Species delimitation of Culicoides complex species can be challenging. Among species within the Culicoides variipennis complex, C. sonorensis is considered the primary vector of bluetongue virus (BTV) and epizootic hemorrhagic disease virus (EHDV) in North America. Morphological identification of C. sonorensis within the C. variipennis complex is laborious, time-consuming, and requires entomology expertise. Therefore, in this study we developed and validated a multiplex real-time PCR for rapid detection and differentiation of C. sonorensis from the two other main cryptic species (C. variipennis and C. occidentalis) within the C. variipennis complex. The assay targets the EF1α gene and has a built-in internal control targeting 18 S. The specificity and the sensitivity of the multiplex real-time PCR were evaluated using morphologically identified reference and field-collected specimens. The multiplex PCR was 100% specific when nucleic acid extracted from C. variipennis, sonorensis, and occidentalis specimens was tested. When nucleic acid extracted from pools of midges was tested, the multiplex PCR was able to detect all three Culicoides species with comparable sensitivity. The multiplex assay, however, failed to detect eight morphologically identified C. sonorensis specimens collected from Alberta in 2014. The EF1α gene sequences of these specimens formed a distinct phylogenetic cluster, amongst those from C. variipennis, sonorensis, and occidentalis, suggesting that they belong to a different species. We hypothesize that those specimens might be C. albertensis, the only other species remaining in the C. variipennis complex with known geographical distribution in North America. We believe that this highly sensitive and specific multiplex real-time PCR assay could be an effective tool for rapid detection and differentiation of C. sonorensis, the known vector of BTV and EHDV, in trap collections in future vector surveillance programs.

When university students are asked to rate their instructors, their evaluations are often influenced by the demographic characteristics of the instructor-such as the instructor's race, gender, or language background. These influences can manifest in unfair systematic biases against particular groups of teachers and hamper movements to promote diversity in higher education. When and how do these biases develop? Here, we begin to address these questions by examining children's sociolinguistic biases against teachers who speak with different accents. To do this, we presented 5-year-old Canadian English-speaking children with pairs of adult talkers. Children were asked to select "who they'd like to be their teacher" then they rated "how good of a teacher" they thought each talker would be on a 5-point scale. In each trial, one talker spoke in the locally dominant variety of Canadian English, and the other spoke in a different accent. Children strongly preferred Canadian-accented teachers over teachers who spoke with non-native (i.e., French or Dutch) accents, but also demonstrated a preference for Canadian teachers over teachers who spoke with non-local regional accents (i.e., Australian or British). In line with the binary choice data, children rated the Canadian talkers more favourably. The relationship between the gender of the teacher and the gender of the child also impacted ratings. This work demonstrates that even at the onset of formal education, children may already exhibit signs of accent-based biases. We discuss these findings in relation to the growing literature on implicit bias in higher education.

The feelings of difficulty and familiarity (FOD and FOF) are two types of metacognitive experiences. Both may influence student engagement and the application of metacognitive strategies, but these relationships are not well understood, in part because many studies have relied on self-report measures of behaviors that may not accurately reflect students' actual behaviors. In this study, FOD and FOF were related to objective measures of off-task behaviors and metacognitive strategies. These measures were extracted from 88 sixth graders' action logs within a computer-based learning environment known as Betty's Brain. Pre- and post-tests were administered to assess learning. Results reveal that high-FOD students showed more off-task behaviors and fewer strategic behaviors than low-FOD students, particularly when this difference was measured in terms of the frequency (as opposed to proportion) of strategic behaviors. FOF was not associated with off-task behaviors and metacognitive strategies but emerged as a moderator in the relationship between FOD and learning gains. Low-FOD students learned more than high-FOD students in the low-FOF group, but such a difference was not found in the high-FOF group.

Summary: Patients with cancer are at increased risk of death from COVID-19 and have reduced immune responses to SARS-CoV2 vaccines, necessitating regular boosters. We performed comprehensive chart reviews, surveys of patients attitudes, serology for SARS-CoV-2 antibodies and T cell receptor (TCR) β sequencing for cellular responses on a cohort of 982 cancer patients receiving active cancer therapy accrued between November-3-2020 and Mar-31-2023. We found that 92 · 3% of patients received the primer vaccine, 70 · 8% received one monovalent booster, but only 30 · 1% received a bivalent booster. Booster uptake was lower under age 50, and among African American or Hispanic patients. Nearly all patients seroconverted after 2+ booster vaccinations (>99%) and improved cellular responses, demonstrating that repeated boosters could overcome poor response to vaccination. Receipt of booster vaccinations was associated with a lower risk of all-cause mortality (HR = 0 · 61, p = 0 · 024). Booster uptake in high-risk cancer patients remains low and strategies to encourage booster uptake are needed.

