Endophytic microorganisms represent promising solutions to environmental challenges inherent in conventional agricultural practices. This study concentrates on the identification of endophytic bacteria isolated from the root, stem, and leaf tissues of four Artemisia plant species. Sixty-one strains were isolated and sequenced by 16S rDNA. Sequencing revealed diverse genera among the isolated bacteria from different Artemisia species, including Bacillus, Pseudomonas, Enterobacter, and Lysinibacillus. AR11 and VR24 obtained from the roots of A. absinthium and A. vulgaris demonstrated significant inhibition on Fusarium c.f. oxysporum mycelial growth. In addition, AR11, AR32, and CR25 exhibited significant activity in phosphatase solubilization, nitrogen fixation, and indole production, highlighting their potential to facilitate plant growth. A comparative analysis of Artemisia species showed that root isolates from A. absinthium, A. campestris, and A. vulgaris have beneficial properties for inhibiting pathogen growth and enhancing plant growth. AR11 with 100% similarity to Bacillus thuringiensis, could be considered a promising candidate for further investigation as microbial biofertilizers. This finding highlights their potential as environmentally friendly alternatives to chemical pesticides, thereby contributing to sustainable crop protection practices.

Despite current efforts to minimize the impact of industry on ecosystems, the environment is polluted by a range of foreign substances, that can have a negative impact on the living organisms. Examples of widely studied substances are polycyclic aromatic hydrocarbons (PAHs) and recently, substances commonly used in conventional agriculture. In our study we focused on the morphology and anatomy of the vegetative organs of pea plants treated with the active substance of herbicides, flurochloridone (FLC), in concentration representing the residual amount in the soil (5 µM), and PAH fluoranthene (FLT) in concentration representing the middle to high environmental load (5 µM). During the long-term cultivation in nutrient solutions modified by the mentioned pollutants, the growth parameters of roots and shoots were observed in the three growth phases (4 and 8 fully developed leaves and the flowering phase). The growth parameters and observation of the morphology were supplemented with root and stem anatomical analysis using the transverse cross-sections. Both xenobiotics caused the decrease in the biomass production, while the more significant inhibition of growth, compared with control plants, was detected in FLC-treated plants, where the root system was reduced up to 75 % and growth parameters of the shoots were reduced about more than 50 %. The decrease in root biomass production was accompanied by changes in root branching. FLT treatment caused milder growth inhibition, it was observed about 50 % reduction of the root system induced by the shortening of the main and lateral roots. Less pronounced, was also the decrease in stem length caused by FLT. Similar information was obtained about the different degrees of effect of FLC and FLT using anatomical analysis. Both studied substances increased the main root diameter accompanied with increase of the average number of the primary cortex layers. Their influence also caused the more intensive formation of exodermis. Changes in anatomical architecture were also observed in stem, where the FLC treatment changed the arrangement of the vascular bundles and decreased their average number. Our elementary morpho-anatomical study suggests that FLC despite its trace concentration could be more detrimental to plants than FLT, known for its harmful effect on living organisms, in relatively higher concentration.

The constant increase in the high number of population, anthropogenic and industrial activities are alarming issues and raise food security issues around the world. The treatment of compost acts as an effective and environmentally friendly tool for an increase in vegetable production. This research article records the effects of three soil compost treatments (1) dry leaves (Ficus bengalensis L.), (2) solid waste (mustard oil cake) and (3) chicken manure, alone and in combination, with seedling growth performances of pea in pot culture experiments with five replications. The results showed that soil compost treatments showed a significant (p < 0.05) impact on the physicochemical features of soil and the physiological characteristics of pea (Pisum sativum L.). Soil compost analysis revealed a difference in soil porosity and pH, electrical conductivity, organic matter, total organic carbon, CaCO3, Na+, K+, phosphorous and nitrogen. Plant tissue analysis of pea seedlings also showed different levels of sodium, potassium and phosphorous values. The findings of the present research work concludes that the treatment of F. bengalensis dry leaves (25%) + garden loam (75%) treatment produced maximum growth performance, and the mustard oil cake solid waste (25%) + garden loam (75%) treatment showed the lowest seedling growth performance. The F. bengalensis dry leaves (25%) + garden loam (75%), soil compost application treatment showed the maximum seedling height of pea. The solid waste (SW) mixed with garden loam (GL) (SW 25% + GL 75%) and mustard oil cake solid waste alone treatment showed the lowest seedling growth of pea. The F. bengalensis dry leaves 25% + garden loam 75% compost treatment also influenced on the soil properties.

Abstract Dry pea (Pisum sativum) seeds are valuable sources of plant protein, dietary fiber, and starch, but their uses in food products are restricted to some extent due to several off‐flavor compounds. Saponins are glycosylated triterpenoids and are a major source of bitter, astringent, and metallic off‐flavors in pea products. β‐amyrin synthase (BAS) is the entry point enzyme for saponin biosynthesis in pea and therefore is an ideal target for knock‐out using CRISPR/Cas9 genome editing to produce saponin deficient pea varieties. Here, in an elite yellow pea cultivar (CDC Inca), LC/MS analysis identified embryo tissue, not seed coat, as the main location of saponin storage in pea seeds. Differential expression analysis determined that PsBAS1 was preferentially expressed in embryo tissue relative to seed coat and was selected for CRISPR/Cas9 genome editing. The efficiency of CRISPR/Cas9 genome editing of PsBAS1 was systematically optimized in pea hairy roots. From these optimization procedures, the AtU6‐26 promoter was found to be superior to the CaMV35S promoter for gRNA expression, and the use of 37°C was determined to increase the efficiency of CRISPR/Cas9 genome editing. These promoter and culture conditions were then applied to stable transformations. As a result, a bi‐allelic mutation (deletion and inversion mutations) was generated in the PsBAS1 coding sequence in a T1 plant, and the segregated psbas1 plants from the T2 population showed a 99.8% reduction of saponins in their seeds. Interestingly, a small but statistically significant increase (~12%) in protein content with a slight decrease (~5%) in starch content was observed in the psbas1 mutants under phytotron growth conditions. This work demonstrated that flavor‐improved traits can be readily introduced in any pea cultivar of interest using CRISPR/Cas9. Further field trials and sensory tests for improved flavor are necessary to assess the practical implications of the saponin‐free pea seeds in food applications.

