Over the last three decades rotary-screen printing has become the most important technique for textile printing. In discussing the challenges for the next decade, this paper identifies the trend towards shorter run lengths, particularly in Western Europe, as a major issue. It goes on to discuss the recent technical developments in the field of machine manufacturing, showing that rotary-screen printing is still very much alive.

I describe the diet and feeding behavior of silver leaf monkeys (Trachypithecus auratus sondaicus) in the Pangandaran Nature Reserve, West Java, Indonesia, and compare a group living in old secondary rain forest with a group living in mixed plantation/secondary forest to determine intraspecific variation in feeding behavior and the importance of the plantation species in the diet of the monkeys. Young leaves and leaf buds made up slightly less than half of their diets, with both groups showing a preference for a few species when seasonally available. Fruits and flowers of a few species were also preferentially selected when available. These included sweet, fleshy fruits, which most other colobines tend to avoid. Young leaf intake was greatest in months when fruit intake was low. Mature leaves were rarely eaten. Both groups spent approximately 20% of feeding time foraging on Moraceae species. Differences in the diet of the two groups were related largely to differences in vegetational composition and the availability and abundance of food items for the species common to both sites. Teak (Tectona grandis) was the top food species of the group living in mixed plantation/secondary forest, with the midribs of young leaves preferentially selected. Young leaves ofT. grandis, available throughout the study, provided a staple food and were eaten when preferred foods were scarce. More favored food items were available to the group living in old secondary forest, though none was a staple food.

Baculoviruses are used as biological control agents of insect pests in agriculture and forestry. The multiple-nucleocapsid nuclear polyhedrosis virus of Autographa californica (AcMNPV) is the prototype baculovirus. Recently, this virus has become widely used as vector for the high-level expression of foreign genes in insect cells. An updated physical map of restriction sites as well as the location of open reading frames (ORFs) and transcripts are presented. Most characteristic is the dispersal of "early", "late", and "very late" genes over the genome and the presence of nested sets of 3' and 5' coterminal transcripts.

Four ellagitannins from boysenberry, a cross between Rubus loganbaccus and Rubus baileyanus Britt., were isolated by preparative HPLC and the exact structures determined by a combination of LC–ESI-MS/MS, MALDI-TOF-MS and NMR spectroscopy. The two most abundant ellagitannins were identified as sanguiin H-6, which is known to be abundant in Rubus species, and the other was identified as an isomer of sanguiin H-10, which has not previously been reported in Rubus. The two less abundant ellagitannins were identified as sanguiin H-2 and [galloyl–bis-HHDP–glucose]2-gallate. Sanguiin H-2 has been previously reported in Rubus, whereas both sanguiin H-2 and [galloyl–bis-HHDP–glucose]2-gallate have been previously reported as hot-water degradation products of lambertianin C. Even though lambertianin C is reported to be a major ellagitannin in other Rubus species, it was not found in any of the fractions, suggesting that both sanguiin H-2 and [galloyl–bis-HHDP–glucose]2-gallate are present naturally in boysenberry.

Samples of leaves and fruits exploited as food items byTrachypithecus auratus sondaicus were analysed for nitrogen content, acid detergent fibre (ADF), pepsin cellulase digestibility (CDIG), condensed tannins (CT), total phenolics (TP) and protein precipitation capacity (PP) and compared with fruits and leaves not eaten. Differences in chemical measures for items eaten and not eaten were not statistically significant but trends indicate that leaves may have been selected for their lower fibre content and greater digestibility. Fruits eaten also had a higher mean level of CDIG and lower mean level of ADF than fruits not eaten but these measures are not considered to be of major importance in fruit selection as CDIG was lower and ADF higher in fruits eaten than in leaves eaten. Levels of CT, TP and PP capacity were higher in fruits eaten than in fruits not eaten but lower in leaves eaten than in leaves not eaten. The role of tannins and phenolics in food selection is discussed. Leaves (and fruits) were not strongly selected on the basis of protein content. Approximately half the dietary intake ofT. auratus sondaicus was leaves, a protein-rich food source. Possibly, protein levels in foliage at Pangandaran were sufficient that selection for this nutrient was not required. A nutrient other than protein (for example, soluble carbohydrates) may have been maximised through food selection. The protein/ADF ratio may provide an indicator of the acceptability of foliage in a habitat as potential food for a colobine. However, this ratio did not govern food selection byT. auratus sondaicus at Pangandaran.

