In this paper, we proposed and analyzed a five-dimensional system of ordinary differential equations modeling the competition of two competing bacteria in a chemostat under the influence of the leachate recirculation and in the presence of a pathogen associated only with the bacteria 1. We suppose that the nutriment is present into two forms, soluble and insoluble nutriment, and both two forms are continuously added to the chemostat. The proposed model takes the form of an “SI” epidemic model and uses general increasing growth functions and general increasing incidence rate. It admits multiple equilibria that we give the conditions under which we assure both the existence and the local stability of each equilibrium point. The possibility of periodic trajectory was excluded, and the uniform persistence of both types of bacteria was proved. Phytopathogenic microbes use secreted effector proteins to increase their virulence in planta. If these effectors or the results of their activity are detected by the plant cell, the plant will mount an immune response which applies evolutionary pressure by reducing growth and success of the pathogen. Bacterial effector proteins in the AvrRps4 family (AvrRps4, HopK1, and XopO) have commonly been used as tools to investigate plant immune components. At the same time, the in planta functions of this family of effectors have yet to be fully characterized. In this minireview we summarize current knowledge about the AvrRps4 effector family with emphasis on properties of the proteins themselves. We hypothesize that the HopK1 C-terminus and the AvrRps4 Cterminus, though unrelated in sequence and structure, are broadly related in functions that counteract plant defense responses. Fertilizers and plant diseases contribute positively and negatively to crop production, respectively. Macro- and micronutrients provided by the soil and fertilizers are transported by various plant nutrient transporters from the soil to the roots and shoots, facilitating growth and development. However, the homeostasis of different nutrients has different effects on plant disease. This review is aimed at providing insights into the interconnected regulation between nutrient homeostasis and immune responses, and it highlights strategies to enhance disease resistance by optimal manipulation of nutrient transporters in rice. First, we highlight the essential roles of six macronutrients (nitrogen, phosphorus, potassium, sulfur, calcium, magnesium) and eight micronutrients (iron, manganese, zinc, copper, boron, molybdenum, silicon, nickel), and summarize the diverse effects of each on rice diseases. We then systematically review the molecular mechanisms of immune responses modulated by nutrient transporters and the genetic regulatory pathways that control the specific nutrient-mediated immune signaling that is regulated by the pathogens and the host plant. This study aimed to obtain and characterize an oil-in-water nanoemulsion (NE) loaded with an in vitro optimized bactericidal essential oil blend of 50% oregano, 40% thyme, and 10% lemongrass and to evaluate its potential at three different concentrations (0.5%, 1%, and 2%) in the inactivation of Escherichia coli, Staphylococcus aureus, and Salmonella enterica serotype Enteritidis inoculated in rainbow trout fillets stored at 4 °C for 9 days. Regarding the NE, the nanometric size (<100 nm) with low polydispersion (0.17 ± 0.02) was successfully obtained through ultrasound at 2.09 W/cm2. Considering the three concentrations used, S. Enteritidis was the most susceptible. On the other hand, comparing the concentrations used, the NE at 2% showed better activity, reducing S. Enteritidis, E. coli, and S. aureus by 0.33, 0.20, and 0.73 log CFU/g, respectively, in the trout fillets. Thus, this data indicates that this is a promising eco-friendly alternative to produce safe fish for consumption and reduce public health risks. [ABSTRACT FROM AUTHOR]

Host–pathogen interactions can be influenced by the host microbiota, as the microbiota can facilitate or prevent pathogen infections. In addition, members of the microbiota can become virulent. Such pathobionts can cause co-infections when a pathogen infection alters the host immune system and triggers dysbiosis. Here we performed a theoretical investigation of how pathobiont co-infections affect the evolution of pathogen virulence. We explored the possibility that the likelihood of pathobiont co-infection depends on the evolving virulence of the pathogen. We found that, in contrast to the expectation from classical theory, increased virulence is not always selected for. For an increasing likelihood of co-infection with increasing pathogen virulence, we found scenario-specific selection for either increased or decreased virulence. Evolutionary changes, however, in pathogen virulence do not always translate into similar changes in combined virulence of the pathogen and the pathobiont. Only in one of the scenarios where pathobiont co-infection is triggered above a specific virulence level we found a reduction in combined virulence. This was not the case when the probability of pathobiont co-infection linearly increased with pathogen virulence. Taken together, our study draws attention to the possibility that host–microbiota interactions can be both the driver and the target of pathogen evolution. At a rapid pace, biologists are learning the many ways in which resident microbes influence, and sometimes even control, their hosts to shape both health and disease. Understanding the biochemistry behind these interactions promises to reveal completely novel and targeted ways of counteracting disease processes. However, in our protocols and publications, we continue to describe these new results using a language that originated in a completely different context. This language developed when microbial interactions with hosts were perceived to be primarily pathogenic, as threats that had to be vanquished. Biomedicine had one dominating thought: winning this war against microorganisms. Today, we know that beyond their defensive roles, host tissues, especially epithelia, are vital to ensuring association with the normal microbiota, the communities of microbes that persistently live with the host. Thus, we need to adopt a language that better encompasses the newly appreciated importance of host-microbiota associations. We also need a language that frames the onset and progression of pathogenic conditions within the context of the normal microbiota. Such a reimagined lexicon should make it clear, from the very nature of its words, that microorganisms are primarily vital to our health, and only more rarely the cause of disease. Pseudogenes are defined as "non-functional" copies of corresponding parent genes. The cognition of pseudogenes continues to be refreshed through accumulating and updating research findings. Previous studies have predominantly focused on mammals, but pseudogenes have received relatively less attention in the field of microbiology. Given the increasing recognition on the importance of pseudogenes, in this review, we focus on several aspects of microorganism pseudogenes, including their classification and characteristics, their generation and fate, their identification, their abundance and distribution, their impact on virulence, their ability to recombine with functional genes, the extent to which some pseudogenes are transcribed and translated, and the relationship between pseudogenes and viruses. By summarizing and organizing the latest research progress, this review will provide a comprehensive perspective and improved understanding on pseudogenes in microorganisms. Introduction: The use of antimicrobials is a permanent challenge as it is constantly confronted with the ability of bacteria to develop resistance mechanisms. Objective: The objective of this research was to evaluate the antimicrobial effect of polyphenolic extracts present in Ananas comosus crown. Materials and Methods: 3.5 kg of Ananas comosus pineapple crown leaves were collected and taken to a tray dryer to reduce humidity, then, they were milled until a fine flour was obtained, for the extraction process of the oleaginous extract a Soxhlet equipment was used, using 70% ethanol as solvent, The identification of phenolic compounds was carried out by ultra-high resolution liquid chromatography with an Orbitrap