101. Examining the Association between Acculturation Indicators and Metabolic Syndrome among Hispanic Adults

Abstract

Purpose: To examine the relationship between acculturation indicators and metabolic syndrome among Hispanic adults living in the Dallas-Fort Worth-Arlington, Texas metropolitan area. Methods: This study utilized secondary data collected from a larger 2014 study among 128 Hispanic adults living in Dallas-Fort Worth-Arlington. The current study examined two research questions: (1) Is there a relationship between acculturation indicators (nativity, duration in the United States, and the Short Acculturation Scale for Hispanics score) and metabolic syndrome among Hispanic adults living in Dallas-Fort Worth-Arlington? (2) Is there a relationship between acculturation indicators and the individual markers associated with metabolic syndrome (high waist circumference, high blood pressure, elevated fasting blood glucose, elevated blood triglycerides, and reduced high density lipoprotein cholesterol among Hispanic adults living in Dallas-Fort Worth-Arlington? Results: Most participants were female (aged 40 and older) born outside of the United States. More than half of the participants had been in the United States fewer than 20 years and revealed loweracculturation scores. More than one-third of the participants also had metabolic syndrome. Prevalence of metabolic syndrome was higher among females compared to males. Most participants had abnormal high-density lipoprotein and waist circumference. Acculturation scores and nativity were not associated with abnormal metabolic syndrome markers. Duration in the United States was not associated with metabolic syndrome markers of high waist circumference, reduced high-density lipoprotein, and elevated blood triglycerides. However, duration in the United States was associated with metabolic syndrome markers of high blood pressure and elevated fasting blood glucose. Conclusions and Recommendations: Acculturation is a multifaceted, dynamic phenomenon that influences health at multiple levels. Future studies can explore a range of social-ecological factors that interact with acculturation and health. Because metabolic syndrome is considered a "lifestyle syndrome," there is an additional need to examine behavioral health and relationships among diet, physical activity, and acculturation. Through a social-ecological and culturally relevant lens, health education specialists can lead interprofessional efforts to promote health literacy and healthy lifestyle behaviors among Hispanic individuals and communities. These action steps can be integral components of professional preparation programs that offer students opportunities to engage in service learning and develop core competencies in their disciplines.

102. Elicitation and Empathy with AI-enhanced Adaptive Assistive Technologies (AATs): Towards Sustainable Inclusive Design Method Education

Abstract

Efforts to include people with disabilities in design education are difficult to scale, and dynamics of participation need to be carefully planned to avoid putting unnecessary burdens on users. However, given the scale of emerging AI-enhanced technologies and their potential for creating new vulnerabilities for marginalized populations, new methods for generating empathy and self-reflection in technology design students (as the future creators of such technologies) are needed. We report on a study with Information Systems graduate students where they used a participatory elicitation toolkit to reflect on two cases of end-user privacy perspectives towards AI-enhanced tools in the age of surveillance capitalism: their own when using tools to support learning, and those of older adults using AI-enhanced adaptive assistive technologies (AATs) that help with pointing and typing difficulties. In drawing on the experiences of students with intersectional identities, our exploratory study aimed to incorporate intersectional thinking in privacy elicitation and further understand its role in enabling sustainable, inclusive design practice and education. While aware of the risks to their own privacy and the role of identity and power in shaping experiences of bias, students who used the toolkit were more sanguine about risks faced by AAT users--assuming more data equates to better technology. Our tool proved valuable for eliciting reflection but not empathy.

Abstract

Can large language models assist in data discovery? Data discovery predominantly happens via search on a data portal or the web, followed by assessment of the dataset to ensure it is fit for the intended purpose. The ability of conversational generative AI (CGAI) to support recommendations with reasoning implies it can suggest datasets to users, explain why it has done so, and provide information akin to documentation regarding the dataset in order to support a use decision. We hold 3 workshops with data users and find that, despite limitations around web capabilities, CGAIs are able to suggest relevant datasets and provide many of the required sensemaking activities, as well as support dataset analysis and manipulation. However, CGAIs may also suggest fictional datasets, and perform inaccurate analysis. We identify emerging practices in data discovery and present a model of these to inform future research directions and data prompt design., Comment: 27 pages, 9 figures

