Aerosol particles have an important role in many scientific and technological issues. Aerosol particle measurements are widely applied for example in clean room technology, in atmospheric measurements and in studying the Particulate Matter (PM) emissions from traffic and industry. This thesis concentrates on developing new aerosol instrumentation both for measurement and calibration purposes. On the measurement side, the driving force has been the urgent need for instruments that have a fast time response and are able to measure nanoparticles with reasonable accuracy. In this respect, the nanoparticle resolution of the Electrical Low Pressure Impactor (ELPI, Dekati Ltd.) was improved by designing, manufacturing and implementing a new impactor stage (cutpoint 16.7 nm) to the ELPI cascade impactor. The new impactor stage divides the particle size range measured by the filter stage (7–30 nm) between the new stage and the filter stage. As a result, the nanoparticle resolution of the ELPI was improved. This made the device more suitable, for example, for vehicle engine emission measurements. The new stage is currently being sold as a part of the new ELPI+ instrument, which is an improved version of the original ELPI. On the calibration side, the main driving force behind aerosol instrument development has been the lack of calibration standards available for calibrating the number concentration responses of the instruments in the sub-micrometer size range. In this size range, the most common method to calibrate an instrument is to use a differential mobility analyzer (DMA), for obtaining monodisperse particles for the calibration, and a Faraday cup aerosol electrometer (FCAE), for measuring the reference number concentration. Even though, in principle, the DMA allows size selection up to 1 μm in diameter, the calibrations are usually limited to particles below 100 nm because of the multiple charging of particles. To solve this problem, a new concept for realizing a number concentration standard in a wide size range was introduced. In this concept, a novel principle of first charging nanoparticles and growing them afterwards to much larger particle sizes is applied for the generation of the singly charged, fairly monodisperse calibration aerosols. Combined with an FCAE this concept, ideally, solves the calibration issues in the sub-micrometer size range. In order to test this concept, a new instrument called the Single Charged Aerosol Reference (SCAR) was designed, built and tested. In the first experiments, the SCAR was verified to produce singly charged, fairly monodisperse particle size distributions between 10 and 500 nm in diameter and to be suitable for calibration purposes. As a result of a rigorous validation process, true SI-traceability was obtained for the particle number concentration output of the SCAR, and the calibration of other instruments on the absolute scale became possible. As a final necessary step in becoming an internationally recognized number concentration standard, an intercomparison between the SCAR and two number concentration standards of the National Institute of Advanced Industrial Science and Technology was conducted in Japan. The results obtained with the three standards were found to agree within the uncertainty limits at all overlapping particle sizes. As a consequence, a new primary number concentration standard, which enables accurate calibration of various instruments in the whole submicrometer range, was established.

In this study we have applied exome sequencing of the X-chromosome in order to identify a mutation in a Finnish family with X–Linked Intellectual Disability (XLID). We identified a novel mutation in the Cullin 4B gene (CUL4B) that has previously shown to cause Cabezas syndrome. The mutation was identified using Agilent array that covers 93% of the coding region of chromosome X. The mutation is located in exon 20 resulting in premature stop codon in exon 21 where aspartic acid is changed to a premature stop codon D806X. Here we present a detailed clinical phenotype of the three affected brothers. CUL4B is a ubiquitin E3 ligase subunit implicated in the regulation of several biological processes, and CUL4B is the first XLID gene that encodes an E3 ubiquitin ligase. Our findings elucidate the functional significance of CUL4B in human cognition and in other aspects of human development.

Advances in measurement technology and wireless signal transfer have enabled the design of new, smaller and portable—even plaster-like—electrocardiographic (ECG) measurement devices that enable patient monitoring at home or in emergency situations. The development of new, miniaturized biomedical sensors has opened up possibilities for their application, but also set new demands on signal analysis and interpretation. In particular, the new small wireless systems often utilize bipolar electrodes that have a shorter interelectrode distance (IED) and different electrode locations from those of the standard 12-lead system. This affects the quality and the information content of the signal. The general objective of this thesis was to evaluate the performance of short IED precordial bipolar ECG leads and to determine their optimal location. This thesis adopted three methods to assess the properties of new bipolar precordial ECG leads: modeling, body surface potential map (BSPM) data, and exercise ECG data. First, two realistic, three-dimensional (3D) thorax models and lead field analysis were used to evaluate whether modeling of the measuring sensitivity of ECG leads could be used as a tool for designing new ECG leads. Second, BSPM data was used to study whether short-distance bipolar leads (IED approximately 6 cm) provide an ECG signal that is adequate for clinical utilization. Third, BSPM data was used to define where a bipolar ECG lead should be located in order to maximize the ECG signal strength within healthy subjects. Finally, the value of bipolar leads for diagnosing two major cardiac conditions—left ventricular hypertrophy (LVH) and coronary artery disease (CAD)—was assessed. It was found that the modeled measuring sensitivity corresponds to the changes in actual ECG signal strength, so modeling can be useful, especially in cases where in vivo measurements are impossible such as in designing implantable applications. Based on ECG data from 236 healthy subjects, all studied bipolar ECG leads with a short IED (approximately 6 cm) provided a detectable signal when compared to a low noise level of 15 μV and considering the P-wave as the smallest parameter. The optimal location of the bipolar lead was diagonally near the chest electrodes of the standard precordial leads V2, V3, and V4 (to maximize QRS amplitude), or above the chest electrodes of leads V1 and V2 (to maximize P-wave amplitude). In the selected clinical applications, LVH and CAD, the performance of bipolar leads was surprisingly good. In differentiating LVH (n=305) and healthy subjects (n=236), the performance of a correctly positioned small bipolar lead was similar to that of the traditional Sokolow-Lyon method. When differentiating CAD (n=255) patients from non-CAD (n=126) or low-likelihood of CAD (n=198) subjects, the overall performance of bipolar lead CM5 corresponded to that of standard lead V5. These results indicate that short IED bipolar leads provide a signal that is adequate for clinical use. Furthermore, the performance of these leads was shown to be similar or even superior to that of the commonly used standard leads. It can be concluded that when correctly positioned, short IED bipolar leads are useful and can give additional value for clinical diagnostics. These results provide promising information on the applicability and potential of short IED bipolar ECG leads, and demonstrate that they are worth developing further.

Internetissä kuluttajan saatavilla on nykyään valtava, koko ajan kasvava määrä tuotteita ja palveluja. Erilaiset tiedonhaku-, suodatus- ja suosittelumenetelmät auttavat kuluttajaa löytämään haluamansa tuotteen. Erityisesti suosittelujärjestelmien rooli korostuu tarjotessa käyttäjälle uusia elämyksiä tai ennen näkemättömiä tuotteita. Suosituimpia tapoja toteuttaa suosittelujärjestelmä on yhteisöllinen suodatus. Se perustuu suositeltavan tuotteen sisällön analysoinnin sijaan järjestelmän käyttäjien arvioihin eri tuotteista. Web-teknologioiden kehitys on tuonut yhteisöllisyyden ja käyttäjälähtöisen näkökulman osaksi nykypäivän verkkopalveluja. Sosiaalisen median palveluista suosiota ovat keränneet erityisesti sosiaaliset verkostoitumispalvelut, joiden voima perustuu yhteisön luomaan sisältöön ja lisäarvoon. Sosiaaliset verkostot kehittyvät yleensä jonkin kiinnostusaiheen ympärille tai keskittyvät tosielämän sosiaalisten suhteiden mallintamiseen. Tässä tutkielmassa esitetään tapoja hyödyntää käyttäjän sosiaalista verkostoa automaattisissa suosittelujärjestelmissä. Tutkimusten mukaan käyttäjät luottavat suositusten saamiseen enemmän ystäviinsä kuin automaattisiin suosittelujärjestelmiin. Hyödyntämällä olemassa olevaa sosiaalista verkostoa voidaan parantaa suosittelutuloksia ja lisätä luottamusta saatuihin tuloksiin. Työn tarkoituksena on tarkastella sosiaalisen verkoston lisäarvoa suositteluissa ja menetelmän yleiskäyttöisyyttä. Sosiaalisen verkoston vaikutusta selvitettiin simuloimalla järjestelmää, joka hyödyntää sosiaalista verkostoa suositteluun. Tutkimuksessa hyödynnettiin aineistoa olemassa olevasta, yhteisöllistä suodatusta käyttävästä suosittelujärjestelmästä. Sosiaaliseen verkostoitumispalveluun kohdistuneella kyselyllä verrattiin käyttäjien arvioita olemassa olevaan dataan. Havainnot osoittivat, että suosittelutulokset paranivat verrattuna oletustoteutukseen. Vaikka saatuja tuloksia ei voida suoraan vertailla, vahvistavat ne kuitenkin aikaisempien tutkimuksien havaintoja. Idean jatkokehittelyllä onkin mahdollista saada aikaan parempia tuloksia. A user browsing the internet today faces a vast amount of products and services offered, which is also growing daily. Different information retrieval and recommendation methods help the user to find relevant or interesting content. Although many implementations for recommender systems exist, the most popular method is called collaborative filtering. Instead of analyzing the content of the recommended product, recommendations are based on the opinions of users with similar taste. Advances in web technologies have brought a communal and a user-centric view to modern websites. Social media sites – especially social networking sites – have proven popular. The strength of these services is based on user-created content and social networking. Networks form usually around users’ subjects of interest. They might also model the real-life social networks of people. The goal of this study is to present different methods of using a user’s social network in an automatic recommender system. Studies show that users prefer recommendations that are based on the opinions of people they know. By utilizing a user’s existing social network, it is possible to provide more accurate and trustworthy recommendations. The purpose is to evaluate social recommendation methods and provide practical suggestions for implementation. The results for social recommendation were simulated by fusing existing data from a collaborative filtering system with data from a social networking site. Ratings from the social networking website were gathered using a questionnaire and then merged with existing data. The results showed noticeable improvement over the default implementation. Even though these results are not comparable, similar results have been gathered from previous studies. These show that fusing social data with collaborative filtering does indeed yield better recommendation results.

Työn tavoitteena on ollut tutkia työssä rakennetulla kannettavalla liekkigeneraattorilla tuotettuja testiaerosoleja ja niiden käyttäytymistä erilaisissa ympäristöissä. Työtä varten on tehty aerosolimittauksia kolmessa paikassa. Laboratoriomittauksissa tutkittiin tuotettuja hiukkasia lähellä liekkiä, ennen hiukkasten suurempaa agglomeroitumista. Saksalaisen tutkimuslaitoksen tuulitunnelissa tehdyissä mittauksissa selvitettiin eri prosessitekijöiden vaikutuksia tuotettuun aerosoliin, sekä minkälaista aerosolia kyseiseen testitilaan voidaan liekkigeneraattorilla tuottaa. Tampereen teknillisen yliopiston paperinjalostus- ja pakkaustekniikan laboratorion pilot-linjalla tutkittiin paperin nanohiukkaspinnoitukseen tuotetun aerosolin emissiota pinnoitusprosessista huoneilmaan. Testiaerosoli tuotettiin kannettavalla liekkigeneraattorilla tai tätä vastaavalla kiinteällä liekkigeneraattorilla. Mittaukset suoritettiin yleisesti käytössä olevilla aerosolimittalaitteilla sekä yhdellä mittalaitteen prototyypillä. Aerosolista mitattiin niin hiukkaskokojakaumaa, lukumääräpitoisuutta kuin pinta-alapitoisuuksiakin, mittaustilanteen ja tarpeen mukaan. Primääriaerosolimittausten perusteella pystytään arvioimaan tuotetun aerosolin kehittymistä muissa ympäristöissä, kun tiedetään liekkigeneraattorilla tuotettujen hiukkasten koko ennen kuin ne pääsevät agglomeroitumaan, ja verrataan tätä kehittyneen aerosolin mittaustuloksiin. Tuotettujen hiukkasten koko vaihtelee liekkigeneraattorin käyttöön liittyvien parametrien vaihtelun mukaan. Tuulitunnelissa suurin vaikutus hiukkaskokoon on tuulitunnelivirtauksella. Suurilla virtauksilla hiukkaset eivät juurikaan pääse agglomeroitumaan suuren laimennuksen vuoksi, kun taas pienemmillä virtauksilla hiukkaskoko voi kasvaa moninkertaiseksi aerosolin kulkeutuessa tuulitunnelin läpi mittauskammioon. Paperin pinnoituksesta pääsee, kohdepoistosta huolimatta, hiukkasia huoneilmaan, ja nämä agglomeroituvat jonkin verran prosessiparametreista riippuen. Hiukkaskoon vaihtelut prosessiparametrien mukaan eivät ole niin selkeitä hiukkasten agglomeroituessa vapaassa huoneilmassa, kuin tuulitunnelissa. Eri tilanteissa tuotettujen hiukkasten keskikoko vaihteli 7–115 nanometrin välillä. Liekkigeneraattorilla tuotetut hiukkaset kehittyvät eri tavoin erilaisissa ympäristöissä. Laimennusilmasta ja muista tekijöistä riippuen tuotetut aerosolihiukkaset voivat agglomeroitua eri tavoin, ja erilaisissa ympäristöissä tehtyjä mittauksia ei voi sinällään suoraan soveltaa uuteen ympäristöön. Aerosolin kehittymistä ei välttämättä voida myöskään arvioida koagulaation teorian avulla luotettavasti, sillä esimerkiksi hiukkastuotto on liekkiruiskun käyttötilanteissa usein jatkuvaa, eikä teoria ota tätä huomioon. Jatkossa paperin pinnoituksen hiukkasemissioita voisi tutkia tarkemmin mittaamalla poistohuuvaan menevää aerosolia. Kannettavan liekkigeneraattorin osalta kehitystyötä puolestaan jatketaan systeemin vielä kompaktimpaan ja helpommin liikuteltavaan muotoon saattamisessa.

Ion gels composed of ionic liquids and block copolymers can be used as gate dielectrics in organic thin-film transistors. Conventional dielectrics used in OTFTs do not have capacitances large enough to enable transistor operation at low voltages, which is needed in many organic electronics applications. Mobile ions in the gel form electric double layers, giving rise to a very high capacitance and therefore high charge carrier concentrations at low operating voltages. The goal of this study was to fabricate a functioning OTFT using an ion gel as gate dielectric, and to examine its properties. The OTFT was prepared with a top-gate geometry, where the ion gel was sandwiched between the gate electrode and the substrate with the semiconductor channel. The semiconductor was amorphous poly(triarylamine), which was deposited by spin-coating. The ion gel gated OTFT properties were compared with a reference OTFT structure, which had a conventional polymeric gate dielectric. The ion gel gated transistor operated with voltages below 2 V and the output current was three orders of magnitude larger than that of the reference OTFT, which required more than ten times the voltage. The capacitance of the ion gel was over 5 uF/cm^2 up to 1 MHz. The charge carrier mobility in the OTFT was on the order of 10^-3 cm^2/(Vs), which was similar to that in previous studies with the semiconductor used. The transistor could be switched on and off at the frequency of 1 kHz, but there was a large overlap capacitance across the gel. Hysteresis and leakage were challenges in the ion gel gated OTFT preparation. The ionic liquid is hygroscopic, making the gel susceptible to water impurities. The impurities can cause leakage and hysteresis due to electrochemical reactions at the electrodes even at low voltages. Future work with ion gel gated OTFTs will require a more controlled environment in terms of ambient humidity. /Kir12

Pyyhkäisytunnelointimikroskopia (eng. Scanning tunneling microscopy, STM) on yksi modernin pintatieteen tärkeimmistä työkaluista. Se mahdollistaa atomirakenteen kuvantamisen reaaliavaruudessa ja pintojen tutkimisen atomimittakaavassa. Menetelmä perustuu kvanttimekaaniseen tunneloitumiseen, jonka ansiosta paikallisen kärjen ja tutkittavan pinnan välille syntyy virta. Kun kärkeä liikuttamalla käydään koko pinta läpi, saadaan topografinen kuva, joka antaa tietoa pinnasta atomitasolla. Kokeelliset tulokset metallipinnalle adsorboituneiden molekyylien pyyhkäisytunnelointimikroskopiasta ovat osoittaneet, että saatu topografinen kuva ei välttämättä vastaa tutkitun systeemin geometrista rakennetta. Siksi kokeellisten STM-kuvien ymmärtämisessä ja tulkitsemisessa STM:n teoreettinen ja laskennallinen mallintaminen on oleellista. Tämä diplomityö keskittyy metallipinnalle adsorboituneiden molekyylien pyyhkäisytunnelointimikroskopian mallintamiseen. Tavoitteena on testata GPAW-ohjelmiston toimivuutta mallinnuksessa. Simulaatioissa tutkitaan, miten adsorbaattimolekyylin lisääminen metallipinnalle vaikuttaa STM-kuviin. Tämä tehdään simuloimalla sekä puhdasta metallipintaa että pintaa, jolle on adsorboitunut molekyyli. Lisäksi työssä simuloidaan elektronien transmissiota molekyylien läpi, jotta adsorbaattimolekyylien vaikutus elektronien tunneloitumiseen saadaan selville. STM-kuvat saadaan puhtaalle Al(111)-pinnalle ja pinnalle, jolle on adsorboitunut happimolekyyli. Kuvissa adsorbaattimolekyyli nähdään pinnalla painaumana eikä kohoumana kuten geometrisen rakenteen perusteella oletettaisiin. Kuvat näyttävät, että adsorbaattimolekyylin lisääminen vaikuttaa laajasti koko STM-kuvaan. Transmissiosimulaatiot eri molekyylien läpi osoittavat molekyylin elektronirakenteen vaikuttavan elektronien tunneloitumiseen. GPAW:n STM-simulaatiopaketin todetaan toimivan vain tiettyjen systeemien STM-simuloinnissa.

