A química analítica utiliza procedimentos empíricos para determinar a concentração de analitos mediante técnicas instrumentais. Tais técnicas permitem relacionar a resposta instrumental (y) e o sinal medido no instrumento, que geralmente é função da concentração do analito, representada por (y = fx) que é comumente denominada curva analítica ou curva- padrão. Funções lineares são muito utilizadas para esta finalidade, assim a linearização desta função torna-se necessária para o cálculo da concentração do analito e para expressar corretamente o resultado é necessário um tratamento estatístico dos dados experimentais. Observa-se que os estudantes de disciplinas de química analítica instrumental apresentam dificuldades no tratamento de dados e na interpretação de resultados quando se utiliza uma curva analítica e na compreensão de seus parâmetros. Nesse sentindo, propõe-se um tutorial de regressão linear de dados experimentais para o cálculo de todos os parâmetros que caracterizam uma curva analítica, através de um procedimento utilizando a planilha eletrônica Excel 2016 do MS Office. Criou-se uma rotina de cálculos utilizando dados experimentais da determinação espectrofotométrica de proteína, usando como padrão a albumina de soro bovino (BSA) e da calibração de pHmetro, mas pode ser estendida a outros procedimentos onde o cálculo se faz necessário.

Historically, administration of dacarbazine to sarcoma patients was limited by frequent treat-ment-related nausea/vomiting and neutropenia. These toxicities are now largely preventable with contemporary antiemetics and growth factor support. In this single-arm, phase II study, dacarbazine 850 mg/m 2 was given on day 1 of each 3-week cycle until disease progression or intolerance with prophylactic serotonin-3 receptor, neurokinin-1 antagonists, corticosteroids, and pegfilgrastim. Coprimary endpoints included clinical benefit rate (CBR), and any grade of nausea/vomiting and/or grade 3–4 neutropenia. The Moroccan locust has been considered one of the most dangerous agricultural pests in the Mediterranean region. The economic importance of its outbreaks diminished during the second half of the 20th century due to a high degree of agricultural industrialization and other human-caused transformations of its habitat. Nevertheless, in Sardinia (Italy) from 2019 on, a growing invasion of this locust species is ongoing, being the worst in over three decades. Locust swarms destroyed crops and pasture lands of approximately 60,000 ha in 2022. Drought, in combination with increasing uncultivated land, contributed to forming the perfect conditions for a Moroccan locust population upsurge. The specific aim of this paper is the quantification of land cover land use (LCLU) influence with regard to the recent locust outbreak in Sardinia using remote sensing data. In particular, the role of untilled, fallow, or abandoned land in the locust population upsurge is the focus of this case study. To address this objective, LCLU was derived from Sentinel-2A/B Multispectral Instrument (MSI) data between 2017 and 2021 using time-series composites and a random forest (RF) classification model. Coordinates of infested locations, altitude, and locust development stages were collected during field observation campaigns between March and July 2022 and used in this study to assess actual and previous land cover situation of these locations. Findings show that 43% of detected locust locations were found on untilled, fallow, or uncultivated land and another 23% within a radius of 100 m to such areas. Furthermore, oviposition and breeding sites are mostly found in sparse vegetation (97%). This study demonstrates that up-to-date remote sensing data and target-oriented analyses can provide valuable information to contribute to early warning systems and decision support and thus to minimize the risk concerning this agricultural pest. This is of particular interest for all agricultural pests that are strictly related to changing human activities within transformed habitats.

Photoinjectors are the main high brightness electron sources for x-ray free electron lasers (XFELs). Photoinjector emittance reduction is one of the key knobs for improving XFEL lasing, so precise emittance measurement is critical. It is well known that rms emittance is very sensitive to low intensity tails of particle distributions in the phase space, whose measurement depends on the signal to noise ratio and image processing procedures. Such sensitivities make the interpretations of beam transverse brightness challenging, leading to different emittance definitions to reduce the impact of tail particles. In this paper, transverse phase space is analyzed in action and phase coordinates for both analytical models and experiments, which gives a more intuitive way to calculate the beam core brightness.

