105. Rapid Differentiation of Seven Species of <italic>Anguilla</italic> Using PNA Clamping-based Asymmetric PCR with Fluorescence Melting Curve Analysis.

Abstract

Differentiation of the genus Anguilla is important for phylogenetic studies, ecological monitoring, and food forensics. Although various methods have been developed to identify these species, current methods are time-consuming and limit the number of species that can be detected. The aim of this study was to develop a rapid and convenient method to identify the genus Anguilla. To differentiate among seven commercially important species of Anguilla (A. japonica, A. anguilla, A. rostrata, A. celebensis, A. pacifica, A. marmorata, and A. bicolor pacifica), fluorescence melting curve analysis (FMCA) with peptide nucleic acid (PNA) probes were used. One primer set was designed to amplify the mitochondrial cytochrome b gene, and seven PNA probes were designed to identify each species from the cyt b gene. The PNA probes were labeled with four different fluorescent reporter dyes (FAM, HEX, Texas Red, Cy5) for multiplex analysis using two tubes. Based on FMCA, each PNA probe showed the highest Tm value when it was 100% complementary to the target sequence. In conclusion, the PNA-based FMCA method is suitable for differentiating among seven species of Anguilla and should be applicable in various fields, such as global marketing and scientific research. For such biomedical applications, SPIONs must have multifunctional characteristics, including optimized size and modified surface. However, the biofunctionality and biocompatibility of SPIONs with various surface functional groups of different sizes have yet to be elucidated clearly. Therefore, it is important to carefully monitor the cytotoxicity and genotoxicity of SPIONs that are surfaced-modified with various functional groups of different sizes. In this study, we evaluated SPIONs with diameters of approximately 10 nm and 100~150 nm, containing different surface functional groups. SPIONs were covered with –O-groups, so-called bare SPIONs. Abstract

The nicotinic acetylcholine receptor (nAChR) is a member of the ligand-gated ion channel (LGIC) family and is composed of five subunits arranged around a central pore. Expressed sequence tag screening and traditional cloning strategies revealed five full-length cDNAs encoding nAChR subunit homologs (Pajα3, Pajα10, Pajα11, Pajα12, and Pajβ1) in the Morotoge shrimp,Pandalopsis japonica. The nAChR subunits exhibited common structural characteristics, including a signal peptide sequence, a large N-terminal extracellular domain with conserved motifs for ligand binding (loops A–F), and a transmembrane (TM) domain with four hydrophobic TM motifs (TM1–TM4). Based on the conserved GEK motifs located just before TM2, all five nAChR subunits fromP. japonicaappear to be cation-selective ion channels. Among the five subunits, Pajα3 and Pajβ1 clustered together with insect core groups, whereas Pajα10, Pajα11, and Pajα12 were classified as a divergent group. Three distinct transcripts were identified in Pajα3, presumably due to alternative splicing between TM3 and TM4, which may be involved in channel formation with other subunits. All five nAChR subunits were expressed predominantly in neuronal tissues, including the brain, sinus gland/X-organ complex, thoracic ganglia, and abdominal ganglia, with no significant differences in subunit expression levels among the neuronal tissues. The five shrimp nAChR subunits could not be functionally expressed inXenopusoocytes, but coexpression of Pajβ1 and rat α4 subunit (Rα4) formed functional channels responding to acetylcholine. Functional expression of vertebrate α subunit (Rα4) with invertebrate β1 subunit (Pajβ1) will expand our knowledge regarding the structural characteristics and molecular gating mechanism of invertebrate nAChRs.
