The ideological paradigms of a complete nursing background; the professional quality one intends to prepare; the ethical, political, and technical commitment, and the complexity of the nursing work processes cause programs to rethink their teaching and adaptation to Brazilian National Curriculum Guidelines. The objective of this study was to identify in interview conversations with 32 undergraduate nursing students the importance attributed to management classes in their professional preparation and to correlate them to the guidelines' governing principles. This is a qualitative research developed as a case study. The data were collected through semi-structured interviews and submitted to Content Analysis. According to undergraduates, teaching administration must have a greater time load; more encompassing content; diversified strategies; reinforcement of skill development, especially ethical-political skills; and consideration of the union which exists between teaching, research, and community clinical work. According to undergraduates managerial skills deserve special attention in restructuring the nursing curriculum.

- Abstract
The ErbB2 receptor tyrosine kinase has been shown to play an important role in cancer cell motility and metastases formation. This receptor is often overexpressed in human tumors of diverse origins, including breast and ovarian cancer. Individuals with ErbB2 over expressing tumors have shown poor clinical outcome. Our studies are focused on signaling molecules that interact with autophosphorylated tyrosine residues of the cytoplasmic tail of the receptor. Two of the sites, Tyr 1201 (YC) and Tyr 1227 (YD) are fully able to restore the migratory phenotype of breast carcinoma cells. Studies of the functional role of ErbB2 phosphorylation sites identified PLCγ1 as an interacting partner of the YC autophosphorylation site, and Memo (Mediator of ErbB2-driven cell Motility) as a binding partner of the YD site that is required for ErbB2 induced cell motility. Memo is encoded by a unique gene that is found in all branches of life, from bacteria to humans. Memo has no characterized domains, nor does it have obvious catalytic activity. Various approaches were used to position Memo in a signaling pathway and to uncover its biochemical function. Memo was initially detected based on its important role in ErbB2- induced cell motility. In fact, tumor cells with a specific knock-down (KD) of Memo failed to grow microtubules in response to Heregulin (HRG)-induced ErbB2 activation and were impaired in their migration. Cell migration proceeds in distinct steps. In response to a chemotactic stimulus, cells extend protrusions at the front that help in attachment. This is followed by contraction of the cell body and tail detachment at the rear allowing movement in the direction of the ligand. The initial event in the process is sensing of the ligand in response to activation of cellular receptors like EGFR or ErbB2. Their activation initiates signaling pathways that lead to polymerization of new actin at the leading edge, which is necessary for generating the protrusive force allowing migration. An important goal of my thesis work has been to investigate the step(s) of the migratory process that require Memo. In the first study, we explored migration using Dunn chambers and analyzed the chemotactic response of tumor cells in a shallow gradient of ligand. By tracing HRG-stimulated cell migration in time-lapse video microscopy, we found that Memo or PLCγ1 KD strongly impairs cell directionality, reflecting an important role for Memo and PLCγ1 in orchestrating directional cell migration. We also demonstrated that depletion of Memo or PLCγ1 resulted in very similar phenotypes, with a strong impairment of HRG-induced cytoskeletal organization. To gain more insight into Memo’s function, we carried out a Yeast-2-Hybrid (YTH) analysis and found a number of interesting new partners of interaction for Memo. Of particular interest is the small protein cofilin, one of the major cellular actin severing and depolymerizing factors that is known to have an essential role in directional sensing during chemotaxis. This interaction was confirmed in vitro using recombinant proteins and in vivo in coimmunoprecipitation experiments where Memo was detected in complexes with cofilin, ErbB2 and PLCγ1. Interestingly, we also found that HRG-induced PLCγ1 phosphorylation was decreased in Memo KD cells, suggesting that Memo regulates PLCγ1 activation. Furthermore, by introducing GFP-tagged cofilin into control, Memo or PLCγ1 siRNA transfected breast tumor cells, we showed that HRG-induced recruitment of GFP-cofilin to lamellipodia is impaired in Memo- and in PLCγ1 KD cells, suggesting that both proteins lie upstream of cofilin in models of ErbB2-driven tumor cell migration. Finally, we examined the effect of Memo on cofilin binding and severing/depolymerizing properties. In vitro F-actin binding assays showed that Memo does not impair cofilin binding to F-actin, and revealed that Memo is a novel F-actin binding protein. In vitro F-actin depolymerization assays indicated that Memo promotes cofilin depolymerizing/severing activity. Altogether, these data suggest a novel role for Memo during the migratory process and its implication in the regulation of actin dynamics through cofilin binding. In the second study, we used two different Memo-defective cellular models to examine Memo’s function in more detail. We demonstrated that inhibition of Memo impairs activation of a number of signaling molecules including Src, Shc, ERK and PLCγ1. We also provide evidence that Memo interacts with the three Shc isoforms, p46shc, p52shc, and p66shc, and showed that Shc is required for Memo binding to the ErbB2 receptor. Control and Memodeficient cells were also scored for their migration and adhesion properties. These assays indicated that Memo is important in both cell migration and adhesion processes. Also, morphological and biochemical analyses of control and Memo-deficient cells suggested that Memo is involved in focal adhesion organization and rear cell deadhesion during the migratory process. Altogether, these two studies revealed important roles for Memo at different steps of cell migration and metastasis, making it a potential interesting target for cancer therapy. Genetic approaches in model organisms have been important for gaining insight into the function of evolutionarily conserved proteins. To position Memo within a genetic network, experiments in the model organism S. cerevisae that lends itself to rapid genetic screening were performed. We investigated cellular localization of Memo in yeast and found that Memo is located in the nucleus and cytoplasm of the cell. A S. cerevisae memo Δ strain has been generated and is viable. Considering the role of Memo in the microtubule and actin networks that we described in mammalian cells, we examined the memo Δ strain for defects in different cytoskeletal dynamics. No significant effect was observed. We also performed a Synthetic Lethal Screen of genetic interactions between a memo Δ strain and an ordered array of 4700 Yeast strains containing non-essential gene deletions. This analysis revealed a limited number of synthetic interactions. Lethality was observed in combination with the plc1Δ strain. PLC1 encodes for the unique isoform of phosphatidylinositol-specific phospholipase C of S. cerevisiae. The results are intriguing and exciting considering the data obtained in the mammalian models; in fact, we demonstrated that Memo and PLCγ1 interact with ErbB2 autophosphorylation sites and are essential for directional migration. We also showed that Memo is found in a complex with PLCγ1 and ErbB2 and that Memo is likely contributing to PLCγ1 activation. We hypothesize that in Yeast, Memo and PLC1 act in the same or in distinct but related pathways, and suggest that the connection between PLC and Memo induced-pathways is also conserved through evolution.