mass detector. The microbiological analysis was evaluated by the standardized method of diffusion with discs using Mueller-Hinton agar, for which the Staphylococcus aureus strain (ATCC 25923) was used, using oxacillin as a positive control and DMSO as a negative control. Results: from the drying obtained, the humidity was reduced by 50%, with which the dry matter obtained was ground and used to carry out the extraction process of the oleaginous extract, obtaining 63 ml of which 27 phenolic compounds were identified. As for the microbiological analysis carried out, inhibition halos varying between 4.5 mm and 6.0 mm were observed. Conclusion: Finally, it was concluded that the polyphenols present in the extract of Ananas comosus showed antibacterial activity on Staphylococcus aureus, with a greater inhibition effect observed when a higher concentration of the extract was applied. Galleria mellonella is used as a model organism to study the innate immune response of insects. In this study, the humoral immune response was assessed by examining phenoloxidase activity, fungal burden, and the expression of phenoloxidase and antimicrobial peptide genes at different time point following separate and combined injections of Hypericum perforatum extract and a nonlethal dose of Candida albicans. The administration of a plant extract at low doses increased phenoloxidase activity, while higher doses had no effect. Similarly, co‐injection of a low dose of the extract with the pathogen allowed half of the yeast cells to survive after 24 h. Co‐injection of plant extract with the pathogen decreased the phenoloxidase activity at the end of 4 h compared to C. albicans mono‐injection. The phenoloxidase gene expressions was reduced in all experimental conditions with respect to the control. When plant extracts and the pathogen were administered together, gallerimycin and hemolin gene expressions were considerably higher compared to mono‐injections of plant extracts and the pathogen. The results of this study reveal that gene activation and regulatory mechanisms may change for each immune gene, and that recognition and signaling pathways may differ depending on the involved immunoregulator. Batrachochytrium dendrobatidis (Bd) is a lethal amphibian pathogen, partly due to its ability to evade the immune system of susceptible frog species. In many pathogenic fungi, the antioxidant glutathione is a virulence factor that helps neutralise oxidative stressors generated from host immune cells, as well as other environmental stressors such as heavy metals. The role of glutathione in stress tolerance in Bd has not been investigated. Here, we examine the changes in the glutathione pool after stress exposure and quantify the effect of glutathione depletion on cell growth and stress tolerance. Depletion of glutathione repressed growth and release of zoospores, suggesting that glutathione is essential for life cycle completion in Bd. Supplementation with <2 mM exogenous glutathione accelerated zoospore development, but concentrations >2 mM were strongly inhibitory to Bd cells. While hydrogen peroxide exposure lowered the total cellular glutathione levels by 42 %, glutathione depletion did not increase the sensitivity to hydrogen peroxide. Exposure to cadmium increased total cellular glutathione levels by 93 %. Glutathione-depleted cells were more sensitive to cadmium, and this effect was attenuated by glutathione supplementation, suggesting that glutathione plays an important role in cadmium tolerance. The effects of heat and salt were exacerbated by the addition of exogenous glutathione. Due to being sessile, plants develop a broad range of defense pathways when they face abiotic or biotic stress factors. Although plants are subjected to more than one type of stress at a time in nature, the combined effects of either multiple stresses of one kind (abiotic or biotic) or more kinds (abiotic and biotic) have now been realized in agricultural lands due to increases in global warming and environmental pollution, along with population increases. Soil-borne pathogens, or pathogens infecting aerial parts, can have devastating effects on plants when combined with other stressors. Obtaining yields or crops from sensitive or moderately resistant plants could be impossible, and it could be very difficult from resistant plants. The mechanisms of combined stress in many plants have previously been studied and elucidated. Recent studies proposed new defense pathways and mechanisms through signaling cascades. In light of these mechanisms, it is now time to develop appropriate strategies for crop protection under multiple stress conditions. This may involve using disease-resistant or stress-tolerant plant varieties, implementing proper irrigation and drainage practices, and improving soil quality. However, generation of both stress-tolerant and disease-resistant crop plants is of crucial importance. The establishment of a database and understanding of the defense mechanisms under combined stress conditions would be meaningful for the development of resistant and tolerant plants. It is clear that leaf pathogens show great tolerance to salinity stress and result in pathogenicity in crop plants. We noticed that regulation of the stomata through biochemical applications and some effort with the upregulation of the minor gene expressions indirectly involved with the defense mechanisms could be a great way to increase the defense metabolites without interfering with quality parameters. In this review, we selected wheat as a model plant and Zymoseptoria tritici as a model leaf pathogen to evaluate the defense mechanisms under saline conditions through physiological, biochemical, and molecular pathways and suggested various ways to generate tolerant and resistant cereal plants. In contemporary times, the sustained aspiration of bioengineering and biomedical applications is the progressive advancement of materials characterized by biocompatibility and biodegradability. The investigation of the potential applications of polymers as natural and non-hazardous materials has placed significant emphasis on their physicochemical properties. Thus, this study was designed to investigate the potential of gelatin–chitosan–moringa leaf extract (G–CH–M) as a novel biomaterial for biomedical applications. The wound-dressing G–CH–M biopolymer was synthesized and characterized. The blood haemolysis, anti-inflammatory, antioxidant, and antibacterial activities of the biopolymer were investigated against Gram-positive (Staphylococcus aureus) and Gram-negative (Escherichia coli) bacterial isolates. Our results showed that S. aureus swarming motility was drastically affected. However, the biopolymer had no significant effect on the swarming motility of E. coli. In addition, the biopolymer showed high antibacterial capacities, especially against S. aureus. Plasmid DNA was observed to be effectively protected from oxidative stresses by the biopolymer. Furthermore, the biopolymer exhibited greatly suppressed haemolysis (lower than 2%), notwithstanding the elevated concentration of 50 mg/mL. A long‐standing debate exists among ecologists as to how diversity regulates infectious diseases (i.e., the nature of diversity‐disease relationships); a dilution effect refers to when increasing host diversity inhibits infectious diseases (i.e., negative diversity‐disease relationships). However, the generality, strength, and potential mechanisms underlying negative diversity‐disease relationships in natural ecosystems remain unclear. To this end, we conducted a large‐scale survey of 63 grassland sites across China to explore diversity‐disease relationships. We found widespread negative diversity‐disease relationships that were temperature‐dependent; non‐random diversity loss played a fundamental role in driving these patterns. Our study provides field evidence for the generality and temperature dependence of negative diversity‐disease relationships in grasslands, becoming stronger in colder regions, while also highlighting the role of non‐random diversity loss as a mechanism. BACKGROUND: Many diseases may be caused by pathogens and oxidative stress resulting from carcinogens. Earlier studies have highlighted the antimicrobial and antioxidant effects of plant essential oils (EO). It is crucial to effectively utilize agricultural waste to achieve a sustainable