Abstract

Diffuse large B-cell lymphoma can be classified by gene expression profiling into germinal center and activated B-cell subtypes with different prognoses after rituximab-CHOP. The importance of previously recognized prognostic markers, such as Bcl-2 protein expression and BCL2 gene abnormalities, has been questioned in the new therapeutic era. We analyzed Bcl-2 protein expression, and BCL2 and MYC gene abnormalities by interphase fluorescence in situ hybridization in 327 patients with de novo disease treated with rituximab-CHOP. Isolated BCL2 and MYC rearrangements were not predictive of outcome in our patients as a whole, but only in those with the germinal center subtype of lymphoma. The prognostic relevance of isolated MYC rearrangements was weaker than that of BCL2 isolated translocations, but was probably limited by the rarity of the rearrangements. Seven of eight patients with double hit lymphoma had the germinal center subtype with poor outcome. The germinal center subtype patients with isolated BCL2 translocations had significantly worse outcome than the patients without BCL2 rearrangements (P=0.0002), and their outcome was similar to that of patients with the activated B-cell subtype (P=0.30), but not as bad as the outcome of patients with double hit lymphoma (P

Abstract

The acquisition of resistance towards FAS-mediated apoptosis may be required for tumor formation. Tumors from various histological origins exhibit FAS mutations, the most frequent being hematological malignancies. However, data regarding FAS mutations or FAS signaling alterations are still lacking in precursor T-cell lymphoblastic lymphomas (T-LBLs). The available data on acute lymphoblastic leukemia, of precursor origin as well, indicate a low frequency of FAS mutations but often report a serious reduction in FAS-mediated apoptosis as well as chemoresistance, thus suggesting the occurrence of mechanisms able to deregulate the FAS signaling pathway, different from FAS mutation. Our aim at this study was to determine whether FAS-mediated apoptotic signaling is compromised in human T-LBL samples and the mechanisms involved. This study on 26 T-LBL samples confirms that the FAS system is impaired to a wide extent in these tumors, with 57.7% of the cases presenting any alteration of the pathway. A variety of mechanisms seems to be involved in such alteration, in order of frequency the downregulation of FAS, the deregulation of other members of the pathway and the occurrence of mutations at FAS. Considering these results together, it seems plausible to think of a cumulative effect of several alterations in each T-LBL, which in turn may result in FAS/FASLG system deregulation. Since defective FAS signaling may render the T-LBL tumor cells resistant to apoptotic cell death, the correct prognosis, diagnosis and thus the success of anticancer therapy may require such an in-depth knowledge of the complete scenario of FAS-signaling alterations.

Abstract

En los últimos años, investigadores de distintos países han concentrado esfuerzos en desarrollar técnicas no contaminantes de desinfección del suelo. El objetivo de este trabajo fue evaluar el efecto de distintas secuencias de solarización y/o biofumigación (BIO). Los tratamientos evaluados consistieron en la combinación de dos desinfecciones realizadas en la primavera 2003 y 2005. Estos fueron 1. Testigo/Testigo, 2. Solarización/Solarización, 3. BIO con Estiércol/BIO con Brócoli, 4. BIO con Colza/BIO con Brócoli. Se observó control de nematodos en los 10 primeros cm del suelo, en el muestreo realizado inmediatamente después de la biofumigación, realizada en noviembre de 2005. Las diferencias entre tratamientos y el testigo se mantuvieron para esta especie (P ≤ 5 %) hasta febrero del 2007. El porcentaje de colonias de Sclerotium rolfsii recuperadas de esclerocios fue siempre mayor en el testigo sin tratar, pero a 35 cm el efecto de los tratamientos fue menor. Fusarium solani fue encontrado solamente en el testigo a 10 cm de profundidad, pero en todos los tratamientos a 35 cm. En noviembre de 2006 se trasplantó un cultivo de tomate. El número de plantas muertas al final del ciclo fue significativamente mayor en el testigo sin tratar. Se obtuvieron diferencias altamente significativas entre tratamientos (P ≤ 1 %) para rendimiento y descarte total y descarte por pequeño y podredumbre apical. El testigo sin tratar mostró los menores rendimientos y mayores porcentajes de descartes. Después de la cosecha se observaron diferencias significativas para el número de agallas y porcentaje de raíces con síntomas de podredumbres en tomate. Los patógenos aislados de las raíces con síntomas de podredumbres fueron Pyrenochaeta lycopersici y Fusarium solani. In the last years, researchers from different countries have made efforts to develop environmentally responsible soil disinfection techniques. The objective of this work was to evaluate the effect of different sequences of soil solarization and/or biofumigation (BIO). The treatments evaluated were the combination of two disinfections performed on spring 2003 and 2005. The sequences evaluated were: 1. Control/Control, 2. Solarization/Solarization, 3. BIO with Chicken manure/BIO with Brocoli, 4. BIO with Brassica napus/BIO with Brocoli. Nematode control was observed at the first 10 cm of soil, inmediately after BIO treatments, performed at november 2005, differences (P ≤ 5 %) between treatments and control were observed for Nacobbus aberrans until february 2007. Percentage of Sclerotium rolfsii colonies obtained from sclerotia was always higher at control plots, but at 35 cm the treatment effect was lower. Fusarium solani was obtained only at control plots at 10 cm but in all treatments at 35 cm. In november 2006 a tomato crop was planted. The number of dead plants at the end of the cycle was significantly higher at control plots. Significant differences (P ≤ 1 %) between treatments were obtained for total yield, yield losses, and losses percentage due to small fruits and blossom end rot. Control plots showed lower yields and higher commercial yield losses. 116. Pecoma pélvico del ligamento ancho con metástasis ganglionares: presentación de un caso y revisión de la literatura

