The solar limb darkening function is well known and is widely employed in models of the solar atmosphere. However, there has been a lack of systematic spectrally resolved measurements. Therefore we recently decided to start an observing campaign with the Gregory Coud\'e Telescope at IRSOL in Locarno in order to produce a spectral atlas obtained at 10 different heliocentric angles $\theta$, chosen so that $\mu = \cos\theta$ covers the interval from 0.1 to 1.0 in step of 0.1. The measurements carried out till now include the spectral range from 439 nm to 666 nm. The collected data provide information about the anisotropy of the emergent radiation field on the solar surface, allowing a better modeling of the Second Solar Spectrum. In recent history, institutional investors have increased allocations to alternative assets in an effort to boost portfolio returns. The attention is now turning to individual investors. Significant partnerships, private platforms, and organizations have emerged to democratize access to alternative investments. In 2021, the US Department of Labor also took a position, suggesting that it might be prudent to include private equity in individual retirement accounts, such as in 401(k) and 403(b) plans. While institutional investors have decades of experience investing in alternative assets, the vast majority of individual investors do not. Consequently, relatively little is known about optimal alternative asset allocations for individual investors. This article presents historical risk and return characteristics from private investments including venture capital, private equity, private credit, and real estate, and constructs portfolios building on a traditional 60/40 (public equity/public debt) allocation, with varying levels of a private markets index, to examine portfolio risk and return trade-offs. Findings reveal that there are portfolio benefits to both returns and risk from adding a significant allocation to alternative investments and that wealth impacts can be sizable over time. Ablative materials are widely employed to protect space vehicles from the extreme thermal conditions experienced during their flight into a planetary atmosphere. Carbon-phenolic ablators are composed of a phenolic matrix and a fibrous carbon reinforcement. In the present study, the fibrous reinforcement has been modified through the deposition of thin protective layers of zirconium oxide and aluminum oxide, with the objective of reducing fiber recession and oxidation. The depositions were carried out via atomic layer deposition (ALD), a method that allows for the controlled deposition of uniform and conformal coatings on the carbon felt fibers. The depositions were subsequently evaluated through SEM-EDS analysis. Pristine and ALD-modified felts were impregnated with a phenolic resin matrix and the ablation performance of the composite materials was evaluated through oxyacetylene flame tests. The results demonstrated that, in comparison to uncoated ablators, the ALD-modified samples exhibited enhanced performance in terms of mass loss and surface recession: compared to uncoated ablators, the former was 14% lower and the latter was diminished by 50%. Moreover, the morphological characterization of the tested specimens revealed a significantly reduced degree of oxidation of the coated fibers which were directly exposed to the flame. 108. Predicting Wall Pressure Fluctuations on Aerospace Launchers Through Machine Learning Approaches.