Synthesis gas fermentation is a microbial process, which uses anaerobic bacteria to convert CO-rich gases to organic acids and alcohols and thus presents a promising technology for the sustainable production of fuels and platform chemicals from renewable sources. Clostridium carboxidivorans and Clostridium autoethanogenum are two acetogenic bacteria, which have shown their high potential for these processes by their high tolerance toward CO and in the production of industrially relevant products such as ethanol, 1-butanol, 1-hexanol, and 2,3-butanediol. A promising approach is the coupling of gasification of biogenic residues with a syngas fermentation process. This study investigated batch processes with C. carboxidivorans and C. autoethanogenum in fully controlled stirred-tank bioreactors and continuous gassing with biogenic syngas produced by an autothermal entrained flow gasifier on a pilot scale >1200 °C. They were then compared to the results of artificial gas mixtures of pure gases. Because the biogenic syngas contained 2459 ppm O2 from the bottling process after gasification of torrefied wood and subsequent syngas cleaning for reducing CH4, NH3, H2S, NOX, and HCN concentrations, the oxygen in the syngas was reduced to 259 ppm O2 with a Pd catalyst before entering the bioreactor. The batch process performance of C. carboxidivorans in a stirred-tank bioreactor with continuous gassing of purified biogenic syngas was identical to an artificial syngas mixture of the pure gases CO, CO2, H2, and N2 within the estimation error. The alcohol production by C. autoethanogenum was even improved with the purified biogenic syngas compared to reference batch processes with the corresponding artificial syngas mixture. Both acetogens have proven their potential for successful fermentation processes with biogenic syngas, but full carbon conversion to ethanol is challenging with the investigated biogenic syngas.

Glioblastoma (GBM) is the most frequently occurring primary malignant brain tumor of astrocytic origin. To change poor prognosis, it is necessary to deeply understand the molecular mechanisms of gliomagenesis and identify new potential biomarkers and therapeutic targets. PIWI-interacting RNAs (piRNAs) help in maintaining genome stability, and their deregulation has already been observed in many tumors. Recent studies suggest that these molecules could also play an important role in the glioma biology. To determine GBM-associated piRNAs, we performed small RNA sequencing analysis in the discovery set of 19 GBM and 11 non-tumor brain samples followed by TaqMan qRT-PCR analyses in the independent set of 77 GBM and 23 non-tumor patients. Obtained data were subsequently bioinformatically analyzed. Small RNA sequencing revealed 58 significantly deregulated piRNA molecules in GBM samples in comparison with non-tumor brain tissues. Deregulation of piR-1849, piR-9491, piR-12487, and piR-12488 was successfully confirmed in the independent groups of patients and controls (all p < 0.0001), and piR-9491 and piR-12488 reduced GBM cells’ ability to form colonies in vitro. In addition, piR-23231 was significantly associated with the overall survival of the GBM patients treated with Stupp regimen (p = 0.007). Our results suggest that piRNAs could be a novel promising diagnostic and prognostic biomarker in GBM potentially playing important roles in gliomagenesis.

Abstract In our most recent study, we found that in Tanzania infection with Treponema pallidum (TP) subsp. pertenue (TPE) is present in four different monkey species. In order to gain information on the diversity and epidemiological spread of the infection in Tanzanian nonhuman primates (NHP), we identified two suitable candidate genes for multi-locus sequence typing (MLST). We demonstrate the functionality of the MLST system in invasively and non-invasively collected samples. While we were not able to demonstrate frequent interspecies transmission of TPE in Tanzanian monkeys, our results show a clustering of TPE strains according to geography and not host species, which is suggestive for rare transmission events between different NHP species. In addition to the geographic stability, we describe the relative temporal stability of the strains infecting NHPs and identified multi-strain infection. Differences between TPE strains of NHP and human origin are highlighted. Our results show that antibiotic resistance does not occur in Tanzanian TPE strains of NHP origin.

