We have generated a monoclonal antibody against prostatic adenocarcinoma, PC-Ab (IgM) was derived from a fusion using the fresh prostatic tissue of adenocarcinoma as the immunogen. Initial screening was performed by enzyme-linked immunosorbent assay (ELISA) on the soluble fraction of the immunogen. The specific analysis was performed by the avidin-biotin-complex immunoperoxidase method on paraffin-embedded sections of normal, benign and malignant neoplastic tissues from the prostate and various organs. PC-Ab reacted with well differentiated adenocarcinoma (83.3%), moderately differentiated adenocarcinoma (92.2%), and poorly differentiated adenocarcinoma (90.2%), respectively. According to classification by stage, the reaction rates with stage T0, T1, T2, T3 and T4 were 77.3%, 80.0%, 95.2%, 91.3% and 92.9%, respectively but no significant differences in the stages were seen among these groups. PC-Ab reacted with the epithelium of the normal prostate and benign prostatic adenoma, and human fetal tissues. Molecular weight and isoelectric point of the antigen recognized by this PC-Ab was estimated to be 57,000 daltons and 7.0, respectively. These results indicate that PC-Ab reacts with the antigen associated with human prostatic adenocarcinoma.

14553 Background: The drug sensitivity of tumor cells is one of the key issues to explore in individualized 5-fluorouracil (5-FU) based chemotherapy for colorectal cancer patients. We reported that growth inhibition rate (IR) and area under the concentration curve (AUC) approximated to a logarithmic curve using the collagen gel droplet embedded culture-drug sensitivity test (CD-DST) in ASCO 2003 (#1283). We also reported that the individual 50% inhibitory area under the concentration curve (AUC IR50) could be obtained using the individual AUC-IR curve in ASCO 2006 (#13560). The aim of this study was to evaluate the achievement of the individual AUCIR50 in 5-FU based adjuvant chemotherapy for the establishment of individualized chemotherapy. Methods: Surgical specimen was obtained from resectable 33 CRC patients without any preoperative chemotherapy during 2001 to 2005. 5-FU based adjuvant chemotherapy was administered in all patients. CD-DST was performed under 6–9 different conditions. The individual AUCIR50 was obtained from the individual AUC-IR curve. The patients were divided into 2 groups, the achieved group and the non-achieved group. In the achieved group, total administered AUC was more than the individual AUCIR50 and in non-achieved group, the total administered AUC was less than the individual AUCIR50. Recurrence rates were evaluated from the 2 groups. Results: In all patients, the recurrence rates were 6.7% (1/15) in the achieved group and 27.8% (5/18) in the non-achieved group. In the colon cancer patients, the recurrence rates were 8.3% (1/12) in the achieved group and 10.0% (1/10) in the non-achieved group. In the rectal cancer patients, the recurrence rates were 0% (0/3) in the achieved group and 50.0% (4/8) in the non-achieved group. Conclusions: The recurrence rate of the achieved group was lower than that of the non-achieved group. Therefore, this study demonstrated that the achievement of the individual AUCIR50 could be a prerequisite of individualized 5-FU based adjuvant chemotherapy. A direct transformation from the g-BC1.6 phase to a diamondlike BC1.6 phase was obtained in a diamond-anvil cell at high temperature, 2230±140K, and high pressure, 45GPa. Such a transition has not been observed in the B–C system before. The recovered samples were examined using both synchrotron-based x-ray diffraction and confocal micro-Raman spectroscopy at ambient conditions. The lattice parameter of the diamondlike phase (3.5745A) is close to that of diamond. The measured visible laser-excited Raman spectra of a diamondlike BC1.6 phase reveal a peak at 1315cm−1 which is attributed to the LO mode. It is also found that the orthorhombic/hexagonal phase was formed under high pressure in the areas where heating was not enough to make transformation in the cubic phase.