agricultural economy and protect the environment. The present study aimed to evaluate the potential benefits of EO extracted from the discarded peels of Citrus depressa Hayata (CD) and Citrus microcarpa Bunge (CM), synonyms of Citrus deliciosa Ten and Citrus japonica Thunb, respectively. RESULTS: Gas chromatography‐mass spectrometry analysis revealed that the main compounds in CD‐EO were (R)‐(+)‐limonene (38.97%), γ‐terpinene (24.39%) and linalool (6.22%), whereas, in CM‐EO, the main compounds were (R)‐(+)‐limonene (48.00%), β‐pinene (13.60%) and γ‐terpinene (12.07%). CD‐EO exhibited inhibitory effects on the growth of common microorganisms, including Candida albicans, Escherichia coli, Pseudomonas aeruginosa and Staphylococcus aureus. However, CM‐EO showed only inhibitory effects on E. coli. Furthermore, CD‐EO exhibited superior antioxidant potential, as demonstrated by its ability to eliminate 1,1‐diphenyl‐2‐picrylhydrazyl and 2,2′‐azinobis‐3‐ethylbenzthiazoline‐6‐sulfonate free radicals. Furthermore, CD‐EO at a concentration of 100 μg mL−1 significantly inhibited 12‐O‐tetradecanoylphorbol‐13‐acetate‐induced cancer transformation in mouse epidermal JB6 P+ cells (P < 0.05), possibly by up‐regulating protein expression of nuclear factor erythroid 2‐related factor 2 and its downstream antioxidant enzymes, such as NAD(P)H:quinone oxidoreductase 1, heme oxygenase‐1 and UGT1A. The widespread use of pesticides poses significant challenges to the safety of agricultural products and the ecological environment. Levan‐type fructans have the ability to act as an induced resistance stimulus, enhancing plant disease resistance, which aligns with the principles of green development. This study showed that levan polysaccharide, produced by Halomonas smyrnnensis levansucrase, could induce the resistance of barley to Fusarium graminearum much better than inulin‐type fructans or low‐molecular‐weight chitosan oligosaccharide. Three different application methods of levan, namely seed soaking, leaf spraying or their combination, all decreased the necrotic areas caused by F. graminearum and Rhizoctonia solani. When inoculated with F. graminearum, the content of hydrogen peroxide, proline or jasmonic acid and the activities of peroxidase and ascorbate peroxidase in levan‐treated barley were higher than those in the control. In contrast, when inoculated with R. solani, the hydrogen peroxide content and peroxidase activity in levan‐treated leaves showed a faster induction than the control at the early priming and the content of proline and methyl salicylate was higher than that of the control. Pathogens are ubiquitous in ecosystems and play a key role in affecting host community structure. In grasslands, large grazing animals such as cattle and sheep have been shown to affect foliar fungal pathogens. However, theory and empirical studies have come to conflicting conclusions because grazers can directly and indirectly impact pathogens through a wide variety of mechanisms and various grazers may impact pathogens in different ways. A better understanding of the mechanisms by which grazers impact pathogens is important for a fundamental understanding of herbivore pathogen interactions and also to optimise grazing managements to reduce pathogen outbreaks.Here, we investigate multiple mechanisms by which livestock grazing impacts foliar fungal pathogens in grasslands. We integrate a large‐scale grazing experiment, with a removal experiment manipulating plant density and litter biomass, to identify direct and indirect effects of two herbivores on pathogens with different life histories (biotrophs and necrotrophs), in a temperate grassland in northeast China.We found that grazing by cattle and sheep had contrasting impacts: cattle grazing significantly reduced pathogen load, of both biotrophs and necrotrophs, whereas sheep grazing increased biotrophic pathogen load, but did not affect the necrotrophs. The grazing effects were mostly indirect and mediated by different impacts of the herbivores on plant community structure. Cattle grazing reduced pathogen load because it reduced the abundance of susceptible, fast‐growing plants, and the overall density of plants, while sheep grazing increased pathogen infection because it reduced the abundance of resistant plant species. Plant diversity also reduced pathogen infection but these effects were independent of the herbivores.Our results show that different herbivores can have contrasting impacts on pathogen infection through contrasting impacts on host community competence. This suggests the importance of considering multiple mechanisms simultaneously to evaluate the impact of herbivores on host‐pathogen interactions. Data support that enterotoxigenic Bacteroides fragilis (ETBF) harbouring the Bacteroides fragilis toxin (bft) gene may promote colorectal tumourigenesis through the serrated neoplasia pathway. We hypothesized that ETBF may be enriched in colorectal carcinoma subtypes with high-level CpG island methylator phenotype (CIMP-high), BRAF mutation, and high-level microsatellite instability (MSI-high). Quantitative PCR assays were designed to quantify DNA amounts of Bacteroides fragilis , ETBF, and each bft gene isotype (bft-1 , bft-2 , or bft-3) in colorectal carcinomas in the Health Professionals Follow-up Study and Nurses' Health Study. We used multivariable-adjusted logistic regression models with the inverse probability weighting method. We documented 4476 colorectal cancer cases, including 1232 cases with available bacterial data. High DNA amounts of Bacteroides fragilis and ETBF were positively associated with BRAF mutation (p ≤ 0.0003), CIMP-high (p ≤ 0.0002), and MSI-high (p < 0.0001 and p = 0.01, respectively). Multivariable-adjusted odds ratios (with 95% confidence interval) for high Bacteroides fragilis were 1.40 (1.06–1.85) for CIMP-high and 2.14 (1.65–2.77) for MSI-high, but 1.02 (0.78–1.35) for BRAF mutation. Multivariable-adjusted odds ratios for high ETBF were 2.00 (1.16–3.45) for CIMP-high and 2.86 (1.64–5.00) for BRAF mutation, but 1.09 (0.67–1.76) for MSI-high. Neither Bacteroides fragilis nor ETBF was associated with colorectal cancer-specific or overall survival. The tissue abundance of Bacteroides fragilis is associated with CIMP-high and MSI-high, whereas ETBF abundance is associated with CIMP-high and BRAF mutation in colorectal carcinoma. Lobsters and crayfish in Australasia can develop a condition known as Tail Fan Necrosis (TFN syndrome). Many attempts have been made to find a primary pathogen or link the syndrome to commercial activities, but a solution remains elusive. TFN syndrome is a 'wicked problem', a problem difficult or impossible to solve because of incomplete and contradictory information forming a matrix of potential outcomes with no simple solution. Reviewing the literature shows TFN syndrome is sometimes reported to develop in association with sterile blisters on the telson and uropods which may rupture permitting invasion by environmental fungal and/or bacterial flora. Whether blisters form prior to, or because of, infection is unknown. TFN syndrome sometimes develops in captivity, sometimes requires a previous insult to the telson and uropods, and prevalence is patchy in the wild. The literature shows the cause of blisters associated with TFN syndrome remains an enigma, for which we suggest several possible initiating factors. We strongly urge that researchers not 'jump to conclusions' as to the aetiology of TFN syndrome. Background: Fleas are ectoparasitic insects with holometabolous development. It has a hematophagous habit with mouthparts adapted to sting and suck its hosts. There are about 3000 species in the world, ∼61 in Brazil, and 19 in Rio Grande do Sul state. The objective of the research is to catalog the diversity of fleas recorded in the state, their respective hosts, and endosymbionts. Materials and Methods: To this end, a search was carried out in the scientific literature, from articles, books, to abstracts submitted to congresses. Results: The 19 species of fleas occurring in Rio Grande do Sul are divided into 7 families and 10 genera. These ectoparasites, in addition to being found in the environment, were associated with 10 different families of hosts in Rio Grande do Sul, and on the endosymbiont, agents found associated with fleas, there were 7 different species. The main agents researched