Schistosomiasis is a public health problem in many developing countries. The mollusc Biomphalaria glabrata is the most important vector of Schistosoma mansoni in South America. The population control of this vector to prevent the spread of schistosomiasis is currently done with the application of highly toxic molluscicide to the environment. The screening of substances in sublethal concentrations that have deleterious effects on physiological parameters is very relevant for the control of schistosomiasis, since the effectiveness of disease prevention increases if it acts on population control of the vector and on reproduction and elimination in S. mansoni cercariae. The objective of this study was to evaluate the reproductive parameters (fecundity and fertility), intra-mollusk effect (sporocysts I (72 h) and II (14 days after)) on the development of cercariae of S. mansoni and the immune cell profile of B. glabrata exposed to sublethal concentrations (LC25 - 0.5 µg/mL and LC50 - 0.92 µg/mL) of the usnic acid potassium salt (potassium usnate). Lymnaeidae/planorbidae snails are the intermediate hosts of Fasciola hepatica and Fasciola gigantica. Distribution of these vector snails explains the infection of fasciolosis as per geography and climatic condition of place. It also plays a vital role in the pattern of fasciolosis transmission. India is a large country, having varied climatic conditions in different states, so that in the present chapter distribution and habitat of snails in different states of India is given, which represents the statewise consolidated information to explore the vector snail presence.

L'extraction et l'identification de l'ADN à partir d'un échantillon environnemental se sont récemment révélées remarquables pour la détection et le suivi non seulement des espèces communes, mais aussi de celles qui sont menacées, envahissantes ou insaisissables. L'utilisation de la méthode d'analyse metabarcoding de l'ADN environnemental (ADNe) pour étudier les communautés de gastéropodes dans le cadre du programme de suivi scientifique RhônEco pourrai fournir des données sur l'état des milieux fluviaux, aider au perfectionnement des modèles de restauration et entretenir la préservation d'espèce emblématique comme Anisus vorticulus. Cette étude à pour objectif de comparer la méthode d'analyse de l'ADNe à la méthode de prélèvement du benthos accompagnée d'une détermination morphotaxonomique pour l'évaluation des communautés de gastéropodes du Rhône. L'étude se fait pour douze sites appartenant aux différents systèmes alluviaux du Rhône. Trois niveaux de comparaison des deux méthodes sont effectués : la comparaison des inventaires d'espèces de gastéropodes d'eau douce détectées, la comparaison des richesses spécifiques des espèces de gastéropodes d'eau douce par l'estimation des diversités Alpha et la comparaison des assemblages de communauté de gastéropodes en fonction du site par l'estimation des diversités Beta. Dans le cadre de l'analyse de l'ADNe par méthode metabarcoding, les deux types d'échantillons (eau filtrée et sédiment) et les deux marqueurs moléculaires (COI et 16S) sont également comparés entre eux. Cette étude se veut interdisciplinaire permettant de fusionner deux domaines différents, le domaine d'analyse écologique et environnementale et celui de la génétique. Les inventaires des espèces de gastéropodes d'eau douce détectées par les deux méthodes ont démontré que la méthode d'ADNe a permis la détection de 68% des espèces détectés par la méthode de prélèvement du benthos. Egalement, la méthode d'analyse d'ADNe a permis la détermination d'espèce là où la méthode de prélèvement du benthos n'a permis que l'identification au genre. Pour la mesure de la richesse spécifique, les deux méthodes ne sont pas corrélées. Cependant, les deux méthodes perçoivent les différences d'assemblages d'espèces de gastéropodes entre les sites de la même façon en fonction du degré de connexion du milieu au chenal principal. Pour l'estimation des abondances de l'espèce Anisus vorticulus, une méthode d'analyse qPCR de l'ADNe permettrai des résultats plus spécifiques et concluants.

Near Infrared Spectroscopy (NIRS) has been applied in epidemiological surveillance studies of insect vectors of parasitic diseases, such as the Dengue’s mosquitoes. However, regarding mollusks, vectors of important worldwide helminth diseases such as schistosomiasis, fascioliasis and angiostrongyliasis, NIRS studies are rare. This work proposes to establish and standardize the procedure of data collection and analysis using NIRS applied to medical malacology, i.e., to mollusk vectors identifications. Biomphalaria shells and live snails were analyzed regarding several operational aspects, such as: moisture, shell side and position of the live animal for acquisition of NIR spectra. Representative spectra of Biomphalaria shells and live snails were collected using an average of 50 scans per sample and resolution of 16 cm-1. For shells, the sample should first be dried for a minimum of 15 days at an average temperature of 26±1°C, and then placed directly in the equipment measurement window with its left side facing the light beam. Live animals should be dried with absorbent paper; placed into a glass jar, and analyzed similarly to the shells. Once standardized, the technique was applied aiming at two objectives: identification of Biomphalaria using only the shells and parasitological diagnosis for Schistosoma mansoni infection. The discrimination of the three Biomphalaria species intermediate hosts of S. mansoni only by shell has technical limit due to the scarcity of organic material. Nevertheless, it was possible to differentiate B. straminea from B. tenagophila and B. glabrata with 96% accuracy. As for the parasitological diagnosis, it was possible to differentiate infected mollusks shedding S. mansoni cercariae from the non-infected ones with 82, 5% accuracy. In conclusion, the Near Infrared Spectroscopy (NIR’s) technique has proven to be an innovative and sound tool to detect infection by S. mansoni in the different species of Biomphalaria intermediate hosts.