ABSTRACT: Chicken diet essentially relies on soybean as the major source of proteins but there are increasing efforts to identify other protein-rich feedstuffs. Of these, some pea cultivars constitute interesting sources of proteins, although some of them contain antinutritional factors that may compromise the digestibility of their protein content. Consequently, chickens exhibit low performance, while undigested compounds rejected in feces have a negative environmental impact. In this article, we analyzed the intestinal content of chickens fed a pea diet (Pisum sativum) to decipher the mechanisms that could explain such a low digestibility. Using gelatin zymography, we observed that the contents of chicken fed the pea diet exhibit altered proteolytic activities compared with intestinal contents from chickens fed a rapeseed, corn, or soybean diet. This pea-specific profile parallels the presence of a 34 kDa protein band that resists proteolysis during the digestion process. Using mass spectrometry analysis, we demonstrated that this band contains the pea-derived Bowman-Birk protease inhibitor (BBI) and 3 chicken proteases, the well-known chymotrypsinogen 2-like (CTRB2) and trypsin II-P39 (PRSS2), and the yet uncharacterized trypsin I-P38 (PRSS3). All 3 proteases are assumed to be protease targets of BBI. Molecular modeling of the interaction of pea BBI with PRSS2 and PRSS3 trypsins reveals that electrostatic features of PRSS3 may favor the formation of a BBI-PRSS3 complex at physiological pH. We hypothesize that PRSS3 is specifically expressed and secreted in the intestinal lumen to form a complex with BBI, thereby limiting its inhibitory effects on PRSS2 and chymotrypsinogen 2-like proteases. These data clearly demonstrate that in chickens, feedstuff containing active pea BBI affects intestinal proteolytic activities. Further studies on the effects of BBI on the expression of PRSS3 by digestive segments will be useful to better appreciate the impact of pea on intestine physiology and function. From these results, we suggest that PRSS3 protease may represent an interesting biomarker of digestive disorders in chickens, similar to human PRSS3 that has been associated with gut pathologies.

The field investigations were conducted at Vegetable Research Farm, Department of vegetable Science, Punjab Agricultural University, Ludhiana with the objective to standarize the agronomic practices for mechical harvesting of garden pea. Therefore, effect of different agronomic traits including time of sowing, spacing and sowing method on yield and its attributing traits were studied on single-harvest garden pea. The experiment was laid out in split-plot design with 30 treatments comprising five dates of sowing in main plots and 2 different planting methods (flat and bed) sown at three different spacing in subplots and replicated three times. The bed size was standardized according to front-loading width of the pea combine. Significant interactions were observed between the date of sowing and spacing for a number of pods per plant, green seeds per pod, green pod yield per plant, green pod yield per plot; date of sowing and sowing method for pod length; spacing and sowing method for plant height. However, for all the traits, there was non-significant 3-way interaction. The late sown (20th December) crop resulted in the least number of pods per plant, green seeds per pod, green pod yield per plant and green pod yield per plot which was due to high temperature and low relative humidity conditions at pod development and filling stage. It is concluded that the garden pea cultivar Punjab-89 sown on 5th November at the spacing of 20 × 7.5 cm on beds (bed width of 1.0 m) resulted in significantly highest green pod yield of 12.75 kg/9 m2 in the single harvest. Therefore, the 3 beds of 1 m width can be harvested together with pea combine (because combine has working front width of 3.1 m) results into maximum yield when pea crops is sown on 5th November @ 20 × 7.5 cm spacing.