in the state are Rickettsia spp. and Bartonella spp. The relationships between parasites, hosts, environment, and etiological agents present different scenarios, whether anthropized or conserved, but unknown. Sometimes, this overlap, a factor that aggravates the possibility of spillovers, either from cosmopolitan fleas in these conserved areas, or from their endosymbionts. Conclusion: Thus, it is important to characterize the environment so that the complexities of each location are known for the adoption of environmental and public health policies in each case. The polyphagous shot hole borer beetle (PSHB, Euwallacea fornicatus) is a pest of global significance. PSHB is an ambrosia beetle which, together with its mutualistic fungi (including Fusarium euwallaceae), can cause the death of more than 100 tree species in invaded ranges. Management of PSHB mostly relies on the removal of infested plant material. Chemical control options have been investigated only in the USA and Israel and only on a few tree species. This study evaluated four chemical treatments for the therapeutic control of PSHB on American sweetgum (Liquidambar styraciflua) in South Africa (1) bifenthrin + surfactant (alcohol ethoxylate), (2) cypermethrin + surfactant (vitamin E) + salicylic acid, (3) emamectin benzoate, and (4) propiconazole. Trees were inoculated with F. euwallaceae and mature PSHB females using a novel technique to document fungal lesion development and PSHB colony establishment success. The bifenthrin and cypermethrin treatments reduced additional PSHB colonisation attempts on treated trees by ca. 40%, while the other treatments had no effect. Colony establishment success was reduced in all treatments by between 20 and 40%. Fungal growth was inhibited only after the application of propiconazole by ca. 36%. Gallery length and the number of PSHB individuals in successful colonies were unaffected by any of the chemical treatments. These results indicate that chemical control of PSHB is only partially effective. In this study, Neofusicoccum mediterraneum, a highly aggressive plant pathogen with a broad host range, including Eucalyptus spp., was investigated to determine its optimum medium and temperature for mycelial growth and chlamydospore production. The results revealed differences in the growth behaviour of N. mediterraneum under different in vitro conditions. Potato Sucrose Agar (PSA) was determined to be the most favourable environment for mycelial growth, achieving the maximum growth rate (GR) of 100% after 3 days of incubation at 25 °C. This was closely followed by cultures on Malt Extract Agar (MEA), exhibiting a GR of 100% after 4 days of incubation at 25 °C, while both substrates displayed optimal growth within the temperature range of 25 to 30 °C. The General Linear Model (GLM) unveiled significant differences in the growth rate across the different media (P = 0.000) and temperatures (P = 0.000). The interaction effect (media*temperature) retained its high significance throughout all incubation days (P < 0.0001). The binomial distribution and multinomial logistic regression analysis revealed that colony texture was significantly influenced by both the media (P = 0.022) and the temperature (P = 0.008). Additionally, temperature affected significantly the surface colour (P = 0.015) and reverse colour (P = 0.023) of the colony. Production of chlamydospores was observed only on PSA at 20 °C, 25 °C, 30 °C and 35 °C and on PDA at 25 °C and 30 °C. Both the medium and temperature had notable influences on the pathogen's growth, highlighting their importance in understanding its biology and behaviour. A zoonosis is an infectious disease that can be transmitted to humans from animals, potentially leading to an epidemic or pandemic. Increasing research into the environmental drivers of zoonoses in a so-called 'Pandemicene' has focused on changes to land use, land cover and climate. But what about the associated changes to earth surface processes, landforms and landscapes that may impact on zoonoses and other infectious diseases with environmental reservoirs (e.g. sapronoses)? This is a crucial question, especially in the context of the Anthropocene, a proposed time interval during which humans have directly and indirectly impacted geomorphological systems and associated abiotic-biotic interactions at local through global scales. Biogeomorphology (and especially zoogeomorphology) potentially has many insights to contribute but is largely divorced from a rapidly expanding body of infectious disease research. By outlining geomorphological principles and providing examples from across dryland, arctic, tropical and temperate environments, we examine the extent to which water, sediment, soil and other organic matter that is directly or indirectly influenced by non-human animal activity may critically influence the emergence and transmission of infectious diseases. In an era of rapid environmental and socioeconomic change, we contend that geomorphological concepts, data and techniques can play a key role in improved characterisation of dynamic 'pathogenic landscapes'. We identify several challenges and opportunities that may provide particular foci for enhanced geomorphological contributions to this growing threat to societal resilience, including: (1) working in transdisciplinary 'One Health' contexts to test hypotheses about geomorphological controls on pathogen emergence and spread; (2) collaborating with archaeologists to examine past disease dynamics and identify potential future pathogen sources; (3) enhancing existing global maps of infection risk/disease emergence at local to regional scales; and (4) contributing to the robust evidence base needed for policies to monitor and manage infectious disease risk. Aggregatibacter actinomycetemcomitans cytolethal distending toxin (Cdt) is capable of intoxicating lymphocytes macrophages, mast cells and epithelial cells. Following Cdt binding to cholesterol, in the region of membrane lipid rafts, the CdtB and CdtC subunits are internalized and traffic to intracellular compartments. These events are dependent upon, cellugyrin, a critical component of synaptic like microvesicles (SLMVCg+). Target cells, such as Jurkat cells, rendered unable to express cellugyrin are resistant to Cdt-induced toxicity. Similar to Cdt, SARS-CoV-2 entry into host cells is initiated by binding to cell surface receptors, ACE-2, also associated with cholesterol-rich lipid rafts; this association leads to fusion and/or endocytosis of viral and host cell membranes and intracellular trafficking. The similarity in internalization pathways for both Cdt and SARS-CoV-2 led us to consider the possibility that cellugyrin was a critical component in both processes. Cellugyrin deficient Calu-3 cells (Calu-3Cg-) were prepared using Lentiviral particles containing shRNA; these cells were resistant to infection by VSV/SARS-CoV-2-spike pseudotype virus and partially resistant to VSV/VSV-G pseudotype virus. Synthetic peptides representing various regions of the cellugyrin protein were prepared and assessed for their ability to bind to Cdt subunits using surface plasmon resonance. Cdt was capable of binding to a region designated the middle outer loop (MOL) which corresponds to a region extending into the cytoplasmic surface of the SLMVCg+. SARS-CoV-2 spike proteins were assessed for their ability to bind to cellugyrin peptides; SARS-CoV-2 full length spike protein preferentially binds to a region within the SLMVCg+ lumen, designated intraluminal loop 1A. SARS-CoV-2-spike protein domain S1, which contains the receptor binding domains, binds to cellugyrin N-terminus which extends out from the cytoplasmic surface of SLMV. Binding specificity was further analyzed using cellugyrin scrambled peptide mutants. An in situ integrated system, consisting of ecological floating islands (EFI), ecological riverbeds (ER), and ecological filter dams (EFD), was built in a ditch only receiving the effluent of sewage plant; the effect of in situ technologies on the distribution of aquatic pathogen was investigated. The results showed the aquatic pathogen decreased along the ditch. Specifically, the relative abundance of Legionella, Aeromonas, and Acinetobacter decreased from 0.032, 0.035, and 0.26 to 0.026%, 0.012%, and 0.08%, respectively. Sedimentation, filtration, and sorption (provided by plant