Abstract Corn snakes are emerging models for animal colouration studies. Here, we focus on the Terrazzo morph, whose skin pattern is characterized by stripes rather than blotches. Using genome mapping, we discover a disruptive mutation in the coding region of the Premelanosome protein (PMEL) gene. Our transcriptomic analyses reveal that PMEL expression is significantly downregulated in Terrazzo embryonic tissues. We produce corn snake PMEL knockouts, which present a comparable colouration phenotype to Terrazzo and the subcellular structure of their melanosomes and xanthosomes is also similarly impacted. Our single-cell expression analyses of wild-type embryonic dorsal skin demonstrate that all chromatophore progenitors express PMEL at varying levels. Finally, we show that in wild-type embryos PMEL-expressing cells are initially uniformly spread before forming aggregates and eventually blotches, as seen in the adults. In Terrazzo embryos, the aggregates fail to form. Our results provide insights into the mechanisms governing colouration patterning in reptiles.

Abstract Introduction The pulmonary Hot Clot artifact (HCa) on 18F-FDG PET/CT is a poorly understood phenomenon, corresponding to the presence of a focal tracer uptake without anatomical lesion on combined CTscan. The hypothesis proposed in the literature is of microembolic origin. Our objectives were to determine the incidence of HCa, to analyze its characteristics and to identify associated factors. Methods All 18F-FDG PET/CT retrieved reports containing the keywords (artifact/vascular adhesion/no morphological abnormality) during the period June 2021–2023 at Brest University Hospital were reviewed for HCa. Each case was associated with 2 control patients (same daily work-list). The anatomical and metabolic characteristics of HCa were analyzed. Factors related to FDG preparation/administration, patient and vascular history were investigated. Case-control differences between variables were tested using Chi-2 test and OR (qualitative) or Student’s t-test (quantitative). Results Of the 22,671 18F-FDG PET/CT performed over 2 years, 211 patients (0.94%) showed HCa. The focus was single in 97.6%, peripheral in 75.3%, and located independently in the right or left lung (51.1% vs. 48.9%). Abstract Background While melanomas commonly harbor losses of 9p21, on which CDKN2A resides, the presence of additional tumor suppressor elements at this locus is incompletely characterized. Here we assess the expression levels and functional role of microRNA-876-3p (miR-876), whose gene also maps to 9p21. Methods Expression of miR-876 was assessed in human tissues and cell lines using quantitative miRNA reverse transcriptase polymerase chain reaction (qRT-PCR). MIR876 copy number was determined in The Cancer Genome Atlas (TCGA) melanoma cohort. The consequences of regulation of miR-876 expression were assessed on melanoma cell colony formation, migration, invasion, apoptosis, cell cycle progression, and drug sensitivity in culture, and on in vivo tumor growth in a xenograft model. Genome-wide transcriptomic changes induced by miR-876 overexpression were determined using RNA sequencing (RNA-Seq). Results miR-876 expression was significantly decreased in primary melanoma samples when compared with nevi, and in human melanoma cell lines when compared with human melanocytes. Analysis of the TCGA cohort revealed deletions in MIR876 in > 50% of melanomas. miR-876 overexpression resulted in decreased melanoma cell colony formation, migration, and invasion, which was accompanied by cell cycle arrest and increased apoptosis. Intra-tumoral injections of miR-876 significantly suppressed melanoma growth in vivo. RNA-Seq analysis of miR-876-treated tumors revealed downregulation of several growth-promoting genes, along with upregulation of tumor suppressor genes, which was confirmed by qRT-PCR analysis. Computational analyses identified MAPK1 (or ERK2) as a possible target of miR-876 action. Overexpression of miR-876 significantly suppressed luciferase expression driven by the MAPK1/ERK2 3’ UTR, and resulted in decreased ERK protein expression in melanoma cells. MAPK1/ERK2 cDNA overexpression rescued the effects of miR-876 on melanoma colony formation. miR-876 overexpression sensitized melanoma cells to treatment with the BRAF inhibitor vemurafenib. Conclusions These studies identify miR-876 as a distinct tumor suppressor on 9p21 that is inactivated in melanoma and suggest miR-876 loss as an additional mechanism to activate ERK and the mitogen activated protein kinase (MAPK) pathway in melanoma. In addition, they suggest the therapeutic potential of combining miR-876 overexpression with BRAF inhibition as a rational therapeutic strategy for melanoma.