This work aims at studying the visual arts panorama in the State of Pará between the 1940’s and the 1980’s departing from local painter and ceramist Ruy Meira (1921-1995). Meira, a member of a distinguished family of politicians and intellectuals in Northern Brazil, played a pivotal role in the ripening of the artistic métier in Pará, having been fundamental to the establishment of many important artistic and intellectual sociability networks, and having synthesized, in his artistic trajectory, the history of the absorption of the main currents of modern art in Pará. In order to study Meira’s artistic and social personality, we searched for support in his personal archive, which contains some important material, most of it unpublished, ranging from active and passive correspondence to pictures and exhibitions catalogues.

MULTIPLE sclerosis , *PATIENT management , *GENE expression , *T cells , *CELL cycle , *NATALIZUMAB , *MATHEMATICAL models - Abstract
MicroRNAs (miRNAs) are a family of noncoding RNAs that play critical roles in the posttranscriptional regulation of gene expression. Accumulating evidence supports their involvement in the pathogenesis of multiple sclerosis (MS). Here, we compare miR-17 expressions in CD4+ T cells from relapsing-remitting (RR) MS patients treated with natalizumab versus untreated patients. miR-17 was downregulated under natalizumab treatment and upregulated during relapse, therefore supporting a possible role of miR-17 in MS immunopathogenesis. Downregulation of miR-17 was associated with upregulation of PTEN, BIM, E2F1, and p21 target genes. In vitro miR-17 inhibition was associated with upregulation of the same targets and resulted in impaired CD4+ T cell activation and proliferation. We further describe deregulated TGFBR2 expression in untreated patients versus healthy volunteers (HVs) and confirm in vitro the link between miR-17 and TGFBR2 expressions. These findings support an effect of natalizumab on expression of specific miRNA and subsequent expression of genes involved in proliferation and control of the cell cycle. [ABSTRACT FROM AUTHOR]