O entrevistado, Prof. Dr. Vilson Leffa, foi escolhido por seu extenso conhecimento na área, trabalhando com o desenvolvimento de matérias de ensino, especialista que é em questões acerca do ensino/aprendizagem de línguas. Seu currículo o confirma como indicado para examinar questões sobre a qualidade do ensino a distância, modalidade que vem crescendo continuamente no país.

Abstract Background Incidence rates of advanced cancer stages are important, e.g., for monitoring cancer screening programs. However, information from cancer registries on tumor stage is often incomplete. Exemplified by colorectal cancer (CRC), we explored the potential of German claims data to estimate incidence rates of advanced cancer stages. Methods We used claims data of the German Pharmacoepidemiological Research Database (GePaRD; information on > 20 million persons) to identify incident patients with advanced CRC based on ICD-10 codes for CRC and secondary malignant neoplasms. We calculated annual age-standardized incidence rates (ASIRs) of advanced CRC per 100,000 for the years 2008–2015 stratified by the presence of affected lymph nodes only (C77) vs. distant metastases (C78-C79) and compared them to ASIRs determined using data (2008–2014) from the German Centre for Cancer Registry Data (ZfKD). Results In GePaRD, the ASIRs of advanced CRC per 100,000 in 2014 were 21.5 among men and 14.9 among women. Compared to ZfKD data the ASIR in GePaRD was 2.58 lower in men and 0.27 higher in women (per 100,000) in 2014. Stratification by presence of distant metastases showed divergent patterns: the ASIRs regarding distant metastases were ~ 50% (women) and ~ 30% (men) higher, and the ASIRs regarding affected lymph nodes only were ~ 40% lower in GePaRD as compared to ZfKD. Conclusion While ASIRs of advanced CRCs overall agreed well between claims and cancer registry data in 2014, the analyses stratified by presence of distant metastases showed differences. Cancer registries might underestimate ASIRs of CRCs with distant metastases.

Slice energy spread is one of the key parameters in free electron laser optimizations, but its accurate measurement is not straightforward. Two recent studies from high energy (≥100 MeV) photoinjectors at SwissFEL and European x-ray free electron laser (XFEL) have reported much higher slice energy spread than expected at their XFEL working points (200–250 pC). In this paper, a new method for measuring slice energy spread at lower beam energy (∼20 MeV) is proposed and demonstrated at the PhotoInjector Test facility at DESY Zeuthen, and the results for 250 pC are much lower than those measured at high energy injectors.

Dactylorhiza majalis is a threatened indicator species for the habitat quality of nutrient-poor grassland sites. Environmentalists utilize the species to validate the success of conservation efforts. Conventionally, plant surveys are field campaigns where the plant numbers are estimated and their spatial distribution is either approximated by GPS or labor-intensively measured by differential GPS. In this study, we propose a monitoring approach using multispectral drone-based data with a very high spatial resolution (~3 cm). We developed the magenta vegetation index to enhance the spectral response of Dactylorhiza majalis in the drone data. We integrated the magenta vegetation index in a random forest classification routine among other vegetation indices and analyzed feature impact on model decision making using SHAP. We applied an image object-level median filter to the classification result to account for image artefacts. Finally, we aggregated the filtered result to individuals per square meter using an overlaying vector grid. The SHAP analysis showed that magenta vegetation index had the highest impact on model decision making. The random forest model could reliably classify Dactylorhiza majalis in the drone data (F1 score: 0.99). We validated the drone-derived plant count using field mappings and achieved good results with an RMSE of 12 individuals per square meter, which is within the error margin stated by experts for a conventional plant survey. In addition to abundance, we revealed the comprehensive spatial distribution of the plants. The results indicate that drone surveys are a suitable alternative to conventional monitoring because they can aid in evaluating conservation efforts and optimizing site-specific management.

