120. CRABP1-CaMKII-Agrnregulates the maintenance of neuromuscular junction in spinal motor neuron

Abstract

Cellular retinoic acid-binding protein 1 (CRABP1) binds retinoic acid (RA) specifically in the cytoplasm with unclear functions. CRABP1 is highly and specifically expressed in spinal motor neurons (MNs). Clinical and pre-clinical data reveal a potential link between CRABP1 and MN diseases, including the amyotrophic lateral sclerosis (ALS). We established a sequenced MN-muscle co-differentiation system to engineer an in vitro functional 3D NMJ model for molecular studies and demonstrated that CRABP1 in MNs contributes to NMJ formation and maintenance. Consistently, Crabp1knockout (CKO) mice exhibited an adult-onset ALS-like phenotype with progressively deteriorated NMJs, characterized with behavioral, EchoMRI, electrophysiological, histological, and immunohistochemical studies at 2–20-months old. Mechanistically, CRABP1 suppresses CaMKII activation to regulate neural Agrnexpression and downstream muscle LRP4-MuSK signaling, thereby maintaining NMJ. A proof-of-concept was provided by specific re-expression of CRABP1 to rescue Agrnexpression and the phenotype. This study identifies CRABP1-CaMKII-Agrnsignaling as a physiological pre-synaptic regulator in the NMJ. This study also highlights a potential protective role of CRABP1 in the progression of NMJ deficits in MN diseases.

Abstract

The biochemical processes of cell membranes are sensitive to the geometry of the lipid bilayer. We show how plasmonic “nanowells” provide label-free real-time analysis of molecules on membranes with detection of preferential binding at negative curvature. 123. Self-directed Transport on Nanostructured Plasmonic Sensors

Abstract

Analytical sensors using varying detection strategies have been widely and successfully employed for advances in areas such as drug discovery, disease diagnosis and study of biological systems. Many of these sensors utilize plasmonic metallic nanostructures which can concentrate electromagnetic fields in nanoscale regions leading to many fold enhancement in optical signal obtained from the molecules. They employ techniques including fluorescence, surface plasmon resonance (SPR)-based refractive index sensing, surface-enhanced Raman spectroscopy (SERS) and other forms of vibrational spectroscopy for molecular characterization. However, the performance of these devices relies on effective transport of the target molecules to these nanoscale detection sites. Guided transport is extremely important for fast detection in cases where the concentration of molecules is really low and for accurate measurements of protein–protein binding kinetics. In this chapter, we discuss nanostructured biosensing substrates which can spontaneously direct the flow of molecules in solution towards the sensing hotspots. These devices demonstrate improved detection sensitivity, while minimizing the limitations and complexity imposed upon the system. Additionally, they can trap biological particles such as organelles and liposomes on the sensor surface, facilitating on-chip analysis of single particles. This chapter discusses a few methods which have been utilized for concentration of molecules on plasmonic sensing surfaces, without the application of external power sources.

Abstract

[EN] In this paper, we introduce a novel energy-efficient dryer that uses hot air as a combined heating method of heat conduction and convection heat transfer. The dryer consisting of rotary cylinders and arched jackets is structurally different from conventional dryers. We designed and built a hybrid 4-cylinder dryer for sheet-form materials. The energy consumption to evaporate unit water is about 930 kcal/kg moisture from the experimental results of a hybrid 4-cylinder dryer. The hybrid 4-cylinders dryer has a compact structure with about 1/3 of the size of a conventional dryer for the drying of sheet-form materials.

Abstract

[EN] The purpose of this study is to analyze the changes of drying efficiency according to the inflow conditions of outside air into the drying equipment during the drying process in order to reduce the energy used in the drying process of sludge. We conducted the experiment using a vertical thin film dryer. Materials used for the experiment are sewage sludge. As a result of the study, higher drying efficiency was obtained in the case of outside air inflow than in the case of no outside air inflow. In addition, optimum condition of outside air inflow was derived.

Abstract

We present a wafer-scale array of resonant coaxial nanoapertures as a practical platform for surface-enhanced infrared absorption spectroscopy (SEIRA). Coaxial nanoapertures with sub-10 nm gaps are fabricated via photolithography, atomic layer deposition of a sacrificial Al2O3 layer to define the nanogaps, and planarization via glancing-angle ion milling. At the zeroth-order Fabry-Pérot resonance condition, our coaxial apertures act as a "zero-mode resonator (ZMR)", efficiently funneling as much as 34% of incident infrared (IR) light along 10 nm annular gaps. After removing Al2O3 in the gaps and inserting silk protein, we can couple the intense optical fields of the annular nanogap into the vibrational modes of protein molecules. From 7 nm gap ZMR devices coated with a 5 nm thick silk protein film, we observe high-contrast IR absorbance signals drastically suppressing 58% of the transmitted light and infer a strong IR absorption enhancement factor of 104∼105. These single nanometer gap ZMR devices can be mass-produced via batch processing and offer promising routes for broad applications of SEIRA.

Abstract

Acoustic plasmon modes tightly coupled between a two-dimensional material and another conducting layer can exhibit optical confinement not possible with conventional plasmons. Here, we investigate acoustic plasmons supported in a monolayer and multilayers of black phosphorus (BP) placed shortly above a conducting plate. In the presence of a conducting plate, the acoustic plasmon dispersion for the armchair direction is found to exhibit the characteristic linear scaling in the mid- and far-infrared regime while it largely deviates from that in the long-wavelength limit and near-infrared regime. For the zigzag direction, such scaling behavior is not evident due to relatively tighter plasmon confinement. Further, we demonstrate a novel design for an acoustic plasmon resonator that exhibits higher plasmon confinement and resonance efficiency than BP ribbon resonators in the mid-infrared and longer wavelength regime. The theoretical framework and new resonator designs studied here provide a practical route toward the experimental verification of acoustic plasmons in BP and open up the possibility to develop novel plasmonic and optoelectronic devices that can leverage its strong in-plane anisotropy and thickness-dependent band gap.

130. Sensitivity of resonance frequency in the detection of thin layer using nano-slit structures.

Abstract

We derive the formulas for the resonance frequencies and their sensitivity when the nano-slit structures are used in the detection of thin layers. For a thin layer with a thickness of |$H$| deposited over the nanostructure, we show quantitatively that for both single and periodic slit structures with slit aperture size |$\delta $|⁠ , the sensitivity of resonance frequency reduces as |$H$| increases. Specifically, the sensitivity is of order |$O(\delta /H)$| if |$H>\delta $| and of order |$O(1+\ln H/\delta)$| otherwise. The evanescent wave modes are present along the interface between the thin dielectric film and ambient medium above. From the mathematical derivations, it is observed that the sensitivity of the resonance frequency highly depends on the effect of evanescent wave modes on the tiny slit apertures.