An automated on-line monitoring system for low-molecular-weight compounds (e.g. sugars) during fermentation processes is described. The applicability of cross-flow ultrafiltration, coupled on-line with high-performance liquid chromatography, is evaluated. Various detection modes for the determination of the test compounds lactose, glucose and fructose) in a complex fermentation broth are compared. In addition, the influence of some system parameters on the performance of several ultrafiltration modules for the removal of cellular and macromolecular broth constituents is investigated. A method for the fully automated determination of sugars during anE. coli batch culture in a brain-heart infusion medium is presented. With the present system five analyses can be performed every hour, using only 35μl of sample, without significant deterioration of the ultrafiltration process. Contamination by external micro-organisms was not observed and good correlation with an off-line method was found.

The Gunung Halimun Reserve has the largest area of primary rain forest left on Java. The reserve is important for the habitat it provides for two endangered primates, the molochHylobates molochand the grizzled langurPresbytis comata,which are endemic to western Java. The author presents the results of a survey conducted in the reserve in July 1989 and makes suggestions for further survey work and scientific research on the primates there.

Ephemeral or seasonal streams tributaries to Magela Creek (Alligator Rivers Uranium province, Northern Territory), containing perennial pools, lie adjacent to a 1 km2 tailings storage dam, still being filled, whose partial seepage collecting system contains an increasing concentration of copper. Acute copper toxicity to local fauna was determined in a flow‐through apparatus at 27°C and pH 6. The 4‐day LC50 values (μg L‐1) for total copper were: rainbowfish Melanotaenia splendida inornata 170–190, pennyfish Denariusa bandata 75, and atyid shrimp Caridina sp. 3–4, and the 3‐day LC50 of the catfish Porochilus rendahli 85. Free ionic copper approximated the total copper. Test water quality was as predicted for seepage‐affected streams during a brief critical period in some years: of low hardness and alkalinity but enriched in Mg2+ and SO4 2‐. The relative LC50 values correspond to the relative occurrence of similar species among zones of the metals‐affected Finniss River. Magela Creek system waters often had ...

We provide new empirical evidence on non- linear liquidity management in Dutch firms. Our results reveal that liquidity adjustment from below the target is significantly faster than from above. We find no evidence for bands of inaction around the target.

MurG is a peripheral membrane protein that is one of the key enzymes in peptidoglycan biosynthesis. The crystal structure of Escherichia coli MurG (S. Ha, D. Walker, Y. Shi, and S. Walker, Protein Sci. 9:1045-1052, 2000) contains a hydrophobic patch surrounded by basic residues that may represent a membrane association site. To allow investigation of the membrane interaction of MurG on a molecular level, we expressed and purified MurG from E. coli in the absence of detergent. Surprisingly, we found that lipid vesicles copurify with MurG. Freeze fracture electron microscopy of whole cells and lysates suggested that these vesicles are derived from vesicular intracellular membranes that are formed during overexpression. This is the first study which shows that overexpression of a peripheral membrane protein results in formation of additional membranes within the cell. The cardiolipin content of cells overexpressing MurG was increased from 1 ± 1 to 7 ± 1 mol% compared to nonoverexpressing cells. The lipids that copurify with MurG were even further enriched in cardiolipin (13 ± 4 mol%). MurG activity measurements of lipid I, its natural substrate, incorporated in pure lipid vesicles showed that the MurG activity is higher for vesicles containing cardiolipin than for vesicles with phosphatidylglycerol. These findings support the suggestion that MurG interacts with phospholipids of the bacterial membrane. In addition, the results show a special role for cardiolipin in the MurG-membrane interaction.