Introduction: Biologics were approved for the treatment of advanced colorectal cancer (CRC) based on favorable benefit-risk-assessments from randomized controlled trials (RCTs), but evidence on their use in the real-world setting is scarce. Based on descriptive analyses we therefore aimed to assess characteristics and survival of CRC patients treated with biologics using large healthcare databases from three European countries (Netherlands, Italy, Germany).Methods: We included CRC patients treated with a biologic in 2010 or 2014 and characterized them regarding age, sex, comorbidities, and absolute survival.Results: Among 4,758 patients, the mean age ranged from 64.8 to 66.8 years, the majority was male, and comorbidities used as exclusion criteria in RCTs were coded in up to 30% of these patients. The proportion of bevacizumab users decreased between 2010 (72–93%) and 2014 (63–85%). In 2014, the absolute 12-month survival in new users was 64% (95% CI 51–77%), 56% (30–80%), and 61% (58–63%) in the Dutch, Italian, and German database, respectively, varying by age and comorbidity.Conclusions: Our study suggests that in the real-world setting, CRC patients treated with biologics are older and less selected regarding comorbidities compared to patients in RCTs, potentially explaining the relatively low 12-month survival we found. Treatment decisions in the real-world setting may require careful evaluation given that the risk-benefit ratio may vary depending on age and co-existing conditions.

Marine litter is a global problem that requires soon management and design of mitigation strategies. Marine litter monitoring is an essential step to assess the abundances, distributions, sinks and hotspots of pollution as well as the effectiveness of mitigation measures. However, these need to be time and cost-efficient, fit for purpose and context, as well as provide a standardized methodology suitable for comparison among surveys. In Europe, the Marine Strategy Framework Directive (MSFD) provides a structure for the effective implementation of long-term monitoring. For beaches, the well-established 100 m OSPAR macrolitter monitoring exists. However, this method requires a high staff effort and suffers from a high spatio-temporal variability of the results. In this study, we test the potential of aerial drones or Unmanned Aerial Vehicles (UAVs) together with a Geographic Information System approach for semi-automatic classification of meso- (1–25 mm) and macrolitter (>25 mm) at four beaches of the southern Baltic Sea. Visual screening of drone images in recovery experiments (50 m2 areas) at 10 m height revealed an accuracy of 99%. The total accuracy of classification using object-based classification was 45–90% for the classification with four classes and 50–66% for the classification with six classes, depending on the algorithm and flight height used. On 100 m beach monitoring transects the accuracy was between 39–74% (4 classes) and 25–74% (6 classes), with very low kappa values, indicating that the GIS classification method cannot be regarded as a reliable method for the detection of litter in the Southern Baltic. In terms of cost-efficiency, the drone method showed high reproducibility and moderate accuracy, with much lower flexibility and quality of data than a comparable spatial-OSPAR method. Consequently, our results suggest that drone based monitoring cannot be recommended as a replacement or complement existing methods in southern Baltic beaches. However, drone monitoring could be useful at other sites and other methods for image analysis should be tested to explore this tool for fast-screening of non-accessible sites, fragile ecosystems, floating litter or heavily polluted beaches.

Coronavirus disease 19 (COVID-19), caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), first emerged in late 2019 and has since rapidly become a global pandemic. SARS-CoV-2 infection causes damages to the lung and other organs. The clinical manifestations of COVID-19 range widely from asymptomatic infection, mild respiratory illness to severe pneumonia with respiratory failure and death. Autopsy studies demonstrate that diffuse alveolar damage, inflammatory cell infiltration, edema, proteinaceous exudates, and vascular thromboembolism in the lung as well as extrapulmonary injuries in other organs represent key pathological findings. Herein, we hypothesize that GPR4 plays an integral role in COVID-19 pathophysiology and is a potential therapeutic target for the treatment of COVID-19. GPR4 is a pro-inflammatory G protein-coupled receptor (GPCR) highly expressed in vascular endothelial cells and serves as a “gatekeeper” to regulate endothelium-blood cell interaction and leukocyte infiltration. GPR4 also regulates vascular permeability and tissue edema under inflammatory conditions. Therefore, we hypothesize that GPR4 antagonism can potentially be exploited to mitigate the hyper-inflammatory response, vessel hyper-permeability, pulmonary edema, exudate formation, vascular thromboembolism and tissue injury associated with COVID-19.