Tässä työssä GNSS-satelliitin (engl. Global Navigation Satellite System) ennustettu kiertorata parametrisoidaan, eli muutetaan parametrimuotoon, jota GPS-järjestelmä (engl. Global Positioning System) käyttää efemerideissään. Kiertoradan ennustuksessa saadaan paikkakoordinaatteja karteesisessa koordinaatistossa tietyillä ajanhetkillä, mihin parametrimuoto sovitetaan. Parametrimuoto ei sovellu täysin ennustettuun kiertorataan, joten parametrit täytyy estimoida siten, että virhettä syntyy mahdollisimman vähän. Tämä on epälineaarinen optimointiongelma, joka ratkaistaan tässä työssä Levenbergin ja Marquardtin ratkaisumenetelmällä. Työssä keskitytään parametrimuodon esittämiseen ja geometrisen merkityksen tutkimiseen sekä epälineaarisen pienimmän neliösumman optimointiongelman numeeriseen ratkaisemiseen. GPS-järjestelmässä satelliittien lähettämä navigointiviesti vastaanotetaan aina parametrimuodossa ja olisi hyvä tietää miten nämä parametrit saadaan laskettua aikasarjasta satelliitin paikkakoordinaatteja. Tällöin eri järjestelmien efemeridit sekä satelliiteille tehdyt rataennusteet saadaan yhtenevään muotoon ja paikannuslaitteessa tehtävä datan käsittely ja tallennus helpottuu. Tässä työssä esitettyä mallia testataan GPS- ja GLONASS-satelliiteille (venäj. Globalnaya Navigatsionnaya Sputnikovaya Sistema) tehdyille rataennusteille, sekä precise-efemerideistä saaduille oikeille kiertoradoille. Sovitusvirhe saadaan kahden tunnin mittaiselle sovitukselle alle 10 cm:n suuruiseksi. Neljän tunnin pituisella sovitusvälilläkin päästään vielä alle 40 cm:n virheeseen. Näin ollen satelliitin ennustetun kiertoradan parametrisointi voidaan katsoa onnistuneeksi. Työssä kehitelty algoritmi sopii hyvin satelliiteille, jotka kulkevat GPS:n ja GLONASS:n kaltaisilla kiertoradoilla, joten soveltaminen onnistuu myös muillekin GNSS-satelliiteille.

Tämä diplomityö on osa laajan SAFIR 2014 (Safety of Nuclear Power Plants –Finnish National Research Programme) -tutkimusohjelman WAPA (Water Chemistry and Plant Operating Reliability) -projektia, jonka tavoitteena on kehittää malli lämpötilan, kiehumisen, hydrodynaamisten olosuhteiden ja rakennemateriaalien vaikutuksista magnetiitin saostumiseen painevesilaitoksen primääri- ja sekundääripiireissä. Työssä perehdyttiin kolloidisten magnetiittipartikkelien (Fe3O4) saostumiseen vaikuttaviin tekijöihin ja tutustuttiin kolloidihiukkasten saostumiseen vaikuttavan zeta-potentiaalin (ζ) mittaamiseen soveltuviin kokeellisiin menetelmiin. Työn tavoitteena oli myös mitata kolloidisten magnetiittipartikkelien ζ-potentiaalia painevesilaitoksen sekundääripiirin olosuhteissa. Kolloidisten hiukkasten stabiilisuuteen ja niiden toisiinsa sekä pinnoille kiinnittymiseen liittyvistä vuorovaikutuksista tärkeimmät ovat van der Waals’n vetovoima ja sähköstaattinen voima. Kolloidipartikkelien pinta on vesiympäristössä yleensä sähköisesti varautunut. Tietoa pinnan sähköisestä tilasta saadaan ζ-potentiaalin avulla. Jos se on lähellä nollaa, hiukkasilla on taipumus kiinnittyä toisiinsa ja muodostaa saostumia pinnoille. Jos sen itseisarvo on suuri, hiukkaset hylkivät toisiaan, mutta saattavat saostua niiden varaukseen verrattuna vastakkaismerkkisesti varatulle pinnalle. Tässä työssä magnetiitin ζ-potentiaalia tutkittiin streaming-potentaaliin perustuvalla menetelmällä. Ammoniakki- ja rikkihappoliuoksissa huoneenlämpötilassa hapettomissa olosuhteissa happamuuden funktiona suoritettujen mittausten perusteella magnetiitin isoelektrinen piste näyttää olevan välillä 4 - 5, jolloin kolloidiset magnetiittipartikkelit pyrkivät siis saostumaan. Huoneenlämpötilassa pH-arvoon 9,2 säädetyssä ammoniakkiliuoksessa hapettomissa olosuhteissa lämpötiloissa 139 °C ja 143 °C ζ- potentiaali oli negatiivinen ja sen itseisarvo laski lämpötilan funktiona. Magnetiitti näytti saavuttavan isoelektrisen pisteensä lämpötilan noustessa hieman yli 143 °C:n. Lämpötilan noustessa kolloidisten magnetiittipartikkelien yhteen kiinnittymisen todennäköisyys vaikuttaa siis kasvavan ja ne saattavat sen seurauksena alkaa saostua pinnoille. Zeta-potentiaalin havaittiin olevan negatiivinen myös morfoliini- ja etanoliamiiniliuoksissa huoneenlämpötilassa hapettomissa olosuhteissa pH-arvon ollessa 9,2. Itseisarvoltaan se oli kuitenkin paljon suurempi etanoliamiiniliuoksessa. Ammoniakkiliuoksessa zeta-potentiaali sijoittui näiden arvojen väliin. Tulosten perusteella näyttää siis siltä, että huoneenlämpötilassa kolloidisten magnetiittipartikkelien saostumiseen voidaan vaikuttaa happamuuden säätämiseen käytettävien kemikaalien avulla. Tulevaisuudessa tarvitaan lisää mittauksia magnetiitin ζ-potentiaalin selvittämiseksi korkeissa lämpötiloissa, sillä aiheesta tehtyjä tutkimuksia on vain muutamia ja saadut tulokset poikkeavat toisistaan. Lisäksi happamuuden säätökemikaalin vaikutusta kolloidisten magnetiittipartikkelien ζ-potentiaalin arvoon tulee tutkia eri lämpötiloissa, koska pH:n säätökemikaaleilla näyttää olevan vaikutusta ζ-potentiaaliin. This Master of Science thesis is a part of the WAPA (Water Chemistry and Plant Operating Reliability) project of SAFIR 2014 (Safety of Nuclear Power Plants -Finnish National Research Programme) research programme. The goal of the project is to develop a model for the deposition of magnetite in pressurized water reactor (PWR) primary and secondary circuit conditions including the effects of temperature, boiling, hydrodynamical conditions and structural materials. The purpose of this thesis was to investigate the factors affecting the deposition of colloidal magnetite (Fe3O4) particles and to get acquainted with the methods for measuring zeta potential (ζ). In addition, the aim was to measure the zeta potential of colloidal magnetite in PWR secondary circuit conditions. The most important factors affecting the stability of colloidal particles, their tendency to form aggregates and their ability to deposit onto surfaces are the van der Waals and electrostatic forces between the particles. The surface of colloidal particles in aqueous media is usually charged. Information on the electrical state of the surface can be obtained with ζ potential measurements. If ζ potential is close to zero the particles tend to form aggregates and deposit onto surfaces. If the absolute value of ζ potential is large the particles tend to repel one another. In this case, however, the particles may form deposits onto oppositely charged surfaces. The current research included measuring the ζ potential of magnetite with a method based on streaming potential. According to the measurements of ζ potential as a function of pH in ammonia and sulphuric acid solutions in anoxic conditions at room temperature it seemed that the isoelectric point of magnetite was between 4 - 5 meaning that the colloidal magnetite particles tend to form aggregates and deposits under these conditions. The ζ potential was found to be negative in anoxic conditions at temperatures 139 °C and 143 °C in ammonia solution, the pH of which was adjusted to 9,2 at room temperature. The absolute value of ζ potential decreased as a function of temperature, and magnetite seemed to reach its isoelectric point when the temperature was just above 143 °C. Therefore, the probability of colloidal magnetite particles to form aggregates and deposits seems to increase as the temperature rises. The ζ potential was also found to be negative in morpholine and ethanolamine solutions of pH 9,2 in anoxic conditions at room temperature. The absolute value was, however, bigger in ethanolamine solution. The ζ potential in ammonia solution was in between those. Thus, it seems that at room temperature the deposition of colloidal magnetite particles can be affected by changing the chemical used to adjust the solution pH. Future work should be devoted to measuring the ζ potential of magnetite at high temperatures because the research already done is scarce and the results obtained differ from each other. In addition, the effect of chemical used to adjust solution pH should be studied at different temperatures because it seems to have an effect on ζ potential.

Electroencephalography (EEG) is the recording of potential fluctuations originating from electrical activity of brain. It is used in diagnosis of neurological disorders, monitoring the depth of anaesthesia and evaluation of sleep. A possible application is long-term continous monitoring of intensive care unit (ICU) patients, with the aim to detect epileptic seizure activity. Currently this is not practised due to poor usability of the available equipment. This thesis is part of a project which aims to develop a novel solution which solves the usability problem. Adding to that, the solution contains an algorithm for automatic online epileptic seizure detection, which enables immediate treatment of patients once epileptic seizures occur. This leads to an increased patient outcome at ICUs. An important factor contributing to poor usability of current EEG equipment is the electrode contact. To ensure good signal quality, skin under the electrodes needs to be prepared by abrasion for example. This is time-consuming especially when multiple electrodes are used. Adding to that, skin preparation damages skin which is undesired especially in long-term applications, as the the presence of electrode and electrolytic gel causes irritation and possibly an infection risk. The quality of the electrode contact is quantified by its electrical impedance. In this thesis the relationship of electrode contact impedance to total electrode contact noise and motion artifact magnitude are studied. These both are factors contributing to biopotential signal quality. Sintered silver-silverchloride electrodes are used in the work. Contact impedance is defined as the magnitude of the impedance vector at 20 Hz. Contact noise is studied by measuring impedance-noise data pairs (n=122) at two body sites of volunteer subjects using two different electrode gels. A univariate analysis of variance is implemented on the data pairs. Motion artifact magnitude is studied with impedanceartifact magnitude data pairs (n=33) while producing a horizontal motion to the electrode. The behavior of the seizure detection algorithm is also studied by adding different amounts of noise to EEG signals, and assessing how its behavior changes. The results show that electrode contact impedance can be used as a rough predictor of total contact noise. With contact impedance in the range of 20 k or less, the contact noise is expected to settle at RMS values less than 5 μV at a 30 Hz bandwidth. It was also found the that the type of electrolytic gel can have a significant effect on total contact noise. Motion artifact magnitude was found to decrease with decreasing contact impedance. With larger contact impedance values, the variations of motion artifact magnitudes were larger. The behavior of the seizure detection algorithm was found to change significantly with a small amount of noise added to EEG signals. By comparing that amount of noise to the measured contact noise data, it can be seen that it is well within the measured values.

Tissue engineering is expected to fulfill its promises to produce knowledge, methods, and products that would benefit the health care sector. The need of a degradable construct is a necessity when trying to grow a living tissue inside a porous structure. The fibers offer a unique approach to this, since the porosity can easily be altered with different methods especially using the textile methods. Textile industry on the other hand offers a broad and variable archive of methods to manufacture products and preforms. These preforms can be further used together with the composite technology to further increase the number of applications to which they can be used. When combining different fields of sciences it is possible to manufacture a living tissue, based on fibrous degradable constructs. Thus fibers and fibrous products are versatile preforms that can be shaped and designed to fulfill many of the demands in the field of tissue engineering. The main objective of this thesis was to study the biodegradable medical grade P(L/D)LA 96/4 polymer fibers and the fibrous products manufactured from them. The thermal degradation behavior of this polymer in single screw extruder environment in melt spinning process was studied. The online hot-drawing was used for fiber orientation. The information based on those studies was analyzed and gathered for future optimization of such melt spinning processes. Different batches and fiber diameters were produced using different processing parameters. Textile methods were further introduced to the melt-spun fibers. The effect of the knitting parameters on fiber properties were studied during the hydrolytic degradation and long term storage. These knits were studied as in vivo soft tissue implant preforms, in vitro spinal fusion cage composite preforms, and in vivo temporomandibular jaw implant preforms. Non-woven fabrics were produced from the melt-spun fibers manufactured by hot-drawing and high-speed drawing. These non-woven felts were used for plasma and solvent based wetting enhancement tests accompanied by cell seeding tests. The felts were also used as a component for temporomandibular jaw implant preforms. The results showed increasing degradation in the extruder barrel in regards to the P(L/D)LA 96/4 polymer molecular weight and viscosity. Lactide monomer was induced into the polymer during the melt spinning process due to the temperature and shear. Monomer content also varied according to the polymer residence time. Fiber showed different hydrolytical degradation behavior depending on the monomer amount. Higher monomer content caused rapid degradation of the mechanical properties and molecular weight. We acquired three different degradation profiles for the fibers that were related to the monomer content. The knitted soft tissue implants degraded faster during the hydrolysis than in in vivo. These knitted soft tissue implants acted as real tissue engineered implants; the cells grew and filled in the scaffolds. While the tissue matured in the scaffolds, the measured mechanical properties of the scaffolds improved despite the loss of mechanical strength of an empty scaffold. Non-woven scaffolds increased their wetting capability due to the oxygen plasma treatment. This also enhanced the cell attachment and proliferation.