Abstract As a contribution to step 3 of the ESG6 blind prediction exercise, we present an application of two different, purely empirical approaches to estimate the strong ground motion at a soft site ("KUMA") from the observed ground motion at a reference rock site ("SEVO") for the two largest shocks of the Kumamoto 2016 sequence. The two methods estimate the non-linear transfer function between a reference rock and a sedimentary site by modifying the linear transfer function derived from weak motion recordings. The modification is based either on a machine learning tool based on a wide collection of Japanese weak and strong motion recordings and the associated site metadata (method 1), or on an estimate of a site-specific parameter related to an average non-linear site response (method 2). The acceleration time series are then derived at the sedimentary site of interest using an estimation of the time delay between wave arrivals at the rock and site stations, and a minimum phase assumption for the site transfer function. These predictions were made blindly, but after the ESG6 conference they could be compared both with the actual ground motion recorded at KUMA during the two shocks, and the average and range of all other predictions preformed for this benchmark. Both of these purely empirical methods provide an honorable prediction of usual engineering ground motion parameters of the two target events. The performance of these two purely empirical approaches is at least comparable to those of the numerical simulation methods for the foreshock—if not better—and slightly worse for the (largest) mainshock. As the methods required only recordings of weak motions at the target and a referent sites and very simple description of the soil profile. The use of moderate motions to constrain the frequency shift prediction for the second method and the consideration of an alternative phase modification are possible ways to improvement. Abstract Aims Right ventricular failure after left ventricular assist device (LVAD) implantation is a major concern that remains challenging to predict. We sought to investigate the relationship between preoperative pulmonary artery pulsatility index (PAPi) and mortality after LVAD implantation. Methods and results A retrospective analysis of the ASSIST‐ICD multicentre registry allowed the assessment of PAPi before LVAD according to the formula [(systolic pulmonary artery pressure − diastolic pulmonary artery pressure)/central venous pressure]. The primary endpoint was survival at 3 months, according to the threshold value of PAPi determined by the receiver operating characteristic (ROC) curve. A multivariate analysis including demographic, echographic, haemodynamic, and biological variables was performed to identify predictive factors for 2 year mortality. One hundred seventeen patients were included from 2007 to 2021. The mean age was 58.45 years (±13.16), with 15.4% of women (sex ratio 5.5). A total of 53.4% were implanted as bridge to transplant and 43.1% as destination therapy. Post‐operative right ventricular failure was observed in 57 patients (48.7%), with no significant difference between survivors and non‐survivors at 1 month (odds ratio 1.59, P = 0.30). The median PAPi for the whole study population was 2.83 [interquartile range 1.63–4.69]. The threshold value of PAPi determined by the ROC curve was 2.84. (1) Background: Multiple point-of-care (POC) glucometers are in use in veterinary medicine, but few are compared to each other. This leaves the potential for clinicians to be unaware of the effect of bias when comparing results from different POC glucometers. (2) Methods: Samples from healthy calves and ewes were simultaneously compared with two POC veterinary glucometers, the Precision Xtra and the AlphaTrak2, under both the “canine” and “feline” settings. The results of each sample were statistically analyzed with linear regression and Bland–Altman analysis. (3) Results: 170 samples from healthy calves and 108 samples from healthy ewes were available for comparison. Calves: The AT2 consistently overestimated blood glucose concentrations when compared to the PX device with the calves. Correlationt with the PX was r = 0.8496 (canine setting) and r = 0.8861 (feline setting). Both the canine and feline settings demonstrated a consistent bias (41.11 and 33.64 mg/dL, respectively). Ewes: The AT2 consistently overestimated blood glucose concentrations when compared to the PX device with the ewes. Correlation with the PX was R = 0.4710 (canine setting) and R = 0.7269 (feline setting). Both the canine and feline settings demonstrated a consistent bias (21.23 and 14.54 mg/dL, respectively). (4) Clinicians should be aware of the potential for consistent bias when evaluating calf and sheep blood glucose concentrations as the AT2 device, at both settings, overestimated blood glucose compared to the previously validated PX. This reliability appears to change when the values are farther from the normal ranges, which should be considered when making clinical decisions based on data from these devices.