The study of the supramolecular organization of the mitochondrial oxidative phosphorylation system (OXPHOS) in various eukaryotes has led to the accumulation of a considerable amount of data on the composition, stoichiometry, and architecture of its constituent superstructures. However, the link between the features of system arrangement and the biological characteristics of the studied organisms has been poorly explored. Here, we report a comparative investigation into supramolecular and functional OXPHOS organization in the mitochondria of etiolated shoots of winter wheat (Triticum aestivum L.), maize (Zea mays L.), and pea (Pisum sativum L.). Investigations based on BN-PAGE, in-gel activity assays, and densitometric analysis revealed both similarities and specific OXPHOS features apparently related to the life strategies of each species. Frost-resistant winter wheat was distinguished by highly stable basic I1III2IVa/b respirasomes and V2 dimers, highly active complex I, and labile complex IV, which were probably essential for effective OXPHOS adaptation during hypothermia. Maize, a C4 plant, had the highly stable dimers IV2 and V2, less active complex I, and active alternative NAD(P)H dehydrogenases. The latter fact could contribute to successful chloroplast–mitochondrial cooperation, which is essential for highly efficient photosynthesis in this species. The pea OXPHOS contained detergent-resistant high-molecular respirasomes I1–2III2IVn, highly active complexes IV and V, and stable succinate dehydrogenase, suggesting an active energy metabolism in organelles of this plant. The results and conclusions are in good agreement with the literature data on the respiratory activity of mitochondria from these species and are summarized in a proposed scheme of organization of OXPHOS fragments. Tomato ketchup is one of the most popular foods eaten all over the world. To improve the texture of these sauces, modified starches are used most commonly. This may be negatively assessed by consumers. The solution to this problem could be the use of legume flours, as beyond thickening potential, they are recognized as plant foods of high nutritional value. The aim of the work was to estimate the applicability of pulse flour as a texture-forming agent for ketchup. A comprehensive assessment of the quality of ketchup was made, both in terms of sensory properties and instrumentally analyzed physicochemical features: acidity, color, texture, rheological properties, and dynamics of water molecules using the LF NMR method. It was stated that pulse flours are suitable for use as forming agents for ketchup, although they have slightly weaker thickening properties with a consistency index ranging from 5.06–6.82 Pa·sn, compared to acetylated distarch adipate (19.48 Pa·sn). Texture, which is the most important parameter for consumer acceptance of ketchup, can be successfully analyzed using instrumental methods. Firmness ranged from 0.51 N for lentil and pea-fortified ketchup to 0.55 N for the lupine variant. Ketchup thickened with different pulses and flours reveals slightly different individual sensory characteristics, so it makes it possible to create a new gamut of healthy tomato sauces. Pea is an important part of minor grains in Ningxia, which is of great significance in solving food security in southern mountainous areas of Ningxia, ensuring farmers’ income and wealth in remote mountainous areas, and promoting sustainable development of agriculture and animal husbandry. In order to screen new pea varieties ( lines) suitable for planting in the south-central mountainous areas of Ningxia, the growth period, main agronomic characters, yield and yield-related characters of 21 introduced fresh pea varieties (lines) were compared and identified with the local main variety Zhongwan 6 as the control. The results showed that the yields of WD21- 07, WD21- 12, WD21- 04, WD21-14 and WD21-05 were high, ranging from 3 520. 18 to 3 935. 08 kg / hm², which was 54. 73% higher than that of the control. It was suggested that WD21-14 should be put into production as high-yield varieties (lines), among which WD21-14 was suitable for mechanized harvesting;the remaining 16 varieties (lines) needed to be further tested and compared or preserved as breeding parents. Introduction: Although pulses are nutritious foods containing high amounts of protein, fiber and phytochemicals, their consumption and use in the food industry have been limited due to the formation of unappealing flavors/aromas described as beany, green, and grassy. Lipoxygenase (LOX) enzymes are prevalent among pulse seeds, and their activity can lead to the formation of specific volatile organic compounds (VOCs) from certain polyunsaturated fatty acids (PUFAs). As a widespread issue in legumes, including soybean, these VOCs have been linked to certain unappealing taste perception of foods containing processed pulse seeds. Methods: To address this problem in pea and as proof of principle to promote the wider use of pulses, a Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR) construct was designed to create null alleles (knockouts) of PsLOX2 which had been implicated in the generation of VOCs in peas. Results and discussion: Successful CRISPR/Cas9-mediated LOX gene editing of stable transgenic pea lines (TGP) was confirmed by DNA sequencing of the wild type (WT) and TGP pslox2 mutant lines. These lines were also assessed for LOX activity, PUFA levels, and VOCs. Compared to WT peas, the TGP lines showed a significant reduction (p < 0.05) in LOX activity and in the concentration of key VOCs, including hexanal, 2-hexenal, heptanal, (E)-2-heptenal, (E,E)-2,4-heptadienal, 1-octen-3-ol, octanal, (E)-2-octenal (E,E)-2,4-nonadienal and furan-2-pentyl. The content of two essential PUFAs, linoleic and α-linolenic acids, the known substrates of LOX in plants, was higher in TGP flours, indicating the efficacy of the CRISPR-mediated gene editing in minimizing their oxidation and the further modification of PUFAs and their products. The collection of VOCs from the headspace of ground pea seeds, using a portable eNose also distinguished the TGP and WT lines. Multiple regression analysis showed that LOX activity correlated with the two VOCs, heptanal and (E,E)-2,4-heptadienal in pea flours. Partial Least Squares Regression (PLS-R) plot for selected PUFAs, VOCs, and sensor responses in WT and TGP lines showed distinct clusters for WT and TGP lines. Together this data demonstrates the utility of CRISPR mediated mutagenesis of PsLOX2 to quickly improve aroma and fatty acid (FA) profiles of pea seeds of an elite Canadian variety. Diabetes prevalence achieved 470B in 2021. Diabetics are looking for foods that allow them to better manage the postprandial glycemia. Owing to its large amylose fraction, pea starch may contribute to formulate recipes with a lower glycemic index (GI). This study measured the rapidly, slowly digested and resistant fractions in pea starch and in a powder mix recipe. Starch fractions were determined according to the Englyst methodology. A nonblind repeat measure crossover design trial in healthy humans was used to study the GI of pea starch and maltodextrin powder mix recipes against glucose. Gastrointestinal symptoms were measured. Thirteen healthy volunteers aged 18–60 years with body mass index <30 kg/m2 and fasting blood glucose <6.1 mmol/L participated in the study. They consumed 25 g available carbohydrate portions of the test products. Blood glucose was measured at −5 and 0 min before consumption till 180 min after starting to eat. The slow digestible starch (SDS) content of native pea starch was 30% of the total starch content. The pea-based powder mix recipe contained 25% SDS in comparison with 9% for the maltodextrin-based recipe. The glucose response after pea starch was significantly lower compared with maltodextrin. The glucose response after pea starch recipe was significantly lower compared with maltodextrin recipe. There was no significant difference in mean scores for well-being and gastrointestinal symptoms after consumption of pea starch and maltodextrin or between the two recipes. In conclusion, this study has demonstrated the presence of high SDS content in pea starch, which reduced postprandial glycemic response compared with maltodextrin. The pea starch recipe did not induce any negative gastrointestinal symptoms. Pea starch may, therefore, prove to be a beneficial ingredient in developing food products for improving glycemic control without undesirable side effects. [ABSTRACT FROM AUTHOR]

The growing interest in the use of zinc oxide nanoparticles (ZnO NPs) in agriculture creates a risk of soil contamination with ZnO NPs, which can lead to phytotoxic effects on germinating seeds and seedlings. In the present study, the susceptibility of germinating seeds/seedlings of pea and wheat to ZnO NPs of various sizes (≤50 and ≤100 nm) applied at concentrations in the range of 100–1000 mg/L was compared. Changes in metabolic profiles in seedlings were analyzed by GC and GC-MS methods. The size-dependent harmful effect of ZnO NPs on the seedling's growth was revealed. The more toxic ZnO NPs (50 nm) at the lowest concentration (100 mg/L) caused a 2-fold decrease in the length of the wheat roots. In peas, the root elongation was slowed down by 20–30% only at 1000 mg/L ZnO NPs. The metabolic response to ZnO NPs, common for all tested cultivars of pea and wheat, was a significant increase in sucrose (in roots and shoots) and GABA (in roots). In pea seedlings, an increased content of metabolites involved in the aspartate–glutamate pathway and the TCA cycle (citrate, malate) was found, while in wheat, the content of total amino acids (in all tissues) and malate (in roots) decreased. Moreover, a decrease in products of starch hydrolysis (maltose and glucose) in wheat endosperm indicates the disturbances in starch mobilization. [ABSTRACT FROM AUTHOR]