Abstract

We study the Moore-Penrose inverse of perturbations by a symmetrically-normed ideal of a closed range operator on a Hilbert space. We show that the notion of essential codimension of projections gives a characterization of subsets of such perturbations in which the Moore-Penrose inverse is continuous with respect to the metric induced by the operator ideal. These subsets are maximal satisfying the continuity property, and they carry the structure of real analytic Banach manifolds, which are acted upon transitively by the Banach-Lie group consisting of invertible operators associated with the ideal. This geometric construction allows us to prove that the Moore-Penrose inverse is indeed a real bianalytic map between infinite-dimensional manifolds. We use these results to study the polar decomposition of closed range operators from a similar geometric perspective. At this point we prove that operator monotone functions are real analytic in the norm of any symmetrically-normed ideal. Finally, we show that the maps defined by the operator modulus and the polar factor in the polar decomposition of closed range operators are real analytic fiber bundles., Comment: 31 pages

Abstract

We investigate the accuracy of the perturbative galaxy bias expansion in view of the forthcoming analysis of the Euclid spectroscopic galaxy samples. We compare the performance of an Eulerian galaxy bias expansion, using state-of-art prescriptions from the effective field theory of large-scale structure (EFTofLSS), against a hybrid approach based on Lagrangian perturbation theory and high-resolution simulations. These models are benchmarked against comoving snapshots of the Flagship I N-body simulation at $z=(0.9,1.2,1.5,1.8)$, which have been populated with H$\alpha$ galaxies leading to catalogues of millions of objects within a volume of about $58\,h^{-3}\,{\rm Gpc}^3$. Our analysis suggests that both models can be used to provide a robust inference of the parameters $(h, \omega_{\rm c})$ in the redshift range under consideration, with comparable constraining power. We additionally determine the range of validity of the EFTofLSS model in terms of scale cuts and model degrees of freedom. From these tests, it emerges that the standard third-order Eulerian bias expansion can accurately describe the full shape of the real-space galaxy power spectrum up to the maximum wavenumber $k_{\rm max}=0.45\,h\,{\rm Mpc}^{-1}$, even with a measurement precision well below the percent level. In particular, this is true for a configuration with six free nuisance parameters, including local and non-local bias parameters, a matter counterterm, and a correction to the shot-noise contribution. Fixing either tidal bias parameters to physically-motivated relations still leads to unbiased cosmological constraints. We finally repeat our analysis assuming a volume that matches the expected footprint of Euclid, but without considering observational effects, as purity and completeness, showing that we can get consistent cosmological constraints over this range of scales and redshifts., Comment: 38 pages, 19 figures

Abstract

We derived galaxy colour selections from Euclid and ground-based photometry, aiming to accurately define background galaxy samples in cluster weak-lensing analyses. Given any set of photometric bands, we developed a method for the calibration of optimal galaxy colour selections that maximises the selection completeness, given a threshold on purity. We calibrated galaxy selections using simulated ground-based $griz$ and Euclid $Y_{\rm E}J_{\rm E}H_{\rm E}$ photometry. Both selections produce a purity higher than 97%. The $griz$ selection completeness ranges from 30% to 84% in the lens redshift range $z_{\rm l}\in[0.2,0.8]$. With the full $grizY_{\rm E}J_{\rm E}H_{\rm E}$ selection, the completeness improves by up to $25$ percentage points, and the $z_{\rm l}$ range extends up to $z_{\rm l}=1.5$. The calibrated colour selections are stable to changes in the sample limiting magnitudes and redshift, and the selection based on $griz$ bands provides excellent results on real external datasets. The $griz$ selection is also purer at high redshift and more complete at low redshift compared to colour selections found in the literature. We find excellent agreement in terms of purity and completeness between the analysis of an independent, simulated Euclid galaxy catalogue and our calibration sample, except for galaxies at high redshifts, for which we obtain up to 50 percent points higher completeness. The combination of colour and photo-$z$ selections applied to simulated Euclid data yields up to 95% completeness, while the purity decreases down to 92% at high $z_{\rm l}$. We show that the calibrated colour selections provide robust results even when observations from a single band are missing from the ground-based data. Abstract