The intra-generic relationships of the Australian genus Calotis, with various chromosome base numbers from x = 8 to x = 4, were examined by the comparison of nucleotide sequences of the complete ITS region of nuclear rDNA and of the matK gene of chloroplast DNA. Within a monophyletic Calotis, four lineages were identified. Reconstruction of ancestral states suggests that the chromosome base number for Calotis is x = 8. Dysploidal reductions in chromosome base number from x = 8 to x = 7 and from x = 8 to x = 5 or 4 have occurred independently at least three times. Lower base numbers of x = 7, 5, and 4 are found predominantly in the arid and semi-arid zone species of Central and Western Australia. Total karyotypic length (genome size) is greater in perennials than in annuals within the genus Calotis. The elaborated pappus and surface structures of cypsela, and life form of species seem to be homoplasous with multiple origins in the evolutionary history of the lineage.

The generic circumscription and intra-generic relationships of the genus Podolepis Labill., with various chromosome numbers from n = 12 to n = 3, were examined by sequences of the internal transcribed spacers (ITS) of nuclear ribosomal DNA and the matK gene of chloroplast DNA. The topology of the ITS tree for 17 species and the matK tree for 18 species of the genus Podolepis sensu Davis (1957) and Anderberg (1991) and 15 taxa from eight related genera (Anderberg 1989, 1991, 1994) are basically concordant. Except for P. georgei Diels andP. kendallii F.Muell., parsimony analyses support the monophyly of the genus Podolepis sensu Davis (1957) and Anderberg (1991). The genera of Asteridea Lindl. and Pterochaeta Steetz are sisters toPodolepis in the combined tree based on the ITS and matK sequences. Within the monophyletic clade of the genus Podolepis, three lineages are identified. The chromosome base number of x = 12 may be ancestral in the genus Podolepis. The dysploidal reduction in chromosome number from n = 12 to n = 10 and 9, from n = 12 to n = 8 and 7, and from n = 12 to n = 11 and 3 in three lineages, respectively, is the primary mode of chromosomal evolution in this genus. Total karyotypic length (= genome size) is much greater in perennials than in annuals within the genus Podolepis. The number of pappus bristles on outer female florets tends to decrease and they are absent in some annuals of this genus, while myxogenic cells on the pericarp become prominent.

Chromosome number determinations from 152 collections representing 42 genera and 106 species of the Australian Gnaphalieae and Plucheeae are reported. The chromosome numbers of 75 of these species have not been previously counted or differ from those previously reported for species. Chromosome numbers have been documented for the first time for 14 genera: Argyroglottis (n = 12), Cephalosorus (2n = 24), Decazesia (n = 14), Dielitzia (2n = 26), Eriochlamys (n = 14), Erymophyllum (n = 11 and 14), Gilruthia (n = 13), Leucochrysum (n = 9), Myriocephalus s. str. (n = 14, 2n = 24), Polycalymma s. str. (n = 14), Pterocaulon (n = 10), Pterochaeta (n = 12), Quinetia (2n – 24) and Sondottia (2n = 6). Remaining counts augment and agree with previously reported determinations. Some problems with generic delimitation and interpretation of chromosome data are outlined. There is an array of karyotypes within the Australian Gnaphalieae and dysploidy is widespread. Polyploidy has also played an important role in the evolution of some taxa. Evidence suggests that the base number for Australian Gnaphalieae is x = 14. This may be the base number for the entire tribe.

The cartilage thickness in both normal hips and in coxarthrosis secondary to hip dysplasia was examined using a computer model. The hip cartilage thickness was calculated in three dimensions from computed tomographic images of the hip joint. In normal hip joints, the anterior cartilage was thicker than the posterior cartilage. In hip joints affected by secondary coxarthrosis, the cartilage thinned as the disease progressed. Approximately 80% of patients demonstrated thinning of the anterior apical cartilage, resulting in a forward and upward displacement of the femoral head.

A high-voltage self-protected thyristor with a well structure formed in its p-base layer whose operation is based on avalanche breakdown is described. The device structure is simple and easy to fabricate compared to other avalanche-type devices. Numerical analyses and experiments demonstrate that the breakover voltage can be controlled by varying the well diameter and/or its depth. The breakdown voltage fluctuation of the device is 10% when the junction temperature is varied from 23 to 100 degrees C. The device is turned on safely at 7300 V. >