BackgroundA composite metric for the quality of glycemia from continuous glucose monitor (CGM) tracings could be useful for assisting with basic clinical interpretation of CGM data.MethodsWe assembled a data set of 14-day CGM tracings from 225 insulin-treated adults with diabetes. Using a balanced incomplete block design, 330 clinicians who were highly experienced with CGM analysis and interpretation ranked the CGM tracings from best to worst quality of glycemia. We used principal component analysis and multiple regressions to develop a model to predict the clinician ranking based on seven standard metrics in an Ambulatory Glucose Profile: very low-glucose and low-glucose hypoglycemia; very high-glucose and high-glucose hyperglycemia; time in range; mean glucose; and coefficient of variation.ResultsThe analysis showed that clinician rankings depend on two components, one related to hypoglycemia that gives more weight to very low-glucose than to low-glucose and the other related to hyperglycemia that likewise gives greater weight to very high-glucose than to high-glucose. These two components should be calculated and displayed separately, but they can also be combined into a single Glycemia Risk Index (GRI) that corresponds closely to the clinician rankings of the overall quality of glycemia (r = 0.95). The GRI can be displayed graphically on a GRI Grid with the hypoglycemia component on the horizontal axis and the hyperglycemia component on the vertical axis. Diagonal lines divide the graph into five zones (quintiles) corresponding to the best (0th to 20th percentile) to worst (81st to 100th percentile) overall quality of glycemia. The GRI Grid enables users to track sequential changes within an individual over time and compare groups of individuals.ConclusionThe GRI is a single-number summary of the quality of glycemia. Its hypoglycemia and hyperglycemia components provide actionable scores and a graphical display (the GRI Grid) that can be used by clinicians and researchers to determine the glycemic effects of prescribed and investigational treatments.

Climate change drives species distribution shifts, affecting the availability of resources people rely upon for food and livelihoods. These impacts are complex, manifest at local scales, and have diverse effects across multiple species. However, for wild capture fisheries, current understanding is dominated by predictions for individual species at coarse spatial scales. We show that species-specific responses to localized environmental changes will alter the collection of co-occurring species within established fishing footprints along the U.S. West Coast. We demonstrate that availability of the most economically valuable, primary target species is highly likely to decline coastwide in response to warming and reduced oxygen concentrations, while availability of the most abundant, secondary target species will potentially increase. A spatial reshuffling of primary and secondary target species suggests regionally heterogeneous opportunities for fishers to adapt by changing where or what they fish. Developing foresight into the collective responses of species at local scales will enable more effective and tangible adaptation pathways for fishing communities.

Mitochondria are classically viewed as ‘on demand’ energy suppliers to neurons in support of their activity. In order to adapt to a wide range of demands, mitochondria need to be highly dynamic and capable of adjusting their metabolic activity, shape, and localization. Although these plastic properties give them a central support role in basal neuronal physiology, recent lines of evidence point toward a role for mitochondria in the regulation of high-order cognitive functions such as memory formation. In this review, we discuss the interplay between mitochondrial function and neural plasticity in sustaining memory formation at the molecular and cellular levels. First, we explore the global significance of mitochondria in memory formation. Then, we will detail the memory-relevant cellular and molecular mechanisms of mitochondrial plasticity. Finally, we focus on those mitochondrial functions, including but not limited to ATP production, that give mitochondria their pivotal role in memory formation. Altogether, this review highlights the central role of mitochondrial structural and functional plasticity in supporting and regulating neuronal plasticity and memory.

Observing System Simulation Experiments (OSSEs) with the Mercator Ocean/Copernicus Marine global 1/12° data assimilation system have been carried out to compare and quantify the expected performance of two high resolution altimetry mission concepts envisioned for the long-term evolution (post-2032) of the Copernicus Sentinel-3 topography mission. The two mission concepts are a constellation of two wide-swath altimeters and a constellation of 12 nadir altimeters. These two configurations greatly improve ocean forecasting and monitoring capabilities. Compared to a constellation of three nadir altimeters (the present configuration), analysis and forecast errors are reduced by a factor of 2. Our results also show that a constellation of two wide-swath altimeters has better performance than a constellation of 12 nadirs. Compared to a constellation of 12 nadirs, the error of the Sea Surface Height (SSH) forecast of a two wide swath constellation is reduced by 14% overall. Improvements are also observed when analyzing surface currents and Lagrangian diagnostics. A constellation of two wide-swath altimeters thus seems to be a very promising concept for the long-term evolution of the Sentinel-3 topography mission.