Weeds are a key component of agroecosystems and compete with crop plants. Crop‐weed competition has been widely investigated over the past decades, although how functional traits mediate these competitive interactions has rarely been explored. In this study, we measured functional traits in 4400 individual plants belonging to 32 weed species, growing among four crops differing in functional traits. We assessed the intraspecific variation in weed functional traits in response to competition with crops but also with the other weeds. We found that the most abundant weeds have a lower intraspecific trait variability than other weed species for leaf dry matter content (LDMC), but this pattern was not observed for canopy height (CH) nor for specific leaf area (SLA). We found that most of the weed species tended to grow taller and have higher LDMC values when crop CH and LDMC increased. We propose that these species display a 'crop matching' strategy, tending to match crop functional trait values in response to competition. We also found that weed‐weed competition affected weed functional traits, sometimes more strongly than crop‐weed competition. This study highlights that intraspecific trait variation is important to consider when investigating the processes underlying weed species community assembly, especially resource competition. The growing demand for sustainable agriculture is raising interest in intercropping for its multiple potential benefits to avoid or limit the use of chemical inputs or increase the production per surface unit. Predicting the existence and magnitude of those benefits remains a challenge given the numerous interactions between interspecific plant-plant relationships, their environment, and the agricultural practices. Soil-crop models are critical in understanding these interactions in dynamics during the whole growing season, but few models are capable of accurately simulating intercropping systems. In this study, we propose a set of simple and generic formalisms (i.e. the structure and mathematical representation necessary for designing a model) for simulating key interactions in bi-specific intercropping systems that can be readily included into existing dynamic crop models. This requires simulating important processes such as development, light interception, plant growth, N and water balance, and yield formation in response to management practices, soil conditions, and climate. These formalisms were integrated into the STICS soil-crop model and evaluated using observed data of intercropping systems of cereal and legumes mixtures, including Faba bean-Wheat, Pea-Barley, Soybean-Sunflower, and Wheat-Pea mixtures. We demonstrate that the proposed formalisms provide a comprehensive simulation of soil-plant interactions in various types of bispecific intercrops. The model was found consistent and generic under a range of spring and winter intercrops (nRMSE = 25% for maximum leaf area index, 23% for shoot biomass at harvest, and 18% for grain yield). This is the first time a complete set of formalisms has been developed and published for simulating bi-specific intercropping systems and integrated into a soil-crop model. With its emphasis on being generic, sufficiently accurate, simple, and easy to parameterize, STICS is well-suited to help researchers designing in silico the agroecological transition by virtually pre-screening sustainable, manageable intercrop systems adapted to local conditions. Trichoderma spp. are considered in the category of biofertilizer microbes that have ability to increase nutrient uptake capacity and contribute in plant development. In the present investigation, recruitment of Trichoderma asperellum T42 with pea seeds increased total nitrogen uptake efficiency that promoted root traits, total biomass and yield. Molecular evidences revealed that T42 interaction with plant roots enhanced nitric oxide (NO) generation at 40, 70 and 90 days involved in several plant physiological processes. Interestingly, these growth parameters were elevated higher when plants were fed with nitrate nutrient as compared to ammonium fed condition at 40 and 70 days. NO generation in nitrogen nutrient confirmed that NO generation was produced via nitrate reductase (NR) dependent pathway. Several nitrate and ammonium transporters help increase in situ NO generation and contributed to increased lateral root initiation, in pea. Higher expression of nitrate transporter (NRT) genes in response to Trichoderma recruitment in nitrate fed condition and suppressive expression effect of ammonium transporter (AMT1.1) suggested that NRTs have more affinity for NO3− acquisition through pea roots. Among the NRTs, NRT2.1 and NRT1.2 were more upregulated as compared to control plant roots. Overall, our findings suggested that Trichoderma recruitment with pea seeds improved plant growth and yield, particularly more in nitrate fed condition providing insight of the strategy for not only pea but probably for other commercial leguminous crops which suffer from fluctuating nitrate availability in soil. Pea (Pisum sativum L.) is a vital leguminous crop farmed worldwide. Pea plays an essential role in China's crop rotation system, but the major restrictions to its cultivation are stability and low yield. Breeding for promising cultivars with a significantly high yield will impact the sustainability of pea production. Additionally, diverse environment trials are crucial in determining the best genotype. The new cultivar "Yunwan 52" was developed by hybridization and subsequently evaluated through yield trials among six pea genotypes across 14 environments during the 2016–2018 growing seasons. The results showed that the average yield of "Yunwan 52" for all tested environments was 2.64 t ha−1 compared to the control cultivar (Yunwan 18, 1.83 t ha−1). Analysis of AMMI variance showed significant differences (p < 0.01) between genotypes, environments, and their interaction. Based on the GGE biplot, some genotypes possessed wide and narrow adaptability to environments, such as Yunwan 52 was considered the most stable and ideal gen-otype across all tested environments. GYT biplot analysis also revealed that this realized cultivar was a superior and stable genotype that can be identified visually by combining all characters in breeding programs. Yunwan 52 distinguishes with purple blossoms and seed coat peas. It is possible to infer that the newly released cultivar "Yunwan 52" has outstanding yield performance and wide adaptability to multiple environmental conditions (resilience to abiotic stress). Pea (Pisum sativum) is an economically important legume crop, cultivated worldwide due to its high nutritional value and diverse culinary applications. However, like any agricultural crop, peas are vulnerable to various biotic and abiotic stresses, including fungal infection. The present paper deals with a study to identify the mycoflora associated with pea seed. The seed borne mycoflora of pea was examined by agar plate method, blotter method and Deep freezing blotter paper method. A total 17 species of fungi viz. We produced homogeneous lines of 227 pea accessions from the Nordic Genetic Resource Center via single seed descent. The genetic diversity among these, mostly Scandinavian accessions, was investigated using three microsatellite markers, A9, AC58 and AA5. The microsatellites were highly informative and separated 153 of 194 accessions on a Neighbor Joining topology. The high polymorphism information content (PIC) values between 0.87 and 0.91 indicated that the gene bank material contains a large number of pea accessions with different breeding histories. The peas were grown in the field for two years and seed protein content showed variation between 9.3% and 34.1% over the years and accessions, respectively. The mean thousand seed weight was 152.05 g. More than 10 accessions had a protein content above 28%, showing that the collection has potential as breeding nursery for high-protein pea cultivars. Background: Basic leucine zipper (bZIP) protein is a plant-specific transcription factor involved in various biological processes, including light signaling, seed maturation, flower development, cell elongation, seed accumulation protein, and abiotic and biological stress responses. However, little is known about the pea bZIP family. Results: In this study, we identified 87 bZIP genes in pea, named PsbZIP1 ~ PsbZIP87, via homology analysis using Arabidopsis. The genes were divided into 12 subfamilies and distributed unevenly in 7 pea chromosomes. PsbZIPs in the same subfamily contained similar intron/exon organization and motif composition. 1 tandem repeat event and 12 segmental duplication events regulated the expansion of the PsbZIP gene family. To better understand the evolution of the PsbZIP gene family, we conducted collinearity analysis using Arabidopsis thaliana, Oryza sativa Japonica, Fagopyrum tataricum, Solanum lycopersicum, Vitis vinifera, and Brachypodium distachyon as the related species of pea. In addition, interactions between PsbZIP proteins and promoters containing hormone- and stress-responsive cis-acting elements suggest that the regulation of PsbZIP expression was complex. We also evaluated the expression patterns of bZIP genes in different tissues and at different fruit development stages, all while subjecting them to five hormonal treatments. Conclusion: These results provide a deeper understanding of PsbZIP gene family evolution and resources for the molecular breeding of pea. In the Canadian prairies, pulse crops such as field pea (Pisum sativum L.) and lentil (Lens culinaris L.) are economically important and widely grown. However, in recent years, root rot, caused by a variety of fungal and oomycete pathogens, including Aphanomyces euteiches, has become a limiting factor on yield. In this study, we examined the impacts of nitrogen (N) fertilization and a commercial arbuscular mycorrhizal fungal (AMF) inoculant on pea and lentil plant health and agronomic production at three locations in Saskatchewan: Swift Current, Indian Head and Melfort. The AMF inoculation had no impact on root rot severity, and therefore is not considered a reliable method to manage root rot in pea and lentil. In contrast, N fertilization led to reductions in root rot in Swift Current, but not the other two sites. However, N fertilization did reduce nodulation. When both pea and lentil are considered, the abundance of A. euteiches in soil increased from pre-seeding to mid-bloom. A negative correlation between soil pH and disease severity was also observed. The high between-site variability highlights the importance of testing root rot mitigation strategies under multiple soil conditions to develop site-specific recommendations. Use of N fertilizer as a root rot management strategy merits further exploration, including investigation into its interactions with other management strategies, soil properties, and costs and benefits. N-palmitoylethanolamine (PEA) plays a key role in preventing Aβ-mediated neuroinflammation and neurotoxicity in murine models. It has been demonstrated that PEA provides anti-neuroinflammatory, pain-relieving and neuroprotective actions even in humans. In this project, we aim to evaluate these anti-neuroinflammatory effects via the cognitive evaluation and biochemical analyses of a 12-month oral administration of PEA in subjects with mild cognitive impairment (MCI). Subjects with MCI will be randomized to placebo or PEA groups, and followed for another 6 months. Cognitive abilities and neurological inflammation will be examined at baseline and after treatment. The specific objectives of the project are to ascertain whether: (i) PEA influences the scores of the neuropsychological and behavioral evaluations after one-year treatment, comparing PEA-treated and placebo subjects in both MCI and control groups; (ii) PEA can change the inflammatory and neuronal damage markers of blood and urine in MCI subjects; and (iii) these changes correlate with the clinical scores of participating subjects. Pea (Pisum sativum L.) plant, a legume species, is used as a vegetable for fresh consumption and frozen, canned or dried and processed and consumed as dry grain. The effects of organic fertilization, which is important for plant nutrition and soil fertility in pea cultivation, on grain yield and some agricultural characteristics were investigated. The experiment was carried out in the experiment field belonging to Selçuk University, Faculty of Agriculture, Prof. Dr. Abdülkadir AKÇİN Research and Application Station in 2021 according to the "Split Plots Experiment in Randomized Blocks" design with 3 replications. In the research, fertilizers were randomly placed in the main plots and varieties were randomly placed in the sub-plots. In the experiment, high yielding 16002, 16011, 16018, 16022 and Ultrello x Rondo genotypes suitable for fresh consumption and registered Betagreen pea variety were used as materials. Grain yield, some agronomic traits, protein yield and protein ratio were analyzed in the study. It was determined that the effects of chemical fertilizer and some organic fertilizer applications on all other traits examined in peas were statistically significant. According to the results of the research, the number of branches in pea plants was 3.18 pieces/plant (control) and 4.13 pieces/plant (chemical fertilizer), plant height was 45.89 cm (sheep manure) and 49.51 cm (chemical fertilizer), the number of pods was 14.31 pieces/plant (control) and 25.90 pieces/plant (chemical fertilizer), the number of grains in pods was 4.90 pieces (control) and 6.64 pieces (chemical fertilizer), the number of grains per plant was 70. 03 (control) to 172.20 (chemical fertilizer), hundred seed weight 29.30 g (control) to 33.22 g (chemical fertilizer), grain yield 128.75 kg da-1 (control) to 300.38 kg da-1 (chemical fertilizer), protein content 22.49% (control) to 23.12% (sheep manure) and protein yield 28.96 kg da-1 (control) to 67.91 kg da-1 (chemical fertilizer). The responses of genotypes to organic fertilizer applications showed differences. As a result, it was determined that organic fertilization in peas gave less grain yield than those grown with chemical fertilization, but protein content had higher values. Salinization is a significant adverse factor that suppresses productivity of agricultural plants. Multispectral imaging and calculation of reflectance indices, including the typical photochemical reflectance index (PRI), can be used for early revealing of changes in plants under salinization. However, the direction of change in typical PRI is dependent on the type of stressor and the experimental conditions. Earlier, we proposed modified PRIs with shifted measuring wavelengths and analyzed their changes under excess light, water deficit, and heating. In the current work, we investigated the sensitivity of these indices to changes in pea plants under salinization (100, 200, and 400 mM NaCl treatment). It was shown that short-wavelength PRIs were increased under this treatment; in contrast, long-wavelength PRIs were decreased. Most of the modified PRIs were strongly related to the maximal yield of photosystem II and the normalized difference vegetation index. Long-wavelength PRIs were more sensitive to plant changes than short-wavelength PRIs, typical PRI, and the normalized difference vegetation index because their stable decrease under moderate salinization (the 200 mM NaCl treatment) was initiated earlier. Our results show that long-wavelength PRIs, which also decreased under the action of the excess light, water deficit, and heating, can be potentially used as a universal tool for early revealing of stress changes in plants. The overall goal of this study was to investigate the effect of pH shifting on the functionality of pea, rice, oat, and hemp proteins and their blends (pea:rice, 60:40; pea:oat/hemp, 50:50). Proteins were processed at pH 2 or 10, with and without heat (80 °C), followed by neutralization at pH 7 and then spray dried. Depending on the protein source and pH treatment changes in the functional properties were observed. Solubility of all untreated samples was improved by at least one of the pH shift treatments whereas foaming stability was not. pH 10-heat-treated oat protein had the highest water-holding capacity at 5.71 g/g compared to 0.77–4.16 g/g for the other proteins. Only oat and hemp showed improved OHC after acidic or alkaline treatment. Emulsion stability was maximized in the pea-rice/oat blends after alkaline treatment, with or without heat. Blending proteins and subjecting them to pH shift treatment can result in unique functionalities that could be tailored to specific food applications. [ABSTRACT FROM AUTHOR]