We investigate the approximations needed to efficiently predict the large-scale clustering of matter and dark matter halos in beyond-$\Lambda$CDM scenarios. We examine the normal branch of the Dvali-Gabadadze-Porrati model, the Hu-Sawicki $f(R)$ model, a slowly evolving dark energy, an interacting dark energy model and massive neutrinos. For each, we test approximations for the perturbative kernel calculations, including the omission of screening terms and the use of perturbative kernels based on the Einstein-de Sitter universe; we explore different infrared-resummation schemes, tracer bias models and a linear treatment of massive neutrinos; we employ two models for redshift space distortions, the Taruya-Nishimishi-Saito prescription and the Effective Field Theory of Large-Scale Structure. This work further provides a preliminary validation of the codes being considered by Euclid for the spectroscopic clustering probe in beyond-$\Lambda$CDM scenarios. We calculate and compare the $\chi^2$ statistic to assess the different modelling choices. This is done by fitting the spectroscopic clustering predictions to measurements from numerical simulations and perturbation theory-based mock data. We compare the behaviour of this statistic in the beyond-$\Lambda$CDM cases, as a function of the maximum scale included in the fit, to the baseline $\Lambda$CDM case. We find that the Einstein-de Sitter approximation without screening is surprisingly accurate for all cases when comparing to the halo clustering monopole and quadrupole obtained from simulations. Our results suggest that the inclusion of multiple redshift bins, higher-order multipoles, higher-order clustering statistics (such as the bispectrum) and photometric probes such as weak lensing, will be essential to extract information on massive neutrinos, modified gravity and dark energy., Comment: 29 pages, 10 figures, 4 tables, 3 appendices. Abstract

The statistical distribution and evolution of key properties (e.g. accretion rate, mass, or spin) of active galactic nuclei (AGN), remain an open debate in astrophysics. The ESA Euclid space mission, launched on July 1st 2023, promises a breakthrough in this field. We create detailed mock catalogues of AGN spectra, from the rest-frame near-infrared down to the ultraviolet, including emission lines, to simulate what Euclid will observe for both obscured (type 2) and unobscured (type 1) AGN. We concentrate on the red grisms of the NISP instrument, which will be used for the wide-field survey, opening a new window for spectroscopic AGN studies in the near-infrared. We quantify the efficiency in the redshift determination as well as in retrieving the emission line flux of the H$\alpha$+[NII] complex as Euclid is mainly focused on this emission line as it is expected to be the brightest one in the probed redshift range. Spectroscopic redshifts are measured for 83% of the simulated AGN in the interval where the H$\alpha$+[NII] is visible (0.892x10^{-16}$ erg s$^{-1}$ cm$^{-2}$, encompassing the peak of AGN activity at $z\simeq 1-1.5$) within the spectral coverage of the red grism. Outside this redshift range, the measurement efficiency decreases significantly. Overall, a spectroscopic redshift is correctly determined for ~90% of type 2 AGN down to an emission line flux of $3x10^{-16}$ erg s$^{-1}$ cm$^{-2}$, and for type 1 AGN down to $8.5x10^{-16}$ erg s$^{-1}$ cm$^{-2}$. Recovered black hole mass values show a small offset with respect to the input values ~10%, but the agreement is good overall. With such a high spectroscopic coverage at z<2, we will be able to measure AGN demography, scaling relations, and clustering from the epoch of the peak of AGN activity down to the present-day Universe for hundreds of thousand AGN with homogeneous spectroscopic information., Comment: 29 pages, 23 figures. Abstract

In April to May 2023, the superBIT telescope was lifted to the Earth's stratosphere by a helium-filled super-pressure balloon, to acquire astronomical imaging from above (99.5% of) the Earth's atmosphere. It was launched from New Zealand then, for 40 days, circumnavigated the globe five times at a latitude 40 to 50 degrees South. Attached to the telescope were four 'DRS' (Data Recovery System) capsules containing 5 TB solid state data storage, plus a GNSS receiver, Iridium transmitter, and parachute. Data from the telescope were copied to these, and two were dropped over Argentina. They drifted 61 km horizontally while they descended 32 km, but we predicted their descent vectors within 2.4 km: in this location, the discrepancy appears irreducible below 2 km because of high speed, gusty winds and local topography. The capsules then reported their own locations to within a few metres. We recovered the capsules and successfully retrieved all of superBIT's data - despite the telescope itself being later destroyed on landing., Comment: 12 pages