We have shown previously that a wide range of mutagenic carcinogens are capable of inducing loss of heterozygosity (LOH) at the endogenous Aprt locus in mouse splenic lymphocytes. To investigate whether LOH might be caused by a single common mechanism, we set out to determine the extent of LOH by microsatellite analysis along (the Aprt gene containing) mouse chromosome 8. Aprt +/- hybrid B6C3F1 mice were treated with mutagens that induce different classes of DNA lesions, i.e. bulky DNA adducts, DNA methylation, DNA inter-strand crosslinks or DNA strand breaks. Aprt mutant frequencies (MF) in this C57Bl/ 6-C3H hybrid background were significantly reduced for mitomycin C (MMC) and methylmethanesulfonate (MMS) in comparison with MF in C57Bl/6 background, suggesting either enhanced repair or reduced formation of MMC-or MMS-induced mutagenic lesions in a hybrid B6C3F1 background. In contrast, Aprt MF after dimethylbenz[a]anthracene (DMBA), methylnitrosurea (MNU) and etoposide treatment were similar in both genetic backgrounds. Microsatellite analysis of Aprt mutant clones indicated a dominant role for mitotic recombination (MR) in generating spontaneous, DMBA- and etoposide-induced LOH at Aprt. However, over 80% of the MMC-induced Aprt LOH mutants had lost heterozygosity for all markers tested, suggesting that either the crossover points were located close to the centromere or that these mutants arose by chromosome loss and duplication of the remaining chromosome 8. A substantial fraction (40%) of MNU-induced Aprt mutants had lost the wild-type Aprt allele, but had retained heterozygosity at all polymorphic markers tested at chromosome 8 indicating an important role for deletions in LOH formation by MNU. Patterns of MR differed quite dramatically for the various chemical mutagens tested, suggesting different mechanisms to be involved in inducing recombination between homologous chromosomes. In addition, non-random adduct formation and repair between chromosomal regions, i.e. heterochromatin versus euchromatin, may contribute to a non-random distribution of recombinational crossover points.

Cellular folate homeostasis is essential to allow continuous use of reduced folate cofactors for a variety of biosynthetic processes including purine and pyrimidine biosynthesis, mitochondrial protein synthesis and amino acid biosynthesis. Beyond this, there is accumulating evidence that the antiproliferative and/or anti-inflammatory effects of methotrexate (MTX) or second/third generation of folate antagonists is dependent on the patients cellular folate status. High intra-/extracellular folate levels may abrogate the efficacy of folate antagonists, while low intra-/extracellular folate levels may play a role in determining toxic effects (1,2).

Heterogeneity in cancer susceptibility exists between patients with an inherited defect in nucleotide excision repair (NER). While xeroderma pigmentosum (XP) patients have elevated skin cancer rates, Cockayne syndrome (CS) patients do not appear to have increased cancer susceptibility. To investigate whether differences in mutagenesis are the basis for the variability in cancer proneness, we studied mutagenesis at the X-chromosomal Hprt gene and the autosomal Aprt gene in splenic T-lymphocytes after 7,12-dimethyl-1,2-benz[a]anthracene (DMBA) exposure in total NER-deficient Xpa mice, global genome repair (GGR)-deficient Xpc mice and transcription coupled repair (TCR)-deficient Csb mice. Surprisingly, while all intraperitoneally-treated Xpc(-/-) mice survived a dose of 40 mg/kg DMBA, a substantial fraction of the treated Xpa(-/-) and Csb(-/-) mice died a few days after treatment with a 20-fold lower dose. Functional TCR of DMBA adducts in Xpc(-/-) mice thus appears to alleviate DMBA toxicity. However, the mutagenic response in Xpc(-/-) mice was +/- 2-fold enhanced at both the Hprt and the Aprt gene compared to heterozygous controls, indicating that GGR at least partially removes DMBA adducts from the genome overall. DMBA-induced SCE frequencies in mouse dermal fibroblasts were significantly enhanced in Xpa- and Csb-, but not in Xpc-deficient background compared to the frequency in normal fibroblasts. These results indicate that both damage-induced cytotoxicity as well as intra-chromosomal recombinational events were not correlated to differences in cancer susceptibility in human NER syndrome patients.