Chronic HIV infection may exacerbate atherosclerotic vascular disease, which at advanced stages presents as necrotic plaques rich in crystalline cholesterol. Such lesions can catastrophically rupture precipitating myocardial infarct and stroke, now important causes of mortality in those living with HIV. However, in this population little is known about plaque structure relative to crystalline content and its chemical composition. Here, we first interrogated plaque crystal structure and composition in atherosclerotic SIV-infected macaques using non-linear optical microscopy. By stimulated Raman scattering and second harmonic generation approaches both amorphous and crystalline plaque lipid was detected and the crystal spectral profile indicated a cholesterol ester (CE) dominated composition. Versus controls, SIV+ samples had a greater number of cholesterol crystals (CCs), with the difference, in part, accounted for by crystals of a smaller length. Given the ester finding, we profiled HIV+ plaques and also observed a CE crystalline spectral signature. We further profiled plaques from Ldlr-/- mice fed a high fat diet, and likewise, found CE-dominate crystals. Finally, macrophage exposure to CCs or AcLDL induced auto-fluorescent puncta that co-stained with the LC3B autophagy sensor. In aggregate, we show that atheromatous plaques from mice, macaques and humans, display necrotic cores dominated by esterified CCs, and that plaque macrophages may induce autophagic vesicle formation upon encountering CCs. These findings help inform our knowledge of plaque core lipid evolution and how the process may incite systemic inflammation.

Os problemas de infraestrutura em armazéns de soja no Brasil são fatores que atrapalham a competitividade da cadeia. Tais fatores estão diretamente relacionados às perdas dos grãos no processo de armazenagem. O objetivo deste trabalho foi mapear as operações relacionadas ao processo de armazenagem da soja em uma cooperativa de grãos com o intuito de identificar etapas onde há maior incidência de perdas. Este estudo, de caráter aplicado e exploratório, fez uso do estudo de caso, desenvolvido com o auxílio de entrevistas semiestruturadas com colaboradores do setor, bem como observações diretas não-participantes para o levantamento dos dados. Como principais resultados destaca-se que as perdas dos grãos podem derivar de diversas operações inerentes ao processo, notadamente na movimentação dos grãos, no transbordo nas moegas, no processo da secagem e da própria armazenagem. Constatou-se também que para mitigar essas perdas são necessários esforços substanciais, e dependem, sobremaneira, de investimentos em ativos específicos.

Spinocerebellar ataxia type-1 (SCA1) is caused by an abnormally expanded polyglutamine (polyQ) tract in ataxin-1. These expansions are responsible for protein misfolding and self-assembly into intranuclear inclusion bodies (IIBs) that are somehow linked to neuronal death. However, owing to lack of a suitable cellular model, the downstream consequences of IIB formation are yet to be resolved. Here, we describe a nuclear protein aggregation model of pathogenic human ataxin-1 and characterize IIB effects. Using an inducible Sleeping Beauty transposon system, we overexpressed the ATXN1(Q82) gene in human mesenchymal stem cells that are resistant to the early cytotoxic effects caused by the expression of the mutant protein. We characterized the structure and the protein composition of insoluble polyQ IIBs which gradually occupy the nuclei and are responsible for the generation of reactive oxygen species. In response to their formation, our transcriptome analysis reveals a cerebellum-specific perturbed protein interaction network, primarily affecting protein synthesis. We propose that insoluble polyQ IIBs cause oxidative and nucleolar stress and affect the assembly of the ribosome by capturing or down-regulating essential components. The inducible cell system can be utilized to decipher the cellular consequences of polyQ protein aggregation. Our strategy provides a broadly applicable methodology for studying polyQ diseases.