BackgroundPolycyclic aromatic hydrocarbons (PAHs) are ubiquitous pollutants originating from petrogenic and pyrogenic sources. PAH compounds can cross the placenta, and prenatal PAH exposure is linked to adverse infant and childhood health outcomes.ObjectiveIn this first human transcriptomic assessment of PAHs in the placenta, we examined associations between prenatal PAH exposure and placental gene expression to gain insight into mechanisms by which PAHs may disrupt placental function.MethodsThe ECHO PATHWAYS Consortium quantified prenatal PAH exposure and the placental transcriptome from 629 pregnant participants enrolled in the CANDLE study. Concentrations of 12 monohydroxy-PAH (OH-PAH) metabolites were measured in mid-pregnancy urine using high performance liquid chromatography tandem mass spectrometry. Placental transcriptomic data were obtained using paired-end RNA sequencing. Linear models were fitted to estimate covariate-adjusted associations between maternal urinary OH-PAHs and placental gene expression. We performed sex-stratified analyses to evaluate whether associations varied by fetal sex. Selected PAH/gene expression analyses were validated by treating HTR-8/SVneo cells with phenanthrene, and quantifying expression via qPCR.ResultsUrinary concentrations of 6 OH-PAHs were associated with placental expression of 8 genes. Three biological pathways were associated with 4 OH-PAHs. Placental expression of SGF29 and TRIP13 as well as the vitamin digestion and absorption pathway were positively associated with multiple metabolites. HTR-8/SVneo cells treated with phenanthrene also exhibited 23 % increased TRIP13 expression compared to vehicle controls (p = 0.04). Fetal sex may modify the relationship between prenatal OH-PAHs and placental gene expression, as more associations were identified in females than males (45 vs 28 associations).DiscussionOur study highlights novel genes whose placental expression may be disrupted by OH-PAHs. Increased expression of DNA damage repair gene TRIP13 may represent a response to double-stranded DNA breaks. Increased expression of genes involved in vitamin digestion and metabolism may reflect dietary exposures or represent a compensatory mechanism to combat damage related to OH-PAH toxicity. Further work is needed to study the role of these genes in placental function and their links to perinatal outcomes and lifelong health.