Abstract

In this paper we investigate the impact of lensing magnification on the analysis of Euclid's spectroscopic survey, using the multipoles of the 2-point correlation function for galaxy clustering. We determine the impact of lensing magnification on cosmological constraints, and the expected shift in the best-fit parameters if magnification is ignored. We consider two cosmological analyses: i) a full-shape analysis based on the $\Lambda$CDM model and its extension $w_0w_a$CDM and ii) a model-independent analysis that measures the growth rate of structure in each redshift bin. We adopt two complementary approaches in our forecast: the Fisher matrix formalism and the Markov chain Monte Carlo method. The fiducial values of the local count slope (or magnification bias), which regulates the amplitude of the lensing magnification, have been estimated from the Euclid Flagship simulations. We use linear perturbation theory and model the 2-point correlation function with the public code coffe. For a $\Lambda$CDM model, we find that the estimation of cosmological parameters is biased at the level of 0.4-0.7 standard deviations, while for a $w_0w_a$CDM dynamical dark energy model, lensing magnification has a somewhat smaller impact, with shifts below 0.5 standard deviations. In a model-independent analysis aiming to measure the growth rate of structure, we find that the estimation of the growth rate is biased by up to $1.2$ standard deviations in the highest redshift bin. As a result, lensing magnification cannot be neglected in the spectroscopic survey, especially if we want to determine the growth factor, one of the most promising ways to test general relativity with Euclid. We also find that, by including lensing magnification with a simple template, this shift can be almost entirely eliminated with minimal computational overhead., Comment: 23 pages, 14 figures

Abstract

The Euclid photometric survey of galaxy clusters stands as a powerful cosmological tool, with the capacity to significantly propel our understanding of the Universe. Despite being sub-dominant to dark matter and dark energy, the baryonic component in our Universe holds substantial influence over the structure and mass of galaxy clusters. This paper presents a novel model to precisely quantify the impact of baryons on galaxy cluster virial halo masses, using the baryon fraction within a cluster as proxy for their effect. Constructed on the premise of quasi-adiabaticity, the model includes two parameters calibrated using non-radiative cosmological hydrodynamical simulations and a single large-scale simulation from the Magneticum set, which includes the physical processes driving galaxy formation. As a main result of our analysis, we demonstrate that this model delivers a remarkable one percent relative accuracy in determining the virial dark matter-only equivalent mass of galaxy clusters, starting from the corresponding total cluster mass and baryon fraction measured in hydrodynamical simulations. Furthermore, we demonstrate that this result is robust against changes in cosmological parameters and against varying the numerical implementation of the sub-resolution physical processes included in the simulations. Our work substantiates previous claims about the impact of baryons on cluster cosmology studies. In particular, we show how neglecting these effects would lead to biased cosmological constraints for a Euclid-like cluster abundance analysis. Importantly, we demonstrate that uncertainties associated with our model, arising from baryonic corrections to cluster masses, are sub-dominant when compared to the precision with which mass-observable relations will be calibrated using Euclid, as well as our current understanding of the baryon fraction within galaxy clusters., Comment: 18 pages, 10 figures, 4 tables, 1 appendix, abstract abridged for arXiv submission; v2 matches published version

Abstract

Deviations from Gaussianity in the distribution of the fields probed by large-scale structure surveys generate additional terms in the data covariance matrix, increasing the uncertainties in the measurement of the cosmological parameters. Super-sample covariance (SSC) is among the largest of these non-Gaussian contributions, with the potential to significantly degrade constraints on some of the parameters of the cosmological model under study - especially for weak lensing cosmic shear. We compute and validate the impact of SSC on the forecast uncertainties on the cosmological parameters for the Euclid photometric survey, obtained with a Fisher matrix analysis, both considering the Gaussian covariance alone and adding the SSC term - computed through the public code $\tt{PySSC}$. The photometric probes are considered in isolation and combined in the '3$\times$2pt' analysis. We find the SSC impact to be non-negligible - halving the Figure of Merit of the dark energy parameters $(w_0, w_a)$ in the 3$\times$2pt case and substantially increasing the uncertainties on $\Omega_{{\rm m}, 0}, w_0$, and $\sigma_8$ for cosmic shear; photometric galaxy clustering, on the other hand, is less affected due to the lower probe response. The relative impact of SSC does not show significant changes under variations of the redshift binning scheme, while it is smaller for weak lensing when marginalising over the multiplicative shear bias nuisance parameters, which also leads to poorer constraints on the cosmological parameters. Finally, we explore how the use of prior information on the shear and galaxy bias changes the SSC impact. Improving shear bias priors does not have a significant impact, while galaxy bias must be calibrated to sub-percent level to increase the Figure of Merit by the large amount needed to achieve the value when SSC is not included., Comment: 22 pages, 13 figures