A high precision wavelength meter in the visible is described, which is based on a Fabry-Perot interferometer with several etalons of different resolution. The interference fringe pattern projected on a photo-diode array detector is computationally processed to give a stepwise refinement of the wavelength value to any adjusted accuracy. The present model intends to provide digital and real-time values of high precision wavelength for dye-laser spectroscopy, and to serve as a monitor or as a pilot for wavelength control of a dye-laser source of nanosecond pulses. The model is, therefore, designed with particular emphasis on its short-pulse capability and on-line mode of operation as well as on its high sensitivity and resolution. Some arrangements of essential necessity are involved therein, such as to avoid an errorneous wavelength readout for a noisy incidence of pulsed field. The ultimate accuracy of wavelength measurement is prescribed by the resolving power of the thickest etalon employed. As applied to the pulsed source, the model determines the wavelength to the accuracy of ±one part in 107 for even a single shot nanosecond incidence of a fraction of μJ energy. The design and performance are described in connection to pulsed dye-laser incidence.

Novel techniques to observe coherent transients are successfully demonstrated on microwave transitions of NH3, H2O, OCS, and O2. The relaxation rate constants in thev 2 excited state of NH3 are obtained by infrared experiments.

A full-automatic wavelength controled pulsed dye laser has been developed. With the control system, dye laser oscillations of nanosecond pulses are tuned to any wavelength in the visible or scanned in any sweep mode keeping the nearly transform-limited bandwidth of 0.45 GHz. A mechanical tuning of the wavelength is employed, such that the motion of three wavelength selectors is synchronized to give a smooth and precise wavelength control. The basic principle underlying the tuning scheme, various requirements to be met in system construction, and the practical performance of automatic wavelength control are described in this paper.

Histoautoradiography was done to evaluate cell turnover by measuring levels of DNA synthesis in the different anatomic areas of the kidney in aging (76 to 103 weeks of age) female F344/NCr rats after injection of tritiated thymidine (3H-thymidine). All areas showed increased 3H-thymidine labeling indices (LI), including cortical and medullary tubules and interstitium. The extent of increase was directly correlated with increasing severity of the nephropathy. Atrophic, degenerative, and hyperplastic cortical tubules in areas of nephropathy had LI eight to nine times greater than nonaffected areas and more normal kidneys. These observations suggest that the hyperplastic responses of renal tubules to the unknown etiologic agents of aging nephropathy may be similar to those of tubules responding to chemical toxins.

Turn-on phenomena of a light-activated thyristor are analyzed using a one-dimensional numerical model that consists of the full set of semiconductor device equations, including the optically generated current and the effect of the shorted emitter. Results indicate that the photocurrent I p is induced by photovoltaic effects of the n_{E}-p_{B} junction and that its flow is in the reverse direction of the anode current I A . Consequently, I p circulates through the p-base, the short path placed across the n_{E}-p_{B} junction and the n-emitter resulting in a contribution to the light triggering, in addition to that of I A caused by the conventional P_{B}-n_{E}-p_{E} transistor action. In order to confirm the predicted behavior of the photocurrents, light-triggered 4-kV devices were fabricated. The experimental results thus obtained confirm the conclusions drawn from the model analysis.

A computer controlled system is presented for a high-precision, time-domain measurement of molecular fluorescence induced by a pulsed dye laser field. In this system three intelligent functions are assembled by the system controller: they are an automatic wavelength control of pulsed dye laser to 0.45 GHz resolution, a digital wavelength meter of 10−7 precision, and a high-speed waveform digitizer with 10 ps inherent resolution. Then the system achieves a unique capability such as to record real-time data of fluorescence decay in the nanosecond regime under an on-line monitor and control of the laser wavelength to milliangstrom precision. The basic constitution and practical performance of the system are described with particular emphasis on its high precision and multi-task capability.

