31st European Photovoltaic Solar Energy Conference and Exhibition; 713-715, We present a novel process scheme for emitter patterning for IBC-SHJ solar cells, employing laser based single step patterning of a dielectric hard mask, and self-alignment for further depositions. With the presented process sequence, we aim at developing a simple, lithography free and contactless emitter patterning method for industrial applications. Laser writing of the hard mask is performed using a picosecond laser operating at 355 nm to reduce the laser damage at the surface of the samples, which then allows for short damage etch durations. We demonstrate the feasibility of this process scheme for IBC-SHJ solar cells. Proof-of-concept cells on planar substrates show a short circuit current density of 33.2 mA/cm2, but still suffer from both low open circuit voltage and low fill factor, the latter being mainly due to a high series resistance. At the current stage of development, analysis of the etched contact groove reveals a rough surface in the overlap zone of the laser pulses, which very likely causes recombination and reduces both open-circuit voltage and fill factor.

28th European Photovoltaic Solar Energy Conference and Exhibition; 2005-2008, In this paper wet anisotropic texturing and subsequent smoothing of c-Si wafers, 350 and 100 μm thick, are investigated for reducing light reflection and improving light confinement within silicon substrate suitable for amorphous/ crystalline silicon heterojunction solar cells. The influence of time of the etching process used in the smoothing procedure is studied. To this aim reflectance measurements and scanning electron microscopy images are used to evaluate the surface morphology after the chemical etching.

The amorphous/crystalline heterojunction promises the goal, considering the world record efficiency targeted by Panasonic. Transparent conducting oxides (TCOs) play an essential role in modern optoelectronic devices through their combination of electrical conductivity and optical transparency. TCO, plays a key role to enhance the HJ cell efficiency. We have studied and characterized the properties of a new trilayer AZO/Ag/AZO as TCO in the c-Si heterojunction solar cell, whose thickness and composition combination ensure also an antireflection coating role. The influence of Ag layer thickness on electrical and optical properties of the tri-layer film was investigated. Several analytical tools such as fourpoint probe, scanning electron microscopy (SEM) and ultraviolet–visible–near infrared (UV–vis–NIR) spectrophotometer were used to explore the causes of the changes in optoelectronic properties and surface micrographs. The highest value is 2.5×10−4Ω cm for the AZO (100nm) /Ag (8nm) /AZO( 50nm) tri-layer film, and the sheet resistance is 15.5 Ω/sq, while the average transmittance is above 78% in the visible light region (400–800 nm).

In particular, the ferric bis- histidyl adduct represents a common accessible ordered state for the ? chains of all tetrameric Hbs isolated from Antarctic and sub-Antarctic fish. Indeed, the crystal structures of known tetrameric Hbs in the bis-His state are characterized by a different binding state of the ? and ? chains. An overall analysis of the bis-histidyl adduct of globin structures deposited in the Protein Data Bank reveals a marked difference between hemichromes in tetrameric Hbs compared to monomeric/dimeric Hbs. Herein, we review the structural, spectroscopic and stability features of hemichromes in tetrameric Antarctic fish Hbs. The role of bis-histidyl adducts is also addressed in a more evolutionary context alongside the concept of its potential physiological role.

Bovine seminal ribonuclease (BS-RNase) is a peculiar member of the pancreatic-like ribonuclease superfamily endowed with unique biological functions. It has been shown that native BS-RNase is a mixture of two distinct dimeric forms. The most abundant form is characterised by the swapping of the N-terminal helix. Kinetic studies have shown that this dimer is allosterically regulated, whereas the minor component, in which no swapping occurs, exhibits typical Michaelian kinetics. In order to correlate the catalytic properties with the structural features of BS-RNase, we have determined the crystal structure of the BS-RNase swapping dimer complexed with uridylyl(2'-5')guanosine. The structure of the complex was refined to an R value of 0.189 at 1.9 A resolution. Surprisingly, the enzyme binds four dinucleotide molecules, all in a non-productive way. In the two active sites, the guanine base is located in the subsite that is specific for pyrimidines. This unusual binding has been observed also in complexes of RNase A with guanine-containing nucleotides (retro-binding). One of the two additional dinucleotide molecules bound to the enzyme is located on the surface of the protein in a pocket generated by crystal packing; the second was found in a cavity at the interface between the two subunits of the swapping dimer. There are indications that the interface site plays a role in the allosteric regulation exhibited by BS-RNase. This finding suggests that domain swapping may not merely be a mechanism that proteins adopt for the transition from a monomeric to oligomeric state but can be used to achieve modulations in catalytic function.