Summary: Background: In industrialized countries, the incidence of inflammatory bowel disease (IBD) appears stabilized. This study examined the incidence and phenotype of IBD in Northern France over a 30-year period. Methods: Including all IBD patients recorded in the EPIMAD population-based registry from 1988 to 2017 in Northern France, we described the incidence and clinical presentation of IBD according to age, sex and time. Findings: A total of 22,879 incident IBD cases were documented (59% (n = 13,445) of Crohn’s disease (CD), 38% (n = 8803) of ulcerative colitis (UC), 3% (n = 631) of IBD unclassified (IBDU)). Over the study period, incidence of IBD, CD and UC was 12.7, 7.2 and 5.1 per 105 person-years, respectively. Addressing the quality of circulating medicines in low- and middle-Income countries is challenging due to limited access to affordable and rapid laboratory analyses. Moreover, traditional analytical methods generate hazardous waste, posing further environmental and health risks, conflicting with several United Nations Sustainable Development Goals, such as reducing pollution and conserving ecosystems.In response to these environmental challenges, the concept of green analytical chemistry emerged promoting environmentally friendly practices through principles such as reducing chemical use, minimizing energy consumption, and managing waste. Various green metric tools have been developed to assess the environmental impact of analytical methods, but these often neglect the reliability and applicability of the methods. To address this gap, approaches like White Analytical Chemistry (WAC) have been proposed, integrating criteria for environmental safety, reliability, and cost-effectiveness. WAC's principles are categorized into red (fitness for purpose), green (environmental safety), and blue (cost-efficiency) criteria.This study aims to use the WAC approach to provide a holistic comparison of different medicines' quality screening devices, helping developing countries with high SF medicine prevalence make informed choices. Three different situations are investigated: ex-ante evaluation, ex-post evaluation of the qualitative performances and ex-post evaluation of the quantitative performances of the most used medicines’ quality screening devices in low- and middle-income countries field settings.

Background Factors associated with the appropriateness of antibiotic prescribing in primary care have been poorly explored. In particular, the impact of computerised decision-support systems (CDSS) remains unknown.Objectives We aim at investigating the uptake of CDSS and its association with physician characteristics and professional activity.Methods Since May 2022, users of a CDSS for antibiotic prescribing in primary care in France have been invited, when registering, to complete three case vignettes assessing clinical situations frequently encountered in general practice and identified as at risk of antibiotic misuse. Appropriateness of antibiotic prescribing was defined as the rate of answers in line with the current guidelines, computed by individuals and by specific questions. Physician’s characteristics associated with individual appropriate antibiotic prescribing (< 50%, 50–75% and > 75% appropriateness) were identified by multivariate ordinal logistic regression.Results In June 2023, 60,067 physicians had registered on the CDSS. Among the 13,851 physicians who answered all case vignettes, the median individual appropriateness level of antibiotic prescribing was 77.8% [Interquartile range, 66.7%–88.9%], and was < 50% for 1,353 physicians (10%). In the multivariate analysis, physicians’ characteristics associated with appropriateness were prior use of the CDSS (OR = 1.71, 95% CI 1.56–1.87), being a general practitioner vs. other specialist (OR = 1.34, 95% CI 1.20–1.49), working in primary care (OR = 1.14, 95% CI 1.02–1.27), mentoring students (OR = 1.12, 95% CI 1.04–1.21) age (OR = 0.69 per 10 years increase, 95% CI 0.67–0.71).Conclusion Individual appropriateness for antibiotic prescribing was high among CDSS users, with a higher rate in young general practitioners, previously using the system. CDSS could improve antibiotic prescribing in primary care.