Työn tarkoituksena oli kartoittaa 4D-mallinnuksen mahdollisuuksia Metso Power Oy:n ja erityisesti asennussuunniteluosaston näkökulmasta. 4D-mallinnusta tarkasteltiin paitsi asennussuunnittelua helpottavana työkaluna, myös välineenä tuottaa materiaalia asennuskonseptia selittävää asennuskuvausta varten. Tavoitteena oli toteuttaa rajoitettu 4D-malli todelliseen voimakattilaprojektiin ja syntyneen kokemuksen kautta määrittää toimintatapa ja ohjeet 4D-mallin muodostamiseksi. Toisena tavoitteena oli kehittää jo aiemmin aloitettua telinemallinnusta uuden alihankkijan kanssa. Työn alussa taustoitetaan kirjallisuuden avulla kattilatoimituksille ominaista projektiympäristöä, esitellään mitä asennussuunnittelulla tarkoitetaan ja kerrotaan, kuinka asennussuunnittelu liittyy toimitusprojektiin. Seuraavaksi esitellään kirjallisuustutkimuksen kautta 4D-konsepti ja sen mahdollisuudet osana asennussuunnittelua. Työn empiirisessä osassa pohditaan 4D-mallinnuksen mahdollisuuksia ja hyötyjä kohdeyrityksen voima- ja soodakattilaliiketoiminnassa. Lisäksi esitellään työn aikana aloitetun mallinnusprojektin keskeiset havainnot ja mallin toteuttamiseen tarvittavat työkalut, sekä menettelytapa mallin tuottamiseksi. Valmista 4D-mallia ei työn puitteissa ehditty saada valmiiksi resurssipulan ja työsuhteen päättymisen vuoksi. Mallinnusta tehtiin kuitenkin niin pitkälle, että keskeiset ongelmakohdat tulivat esiin ja mallin tuottamiseksi tarvittavat toimenpiteet saatiin määritetyksi. Telinemallinnukseen löydettiin työn aikana tehokkaampi ja toimivampi toteutustapa aikaisempaan verrattuna. Työn lopputuloksena todetaan, että 4D-mallinnus on työkalu, jossa on paljon potentiaalia kohdeyritykselle ja asennussuunnitteluosastolle. Suurimpana haasteena mallinnuksen käyttöönotossa on mallin tekemiseen ja ylläpitämiseen vaadittava suuri työmäärä. Myös tarvittavan tietotekniikan omaksuminen kohdeyrityksen asennussuunnitteluosastolla voi hidastaa mallinnuksen hyödyntämistä. Lisäksi konservatiivinen suhtautuminen vastaavanlaisiin uusiin työkaluihin, erityisesti asennustyömaalla, hankaloittaa 4D-mallinnuksen käyttöönottoa.

Tässä tutkimuksessa analysoidaan Suomessa vuosina 2004-2008 poliisin rekisteröimää 83509 tieliikenneonnettomuutta, joissa loukkaantumisiin johti enemmän kuin joka viides ja kuolemaan 1203 onnettomuutta; näissä enemmän kuin yksi ihmishenki menettiin 90 tapauksessa. Vaikka tieliikennekuolemat ovat lukumääräisesti olleet viime vuosina laskussa, on yksikin kuollut tai loukkaantunut liikaa. Kaikki mikä voidaan tehdä onnettomuuksien vähentämiseksi, on tehtävä. Analysoimalla onnettomuusdataa voidaan mahdollisesti löytää sellaisia syitä ja riippuvuuksia, jotka eivät ole olleet liikennesuunnittelijoiden, lainsäätäjien ja muiden asiantuntijoiden tiedossa ja joihin ei sen vuoksi ole osattu tarttua.Tutkimus on osa Tampereen teknillisen yliopiston matematiikan laitoksen sovelletun logiikan tutkimusryhmän toimintaa. Käyttämämme tutkimusmetodi on GUHA-tiedonlouhintamenetelmä, jonka soveltuvuutta tieliikenneonnettomuusdatan analysoimiseen tässä tutkimusprojektissa selvitetään. Sovelletun logiikan tutkimusryhmällä ei ole erityistä tieliikenneproblematiikan asiantuntemusta, mutta valitsemalla tieliikenneonnettomuusaineisto GUHA-menetelmän testiaineistoksi haluamme toisaalta testata menetelmän käyttökelpoisuutta reaaliseen, kaikille suhteellisen helposti ymmärrettävään dataan, ja toisaalta toteuttaa osaltamme uuden yliopistolain yliopistolle asettamaa tehtävää toimia vuorovaikutuksessa muun yhteiskunnan kanssa sekä edistää tutkimustulosten yhteiskunnallista vaikuttavuutta. Tutkittua tieliikenneonnettomuusdataa on analysoitu myös Jyväskylän yliopistossa louhimalla sitä muilla tiedonlouhintamenetelmillä. Tämän selvityksen eräänä tavoitteena on tutkia, voidaanko GUHA-menetelmallä löytää myös sellaisia riippuvuuksia, joita tutkimuksessa ei ole raportoitu.The suitability of GUHA-data mining method for analyzing a data set containing more than 80.000 road accidents occurred in Finland in 2004 - 2008 is examined in this report. By GUHA-method, implemented to LISp-Miner software, more than 10.000 dependencies was found; about 100 easily understandable of them are presented here. Our conclusion is that GUHA-method is useful when one wants to explore relatively small size, but still significant dependencies between subsets of a given data.

The Ecosan Promotion Project (EPP) in Kenya, funded by EU, GTZ and Sida, was promoting appropriate ecological sanitation (ecosan) technologies during the project period (2006-2010). Urine Diversion Dry Toilets (UDDTs) were built in 73 primary and secondary schools throughout Kenya with four units in each school. The purpose of the research in question was to monitor and post-evaluate UDDTs implemented by the EPP, two years after the constructions. In November 2011 and January 2012 five schools were visited and evaluated in southern Nyanza province and five in Western province in Kenya. The schools were pre-evaluated and selected for this research on the basis of their good performance in operation and maintenance (O&M) of their UDDTs. The constructions of the UDDTs were financed by EPP but maintenance is organized and funded by the schools themselves. Generally a common problem for sanitation projects is poor long term maintenance of the facilities, after the donor funding for construction and initial training has ended. Monitoring and post-evaluation of the UDDTs focused on how the facilities are being managed and used. Methods for this were on-site observations, interviews and discussions together with teachers, pupils and caretakers. The research objective was to determine the reasons for the good practice, but also to identify the problems and reflect the results to improve O&M systems in the future. One major problem observed was gradual or sudden disappearance of the initial motivation and knowledge from the school, often when the person in charge of sanitation left the school. According to the results of this research, an important factor motivating the schools to keep their UDDT facilities clean and in good condition, were the actual benefits received from the sanitation systems such as clean and comfortable toilet facilities, healthier environment, e.g. very low risk for ground water pollution and spreading of pathogens into the surroundings, saved space on the school yard as UDDTs can last long at the same spot, and possibility to use the UDDT products either as fertilizers or soil conditioner. These benefits and good results lead to motivation and deeper engagement and to continuity. Especially utilization of UDDT products indicates that the concept of ecological sanitation and technology of UDDTs have been accepted.

Internetin räjähdysmäinen kasvu on tehnyt entistä suuremmaksi haasteeksi löytää merkitykselliset ja luotettavat sivustot harmaasta massasta. Tähän tarkoitukseen on kehitetty erilaisia arvostusalgoritmeja, joilla pyritään asettamaan eri sivustot objektiiviseen paremmuusjärjestykseen. Merkittävämmät tällaiset algoritmit ovat Pagerank ja HITS, jotka molemmat ovat lähtöisin internetin hakukoneista, ja perustuvat internetin linkkirakenteeseen. Näiden algoritmien hyödyntämismahdollisuudet eivät kuitenkaan rajoitu ainoastaan internetin hakukoneisiin, ja näillä on käytännössä mahdollista tarkastella lähes minkälaista verkostoa tahansa. Näiden arvostusalgoritmien matemaattinen käsittely perustuu pitkälti graafiteoriaan sekä matriisilaskentaan, graafiteorian tarjoten työkalut verkostojen mallinnukseen ja visualisointiin, ja matriisilaskennan luodessa pohjan arvostusten laskemiselle. Nämä kaksi matematiikan osa-aluetta nivoutuvat siististi yhteen, muodostaen elegantin kokonaisuuden arvostusalgoritmien käsittelylle. Tässä diplomityössä perehdytään eri arvostusalgoritmeihin, sekä näiden matemaattiseen taustaan. Esimerkkidatana käytetään Tampereen teknillisen yliopiston vuoden 2010 opinto-oppaan kurssien esitietoketjuja, joista muodostuu käsiteltävä verkosto. Ilmiönä esitietoketjut eroavat jonkin verran internetin linkkirakenteesta, mutta tuloksissa havaitaan että algoritmit toimivat hyvin myös tämänkaltaisen verkoston tapauksessa. Pääsääntöisesti eri algoritmit tuottavat saman suuntaisia arvostuksia eri kursseille, mutta eri algoritmit painottavat kukin hieman eri asioita. Näin ollen eri algoritmien toiminnan tunteminen on ensiarvoisen tärkeää, kun pohditaan näiden hyödyntämistä eri ilmiöiden tarkastelussa.

This thesis is made to UPM Energy for FUSEC (Future Fuels for Sustainable Energy Conversion) project of Tekes and the aim of this thesis is to increase knowledge and gather information about the properties of critical solid fuels and fuel mixtures, to find out the problems caused by biomass power plants, especially bubbling fluidized bed (BFB) boilers, and to find correlations between fuels and failures. These problems affect the availability of the power plant and maintenance costs, so the thesis also aims to create a procedure tools for continuous benchmarking and managing these problems. Legal obligations, as well as climate change caused pressures to increase the use of biomass based fuels in combustion plants. Fuels become more difficult and demanding, so finding correlations between fuels and the problems is justified. Knowledge of composition and characteristics of the fuel mixture are an important part of the controlling of the problems of boilers. The work is divided into theoretical and practical parts. In the theoretical part are introduced characteristics of the fuels and the combustion process, combustion technologies and problems in the combustion process. The sources of the theory have been used in the literature, field studies, as well as the knowledge and experience from power plants and experts. The practical part is divided into two parts. In the first part, the tool has been created, which can be estimated due to the problems of fuels incidence. The second part of has been studied in four different BFB boilers of UPM and fuels used in them. It was the aim to find explanations between used fuels and observed problems with the analysis of the given fuel data. The result of the analysis showed that the systematic gathering of fuel data played a key role in detecting problems. In order to detect problems, must fractions of fuel, the fuel mixture and mixture ratios know well. The estimation tool of the properties of the fuel mixture and the problems will help with this issue. On the grounds of fuel data from power plants was plotted graphs, which support the data of the characteristics of the fuel fractions. Between of used fuels and the problems, the graphs did not found clear correlations, because the problems are complex and due to many different reasons. The fuel data from the power plants was also partially incomplete and variety, so deep analysis was challenging to make. As further actions, I propose a systematic fuel data collection on a monthly basis, as well as the survey of potential of real-time measurement systems. After better fuel data receiving, can be gone deeply into the correlations between fuels and the failures. Tämä diplomityö on tehty UPM Energialle Tekesin FUSEC (Future Fuels for Sustainable Energy Conversion) projektin puitteissa ja tavoitteena on lisätä tietoutta polttoaineiden ominaisuuksista, kerätä tietoa kriittisistä polttoaineista sekä niiden vaikutuksista kerrosleijukattiloiden (BFB) ongelmiin etsimällä korrelaatioita polttoaineiden ja ongelmien välille. Nämä ongelmat heijastuvat suoraan kattiloiden käytettävyyteen ja sitä kautta kustannuksiin. Työn tavoitteena on myös luoda työkaluja ongelmien arviointiin ja hallintaan. Lakisääteiset velvoitteet sekä ilmastonmuutoksen aiheuttamat paineet kasvattavat biopohjaisten polttoaineiden käyttöä polttolaitoksissa. Polttoaineet muuttuvat vaikeammiksi ja vaativammiksi, joten syy-seuraussuhteiden löytäminen polttoaineiden ja ongelmien välille on perusteltua. Polttoaineseoksen koostumuksen ja ominaisuuksien tunteminen on tärkeä osa kattilaongelmien hallinnassa. Työ on jaettu teoria- sekä käytännön osaan. Teoriaosassa tutustutaan polttoaineiden ja polttoprosessin ominaisuuksiin, polttotekniikkaan ja poltosta aiheutuviin ongelmiin. Lähteinä teoriaosalle on käytetty kirjallisuutta, alan tutkimuksia sekä yrityksen sisältä saatuja tietoja ja kokemuksia. Käytännön osa jakautuu kahteen osaan. Ensimmäisessä osassa on luotu työkalu, jolla voidaan arvioida polttoaineseoksen aiheuttamia ongelmia. Toisessa osassa on tutkittu yrityksen neljää eri BFB-kattilaa ja niissä käytettyjä polttoaineita. Tarkoituksena oli löytää korrelaatioita käytettyjen polttoaineiden ja havaittujen ongelmien välille analysoinnin avulla. Analysoinnin tuloksena havaittiin, että polttoainetietojen järjestelmällinen kerääminen on avain asemassa ongelmien havaitsemisessa. Jotta ongelmia voidaan havaita, polttoainejakeet, seos ja seossuhteet täytyy tuntea hyvin. Polttoaineseoksen ominaisuuksien ja niistä aiheutuvien ongelmien arviointityökalu edes auttaa tässä ongelmassa. Laitoksilta saadun polttoainedatan perusteella tehtiin kuvaajia, jotka tukevat tietoja kyseisten polttoainejakeiden ominaisuuksista. Käytettyjen polttoainemäärien ja ongelmien välille ei kuvaajien avulla löytynyt selviä korrelaatioita, sillä ongelmat ovat monitahoisia ja johtuvat monista eri syistä. Laitoksilta saatu polttoainedata oli myös osittain puutteellista, joten syvällisiä analysointeja oli haastava tehdä. Jatkotoimenpiteinä ehdotan järjestelmällistä polttoainedatan keräämistä kuukausittain sekä mahdollisten reaaliaikaisten mittausjärjestelmien kartoittamista. Tarkemman polttoainedatan saamisen jälkeen voidaan syventyä polttoaineiden ja ongelmien välisiin riippuvuuksiin.

The multiple melting behavior is a common phenomenon observed by differential scanning calorimetry (DSC) for polymorphic polymers. In the case of poly(lactic acid) (PLA) this is detected as double melting peaks, which have been proposed to arise from α- and α’- crystalline forms of the polymer. Various explanations have been presented for the mechanism behind this behavior, namely melt-recrystallization, multiple lamellae population and multiple crystal structure, but so far none of them has been fully confirmed. Determination of the thermal properties of PLA is crucial, because the polymer is used as a material for medical applications alongside food packaging. PLA is known to degrade via hydrolysis under certain thermal and humidity conditions, for which reason the degradation time and rate should be taken into account. At body temperature the plasticizing effect of water can be utilized in the activation of the shape-memory of amorphous poly(D,L-lactic acid) (PDLLA). In this thesis, an investigation of the double melting behavior of the poly(L,D-lactic acid) (P(L/D)LA) samples with two different D-contents by a new temperature-modulated differential scanning calorimetry (TMDSC) method, TOPEM is presented. The aim of the study is to determine the suitable parameters for TOPEM studies in the melting region and establish, whether this method is convenient to measure the phenomenon in question. The other part of the study concentrates on the examination of the plasticizing effect of water by Fourier transform infrared (FTIR) spectroscopy. In addition, the effect of gamma sterilization on the dry and wet samples is investigated. The results obtained by TOPEM correlate well with the ones attained by the conventional DSC method. The required crystallization temperature and time for the appearance of double melting peaks in the DSC curves are different for the samples with different D-contents. In both cases, the major part of melting is observed to be non-reversing. This is explained by superheating of the samples, which is assumed to be due to the slow melting kinetics during TOPEM measurements. Reversible melting in turn occurs simultaneously with the crystallization process, which is detected in the non-reversing heat flow curve. This supports the melting-recrystallization model suggested in previous studies. The plasticizing effect of water is observed clearly in certain IR-absorption bands in the region of 3800–3400 cm-1. A new band appearing in the region of 1700–1500 cm-1 is assigned to the bending mode of free water present in the samples. The changes in the spectra indicate that PLA degrades because of water treatment. Gamma sterilization, in turn, is not observed to have significant effect on the bands. Further investigation would be required to determine, whether this is due to alternating quality and thickness of the samples.