Abstract

The technique of photometric redshifts has become essential for the exploitation of multi-band extragalactic surveys. While the requirements on photo-zs for the study of galaxy evolution mostly pertain to the precision and to the fraction of outliers, the most stringent requirement in their use in cosmology is on the accuracy, with a level of bias at the sub-percent level for the Euclid cosmology mission. A separate, and challenging, calibration process is needed to control the bias at this level of accuracy. The bias in photo-zs has several distinct origins that may not always be easily overcome. We identify here one source of bias linked to the spatial or time variability of the passbands used to determine the photometric colours of galaxies. We first quantified the effect as observed on several well-known photometric cameras, and found in particular that, due to the properties of optical filters, the redshifts of off-axis sources are usually overestimated. We show using simple simulations that the detailed and complex changes in the shape can be mostly ignored and that it is sufficient to know the mean wavelength of the passbands of each photometric observation to correct almost exactly for this bias; the key point is that this mean wavelength is independent of the spectral energy distribution of the source}. We use this property to propose a correction that can be computationally efficiently implemented in some photo-z algorithms, in particular template-fitting. We verified that our algorithm, implemented in the new photo-z code Phosphoros, can effectively reduce the bias in photo-zs on real data using the CFHTLS T007 survey, with an average measured bias Delta z over the redshift range 0.4

Abstract

We report the discovery, mass, and radius determination of TOI-1801 b, a temperate mini-Neptune around a young M dwarf. TOI-1801 b was observed in TESS sectors 22 and 49, and the alert that this was a TESS planet candidate with a period of 21.3 days went out in April 2020. However, ground-based follow-up observations, including seeing-limited photometry in and outside transit together with precise radial velocity (RV) measurements with CARMENES and HIRES revealed that the true period of the planet is 10.6 days. These observations also allowed us to retrieve a mass of 5.74 $\pm$ 1.46 $M_\oplus$, which together with a radius of 2.08 $\pm$ 0.12 $R_\oplus$, means that TOI-1801 b is most probably composed of water and rock, with an upper limit of 2\% by mass of H$_{2}$ in its atmosphere. The stellar rotation period of 16 days is readily detectable in our RV time series and in the ground-based photometry. We derived a likely age of 600--800 Myr for the parent star TOI-1801, which means that TOI-1801 b is the least massive young mini-Neptune with precise mass and radius determinations. Our results suggest that if TOI-1801 b had a larger atmosphere in the past, it must have been removed by some evolutionary mechanism on timescales shorter than 1 Gyr., Comment: Accepted in A&A. 29 pages, 21 figures

Abstract

Up to 150000 asteroids will be visible in the images of the ESA Euclid space telescope, and the instruments of Euclid offer multiband visual to near-infrared photometry and slitless spectra of these objects. Most asteroids will appear as streaks in the images. Due to the large number of images and asteroids, automated detection methods are needed. A non-machine-learning approach based on the StreakDet software was previously tested, but the results were not optimal for short and/or faint streaks. We set out to improve the capability to detect asteroid streaks in Euclid images by using deep learning. We built, trained, and tested a three-step machine-learning pipeline with simulated Euclid images. First, a convolutional neural network (CNN) detected streaks and their coordinates in full images, aiming to maximize the completeness (recall) of detections. Then, a recurrent neural network (RNN) merged snippets of long streaks detected in several parts by the CNN. Lastly, gradient-boosted trees (XGBoost) linked detected streaks between different Euclid exposures to reduce the number of false positives and improve the purity (precision) of the sample. The deep-learning pipeline surpasses the completeness and reaches a similar level of purity of a non-machine-learning pipeline based on the StreakDet software. Additionally, the deep-learning pipeline can detect asteroids 0.25-0.5 magnitudes fainter than StreakDet. The deep-learning pipeline could result in a 50% increase in the number of detected asteroids compared to the StreakDet software. There is still scope for further refinement, particularly in improving the accuracy of streak coordinates and enhancing the completeness of the final stage of the pipeline, which involves linking detections across multiple exposures., Comment: 18 pages, 11 figures