Free-electron laser based x-ray facilities require high-brightness photoinjectors to provide low emittance electron beams at a fixed bunch charge. The emittance optimization in the injector determines the lowest achievable emittance. Based on experimental emittance optimization at the photoinjector test facility at DESY in Zeuthen, a space-charge affected emission regime is identified, in which the optimum transverse beam emittance is achieved and thus, the injector is routinely operated in this regime. An advanced modeling approach is proposed to consider a dynamic emission process in the simulation of injector beam dynamics, meanwhile allowing detailed studies of the impact of strong space-charge fields during emission on the slice formation of the emitted electron bunch at the cathode. As an application, the proposed approach is used to analyze the budget of the optimized transverse beam emittance. An interplay, taking place in the identified emission regime, between intrinsic cathode emittance and space-charge induced emittance is demonstrated. The resolved behavior by simulation is consistent with the corresponding measurement under practical operation conditions of interest. The obtained results are reported.

Surface protein CD20 serves as the critical target of immunotherapy in various B-cell malignancies for decades, however its biological function and regulation remain largely elusive. Better understanding of CD20 function may help to design improved rational therapies to prevent development of resistance. Using CRISPR/Cas9 technique, we have abrogated CD20 expression in five different malignant B-cell lines. We show that CD20 deletion has no effect upon B-cell receptor signaling or calcium flux. Also B-cell survival and proliferation is unaffected in the absence of CD20. On the contrary, we found a strong defect in actin cytoskeleton polymerization and, consequently, defective cell adhesion and migration in response to homeostatic chemokines SDF1α, CCL19 and CCL21. Mechanistically, we could identify a reduction in chemokine-triggered PYK2 activation, a calcium-activated signaling protein involved in activation of MAP kinases and cytoskeleton regulation. These cellular defects in consequence result in a severely disturbed homing of B cells in vivo.

Background and purpose: Acute pain during weekly radiotherapy (RT) to the head and neck is not well characterized. We studied dose-volume metrics and clinical variables that are plausibly associated with throat or esophageal pain as measured with a weekly questionnaire during RT. Materials and methods: We prospectively collected weekly patient-reported outcomes from 122 head and neck cancer patients during RT. The pain score for each question consisted of a four-level scale: none (0), mild (1), moderate (2), and severe (3). Univariate and multivariate ordinal logistic regression analyses were performed to investigate associations between both esophageal and throat pain and clinical as well as dosimetric variables. Results: In multivariate analysis, age was significantly associated with both types of pain, leading to odds ratio (OR) = 0.95 (p = 0.008) and OR = 0.95 (p = 0.007) for esophageal and throat pain, respectively. For throat pain, sex (OR = 4.12; p = 0.010), with females at higher risk, and fractional organ at risk (OAR) mean dose (OR = 3.30; p = 0.014) were significantly associated with throat pain. Conclusions: A fractional OAR mean dose of 1.1 Gy seems a reasonable cutoff for separating no or mild pain from moderate to severe esophageal and throat pain. Younger patients who received RT experienced more esophageal and throat pain. Females experienced more throat pain, but not esophageal pain. O presente estudo discute os efeitos da Lei 13.467/2017 na litigância perante a Justiça do Trabalho, a partir das ferramentas proporcionadas pela análise econômica do direito. Partindo da premissa de que a utilização excessiva do Judiciário compromete a celeridade e a qualidade da prestação jurisdicional, a adequação das regras processuais trabalhistas tornou-se socialmente desejável, para o fim de desincentivar a litigância frívola –ações com baixa probabilidade de êxito. Propondo adequar o antigo modelo de concessão do benefício da gratuidade de justiça, a Lei 13.467/2017 implementa a sistemática de dispensa provisória de despesas processuais, honorários advocatícios e honorários periciais decorrentes de sucumbência. Considerando que a decisão do autor pelo ajuizamento ou não do processo judicial passa pela ponderação dos riscos e probabilidades do resultado esperado, as alterações propostas pela reforma, de um modo geral, tornam a litigância mais racional na seara processual. Assume-se que a maximização das riquezas no modelo dá base a uma justiça distributiva e corretiva, reduzindo o excesso de litigância para, no fim, assegurar a tutela jurisdicional célere e efetiva aos direitos trabalhistas.