Terrestrial laser scanning provides accurate forest measurements, for example, from single trees. However, the analysis of such measurements requires complicated algorithms to find useful information about the scanned trees. In this thesis a new method for reconstructing a tree model from single tree laser scanning measurements is presented. The algorithm is based on topological properties and relative measures which makes it scale independent and fast. The output of the algorithm is a reconstruction of the tree surface and volume as a three-dimensional cylinder model. The resulting model is complete and quantitative in the sense that it contains accurate information about the sizes and locations of each of the branches and also information about the relations between branches. This thesis describes the outline of the tree analysis algorithm and the mathematical concepts it is based on. Furthermore, several examples of the usage of the algorithm are provided. Maanpäällinen laserskannaus tarjoaa mahdollisuuden tehdä tarkkoja met\-sä\-mit\-ta\-uk\-si\-a esimerkiksi yksittäisistä puista. Mittaaminen on kuitenkin vasta alkua, sillä puusta saatavat mittaustulokset eivät itsessään ole useinkaan mielenkiintoisia vaan tuloksista jonkin algoritmin avulla lasketut ominaisuudet ovat todellinen mielenkiinnon kohde. Tässä opinnäytetyössä esitellään uusi menetelmä puumallin rekonstuktuointiin yksittäisestä puusta saatujen lasermittausten perusteella. Menetelmä perustuu topologisiin ominaisuuksiin sekä suhteellisiin mittoihin, mikä tekee siitä mittakaavasta riippumattoman ja nopean. Algoritmi tuottaa lopputuloksenaan kattavan ja kvantitatiivisen kolmiulotteisen sylinterimallin, joka kuvastaa mitatun puun pintaa ja tilavuutta. Sylinterimalli on kattava ja kvantitatiivinen siinä mielessä, että se sisältää jokaisen oksan koon ja paikan sekä tiedon oksien välilistä suhteista. Tässä työssä esitellään algoritmi yleisellä tasolla sekä matemaattiset käsitteet, joiden varaan se rakentuu. Mukaan on sisällytetty myös useita esimerkkejä algoritmin käytöstä.

This master of science thesis introduces an implantable measurement device that can be used to measure oxygen saturation (SpO2) with pulse oximetry methods. The device is intended to be incorporated into a implantable measurement device developed earlier at Tampere University of Technology (TUT). Two prototype devices were built and tested externally on a human subject with number of different measurement setups to determine how it would function in vivo. Respiratory diseases are the cause of approximately 50 % of all mortality in cattle. They can be hard to diagnose in early stages since there are no obvious external symptoms, this can cause outbreaks in groups of cattle. SpO2 gives a good measurement on how the respiratory system is functioning in the cattle by measuring the amount of oxygenated hemoglobin versus deoxygenated hemoglobin. The developed device measures the SpO2 with a probe made out of two light emitting diodes (LED) and a photosensor. In the thesis two types of coating methods where used to seal the probe, 3.5mm layer of medical grade epoxy and 15 µm layer of Parylene-C. The effect of these coatings on the probe components and signals where determined with measurements prior to coating and after. Parylene-C coating had much less effect on the signal acquisition than the epoxy coating, where the amplitude of the non-pulsatile signal increased on average over 1V and the pulsatile part decreased in amplitude. For Parylene-C there was a minor decrease in the amplitude of the non-pulsatile part but the pulsatile part had similar amplitude to non-coated probe. This difference is partially explained with the fact that thicker layer of coating creates internal scattering of light inside the coating. This light hits the photosensor before being absorbed by tissue and thus increases the DC level. A signal processing script was written in MATLAB to calculate the uncalibrated SpO2 from the raw signal. The noise level in all measurements was estimated with the standard deviation since the signal is unambigous and it was concluded that with a moving average filter of 4- or 8-points it is possible to reduce the noise significantly. Thermal radiation of the probe was estimated with test measurement of two different LED drive currents and theoretical calculations, neither case showed any significant increase in temperature. The effect of fat tissue that will surround the implant was also tested in a practical way with cow fat from a local supermarket. According to theory, light penetrates well through fat tissue and this was confirmed with measurements where the increased thickness of fat tissue decreased the amplitude of the signal. By applying a 8-point moving average filter it was possible to acquire a signal through ∼1cm thick layer of tissue with no perfusion. Number of other minor topics were solved some theoretically and others practically. The output of the thesis is a novel device that could be easily implanted in a dairy-cow as well as other mammals. The thesis also presents new information on the effects of coating SpO2 probes and the effects of fat tissue in cattle on the SpO2 signal. Pulse oxygen saturation measurements have not been conducted with an implantable meter before in any type of animal and thus certain uncertainty of measurements can only be eliminated with an implantation of a real device.

Euroopan unionissa hyväksytty rakennusten energiatehokkuutta parantava direktiivi on johtanut Suomen rakentamismääräysten tiukentumiseen. Tiukentuneiden rakentamismääräysten myötä merkitys rakennusten sisäilman määräystenmukaisuudesta ja lämpöviihtyvyydestä korostuu. Työssä tarkasteltiin kirjallisuuden avulla asuinrakennusten sisäilmastoon vaikuttavia tekijöitä sekä Suomen rakentamismääräysten vaatimuksia asuinrakennusten lämpöoloihin. Suomen rakentamismääräysten ja ohjeiden asettamien vaatimuksien täyttämiseksi haettiin asuinrakennuksen lämpöoloihin vaikuttavia tekijöitä. Ylilämpötilojen hallitsemiseksi mahdollisia ratkaisuja löytyi rakenteellisista-, ilmanvaihdollisista- ja passiivisista auringonsuojaratkaisuista sekä aktiivisesta jäähdytyksestä. Rakenteellisten, ilmanvaihdollisten ja passiivisten auringonsuojaratkaisujen vaikutuksia asuinrakennusten lämpöoloihin ja lämpöviihtyvyyteen tutkittiin case-kohteiden avulla. Auringonsuojaratkaisujen vaikutuksia case-kohteiden lämpöoloihin tarkasteltiin vertailemalla valittuja rakenteellisia-, ilmanvaihdollisia- ja passiivisia auringonsuojaratkaisuja. Tehokkaimmista passiivisista auringonsuojaratkaisuista luotiin toteuttamiskelpoiset yhdistelmät, jotka valittiin tarkempaan tarkasteluun. Auringonsuojaratkaisujen yhdistelmien avulla tutkittiin valittujen kriittisten asuntojen lämpöviihtyvyyttä, asetettujen määräysten täyttymistä sekä jäähdytystehontarvetta todellista käyttöä ja standardikäyttöä vastaavilla sisäisillä lämpökuormilla. Asuinrakennusten ylilämpötilojen hallitsemiseksi luotiin kahdeksan passiivista auringonsuojaratkaisua, joilla tarkasteltujen case-kohteiden kriittisten asuntojen lämpöolot olivat lämpöviihtyisät ja määräystenmukaiset. Ylilämpötilojen hallitsemiksi luotujen ratkaisujen avulla voitiin oleskelutilojen lämpöoloja hallita tehokkaasti. Auringonsuojalaseilla saavutettiin keskimäärin 2,5-6 asteen parannus huonelämpötiloihin, ilmanvaihdon tehostuksella noin 1,8-4,5 asteen ja auringonsuojaratkaisuilla 2-7 asteen oleskelutilojen lämpötilojen lasku. Auringonsuojayhdistelmillä tarkasteltujen kohteiden lämpöviihtyvyyttä haluttiin tehostaa pienentämällä kesäajan lämpötilahuippuja aktiivisella jäähdytyksellä. Tällöin aktiivinen jäähdytys oli mahdollista mitoittaa 39-62 prosenttia pienemmäksi perustarkastelun auringonsuojaratkaisuilla kuin ilman auringonsuojaratkaisuja. Kesäjakson aikaisena jäähdytystarpeena ero on suurimmillaan 6,5-14 -kertainen, tarkasteltavasta auringonsuojayhdistelmästä riippuen.

Vesihuoltopalveluiden toiminnan suunnittelua edellytetään lainsäädännössä vesihuolto-, terveydensuojelu-, valmius- ja pelastuslaeissa. Vesihuoltolaitoksen kannalta varautumista koskevat säädökset ovat yleisluontoisia eivätkä viittaa yksiselitteisesti oman valmiusjärjestelmän luomiseen. Maa- ja metsätalousministeriön vuonna 2004 asettama erityistilannetyöryhmä lanseerasi käyttöön niin kutsutun varautumissuunnitelman, jolla vesihuoltolaitos voisi täyttää osaltaan pelastus-, valmius- ja terveydensuojelulakien suunnitteluvelvoitteet ja varautua sekä normaaliolojen että poikkeusolojen erityistilanteisiin ja ehdotti vesihuoltolaitoksia koskevan varautumissuunnitteluvelvoitteen lisäämistä vesihuoltolakiin. Vuonna 2008 asetettu vesihuoltolain tarkastamistyöryhmä esitti erityistilannetyöryhmän ehdotuksen pohjalta varautumissuunnitteluvelvoitteen säätämistä vesihuoltolakiin. Työn tavoitteena on selvittää, millainen työkalu Maa- ja metsätalousministeriön lanseeraama vesihuoltolaitoksen varautumissuunnitelma olisi vesihuollon erityistilanteiden varautumisen suunnittelussa ja miten se käytännössä laadittaisiin. Työn yleisosuudessa käsitellään vesihuollon erityistilanteisiin varautumisen suunnittelua eri toimijoiden ja eri suunnittelujärjestelmien kautta. Vesihuoltolaitoksen lisäksi vesihuollon varautumiseen liittyviä suunnitteluvelvoitteita on mm. kunnan terveydensuojeluviranomaisella, pelastusviranomaisella sekä kunnan eri toimijoilla. Varautumisnäkökulma ja/tai riskien tarkastelu sisällytetään nykyään lukuisiin suunnittelujärjestelmiin, mikä saattaa aiheuttaa päällekkäisyyttä ja ristiriitoja. Työn suunnitelmaosassa luodaan varautumissuunnitelmapohja pienelle tai keskisuurelle vesihuoltolaitokselle ja kuvataan laatimisprosessi konsulttityönä. Suunnitelmapohjaa kehitettiin VVVasun mukaisesta valmiussuunnitelmapohjasta. Pohjaa pyrittiin muokkaamaan nykyistä käytännönläheisemmäksi ja käyttökelpoisemmaksi. Suunnitelman laatimisen tärkeimpinä vaiheina voidaan pitää uhkatekijöiden kartoitusta ja riskien arviointia sekä sidosryhmäkokousta viranomaisten kanssa. Työ osoittaa, että vesihuoltolaitoksen varautumissuunnitelmasta voidaan luoda käytännöllinen suunnittelutyökalu pienille ja keskisuurille vesihuoltolaitoksille. Suunnitelma sopii laadittavaksi myös ulkopuolisen konsultin toimesta, mutta vesihuoltolaitoksen henkilökunnan ja konsultin välillä tulee olla keskinäinen luottamus.

The aim of this Master of Science Thesis was to develop an MS Excel-based calculation tool for cooling and heating capacities in Halton Group´s new product, the Fan Coil Unit. The Fan Coil Unit is commonly used in passenger cabins of cruisers to cool or heat the cabin air. The number of customer orders of the Fan Coil Unit has been increasing, therefore, Halton Group’s subdivision Marine had a need to get this tool to save time, which had earlier gone to calculations for each unit order separately. Cooling demand consists of two kinds of heat loads: latent and sensible heat load. The part of the heat load which is caused by evaporation, is called the latent heat load. The latent heat load comes from for example the cabin, when the passenger's skin is sweating and from the shower facilities when water vapor is released into the cabin air. Sensible heat is heat exchanged by a body or thermodynamic system that has as its sole effect a change of temperature. This Thesis is divided into two parts. The first part will offer a basic theoretical background for the features that will be dealt with in the second part. The basic principles of Fan Coil Unit, heat transfer properties between air and water and other mathematical functions that have been used in this work are presented in the first part. The structure of the calculation tool and operations manual, are presented in the second part. As a result of this work, the calculation tool was developed and found to be appropriate for designing as well as improving the Fan Coil Unit.

Spray application can bring various benefits when applying chemicals on wet and dry paper or board such as precise adjustability of dosage and good coverage. Special advantage of spray application is in creating a uniform coverage onto irregular surfaces. This thesis is divided into two major sections. In the literary section the basic aspects of spraying are covered. Some of the current applications in paper and board industry, with ongoing research projects included, are reviewed with benefits and problems arising from using spray technology. The aim of the study was to determine the possibilities and restrictions of application of strengthening chemicals via spraying. Based on the literary section, xyloglucan, a water soluble polysaccharide, was chosen as focus. Xyloglucan (XG) was selected as the main focus as according to previous studies the strength properties, when sprayed onto paper web, increase significantly. Viscosity measurements were carried on a variety of solutions to characterize their behaviour when sprayed. Surface tension, according to general theories, factors in to good sprayability and was therefore included in the measurements – although the results from surface tension measurements were inconsistent. The power of ultrasound was taken in use to determine the effect of deflocculation and shorter molecules on the viscosity of the xyloglucan solution. The main objective of the experimental part was to study how the sprayed solutions which varied in viscosity and composition affect strength properties. Goal was also to determine whether it is possible to increase the dry weight fraction of sprayed xyloglucan solutions on to the level where the amount of water in the solutions would not require too much energy to be evaporated at drying sections in paper or board machines. Amount of borax in Tamarind Kernel Powder (TKP) and XG solutions was the most significant factor increasing the strength properties of the handsheets. Borax addition lowered viscosity to a certain point at lower dosages and increased the dry weight proportion of the solutions. XG with an addition of borax had the highest percentage of strain at break by far. Addition of only TKP or XG to the handsheet did not improve the sustained strain notably thus the runnability improving properties of borax seem to be excellent with conjunction of XG. Introducing ultrasound to higher concentrations of TKP showed a vast decline in viscosity already at short processing times. It can be assumed that sonication reduces viscosity via deflocculation to a certain point. The strength properties were significantly worse when compared to unmodified TKP. The results show that spraying of XG or TKP with borax provides best total increase in almost all strength properties. Borax can also be used effectively to increase the dry weight fraction of the sprayed solution without increasing viscosity too much.

Artificial groundwater recharge (AGR) and bank filtration (BF) are old, widely applied methods to produce drinking water by the infiltration of surface water through soil. These methods are increasingly used because they are considered to offer a relatively inexpensive and sustainable way to increase the water quality by physical, chemical and biological processes. One of the primary goals of AGR and BF is the removal of natural organic matter (NOM) that has several indirect adverse effects. Pollutants possibly existing in aquatic environments may pose a risk of aquifer contamination and have harmful effects for human health. Biodegradation has an important role in sustainable removal of NOM and pollutants in AGR and BF. However, the mechanisms affecting the natural removal processes are still poorly understood, and therefore more knowledge of underlying biogeochemical processes is needed. A research project TEVA (Improving artificial groundwater recharge by the development of on-line monitoring and control) was established to improve the understanding of the removal mechanisms of NOM during AGR and to enhance the AGR process control by on-line monitoring. As a part of the TEVA-project, the aim of this thesis was to study the effects of surface water infiltration on a pristine aquifer by monitoring physico-chemical water and bacterial community changes in Virttaankangas AGR site, southwest Finland, during experimental river water infiltration. In addition, the feasibility of biofilm collector slides for aquifer biofilm sampling was evaluated. The progress of infiltrated water in the aquifer was evident from increased dissolved organic carbon (DOC) concentrations. Compared to the initial infiltrated water concentration, DOC removal was observed along the soil passage. However, no preferential removal of any molecular fraction of DOC could be demonstrated. Due to infiltration, the native groundwater bacterial community profile changed significantly towards the community composition of infiltrated water. Attached bacterial community composition was not strongly influenced by water infiltration. Leucine incorporation method confirmed bacterial activity in the subsurface, whilst the RNA-based community analysis indicated that only a minor part of the attached and unattached communities were responsible for the activity in the aquifer. Differences in the minor bacterial groups between soil and biofilm collector community compositions were observed. However, the biofilm collectors accumulated dominating soil bacterial groups and therefore the collectors reliably collected the dominant soil bacteria.