Abstract

The effect of stellar multiplicity on planetary architecture and orbital dynamics provides an important context for exoplanet demographics. We present a volume-limited catalog up to 300 pc of 66 stars hosting planets and planet candidates from Kepler, K2 and TESS with significant Hipparcos-Gaia proper motion anomalies, which indicate the presence of companions. We assess the reliability of each transiting planet candidate using ground-based follow-up observations, and find that the TESS Objects of Interest (TOIs) with significant proper motion anomalies show nearly four times more false positives due to Eclipsing Binaries compared to TOIs with marginal proper motion anomalies. In addition, we find tentative evidence that orbital periods of planets orbiting TOIs with significant proper motion anomalies are shorter than those orbiting TOIs without significant proper motion anomalies, consistent with the scenario that stellar companions can truncate planet-forming disks. Furthermore, TOIs with significant proper motion anomalies exhibit lower Gaia differential velocities in comparison to field stars with significant proper motion anomalies, suggesting that planets are more likely to form in binary systems with low-mass substellar companions or stellar companions at wider separation. Finally, we characterize the three-dimensional architecture of LTT 1445 ABC using radial velocities, absolute astrometry from Gaia and Hipparcos, and relative astrometry from imaging. Our analysis reveals that LTT 1445 is a nearly flat system, with a mutual inclination of 2.88 deg between the orbit of BC around A and that of C around B. Abstract

Neuroendocrine carcinomas, such as neuroendocrine prostate cancer and small-cell lung cancer, commonly have a poor prognosis and limited therapeutic options. We report that ubiquitin carboxy-terminal hydrolase L1 (UCHL1), a deubiquitinating enzyme, is elevated in tissues and plasma from patients with neuroendocrine carcinomas. Loss of UCHL1 decreases tumor growth and inhibits metastasis of these malignancies. UCHL1 maintains neuroendocrine differentiation and promotes cancer progression by regulating nucleoporin, POM121, and p53. UCHL1 binds, deubiquitinates, and stabilizes POM121 to regulate POM121-associated nuclear transport of E2F1 and c-MYC. Treatment with the UCHL1 inhibitor LDN-57444 slows tumor growth and metastasis across neuroendocrine carcinomas. The combination of UCHL1 inhibitors with cisplatin, the standard of care used for neuroendocrine carcinomas, significantly delays tumor growth in pre-clinical settings. Our study reveals mechanisms of UCHL1 function in regulating the progression of neuroendocrine carcinomas and identifies UCHL1 as a therapeutic target and potential molecular indicator for diagnosing and monitoring treatment responses in these malignancies.

Recent research indicates that paired comparisons can accelerate perceptual learning of challenging dermatological lesion categories. Here we investigated whether the role of object categories as targets or distractors differentially influences learning outcomes. The frequency with which a given category occupied the target position was manipulated across three learning conditions: Always-Never, where half of 10 categories were always shown as target and the other half never shown as target; Often-Rarely, where half of categories appeared 75% as targets and 25% as distractors, with reversed presentation frequency for the other half; and Equal Split learning, in which all categories appeared as targets or distractors equally often. After learning, transfer results indicated that all conditions yielded equivalent overall learning, but categories prioritized more often as targets exhibited greater learning gains. These findings implicate differential processing of images in comparisons, even when no information regarding target vs. distractor was given prior to feedback.

Variability is used to calculate critical change values (CCVs) that are thresholds representing real change in a measure not associated with observer variability. Hypothesis Observed intra‐ and interobserver accuracy and variability in healthy dogs help model CCV for RFMR. Animals Reliability cohort of 34 healthy dogs; allometric scaling cohort of 99 dogs with heart disease and 25 healthy dogs. Methods Accuracy, variability, and CCV of 2 observers using geometric and flow‐based echocardiography were prospectively compared against a standard of RFMR = 0% and extrapolated across a range of expected RFMR values in the reliability cohort partly derived from cardiac dimensions predicted by the allometric cohort. Results Accuracy of methods to determine RFMR in descending order was 4‐chamber bullet (Bullet4CH), mitral inflow, cube formula, and Simpson's method of disks. Intraobserver variability was relatively high. The CCV for RFMR ranged from 28% to 88% and was inversely related to RFMR when extrapolated for use in affected dogs. For both observers, the Bullet4CH method had the lowest intraobserver CCV (Operator 1:28%, Operator 2:41%). Interobserver strength of agreement was low with intraclass correlation coefficients ranging from 0.210 to 0.413. Conclusions and Clinical Importance Echocardiographic volumetric methods used to calculate stroke volume and RFMR have low accuracy and high variability in healthy dogs. Extrapolation of observed CCV to a range of expected RFMR suggests observers and methods are not interchangeable and variability might hinder routine clinical usage. Individual observers should be aware of their own variability and CCV.