Abstract

Objetivos: Describimos y documentamos el primer caso de PEComa publicado en España según la base de datos Pub Med. Y revisamos la bibliografía existente sobre este tipo de tumores. Métodos/Resultados: Se Trata de una paciente de 39 años con un tumor pélvico de 9 cms. descubierto en una revisión ginecológica rutinaria. El TAC muestra múltiples adenopatías periaórticas que se extienden hasta vena renal izquierda El diagnóstico de PEComa se obtiene por punción con aguja gruesa. Se realiza exéresis del tumor junto a anexo izquierdo y linfadenectomía aortoiliaca. El resultado patológico confirma el diagnóstico tras la inmunohistoquímica con actina de músculo liso y HMB-45. No recibió tratamiento adyuvante. Tras un año de seguimiento la paciente no muestra en el TAC signos de recidiva. Se realiza una amplia búsqueda bibliográfica en Pub Med encontrando 73 referencias a este tipo de tumor cuyas conclusiones se exponen en este articulo. Conclusiones: El tumor de celulas epitelioides perivasculares, PEComa, es una neoplasia mesenquimal de muy baja incidencia y con malignidad incierta. Tumores muy raros: el angiomiolipoma epitelioide, la linfangioleiomiomatosis, el linfangiomioma, el tumor miomelanocitico del ligamento falciforme, el tumor pulmonar de celulas claras "de azucar" y su variante extrapulmonar; son antiguas descripciones de lo que ahora es una única entidad tumoral denominada PEComa. Aun no de ha descrito la célula que en tejidos normales da origen a este tumor. Afectando principalmente a mujeres en edad media, pueden encontrarse en cualquier lugar anatómico. Su tratamiento es quirúrgico aunque se desconoce el papel de la linfadenectomía y del tratamiento adyuvantes. Objective: We describe and document the first case of PEcoma published in Spain following the PubMed database. We review the bibliography about these tumors. Methods/Results: 39-year-old female patient with a 9 cm pelvic tumor discovered in a routine gynecologic review. CT scan showed multiple periaortic adenopathies extending up to the left renal vein. The diagnosis of PEcoma was obtained by needle biopsy. Tumor excision including left annex and aortoiliac lymphadenectomy were performed. Pathology confirmed the diagnosis after immunohistochemical study with smooth muscle actin and HMB-45. No adjuvant treatment was given. After one year of follow-up the patient does not show signs of recurrence of the CT scan. A PubMed search was performed finding 73 references about this kind of tumor the conclusions of which are exposed in this article. Conclusions: Perivascular epithelioid cell tumor, PEcoma, is a very low incidence mesenchymal neoplasia with uncertain malignancy. Very rare tumors such as epithelioid angiomyolipoma, lymphangioleiomyomatosis, lymphangiomyoma, myelomelanocytic tumor of the falciform ligament, "sugar" clear cell lung tumor and its extrapulmonary variant are all descriptions of what is now a unique tumoral entity named PEcoma. It has not been described yet the normal tissue cell giving origin to this tumor. It affects mainly middle age women, and may appear in any anatomical site. Treatment is surgical, although the role of lymphadenectomy and adjuvant treatment is unknown.