Mammalian receptor-mediated endocytosis (RME) often involves at least one of three isoforms of the large GTPase dynamin (Dyn). Dyn pinches-off vesicles at the plasma membrane and mediates uptake of many viruses, although some viruses directly penetrate the plasma membrane. RME is classically interrogated by genetic and pharmacological interference, but this has been hampered by undesired effects. Here we studied virus entry in conditional genetic knock-out (KO) mouse embryonic fibroblasts lacking expression of all three dynamin isoforms (Dyn-KO-MEFs). The small canine parvovirus known to use a single receptor, transferrin receptor, strictly depended on dynamin. Larger viruses or viruses known to use multiple receptors, including alphaviruses, influenza, vesicular stomatitis, bunya, adeno, vaccinia, severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and rhinoviruses infected Dyn-KO-MEFs, albeit at higher dosage than wild-type MEFs. In absence of the transmembrane protease serine subtype 2 (TMPRSS2), which normally activates the SARS-CoV-2 spike protein for plasma membrane fusion, SARS-CoV-2 infected angiotensin-converting enzyme 2 (ACE2)-expressing MEFs predominantly through dynamin- and actin-dependent endocytosis. In presence of TMPRSS2 the ancestral Wuhan-strain bypassed both dynamin-dependent and -independent endocytosis, and was less sensitive to endosome maturation inhibitors than the Omicron B1 and XBB variants, supporting the notion that the Omicron variants do not efficiently use TMPRSS2. Collectively, our study suggests that dynamin function at endocytic pits can be essential for infection with single-receptor viruses, while it is not essential but increases uptake and infection efficiency of multi-receptor viruses that otherwise rely on a functional actin network for infection.

With an expected increase in population by 2050, the demand for high-quality protein for human consumption will increase. To improve the sustainability of US cattle production, producers and researchers have historically focused on improving the feed efficiency of steers through improvements in genetics, nutrition, and microbiome management, producing a more marketable beef product. However, without a successful pregnancy, there would be no marketable animal to feed. The objective of this study was to examine the impact of pregnancy on the rumen environment, microbial communities, and the correlation to feed efficiency status in Angus heifers. Utilizing 17 cannulated Angus heifers, feed efficiency status was determined using the GrowSafe 8000 feed intake monitoring system to monitor feed intake for a 70d trial period to calculate residual feed intake. Following the trial, heifers were bred and 40mL rumen fluid and content samples were collected every two weeks during gestation. Metagenomic DNA was extracted from the rumen samples, sequencing libraries were prepared targeting the bacterial V4 region of the 16S rRNA gene, and resultant libraries were sequenced on an Illumina MiSeq. All microbial analyses were conducted in the R environment v2.6.2 where alpha diversity, beta diversity, and differential abundances were determined. Metabolites were identified using ultra high-performance liquid chromatography high resolution mass spectrometry, and Metabolomic Analysis and Visualization Engine was used to select metabolite peaks. The eco-epidemiology of tick-borne diseases hinges on the abundance and distribution of hosts that sustain tick populations and the pathogens they carry. Research into the role of bird species in the feeding of Ixodes ricinus ticks, the primary tick species of veterinary and public health importance in Europe, remains scarce. This study endeavors to bridge these knowledge gaps by (i) assessing the density of feeding ticks (DFT) within a bird community to pinpoint species making substantial contributions, and (ii) exploring interannual variations in DFT over an extended timeline. Furthermore, we investigate whether variations in individual tick burden (TB) were more closely associated with the characteristics of bird species or interannual variations affecting the density of questing tick, using interannual TB variation as a surrogate. To fulfill these aims, we conducted a 13-year longitudinal study monitoring I. ricinus ticks feeding on a bird community in a periurban forest in France, covering breeding periods from 2007 to 2019. Within this community, we identified seven principal bird species significantly contributing to I. ricinus tick feeding: the Common Blackbird (Turdus merula), the Song Thrush (Turdus philomelos), the European Robin (Erithacus rubecula), the Dunnock (Prunella modularis), the Eurasian Blackcap (Sylvia atricapilla), the Great Tit (Parus major), and the Common Nightingale (Luscinia megarhynchos). Our results show that the bird community's contribution to tick feeding remained relatively consistent from year-to-year, though certain years displayed higher or lower DFT values related to the average over the study period. Moreover, five out the seven major species accounted for 80 % to 95 % of DFT annually. Consequently, we emphasized the need to broaden the scope of future research on bird contributions to tick population dynamics beyond merely thrushes (Turdidae species), to encompass a more diverse range of species, particularly those common birds that engage in ground foraging activities. Furthermore, variations in individual tick burden were predominantly influenced by the characteristics of bird species rather than by interannual variability in infestation rates. This finding suggests a significant role for species-specific traits in determining tick exposure and susceptibility. In conclusion, our study offers new insights into the medium-term dynamics of tick-bird ecological systems, underscoring the need for future study of tick populations and their interactions with vertebrate hosts to improve our understanding of tick-borne disease circulation.