Abstract Background Continuous cropping is a significant obstacle to sustainable development in the pea (Pisum sativum L.) industry, but the underlying mechanisms of this remain unclear. In this study, we used 16 S rDNA sequencing, transcriptomics, and metabolomics to analyze the response mechanism of roots and soil bacteria to continuous cropping and the relationship between soil bacteria and root phenotypes of different pea genotypes (Ding wan 10 and Yun wan 8). Results Continuous cropping inhibited pea growth, with a greater effect on Ding wan 10 than Yun wan 8. Metabolomics showed that the number of differentially accumulated metabolites (DAMs) in pea roots increased with the number of continuous cropping, and more metabolic pathways were involved. Transcriptomics revealed that the number of differentially expressed genes (DEGs) increased with the number of continuous cropping. Continuous cropping altered the expression of genes involved in plant-pathogen interaction, MAPK signal transduction, and lignin synthesis pathways in pea roots, with more DEGs in Ding wan 10 than in Yun wan 8. The up-regulated expression of genes in the ethylene signal transduction pathway was evident in Ding wan 10. Soil bacterial diversity did not change, but the relative abundance of bacteria significantly responded to continuous cropping. Integrative analysis showed that the bacteria with significant relative abundance in the soil were strongly associated with the antioxidant synthesis and linoleic acid metabolism pathway of pea roots under continuous cropping once. Under continuous cropping twice, the bacteria with significant relative abundance changes were strongly associated with cysteine and methionine metabolism, fatty acid metabolism, phenylpropanoid biosynthesis, terpenoid backbone biosynthesis, linoleic acid, and amino sugar and nucleotide sugar metabolism. Conclusion Ding wan 10 was more sensitive to continuous cropping than Yun wan 8. Continuous cropping times and pea genotypes determined the differences in root metabolic pathways. There were common metabolic pathways in the two pea genotypes in response to continuous cropping, and the DEGs and DAMs in these metabolic pathways were strongly associated with the bacteria with significant changes in relative abundance in the soil. This study provides new insights into obstacles to continuous cropping in peas.