Small hydrophobic peptides were studied as possible substrates of the multidrug resistance protein (MRP)-1 (ABCC1) transmembrane transporter molecule. As observed earlier for P-glycoprotein- (Pgp; ABCB1) overexpressing cells, MRP1-overexpressing cells, including cells stably transfected with the MRP1 cDNA, showed distinct resistance to the cytotoxic peptide N-acetyl-Leu-Leu-norleucinal (ALLN). Resistance to this peptide and another toxic peptide derivative, which is based on a Thr-His-Thr-Nle-Glu-Gly backbone conjugated to butyl and benzyl groups (4A6), could be reversed by MRP1 inhibitors. The reduced toxicity of 4A6 in MRP1-overexpressing cells was found to be associated with lower accumulation of a fluorescein-labeled derivative of this peptide. Glutathione (GSH) depletion had a clear effect on resistance to ALLN but hardly affected 4A6 resistance. In a limited structure-activity study using peptides that are analogous to 4A6, MRP1-overexpressing cells were found to be resistant to these peptides as well. Remarkably, when selecting A2780 ovarian cancer cells for resistance to ALLN, even in the absence of Pgp blockers, resulting cell lines had up-regulated MRP1, rather than any of the other currently known multidrug resistance transporter molecules including Pgp, MRP2 (ABCC2), MRP3 (ABCC3), MRP5 (ABCCS), and the breast cancer resistance protein ABCG2. ALLN-resistant, MRP1-overexpressing cells were found to be cross-resistant to 4A6 and the classical multidrug resistance drugs doxorubicin, vincristine, and etoposide. This establishes MRP1 as a transporter for small hydrophobic peptides. More extensive structure-activity relationship studies should allow the identification of clinically useful peptide antagonists of MRP1.

Tumor cells may display a multidrug resistance phenotype by overexpression of ATP binding cassette transporter genes such as multidrug resistance (MDR) 1 P-glycoprotein (P-gp) or the multidrug resistance protein 1 (MRP1). MDR3 P-gp is a close homologue of MDR1 P-gp, but its role in MDR is probably minor and remains to be established. The MRP1 protein belongs to a family of at least six members. Three of these, i.e., MRP1, MRP2, and MRP3, can transport MDR drugs and could be involved in MDR. The substrate specificity of the other family members remains to be defined. Specific monoclonal antibodies are required for wide-scale studies on the putative contribution of these closely related transporter proteins to MDR. In this report, we describe the extensive characterization of a panel of monoclonal antibodies (Mabs) detecting several MDR-related transporter proteins in both human and animal tissues. The panel consists of P3II-1 and P3II-26 for MDR3 P-gp; MRPr1, MRPm6, MRPm5, and MIB6 for MRP1; M2I-4, M2II-12, M2III-5 and M2III-6 for MRP2; M3II-9 and M3II-21 for MRP3; and M5I-1 and M5II-54 for MRP5. All Mabs in the panel appeared to be fully specific for their cognate antigens, both in Western blots and cytospin preparations, as revealed by lack of cross-reactivity with any of the other family members. Indeed, all Mabs were very effective in detecting their respective antigens in cytospins of transfected cell lines, whereas in flow cytometric and immunohistochemical analyses, distinct differences in reactivity and suitability were noted. These Mabs should become valuable tools in studying the physiological functions of these transporter proteins, in screening procedures for the absence of these proteins in hereditary metabolic (liver) diseases, and in studying the possible contributions of these molecules to MDR in cancer patients.

Intrinsic and/or acquired resistance to chemotherapy is the major obstacle to overcome in the treatment of patients with ovarian carcinoma. The aim of the present study was to investigate the prognostic value of drug resistance-associated proteins P-glycoprotein (P-gp), multidrug resistance-associated protein 1 (MRP1), canalicular multispecific organic anion transporter (c-MOAT/MRP2), and lung resistance protein (LRP) in ovarian carcinoma. Expression of P-gp, MRP1, MRP2, and LRP was determined by immunohistochemistry of frozen tissue sections of 115 ovarian carcinoma patients and related to clinicopathological factors, response to chemotherapy, and progression-free survival. P-gp expression was observed in 20 of 115 (17%), MRP1 in 51 (44%), MRP2 in 19 (16%), and LRP in 85 (74%) tumors. Expression of MRP1 was related to MRP2 (P0.0001) and P-gp (P0.001) expression, whereas LRP expression was more frequently observed in patients with early stage (P0.01), lower grade (P0.05), and smaller residual tumor (P0.05). Early stage (P0.001), smaller residual tumor (P0.001), and lower differentiation grade (P0.05) were related to longer (progression-free) survival. P-gp, MRP1, MRP2, and LRP expression were neither related to response to first-line chemotherapy in 59 evaluable patients nor to progression-free survival in all patients. On multivariate analysis, only stage and residual tumor were independent prognostic factors for survival. In conclusion, in ovarian carcinoma, MRP1 expression is associated with MRP2 and P-gp expression, whereas LRP expression is associated with favorable clinicopathological characteristics. Assessment of P-gp, MRP1, MRP2, or LRP does not allow prediction of response to chemotherapy or survival in ovarian carcinoma.