Background The Lake Victoria basin is one of the most persistent hotspots of schistosomiasis in Africa, the intestinal form of the disease being studied more often than the urogenital form. Most schistosomiasis studies have been directed to Schistosoma mansoni and their corresponding intermediate snail hosts of the genus Biomphalaria, while neglecting S. haematobium and their intermediate snail hosts of the genus Bulinus. In the present study, we used DNA sequences from part of the cytochrome c oxidase subunit 1 (cox1) gene and the internal transcribed spacer 2 (ITS2) region to investigate Bulinus populations obtained from a longitudinal survey in Lake Victoria and neighbouring systems during 2010–2019. Methods Sequences were obtained to (i) determine specimen identities, diversity and phylogenetic positions, (ii) reconstruct phylogeographical affinities, and (iii) determine the population structure to discuss the results and their implications for the transmission and epidemiology of urogenital schistosomiasis in Lake Victoria. Results Phylogenies, species delimitation methods (SDMs) and statistical parsimony networks revealed the presence of two main groups of Bulinus species occurring in Lake Victoria; B. truncatus/B. tropicus complex with three species (B. truncatus, B. tropicus and Bulinus sp. 1), dominating the lake proper, and a B. africanus group, prevalent in banks and marshes. Although a total of 47 cox1 haplotypes, were detected within and outside Lake Victoria, there was limited haplotype sharing (only Haplotype 6 was shared between populations from Lake Victoria open waters and neighbouring aquatic systems) – an indication that haplotypes are specific to habitats. Conclusions The Bulinus fauna of Lake Victoria consists of at least B. truncatus, B. tropicus, Bulinus sp. 1 (B. trigonus?) and B. ugandae. The occurrence and wide distribution of Bulinus species in Lake Victoria potentially implies the occurrence of urogenital schistosomiasis in communities living along the shores and on islands of the lake who depend solely on the lake for their livelihood. More in-depth studies are needed to obtain a better picture of the extent of the disease in the Lake Victoria basin.

The Tonle Sap Lake in Cambodia is a crucial freshwater biodiversity hotspot and supports one of the world’s largest inland fisheries. Within the Tonle Sap basin, freshwater molluscs provide vital ecosystem services and are among the fauna targetted for commercial harvesting. Despite their importance, freshwater molluscs of the Tonle Sap basin remain poorly studied. The historical literature was reviewed and at least 153 species of freshwater molluscs have been previously recorded from throughout Cambodia, including 33 from the Tonle Sap basin. Surveys of the Tonle Sap Lake and surrounding watershed were also conducted and found 31 species, 15 bivalves (five families) and 16 gastropods (eight families), in the Tonle Sap basin, including three new records for Cambodia (Scaphula minuta, Novaculina siamensis, Wattebledia siamensis), the presence of globally invasive Pomacea maculata and potential pest species like Limnoperna fortunei. This study represents the most comprehensive documentation of freshwater molluscs of the Tonle Sap basin, and voucher specimens deposited at the Inland Fisheries Research and Development Institute, Cambodia, represent the first known reference collection of freshwater molluscs in the country. In order to combat the combined anthropogenic pressures, including invasive species, climate change and dams along the Mekong River, a multi-pronged approach is urgently required to study the biodiversity, ecology, ecosystem functioning of freshwater molluscs and other aquatic fauna in the Tonle Sap basin.

Trematode-induced castration of snails is widespread and can lead to other life history changes of snails such as changes in trajectories of size and growth or survival. The changes produced likely depend on whether the parasite or host controls allocation of host resources remaining after partial or complete cessation of host current reproduction by castrating trematodes. Documenting host life history changes, like changes in host size in response to castration, is a first step in assessing whether these changes are beneficial to the parasite (increasing transmission success) or to the host (outliving the infection) or to neither. Herein, we test for differences in size and survival among individuals of two snail species in relation to infection by Echinostoma spp. trematodes. Active shedding of Echinostoma spp. was associated with castration of all Stagnicola elodes snails from a site in Eastern Ontario. Snails actively shedding cercariae were not different in size from non-shedding, egg-laying snails but had a higher mortality than egg-laying snails. Active shedding of Echinostoma spp. cercariae was also associated with castration of nearly all Helisoma trivolvis monitored, from a site in Southwestern Ontario. Actively shedding, non-laying H. trivolvis hosts were smaller on average than non-shedding egg-laying hosts, but both non-laying and egg-laying snails survived equally well. We discuss these results in light of what is known about effects of castration on snail hosts in terms of growth and survival for these and other trematode species and speculate on whether changes in size or survival benefits parasite or host.