Artificial intelligence (AI) can be used to optimize the prediction of pressure fluctuations over the external surfaces of aerospace launchers and minimize the number of wind tunnel tests. In the present research, various machine learning (ML) techniques capable of predicting the acoustic load were tested and validated. The methods included decision trees, Gaussian Process Regression (GPR), Support Vector Machines (SVMs), artificial neural networks (ANNs), linear regression, and ensemble methods such as bagged and boosted trees. These algorithms were trained using experimental data from an extensive wind tunnel test campaign conducted to support the design of a VEGA (Advanced Generation European Vehicle) launcher vehicle and provide wall pressure fluctuations in many configurations. The main objective of this study was to identify, among several algorithms, the most suitable method able to process such complex databases efficiently and to provide reliable predictions. Different statistical indices, including the root mean square error (RMSE), the mean square error (MSE), and a correlation coefficient (R-squared), were employed to evaluate the performance of the ML methods. Among all the methods, the bagged tree algorithm outperformed the others, providing the most accurate predictions, with low RMSE and high R-squared values across all test cases. Other methods, such as the ANNs and GPR, exhibited higher errors, indicating their reduced suitability for this dataset. The results demonstrate that ensemble decision tree methods are highly effective in predicting acoustic loads, offering reliable predictions, even for configurations outside the training database. Simple Summary: A growing world population has increased the demand for animal protein, and the poultry industry is a major contributor to meeting this demand. Increasing feed costs and unreliable availability have prompted the poultry industry to identify and evaluate alternative feed ingredients. In this study, we evaluate the energy and amino acid availability of spirulina (Arthrospira platensis), a filamentous blue-green microalgae. The results show that spirulina is a high-quality feed ingredient for possible inclusion in poultry diets. Our study objective was to determine the apparent metabolizable energy (AME) and apparent ileal amino acid digestibility (AIAAD) of spirulina fed to broiler chickens and laying hens using the difference method. In both experiments, birds were either fed corn-soybean meal basal diets, containing no spirulina and formulated to provide the nutrient requirements of either broilers or layers, or fed test diets consisting of 25% spirulina and 75% of the appropriate basal diet. Titanium dioxide was added to all diets as an indigestible marker. The diets were fed to 10 replicate cages/treatment of broilers and 15 replicate cages/treatment of laying hens. The AME of spirulina for broilers was significantly lower (p < 0.05) (2368 ± 104 kcal/kg, as fed) than for laying hens (3144 ± 173 kcal/kg, as fed), suggesting bird type and age may influence energy utilization. The AIAAD of spirulina fed to broiler chickens did not differ from that of laying hens (p > 0.05), except for valine, alanine, and glycine, which were all significantly higher in laying hens (p < 0.05). Overall, the AIAAD for nonessential amino acids averaged 81.1%, with no significant difference between essential and nonessential amino acids. Differences in spirulina nutrient content cited in the literature support further research to determine the optimal inclusion of this alternative ingredient in broiler and layer diets. Background: The shortage of hospital beds for COVID-19 patients has been one critical cause of Emergency Department (ED) overcrowding. Purpose: We aimed at elaborating a strategy of conversion of hospital beds, from non-COVID-19 to COVID-19 care, minimizing both ED overcrowding and the number of beds eventually converted. Research Design: Observational retrospective study. Study Sample: We considered the centralized database of all ED admissions in the Lombardy region of Italy during the second "COVID-19 wave" (October to December 2020). Data collection and Analysis: We analyzed all admissions to 82 EDs. We devised a family of Monte Carlo simulations to evaluate the performance of hospital beds' conversion strategies triggered by ED crowding of COVID-19 patients, determining a critical number of beds to be converted when passing an ED-specific crowding threshold. Results: Our results suggest that the maximum number of patients waiting for hospitalization could have been decreased by 70% with the proposed strategy. Such a reduction would have been achieved by converting 30% more hospital beds than the total number converted in the region. Conclusions: The disproportion between reduction in ED crowding and additionally converted beds suggests that a wide margin to improve the efficiency of the conversions exists. Coffee is one of the most enjoyed beverages worldwide and whilst a wide array of secondary metabolites has been qualitatively and quantitatively characterised within the coffee bean, little is known about their distribution in different bean tissues. In this work, complementary mass spectrometry (MS) based approaches were utilised to determine the spatial metabolomic content of coffee beans, focusing on highlighting those which presented a regiospecific distribution and distinguished the different endosperm regions and the embryo. Preliminary exploration of the spatial metabolome of Coffea arabica coffee beans highlighted tissue-specific molecular features through an unsupervised statistical analysis. Then, the complementary information derived from higher resolution MS approaches resulted in the annotation of 13 metabolites which showed a specific enrichment in at least one tissue, and most of these were noted to be bioactive compounds or aroma/flavour precursors. Knowledge of the tissue distribution of these compounds could shed light on their biological role in plant physiology and bean development, but could also have relevant implications for their positive impact on human health due to coffee consumption as well as their possible effect on aroma and flavour upon roasting of green coffee beans. Blood serves as the primary global biological matrix for health surveillance, disease diagnosis, and response to drug treatment, holding significant promise for personalized medicine. The diverse array of lipids and metabolites in the blood provides a snapshot of both physiological and pathological processes, with many routinely monitored during conventional wellness checks. The conventional method involves intravenous blood collection, extracting a few milliliters via venipuncture, a technique limited to clinical settings due to its dependence on trained personnel. Microsampling methods have evolved to be less invasive (collecting ≤150 µL of capillary blood), user-friendly (enabling self-collection), and suitable for remote collection in longitudinal studies. Dried blood spot (DBS), a pioneering microsampling technique, dominates clinical and research domains. Recent advancements in device technology address critical limitations of classical DBS, specifically variations in hematocrit and volume. This review presents a comprehensive overview of state-of-the-art microsampling devices, emphasizing their applications and potential for monitoring metabolites and lipids in blood. The scope extends to diverse areas, encompassing population studies, nutritional investigations, drug discovery, sports medicine, and multi-omics research.

The study of bioactive molecules of marine origin has created an important bridge between biological knowledge and its applications in biotechnology and biomedicine. Current studies in different research fields, such as biomedicine, aim to discover marine molecules characterized by biological activities that can be used to produce potential drugs for human use. In recent decades, increasing attention has been paid to a particular group of marine invertebrates, the Ascidians, as they are a source of bioactive products. We describe omics data and computational methods relevant to identifying the mechanisms and processes of innate immunity underlying the biosynthesis of bioactive molecules, focusing on innovative computational approaches based on Artificial Intelligence. Since there is increasing attention on finding new solutions for a sustainable supply of bioactive compounds, we propose that a possible improvement in the biodiscovery pipeline might also come from the study and utilization of marine invertebrates’ innate immunity.