Abstract

Background: Optimal therapeutic management of intravascular lymphoma (IVL) lacks precise guidelines. Patients and methods: The clinico-pathological features of 38 HIV-negative patients with IVL were reviewed to define efficacy of chemotherapy in these malignancies. Clinical characteristics of 22 patients treated with chemotherapy and of 16 untreated patients were compared in order to understand better the impact and causes of potential patient selection. Results: Median age was 70 years (range 34-90), with a male/female ratio of 0.9; 23 (61%) patients had Eastern Cooperative Oncology Group performance status (ECOG-PS) >1; 21 (55%) had systemic symptoms. Cutaneous lesions and anemia were significantly more common among patients treated with chemotherapy; central nervous system (CNS) and renal involvement were significantly more common among untreated patients. Chemotherapy was associated with a response rate of 59% and a 3-year overall survival of 33 ± 11%. Five of six patients with CNS involvement received chemotherapy: four of them died early; only one patient, treated with adriamycin, cyclophosphamide, vincristine, methotrexate, bleomycin and prednisolone (MACOP-B) followed by high-dose chemotherapy and autologous stem cell transplantation (ASCT), was alive at 19 months. High-dose chemotherapy supported by ASCT was indicated at diagnosis in another patient (43 years of age, stage I), who was alive at 71 months, and at relapse after cyclophosphamide, doxorubicin, vincristine and prednisone (CHOP) in two patients who died early after transplantation. PS ≤1, disease limited to the skin, stage I, and use of chemotherapy were independently associated with better outcome. Conclusions: Anthracycline-based chemotherapy is the standard treatment for IVL. However, survival is disappointing, with a relevant impact of diagnostic delay and lethal complications. More intensive combinations, containing drugs with higher CNS bioavailability, are needed in cases with brain involvement, and the role of high-dose chemotherapy supported by ASCT should be further investigated in younger patients with unfavorable features. © 2004 European Society for Medical Oncology.

Abstract

In diffuse large B-cell lymphoma (DLBCL), the clinical and biological significance of concordant and discordant bone marrow (BM) involvement have not been well investigated. We evaluated 712 de novo DLBCL patients with front-line rituximab-containing treatment, including 263 patients with positive and 449 with negative BM status. Compared with negative BM disease, concordant BM adversely impacted overall and progression-free survival, independent of the International Prognostic Index (IPI) and cell-of-origin classification. Once BM is concordantly involved, poor prognosis was not associated with the extent of BM involvement. Conversely, patients with discordant BM showed favorable overall survival similar to stage I–II DLBCL. A BM-adjusted IPI, using three parameters: concordant BM involvement, age >60 years, and performance status >1, improves the risk stratification for DLBCL with positive BM. Intensive immunochemotherapy seemingly rendered survival benefit for patients with concordant BM, as did rituximab maintenance for the discordant BM group. Frequently revealing adverse clinical and molecular characteristics, patients with concordant BM demonstrated gene expression signatures relevant to tumor cell proliferation, migration and immune escape. In conclusion, clinical and biological heterogeneity is seen in DLBCL with positive BM but concordant BM involvement represents a distinct subset with unfavorable gene signatures, high-risk clinicopathologic features and poor prognosis.

The International Cancer Genome Consortium (ICGC) was launched to coordinate large-scale cancer genome studies in tumours from 50 different cancer types and/or subtypes that are of clinical and societal importance across the globe. Systematic studies of more than 25,000 cancer genomes at the genomic, epigenomic and transcriptomic levels will reveal the repertoire of oncogenic mutations, uncover traces of the mutagenic influences, define clinically relevant subtypes for prognosis and therapeutic management, and enable the development of new cancer therapies. © 2010 Macmillan Publishers Limited. Abstract

In the last years, researchers from different countries have made efforts to develop environmentally responsible soil disinfection techniques. The objective of this work was to evaluate the effect of different sequences of soil solarization and/or biofumigation (BIO). The treatments evaluated were the combination of two disinfections performed on spring 2003 and 2005. The sequences evaluated were: 1. Control/Control, 2. Solarization/Solarization, 3. BIO with Chicken manure/BIO with Brocoli, 4. BIO with Brassica napus/BIO with Brocoli. Nematode control was observed at the first 10 cm of soil, inmediately after BIO treatments, performed at november 2005, differences (P ≤ 5 %) between treatments and control were observed for Nacobbus aberrans until February 2007. Percentage of Sclerotium rolfsii colonies obtained from sclerotia was always higher at control plots, but at 35 cm the treatment effect was lower. Fusarium solani was obtained only at control plots at 10 cm but in all treatments at 35 cm. In November 2006 a tomato crop was planted. The number of dead plants at the end of the cycle was significantly higher at control plots. Significant differences (P ≤ 1 %) between treatments were obtained for total yield, yield losses, and losses percentage due to small fruits and blossom end rot. Control plots showed lower yields and higher commercial yield losses. Abstract

The potential energy curves of P2 and P2+ have been calculated using an approximate, albeit strictly N-representable, energy functional of the one-particle reduced density matrix: PNOF5. Quite satisfactory accord is found for the equilibrium bond lengths and dissociation energies for both species. The predicted vertical ionisation energy for P2 by means of the extended Koopmans' theorem is 10.57 eV, in good agreement with the experimental data. Comparison of the vibrational energies and anharmonicities with their corresponding experimental values supports the quality of the resultant potential energy curves. [ABSTRACT FROM AUTHOR]