BACKGROUND: Spontaneous preterm birth accounts for most preterm births and leads to significant morbidity in the newborn and childhood period. This subtype of preterm birth represents an increasing proportion of all preterm births when compared with medically indicated preterm birth, yet it is understudied in omics analyses. The placenta is a key regulator of fetal and newborn health, and the placental transcriptome can provide insight into pathologic changes that lead to spontaneous preterm birth. OBJECTIVE: This analysis aimed to identify genes for which placental expression was associated with spontaneous preterm birth (including early preterm and late preterm birth). STUDY DESIGN: The ECHO PATHWAYS consortium extracted RNA from placental samples collected from the Conditions Affecting Neurocognitive Development and Learning in Early Childhood and the Global Alliance to Prevent Prematurity and Stillbirth studies. Placental transcriptomic data were obtained by RNA sequencing. Linear models were fit to estimate differences in placental gene expression between term birth and spontaneous preterm birth (including gestational age subgroups defined by the American College of Obstetricians and Gynecologists). Models were adjusted for numerous confounding variables, including labor status, cohort, and RNA sequencing batch. This analysis excluded patients with induced labor, chorioamnionitis, multifetal gestations, or medical indications for preterm birth. Our combined cohort contained gene expression data for 14,023 genes in 48 preterm and 540 term samples. Species-focused conservation requires a thorough understanding of species' distributions. Delineating a species' distribution requires taxonomic knowledge and adequate occurrence data. For plants and fungi, herbaria represent a valuable source of large-scale occurrence data. Advances in digital technology mean that data from many herbarium collections worldwide are now easily accessible. However, species concepts can change over time requiring herbarium records to be re-examined and databases updated, which does not always occur synchronously across all collections. Therefore, non-critical use of these data can promote inaccuracies in understanding species distributions. Taxonomic revisions are common in understudied organisms, such as lichens. Here, we illustrate how changing taxonomy and non-critical acceptance of online data affects our understanding of disjunct distributions, using the lichen Brodoa oroarctica (Krog) Goward as an example. Defining the distribution of the arctic lichen B. oroarctica is confounded by changing taxonomy and uncertainty of herbarium records that pre-date taxonomic revisions. We review the distribution of this species in the literature and in aggregate occurrence databases, and verify herbarium specimens that represent disjunct occurrences in eastern North America to present an updated account of its distribution and frequency in eastern North America. We show that knowledge of changing species taxonomy is essential to depicting accurate species distributions. Background: Current measures to prevent spinal surgical site infection (SSI) lack compliance and lead to antimicrobial resistance. We aimed to examine the effectiveness of bundled preoperative intranasal photodynamic disinfection therapy (nPDT) and chlorhexidine gluconate (CHG) body wipes in the prophylaxis of spine SSIs in adults, as well as determine our institutional savings attributable to the use of this strategy and identify adverse events reported with nPDT-CHG. Methods: We performed a 14-year prospective observational interrupted time-series study in adult (age > 18 yr) patients undergoing emergent or elective spine surgery with 3 time-specific cohorts: before rollout of our institution's nPDT-CHG program (2006-2010), during rollout (2011-2014) and after rollout (2015-2019). We used unadjusted bivariate analysis to test for temporal changes across patient and surgical variables, and segmented regression to estimate the effect of nPDT-CHG on the annual SSI incidence rates per period. We used 2 models to estimate the cost of nPDT-CHG to prevent 1 additional SSI per year and the annual cumulative cost savings through SSI prevention. Results: Over the study period, 13 493 patients (mean 964 per year) underwent elective or emergent spine surgery. From 2006 to 2019, the mean age, mean Charlson Comorbidity Index (CCI) score and mean Spine Surgical Invasiveness Index (SSII) score increased from 48.4 to 58.1 years, from 1.7 to 2.6, and from 15.4 to 20.5, respectively (p < 0.001). Unadjusted analysis confirmed a significant decrease in the annual number (74.6 to 26.8) and incidence (7.98% to 2.67%) of SSIs with nPDT-CHG (p < 0.001). After adjustment for mean age, mean CCI score and mean SSII score, segmented regression showed an absolute reduction in the annual SSI incidence rate of 3.36% per year (p < 0.001). The estimated annual cost to prevent 1 additional SSI per year was about $1350-$1650, and the estimated annual cumulative cost savings were $2 484856-S2 495 016. No adverse events were reported with nPDT-CHG. Conclusion: Preoperative nPDT-CHG administration is an effective prophylactic strategy for spinal SSIs, with significant cost savings. Given its rapid action, minimal risk of antimicrobial resistance, broad-spectrum activity and high compliance rate, preoperative nPDT-CHG decolonization should be the standard of care for all patients undergoing emergent or elective spine surgery. Contexte : L'application des mesures de prévention des infections des plaies opératoires (IPO) du rachis laisse actuellement à désirer et pourrait contribuer à la résistance aux antibiotiques. Nous avons voulu vérifier l'efficacité de la thérapie photodynamique antimicrobienne (TPA) intranasale préopératoire alliée à l'utilisation de lingettes désinfectantes à la chlorhexidine (LDC) pour le corps en prophylaxie des IPO du rachis chez des adultes, calculer les économies réalisées par notre établissement grâce à la stratégie TPD-LDC et recenser ses effets indésirables. Méthodes : Nous avons procédé à une étude observationnelle de séries chronologiques interrompues échelonnée sur une période de 14 ans chez des adultes (âgés de >18 ans) soumis à une chirurgie urgente ou non urgente du rachis auprès de 3 cohortes chronologiques spécifiques : avant le déploiement de la stratégie dans notre établissement (2006-2010), puis durant (2011-2014) et après son déploiement (2015-2019). Nous avons appliqué une analyse bivariée non ajustée pour vérifier les changements dans le temps selon la patientèle et les paramètres chirurgicaux, et segmenté la régression pour estimer l'effet de la stratégie TPA-LDC sur les taux d'incidence annuels moyens d'IPO par période. Nous avons utilisé 2 modèles pour estimer le coût de la stratégie TPA-LDC apte à prévenir une IPO additionnelle par année et les économies cumulatives annuelles moyennes générées par la prévention des IPO. Résultats : Durant la période de l'étude, 13 493 malades (964 par année en moyenne) ont subi une chirurgie urgente ou non urgente du rachis. Entre 2006 et 2011 et entre 2015 et 2019, l'âge moyen, l'indice moyen de comorbidité de Charlson (ICC) et l'indice moyen d'invasivité de la chirurgie du rachis (IICR) sont passés de 48,4 à 58,1 ans, de 1,7 à 2,6 et de 15,4 à 20,5, respectivement (p < 0,001). L'analyse non ajustée a confirmé une baisse significative du nombre annuel (de 74,6 à 26,8) et de l'incidence (de 7,98% à 2,67%) des IPO associée à la stratégie TPA-LDC (p < 0,001). Après ajustement pour tenir compte de l'âge, de l'ICC et de l'IICR moyens, la régression segmentée a montré une réduction absolue du taux d'incidence annuel des IPO de 3,36% par année (p < 0,001). Le coût annuel moyen estimé pour prévenir une IPO additionnelle a été d'environ 1350$-1650$, et les économies cumulatives annuelles moyennes estimées ont été de 2484856 $-2 495 016$. Aucun effet indésirable n'a été signalé avec la stratégie TPA-LDC. Conclusion : L'application préopératoire de la stratégie TPA-LDC est une mesure prophylactique efficace pour les IPO du rachis, et donne lieu à des économies significatives. Transition metrics, which quantify the propensity for one event to follow another, are often utilized to study sequential patterns of behaviors, emotions, actions, and other states. However, little is known about the conditions in which application of transition metrics is appropriate. We report on two experiments in which we simulated sequences of states to explore the properties of common transition metrics (conditional probability, D'Mello's "L," lag sequential analysis, and Yule's "Q") where results should be null (i.e., random sequences). In experiment 1, we found that transition metrics produced statistically significant results with non-null effect sizes (e.g., "Q" > 0.2) when sequences of states were short. In experiment 2, we explored situations where consecutively repeated states (i.e., loops, or self-transitions) are impossible -- e.g., in digital learning environments where actions such as hint requests cannot be made twice in a row. We found that impossible loops affected all transition metrics (e.g., "Q" = 0.646). Based on simulations, we recommend sequences of length 50 or more for transition metric analyses. Our software for calculating transition metrics and running simulated experiments is publicly available.