Relevance-based selectivity and high energy cost are two distinct features of long-term memory (LTM) formation that warrant its default inhibition. Spaced repetition of learning is a highly conserved cognitive mechanism that can lift this inhibition. Here, we questioned how the spacing effect integrates experience selection and energy efficiency at the cellular and molecular levels. We showed in Drosophila that spaced training triggers LTM formation by extending over several hours an increased mitochondrial metabolic activity in neurons of the associative memory center, the mushroom bodies (MBs). We found that this effect is mediated by PKCδ, a member of the so-called ‘novel PKC’ family of enzymes, which uncovers the critical function of PKCδ in neurons as a regulator of mitochondrial metabolism for LTM. Additionally, PKCδ activation and translocation to mitochondria result from LTM-specific dopamine signaling on MB neurons. By bridging experience-dependent neuronal circuit activity with metabolic modulation of memory-encoding neurons, PKCδ signaling binds the cognitive and metabolic constraints underlying LTM formation into a unified gating mechanism.

Our research aims to address the gap in understanding the sensitivity of multipurpose active pools of soil organic matter (SOM) and microbiological indicators, in addition to the usual physico-chemical measurements, to assess the effects of the shift from conventional to sustainable management practices under various pedoclimatic conditions. We conducted a meta-analysis (166 articles) on the sensitivity of these indicators on six practices: exogenous organic matter input (EOM), crop residue retention (RR), cover crop introduction (CC), crop rotation /diversification (CR), tillage reduction (TR), and conversion from conventional to innovative or organic systems (IOS). Random effects models considering the natural log of the response ratio were applied.Our study showed that climatic factors did not significantly influence indicator responses, while clay content, organic matter, pH, trial durations, and sampling depth significantly modulate the response of soil indicators. In detail, our results emphasized that both physico-chemical (soil organic carbon (SOC) and total nitrogen (STN) contents) and microbiological indicators (e.g., microbial biomass carbon (MBC), total phospholipids fatty acids (PLFA), and B-glucosidase (B-GLU)) were sensitive to EOM, RR, CC, and TR. For instance, CC significantly increased SOC (+7%) and STN (+8%), as well as MBC (+14 %), total PLFA (+19 %), and B-GLU (+11 %). On the other hand, CR and IOS practices did not impact SOC and STN, contrary to some microbiological indicators. For these two practices, MBC increased between 7 and 34 % and PLFA between 8 and 24 %. Moreover, B-GLU raised by + 58 % with IOS practice. This suggests that the microbiological indicators may be more responsive to specific agricultural practices, such as CR and IOS, even if the changes they induce are not immediately related to carbon inputs. SOC and STN provide information on the changes in the organic status of soil under carbon input practices and TR, though they are less sensitive to crop diversification and innovative farming in short-term studies, highlighting the time needed for sustainable management practices to modify soil health. Microbiological indicators (MBC and PLFA) and enzyme activities (FDA, B-GLU, and URE) showed greater sensitivity to a large range of agricultural practices, regardless of the age of agricultural practice, suggesting their potential use as more responsive, early, and versatile indicators in evaluating soil health.