As development of mushroom industry in Russia is growing, and the amount of spent mushroom composts is increasing, they are actively used as organomineral fertilizers for agricultural crops. Therefore, from the rather large problem of storing waste composts, these fertilizers began to be in great demand in the agricultural industry. Organic fertilizers are characterized by a high percentage of nutrients. They are able to improve physical properties of soil by increasing aggregative stability and reducing soil density. Organic fertilizers improve biological and biochemical properties of soil, and positively affect the development of soil microbial community. In the experiments, the content of nutrients in fresh compost and compost after one year of storage averaged: ash content - 8.0 and 74.1 %; organic matter - 66.3 and 25.9 %; NPK (%) - 0.50, 0.63, 0.44 and 0.45, 0.51, 0.39, respectively, under a favorable soil acidity (pH 8.0-7.3). In studies carried out on dark grey forest soils of the Ryazan region, the maximum productivity of agricultural crops was noted in variants with application of half-ripened compost: the yield of spring wheat was 2.82 t/ha (+ 0.23 t/ha compared to the control); spring barley - 3.21 t/ha (+0.17); spring rapeseed - 1.77 t/ha (+0.24); potato cv. Vympel - 27.91 (+ 4.41); potato cv. Kolobok - 11.21 t/ha (+1.63); pea - 2.10 t/ha (+0.21). The high efficiency of spent mushroom composts as fertilizers for increasing yields of spring barley and wheat, spring rapeseed, potatoes and peas was confirmed.

Abstract Background Seaweeds are a viable bioresource for suffering plants against salt stress, as they abundant in nutrients, hormones, vitamins, secondary metabolites, and many other phytochemicals that sustain plants' growth under both typical and stressful situations. The alleviating capacity of extracts from three brown algae (Sargassum vulgare, Colpomenia sinuosa, and Pandia pavonica) in pea (Pisum sativum L.) was investigated in this study. Methods Pea seeds were primed for 2 h either with seaweed extracts (SWEs) or distilled water. Seeds were then subjected to salinity levels of 0.0, 50, 100, and 150 mM NaCl. On the 21st day, seedlings were harvested for growth, physiological and molecular investigations. Results SWEs helped reduce the adverse effects of salinity on pea, with S. vulgare extract being the most effective. Furthermore, SWEs diminished the effect of NaCl-salinity on germination, growth rate, and pigment content and raised the osmolytes proline and glycine betaine levels. On the molecular level, two low-molecular-weight proteins were newly synthesized by the NaCl treatments and three by priming pea seeds with SWEs. The number of inter-simple sequence repeats (ISSR) markers increased from 20 in the control to 36 in 150 mM NaCl-treated seedlings, including four unique markers. Priming with SWEs triggered more markers than the control, however about ten of the salinity-induced markers were not detected following seed priming before NaCl treatments. By priming with SWEs, seven unique markers were elicited. Conclusion All in all, priming with SWEs alleviated salinity stress on pea seedlings. Salinity-responsive proteins and ISSR markers are produced in response to salt stress and priming with SWEs.