Transfection of multidrug resistance proteins (MRPs) MRP1 and MRP2 in human ovarian carcinoma 2008 cells conferred a marked level of resistance to short-term (1-4 h) exposure to the polyglutamatable antifolates methotrexate (MTX; 21-74-fold), ZD1694 (4-138-fold), and GW1843 (101-156-fold). Evidence for MRP-mediated antifolate efflux relies upon the following findings: (a) a 2-3.3-fold lower accumulation of [3H]MTX and subsequent reduced formation of long-chain polyglutamate forms of MTX; (b) reversal of MTX resistance by probenecid in both transfectants, and (c) ATP-dependent uptake of [3H]MTX in inside-out vesicles of MRP1 and MRP2 transfectants. This report provides a mechanistic basis for resistance to polyglutamatable antifolates through an MRP-mediated drug extrusion.

The human multidrug resistance protein (MRP) family contains at least six members: MRP1, the gene encoding the multidrug resistance protein; cMOAT or MRP2, encoding the canalicular multispecific organic anion transporter; and four homologues, called MRP3, MRP4, MRP5, and MRP6. The most recently discovered member of the family, MRP6, is analyzed in this report. The MRP6 gene is located on chromosome 16, immediately next to MRP1, and encodes a protein of 1503 amino acids with a predicted molecular weight of Mr 165,000. The 3' end of the MRP6 protein was found to be almost identical with the anthracycline resistance associated (ARA) protein identified previously in epirubicin-selected leukemia cells. Using both 3'- and 5'-derived MRP6 probes, we found that MRP6 is highly expressed in liver and kidney and to a low or very low extent in a few other tissues. No evidence was obtained for an independent expression of the ARA part of the MRP6 gene in normal tissues. To assess a possible role for MRP6 in multidrug resistance, we examined a large panel of resistant cell lines for the (over)expression of MRP6. We found overexpression of the complete MRP6 gene or part of it only in those cell lines with high overexpression and amplification of the MRP1 gene. DNA blot (Southern) analysis showed that MRP6 or a part of it is also amplified in these cell lines. Our results suggest that MRP6 does not play a role in the resistance of the resistant cells analyzed, and that MRP6/ARA is only coamplified with MRP1 because of its location immediately next to it on the same chromosome.

Allergic asthma is thought to be regulated by Th2 cells, and inhibiting this response is a promising mode of intervention. Many studies have focused on differentiation of Th cells to the Th1 or Th2 subset in vitro. IL-4 is essential for Th2 development, while IL-12 induces Th1 development, which can be enhanced by IL-18. In the present study, we investigated whether IL-12 and IL-18 were able to interfere in Th2 development and the associated airway symptoms in a mouse model of allergic asthma. Mice were sensitized with OVA using a protocol that induces IgE production. Repeated challenges by OVA inhalation induced elevated serum levels of IgE, airway hyperresponsiveness, and a predominantly eosinophilic infiltrate in the bronchoalveolar lavage concomitant with the appearance of Ag-specific Th2-like cells in lung tissue and lung-draining lymph nodes. Whereas treatments with neither IL-12 nor IL-18 during the challenge period were effective, combined treatment of IL-12 and IL-18 inhibited Ag-specific Th2-like cell development. This inhibition was associated with an absence of IgE up-regulation, airway hyperresponsiveness, and cellular infiltration in the lavage. These data show that, in vivo, the synergistic action of IL-12 and IL-18 is necessary to prevent Th2-like cell differentiation, and consequently inhibits the development of airway symptoms in a mouse model of allergic asthma.

