117. Accumulation of a putative guanidine compound in relation to other early defence reactions in epidermal cells of barley and wheat exhibiting resistance to Erysiphe graminis f.sp. hordei

Abstract

The infection process was compared in interactions between Erysiphe graminis f.sp. hordei isolate A6 and two near-isogenic barley lines (the susceptible line P-01 and the resistant line P-02), and the non-host wheat. A low frequency of successful infections, measured as the percentage of germinated conidia forming primary haustoria, was detected in both susceptible (7·5%), and resistant (73%) barley isolines and wheat plants (223%). In all three types of plants, papilla formation around the sites of attempted penetration seems to be a general resistance factor and thus not the determining step for distinguishing between race-specific and non-host resistance. In wheat plants, further resistance was manifested early after penetration and characterized by hypersensitively reacting (HR) single cells, no or limited growth of elongating secondary hyphae and no visible sign of infection. The race-specific resistance of barley isoline P-02 occurred later after penetration, and was characterized by plant tissue necrosis and no or weak fungal sporulation. In the susceptible barley isoline P-01, successful infections resulted in heavily sporulating powdery mildew colonies. Putative guanidine compounds, indicated by the Sakaguchi test, occurred in papillae 4-5 h after inoculation and subsequently in HR cells too, and are apparently associated with the very early resistance responses in barley and wheat. The highly basic guanidine compounds, not identified previously in plant defence responses, were found to be bound to the cell wall, and referred to as “basic staining material”. Accumulation of polyphenolic substances and callose were demonstrated in papillae and HR cells of both barley and wheat. Lignification occurred in wheat, but did not occur or did so only minimally in barley.

Abstract

The results presented here show that inoculation of barley with wheat powdery mildew (Erysiphe graminis f.sp. tritici), which is non-pathogenic on barley, is followed by an early and rapid activation of a specific set of genes as indicated by the accumulation of specific mRNAs. Two-dimensional polyacrylamide gel electrophoresis of in vitro translation products made from RNA isolated from inoculated and uninoculated barley leaves showed several new polypeptide products at 4 h after inoculation. These products were not apparent at 2 h and they are most probably of plant origin. Induction of new mRNA species correlated with interactions between the primary germ tubes of mildew spores and the epidermis. Analysis of RNA preparations from epidermal strips indicates that at least some of the induced mRNAs are also localized in the epidermis. The results are discussed in relation to other reports of gene expression in barley accompanying resistance responses, and in relation to induced resistance in barley against powdery mildew.

Abstract

Radiotherapy to the affected breast or chest wall is well established as an integral part of postoperative management of breast cancer. However, it is known to be associated with increased cardiac and pulmonary morbidities and mortalities. Modern technologies, such as CT planning, have shown to improve treatment planning by accurately delivering optimal doses to the target volumes, while minimizing doses to sensitive structures, thus reducing potential treatment-related adverse effects. Abstract

We investigate the possibility of transforming, under local operations and classical communication, a general bipartite quantum state on a dA x dB tensor-product space into a final state in 2 x 2 dimensions, while maintaining as much entanglement as possible. For pure states, we prove that Nielsen’s theorem provides the optimal protocol, and we present quantitative results on the degree of entanglement before and after the dimensional reduction. For mixed states, we identify a protocol that we argue is optimal for isotropic and Werner states. In the literature, it has been conjectured that some Werner states are bound entangled and in support of this conjecture our protocol gives final states without entanglement for this class of states. For all other entangled Werner states and for all entangled isotropic states some degree of free entanglement is maintained.