Optical pulses are used as bits to transmit information. Controlled pulse generation is required for error-free communication and data processing. Semiconductor disk lasers (SDLs) are compact light sources that have shown potential for inexpensively generating intense but focusable ultrashort light pulses of a chosen color at gigahertz repetition rates. In this thesis, the generation of light pulses with SDLs is studied. Different methods for initiating pulsed operation are reviewed. A state-of-the-art high-power picoseconds pulse source emitting in the 1.55-μm telecommunications band is demonstrated by employing wafer fusion technology and advanced heat management in an SDL pulsed with a dilute nitride semiconductor saturable absorber mirror (SESAM). A simple way to generate picosecond pulses from an SDL by feeding frequency-shifted light back to the gain element of the laser is presented. The dominant way to generate pulses from SDLs, using a SESAM to organize the phases of the lasing cavity modes, is studied in detail. Models for pulse shaping in such passively mode-locked SDLs are reviewed. Stable mode locking (ML) at a very low saturation of absorption, relevant for SDLs pursuing high pulse repetition rates, is demonstrated and qualitatively explained. The regime of harmonic ML where multiple pulses circulate in the SDL cavity is reported and investigated both experimentally and numerically. Saturation and fast recovery of the gain are identified as being responsible for the strong ordering of pulses and for the changes in pulse characteristics observed between states with different numbers of pulses in the cavity. Hysteresis in the properties of the optical output is observed and controlled by tailoring the SDL gain element design. The presented results provide a means to precisely manipulate pulse characteristics in SDLs, paving the way for flexible high-speed data transfer and processing.

Matematiikan verkko-opetukseen voidaan tuoda lisäarvoa tarjoamalla opiskelijoille automaattisesti tarkastettavia matematiikan tehtäviä. Tässä työssä tarkastellaan tällaisten tehtävien luomista STACK-järjestelmällä. Työn tavoitteet ovat selvittää, millainen järjestelmä STACK on, kuinka STACKia tulisi käyttää teknisesti ja opetusjärjestelyjen näkökulmasta ja millaisia ohjeita STACKia käyttävälle opettajalle pitäisi antaa. Aluksi esitellään työn konteksti kertomalla matematiikan verkko-opetuksesta yleisesti ja Tampereen teknillisellä yliopistolla. Seuraavaksi esitellään STACKin eri versiot ja tarkastellaan lähemmin STACK2:n ominaisuuksia ja teknistä toteutusta sekä arvioidaan sen soveltuvuutta opetuskäyttöön. STACKia testattiin osana Insinöörimatematiikka B 4u -opintojakson kevään 2012 toteutusta. Tulosten perusteella tehdään ylläpitosuunnitelma ja annetaan jatkokehitysideoita. Lopuksi esitellään työn päätuloksena syntyneet opettajan ohjeet. Tutkimus osoittaa, että STACKilla on mahdollista luoda yliopistotasoisia, opetuksen kannalta mielekkäitä matematiikan tehtäviä. STACK ei kuitenkaan kerro opettajalle, mitä tai miten tulisi opettaa, eikä pelkkä järjestelmän olemassaolo motivoi opiskelijoita käyttämään sitä. STACKin käyttöä hankaloittavat epäintuitiiviset käyttöliittymät, useat käytettävät syntaksit ja ajonaikaiset tekniset ongelmat. Opettajan ohjeet esittelevät STACKin ominaisuuksia ja antavat konkreettisia ohjeita sen käyttöön.

This user guide accompanies the surrogate model toolbox for global optimization problems. The toolbox is made for computationally expensive black-box global optimization problems with continuous, integer, or mixed-integer variables. Problems where several or all variables have to be integers may also have black-box constraints, whereas purely continuous problems may only have box constraints. For problems with computationally cheap function evaluations the toolbox may not be very efficient. Surrogate models are intended to be used when function evaluations take from several minutes to several hours or more. When reading this manual it is recommended to simultaneously take a look at the code. The code is set up such that the user only has to define his/her optimization problem in a Matlab file (see Section 6.1). Additional input such as the surrogate model to be used, the sampling strategy, or starting points are optional (see Section 6). This document is structured as follows. In Section 2 the general structure of a surrogate model algorithm is summarized. The installation is described in Section 3. The dependencies of the single functions in the code are shown in Section 4. Section 5 briefly summarizes how the surrogate model algorithm works in general. Section 6 describes the options for the input of the main function. In Section 7 the input and output of the single subfunctions of the algorithm are described. Examples for using the surrogate model algorithm are given in Section 8. In Section 9 it is explained how the user can define an own (mixture) surrogate model and an example is given. The elements of the saved results are described in Section 10.

Epithelium forms tight membranes that efficiently take part in secretion and absorption between the two lining tissues. A tight membrane of this nature has a low DC conductivity and this is generally used to assess the integrity of cultured epithelium cell layers. Recent studies have shown that impedance spectroscopy gives more information about the electrophysiological structure of the cells. Collaborative research by Department of Biomedical Engineering at Tampere University of Technology and stem cell research group at University of Tampere has shown that electrochemical impedance spectroscopy is useful in assessing the maturity and functionality of differentiated retinal pigment epithelium (RPE) cells. However the time expenditure of the traditional frequency sweeping method is a poor candidate for drug permeability or multichannel studies where several frequency responses have to be measured within a short time. The aim of this Thesis was to develop the front end electronics for fast impedance spectroscopy measurement employing inverse-repeat binary sequence as the broadband excitation signal. Also the DC potential across the cell membrane was to be measured with the device. The developed device was first tested using a custom built test box and plastic film as artificial membrane. In addition several electrode materials were used to study the observed polarization impedances. Further testing with differentiated RPE cell layers was done using the Ussing chamber and the well plate setups that are commonly used in cell culturing studies. All the measured frequency responses were referenced with a commercial device widely used in epithelium research. The observed measurement differences between the device and the reference were largely caused by the load dependent output current of the device and by the electrode polarization taking place at the voltage measurement electrodes. Due to the input current error the relative difference of the measured impedance levels was typically from 5 % to 10 % with load impedances larger than 700 ohms. With lower load impedances the measured relative difference increased rapidly. A method to compensate for the input current error is presented in this thesis. DC potential measurements with the device were not successful as the electrodes used had very high offset voltages. The frequency responses measured with the device give a good measure of the capacitance present in the cell layer. Capacitance of the cell layer can be used to assess the maturity of the cell layer and for such purpose the device suits well. For impedance level measurements the device has a relatively large error margin and further research needs to be done to improve the accuracy and to eliminate the DC current flow. In addition the accuracy of the measurement system would improve by dividing the input stage between the DC potential and frequency response measurements. Also more carefully designed electrodes would help to control the electrode offset voltages. Epiteelisolut muodostavat tiivisliitoksia, jotka ovat tärkeässä roolissa kudosten välillä tapahtuvassa erityksessä sekä absorptiossa. Tiivisliitoksen tasavirtajohtavuus on heikko ja tätä ominaisuutta hyödynnetään yleisesti viljeltyjen epiteelisolukerrosten tiiviyden sähköisessä tarkastelussa. Tutkimukset ovat kuitenkin osoittaneet, että impedanssispektroskopia antaa enemmän tietoa solujen elektrofysiologisesta rakenteesta kuin yksinkertainen resistanssimittaus. Tampereen Teknillisen Yliopiston Biolääketieteen laitos sekä Tampereen Yliopiston kantasolututkimusryhmä ovat yhteistyössä osoittaneet elektrokemiallisen impedanssispektroskopian soveltuvan kantasoluista erikoistettujen retinan pigmenttiepiteelisolujen (RPE) kypsyyden ja toiminnallisuuden arviointiin. Perinteiset taajuuspyyhkäisyä hyödyntävät taajuusvasteanalysaattorit soveltuvat kuitenkin hitaudeltaan heikosti tutkimuksiin, joissa mittauskohteessa tapahtuu nopeita muutoksia tai missä useita taajuusvasteita mitataan lyhyellä aikavälillä. Tämän diplomityön tavoitteena oli kehittää pääteasteen elektroniikka mittausjärjestelmälle, joka hyödyntää laajakaistaista binäärijaksoa herätesignaalina ja mahdollistaa näin huomattavasti nopeamman impedanssispektroskopian. Kehitettävän laitteen tuli myös mitata solukerroksen yli oleva DC potentiaali. Diplomityössä kehitettyä laitetta testattiin aluksi in vitro mittauksia varten kehitetyllä testijärjestelmällä, jossa solukerrosta jäljiteltiin ohuella muovikalvolla. Näissä mittauksissa testattiin myös eri elektrodimateriaalien vaikutus havaittuun polarisaatioimpedanssiin. Viljeltyjen RPE solujen taajuusvasteita mitattiin työssä käyttäen sekä Ussingin kammio- että kuoppalevymittausasetelmia. Laitteella mitattujen taajuusvasteiden hyvyyttä arvioitiin vertaamalla tuloksia kaupallisella taajuusvasteanalysaattorilla mitattuihin vasteisiin. Kehitetyllä laitteella mitattujen vasteiden ero analysaattorilla mitattuihin johtui suurilta osin herätevirran riippuvuudesta kuormasta sekä jännitemittauselektrodien polarisaatioimpedansseista. Yli 700 ohmin kuormilla herätevirrasta aiheutuva virhe oli tyypillisesti 5% – 10 %, kun taas matalimmilla kuormilla virhe kasvoi nopeasti. Tämän virheen kompensoimiseksi on kuitenkin esitetty metodi tässä diplomityössä. Epiteelisolukerroksen yli olevaa DC jännitettä ei onnistuttu mittaamaan johtuen käytettyjen elektrodien korkeista offset-jännitteistä. Tässä diplomityössä kehitetyllä laitteella mitatut taajuusvasteet noudattavat hyvin analysaattorilla mitattujen vasteiden käyrämuotoja ja laite soveltuu täten solukerroksen kapasitanssin arviointiin. Solukerroksen kapasitanssia voidaan käyttää apuna solujen kypsyyden arvioinnissa. Laitteella mitatut impedanssitasot eroavat kuitenkin analysaattorilla mitatuista ja jatkokehitys DC virtojen eliminoimiseksi sekä elektrodien erisuuruisten offset-jännitteiden kompensoimiseksi on suositeltavaa.

Microalgae are a promising feedstock for biofuel and bioenergy production due to their high photosynthetic efficiencies, high growth rates and no need for external organic carbon supply. However, microalgal biomass cultivation for energy production purposes is still rare in commercial scale. Further research and development is needed to make microalgal derived energy sustainable and economically competitive. This work investigated cultivation of fresh water microalga Chlorella vulgaris and marine microalga Dunaliella tertiolecta and their utilization in production of hydrogen, methane, electricity, butanol and bio-oil after bulk harvesting the biomass. Growth of the two microalgae was studied in five different photobioreactor (PBR) configurations especially concentrating on the quantification and characterization of heterotrophic bacteria in non-axenic microalgal cultivations and microalgal utilization of different nitrogen sources. Anaerobic cultures used for the energy conversion processes were enriched from a mesophilic municipal sewage digester separately for production of H₂, CH₄ and electricity from the two microalgal species. After culture enrichment, energy conversion yields of microalgal biomass to the different energy carriers were compared. Anaerobic microbial consortia utilizing microalgal biomass were characterized based on the 16S rRNA gene sequence analysis. H₂ and CH₄ production potentials were tested in anaerobic serum bottles and electricity and butanol production in fed-batch operated two-chamber microbial fuel cells (MFCs). All the PBR configurations tested were amenable to C. vulgaris and D. tertiolecta biomass production. Highest biomass concentrations and productivities by C. vulgaris and D. tertiolecta were 3.8 and 3.2 g L⁻¹, and 0.60 and 0.83 g L⁻¹ d⁻¹,, respectively. They were obtained in bubble column PBRs at 12% CO₂, 10 mM NO₃⁻ and 350 μmol photons m⁻² s⁻¹. Static mixers used in the flat plate PBRs did not generally enhance the growth of either C. vulgaris or D. tertiolecta at the low light intensities (50 μmol photons m⁻² s⁻¹) used. However, the low light intensity resulted in high growth rates at the early stages of growth. The highest specific growth rates were obtained in the flat plate PBRs and were 2.0 and 1.4 d⁻¹ for C. vulgaris and D. tertiolecta, respectively. Bacterial growth occurred simultaneously with microalgal growth and correlated generally well with algal exuded dissolved organic carbon (DOC) concentrations. The bacterial communities were relatively stable and reproducible in both C. vulgaris and D. tertiolecta cultivations. However, algal associated bacterial communities were vastly different in C. vulgaris and D. tertiolecta cultures due to different growth medium salinity (

Dry eye syndrome (DES) is disease of the eye surface, where tear film is ruptured and lubrication or moistness of the corneal and conjunctival epithelia do not work properly. DES is most common in older people, but also people with rheumatoid diseases, damaged corneal epithelia and certain medications are more likely to have DES. There are lubricating and moisturizing eye drops to relieve the symptoms of DES, administered at least twice a day depending on the severity, but they are only palliative. Eye drops for DES are composed of natural and synthetic origin polymers and their blends. Retaining methods are mainly viscosity and adherence to surfaces by weak interactions. In situ gelation by temperature, pH and osmotic change have been under research in recent years. Many studies have concentrated on ambiphilic polymer conjugates, which fit to the oily and aqueous layer of the tear film and have a better ability to carry a hydrophobic therapeutic drug molecule. Interest has also been pointed to mucoadhesive polymers, which are adhering to mucin by covalent bond. Thiolation of the polymer has been seen as a very promising method to achieve better muchoadhesion and thus has been extensively studied. A chitosan and hyaluronan are natural origin polymers, which have been thiolated and these polymer matrixes can be loaded with trehalose or albumin as therapeutic agent. In this thesis the anatomy of the eye, and especially the corneal and precorneal area and the conjunctiva is discussed. Ethiology of DES and commercial available therapies are covered in this thesis, but also therapies under research and corneal simulation methods in vitro. Kuivasilmäisyys on sairaus, jossa silmän sarveiskalvon pinnalla oleva kyynelkalvo rikkoutuu. Sarveis- ja sidekalvon kosteutus ja voitelu häiriintyvät kyynelkalvo epätasapainosta. Kuivasilmäisyys on ikääntyvien ihmisten sairaus, mutta sitä tavataan myös sarveiskalvovaurio- ja reumapotilailla. Joidenkin lääkkeiden käyttöön liittyy sivuoireina kuivasilmäisyyttä. Kuivasilmäisyyteen on saatavilla kaupallisia oireita lievittäviä silmätippoja, jotka kosteuttavat ja voitelevat silmän pintaa, mutta eivät hoida sairautta. Kuivan silmän tipat on valmistettu luonnon polymeereistä tai synteettisistä polymeereistä, joita annostellaan vähintään kahdesti päivässä. Terapeuttisen vaikutuksen kesto perustuu viskositeettiin tai tarttumiseen silmän pintaan heikoilla sidoksilla. In situ geeliytyminen lämpötilan, pH:n tai suolapitoisuuden muutoksesta on ollut tutkijoiden mielenkiinnon kohteena viime vuosina. Myös polymeerien tarttumista limakalvon pintaan (mukoadheesio) heikoilla vuorovaikutuksilla tai vahvemmilla kovalenttisilla sidoksilla on tutkittu. Amfifiliset polymeerit ovat myös vallanneet alaa, sillä niiden oletetaan sopivan kyynelkalvon pinnan öljykerrokseen ja edelleen sen alla olevaan vesikerrokseen. Amfifilisella polymeerillä on myös kyky kantaa hydrofobista lääkemolekyyliä. Polymeerien tiolointiin ja sen tutkimiseen on panostettu viime aikoina paljon. Tiolointi on menetelmä, jolla parannetaan polymeerin mukoadheesiota. Erityistä mielenkiintoa on herättänyt kitosaanin ja hyaluronihapon tiolointi, jolloin myös terapeuttinen molekyyli, kuten trehaloosi tai albumiini voitaisiin ladata polymeerimatriisiin. Tässä kirjallisuusselvityksessä käsitellään silmän anatomiaa erityisesti kyynelkalvon, sarveiskalvon ja sidekalvon osalta. Tässä opinnäytetyössä on käsitelty kuivasilmäisyyteen johtavia syitä ja nykyisiä sekä kehityksen kohteena olevia hoitomenetelmiä. Lopuksi esitellään sarveiskalvon simulointimenetelmiä, joita on käytetty laboratorio-olosuhteissa.