San Nicola Island, pertaining to the Tremiti Archipelago (Southern Adriatic Sea, Italy), is widely affected by cliff retreat and gravitational phenomena which severely threaten its monumental historical and natural value. In this study, geomorphological features of the area were derived following a stepwise approach, combining multitemporal stereoscopic aerial photo interpretations with morphometric analyses and detailed field surveys. A rockfall susceptibility map was created following a heuristic approach based on morphometric and geothematic parameters, accounting for slope, slope aspect, outcropping lithologies, structural discontinuities density, distance from landslide scarps, and presence of anthropic caves. Cliff sectors set on dolomitic limestones feature the highest susceptibility values, especially along the southeastern sector; medium values, instead, are found along the island flanks and along scarps located within the inner sectors; and the lowest values are detected on summit tabular surfaces. The achieved results were compared with historical maps and seismic data derived from local and national archives and catalogues, respectively. These analyses allowed us to define the role played by litho-structural and tectonic features on landslide occurrence and distribution, and their interplay in driving landscape evolution over centuries. Finally, this work represents a valuable scientific tool to support geomorphological studies for landslide hazard assessment and proper territorial planning in any other small insular areas, showing similar geological–geomorphological features and landscape values.

Domestic chimney cleaning is still mostly a manual procedure which can be overly complex, dangerous, and expansive. This paper describes the design of a novel robotic device for chimney cleaning that aims to provide a valuable alternative solution to the traditional manual techniques with user-friendly and low-cost features. The proposed device enables a significant reduction in operator risks, including roof falling and soot dust contact. The paper’s content describes, in detail, the design process, including a definition of the main design requirements and steps towards the manufacturing of a preliminary prototype. Moreover, a preliminary validation is described through laboratory tests to demonstrate the engineering feasibility and effectiveness of the proposed design solution for the intended semi-autonomous chimney-cleaning application.

The auditory brainstem response (ABR) by evoked potentials is a widespread auditory pathway assessment technique. This is largely applied due to its cost-effectiveness, practicality and ease of use. In contrast, it requires a trained professional to carry out the analysis of the results. This motivates several research efforts to increase the independence of the diagnostician. To this end, the present work shows the ability of three signal classification tools to differentiate ABR studies of normal hearing subjects from those who may have some pathology. As a starting point, the PhysioNet short term auditory evoked potentials databases are used to calculate the features later applied to construct the dataset. The features used are diverse classes of permutation entropy, fractal dimension, the Lyapunov exponent and the zero crossing rate. To ensure more accurate results, a Montecarlo simulation of one thousand trials is employed to train the classifiers and test the results. The goodness of the classification is performed upon the basis of the computation of quality parameters, such as the area under the receiver operating characteristic curve (ROC), the F1 score coefficient and the accuracy. In all the cases the methodology proposed gives high performance results that encourage the line of research of the present work.

121. Metabolomics Reveals the Molecular Mechanisms of Copper Induced Cucumber Leaf (Cucumis sativus) Senescence

Excess copper may disturb plant photosynthesis and induce leaf senescence. The underlying toxicity mechanism is not well understood. Here, 3-week-old cucumber plants were foliar exposed to different copper concentrations (10, 100, and 500 mg/L) for a final dose of 0.21, 2.1, and 10 mg/plant, using CuSO4 as the Cu ion source for 7 days, three times per day. Metabolomics quantified 149 primary and 79 secondary metabolites. A number of intermediates of the tricarboxylic acid (TCA) cycle were significantly down-regulated 1.4-2.4 fold, indicating a perturbed carbohydrate metabolism. Ascorbate and aldarate metabolism and shikimate-phenylpropanoid biosynthesis (antioxidant and defense related pathways) were perturbed by excess copper. These metabolic responses occur even at the lowest copper dose considered although no phenotype changes were observed at this dose. High copper dose resulted in a 2-fold increase in phytol, a degradation product of chlorophyll. Polyphenol metabolomics revealed that some flavonoids were down-regulated, while the nonflavonoid 4-hydroxycinnamic acid and trans-2-hydroxycinnamic acid were significantly up-regulated 4- and 26-fold compared to the control. This study enhances current understanding of copper toxicity to plants and demonstrates that metabolomics profiling provides a more comprehensive view of plant responses to stressors, which can be applied to other plant species and contaminants.