Abstract

Abstract Imidazole Propionate (ImP), a gut-derived metabolite from histidine, affects insulin signaling in mice and is elevated in type 2 diabetes (T2D). However, the source of histidine and the role of the gut microbiota remain unclear. We conducted an intervention study in mice and humans, comparing ImP kinetics in mice on a high-fat diet with varying histidine levels and antibiotics, and assessed ImP levels in healthy and T2D subjects with histidine supplementation. Results show that dietary histidine is metabolized to ImP, with antibiotic-induced gut microbiota suppression reducing ImP levels in mice. In contrast, oral histidine supplementation resulted in increases in circulating ImP levels in humans, whereas antibiotic treatment increased ImP levels, which was associated with a bloom of several bacterial genera that have been associated with ImP production, such as Lactobacilli. Our findings highlight the gut microbiota’s crucial role in regulating ImP and the complexity of translating mouse models to humans.

Abstract

Abstract Background Smoking in pregnancy continues to cause significant morbidity to mothers and babies and contributes to tremendous costs to society. Maternal-fetal attachment (MFA) may differentiate smokers who quit or pregnant smokers from non-smokers. Researchers have recommended utilizing interventions that improve MFA to help decrease smoking within pregnancy. Methods We performed a randomized clinical trial of pregnant smokers (n = 33) using an MFA-informed, intention-to-treat protocol. We recruited pregnant smokers and provided timeline follow back (TLFB) interviews from 27 weeks of pregnancy until 6 weeks post-partum. Salivary cotinine was also collected at five different time points. 3D ultrasonography was performed, and patients were randomly assigned a 3D picture or a 3D model of their fetus. Results Overall, the average percent reduction in cigarette use was 37.03% (SD = 31.18). The main effect of 3D type was not significant (3D Model vs. 3D Print Estimate = -0.09, 95% CI: − 0.19 to 0.01, p = 0.066). A total of 4 patients (12%) quit smoking within one week of delivery. A 10% reduction in cigarette use was associated with a 30.57 g increase in birth weight (Estimate = 30.57, 95% CI: -14.15 to 75.29); a 10% reduction in cigarette use was associated with a 0.14 week increase in estimate gestational age at delivery (Estimate = 0.14, 95% CI: -0.01 to 0.28). Conclusions Patients who smoke in pregnancy decrease the number of cigarettes smoked after receiving either a 3D picture or 3D model of their fetus. Abstract Background High-risk strains of HPV are known to cause cervical cancer. Multiple clinical studies have emphasized that smoking and drinking are critical risk factors for cervical cancer and its high-grade precursors. In this study, we investigated if smoking and/or drinking augment the molecular mechanisms of cervical carcinogenesis and defined a potential therapeutic approach for their attenuation. Methods The impact of benzo[a]pyrene (B[a]P) and/or ethanol (EtOH) exposure on cervical cancer cells was assessed by measuring changes in their cell migration and invasion characteristics. Expression of HPV16 E6/E7, NF-κB, cytokines, and inflammation mediators was determined using qRT-PCR, immunoblotting, ELISA, luciferase reporter assay, and confocal microscopy. Herein, we used curcumin (Cur), and PLGA nanoparticle formulation of curcumin (PLGA-Cur) and determined effectiveness of free Cur and PLGA-Cur formulation on smoking and drinking activated NF-κB/IL-6 mediated inflammatory signaling pathways using in vitro cervical cancer models. Results Treatments with B[a]P and/or EtOH altered the expression of HPV16 E6/E7 oncogenes and EMT markers in cervical cancer cells; it also enhanced migration and invasion. In addition, B[a]P and/or EtOH exposure promoted inflammation pathways through TNF-α and NF-κB signaling, leading to IL-6 upregulation and activation of VEGF. The molecular effects caused by B[a]P and/or EtOH exposure were effectively attenuated by curcumin (Cur)/PLGA-Cur treatment. Conclusions These data suggest a molecular link between smoking, drinking, and HPV infectivity in cervical carcinogenesis. In addition, attenuation of these effects by treatment with Cur/PLGA-Cur treatment, implies the role of curcumin in cervical cancer prevention and treatment.