Lung surfactant is a thin lining covering the air-water interface inside the alveoli. Its main function is to reduce the surface tension at this interface and thus prevent the alveoli from collapsing at the end of the breathing cycle. In addition to its importance in maintaining proper lung function, it is suggested that this monolayer aects the permeation of small non-electrolytes such as molecular oxygen from inhaled air towards circulation. However, the results on this topic are extremely diverse. Earlier computational studies have been inadequate in terms of monolayer composition and state, simulation conditions and force eld parametrisations and the amount of employed computational resources. They have also often considered either energetic or kinetic factors on permeability alone. On the other hand, lack of resolution has been the main issue with experimental studies on oxygen permeability. To overcome these limitations, the permeation properties of protein-free surfactant membrane systems were studied through long atomistic molecular dynamics simulations with high spatial resolution. Monolayers at dierent compression states were employed with the lipid composition and simulation conditions of the systems mimicking that of the real surfactant. In addition to the permeation properties of the monolayer, information about oxygen pathways below the interface were also discussed with the data obtained from bilayer simulations with the same composition. A new model for molecular oxygen was sought in this study and its performance was verified with multiple benchmarks. A special free energy technique, the z constraint method was employed together with the inhomogeneous solubility{diusion model to study the permeabilities of the membrane systems as it provides one with detailed information about both energetic and kinetic factors on permeation. The key values of the spatially resolved free energy and diusion coe cient pro les along the membrane normal agree with those of earlier studies yet the detailed pro les also provide new and interesting information. The calculated permeabilities of the monolayer systems are successfully linked to the other physical quantities characterizing the monolayers. The possible suggested structures lying below the surfactant monolayer are compared based on their eect on oxygen transport. Finally, thorough comparison is performed between our calculations and earlier studies.

Tämä diplomityö on osa Tampereen teknillisen yliopiston konstruktiotekniikan laitoksen projektia ”Julkisten rakennusten energiatehokkuuden parantaminen”. Työn päätavoitteina on tutkia, mitä mittauksia vaaditaan, että saataisiin kohtalaisen hyvin selvitettyä koulurakennusten energiatehokkuus ja sisäilmaston taso. Lisäksi keskeisenä osana on saada selville yksityiskohtaista tietoa kahden lämmönkulutukseltaan poikkeavan koulurakennuksen energiatehokkuudesta ja sisäilmastosta erilaisilla energiatehokkuuden ja sisäilmaston mittauksilla. Taustalla on vaikuttanut myös riittävien lähtötietojen saaminen koulurakennusten energiankulutusta paremmin kuvaavien laskentamallien taustalle, jotka ovat osa projektia, mutta eivät kuulu tähän diplomityöhön. Työssä on painotettu energiatehokkuuden osalta erityisesti lämmönkulutusta, koska siinä on huomattu edellisten tutkimusten perusteella olevan suuri hajonta. Koulurakennusten lämmönkulutukseen vaikuttavia tekijöitä ovat ulkovaipan lämmöneristävyys ja ilmanpitävyys, ilmanvaihto- ja lämmitysjärjestelmät, rakennusautomaatio ja sen taso sekä koulun kiinteistönhoitajat ja käyttäjät. Koulurakennusten lämmönkulutukseen vaikuttavat tekijät eivät ole siis pelkästään teknisiä. Energiatehokkuus ei saisi koskaan mennä ihmisten terveyden edelle, minkä johdosta työssä on tarkasteltu myös sisäilmaston tasoa mittauskohteissa. Sisäilmastoon liittyviä asioita ovat muun muassa lämpötila, veto ja ilmanlaatu. Edellä mainittuihin lämmönkulutuksen osatekijöihin liittyviä mittauksia on tehty kahdessa koulussa, joista käytetään tunnuksia A ja B. Koulu A on lämmönkulutukseltaan vähän kuluttava koulu ja koulu B on paljon kuluttava. Lämpötilaolosuhteita ja ilmanlaatua on mitattu kouluissa laajoilla langattomilla mittausverkoilla. Lisäksi kohteissa on tehty lämpökamerakuvaus sekä määritetty ilmavirrat. Koulussa A on myös suoritettu ilmanpitävyyden mittaus ja määritetty ilmanvaihtokoneiden lämmön talteenoton hyötysuhteet. Tehtyjen mittausten perusteella koulujen A ja B välillä on jonkin verran teknisiä eroja, esim. ilmanvaihdon lämmön talteenotossa, jotka vaikuttavat lämmönkulutuksen eroihin. Työn aikana on kuitenkin käynyt selville, että syyt eivät ole pelkästään rakenteisiin ja järjestelmiin liittyviä. Myös kiinteistönhoitajilla ja ylemmillä kiinteistöistä vastaavilla henkilöillä on iso vaikutuksensa rakennusten energiatehokkuuteen. Kuitenkin sisäilman lämpötilojen, hiilidioksidipitoisuuksien ja ilmavirtojen mittaus antaa arvokasta energiankulutusta selittävää tietoa.

Due to rising global demands for energy and growing concerns about the accelerating greenhouse effect, efficient and clean energy generation is increasingly under a spotlight. At the moment and in the future the weather depending renewable energy production is increasing. This growth leads to higher demand for fast balancing energy production during unfavorable generation conditions. Additionally, overall electricity consumption is increasing which results in higher peak loads and decreasing transmission and distribution capacity in the electric grid. This requires more fast support for the grid and decentralized energy production. Combined heat and power (CHP) is the simultaneously generation of electricity and useful heat from a single fuel source at high efficiency. A high number of small internal combustion engine based CHP units can provide fast balancing energy, due to good dynamic behaviour, and simultaneously heat for heat sinks, such as residential and commercial buildings. The distributed structure of the heat sinks leads to a transformation towards a decentralized energy system in which small-scale distributed generation is connected to the distribution network instead of large central generators connected to the transmission network. This thesis models and examines a small CHP system operation under a new concept where the electricity utility can remote control a contracted CHP units within different heat sinks and gain a benefit by generating electricity during unfavourable weather conditions and peak loads. The main goal of this thesis was to construct a single CHP system model into the Matlab/Simulink and investigate the behaviour of the system. The thesis is divided into two parts. In the literature study part, combined heat and power and different prime movers are introduced. Additionally, different CHP configurations and components are presented. Finally, the market research was conducted and market available internal combustion based small- and micro CHP units were introduced. In the second part, the model construction methodology was presented and results of two test cases were discussed. Finally, the optimization was performed using the model. The result indicates that small CHP operation on the liberalized electricity market is profitable from the electricity utility point of view. In this case the CHP unit requires large thermal storage which enables long storage time and operation only during the high electricity price. This enables also to provide fast balancing energy for weather depending renewables energies and support for the grid during peak loads. The large storage decouples heat- and power driven CHP system because it can respond to both demand with the storage. Until now the heat-driven systems have been commonly used and electricity is produced as a by-product.

The development of computers has enabled the collection and storage of terabytes of data and the creation of large data warehouses. The main problems with such data are their size and structure. The fundamental intellectual challenges at present are the analysis and understanding of the data in the decision-making process. This thesis introduces and compares the methods of GUHA and Weka software. The thesis highlights the differences between GUHA and Weka software through taking 2 methods which reveal the association rules of the Weka programme and comparing them with three methods which reveal the association rules of the GUHA programme. The difficulty of the task is the amount of computation which has to be done to explain whether the methods have any differences or not. The work has been done by taking the results from one of the Weka methods and comparing these with all the methods of GUHA. The second Weka method provides the same results as the first one, but in a different order. The results have been carefully compared and there are some comments in the discussion part of the thesis.

UV-kovettuvat kuumasulaliimat ovat olleet viime vuosina suuren kiinnostuksen kohteena liimateollisuudessa. Suurin osa käytössä olevista liimoista ovat liuotin- tai vesipohjaisia, jolloin liimapinnan kovettuminen vaatii aina liuottimen tai veden poiston. UV-kovettuvilla liimoilla ei tarvita liuottimen poistoa, koska ne ovat valmiiksi 100 % kiinteitä. UV-kovettuvat kuumasulaliimat sulatetaan ennen levitystä ja ne kovettuvat UV-valon vaikutuksesta. Ennen tätä työtä UV-kovettuvia liimoja on käytetty pääsääntöisesti vain muovipinnoille, mutta tässä työssä on tarkoitus tutkia niiden soveltuvuutta paperipinnoille. UV-kovettuvat liimat polymeroituvat radikaalipolymeraatiolla. Radikaalipolymeraatiossa fotoinitiaattori absorboi ultraviolettisäteilyn, jolloin syntyy vapaa radikaali. Se reagoi monomeerin kanssa muodostaen radikaalipolymeerin. Radikaalipolymeeri reagoi edelleen monomeerien tai oligomeerien kanssa, kunnes tapahtuu terminaatio kahden radikaalin reaktiossa. Työtä varten suoritettiin kaksi koeajoa. Ensimmäisen koeajon perusteella valittiin tutkittavat kahdeksan näytettä. Sivelymäärä, pintapaperi ja kovetusaste valittiin muuttujiksi, liiman pysyessä vakiona. Vertailemalla tuloksia keskenään voitiin vetää johtopäätöksiä näiden vaikutuksista tarraetiketin ominaisuuksiin. Tarraetiketin tärkeimmät ominaisuudet ovat tarttuvuus, adheesio ja koheesio. Tarttuvuutta mitataan tack-testillä, adheesiota peel-testillä ja koheesiota shear-testillä. Näiden lisäksi tarraetiketeille suoritettiin muita visuaalisia testejä, kuten vedenkestotesti, tarttuvuus kylmälle ja kostealle pinnalle sekä koeputkiliputustesti. Lisäksi näytteille tehtiin DMTA-mittaus, joka mittaa materiaalin viskoelastisia ominaisuuksia. Työn tarkoituksena on selvittää voidaanko UV-kovettuvat tarraliiman ominaisuuksia muuttaa kovettumisastetta muuttamalla. Lisäksi tutkittiin pintapaperin valinnan ja sivelymäärän vaikutuksia tarraetiketin ominaisuuksiin. Huomattiin, että kovettumisasteella on vaikutusta joihinkin liiman ominaisuuksiin. Eroavaisuudet kuitenkin pienenivät, kun näytteitä oli vanhennettu eri olosuhteissa. Myös sivelymäärällä oli vaikutusta tarraliiman ominaisuuksiin. Suurin vaikutus liiman vanhenemiseen oli pintapaperilla. Työ tehtiin yhteistyössä UPM Raflatac Oy:n kanssa.