By screening databases of human expressed sequence tags, we have identified three new homologues of MRP1, the gene encoding the multidrug resistance-associated protein, and cMOAT (or MRP2), the canalicular multispecific organic anion transporter gene. We call these new genes MRP3, MRP4, and MRP5. MRP3, like cMOAT, is mainly expressed in the liver. MRP4 is expressed only at very low levels in a few tissues, and MRP5, like MRP1, is expressed in almost every tissue tested. To assess a possible role of these new MRP homologues in multidrug or cisplatin resistance, a large set of resistant cell lines was examined for the (over)expression of MRP1, cMOAT, MRP3, MRP4, and MRP5. We find that even in cells selected for a low level of resistance, several MRP-related genes can be up-regulated simultaneously. However, MRP4 is not overexpressed in any of the cell lines we analyzed; MRP3 and MRP5 are only overexpressed in a few cell lines, and the RNA levels do not seem to correlate with resistance to either doxorubicin or cisplatin. cMOAT is substantially overexpressed in several cell lines, and cMOAT RNA levels correlate with cisplatin but not doxorubicin resistance in a subset of resistant cell lines. Our results emphasize the need for gene-specific blocks in gene expression to define which transporter contributes to resistance in each resistant cell line.

Rabbit epithelial basolateral chloride conductance regulator (EBCR) and rat canalicular multispecific organic anion transporter (Cmoat) are found to be homologues based on protein sequence comparison and Northern blot analysis. EBCRis, therefore, renamed as rabbit Cmoat. The gene encoding CMOAT, a transporter possibly involved in Dubin-Johnson syndrome in humans, is mapped on human chromosome 10q24 and mouse chromosome 19D2.

From My 1992, Petroleum Development Oman (PDO) embarked on a series of field trials to assess the operational feasibility and commercial benefits of Underbalanced Drilling (UBD) for improving the company's oil production. The target formations were depleted sandstone reservoirs and fractured carbonate reservoirs, known for deferred oil production due to flushing by lost circulation. After four foam drilling trials in depleted reservoirs, it was concluded that, although a production increase was identified, a clear commercial quantification of UBD benefits was still lacking. A larger campaign in a single field with considerable offset production data was therefore desirable to establish the production improvement as a result of reduced impairment. Furthermore, UBD had to be extended to Underbalanced Operations (UBO) which includes logging and completing, without killing the well at any point to reap the full benefits. The scope for UBO had to be increased to include free flowing wells. The Nimr field (in South Oman) was identified as a suitable area where many offset wells are available (some 100 wells drilled to date) and where a large application of UBO is envisaged, once it has proven to be commercially attractive. The Nimr field contains viscous (400 cP) oil with traces of H2S in sandstone reservoir at a depth of 950 m with a pressure of 810.4 kPa/m. The wells are completed with a horizontal 7" predrilled liner and 4 1/2" wire wrapped screens over the reservoir. Technical solutions are presented to drill, evaluate and complete these wells without the need to kill. Health, Safety and Environment (HSE) issues of UBO have been addressed and where necessary appropriate measures have been taken to maintain the same level of HSE standards as applicable for conventional overbalanced operations. The criteria and the means to determine the economic merits of UBO were established before embarking on a campaign in the Nimr field. A maximum incremental well cost for the campaign has been set on the basis of the possible productivity increase. A possible increase in rate of penetration (ROP) during the campaign would be an added benefit, but is not considered the main driver behind the project. Horizontal wells in the Nimr field show a significant spread in inflow performance and a statistical approach was applied to determine the number of wells needed to prove with confidence that the break-even productivity increase has been achieved. Pre-campiagn trials for Coiled Tubing logging and shale stability have been conducted. Results of the shale stability trial have led to the decision to discontinue the Nimr UBO campaign because it caused the project to become uneconomical. However, PDO will continue to look for areas where UBO may offer economic opportunities, following the same approach as outlined for the Nimr field! Based on PDO's past experience and the recent developments in coiled tubing operations, it is envisaged that in 1996 a merger will take place between these two key technologies to allow for a technically integrated and commercially more attractive approach to underbalanced operations.

