The purpose of this study was to investigate the mechanism of EMP1 action in high glucose (HG)‐induced H9c2 cardiac cell pyroptosis and oxidative injury. Rat cardiomyocytes H9c2 were exposed to 33 mM glucose for 24, 48, or 72 h to induce cytotoxicity. EMP1‐siRNA, NLRP3 agonist Nigericin, and pcNDA‐RAS were used to treat H9c2 cells under HG conditions. Cell Counting Kit (CCK)‐8 assay showed that cell proliferation was decreased following HG induction, which was rescued by EMP1 knockdown. Our results also suggested that EMP1 siRNA transfection significantly decreased the apoptosis and pyroptosis of HG‐induced cells, as indicated by the reduction of NLRP3 IL‐1β, ASC, GSDMD, cleaved‐caspase1 and cleaved‐caspase3 levels in HG‐induced H9c2 cells. In addition, EMP1 knockdown alleviated HG‐induced mitochondrial damage and oxidative stress in H9c2 cells. NLRP3 activation reversed the inhibitory effects of EMP1 knockdown on pyroptosis and oxidative stress in HG‐induced H9c2 cells. Mechanistically, we found that EMP1 knockdown suppressed the RAS/RAF/MAPK signaling pathway in HG‐induced H9c2 cells. RAS overexpression blocked the protective effect of EMP1 knockdown on HG‐induced H9c2 cell apoptosis, pyroptosis, and oxidative injury. The recovery of valuable metals from spent ternary lithium-ion batteries (LIBs) has recently garnered significant attention due to the imperatives of the circular economy and environmental management. While the reclamation of lithium is generally straightforward, the hydrometallurgical methods most frequently employed for leaching and separating the remaining nickel, cobalt, and manganese from spent electrode material often yield secondary liquid and solid wastes. In this study, we present a mechanochemical strategy aimed at repurposing lithium-removed spent ternary LIBs cathode material as a precursor for perovskite oxides through a straightforward and scalable solid-state high-energy ball-milling synthesis. By optimizing the synthesis procedure, we have obtained a perovskite catalyst composed of LaNi0.6Co0.2Mn0.2O3 with a trace amount of phase-separated surface NiO nanocrystals. This catalyst demonstrates outstanding performance in the low-temperature oxidation of CO, exhibiting no degradation in performance over extended periods of service. Notably, it achieves a T50 of 162 °C and a T90 of 197 °C, which compares favorably with previously reported perovskite catalysts prepared via wet synthesis, utilizing fine chemicals as precursors. Purpose: Teaching Games for Understanding (TGfU) has received positive support. However, implementing TGfU-based physical education (PE) can reveal notable differences between urban and rural contexts depending on the prevailing cultural capital. This study aimed to explore the differences in TGfU-based PE between one urban and one rural primary school from the perspective of cultural capital.Method: The participants were 44 fifth-graders from one urban and one rural primary school. Twenty TGfU-based PE sessions were conducted over a 10-week period. Participatory observation and semistructured interviews were employed to collect the data.Findings: The differences observed in TGfU-based PE between these urban and rural primary schools included disparities in the acceptance of TGfU, (2) adaptation to the changing game rules in TGfU, and (3) volleyball skills and game performance.Discussion/Conclusion: Urban–rural differences were discerned in the implementation of TGfU-based PE as the resources in the rural school might be limited. Given the relatively constrained cultural capital and environmental resources at the rural primary school, the adoption of TGfU-based PEs might necessitate tailored strategies for rural contexts. Background: Lymph node metastasis (LNM) is a critical factor affecting the outcomes of head and neck squamous cell carcinoma (HNSCC) and the main reason for treatment failure. This study was designed to examine the effects of the key genes involved in the LNM of HNSCC. Methods: Tissue samples (HNSCC) were examined by transcriptome sequencing, and the core genes associated with LNM were detected via bioinformatics analysis. The functions of these core genes were then validated using the TCGA biological database and their effects on the propagation, invasion, and metastasis of HNSCC cells were evaluated through cell culture experiments. Moreover, the effect of core gene expression on the LNM capability of HNSCC was confirmed via a footpad xenograft mice model. Results: In the findings, a key gene involved in the LNM of HNSCC was identified as SLC7A2. It was correlated with adverse clinical prognosis and expressed with low expression in HNSCC tissues. As shown in cell culture experiments, FaDu and SCC15 cell growth, invasion, and migration were inhibited when SLC7A2 was overexpressed. Further, cell apoptosis was stimulated, and lymphangiogenesis was suppressed through the downregulation of CPB2 expression. Animal studies demonstrated that the growth and LNM of HNSCC cells were inhibited by SLC7A2 overexpression. Conclusion: It is concluded that SLC7A2 is involved in HNSCC lymphatic metastasis by controlling CPB2 function. Spontaneous vegetation within a managed green space is often regarded as unwelcoming and insignificant weeds. This perception is still deep-rooted among green-space managers and the general public worldwide; they are generally