The blood of the sub-Antarctic fish Eleginops maclovinus (Em) contains three haemoglobins. The major haemoglobin (Hb1Em) displays the Root effect, a drastic decrease in the oxygen affinity and a loss of cooperativity at acidic pH. The carbomonoxy form of HbEm1 has been crystallized in two different crystal forms, orthorhombic (Ortho) and hexagonal (Hexa), and high-resolution diffraction data have been collected for both forms (1.45 and 1.49 Å resolution, respectively). The high-frequency resonance Raman spectra collected from the two crystal forms using excitation at 514 nm were almost indistinguishable. Hb1Em is the first sub-Antarctic fish Hb to be crystallized and its structural characterization will shed light on the molecular mechanisms of cold adaptation and the role of the Root effect in fish haemoglobins.

Fishes of the perciform suborder Notothenioidei provide an excellent opportunity for studying the evolution and functional importance of evolutionary adaptations to temperature. To understand the unique biochemical features of high-Antarctic notothenioids, it is important to improve our knowledge of these highly cold-adapted stenotherms with new information on their sub-Antarctic relatives. This paper focuses on the oxygen-transport system of two non-Antarctic species, Eleginops maclovinus and Bovichtus diacanthus. Unlike most Antarctic notothenioids, the blood of E. maclovinus and B. diacanthus displays high hemoglobin (Hb) multiplicity. E. maclovinus, the sister group of Antarctic notothenioids, has one cathodal (Hb C) and two anodal components (Hb 1, Hb 2). B. diacanthus, one of the most northern notothenioids, has three major Hbs. The multiple Hbs may have been maintained as a response to temperature differences and fluctuations of temperate waters, much larger than in the Antarctic. Although non-Antarctic notothenioids have never developed cold adaptation, the amino-acid sequence reveals high identity with the globins of Antarctic notothenioids. Hbs of sub-Antarctic notothenioids are characterised by high oxygen affinity and Root effect. Phylogenetic analyses are consistent with the hypothesis that Bovichtidae and Eleginopidae diverged after they became established in more temperate waters north of the Antarctic Polar Front.

The dominant perciform suborder Notothenioidei is an excellent study group for assessing the evolution and functional importance of biochemical adaptations to temperature. The availability of notothenioid taxa in a wide range of latitudes (Antarctic and non-Antarctic) provides a tool to enable identification of physiological and biochemical characteristics gained and lost during evolutionary history. Non-Antarctic notothenioids belonging to the most basal families are a crucial source for understanding the evolution of hemoglobin in high-Antarctic cold-adapted fish. This paper focuses on the structure, function and evolution of the oxygen-transport system of Cottoperca gobio, a sub-Antarctic notothenioid fish of the family Bovichtidae, probably derived from ancestral species that evolved in the Antarctic region and later migrated to lower latitudes. Unlike most high-Antarctic notothenioids, but similar to many other acanthomorph teleosts, C. gobio has two major hemoglobins having the b chain in common. The oxygenbinding equilibria and kinetics of the two hemoglobins have been measured. Hb1 and Hb2 show strong modulation of oxygen-binding equilibria and kinetics by heterotropic effectors, with marked Bohr and Root effects. In Hb1 and Hb2, oxygen affinity and subunit cooperativity are slightly higher than in most high-Antarctic notothenioid hemoglobins. Hb1 and Hb2 show similar rebinding rates, but also show significant dynamic differences that are likely to have functional consequences. Molecular dynamic simulations of C. gobio Hb1 were performed on the dimeric protein in order to obtain a better understanding of the molecular basis of structure D function relationships.

