitatum . Here, we investigated the in vitro and in vivo efficacy of vapors of thymol, a natural compound found in the essential oil of various plants and the primary constituent of thyme ( Thymus vulgaris ) oil, as a potential solution for the management of GM and SER. Thymol vapors at concentrations lower than 10 mg L -1 significantly inhibited the mycelial growth of both pathogens, causing severe ultrastructural damage to P. digitatum conidia. In in vivo trials, the incidence and lesion area of GM and SER on inoculated grapefruit were significantly reduced after a 5 d exposure to 50 mg L -1 thymol vapors. In addition, the in vitro and in vivo sporulation of P. digitatum was suppressed by thymol. When applied in its vapor phase, thymol had no negative effect on the fruit, neither introducing perceivable off-flavor nor causing additional weight loss. Our findings support the pursuit of further studies on the use of thymol, recognized as safe for human health and the environment, as a promising strategy for grapefruit postharvest disease management.

Beverage mixtures based on pineapple ( Ananas comosus ) and turmeric ( Curcuma longa ) juice as a ready-to-drink product were developed, and their physicochemical, nutritional, and sensory properties were evaluated. Four different concentrations of turmeric juice (5%, 10%, 15%, and 20% ( v / v )) were added to pineapple juice to make turmeric-fortified pineapple (TFP) juice samples. Pineapple juice without turmeric was the control. The L *, a *, b *, titratable acidity (TA), total antioxidant capacity, and %DPPH scavenging values, as well as the concentrations of the phenolic compounds curcumin and demethoxycurcumin, were significantly increased with increasing turmeric concentration. Thirty volatile compounds were detected in the mixed juice samples with turmeric. Most of the turmeric-specific compounds, including monoterpenes, sesquiterpenes and turmerones, were detected in the TFP juice samples. While the antioxidant activity of the juice samples increased with increasing turmeric concentration, the pineapple juice fortified with 10% turmeric (10%T) had the best overall quality as determined by panelists. Greater concentrations of turmeric were associated with decreased palatability due to reduced mouthfeel and sweetness and increased aftertaste and sourness. These results suggest that the 10%T juice could be developed into a commercial functional beverage with increased overall flavor and nutritional quality.