uncertain about the management needs after allowing these groups of flora to take root. The short-term growth dynamics of both spontaneous and planted vegetation should be analyzed, and a widely acceptable, feasible management plan to balance aesthetic and ecological functions should be formulated with the backing of data and analysis for such fast-growing flora in tropical and subtropical regions. A manicured, extensive green roof with only seven (two native, five exotic) plant species was transformed into a renaturalized biotope by replacing 15 native ferns and forb species over 15 months. After planting, a baseline plant survey was conducted, with 54 plant species representing spontaneous growth and 14 planted species alive (7 planted native species survived, plus 7 species planted prior to renaturalization revived). Three quarterly plant surveys recorded the cover-abundance of each species, and the growth dynamics of the planted and spontaneous plant species were evaluated over the first year of study. During each quarterly survey, the number of planted and spontaneous plant species remained stable (ranging from 14 to 16 species and 51 to 54 species, respectively), with a constant turnover of 11 to 12 die-out species and 11 to 12 newly colonized or revived species. Plant coverage of different plant forms fluctuated slightly (within 7%) in the quarterly surveys according to seasonal changes, except for ferns, which outperformed (12% increase in coverage in a year) all the other plant forms. The height of the planted vegetation fluctuated in a year, being shorter during the summer, while the height of spontaneous vegetation remained stable throughout the year, exhibiting resilience to scouring heat. The seasonal growth tendencies of both planted and spontaneous plants were illustrated in relation to their species ranks, and further hierarchical cluster analysis was conducted for the clustering of spontaneous species. Their differential growth patterns provided comprehensive information or supported decisions regarding plant selection and maintenance, which is a scientific novelty within this unexplored topic. Management recommendations based on the findings were suggested to fulfill both aesthetic and ecological needs. Species with stable and less stable growth patterns could be useful to meet maintenance efficiency and biodiversity enhancement needs, respectively. BACKGROUND: Bombyx mori nuclear polyhedrosis virus (BmNPV), as a typical baculovirus, is the primary pathogen that infects the silkworm B. mori, a lepidopteran species. Owing to the high biological safety of BmNPV in infecting insects, it is commonly utilized as a biological insecticide for pest control. Apoptosis is important in the interaction between the host and pathogenic microorganisms. MicroRNAs (miRNAs) influence immune responses and promote stability of the immune system via apoptosis. Therefore, the study of apoptosis‐related miRNA in silkworms during virus infection can not only provide support for standardizing the prevention and control of diseases and insect pests, but also reduce the economic losses to sericulture caused by the misuse of biological pesticides. RESULTS: Through transcriptome sequencing, we identified a miRNA, miR‐31‐5p, and demonstrated that it can inhibit apoptosis in silkworm cells and promote the proliferation of BmNPV in BmE‐SWU1 cells. We identified a target gene of miR‐31‐5p, B. mori cytochrome P450 9e2 (BmCYP9e2), and demonstrated that it can promote apoptosis in silkworm cells and inhibit the proliferation of BmNPV. Moreover, we constructed transgenic silkworm strains with miR‐31‐5p knockout and confirmed that they can inhibit the proliferation of BmNPV. CONCLUSION: These data indicate that miR‐31‐5p may exert functions of inhibiting apoptosis and promoting virus proliferation by regulating BmCYP9e2. The findings demonstrate how miRNAs influence host cell apoptosis and how they are involved in the host immune system response to viruses, providing important insights into the applications of biological insecticides for pest control. © 2024 Society of Chemical Industry. Nicotinic acetylcholinergic receptors (nAChR's) have been implicated in several brain disorders, including addiction, Parkinson's disease, Alzheimer's disease and schizophrenia. Here we report in vitro selectivity and functional properties, toxicity in rats, in vivo evaluation in humans, and comparison across species of [18 F]Nifene, a fast acting PET imaging agent for α4β2* nAChRs. Nifene had subnanomolar affinities for hα2β2 (0.34 nM), hα3β2 (0.80 nM) and hα4β2 (0.83 nM) nAChR but weaker (27-219 nM) for hβ4 nAChR subtypes and 169 nM for hα7 nAChR. In functional assays, Nifene (100 μM) exhibited 14% agonist and >50% antagonist characteristics. In 14-day acute toxicity in rats, the maximum tolerated dose (MTD) and the no observed adverse effect level (NOAEL) were estimated to exceed 40 μg/kg/day (278 μg/m2 /day). In human PET studies, [18 F]Nifene (185 MBq; 15% than males. No significant aging effect was observed in [18 F]Nifene binding over 5 decades. In all species (mice, rats, monkeys, and humans) thalamus showed highest [18 F]Nifene binding with reference region ratios >2 compared to extrathalamic regions. Our findings suggest that [18 F]Nifene PET may be used to study α4β2* nAChRs in various CNS disorders and for translational research.