Pea (Pisum sativum L.) is an annual cool-season legume crop. Owing to its role in sustainable agriculture as both a rotation and a cash crop, its global market is expanding and increased production is urgently needed. For both technical and regulatory reasons, neither conventional nor transgenic breeding techniques can keep pace with the demand for increased production. In answer to this challenge, CRISPR/Cas9 genome editing technology has been gaining traction in plant biology and crop breeding in recent years. However, there are currently no reports of the successful application of the CRISPR/Cas9 genome editing technology in pea. We developed a transient transformation system of hairy roots, mediated by Agrobacterium rhizogenes strain K599, to validate the efficiency of a CRISPR/Cas9 system. Further optimization resulted in an efficient vector, PsU6.3-tRNA-PsPDS3-en35S-PsCas9. We used this optimized CRISPR/Cas9 system to edit the pea phytoene desaturase (PsPDS) gene, causing albinism, by Agrobacterium-mediated genetic transformation. This is the first report of successful generation of gene-edited pea plants by this route.

Background: Changes in the structural and physicochemical properties of pea starch could be significantly affected by germination treatment, which can provide a theoretical basis for promoting the use of this starch in the food industry. Methods: This work aims to evaluate the effect of germination time on the structural, thermal, rheological and functional properties of pea starches to determine their potential in the production of fermented beverages. The physicochemical changes during the germination process of peas and native and germinated starch granules were evaluated. Results: For germination critical time was 0.985 days, with 95% of germinated grains. The starch grains did not undergo any morphological change during the germination process as seen in the scanning electron microscopy images, indicating the absence of the α and ß - amylases responsible for the starch splitting. The X-ray patterns revealed that the crystalline structures of pea starch with and without germination were unchanged and contained by hexagonal and orthorhombic glucopyranose crystals. The viscosity profiles of the starches do not show significant changes; the most representative change is the increase in the gelatinization onset temperature of the paste from germinated starches compared to native starch. The functional properties of starches showed generally low values, with statistically significant differences between water absorption index, water solubility index, and starch swelling power and germination time. Conclusions: In general terms, it can be concluded that lentil starch does not undergo significant changes in its physicochemical and functional integrity with respect to the grain germination process.

This study aimed to explore the feasibility of substituting wheat flour with varying levels (10%, 15%, 20%, and 25%) of flour derived from field bean, chickpea, lentil, and pea seeds. The investigation focused on assessing the physical properties of wheat dough and the physicochemical characteristics of bread samples. The addition of legume seed flours significantly influenced the dough’s development time, particularly with chickpea flour causing a notable increase in this parameter. While dough stability was generally shorter for mixtures containing wheat flour and legume seed flour, chickpea flour was an exception, significantly prolonging dough stability time. Furthermore, the inclusion of legume flours resulted in increased protein, ash, fiber, fat, and phenolic contents in the enriched bread, while the carbohydrate content decreased. Additionally, the crumb exhibited increased redness and yellowness and decreased lightness due to the enrichment of the bread. Notably, the antioxidant activity of bread containing legume flour also increased, with the most significant increase observed when pea flour was utilized. Conversely, negative effects on bread volume, crumb density, and texture parameters were noted with the incorporation of legume additives. Taking into consideration the results of both physicochemical analyses and sensory evaluation, it is recommended that the incorporation of the specified legume flours should not exceed 15% in relation to the quantity of wheat flour used.

There is a pressing need for efficacious therapies in the field of respiratory diseases and infections. Lipid nanocarriers, administered through aerosols, represent a promising tool for maximizing therapeutic concentration in targeted cells and minimizing systemic exposure. However, this approach requires the application of efficient and safe nanomaterials. Palmitoylethanolamide (PEA), an endocannabinoid-like endogenous lipid, plays a crucial role in providing protective mechanisms during inflammation, making it an interesting material for preparing inhalable lipid nanoparticles (LNPs). This report aims to preliminarily explore the in vitro behavior of LNPs prepared with PEA (PEA-LNPs), a new inhalable inflammatory-targeted nanoparticulate drug carrier. PEA-LNPs exhibited a size of about 250 nm, a rounded shape, and an marked improvement in PEA solubility in comparison to naked PEA, indicative of easily disassembled nanoparticles. A twin glass impinger instrument was used to screen the aerosol performance of PEA-LNP powders, obtained via freeze-drying in the presence of two quantities of mannose as a cryoprotectant. Results indicated that a higher amount of mannose improved the emitted dose (ED), and in particular, the fine particle fraction (FPF). A cytotoxicity assay was performed and indicated that PEA-LNPs are not toxic towards the MH-S alveolar macrophage cell line up to concentrations of 0.64 mg/mL, and using coumarin-6 labelled particles, a rapid internalization into the macrophage was confirmed. This study demonstrates that PEA could represent a suitable material for preparing inhalable lipid nanocarrier-based dry powders, which signify a promising tool for the transport of drugs employed to treat respiratory diseases and infections.

Analysis of the viromes of three symptomatic Fabaceae plants, i.e., red clover (Trifolium pratense L.), pea (Pisum sativum L.), and common bean (Phaseolus vulgaris L.), using high-throughput sequencing revealed complex infections and enabled the acquisition of complete genomes of a potyvirus, bean yellow mosaic virus (BYMV). Based on phylogenetic analysis, the Slovak BYMV isolates belong to two distinct molecular groups, i.e., VI (isolate FA40) and XI (isolates DAT, PS2). Five commercial pea genotypes (Alderman, Ambrosia, Gloriosa, Herkules, Senator) were successfully infected with the BYMV-PS2 inoculum and displayed similar systemic chlorotic mottling symptoms. Relative comparison of optical density values using semi-quantitative DAS-ELISA revealed significant differences among virus titers in one of the infected pea genotypes (Ambrosia) when upper fully developed leaves were tested. Immunoblot analysis of systemically infected Alderman plants showed rather uneven virus accumulation in different plant parts. The lowest virus accumulation was repeatedly detected in the roots, while the highest was in the upper part of the plant stem.