Confusion may benefit learning when it is resolved or partially resolved. Metacognitive strategies (MS) may help learners to resolve confusion when it occurs during learning and problem solving. This study examined the relationship between confusion and MS that students evoked in Betty's Brain, a computer-based learning-by-modelling environment where elementary and middle school students learn science by building causal maps. Participants were sixth graders. Emotion data were collected from real-time observations by trained researchers. MS and task performance information were determined by analyzing the action logs. Pre- and post-tests were used to assess learning gains. The results revealed that the use of MS was a function of the state of student confusion. However, confusion resolution was not related to MS behaviour, and MS did not moderate the effect of confusion on student task performance in Betty's Brain or on learning gains.

PurposeExposures early in life, beginning in utero, have long-term impacts on mental and physical health. The ECHO prenatal and early childhood pathways to health consortium (ECHO-PATHWAYS) was established to examine the independent and combined impact of pregnancy and childhood chemical exposures and psychosocial stressors on child neurodevelopment and airway health, as well as the placental mechanisms underlying these associations.ParticipantsThe ECHO-PATHWAYS consortium harmonises extant data from 2684 mother-child dyads in three pregnancy cohort studies (CANDLE [Conditions Affecting Neurocognitive Development and Learning in Early Childhood], TIDES [The Infant Development and Environment Study] and GAPPS [Global Alliance to Prevent Prematurity and Stillbirth]) and collects prospective data under a unified protocol. Study participants are socioeconomically diverse and include a large proportion of Black families (38% Black and 51% White), often under-represented in research. Children are currently 5-15 years old. New data collection includes multimodal assessments of primary outcomes (airway health and neurodevelopment) and exposures (air pollution, phthalates and psychosocial stress) as well as rich covariate characterisation. ECHO-PATHWAYS is compiling extant and new biospecimens in a central biorepository and generating the largest placental transcriptomics data set to date (N=1083).Findings to dateEarly analyses demonstrate adverse associations of prenatal exposure to air pollution, phthalates and maternal stress with early childhood airway outcomes and neurodevelopment. Placental transcriptomics work suggests that phthalate exposure alters placental gene expression, pointing to mechanistic pathways for the developmental toxicity of phthalates. We also observe associations between prenatal maternal stress and placental corticotropin releasing hormone, a marker of hormonal activation during pregnancy relevant for child health. Other publications describe novel methods for examining exposure mixtures and the development of a national spatiotemporal model of ambient outdoor air pollution.Future plansThe first wave of data from the unified protocol (child age 8-9) is nearly complete. Future work will leverage these data to examine the combined impact of early life social and chemical exposures on middle childhood health outcomes and underlying placental mechanisms.