Transplantable renal adenocarcinoma can be readily induced in Wistar strain male rats by initiation with N-ethyl-N-hydroxy-ethylnitrosamine followed by promotion with beta-cyclodextrin. The transplantability rates of the tumors by s.c. inoculation in newborn rats were 33 and 50%, respectively, for tumors of the first and second passages, and 100% for both third and fourth passages. The transplantability rates were affected by route of inoculation; rates of 50 and 100% were observed for s.c. and i.p. inoculations, respectively. The growth rate of tumors induced by i.p. inoculation was 3-fold higher than that induced by s.c. injection. Macroscopically, most of the tumors grew in the s.c. tissue of inoculation sites. However, invasive growth of tumors in spleen, liver, stomach, peritoneum, and intestine were seen in 50% of the animals inoculated i.p.; metastatic cancers to lung were seen in 16%. Histologically, the tumors were well-differentiated adenocarcinomas composed of uniform cells resembling kidney tubular cells and appeared to be derived from normal kidney tissues. A 5-fold decrease in gamma-glutamyl transferase activity in tumor tissues was found as compared with that of nontumorous kidney tissues. Electrophoretic analysis of cellular proteins in polyacrylamide gels revealed that tumor tissues exhibited five new polypeptides with molecular weights of 81,000, 64,000, 59,000, 50,000, and 36,000 which were either lacking or undetectable in the nontumourous area and control kidney. In addition, protein banding patterns of transplantable renal tumor appeared to be more heterogeneous than those of primary kidney tumor.

The effect of 1000 ppm potassium perchlorate (KClO4), 1000 ppm potassium iodide (KI) or 1000 ppm propylthiouracil (PTU) in the diet on the development of thyroid tumors was studied histologically and biochemically in Wistar rats given a single ip injection of 280 mg of N-bis(2-hydroxypropyl)nitrosamine (DHPN) per 100 g body weight. Basal diet containing 100 ppm KClO4, 1000 ppm KI or 1000 ppm PTU was given for 19 weeks from week 2 to week 20. The incidence of thyroid adenomas at the end of week 20 of the experiment was 100% (20/20) in rats treated with DHPN followed by KClO4, 85% (17/20) in rats given DHPN followed by KI, 95% (19/20) in rats given DHPN followed by PTU, and 5% (1/20) in rats given DHPN alone. The incidence of thyroid cancers was 100% (20/20) in rats treated with DHPN followed by KClO4, 65% (13/20) in rats treated with DHPN followed by KI and 0% (0/20) in rats treated with DHPN followed by or not followed by PTU. Rats given KClO4, KI or PTU alone and untreated rats had no thyroid tumors. The mean values of TSH in serum were 2.94 +/- 0.79 ng/ml in rats treated with DHPN followed by KClO4, 9.40 +/- 16.0 ng/ml in rats treated with DHPN followed by KI and 60.94 +/- 20.60 ng/ml in rats treated with DHPN followed by PTU. It was confirmed that (1) KClO4, PTU and KI promote the development of thyroid tumor in rats treated with DHPN, (2) the promoting effect of KClO4 or KI is stronger than that of PTU and (3) the value of TSH in serum is not parallel to the promoting effect on the development of thyroid tumor.

The effect of trisodium nitrilotriacetate monohydrate [(Na3NTA X H2O) CAS: 18662-53-8] on development of urinary bladder tumors in rats initiated with N-butyl-N-(4-hydroxybutyl)nitrosamine [(BBN) CAS: 3817-11-6] was studied. Twenty-one male inbred W rats 6 weeks of age were given drinking water containing 500 ppm of BBN for 4 weeks and then put on diet containing 10,000 ppm of Na3NTA X H2O for 28 weeks. Na3NTA X H2O promoted the development of urinary bladder tumors in rats treated with BBN. The incidences of papilloma and transitional cell carcinomas in the urinary bladder were 90% (18/20) and 25% (4/20), respectively, in rats treated with BBN and then Na3NTA X H2O and 0 in those treated with BBN or Na3NTA X H2O alone. The incidence of papillary or nodular hyperplasia in week 32 was 100% (20/20) in rats treated with BBN and then Na3NTA X H2O and 61% (13/21) in rats treated with BBN only.

Subcutaneous injection of DL-serine increased the number and size of renal tubular cell tumors in male W rats treated with 500 or 1,000 ppm N-ethyl-N-hydroxyethylnitrosamine [(EHEN) CAS: 13147-25-6, 2-(ethylnitrosamino)ethanol]. At the end of the 32-week experiment, the incidences of renal tumors were 95% in rats treated with 1,000 ppm EHEN for 2 weeks and then given three sc injections of DL-serine every 2 weeks, 33% in rats treated with 1,000 ppm EHEN for 2 weeks, and 28% in rats treated with 500 ppm EHEN for 2 weeks and then given three sc injections of DL-serine every 2 weeks. No renal tumors were found in rats treated with 500 ppm EHEN alone or given three sc injections of DL-serine alone every 2 weeks.