Neonicotinoids emerged as an environmentally safe alternative to previous generations of insecticides becoming one of the most widely applied in modern agriculture. Nevertheless, they have been reported to affect several non-target organisms. Most toxicity studies focus on the effects on pollinators or terrestrial invertebrates and evaluate either the active ingredient or the commercial formulation. In the present study, we aimed to assess the long-term effects of the active ingredient acetamiprid and a broadly used commercial formulation (Assail® 70) on the non-target freshwater gastropod Biomphalaria straminea using a battery of biomarkers. A 14 day-exposure of adult organisms to both active ingredient and commercial formulation increased carboxylesterase activity and glutathione content, inhibited superoxide dismutase activity and decreased reactive oxygen species levels. The commercial formulation additionally increased glutathione S-transferase activity and inhibited catalase activity. The results indicate a greater toxicity of the commercial formulation than that of the active ingredient alone. Cholinesterase activity, development and offspring survival of B. straminea were not impaired. We conclude that the toxicity of acetamiprid on this gastropod species is mainly related to effects on detoxification and oxidative metabolism responses. This study provides novel information about the adverse effects of the active ingredient and a commercial formulation of a widely used neonicotinoid on a non-target aquatic species. A new fossil species of Biomphalaria Preston is described for the Upper Cretaceous of Uruguay (Queguay Formation). The new species is characterized by a conspicuous spiral ornamentation, a unique character within the genus. This new Biomphalaria species is the third known for the Queguay Formation, in addition to B. walteri (Parodiz) and B. reversa Cabrera, Martínez and Norbis. They represent the oldest record of this genus worldwide.

Individual growth of 15 great ramshorn snail Planorbarius corneus individuals was studied in aquaculture at 20°С starting from 2 weeks after hatching until the death of the animals. It was demonstrated that the weight of mollusks increases throughout the study period by an S-type curve. The linear growth (shell diameter) follows a parabolic (convex) curve. Both weight and linear growth on a specified age range is well approximated by the Bertalanffy equation, while the interdependence between the shell weight and diameter by an allometric equation. The value of Bertalanffy equation coefficients is different in different individuals. The mean values of the growth constants in these equations for the weight and linear growth do not differ. Their value is 0.041 ± 0.004 weeks–1. The species lifespan, which is determined by a product of the growth constant into maximal lifespan, is ≈4.5.

The freshwater snail Physella acuta was selected to expand the perspective of comparative snail immunology. Analysis of Physella acuta, belonging to the Physidae, taxonomic sister family to Planorbidae, affords family-level comparison of immune features characterized from Biomphalaria glabrata, the model snail often used to interpret general gastropod immunity. To capture constitutive and induced immune sequences, transcriptomes of an individual Physella acuta snail, 12 h post injection with bacteria (Gram -/+) and one sham-exposed snail were recorded with 454 pyrosequencing. Assembly yielded a combined reference transcriptome containing 24,288 transcripts. Additionally, genomic Illumina reads were obtained (∼15-fold coverage). Recovery of transcripts for two macin-like antimicrobial peptides (AMPs), 12 aplysianins, four LBP/BPIs and three physalysins indicated that Physella acuta shares a similar organization of antimicrobial defenses with Biomphalaria glabrata, contrasting a modest AMP arsenal with a diverse set of antimicrobial proteins. The lack of predicted transmembrane domains in all seven Physella acuta PGRP transcripts supports the notion that gastropods do not employ cell-bound PGRP receptors, different from ecdysozoan invertebrates yet similar to mammals (vertebrate deuterostomes). The well-documented sequence diversification by Biomphalaria glabrata FREPs (immune lectins comprising immunoglobulin superfamily domains and fibrinogen domains), resulting from somatic mutations of a large FREP gene family is hypothesized to be unique to Planorbidae; Physella acuta revealed just two bonafide FREP genes and these were not diversified. Furthermore, the flatworm parasite Echinostoma paraensei, confirmed here to infect both snail species, did not evoke from Physella acuta the abundant expression of FREP proteins at 2, 4 and 8 days post exposure that was previously observed from Biomphalaria glabrata. The Physella acuta reference transcriptome also revealed 24 unique transcripts encoding proteins consisting of a single fibrinogen-related domain (FReDs), with a short N-terminal sequence encoding either a signal peptide, transmembrane domain or no predicted features. The Physella acuta FReDs are candidate immune genes based on implication of similar sequences in immunity of bivalve molluscs. Overall, comparative analysis of snails of sister families elucidated the potential for taxon-specific immune features and investigation of strategically selected species will provide a more comprehensive view of gastropod immunity.

The radular apparatus of Biomphalaria occidentalis Paraense, 1981, Biomphalaria oligoza Paraense, 1971, Biomphalaria peregrina (d'Orbigny, 1835), Biomphalaria schrammi (Crosse, 1864) and Biomphalaria tenagophila (d'Orbigny, 1835) from the Metropolitan Region of Sao Paulo, Brazil were analyzed from Scanning Electron Microscope (SEM) micrographs. The data support the usage of characters of central, lateral and intermediate teeth in the species identification of Biomphalaria. The use of SEM provided additional quantitative and structural details to radula descriptions, which are extremely important for elucidating the taxonomy of Biomphalaria species.