roots and biofilms on substrates) were principal processes for the removal. The nitrogen removal bacteria to prevent the potential risk of eutrophication were also evaluated. The EFI and ER were the dominant sites for Nitrosomonas (34.96%, 32.84%) and Nitrospira (35.74%, 54.73%) enrichment, while EFI and EFD facilitated the enrichment of denitrification bacteria. Notably, the relative abundance of endogenous denitrifiers (DNB-en) (including Dechloromonas at 9.72%, Thermomonas at 0.58%, and Saccharibacteria at 2.55%) exceeded those of exogenous denitrifiers (DNB-ex) (Thauera at 0.20%, Staphylococcus at 0.005%, and Rhodobacter at 0.27%). This study demonstrated that the in situ integrated system was effective in reducing the abundance of pathogens in the drainage channel, and the deficiency of DNB-ex and carbon sources made nitrate removal difficult. Secondary metabolites produced by primary producers have a wide range of functions as well as indirect effects outside the scope of their direct target. Research suggests that protease inhibitors produced by cyanobacteria influence grazing by herbivores and may also protect against parasites of cyanobacteria. In this study, we asked whether those same protease inhibitors produced by cyanobacteria could also influence the interactions of herbivores with their parasites. We used the Daphnia‐Metschnikowia zooplankton host‐fungal parasite system to address this question because it is well documented that cyanobacteria protease inhibitors suppress trypsin and chymotrypsin in the gut of Daphnia, and because it is known that Metschnikowia infects via the gut. We tested the hypothesis that Daphnia gut proteases are necessary for Metschnikowia spores to be released from their asci. We then also tested whether diets that decrease trypsin and chymotrypsin activity in the guts of Daphnia lead to lower levels of infection. Our results show that chymotrypsin promotes the release of the fungal spores from their asci. Moreover, a diet that strongly inhibited chymotrypsin activity in Daphnia decreased infection levels, particularly in the most susceptible Daphnia clones. Our results support the growing literature that cyanobacterial diets can be beneficial to zooplankton hosts when challenged by parasites and uncover a mechanism that contributes to the protective effect of cyanobacterial diets. Specifically, we demonstrate that host chymotrypsin enzymes promote the dehiscence of Metschnikowia spores; when cyanobacteria inhibit the activity of chymotrypsin in hosts, this most likely traps the spore inside the ascus, preventing the parasite from puncturing the gut and beginning the infection process. The mitogen-activated protein kinase (MAPK/MPK) cascade is an important intercellular signaling module that regulates plant growth, development, reproduction, and responses to biotic and abiotic stresses. A MAPK cascade usually consists of a MAPK kinase kinase (MAPKKK/MEKK), a MAPK kinase (MAPKK/MKK/MEK), and a MAPK. The well-characterized MAPK cascades in plant immunity to date are the MEKK1-MKK1/2-MPK4 cascade and the MAPKKK3/4/5-MKK4/5-MPK3/6 cascade. Recently, major breakthroughs have been made in understanding the molecular mechanisms associated with the regulation of immune signaling by both of these MAPK cascades. In this review, we highlight the most recent advances in understanding the role of both MAPK cascades in activating plant defense and in suppressing or fine-tuning immune signaling. We also discuss the molecular mechanisms by which plants stabilize and maintain the activation of MAPK cascades during immune signaling. Based on this review, we reveal the complexity and importance of the MEKK1-MKK1/2-MPK4 cascade and the MAPKKK3/4/5-MKK4/5-MPK3/6 cascade, which are tightly controlled by their interacting partners or substrates, in plant immunity. Objective To study the pathogen composition and antimicrobial resistance characteristics in neonates with sepsis in the Hebei area, providing a basis for optimizing the management strategies for neonatal sepsis in the region. Methods The medical data of neonates with sepsis confirmed by blood culture from 23 hospitals in Hebei Province were collected prospectively from November 1, 2021 to December 31, 2022. The pathogen composition and antimicrobial resistance characteristics were analyzed in early-onset sepsis (EOS), community-acquired late-onset sepsis (CALOS), and hospital-acquired late-onset sepsis (HALOS) cases. Results A total of 289 neonates were included, accounting for 292 infection episodes, with 117 (40.1%) EOS, 95 (32.5%) CALOS, and 80 (27.4%) HALOS cases. A total of 294 pathogens were isolated, with 118 (40.1%) from the EOS group, where Escherichia coli (39 strains, 33.0%) and coagulase-negative staphylococci (13 strains, 11.0%) were the most common. In the CALOS group, 95 pathogens were isolated (32.3%), with Escherichia coli (35 strains, 36.8%) and group B Streptococcus (14 strains, 14.7%) being predominant. The HALOS group yielded 81 isolates (27.6%), with Klebsiella pneumoniae (24 strains, 29.6%) and Escherichia coli (21 strains, 25.9%) as the most frequent. Coagulase-negative staphylococci showed over 80% resistance to penicillin antibiotics. Escherichia coli and Klebsiella pneumoniae showed a resistance rate of 2.2% to 75.0% to common β-lactam antibiotics. The resistance rates of predominant Gram-negative bacteria in the HALOS group to common β-lactam antibiotics were higher than those in the CALOS group. Conclusions In the Hebei region, Escherichia coli is the most common pathogen in both EOS and LOS. There are differences in the composition and antimicrobial resistance of pathogens among different types of neonatal sepsis. The COVID-19 pandemic was a challenge for the whole world, and it had major secondary effects on humans and environmental health. The viral infection induced, in many situations, secondary bacterial infections, especially enteric infections, by destabilizing the balance of the gastrointestinal microbiota. The large-scale use of antibiotics and biocides for both curative and preventive purposes has resulted in an increase in bacterial resistance, and at the same time, the possibility of pathogenic microorganism multiplication and their dissemination to natural environments. Wastewater is the main vector of fecal microorganisms that favors their dissemination into natural aquatic ecosystems. The present paper aims to analyze the effect of the COVID-19 pandemic on the microbiological quality of wastewater from sewage treatment plants in Romania and its impact on receiving rivers. In order to highlight different and important areas in Romania, three cities from the east, center and west were selected for a microbiological evaluation of their WWTP influents and effluents from the pre-COVID-19 period and during the COVID-19 pandemic peak period, when the COVID-19 pandemic had a direct impact on WWTP microbial compositions. Our study shows that a higher level of contamination with fecal bacteria is linked to a higher COVID-19 incidence. Vaccines have significantly reduced the impact of numerous deadly viral infections. However, there is an increasing need to expedite vaccine development in light of the recurrent pandemics and epidemics. Also, identifying vaccines against certain viruses is challenging due to various factors, notably the inability to culture certain viruses in cell cultures and the wide-ranging diversity of MHC profiles in humans. Fortunately, reverse vaccinology (RV) efficiently overcomes these limitations and has simplified the identification of epitopes from antigenic proteins across the entire proteome, streamlining the vaccine development process. Furthermore, it enables the creation of multiepitope vaccines that can effectively account for the variations in MHC profiles within the human population. The RV approach offers numerous advantages in developing precise and effective vaccines against viral pathogens, including extensive proteome coverage, accurate epitope identification, cross-protection capabilities, and MHC compatibility. With the introduction of RV, there is a growing emphasis among researchers on creating multiepitope-based vaccines aiming to stimulate the host's immune responses against multiple serotypes, as opposed to single-component monovalent alternatives. Regardless of how promising the RV-based vaccine candidates