- Abstract

Introduction: Integrality is associated with the various dimensions of life, considering the tangible and subjective aspects related to the complex unity of the human being. Objective: To promote a critical reflection on the integrality of the human being, in the scope of care, as the essence of health practices. Method: This is a study developed as a reflective essay that approached care in an existential philosophical perspective, aiming at integrality for health promotion. Development of the theme: Human care, understood as much more than doing or providing care, is projected beyond duty and becomes care and compassion. It covers attitudes that should be part of the analysis and critical reflection of professionals working in the health area. Conclusion: The recovery of integrality in the health care provided during the practice requires a reflexive posture of the health professional in order to value all the dimensions of the human being, including the spiritual one. - Abstract
As far as we know, no report uses the Swenson transanal endorectal pull-through technique in an animal model. Our objective is to describe the use of this technique as an experimental model for training and research purposes. Ten Norfolk hybrid rabbits were randomly selected from our experimental laboratory, with a mean weight of 3539.3 (± 678.4) g. Neither colon preparation nor fast were used before the procedures. The surgical technique was based on the description performed by Levitt et al. (2013, J Pediatr Surg . 2013;48(11):2289-2295). Information related to the surgical procedures and the clinical evolution in the postoperative period were recorded and analyzed. There were no deaths or severe complications. The anesthetic and the surgical times were significantly higher for the first three animals of the experiment. Our animal model proved adequate to perform the transanal endorectal Swenson pull-through technique, allowing the training of surgical skills through a model similar to the human, with few anesthetic complications and good postoperative evolution, including postoperative follow-up. We believe that it will serve as a learning tool in many institutions that are continuously searching for improved new techniques and will support new researches in this area.

- Abstract
Therapeutic efficacy with durable responses has been demonstrated with several antibody drugs that block key immune checkpoint receptors, including PD-1, PD-L1, and CTLA-4. Despite the success of these drugs, a substantial proportion of patients do not benefit. Targeting multiple inhibitory pathways simultaneously to augment anti-tumor immunity has proven to be a promising approach. The emergence of Repulsive Guidance Molecule b (RGMb), a ligand for PD-L2, as a novel co-inhibitory pathway in T cells, together with its regulation by the gut microbiome, encouraged the discovery and development of fully human anti-RGMb antibodies. Here, we describe phage display-derived monoclonal antibodies (mAbs) 2C11 and 5C10 that bind human RGMb with high affinities of 1.4 nM and 0.72 nM, respectively. Both mAbs 2C11 and 5C10 potently inhibited RGMb interaction with PD-L2. MAb 2C11 effectively inhibited RGMb interaction with bone morphogenetic proteins 2 and 4 (BMP2-4), while leaving RGMb interaction with Neogenin 1 (Neo1) unaffected. Conversely, mAb 5C10 disrupted RGMb interaction with Neo1 while maintaining RGMb binding to BMP2-4. These findings map the 2C11 epitope at the membrane-distal N-terminal region of RGMb, which coincides with both PD-L2- and BMP2-4-binding sites. The PD-L2 binding interface is likely positioned between RGMb's N-terminal BMP-binding and C-terminal Neo1-binding regions. The in vivo activity of mAb 2C11 in combination with anti-PD-1 or anti-PD-L1 was tested in MC38 and B16-OVA cancer models and demonstrated synergistic effects by significantly enhancing anti-tumor responses. These properties make mAb 2C11 a promising candidate for therapeutic use to overcome immune checkpoint inhibitor resistances, warranting further exploration in clinical settings.