Ever since World War II, antibiotics have been medicine’s number one asset in fighting microbial infection, one of the leading causes of death worldwide. Misuse of antibiotics has, however, led to rapid spread of antibiotic resistance among bacteria and ensuing development of multiple resistant pathogens. Therefore, antibiotics are rapidly losing their antimicrobial value, which can be seen a failure of society to protect one of its valuable resources.The use of antibiotics in food production animals is strictly controlled by the European Union. Veterinary use is regulated to prevent spreading of resistance due to unwarranted use and to prevent antibiotic residues in food products. EU legislation establishes maximum residue limits (MRLs) of veterinary medicinal products in foodstuffs of animal origin, and enforces countries to establish and execute a national monitoring plan of animal products to implement food control measures. Among samples selected for monitoring, suspect noncompliant samples are screened for and then subjected to confirmatory analysis to establish the identity and concentration of the contaminant. Screening methods for antibiotic residues are typically based on microbiological growth inhibition, whereas physico-chemical methods are used for confirmatory analysis.In this study, antibiotic whole-cell biosensor assays were examined as a novel screening method. Utilizing a tetracycline-specific bioluminescent whole-cell biosensor, a screening method for tetracycline residues in poultry meat was developed. Assay sensitization to meet the EU MRLs was achieved by improving tetracycline accumulation into the biosensor cells with a combination of membrane-permeabilizing agent polymyxin B and chelating agent EDTA. The result was a rapid, simple and cost-effective high-throughput screening method that could detect all four veterinary relevant tetracyclines and their 4-epimer metabolites in poultry meat with sensitivity below the MRLs. The study also provided proof of antimicrobial activity of tetracycline 4-epimer metabolites, a quality previously thought absent from 4-epidoxycycline.Nisin is a lantibiotic, a peptide antibiotic produced by lactococci. The industrial use of nisin as a food preservative (E234) and maximum allowed levels set by the EU warrant developing methods for nisin quantification in foods. In this study, a bioluminescent whole-cell biosensor for nisin was constructed and utilized in determining nisin concentrations in milk. The developed assay was rapid and simple to perform, and required no sample pretreatment except dilution. Sensitivity of the assay was in the sub-picogram per ml level, exceeding the performance of all previously published methods. The assay was also used in determining nisin-production efficiency by quantifying nisin in growth medium of a nisin-producing Lactococcus strain. Simultaneously, nisin producers could be distinguished from non-producers. This idea was expanded in a follow-up study, which utilized the nisin biosensor in screening for nisin producers in raw milk. Screening was based on simple overlay of raw milk cultures and identification of nisin producers by a bioluminescent zone surrounding the nisinogenic colony. The seven identified nisinogenic colonies were divided in three groups by genetic fingerprinting,and characterized as nisin variant Z producing Lactococcus lactis subsp. lactis. In addition, four nisin A producers were identified in a panel of 91 dairy lactococcal strains. Specificity studies showed that only nisin and not other bacteriocin peptides induced bioluminescence in the sensor strain. Also, all nisin-gene harboring colonies induced bioluminescence, with the exception of one lactococcal strain shown to carry a nonfunctional nisin gene.The development of novel inducible whole-cell biosensors for different groups of antimicrobials can be limited by the lack of regulatory elements specifically responsive for these substances. In this study, we characterized DNA and ligand binding of the macrolide antibiotic-responsive repressor protein, MphR(E). The protein was modified by rational design of mutations to improve DNA affinity and dimerization. DNA and ligand binding as well as macrolide-induced dissociation from DNA were studied by fluorescence anisotropy and mass spectrometry. Mutants with improved DNA affinity and retained ligand binding and dissociation characteristics were identified. One mutant surprisingly formed a covalent dimer through disulfide bridge formation. This was shown to improve DNA affinity, but ligand binding and induction was impaired. Ligand binding spectrum of MphR(E) was shown to cover macrolides with a 14-membered lactone ring structure, but macrolides with a 16-membered ring or lincosamides showed no binding. MphR(E) and its mutants showed interesting novel characteristics that could benefit biosensor design.In conclusion, this study shows the applicability of whole-cell biosensors in developing simple, robust and cost-effective screening methods for antimicrobials in food products. These methods show high sensitivity and specificity towards the target analyte, and can be used in semi-quantitative to quantative analysis. In addition to residue monitoring, whole-cell biosensors can be used for producer identification. The identified nisin producers can find use as protective starter cultures in fermented food production. The modified repressor MphR(E) shows promise as an improved regulator of reporter gene production in whole-cell biosensor applications, and is an example of purposeful effort to develop regulatory elements for novel biosensor designs. Antibiootit ovat olleet tärkein bakteeri-infektioiden hoitokeino aina toisesta maailmansodasta lähtien. Bakteeri-infektiot ovat maailmanlaajuisten kuolemansyytilastojen kärkisijoilla. Antibioottien huolimaton käyttö on kuitenkin johtanut antibioottiresistenssin nopeaan leviämiseen bakteerien keskuudessa sekä useille antibiooteille resistenttien patogeenien kehittymiseen. Tämän vuoksi antibiootit menettävät nopeasti antimikrobiaalista voimakkuuttaan, mitä voidaan pitää yhteiskunnan kyvyttömyytenä suojella arvokasta pääomaansa. Euroopan Unioni valvoo antibioottien käyttöä eläimissä, joita hyödynnetään elintarvikkeiden tuotannossa. Eläinlääketieteellistä käyttöä säädellään resistenssin leviämisen ja ruoassa esiintyvien antibioottijäämien estämiseksi. EU:n lainsäädäntö osoittaa enimmäisjäämärajat eläinlääkinnällisille aineille eläinperäisissä elintarvikkeissa ja velvoittaa jäsenvaltiot laatimaan ja toteuttamaan eläimistä saatavien elintarvikkeiden kansallisen vierasainevalvontaohjelman. Valvontaan valittujen näytteiden joukosta seulotaan näytteet, joiden epäillään sisältävän jäämiä, joiden luonne ja pitoisuus varmistetaan lisäanalyysillä. Seulonnassa käytetään tavallisesti mikrobiologiseen kasvuinhibitioon perustuvia menetelmiä, ja varmistukseen fysikaalis-kemiallisia analyysejä. Tässä tutkimuksessa tarkasteltiin kokosolubioantureilla tehtävien antibioottimääritysten soveltuvuutta antibioottijäämien seulontamenetelmäksi. Tetrasykliinispesifistä bioluminoivaa kokosolubiosensoria käyttäen kehitettiin seulontamenetelmä tetrasykliinijäämien tunnistamiseksi kananlihasta. Määritys saatiin herkistettyä EU:n enimmäisjäämärajojen tasolle helpottamalla tetrasykliinien pääsyä bioanturisoluun solukalvon läpäisykykyä lisäävällä polymyksiini B:llä sekä kahdenarvoisia kationeja kelatoivalla EDTA:lla. Tuloksena oli nopea, yksinkertainen ja kustannustehokas seulontamenetelmä, jolla oli korkea suoritusteho. Menetelmä kykeni havaitsemaan kaikki neljä eläinlääketieteessä käytettävää tetrasykliiniä sekä niiden 4-epimeeri aineenvaihduntatuotteet kananlihassa enimmäisjäämärajat alittavissa pitoisuuksissa. Tutkimus myös tuotti todisteita tetrasykliinien 4-epimeerien antimikrobiaalisesta aktiivisuudesta, joka aiemmin arveltiin puuttuvan 4-epidoksisykliiniltä. Nisiini on laktokokkien tuottama lantibiootti eli peptidiantibiootti. Nisiinin käyttö elintarviketeollisuudessa säilöntäaineena (E234) sekä EU:n nisiinille asettama sallittu enimmäismäärä elintarvikkeissa luovat tarpeen nisiinin määritysmenetelmille. Tässä tutkimuksessa rakennettiin bioluminoiva nisiinispesifinen kokosolubioanturi, jota käytettiin määrittämään nisiinipitoisuuksia maidossa. Kehitetty määritys oli nopea ja yksinkertainen, eikä vaatinut laimennusta monimutkaisempaa näytteen esikäsittelyä. Määrityksen herkkyys oli alle pg/ml mittaluokassa, ja se oli herkin koskaan julkaistu nisiinimääritys. Määritystä käytettiin myös nisiinintuotannon tehokkuuden arvioinnissa mittaamalla nisiinipitoisuus sitä tuottavan Lactococcus lactis -kannan kasvumediumista. Samanaikaisesti nisiinintuottaja voitiin erottaa nisiiniä tuottamattomista kannoista. Tätä ajatusta tarkasteltiin laajemmin jatkotutkimuksessa,jossa nisiinibioanturia käytettiin seulomaan nisiinintuottajakantoja raakamaidosta. Yksinkertainen seulontamenetelmä perustui raakamaitobakteerien viljelmien peittämiseen ohuella bioanturikerroksella, jolloin nisiinintuottajapesäkkeiden ympärille muodostui biolumine-senssivyöhyke. Seitsemän tunnistettua nisiinintuottajapesäkettä jakautuivat geneettisen sormen-jäljen perusteella kolmeen ryhmään, ja ne olivat kaikki Lactococcus lactis subsp. lactis -alalajiin kuuluvia nisiini Z -variantin tuottajia. Lisäksi 91 laktokokkikannan paneelista tunnistettin kolme nisiini A -variantin tuottajaa. Spesifisyystutkimukset osoittivat, että vain nisiini indusoi bioluminesenssin bioanturibakteerissa eivätkä muut bakteriosiinipeptidit. Lisäksi kaikki nisiinigeeniä kantavat pesäkkeet indusoivat bioluminesenssin. Poikkeuksena oli yksi laktokokkikanta, jonka todettiin kantavan toimimatonta nisiinigeeniä. Tietyille antibioottiryhmille spesifisen vasteen antavien säätelyelementtien puute saattaa vaikeuttaa kokosolubioanturien kehittämistä. Tässä tutkimuksessa karakterisoitiin makrolidispesifisen repressoriproteiini MphR(E):n DNA- ja ligandinsitomisominaisuuksia. Proteiinia muokattiin rationaalisen mutaatiosuunnittelun keinoin tarkoituksena tuottaa DNA- ja dimerisoitumisominaisuuksiltaan parempia mutantteja. DNA- ja ligandisitoutumista sekä makrolidiligandien indusoimaa dissosiaatiota DNA:sta tutkittiin fluoresenssianisotropialla ja massaspektrometrialla. Tutkimuksessa löydettiin mutantteja, joilla oli villityypin proteiinia parempi DNA-affiniteetti sekä ennallaan säilynyt kyky sitoa ligandeja ja indusoitua sitomisen vaikutuksesta. Yksi mutanteista muodosti rikkisillan avulla kovalenttisen dimeerin vastoin odotuksia. Kovalentti dimerisaatio paransi DNA-affiniteettia, mutta haittasi ligandin sitomista sekä induktiota. MphR(E):n ligandikirjo kattoi 14-jäsenisen laktonirenkaan makrolidit, mutta ei 16-jäsenisen renkaan makrolideja eikä linkosamideja. MphR(E) ja sen mutantit osoittivat mielenkiintoisia uusia ominaisuuksia, jotka voivat hyödyttää bioantureiden suunnittelua. Johtopäätöksenä voidaan sanoa, että tämä tutkimus osoittaa kokosolubioanturien soveltuvan yksinkertaisten, luotettavien ja kustannustehokkaiden seulontamenetelmien kehittämiseen antibioottijäämien osoittamiseen elintarvikkeista. Nämä menetelmät osoittavat suurta herkkyyttä ja spesifisyyttä analyyttimolekyyliä kohtaan, ja niitä voidaan käyttää semikvantitatiiviseen sekä kvantitatiiviseen analysiin. Jäämien havainnoinnin lisäksi bioantureita voidaan käyttää antimikrobiaalisten aineiden tuottajien tunnistamiseen. MphR(E)-mutantit ovat lupaavia paranneltua säätelykykyä osoittavia reportterigeenin tuoton repressoreita käytettäväksi bioanturisovelluksissa. Ne ovat myös esimerkki rationaalisesta säätelyelementtien parantelusta uusien bioanturisovellusten kehittämiseksi.

Alasenjärven ja Sylvöjärven välinen järviketju sijaitsee Kymijoen vesistöalueen latvoilla Lahden kaupungin ja Nastolan kunnan alueella. Se on osa Arrajoen vesistöaluetta, joka laskee Kymijokeen. Järviketjuun kuuluvat Lahden Alasenjärvi, Lahden ja Nastolan rajalla sijaitseva Kymijärvi sekä Nastolassa sijaitsevat Kärkjärvi, Alvojärvi, Villähteen Kukkanen, Pikku-Kukkanen, Iso-Kukkanen, Salajärvi, Ruuhijärvi ja Sylvöjärvi sekä järvien väliset joet. Tavoitteena oli selvittää järvien vedenkorkeuksien ja virtaamien nykytila, niissä mahdollisesti tapahtuneet muutokset ja etsiä kehittämistarpeita. Nykytilan selvittämisen lisäksi työssä kartoitettiin vesistössä tehtyjä toimenpiteitä, joilla on vaikutettu veden virtaukseen ja vedenpinnan tasoon. Kehittämistarpeita ja niiden ratkaisemiseksi ehdotet-tuja toimenpiteitä tarkasteltiin kokonaisuutena vesiekosysteemin ja vesistön käyttäjien kannalta. Työssä käytettyjä tutkimusmenetelmiä olivat kirjallisuusselvitys, arkistoselvitys, ve-sistön käyttäjille tehty kyselytutkimus ja maastotyöt. Kirjallisuuden lisäksi työssä on käytetty lähteinä Hämeen elinkeino-, liikenne- ja ympäristökeskuksen arkistossa olevia asiakirjoja, ympäristöhallinnon tietojärjestelmiä ja alueella aiemmin tehtyjä selvityksiä. Työn teoriaosassa selvitettiin vedenkorkeuksien ja virtaamien vaihtelua, niihin vaikuttavia tekijöitä sekä esiteltiin mittaus- ja seurantamenetelmiä. Järviketjun järvet eroavat toisistaan sekä hydrologisten että vedenlaatuun liittyvien ominaisuuksien osalta. Kaikkien järvien virkistyskäyttö on runsasta. Säännöstelemättömillä Salajärvellä, Ruuhijärvellä ja Kymijärvellä vedenkorkeuden suuri vaihteluväli ja loppukesän matalat vedenkorkeudet on koettu haitalliseksi virkistyskäytön kannalta. Iso-Kukkasen, Pikku-Kukkasen ja Villähteen Kukkasen säännöstely Kumian padolla ei aina ole toteutunut tavoitteiden mukaisesti. Alasenjärven vedenkorkeuden laillinen taso on epäselvä ja sen luusuassa oleva Potilanjoen pato estää kalan kulun. Järviketjussa alimpana olevan Sylvöjärven ongelmana on läpi vuoden tasaisena pysyvä vedenkorkeus. Toimenpiteiksi ehdotetaan muun muassa vedenkorkeuden ja virtaaman seurannan aloittamista koko selvitysalueella, Potilanjoen puupadon muuttamista pohjapadoksi sekä selvityksiä Iso-Kukkasen ja yläpuolisten järvien säännöstelyn purkumahdollisuudesta ja Sala- ja Ruuhijärven kesäaikaisen vedenkorkeuden nostamisesta. Sylvöjärven tilan parantamiseksi ehdotetaan selvitystä laskujoesta peratun kosken ennallistamisen vaikutuksista Sylvöjärveen sekä valuma-aluekunnostusta. Toimenpiteiden suunnittelussa tulee huomioida niiden vaikutukset järven ja rantavyöhykkeen ekologiseen tilaan sekä vesistön eri käyttömuotoihin. /Kir12

Indoles have high structural flexibility, and the functionalization of the indole skeleton has extensively been investigated. Therefore various possibilities are offered for the design of catalytic ligands with an indole core. The theory part of this thesis is divided into three parts: the first part is an introduction on the use of chiral catalytic ligands with focus on oxazoline-containing ligands; the second part introduces the modification of the indole ring system at the N1-, C2- and C3-position; the third part gives an overall introduction on catalytic asymmetric allylic substitution with hetero-nucleophiles. In the results part of the thesis, the design and preparation of novel indole-phosphine-oxazoline (IndPHOX) ligands from readily available starting materials are presented. These modular ligands include an indole skeleton with either a phosphine moiety at the 2- or 3-position or an oxazoline ring at the 1-, 2- or 3-position, respectively. The IndPHOX ligands were first evaluated in palladium-catalyzed allylic alkylation with dimethyl malonate, generating the product in good yield and high ee. The ligands with a diphenylphosphine group and an oxazoline moiety at the 2 or 3-position, respectively, demonstrated high catalytic efficiency in palladium-catalyzed allylic amination with various N-nucleophiles, even with aromatic aniline having low nucleophilicity. N-functionalized IndPHOX ligands bearing various groups were also synthesized. The effects of N1-substituents to the reaction rate, yield and asymmetric induction in palladium-catalyzed asymmetric allylic amination have been discussed. In addition, the enhancement of enantioselectivity due to the presence of an oxygen atom in ligands with a N-MOM or a N-THP group has been presented in the thesis. Moreover, an IndPHOX ligand was utilized to afford 2-aryl substituted chromans with sterocontrol at C-2 via asymmetric allylic etherification using a novel synthetic route.