Millimetre-wave (mmW) imaging is an emerging technique for non-destructive testing. Since many polymers are transparent in this frequency range, mmW imaging is an attractive means in the testing of polymer devices, and images of relatively high resolution are possible. This contribution presents an algorithm for the precise imaging of arbitrarily shaped dielectric objects. The reconstruction algorithm is capable of automatically detecting the object's contour, followed by a material-sensitive reconstruction of the object's interior. As an example we examined a polyethylene device with simulated material defects, which could be depicted precisely.

Along the Baltic coastline of Germany, drifting vegetation and beach cast create overlays at the otherwise sandy or stony beaches. These overlays influence the morphodynamics and structures of the beaches. To better understand the influence of these patchy habitats on coastal environments, regular monitoring is necessary. Most studies, however, have been conducted on spatially larger and temporally more stable occurrences of aquatic vegetation such as floating fields of Sargassum. Nevertheless, drifting vegetation and beach cast pose a particular challenge, as they exhibit high temporal dynamics and sometimes small spatial extent. Regular surveys and mappings are the traditional methods to record their habitats, but they are time-consuming and cost-intensive. Spaceborne remote sensing can provide frequent recordings of the coastal zone at lower cost. Our study therefore aims at the monitoring of drifting vegetation and beach cast on spatial scales between 3 and 10 m. We developed an automated coastline masking algorithm and tested six supervised classification methods and various classification ensembles for their suitability to detect small-scale assemblages of drifting vegetation and beach cast in a study area at the coastline of the Western Baltic Sea using multispectral data of the sensors Sentinel-2 MSI and PlanetScope. The shoreline masking algorithm shows high accuracies in masking the land area while preserving the sand-covered shoreline. We could achieve best classification results using PlanetScope data with an ensemble of a random forest classifier, cart classifier, support vector machine classifier, naïve bayes classifier and stochastic gradient boosting classifier. This ensemble accomplished a combined f1-score of 0.95. The accuracy of the Sentinel-2 classifications was lower but still achieved a combined f1-score of 0.86 for the same ensemble. The results of this study can be considered as a starting point for the development of time series analysis of the vegetation dynamics along Baltic beaches.

In embedded applications that use neural networks (NNs) for classification tasks, it is important to not only minimize the power consumption of the NN calculation, but of the whole system. Optimization approaches for individual parts exist, such as quantization of the NN or analog calculation of arithmetic operations. However, there is no holistic approach for a complete embedded system design that is generic enough in the design process to be used for different applications, but specific in the hardware implementation to waste no energy for a given application. Therefore, we present a novel framework that allows an end-to-end ASIC implementation of a low-power hardware for time series classification using NNs. This includes a neural architecture search (NAS), which optimizes the NN configuration for accuracy and energy efficiency at the same time. This optimization targets a custom designed hardware architecture that is derived from the key properties of time series classification tasks. Additionally, a hardware generation tool is used that creates a complete system from the definition of the NN. This system uses local multi-level RRAM memory as weight and bias storage to avoid external memory access. Exploiting the non-volatility of these devices, such a system can use a power-down mode to save significant energy during the data acquisition process. Detection of atrial fibrillation (AFib) in electrocardiogram (ECG) data is used as an example for evaluation of the framework. It is shown that a reduction of more than 95% of the energy consumption compared to state-of-the-art solutions is achieved.