Background: Providing adaptive scaffolds to help learners develop effective self-regulated learning (SRL) behaviours has been an important goal for intelligent learning environments. Adaptive scaffolding is especially important in open-ended learning environments (OELE), where novice learners often face difficulties in completing their learning tasks. Objectives: This paper presents a systematic framework for adaptive scaffolding in Betty's Brain, a learning-by-teaching OELE for middle school science, where students construct a causal model to teach a virtual agent, generically named Betty. We evaluate the adaptive scaffolding framework and discuss its implications on the development of more effective scaffolds for SRL in OELEs. Methods: We detect key cognitive/metacognitive "inflection points," that is, moments where students' behaviours and performance change during learning, often suggesting an inability to apply effective learning strategies. At inflection points, Mr. Davis ("a mentor agent in Betty's Brain") or Betty ("the teachable agent") provides context-specific conversational feedback, focusing on strategies to help the student become a more productive learner, or encouragement to support positive emotions. We conduct a classroom study with 98 middle schoolers to analyse the impact of adaptive scaffolds on students' learning behaviours and performance. We analyse how students with differential pre-to-post learning outcomes receive and use the scaffolds to support their subsequent learning process in Betty's Brain. Results and Conclusions: Adaptive scaffolding produced mixed results, with some scaffolds (viz., strategic hints that supported debugging and assessment of causal models) being generally more useful to students than others (viz., encouragement prompts). Additionally, there were differences in how students with high versus low learning outcomes responded to some hints, as suggested by the differences in their learning behaviours and performance in the intervals after scaffolding. Overall, our findings suggest how adaptive scaffolding in OELEs like Betty's Brain can be further improved to better support SRL behaviours and narrow the learning outcomes gap between high and low performing students. Implications: This paper contributes to our understanding and impact of adaptive scaffolding in OELEs. The results of our study indicate that successful scaffolding has to combine context-sensitive inflection points with conversational feedback that is tailored to the students' current proficiency levels and needs. Also, our conceptual framework can be used to design adaptive scaffolds that help students develop and apply SRL behaviours in other computer-based learning environments.

It is widely recognized that debugging is challenging for novice programmers and, as such, computing educators and researchers have called for explicit debugging instruction. Debugging requires various knowledge and skills, and different students may show different strengths and weaknesses. An understanding of such individual differences is important as it may guide personalized instruction. The current study investigated individual differences in debugging in an undergraduate introductory computer science course. We extracted variables related to debugging from students' submission traces to programming problems in the first month of the course. We applied latent profile analysis to these variables and identified three distinctive profiles. Profile A showed higher debugging accuracy and speed. Profile B showed lower debugging performance in runtime and logic errors, while profile C had lower performance in syntactic errors and tended to make large code edit every submission. Students' gender and self-rated programming ability predicted profile membership. Moreover, profile A got higher scores than the others in the first exam, and this difference persisted in the second and third exam, even controlling for background variables and score on the first exam. We investigated how students transitioned across debugging profiles over the duration of the course. From the beginning to the end of the course, a large part of students stayed in lower performance profiles. Overall, these findings support the call that debugging should be taught at an early stage and suggest that different groups may need different debugging instructions or support.

This qualitative research study aimed to identify factors, as perceived by Administration and Leadership Studies doctoral student graduates and faculty mentors, that were helpful in securing terminal degree success. Strategies employed and obstacles overcome are detailed from both groups' perspectives. The researchers conducted the study at a public university in the mid-Atlantic region. The purpose was to explain the experiences of doctoral student graduates and their faculty mentors to explore what traits and strategies were employed to complete and defend a dissertation. Some of the results revealed the importance of establishing clear communication, persistently engaging in the process, and enjoying the research journey. Approximately 50% of those who attempt a doctoral program are successful and the other 50% are "all-but-dissertation" (ABD) (Ames et al., 2018; Hawley, 2010; Jimenez y West et al., 2011; Landi, 2017; Scott & Johnson, 2021). By identifying and proactively avoiding common pitfalls, the didactic doctoral student has an increased probability of achieving terminal degree success.