may appear, they must undergo experimental validation to probe their protection efficacy for real-world applications. The time, effort, and resources allocated to the laborious epitope identification process can now be redirected toward validating vaccine candidates identified through the RV approach. However, to overcome failures in the RV-based approach, efforts must be made to incorporate immunological principles and consider targeting the epitope regions involved in disease pathogenesis, immune responses, and neutralizing antibody maturation. Integrating multi-omics and incorporating artificial intelligence and machine learning-based tools and techniques in RV would increase the chances of developing an effective vaccine. This review thoroughly explains the RV approach, ideal RV-based vaccine construct components, RV-based vaccines designed to combat viral pathogens, its challenges, and future perspectives. Multiple stress resistant variants of Listeria monocytogenes with mutations in rpsU encoding ribosomal protein RpsU have previously been isolated after a single exposure to acid stress. These variants, including L. monocytogenes LO28 variant V14 with a complete deletion of the rpsU gene, showed upregulation of the general stress sigma factor Sigma B-mediated stress resistance genes and had a lower maximum specific growth rate than the LO28 WT, signifying a trade-off between stress resistance and fitness. In the current work V14 has been subjected to an experimental evolution regime, selecting for higher fitness in two parallel evolving cultures. This resulted in two evolved variants with WTlike fitness: 14EV1 and 14EV2. Comparative analysis of growth performance, acid and heat stress resistance, in combination with proteomics and RNAsequencing, indicated that in both lines reversion to WT-like fitness also resulted in WT-like stress sensitivity, due to lack of Sigma B-activated stress defense. Notably, genotyping of 14EV1 and 14EV2 provided evidence for unique pointmutations in the ribosomal rpsB gene causing amino acid substitutions at the same position in RpsB, resulting in RpsB22Arg-His and RpsB22Arg-Ser, respectively. Combined with data obtained with constructed RpsB22Arg-His and RpsB22Arg-Ser mutants in the V14 background, we provide evidence that loss of function of RpsU resulting in the multiple stress resistant and reduced fitness phenotype, can be reversed by single point mutations in rpsB leading to arginine substitutions in RpsB at position 22 into histidine or serine, resulting in a WT-like high fitness and low stress resistance phenotype. Baby chicks administered a fecal transplant from adult chickens are resistant to Salmonella colonization by competitive exclusion. A two-pronged approach was used to investigate the mechanism of this process. First, Salmonella response to an exclusive (Salmonella competitive exclusion product, Aviguard®) or permissive microbial community (chicken cecal contents from colonized birds containing 7.85 Log10 Salmonella genomes/gram) was assessed ex vivo using a S. typhimurium reporter strain with fluorescent YFP and CFP gene fusions to rrn and hilA operon, respectively. Second, cecal transcriptome analysis was used to assess the cecal communities' response to Salmonella in chickens with low (≤5.85 Log10 genomes/g) or high (≥6.00 Log10 genomes/g) Salmonella colonization. The ex vivo experiment revealed a reduction in Salmonella growth and hilA expression following co-culture with the exclusive community. The exclusive community also repressed Salmonella's SPI-1 virulence genes and LPS modification, while the anti-virulence/inflammatory gene avrA was upregulated. Salmonella transcriptome analysis revealed significant metabolic disparities in Salmonella grown with the two different communities. Propanediol utilization and vitamin B12 synthesis were central to Salmonella metabolism co-cultured with either community, and mutations in propanediol and vitamin B12 metabolism altered Salmonella growth in the exclusive community. There were significant differences in the cecal community's stress response to Salmonella colonization. Cecal community transcripts indicated that antimicrobials were central to the type of stress response detected in the low Salmonella abundance community, suggesting antagonism involved in Salmonella exclusion. Rice is a major cereal crop which ensure food security to more than half of the global population. Several biotic factors impact rice grain quality and its final production. White stripe disease, caused by pathogen Stenotrophomonas maltophilia is considered among the major limiting factor for reducing rice yields and quality. Present study was performed to understand the white stripe disease, which has been frequently misdiagnosed as bacterial leaf blight (BLB) due to similar symptoms. A survey was carried out based on accessibility and farmer participation to monitor incidence and sample collection. The survey was conducted in districts Faisalabad, Gujranwala, Sialkot, Sheikhupura, and Hafizabad, these districts were selected for their importance for rice cultivation in Pakistan. The total sample size was around 500 leaves distributed evenly throughout each study area. The results of study indicated presence of new pathogen of rice. These isolates were biochemically identified and confirmed by gram staining negative, 3% KOH positive, 5% salt tolerance positive, oxidase test negative, catalase test positive, starch hydrolysis test negative, nitrate reductase test positive, indole test negative, lactose test positive, maltose test positive, methyl red test negative, Vogesprokauer test negative and urea hydrolysis test negative. The pathogenicity test was confirmed that pathogen and the Sialkot isolate were the most aggressive isolate among the five isolates collected form the studied areas. The molecular characterization was accomplished by PCR and sequencing. The results of the phylogenetic study indicate that this pathogen belongs to a distinct group, as it is distantly related to Xanthomonas oryzae pv. oryzae (Xoo). Pyricularia oryzae (syn. Magnaporthe oryzae), is a filamentous ascomycete that causes a major disease called blast on cereal crops, as well as on a wide variety of wild and cultivated grasses. Blast diseases have a tremendous impact worldwide particularly on rice and on wheat, where the disease emerged in South America in the 1980s, before spreading to Asia and Africa. Its economic importance, coupled with its amenability to molecular and genetic manipulation, have inspired extensive research efforts aiming at understanding its biology and evolution. In the past 40 years, this plant‐pathogenic fungus has emerged as a major model in molecular plant–microbe interactions. In this review, we focus on the clarification of the taxonomy and genetic structure of the species and its host range determinants. We also discuss recent molecular studies deciphering its lifecycle. Taxonomy: Kingdom: Fungi, phylum: Ascomycota, sub‐phylum: Pezizomycotina, class: Sordariomycetes, order: Magnaporthales, family: Pyriculariaceae, genus: Pyricularia. Host range: P. oryzae has the ability to infect a wide range of Poaceae. It is structured into different host‐specialized lineages that are each associated with a few host plant genera. The fungus is best known to cause tremendous damage to rice crops, but it can also attack other economically important crops such as wheat, maize, barley, and finger millet. Disease symptoms: P. oryzae can cause necrotic lesions or bleaching on all aerial parts of its host plants, including leaf blades, sheaths, and inflorescences (panicles, spikes, and seeds). Perilla frutescens (L.) Britton, an annual plant in the mint family, is considered invasive in the United States where it excludes native vegetation in natural areas and can cause respiratory illness in livestock. An effective biological control agent would improve perilla management, but none have been developed or proposed. This study investigated the biocontrol potential of a fungal pathogen, Colletotrichum shisoi, recently found infecting perilla in North America. The growth rates and morphology of seven Colletotrichum shisoi isolates were recorded. A virulence assay was performed on two perilla accessions, and the number and size of foliar lesions produced were used to select a single isolate, Colletotrichum