We have developed a vector system for high-copy-number integration into the ribosomal DNA of the yeast Kluyveromyces lactis. This system is analogous to the pMIRY-system developed for Saccharomyces cerevisiae. Plasmids containing a portion of K. lactis rDNA for targeted homologous recombination, as well as the S. cerevisiae TRP1 gene with various promoter deletions, were constructed and, after transformation to K. lactis, analyzed for both copy number and stability. These plasmids were found to be present in about 60 copies per cell and were stably maintained during growth under non-selective conditions. Using this vector system, we expressed a fusion construct containing the S. cerevisiae GAL7 promoter, the SUC2 (invertase) signal sequence and the gene coding for alpha-galactosidase from the plant Cyamopsis tetragonoloba. Although the maximum copy number of these integrated plasmids was only about 15, we nevertheless obtained a high level of alpha-galactosidase production (250 mg/l) with a secretion efficiency of about 95%. When compared to extrachromosomal K. lactis vectors containing the same fusion construct, the multicopy integrants showed a much higher alpha-galactosidase production level and a considerably higher stability under non-selective conditions.

It was analysed to what extent the functional T-cell responses that result from T-cell receptor (TcR)/CD3 triggering differ from responses that are induced after simultaneous ligation of CD2 and CD28 accessory molecules. To allow a quantitative comparison of these activation pathways, purified lymphocytes were stimulated with either graded densities of immobilized anti-CD3 monoclonal antibodies (mAb) or with increasing amounts of anti-CD28 mAb in the presence of a constant concentration of anti-CD2 mAb. Both activation systems were sensitive to the regulatory properties of CD11a/CD18 molecules. T-cell stimulation via CD2/CD28 molecules induced a more potent release of interleukin-2 (IL-2) and more pronounced T helper (Th) cell responses than T-cell stimulation via the TcR/CD3 complex, whereas CD25 expression was more readily initiated after T-cell activation via the TcR/CD3 complex. Optimal Th cell differentiation was detected under conditions of suboptimal receptor occupancy whereas, in contrast, optimal cytolytic T lymphocyte (CTL) differentiation required optimal TcR/CD3 or CD2/CD28 engagement. The findings indicate that T-cell differentiation can be influenced in a qualitative manner by the strength of the activation signal provided, and suggest that antigen-specific T-cell responses might be regulated in a quantitative manner through CD2 and CD28 accessory molecules.

The insect cell-baculovirus expression system has significant potential for producing proteins requiring some degree of posttranslational modification. T. ni cells appear to be as good a host as S. frugiperda cells for heterologous protein production as demonstrated by production of beta-galactosidase. Attachment-dependent cells of T. ni can be effectively cultured in a packed-bed reactor using glass beads. When cell in such a reactor were infected, they produced 35% of the total protein as beta-galactosidase. No cell detachment was observed even 70 h postinfection. A model of viral entry has been proposed and tested.

Many insurers offer policies which provide a combination of insurance coverage up to a fixed retirement age and an annuity income after retirement. Premiums are normally payable until the retirement age is reached. The retirement income is often at the rate of $10 per month for each $1000 of face amount and is customarily payable for a fixed number of years certain and the remainder of life. Under normal circumstances the present value at the retirement age of the annuity income of $10 per month exceeds the face amount of $1000. This means that the policy must nature at the retirement age for an amount greater than the face amount, and that the policy reserves and cash values, in building up to the required maturi ty value, will also exceed the face amount for a period prior to retirement. Since it is impractical to have the face amount of insurance loss than the cash value, these policies provide that the death benefit will be equal to the cash value during the period when the cash value exceeds the face amount. Let 1 ~ k denote the present value at the retirement age, x ~ n, of the annuity to be provided at that age for each unit of face amount issued at age x. Then the policy can be regarded as a combination of a pure endowment of 1 ~ k due at the retirement age and a term insurance up to that age providing a death benefit equal to 1 or the total cash value on the policy whichever is greater.