The effects of sodium chloride and alcohol on esophageal carcinogenesis induced by N-nitrosopiperidine (NNP) were studied in male F-344 rats. Sodium chloride given concomitantly with NNP during the first 8 weeks of the experiment and followed by sodium chloride increased the incidence of esophageal tumors in week 20. Sodium chloride given after NNP during the second 12 weeks of the experiment increased the incidence of esophageal hyperplasia. In the present experiment, no clear effect of alcohol on esophageal carcinogenesis was demonstrated, but alcohol slightly increased the incidence of preneoplastic esophageal tumors in groups given alcohol with and after NNP. These results suggest that sodium chloride may be able to be a potent promoter of the development of esophageal tumors in rats if the conditions of initiation are appropriate. Examination of the incidences of tumors in 9 segments of the esophagus gave no evidence of any portion that was especially susceptible to induction of esophageal tumors.

The effect of trisodium nitrilotriacetate monohydrate [N,N-bis(carboxymethyl)glycine trisodium salt] (Na3NTA X H2O) on development of renal tubular cell tumors induced with N-ethyl-N-hydroxyethylnitrosamine [CAS:13147-25-6; 2-(ethylnitrosamino)-ethanol] (EHEN) was studied. Six-week-old male inbred W rats were given a diet containing 1,000 ppm of EHEN for 2 weeks and then a diet containing a high (10,000 ppm) or low (500 ppm) concentration of Na3NTA X H2O for 30 weeks. The rats were killed during week 32. The higher concentration of Na3NTA X H2O enhanced the development of renal tubular cell tumors and increased the number and size of tumors in rats treated with EHEN, but the lower concentration of Na3NTA X H2O did not. The incidence of renal tubular cell tumors in week 32 was 33% in rats treated with 1,000 ppm EHEN for 2 weeks, 100% in rats treated with 1,000 ppm EHEN for 2 weeks plus high Na3NTA X H2O diet for 30 weeks, and 39% in rats treated with 1,000 ppm EHEN for 2 weeks and then given low Na3NTA X H2O diet for 30 weeks. Numbers of atypical cell foci per kidney area (No./cm2) were 17.0 +/- 7.6 in rats treated with EHEN and high Na3NTA X H2O, 7.3 +/- 2.2 in rats treated with EHEN and low Na3NTA X H2O, 3.7 +/- 1.4 in rats treated with EHEN alone, and 1.0 +/- 2.4 in rats treated with high Na3NTA X H2O diet alone. Atypical cell foci retained a tubular pattern and consisted of basophilic cells with a large nucleus or clear cells with a small nucleus.

Studies were made on the dose and sex dependence of thyroid tumor development in rats pretreated with N-bis(2-hydroxypropyl)nitrosamine (DHPN) followed by exposure to various doses of phenobarbital (PB). A direct dose-response relationship in induction of thyroid tumors was found in both male and female rats. Upon feeding the DHPN-treated rats with basal diet containing 20, 100, 500, and 2500 ppm of PB, the incidences of follicular adenoma were, respectively, 8, 45, 70, and 66% in male rats and 12, 17, 50, and 58% in female rats. Development of papillary adenomas in male rats was observed only at the higher doses of PB, at incidences of 12 and 20% for doses of 500 and 2500 ppm. Follicular carcinoma was also seen at higher doses of PB, at 16 and 12%, respectively, for the 500- and 2500-ppm groups. Neither follicular nor papillary carcinomas were induced in female rats; only a low incidence of papillary adenoma (4%) was observed with a PB concentration as high as 2500 ppm. A single injection of DHPN resulted in production of approximately 1 tumor/female rat and 2.5 tumors/male rat. DHPN combined with posttreatment with PB at doses up to 500 ppm did not increase tumor yield in female rats, whereas a 3-fold increase was observed in male rats for the 500-ppm-treated groups. When PB was increased to 2500 ppm a marked increase (8-fold) in tumor yield in male rats was observed, in contrast to a less than 3-fold increase in similarly treated female rats.