shisoi isolate 21-072, exhibiting consistently high virulence for further evaluation of biocontrol efficacy and host specificity. Treatment with 0.5 mL of 1 × 107 spores/mL H2O caused a 52% reduction in shoot height and 84% reduction in root length of invasive perilla seedlings compared to a control treatment (P < 0.001). Seedlings treated with 1 × 108 spores/mL had a 100% mortality rate. Formulation of inoculum in a 1:1 water:oil emulsion reduced dew period requirements for causing >50% plant mortality from 48 to 24 h, but oil emulsions exhibited a mild phytotoxic effect on seedlings. Four additional perilla accessions were susceptible to C. shisoi, and none of 34 other species included in host range experiments developed disease symptoms following inoculation. One-Health is an umbrella term that integrates the health of the environment, humans and non-human animals. This approach is applied here to elucidate the impact of non-native invasive species on aquaculture and food security. Despite inherent biases against these species, a better understanding of their characteristics allows for the identification of those of greatest concern, minimizing the risk of food shortages and infectious diseases. This review summarises the positive and negative impacts of non-native species, delineating the specific areas they may impact. Additionally, this review gives an insight to the expertise and stakeholders that would need to be included if a "One-Health" approach were to be implemented by policymakers to better control non-native species. Detailed examples illustrate the consequences of non-native species on trophic dynamics, ecosystem health, water chemistry, and human health, emphasizing the importance of managing them within a multidimensional framework. The "One-Health" approach is explained, and suggestions are made on how certain non-native species could be used to contribute to food security in low- and middle-income countries. Clubroot is one of the important diseases affecting members of the Cruciferae family. This disease is caused by soil-borne pathogen, called Plasmodiophora brassicae. The pathogen produces motile spores called zoospores. This pathogen results in a drop of cruciferous plant productivity. The objectives of this research were to identify specific Trichoderma species in three districts in Sumatra Utara and to evaluate the potency of T. harzianum (both local species from Berastagi and species developed by Indonesian Vegetable Research Institute (IVEGRI)) to control the disease and then comparing them with the application of synthetic fungicide. The research was conducted in a greenhouse of Research Installation and Application of Agricultural Technology (IP2TP), Berastagi, North Sumatera from April-September 2018, using Completely Randomized Design (CRD) with eight treatments: C1 (control-without Trichoderma), C2 (T. harzianum IVEGRI obtained from corn substrate 2 g/polybag), C3 (T. harzianum IVEGRI obtained from rice substrate 2 g/polybag), C4 (T. harzianum IVEGRI obtained from corn substrate 4 g/polybag, C5 (T. harzianum IVEGRI obtained from rice substrate 4 g/polybag), C6 (local T. harzianum obtained from corn substrate 2 g/polybag), C7 (local T. harzianum obtained from corn substrate 4 g/polybag) and C8 (synthetic fungicide Nebijin). Each treatment contained 10 polybags of plants. These treatments were replicated four times. The results exhibited there were 3 Trichoderma species found in Berastagi: T. harzianum, T. viride and T. koningii. Also, 4 g of local T. harzianum (corn substrate) has better performance (0% disease incidence and 0% disease severity) compared to other treatments. [ABSTRACT FROM AUTHOR]

Alone, pathogen-associated molecular patterns (PAMPs) or danger-associated molecular patterns (DAMPs) elicit the activation of single-sensor inflammasomes, activating caspases. Live pathogens harbor multiple PAMPs/DAMPs that may prompt recognition by multiple-sensor inflammasomes. The synchronized detection and activation in response to diverse PAMPs and DAMPs are regulated by collaboration among multiple inflammasome sensors, instigating cell death pathways and bolstering the host's immune response. Inflammasomes have an indispensable role in integrating various cell death pathways, showcasing their adaptability in modulating immune responses and influencing outcomes following pathogen infection. Elucidating the nuanced mechanisms through which inflammasome sensors can orchestrate immune responses and programmed cell death pathways presents promising avenues for developing targeted therapeutics against infectious diseases and inflammatory disorders. Insights into the orchestration of multiple sensor inflammasomes highlight the intricate mechanisms of mammalian immunity. Understanding how diverse pathogens activate multiple sensors simultaneously can enhance our comprehension of the innate immune system's ability to mount tailored responses against various threats. This advanced understanding of immune system dynamics may provide avenues for developing targeted therapeutic interventions for infectious and autoimmune diseases, as well as cancers. Pathogens elicit complex mammalian immune responses by activating multiple sensors within inflammasomes, which recognize diverse pathogen-associated molecular patterns (PAMPs) and damage-associated molecular patterns (DAMPs). This simultaneous activation induces the formation of protein complexes referred to as multiple inflammasomes, that orchestrate a spectrum of programmed cell death pathways, including pyroptosis, apoptosis, and necroptosis. This concept is crucial for comprehending the complexity of the innate immune system's response to diverse pathogens and its implications for various diseases. Novel contributions here include emphasizing simultaneous sensor activation by pathogens, proposing the existence of multiple inflammasome complexes, and advocating for further exploration of their structural basis. In this study, the antibacterial effects of peels obtained from Zivzik pomegranate against Staphylococcus aureus, Escherichia coli, Bacillus cereus, Enterococcus faecalis, and Pseudomonas aeruginosa were investigated. The pomegranate peel was ground into powder form and their control (0%), 2%, 4% and 8% solutions were prepared, and firstly pH, Oxidation/Reduction (O/R) and electrical conductivity (EC) values were measured. Then, these bacteria were inoculated with selected standard antibiotics according to the disc diffusion method and with pomegranate peel powder according to the well/hole method and the zone diameters formed because of incubation were determined. In addition, the counts of bacteria formed after incubation was determined by inoculating these bacteria and each peel solution on special media. In analyses, the pH value of the 0% concentration solution was determined as 5.96, O/R value 83.17 mV and EC value 0.13 µS cm-1. However, the pH decreased to 3.7 in the 8% concentration solution, O/R and EC also increased to195.67 mV and 0.42 µS cm-1, respectively. These changes increased the antibacterial effect of solutions prepared from pomegranate peel powder. Pomegranate peel solutions showed similar effects to most of the standard antibiotics used. Also, by influencing the numbers of bacteria used, especially the 8% solution provided about 1.5-2 log reduction compared to the control. Consequently, while it was determined that 4% and 8% solutions of the powders obtained from this pomegranate peel could be used against these bacteria, it was understood that it would be beneficial for the food industry to conduct research against different microorganisms. Over 80% of agricultural crops rely on insect pollination. Thus, pollinator biodiversity is critical for augmented plant yield and quality.Blow flies (Diptera: Calliphoridae) are acknowledged pollinators of several crops. However, there are many open questions regarding their importance in pollination ecology. For instance, there has not been a comprehensive assessment of pollen use by blow flies, and thus, it is unknown whether individuals visit few or many plant types per trip.At least one blow fly species (Lucilia sericata) can digest pollen and use the protein therein as an oogenic resource. However, it is unknown whether this is a commonly used strategy for protein acquisition, given that they are more commonly associated with