Tetrameric hemoglobins represent the most commonly used model for the description of the basic concepts of protein allostery. The classical stereochemical model assumes a concerted transition of the protein, upon oxygen release, from the relaxed (R) to the tense (T) state. Despite the large amount of data accumulated on the end-points of the transition, scarce structural information is available on the intermediate species along the pathway. Here we report a spectroscopic characterization of the autoxidation process of the Trematomus newnesi major Hb component and the atomic resolution structure (1.25 A) of an intermediate form along the pathway characterized by a different binding and oxidation state of the alpha and beta chains. In contrast to the alpha-heme iron, which binds a CO molecule, the beta iron displays a pentacoordinated oxidized state, which is rare in tetrameric hemoglobins. Interestingly, the information provided by the present analysis is not limited to the characterization of the peculiar oxidation process of Antarctic fish hemoglobins. Indeed, this structure represents the most detailed snapshot of hemoglobin allosteric transition hitherto achieved. Upon ligand release at the beta heme, a cascade of structural events is observed. Notably, several structural features of the tertiary structure of the alpha and beta chains closely resemble those typically observed in the deoxygenated state. The overall quaternary structure also becomes intermediate between the R and the T state. The analysis of the alterations induced by the ligand release provides a clear picture of the temporal sequence of the events associated with the transition. The implications of the present findings have also been discussed in the wider context of tetrameric Hbs.

Crystallographic and spectroscopic evidences on Antarctic fish hemoglobins (AFHbs) have revealed that their ferric tetramers at physiological pH are in a mixed ±(aquo-met)/²(bis-histidyl) coordination state and show a quaternary structure intermediate between the classical R and T states (H state). Ferric bis-histidyl adducts (hemichromes) have been also observed in some mammalian Hbs. In order to clarify whether hemichrome in AFHbs can be converted into a ferrous bis-histidyl adduct (hemochrome), at least in the crystal phase, chemical reduction of ferric hemoglobins from Trematomus bernacchii (HbTb) single crystals has been followed via Raman microscopy. The results of this analysis reveal that in HbTb, upon reduction, the bis-histidyl coordination is disrupted in favor of a penta-coordinated ferrous deoxy state, with no evidence of hemochrome. These data are in agreement with UV/Vis absorption spectra in solution. Furthermore, our data are also indirectly supported by the observation that upon reduction with dithionite, the ferric HbTb crystals crack and lose their diffraction power: in the crystalline state, the quaternary structure transition from the H to the T state is not compatible with the crystal packing. Altogether these data indicate that if bis-histidyl adducts have a functional significance in AFHbs, this function refers to a stable ferric state, or to a transient, though never detected, ferrous species.

Hemoglobins (Hbs) are important proteins devoted to oxygen transport. Hbs carry out their function by keeping the iron atom, which binds the oxygen molecule, in its reduced Fe(II) state. Nonetheless, it is well known that Hbs frequently undergo, even under physiological conditions, spontaneous oxidation. Although these processes have been widely investigated, their role and impact in different biological contexts are still highly debated. In vertebrate Hbs, assembled in ±2²2 tetramers, it has traditionally been assumed that oxidized forms endowed with nativelike structures are either aquo-met or hydroxy-met states, depending on the pH of the medium. This view has been questioned by several independent investigations. In the past, indirect evidence of the existence of alternative nativelike oxidized forms was obtained from spectroscopic analyses. Indeed, it was suggested that, in tetrameric Hbs, bis-histidyl hemichrome states could be compatible with folded structures. Recent studies performed by complementing spectroscopic and crystallographic methodologies have provided a detailed picture of hemichrome structure and formation in these proteins. Here we review the methodological approaches adopted to achieve these results, the main structural features of these states, and the current hypotheses on their possible functional implications.