This paper presents a real-time solution for collision detection between objects based on the physics properties. Traditional approaches on collision detection often rely on the geometric relationships that computing the intersections between polygons. Such technique is very computationally expensive when applied for deformable objects. As an alternative, we approximate the 3D mesh in an spherical surface implicitly. This allows us to perform a coarse-level collision detection at extremely fast speed. Then a dynamic programming based procedure is applied to identify the collision in fine details. Our method demonstrates better prevention to collision tunnelling and works more efficiently than the state-of-the-arts., Comment: Computer-Aided Design, 2015

The early and non-invasive diagnosis of tumor diseases has been widely investigated by the scientific community focusing on the development of sensors/biomarkers that act as a way of recognizing the adhesion of circulating tumor cells (CTCs). As a challenge in this area, strategies for CTCs capture and enrichment currently require improvements in the sensors/biomarker’s selectivity. This can be achieved by understanding the biological recognition factors for different cancer cell lines and also by understanding the interaction between surface parameters and the affinity between macromolecules and the cell surface. To overcome some of these concerns, electrochemical sensors have been used as precise, fast-response, and low-cost transduction platforms for application in cytosensors. Additionally, distinct materials, geometries, and technologies have been investigated to improve the sensitivity and specificity properties of the support electrode that will transform biochemical events into electrical signals. This review identifies novel approaches regarding the application of different specific biomarkers (CD44, Integrins, and EpCAm) for capturing CTCs. These biomarkers can be applied in electrochemical biosensors as a cytodetection strategy for diagnosis of cancerous diseases.

This paper presents a morphometric and structural-geomorphological approach to identifying morphotectonic features across an area underlain by lithologies that do not easily record tectonic deformations but are widely affected by seismic activity. The middle Biferno River Valley (Central Italy) was chosen as a study area. It was investigated through analyses performed from the drainage basin scale (Biferno River) to the local scale (Ponte Liscione Dam). This methodological approach was applied to investigate the impact of morphotectonic processes in the study area, providing a geomorphology-based contribution to landscape evolution. The aim of the work was to discriminate the main morphotectonic elements acting in the complex tectonic framework of the study area by means of a combination of morphometric, morphotectonic, and structural-geomorphological analyses. The resulting data allowed us to identify three main systems of tectonic elements (F1, F2, and F3), which are attributable to compressive and extensional kinematics, as already reported in previous thematic studies. The tectonic setting reflects the geodynamic framework of the Periadriatic region, as highlighted by the distribution of seismogenic sources and the historical to recent seismicity. Finally, the present work could act as a valuable scientific tool for any geomorphological studies aimed at better defining the impact of morphotectonic processes in similar tectonically active regions hosting important and strategic artificial dams.

The temperature dependence of biological processes has been studied at the levels of individual biochemical reactions and organism physiology (e.g. basal metabolic rates) but has not been examined at the metabolic network level. Here, we used a systems biology approach to characterize the temperature dependence of the human red blood cell (RBC) metabolic network between 4 and 37 °C through absolutely quantified exo- and endometabolomics data. We used an Arrhenius-type model (Q10) to describe how the rate of a biochemical process changes with every 10 °C change in temperature. Multivariate statistical analysis of the metabolomics data revealed that the same metabolic network-level trends previously reported for RBCs at 4 °C were conserved but accelerated with increasing temperature. We calculated a median Q10 coefficient of 2.89 ± 1.03, within the expected range of 2-3 for biological processes, for 48 individual metabolite concentrations. We then integrated these metabolomics measurements into a cell-scale metabolic model to study pathway usage, calculating a median Q10 coefficient of 2.73 ± 0.75 for 35 reaction fluxes. The relative fluxes through glycolysis and nucleotide metabolism pathways were consistent across the studied temperature range despite the non-uniform distributions of Q10 coefficients of individual metabolites and reaction fluxes. Together, these results indicate that the rate of change of network-level responses to temperature differences in RBC metabolism is consistent between 4 and 37 °C. More broadly, we provide a baseline characterization of a biochemical network given no transcriptional or translational regulation that can be used to explore the temperature dependence of metabolism.

The increasing availability of metabolomics data necessitates novel methods for deeper data analysis and interpretation. We present a flux balance analysis method that allows for the computation of dynamic intracellular metabolic changes at the cellular scale through integration of time-course absolute quantitative metabolomics. This approach, termed "unsteady-state flux balance analysis" (uFBA), is applied to four cellular systems: three dynamic and one steady-state as a negative control. uFBA and FBA predictions are contrasted, and uFBA is found to be more accurate in predicting dynamic metabolic flux states for red blood cells, platelets, and Saccharomyces cerevisiae. Notably, only uFBA predicts that stored red blood cells metabolize TCA intermediates to regenerate important cofactors, such as ATP, NADH, and NADPH. These pathway usage predictions were subsequently validated through 13C isotopic labeling and metabolic flux analysis in stored red blood cells. Utilizing time-course metabolomics data, uFBA provides an accurate method to predict metabolic physiology at the cellular scale for dynamic systems.