Pea is a major pulse crop in temperate regions and a model plant in genetics. Large genetic marker resources are needed to assess the genetic diversity in the species genepool and to provide selection tools for breeders. In this study, we used second-generation sequencing to perform an exome-capture protocol using a diverse pea germplasm collection, and produced a resource of over 2 million Single Nucleotide Polymorphisms. This dataset was then used to characterize the genetic diversity present in the panel and compute phylogenetic and structure analyses. The development of this resource paves the way for Genome-wide association studies and the development of powerful genotyping tools.

IntroductionAlthough pulses are nutritious foods containing high amounts of protein, fiber and phytochemicals, their consumption and use in the food industry have been limited due to the formation of unappealing flavors/aromas described as beany, green, and grassy. Lipoxygenase (LOX) enzymes are prevalent among pulse seeds, and their activity can lead to the formation of specific volatile organic compounds (VOCs) from certain polyunsaturated fatty acids (PUFAs). As a widespread issue in legumes, including soybean, these VOCs have been linked to certain unappealing taste perception of foods containing processed pulse seeds.MethodsTo address this problem in pea and as proof of principle to promote the wider use of pulses, a Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR) construct was designed to create null alleles (knockouts) of PsLOX2 which had been implicated in the generation of VOCs in peas.Results and discussionSuccessful CRISPR/Cas9-mediated LOX gene editing of stable transgenic pea lines (TGP) was confirmed by DNA sequencing of the wild type (WT) and TGP pslox2 mutant lines. These lines were also assessed for LOX activity, PUFA levels, and VOCs. Compared to WT peas, the TGP lines showed a significant reduction (p < 0.05) in LOX activity and in the concentration of key VOCs, including hexanal, 2-hexenal, heptanal, (E)-2-heptenal, (E,E)-2,4-heptadienal, 1-octen-3-ol, octanal, (E)-2-octenal (E,E)-2,4-nonadienal and furan-2-pentyl. The content of two essential PUFAs, linoleic and α-linolenic acids, the known substrates of LOX in plants, was higher in TGP flours, indicating the efficacy of the CRISPR-mediated gene editing in minimizing their oxidation and the further modification of PUFAs and their products. The collection of VOCs from the headspace of ground pea seeds, using a portable eNose also distinguished the TGP and WT lines. Multiple regression analysis showed that LOX activity correlated with the two VOCs, heptanal and (E,E)-2,4-heptadienal in pea flours. Partial Least Squares Regression (PLS-R) plot for selected PUFAs, VOCs, and sensor responses in WT and TGP lines showed distinct clusters for WT and TGP lines. Together this data demonstrates the utility of CRISPR mediated mutagenesis of PsLOX2 to quickly improve aroma and fatty acid (FA) profiles of pea seeds of an elite Canadian variety.

Background/Purpose: Pulse palpation is an unreliable method for diagnosing cardiac arrest. To address this limitation, continuous hemodynamic monitoring may be a viable solution. Therefore, we developed a novel, hands-free Doppler system, RescueDoppler, to detect the pulse continuously in the carotid artery. Methods: In twelve pigs, we evaluated RescueDoppleŕs potential to measure blood flow velocity in three situations where pulse palpation of the carotid artery was insufficient: (1) systolic blood pressure below 60 mmHg, (2) ventricular fibrillation (VF) and (3) pulseless electrical activity (PEA). (1) Low blood pressure was induced using a Fogarty balloon catheter to occlude the inferior vena cava. (2) An implantable cardioverter-defibrillator induced VF. (3) Myocardial infarction after microembolization of the left coronary artery caused True-PEA. Invasive blood pressure was measured in the contralateral carotid artery. Time-averaged blood flow velocity (TAV) in the carotid artery was related to mean arterial pressure (MAP) in a linear mixed model. Results: RescueDoppler identified pulsatile blood flow in 41/41 events with systolic blood pressure below 60 mmHg, with lowest blood pressure of 19 mmHg. In addition the absence of spontaneous circulation was identified in 21/21 VF events and true PEA in 2/2 events. The intraclass correlation coefficient within animals for TAV and MAP was 0.94 (95% CI. 0.85–0.98). Conclusions: In a porcine model, RescueDoppler reliably identified pulsative blood flow with blood pressures below 60 mmHg. During VF and PEA, circulatory arrest was rapidly and accurately demonstrated. RescueDoppler could potentially replace unreliable pulse palpation during cardiac arrest and cardiopulmonary resuscitation.

Agricultural biotechnology has become familiar with nanomaterials' properties and potential use. The present experiment was conducted to examine the effect of 8.1 ± 1.6 nm sized silver nanoparticles (AgNPs) on the growth performance and yield quality of pea (Pisum sativum) as well as Arbuscular Mycorrhizal Fungi (AMF) and Rhizobium population in the soil. Pea seeds were separately treated in 3 h with 1 mM, 2.5 mM, and 5 mM of AgNPs aqueous solution, and after 2 weeks their effect on seed germination, leaf number, shoot, and root length, shoot and root dry weight, fresh biomass, and dry biomass weight were investigated in laboratory condition using Murashige and Skoog (MS) Basal Medium. Results showed that 2.5 mM AgNPs had a significant positive impact and notably increased all the experimental growth parameters mentioned above of pea compared with the control. The experiment was also conducted in an open environment under a natural condition where 2.5 mM AgNPs improved 31.57 % weight of fruit and 9.09 % of seed weight without causing any harm to the symbiotic microbes (AMF and Rhizobium bacteria). Whereas, the other concentrations of nanoparticles (1.0 mM and 5.0 mM) showed varied impacts on the germination, growth, and yield of pea in comparison with the control. For the first time, the outcome demonstrated the successful use of 2.5 mM AgNPs in enhancing the growth and yield of peas, and increased AMF colonization but showed no impact on Rhizobium compared to the control.