A new type of light activated bi-directional thyristor device, which is well suited for control of full wave ac power, is presented. This device consists of two thyristors connected in inverse parallel and one or two light emitting diodes provided between the thyristors, which are optically coupled so that either thyristor can be triggered by the light signal from the LED. It has an improved commutating dv/dt characteristic of 100 ν/μs and has ratings of 1,000 V for blocking voltage and 100 A for current carrying capability.

Injection (sc) of beta-cyclodextrin (beta-C) increased the number and size of renal tubular cell tumors in inbred Wistar (W) rats treated with 1,000 ppm of N-ethyl-N-hydroxyethylnitrosamine (EHEN). The incidence of renal tumors at the end of the 32-week experiment was 50% in rats treated with 1,000 ppm EHEN for 2 weeks and 100% in rats treated with 1,000 ppm EHEN for 2 weeks and then given daily sc injections of beta-C for 1 week. The incidence of renal tumors more than 3 mm in diameter was 70% in rats treated with 1,000 ppm EHEN before beta-C but 0% in rats treated with EHEN alone. In addition, beta-C promoted the development of renal tumors in rats treated with 500 ppm EHEN, which is a subthreshold dose for renal tubular cell tumorigenesis. These results show that beta-C promotes EHEN-induced renal tubular cell tumorigenesis.

In trochanteric fractures of the femur it is difficult to know the exact type and location of fracture lines from conventional radiography. We therefore made a program to reconstruct a three-dimensional image from CT figures and observed closely thirty-six fractures to obtain the following results. These fractures were basically composed of three major fracture lines, the anterior, the posterior and the lateral. The posterior fracture lines were seen in all cases in the same location, a shallow groove located just below the trochanteric fossa and extending to the upper indented portion of the minor trochanter (the paratrochanteric groove). The posterolateral destruction could be observed more frequently than in conventional radiography. From these observations, we have suggested a new anatomical classification based on the contact areas of the proximal and distal fragments classifying trochanteric fractures into five types.

Thymic T-cell lymphomas were induced by administration of nitroso-2-hydroxypropylurea (NHPU) at a dose of 1.6 mg by gavage twice weekly for each of 20 weeks in F344/NCr rats. Lymphomas began to appear 5-10 weeks after the first intubation. Primary and transplantable tumors were subjected to morphological, histogenetic and immunological characterization. Lymphomas composed of medium-sized lymphocytes (prolymphocytes) arose within individual thymic lobules, often in atrophic thymic lobes and metastasized slowly to the T-cell zones of the splenic white pulp. The tissue fixative was important for tumor morphology and classification, with Bouin's fluid markedly superior to formalin. Transplantable tumors were similar morphologically except for one lymphoblastic lymphoma, but all transplants metastasized quickly to spleen, liver, thymus, and other tissues. Fluorescence-activated cell sorter (FACS) analysis of primary and transplantable lymphomas revealed cells of the T-cell lineage, with tumor cells expressing both OX-8 (CD8) and W3/25 (CD4) antigens. Immunoperoxidase studies of spleen showed infiltration of OX-8+ tumor cells first into T-cell dependent areas of the splenic white pulp.

The effect of castration on the development of thyroid tumors was studied histologically and biochemically in Wistar rats given a single ip injection of 210 mg of N-bis (2-hydroxypropyl)-nitrosamine (DHPN) followed by phenobarbital (Pb). Castration was performed one week after, or one week before the injection of DHPN. The injection of DHPN was given at the end of the first week, and the rats were fed 500 ppm Pb in the basal diet for 38 weeks from week 3 to week 40. The incidence of thyroid adenomas and cancers was 20% (4/20) and 10% (2/20) in rats treated with DHPN alone. It was 75% (15/20) and 40% (8/20) in rats treated with DHPN and Pb; 30% (6/20) and 15% (3/20) in rats treated with DHPN and Pb, and castrated after DHPN; 20% (4/20) and 0% in rats treated with DHPN and Pb, and castrated before DHPN and Pb; and 0% and 0% in rats castrated either before or after receiving DHPN. Castration thus inhibited the development of thyroid tumor in rats treated with DHPN. The inhibition of tumor development in rats treated with DHPN and Pb, and castrated before receiving DHPN, was greater than in the rats castrated after receiving DHPN.