- Abstract
Objectives: To assess messenger RNA (mRNA) expression of POU2AF1 and Spi-B and their potential regulatory microRNAs (miRNAs) in natalizumab-treated patients with multiple sclerosis and in therapy-associated progressive multifocal leukoencephalopathy (PML)., Methods: Expression of POU2AF1/Spi-B was analyzed by using real-time reverse transcription PCR assays on isolated B/CD8(+) T lymphocytes and peripheral blood mononuclear cells (PBMCs) from cohorts of untreated and natalizumab-treated patients with and without PML. Longitudinal expression analysis was performed on CD4(+), CD8(+) T and B cells from 14 patients who interrupted natalizumab therapy for 8 weeks. The miRNA profiling was conducted in PBMCs from 5 untreated and 5 natalizumab-treated patients using low-density arrays followed by validation with single miRNAs assays in untreated and natalizumab-treated patients., Results: POU2AF1 and Spi-B mRNAs were upregulated in B and CD8(+) T cells from natalizumab-treated patients, which was validated in PBMCs from different cohorts of natalizumab-treated patients with and without PML, with a noteworthy higher expression of Spi-B in patients with PML. In contrast, downregulation of POU2AF1/Spi-B expression was measured in B and CD8(+) T cells after natalizumab discontinuation. Seventeen differentially expressed miRNAs including miR-10b, a regulator of POU2AF1 mRNA, were identified in long-term natalizumab-treated patients compared with untreated ones., Conclusions: Upregulation of POU2AF1 and Spi-B, known transactivators of the JC virus, the causative agent for PML, and its association with occurrence of PML in natalizumab-treated patients, corroborates POU2AF1/Spi-B as potential biomarkers for PML risk, which merits further evaluation.

- Abstract
Background: MicroRNAs (miRNAs) have emerged as a family of post-transcriptional regulators of gene expression that mediate diverse aspects of immunity. MiRNA dysregulation has been found in multiple sclerosis (MS), reflecting the growing need to identify disease-specific miRNA expression signatures. Our previous low-density array studies reveal differential miR-126 expression in the CD4(+)T cells of untreated relapsing-remitting MS (RRMS) patients. Here, we investigated miR-126 expression in natalizumab-treated patients., Methods: We isolated CD4(+) T cells from untreated (n = 12) and natalizumab-treated MS patients (n = 24), and from healthy volunteers (n = 12). We analyzed the expression of miRNAs and potential targets by real time reverse transcription polymerase chain reaction (RT-PCR). We assessed specific inhibition of miR-126, in vitro., Results: MiR-126 was down-regulated in cells of patients under natalizumab treatment and up-regulated during relapse, supporting a regulatory role in MS immunopathogenesis. MiR-126 expression correlated with the expression of POU2AF1, a regulator of Spi-B that binds to the promoter/enhancer sequences of JC virus (JCV), the pathogen of progressive multifocal leukoencephalopathy (PML), a rare complication of natalizumab treatment. The same trend was found for Spi-B. Strong up-regulation of both genes appeared to be treatment duration-dependent. Specific inhibition experiments supported the link between the expression of miR-126 and POU2AF1/Spi-B., Conclusions: Our findings provided deeper insight into the mode of action of natalizumab, with possible implications for understanding both the effects of natalizumab on MS activity and its specific adverse event profile., (© The Author(s) 2014.)

- Abstract
Every day, college graduates face complex situations that make them check skills developed during the course in comparison with the skills needed at the professional level. This experience helps them evaluate the adequacy of the course's teaching structure and recover intervening aspects of this process. This text discusses the Evaluation of College Courses, recovering concepts in the field of Education Evaluation; describes the practices developed in Brazil, directed towards course evaluation, based on the graduate's perspective. The following descriptors were used: college graduates, indicators, evaluation and graduation available in the databases: PeriEnf, BDEnf, LILACS and DEDALUS. The analysis of the surveyed studies highlights the need to consider the opinion of college graduates in the curriculum proposals with a view to implementing an educational process with emancipating and transforming characteristics.