faeces, carrion and myiasis.We surveyed the pollen found in blow fly guts to determine whether blow flies (a) consume pollen, and if so, which types of pollen; (b) exhibit species‐specific pollen use and (c) demonstrate sex‐specific pollen use.We collected blow flies from natural habitats around Central Texas from 2016 to 2019 and dissected their guts to look for the presence of pollen. We found widespread use of pollen from a variety of plants by six blow fly species and discovered a bias in the amount of pollen collected by female blow flies compared with males in some, but not all fly species.Pollen represents a relatively underappreciated aspect of blow fly biology, with the potential for several underappreciated ecological impacts ranging from disease ecology to plant diversity. The order Procellariiformes includes several species of seabirds that perform long-distance migrations crossing all the oceans. These movements may contribute to the dispersal and exchange of hemoparasites, such as haemosporidians. There is a lack of studies regarding the order Haemosporida in Procellariiformes, and, to date, only the genus Plasmodium has been reported. This survey investigated the occurrence of the three genera of haemosporidians, Plasmodium, Haemoproteus, and Leucocytozoon, in samples collected between 2013 and 2022 from 95 individuals of 14 species of Procellariiformes from southern Brazil, including live animals in rehabilitation centers, individuals caught as incidental bycatch, and carcasses found along the coast. A total of 171 samples of blood and fragments of liver and spleen were analyzed, with extracted DNA being subjected to a nested PCR followed by phylogeny analysis. All animals were negative for Plasmodium spp. and Leucocytozoon spp., but one Black-browed Albatross (Thalassarche melanophris) and one Manx Shearwater (Puffinus puffinus) specimen were positive for Haemoproteus spp. The sequences obtained from positive seabirds did not show 100% similarity with other known lineages available in the MalAvi database and thus were probably novel lineages. However, one sequence clustered together with Haemoproteus noctuae, a parasite from Strigiformes, while the other was grouped with Haemoproteus columbae, which is classically related to Columbiformes. These results suggest that both positive animals may have become infected when beached or in rehabilitation centers by a spillover of vectors from local birds. This highlights the importance of surveillance of the health of Procellariiformes regarding the possibility of dissemination of new pathogens in different bird populations. [ABSTRACT FROM AUTHOR]

Staka is a traditional Greek sour cream made mostly from spontaneously fermented sheep milk or a mixture of sheep and goat milk. At the industrial scale, cream separators and starter cultures may also be used. Staka is sometimes cooked with flour to absorb most of the fat. In this study, we employed culture-based techniques, amplicon sequencing, and shotgun metagenomics to analyze the Staka microbiome for the first time. The samples were dominated by Lactococcus or Leuconostoc spp. Most other bacteria were lactic acid bacteria (LAB) from the Streptococcus and Enterococcus genera or Gram-negative bacteria from the Buttiauxella, Pseudomonas, Enterobacter, Escherichia-Shigella, and Hafnia genera. Debaryomyces, Kluyveromyces, or Alternaria were the most prevalent genera in the samples, followed by other yeasts and molds like Saccharomyces, Penicillium, Aspergillus, Stemphylium, Coniospotium, or Cladosporium spp. Shotgun metagenomics allowed the species-level identification of Lactococcus lactis, Lactococcus raffinolactis, Streptococcus thermophilus, Streptococcus gallolyticus, Escherichia coli, Hafnia alvei, Streptococcus parauberis, and Enterococcus durans. Binning of assembled shotgun reads followed by recruitment plot analysis of single reads could determine near-complete metagenome assembled genomes (MAGs). Culture-dependent and culture-independent analyses were in overall agreement with some distinct differences. For example, lactococci could not be isolated, presumably because they had entered a viable but not culturable (VBNC) state or because they were dead. Finally, several LAB, Hafnia paralvei, and Pseudomonas spp. isolates exhibited antimicrobial activities against oral or other pathogenic streptococci, and certain spoilage and pathogenic bacteria establishing their potential role in food bio-protection or new biomedical applications. Transcriptomic analyses across large scales of evolutionary distance have great potential to shed light on regulatory evolution but are complicated by difficulties in establishing orthology and limited availability of accessible software. We introduce here a method and a graphical user interface wrapper, called Annotator-RNAtor, for performing interspecies transcriptomic analysis and studying intragenus evolution. The pipeline uses third-party software to infer homologous genes in various species and highlight differences in the expression of the core-genes. To illustrate the methodology and demonstrate its usefulness, we focus on the emergence of the highly virulent Leptospira subclade known as P1+, which includes the causative agents of leptospirosis. Here, we expand on the genomic study through the comparison of transcriptomes between species from P1+ and their related P1- counterparts (low-virulent pathogens). In doing so, we shed light on differentially expressed pathways and focused on describing a specific example of adaptation based on a differential expression of PerRA-controlled genes. We showed that P1+ species exhibit higher expression of the katE gene, a well-known virulence determinant in pathogenic Leptospira species correlated with greater tolerance to peroxide. Switching PerRA alleles between P1+ and P1- species demonstrated that the lower repression of katE and greater tolerance to peroxide in P1+ species was solely controlled by PerRA and partly caused by a PerRA amino-acid permutation. Overall, these results demonstrate the strategic fit of the methodology and its ability to decipher adaptive transcriptomic changes, not observable by comparative genome analysis, that may have been implicated in the emergence of these pathogens. The manifestation of physiological stresses can lead to a variety of unfavorable outcomes such as oxidative stress, imbalances in nutrient homeostasis, and in extreme scenarios, plant mortality. Rose is a popular ornamental plant that exhibits vulnerability to crown gall, a pathological condition primarily attributed to the presence of Rhizobium radiobacter (also known as Agrobacterium tumefaciens). In the present study, impacts of R. radiobacter inoculation on the graft strength, nutrient levels, and oxidative stress of two distinct rose cultivars ('Angelina' and 'Pearl') in relation to their sensitivity to R. radiobacter were investigated. Rhizobium radiobacter was inoculated at the grafting site, and the grafted plants were grown for a period of 90 days. The findings indicated that the presence of bacteria in the samples led to more robust graft unions, owing to the development of enlarged calluses (crown galls) that functioned as a potent inherent adhesive. The inoculation of R. radiobacter reduced shoot biomass, while promoted root biomass. The nutrient levels in the infected samples were higher than their levels in the non-infected samples. Consequently, the non-infected samples exhibited higher relative water content (RWC) than the RWC of infected samples. The infection caused by R. radiobacter-induced oxidative stress, as evidenced by the elevated levels of malondialdehyde and electrolyte leakage. Following to bacterial infection, decline in catalase (CAT) and increase in guaiacol peroxidase (GPX) activity were detected. The inoculation of R. radiobacter into the host plant altered the sink/source balance, leading to more accumulation of nutrients in the leaves. Occurrence of oxidative stress, reduction in leaf area, and shoot biomass in the inoculated 'Angelina' cultivar were indicative of higher susceptibility of this cultivar to R. radiobacter infection, while, 'Pearl' cultivar exhibited higher resistance to R. radiobacter infection, as evidenced by its higher: soluble carbohydrates, soluble protein, and proline contents, and GPX activity compared to 'Angelina' cultivar.