We present an algorithm for computing ice drift in the marginal ice zone (MIZ), based on partial shape recognition. With the high spatial resolution of Sentinel-1 and Sentinel-2 images, and the low sensitivity to atmospheric influences of Sentinel-1, a considerable quantity of ice floes is identified using a mathematical morphology method. Hausdorff distance is used to measure the similarity of segmented ice floes. It is tolerant to perturbations and deficiencies of floe shapes, which enhances the density of retrieved sea ice motion vectors. The PHD algorithm can be applied to sequential images from different sensors, and was tested on two combined image mosaics consisting of Sentinel-1 and Sentinel-2 data acquired over the Fram Strait; the PHD algorithm successfully produced pairs of matched ice floes. The matching result has been verified using shape and surface texture similarity of the ice floes. Moreover, the present method can naturally be extended to the problem of multi-source sea ice image registration.

This study aimed to present our case series of patients with early-stage cervical cancer undergoing simple trachelectomy (ST). Currently, radical trachelectomy is considered the most appropriate fertility-preserving procedure for the treatment of early-stage cervical cancer. However, there is increasing debate on the appropriate radicality of the surgery to preserve oncologic safety. Descriptive retrospective analysis of patient records and evaluation of questionnaires. 2 gynecologic oncologic centers, surgeries performed by one surgical team. 36 women with early-stage cervical cancer undergoing ST. Laparoscopic assisted simple vaginal trachelectomy. Demographic, histologic, fertility, and follow-up data of all patients who underwent ST between April 2007 and July 2021 were prospectively recorded and retrospectively analyzed. A total of 36 women (mean age: 28 years) underwent ST of whom 81% were nulliparous. Indications for ST were multifocal International Federation of Gynecology and Obstetrics stage IA1 (n = 30), stage IA1 L1 (n = 1), stage IA2 (n = 2), and stage IB1 (n = 3). Mandatory staging procedure was laparoscopic pelvic lymphadenectomy, including bilateral sentinel biopsy in 92% of the cases and systematic in 8%. Residual tumor was histologically confirmed in 8 specimens (22%); 18 women (50%) were seeking parenthood, and 13 succeeded (72%). There were 16 live births, all on term, with a median fetal weight of 3110 grams (2330–4420). One patient had a medical abortion owing to fetal congenital malformation. One pregnancy is ongoing. After a median follow-up of 91.5 months (9–174), all women are alive with no evidence of disease. ST represents a de-escalation compared with radical trachelectomy and provides excellent oncologic results with an outstanding fertility rate and obstetric outcome for patients with early cervical cancer. Feline coronavirus (FCoV) is a complex pathogen causing feline infectious peritonitis (FIP). Host genetics represents a factor contributing to the pathogenesis of the disease. Differential susceptibility of various breeds to FIP was reported with controversial results. The objective of this study was to compare the genetic diversity of different breeds on a panel of candidate genes potentially affecting FCoV infection. One hundred thirteen cats of six breeds were genotyped on a panel of sixteen candidate genes. SNP allelic/haplotype frequencies were calculated; pairwise FST and molecular variance analyses were performed. Principal coordinate (PCoA) and STRUCTURE analyses were used to infer population structure. Interbreed differences in allele frequencies were observed. PCoA analysis performed for all genes of the panel indicated no population substructure. In contrast to the full marker set, PCoA of SNP markers associated with FCoV shedding (NCR1 and SLX4IP) showed three clusters containing only alleles associated with susceptibility to FCoV shedding, homozygotes and heterozygotes for the susceptibility alleles, and all three genotypes, respectively. Each cluster contained cats of multiple breeds. Three clusters of haplotypes were identified by PCoA, two clusters by STRUCTURE. Haplotypes of a single gene (SNX5) differed significantly between the PCoA clusters.