Abstract

The increased use of immune checkpoint inhibitors (ICIs) across cancer programs has created the need for standardized monitoring and management of immune-related adverse events (irAEs). Delayed recognition without appropriate treatment can have serious and life-threatening consequences. The management of irAEs presents a unique set of challenges that must be addressed at a multidisciplinary level. Although various national and international guidelines and working groups provide high-level recommendations for the management of irAEs, practical guidance is lacking. Furthermore, timely collaboration between specialists requires institutional protocols that enable the early recognition, assessment, and treatment of irAEs. Such protocols should be developed by institution specialists and include algorithms for all healthcare providers involved in the care of patients treated with ICIs. At William Osler Health System in Brampton, Ontario, practical step-by-step multidisciplinary treatment approaches with recommendations for the management of irAEs were developed in collaboration with experts across Canada. Here, we provide an in-depth description of the approaches, outlining baseline investigations prior to the initiation of ICIs, as well as the monitoring and management of irAEs based on symptoms, severity, and involved organ systems. We encourage other centres to adapt and modify our approaches according to their specific needs and requirements.

Abstract

Abstract The widespread use of pesticides poses significant threats to both environmental and human health, primarily due to their potential toxic effects. The study investigated the cardiovascular toxicity of selected pesticides, focusing on their interactions with Toll-like receptor 4 (TLR4), an important part of the innate immune system. Using computational tools such as molecular docking, molecular dynamics (MD) simulations, principal component analysis (PCA), density functional theory (DFT) calculations, and ADME analysis, this study identified C160 as having the lowest binding affinity (-8.2 kcal/mol), followed by C107 and C165 (-8.0 kcal/mol). RMSD, RMSF, Rg, and hydrogen bond metrics indicated the formation of stable complexes between specific pesticides and TLR4. PCA revealed significant structural changes upon ligand binding, affecting stability and flexibility, while DFT calculations provided information about the stability, reactivity, and polarity of the compounds. ADME studies highlighted the solubility, permeability, and metabolic stability of C107, C160, and C165, suggesting their potential for bioavailability and impact on cardiovascular toxicity. C107 and C165 exhibit higher bioactivity scores, indicating favourable absorption, metabolism, and distribution properties. C165 also violated rule where molecular weight is greater than 500 g/mol. Further, DFT and NCI analysis of post MD conformations confirmed the binding of ligands at the binding pocket. The analysis shed light on the molecular mechanisms of pesticide-induced cardiovascular toxicity, aiding in the development of strategies to mitigate their harmful effects on human health.

Abstract

Abstract Reprogrammed glucose metabolism is considered as the hallmark of cancer with therapeutic implications. Phytocompounds have potential to inhibit cancer metabolism. Here, we tested the ability of Withaferin A (WA), a withanolide derived from Withania somnifera, in modulating cancer metabolism. The assessed effect of WA on aerobic glycolysis in breast cancer cell lines showed that WA decreases the glucose uptake, lactate production and ATP generation by inhibiting the expression of key glycolytic enzymes i.e., GLUT1, HK2 and PKM2. We also identified that WA induced inhibition of cancer glycolysis by targeting c-myc as validated by silencing experiments followed by metabolic readouts. Decreased glycolysis resulted in reduced cell viability, biomass and colony forming ability of breast cancer cells. To further validate our in vitro findings in breast cancer patients, we analyzed 90 metabolic pathways in ~ 2000 breast tumors and observed that glycolysis is the most deregulated pathway in breast tumors. Deregulated glycolysis also predicted poor prognosis in breast cancer patients. In addition, patient data showed correlation between c-myc expression and glycolytic deregulation in breast cancer. Taken together, our results highlight the role of WA in inhibiting breast cancer metabolism via c-myc/glycolysis axis.