Whole-pelvic radiotherapy (WPRT) has shown superiority over nodal stereotactic body radiotherapy (SBRT) in two retrospective cohorts. We aimed to compare both modalities and assess factors associated with treatment outcomes.Materials and methodsThis retrospective multicentric cohort study included patients from five institutions spanning from 2010 to 2022. Patients had a history of prostatic adenocarcinoma classified as N0 M0 at diagnosis with a first nodal-only pelvic castration-sensitive recurrence. Failure-free survival (FFS) was defined as the time from the end of RT to the first failure event–biochemical or imaging recurrence, or death.ResultsA total of 147 patients (pts) were analyzed, mainly treated for a recurrence after initial prostatectomy (87%), with 64 (43.5%) undergoing SBRT and 83 (56.5%) undergoing WPRT. SBRT was chosen mainly for dosimetric constraints (67%) and was associated with a lower rate of concomitant androgen deprivation therapy (ADT) prescription. With a median follow-up of 68 months [inter-quartile range (IQR) = 51], FFS was significantly lower in the SBRT group (p < 0.0001). In multivariable analysis, WPRT and ADT were associated with a longer FFS. Factors associated with a longer FFS after SBRT included associated ADT, lower prostate-specific antigen (PSA) levels, a PSA doubling time >6 months, and a Gleason score

The EU funded project EuroSea brought together key actors of the European ocean observing and forecasting communities with key users of the ocean observing products and services in order to better integrate existing ocean observation systems and tools, and to improve the delivery of ocean information to users. EuroSea was constructed around the ocean observing value chain that connects observations to users of ocean information, and, just as intended, the value chain concept was a useful prism to improve the system. In this article, we summarize some of the main take-home messages from EuroSea on the needs for developing the European Ocean Observing System and its links with modeling and forecasting systems. During the project, the challenges and gaps in the design and coordination of the European ocean observing and forecasting system were identified and mapped. Many gaps and challenges related to the observations of physical, chemical and biological Essential Ocean Variables were identified. Some of these gaps are related to technological developments, while others are caused by insufficient and short-term funding leading to a not sustainable system, management, and cooperation between different entities, as well as limitations in foresight activities, policies and decisions. This article represents a compilation of the broader needs for advancing the observing and forecasting system, and is meant as a guide for the community, and to funders and investors to advance ocean observing and the delivery of ocean information in Europe. To enhance the sustainability of ocean observations, which is paramount for a reliable provision of quality oceanographic data and services, several recommendations were compiled for ocean observing networks, frameworks, initiatives, as well as the ocean observing funders within the European nations, and the European Commission.

Passive hydroacoustic studies have underscored the efficiency and relevance of deploying autonomous hydrophones for the surveillance of underwater geophony. In particular, monitoring networks have been deployed for detecting SOFAR-propagating hydroacoustic waves generated by seismic events and locating their sources. The technique has been extended to study other hydroacoustic signals, such as P-waves from teleseismic events or impulsive waves generated by sea water-lava interactions. A significant challenge in this endeavor lies in the time required for the manual detection and annotation of these signals in long-term records. To address this issue, we tested the feasibility of implementing automated algorithms based on machine learning to detect and identify these various signals, and obtained satisfying classification and time picking accuracies. We incorporated those models in a benchmarking framework, proposing a training dataset, two evaluation datasets, two tasks to solve and the evaluations of the mentionned models on them. The goal of this framework is to foster the development of new models in the community, as it gives a clear way to evaluate them.

Developmental psychologists have long-established socio-cognitive abilities as fundamental to human intelligence and development. These abilities enable individuals to enter, learn from, and contribute to a surrounding culture. This drives the process of cumulative cultural evolution, which is responsible for humanity's most remarkable achievements. AI research on social interactive agents mostly concerns the emergence of culture in a multi-agent setting (often without a strong grounding in developmental psychology). We argue that AI research should be informed by psychology and study socio-cognitive abilities enabling to enter a culture as well. We draw inspiration from the work of Michael Tomasello and Jerome Bruner, who studied socio-cognitive development and emphasized the influence of a cultural environment on intelligence. We outline a broader set of concepts than those currently studied in AI to provide a foundation for research in artificial social intelligence. Those concepts include social cognition (joint attention, perspective taking), communication, social learning, formats, and scaffolding. To facilitate research in this domain, we present The SocialAI school—a tool that offers a customizable parameterized suite of procedurally generated environments. This tool simplifies experimentation with the introduced concepts. Additionally, these environments can be used both with multimodal RL agents, or with pure-text Large Language Models (LLMs) as interactive agents. Through a series of case studies, we demonstrate the versatility of the SocialAI school for studying both RL and LLM-based agents. Our motivation is to engage the AI community around social intelligence informed by developmental psychology, and to provide a user-friendly resource and tool for initial investigations in this direction.