Ultraviolet (UV) radiation is an important factor affecting the life-time of polymer products. When polymers are exposed to UV radiation in the presence of oxygen they degrade due to photo-oxidative reactions. The changes in the mechanical properties and appearances of polymers are the result of chemical changes in the polymer structure. The observation of the early chemical changes in the photo-oxidative degradation of polypropylene (PP) and polystyrene (PS) was examined in this thesis. In this thesis Fourier transform infrared (FTIR) spectroscopy, ultraviolet-visible (UV-Vis) spectroscopy and differential scanning calorimetry (DSC) were used to study the changes that occurred in polypropylene and polystyrene when they were irradiated with a xenon arc lamp in air. The spectroscopic methods were used to observe the chemical changes in the polymer structure and the addition of new chemical groups and DSC was used to observe changes in the thermal properties and oxidative stability of the polymers. Differential photocalorimetry (DPC) was used as a novel method to observe changes in the oxidative stability of polymers with simultaneous irradiation. The DPC results were compared with the DSC results to examine how irradiation affects the oxidative stability of PP and PS. The largest change observed in the polymer samples during irradiation was the formation of a carbonyl group absorption band in the IR spectra located at ν ̃≈1720 cm^(-1). A substantial increase in the absorbance between λ=270–350 nm was observed in the UV-Vis spectra of PS, due mostly to the formation of conjugated double bonds. A small increase in the absorbance between λ=250–300 nm was observed in the UV-Vis spectra of PP, assigned to the formation of carbonyl groups. The melting temperature of PP decreased after irradiation, which was assigned to chain scission. The glass transition temperature of PS rose steadily for the first irradiation sequences, but decreased noticeably during the last one. The increase was assigned to crosslinking whereas the decrease was assigned to either chain scission or the photochain oxidation mechanism. The oxidation induction time (OIT) of PP decreased over 90 % during the first irradiation sequence. The oxidation occurred practically immediately for the irradiated samples, which indicates a large and fast decrease in thermal oxidative stability. The OIT test was not applicable for PS. The oxidation induction temperature (T_ox) decreased for PP and increased for PS due to photodegradation. This may be due to changes in the average molecular mass, with a decrease in mass indicating a decrease in the oxidation induction temperature and vice versa. Kun polymeerejä altistetaan UV-säteilylle hapen läsnä ollessa, ne rappeutuvat valon aikaansaamien hapetusreaktioiden ansiosta. Muutokset polymeerien mekaanisissa ja ulkoisissa ominaisuuksissa aiheutuvat kemiallisista muutoksista polymeerirakenteessa. Tässä diplomityössä tutkittiin varhaisten kemiallisten muutosten havaitsemista polypropeenin (PP) ja polystyreenin (PS) rappeutumisessa valon vaikutuksesta. Tässä työssä käytettiin Fourier’n muunnos infrapunaspektroskopiaa (FTIR), ultravioletti-näkyvä valo (UV-Vis) -spektroskopiaa sekä differentiaalista pyyhkäisykalorimetriaa (DSC) polypropeenissa ja polystyreenissä tapahtuvien muutosten havaitsemiseen, kun niitä säteilytettiin ksenon-kaarilampulla ilman läsnäollessa. Spektroskopisia menetelmiä käytettiin polymeerirakenteessa tapahtuvien muutosten ja uusien funktionaalisten ryhmien muodostumisen havaitsemiseen, ja DSC:tä käytettiin havaitsemaan muutoksia polymeerien termisissä ominaisuuksissa ja hapettumisen kestossa. Differentiaalista fotokalorimetriaa (DPC) käytettiin uutena menetelmänä muutosten havaitsemiseen hapettumisen kestossa samanaikaisen säteilytyksen kanssa. DPC:llä mitattua hapettumisen kestoa verrattiin DSC:llä mitattuun säteilytyksen vaikutuksen määrittämiseksi PP:n ja PS:n hapettumisen kestossa. Suurin muutos säteilytetyissä polymeerinäytteissä oli IR-spektreihin aaltoluvulle ν ̃≈1720 cm^(-1) muodostunut karbonyyliryhmän absorptiovyö. Polystyreenin UV-Vis-spektreissä havaittiin huomattava absorbanssin kasvu välillä λ=270–350 nm, mikä johtui konjugoituneiden kaksoisidosten muodostumisesta. Polypropeenin UV-Vis spektreissä havaittiin pieni absorbanssin kasvu välillä λ=250–300 nm, mikä johtui karbonyyliryhmien muodostumisesta. Polypropeenin sulamislämpötila laski säteilytyksen jälkeen, minkä aiheutti polymeeriketjujen katkeaminen. Polystyreenin lasittumislämpötila kasvoi tasaisesti ensimmäiset säteilytysjaksot, mutta laski huomattavasti viimeisen aikana. Kasvu johtui polymeeriketjujen silloittumisesta, kun taas lasku oli seurausta joko ketjujen katkeamisesta tai valoketjuhapettumismekanismista. Polypropeenin hapettumisen alkamisaika (OIT) laski yli 90 % ensimmäisen säteilytysjakson aikana. Säteilytettyjen näytteiden hapettuminen tapahtui käytännössä välittömästi, mikä viittaa huomattavaan ja nopeaan laskuun termisen hapettumisen kestossa. OIT-mittaus ei soveltunut PS:lle. Hapettumisen alkamislämpötila (T_ox) laski PP:lle ja kasvoi PS:lle valon aikaansaaman hajoamisen seurauksena. Tämä voi olla seurausta keskimääräisen molekyylimassan muutoksista, jolloin massan lasku vastaa laskua hapettumisen alkamislämpötilassa ja päinvastoin.

Respiratory Complex I is the main generator of Reactive Oxygen Species (ROS) inside the cell. ROS are generated during aerobic respiration, which occurs inside the mitochondria and generates most of the energy used by aerobic organisms. In this study we have provided evidence supporting the Mitochondrial Free Radical Theory of Aging (MFRTA), which states that ROS, produced inside the mitochondria cause aging. To test this we have utilised the power of Drosophila genetics to study the effects knocking-down one of the subunits of Complex I has on the Complex I assembly, energy homeostasis, and lifespan in Drosophila melanogaster. A series of crosses were performed, in which an RNAi construct for the gene CG6020 (encoding the 39kDa subunit of complex I) was combined with ectopic expression of the yeast Ndi1 protein to compensate the redox disequilibria caused by a lack of NADH re-oxidation in flies expressing only the knock-down. Both the RNAi construct and the NDI1 transgene were expressed using the binary UAS/GAL4 expression system. The resulting progeny were used to evaluate the knockdown of Complex I and to perform lifespan studies and complementary experiments. The knockdown of the nuclear encoded subunit of complex I (CG6020) was verified at the RNA level, protein level and at the whole-complex level by qPCR, western blotting and Blue Native Gel electrophoresis, respectively. The decrease in complex I respiration was also quantified by polarographic measurements using an Oroboros instrument (Oxygraph 2k), and the change in hydrogen peroxide production was studied in isolated mitochondria. Moreover, other physiological parameters –fertility, activity, food intake- were also studied to understand how longevity was affected in flies expressing the complex I knockdown. The results from shown here illustrate that, the knockdown of the 39kDa subunit (encoded by CG6020) specifically decreases the concentration of complex I. This specifically affects complex I-linked respiration, and at the same time reduces the generation of hydrogen peroxide in isolated mitochondria. Moreover, knock-down of complex I increases life-span without affecting fly mobility or food intake. Despite a few shortcomings, substantial evidence has been provided to show that, the increase in life-span of the flies is only due to reduction in the concentration of complex I.

The main focus in this thesis was to study the suitability of flexographic printing and new electrode materials in the manufacture of flexible piezoelectric pressure sensors. There are numerous application where flexible and economical pressure sensors can be used. Such are process controll in industry and measurement of vital functions and pressure stress in health care. Flexography would enable economical and efficient mass-production of such sensors. Function of the pressure sensors fabricated in this work is based on piezoelectricity of the uniaxially stretched and poled polyvinylidene fluoride (PVDF) that is used as substrate in sensors. When piezoelectric PVDF is exposed to mechanical stress, electric charges of opposite signs (voltage difference) form on opposite sides of the substrate. If conducting electrodes are fabricated on surface of the substrate, formed charges can be conducted to measuring unit that analyses the magnitude of the charge. Because formed charge is proportional to the force applied on PVDF, force or pressure applied on sensor can be solved. Because piezoelectric PVDF cannot resist high temperatures, no inks that require high sintering temperatures can be used as electrode materials. Manufacture of metal electrodes by sputtering or evaporation is slow and expensive. In this work carbon nanotubes and conducting poly(3,4-ethylenedioxythiophene) polymer are studied as alternative electrode materials. Electrodes are fabricated by using RK Flexiproof 100 flexo-printer or CX202 Motorized bar coater. For lower price and better availability also non-piezoelectric substrates were used to evaluate printing process and electrode materials. Fabricated samples were electrically characterized. Main focus was in sheet resistance of the electrodes and sensitivity of the sensor elements. Obtained sheet resistance values varied a lot. Highest values were up to over four orders of magnitude larger than those of metal electrodes. Despite the high sheet resistance, fabricated samples showed sensitivities comparable to reference samples. According to the results of this work, studied new electrode materials are most likely suitable to be used in flexible pressure sensors.

Surrogate models (also called response surface models or metamodels) have been widely used in the literature to solve continuous black-box global optimization problems that have computationally expensive objective functions. Different surrogate models such as radial basis functions, kriging, multivariate adaptive regression splines, and polynomial regression models have been used in various applications. It is in general however unknown which model will perform best for a given application, and computation time restrictions do not allow trying different models. Thus, in the first part of this thesis, a family of algorithms (SO-M, SO-M-c, SO-M-s) based on using a mixture of surrogate models is developed. The second part of the thesis extends the research in using surrogate models for mixed-integer (algorithm SO-MI) and purely integer (algorithms SO-I) optimization problems. Finally, a real world application problem arising in the agricultural land use management of a watershed is examined (algorithms SO-Ic). The algorithm SO-M uses Dempster-Shafer theory to combine information derived from various model characteristics in order to determine the influence of individual models in the mixture. Extensions of SO-M with respect to the sampling strategy (algorithms SO-M-c and SO-M-s) have been compared in numerical experiments, and it was found that whenever it is a priori unknown which surrogate model should be used, it is advisable to use a mixture model in order to prevent accidentally selecting the worst model. It could be shown that mixture models containing radial basis function interpolants generally work very well, whereas using only polynomial regression models should be avoided. Moreover, algorithms using mixture models often outperform the algorithms that use only the single models that are contributing to the mixture. Although there are many computationally expensive black-box optimization applications that have besides continuous also integer variables, or that have only integer variables, algorithms for solving these types of problems are scarce. In the second part of this thesis two algorithms, namely SO-MI for mixed-integer problems, and SO-I for purely integer problems have been developed and were shown to find accurate solutions for computationally expensive problems with black-box objective functions and possibly black-box constraints. The constraints were treated with a penalty approach and numerical experiments showed that the surrogate model based algorithms outperformed commonly used algorithms for (mixed-) integer problems such as branch and bound, and genetic algorithms. Also NOMAD (Nonsmooth Optimization by Mesh Adaptive Direct Search) has been included in the comparison. NOMAD is suitable for integer and mixed-integer black-box problems, but its performance for these problem types has not been studied in the literature. In the numerical experiments, NOMAD also proved superior as compared to branch and bound and the genetic algorithm, but it performed worse than SO-I and SO-MI for most test problems. Lastly, the algorithm SO-I has been further extended to directly handling constraints with a response surface. The algorithm, SO-Ic, has been developed specifically for a watershed management problem that has only one constraint, but SO-Ic is easily generalizable for problems with more constraints. In the considered application problem parts of the agricultural land in the Cannonsville reservoir watershed in upstate New York have to be retired in order to decrease the total phosphorus runoff to a given limit at minimal cost. A computationally expensive simulation model has to be used to compute the costs and phosphorus runoff. The performance of SO-Ic has been compared to a genetic algorithm, NOMAD, and the discrete dynamically dimensioned search algorithm on three problem instances with different sizes of the feasible region. The surrogate model based algorithm SO-Ic performed also for these problems significantly better than all other algorithms and could be shown to be the most robust.

Tässä työssä testataan Complex Systems Computation Group -tutkijaryhmän kehittämän B-course-ohjelmiston riippuvuuksien mallinnusmenetelmän soveltuvuutta liikenneonnettomuusdatan tutkimiseen. Ohjelmiston datan analysointi perustuu bayesverkkoihin, jotka ovat todennäköisyysmalleja. Riippuvuuksien mallinnusmenetelmä etsii mallin, joka parhaiten kuvaa aineiston muuttujien välisiä riippuvuussuhteita. B-course-ohjelmistossa on Java Playground -sovellus, jolla riippuvuusmallia voidaan tutkia tarkemmin. Tutkittavana aineistona on tietoja Pirkanmaalla vuosien 2004 - 2008 aikana tapahtuneista liikenneonnettomuuksista. Riippuvuusmallinnuksen testauksessa aineistosta tutkitaan muuttujien välisiä riippuvuussuhteita, niiden vahvuuksia sekä kausaalisuussuhteiden suuntaa ja laatua. Lisäksi testataan Java Playground -sovellusta tarkastelemalla lähemmin taajamissa ja taajamien ulkopuolella tapahtuneiden liikenneonnettomuuksien eroja, sekä eläinonnettomuuksia ja yli 65-vuotiaitten aiheuttamia liikenneonnettomuuksia. Työn tuloksena voidaan todeta, että B-course-ohjelmiston riippuvuuksien mallinnusmenetelmä soveltuu hyvin riippuvuussuhteiden löytämiseen käytetyn liikenneonnettomuusdatan tyyppisille aineistoille. Paljon muuttujia sisältävä aineisto on syytä jakaa pienempiin osiin tuloksena saatavan riippuvuusmallin tarkastelun helpottamiseksi. Tuloksina saatujen riippuvuus- ja kausaalisuusmallien muuttujien välisten suhteiden laatuun on hyvä suhtautua kriittisesti. Erityisesti kausaalisuussuhteiden suunta ei vaikuttaisi aina olevan sen suuntainen kuin loogisin perustein pitäisi olla.

Concerns about oil price, the strengthening climate change and traffic related health effects are all reasons which have promoted the research of renewable fuels. One renewable fuel candidate is diesel consisting of hydrotreated vegetable oils (HVO). The fuel is essentially paraffinic, has high cetane number (>80) and contains practically no oxygen, aromatics or sulphur. Furthermore, HVO fuel can be produced from various feedstocks including palm, soybean and rapeseed oils as well as animal fats. HVO has also been observed to reduce all regulated engine exhaust emissions compared to conventional diesel fuel. In this thesis, the effect of HVO fuel on engine exhaust emissions has been studied further. The thesis is roughly divided into two parts. The first part explores the emission reductions associated with the fuel and studies techniques which could be applied to achieve further emission reductions. One of the studied techniques was adjusting engine settings to better suit HVO fuel. The settings chosen for adjustments were injection pressure, injection timing, the amount of EGR and the timing of inlet valve closing (with constant inlet air mass flow, i.e. Miller timing). The engine adjustments were also successfully targeted to reduce either NOx or particulate emissions or both. The other applied emission reduction technique was the addition of oxygenate to HVO fuel. The chosen oxygenate was di-n-pentyl ether (DNPE), and tested fuel blend included 20 wt-% DNPE and 80 wt-% HVO. Thus, the oxygen content of the resulting blend was 2 wt-%. Reductions of over 25 % were observed in particulate emissions with the blend compared to pure HVO while NOx emissions altered under 5 %. On the second part of this thesis, the effect of the studied fuels on chosen surface properties of exhaust particles were studied using tandem differential mobility analyzer (TDMA) techniques and transmission electron microscopy (TEM). The studied surface properties were oxidizability and hygroscopicity. The oxidation properties were compared between soot particles emitted with HVO and with conventional diesel. The effects of fuel on hygroscopicity, on the other hand, were compared between HVO and oxygenated HVO fuel. In general, no considerable differences were observed in the exhaust particle surface properties due to the change of fuel.

Through research and technological advances, modern society has developed a variety of implantable devices that helps to optimize human’s life quality. One of these devices is the implantable cardioverter defibrillator (ICD). Our unit, ERP Environmental Health, in previous investigations has found that, in some cases, the ICD receives certain disturbances under the power lines. Therefore it is important to study the degree of immunity that implants have against electromagnetic fields, because even small interference that result from EMF exposure could deliver a major public health impact. The aim of this master thesis is to study the effect of extremely low frequency (ELF) non-uniform electric fields on implantable cardioverter defibrillators (ICD). This test was conducted in the laboratory of High Voltage Technology, University of Tampere, in a controlled climate room. The experiment consisted in introducing the ICD in the interior of a Human-Shaped phantom which was filled with a saline solution to emulate the conductivity within the human body; the phantom was subsequently exposed to an electric field between two parallel plates. Following the experiment, we created a three-dimensional model of the electromagnetic fields generated by a phantom. This allowed us to corroborate the experiment and calculate quantities that it was impossible to obtain physically, such equipotential lines and the electric field lines between the parallel plates. We also calculated the total induced current and compared with experimental results, obtaining in a worse case a mean relative error of 10.53%, which suggests that our calculations are in agreement with the experimental result. However, an additional comprehensive study is needed to obtain more conclusive results.

This work investigates the tuning of photophysical properties of two different classes of photoactive molecules, 2-(2’-hydroxyphenyl)benzothiazole (HBT) and perylene bisimide (PBI). The Suzuki coupling reaction was employed to synthesize aryl-substituted derivatives of HBT and PBI. The tuning of the photophysical properties of both HBT and PBI was successfully carried out by attaching either electrondonating (ED) or electron-withdrawing (EW) aryl moieties to the parent compounds. For HBT, a series of 5-substituted derivatives were synthesized to modify the highest occupied molecular orbital (HOMO) energy levels of zinc-benzothiazole (Znb₂) complexes. Perylene bay substitutions were used to tune the HOMO-LUMO (LUMO: lowest unoccupied molecular orbital) energy gap of the diaryl-PBIs. Spectroscopic and electrochemical techniques were used to determine the effects of substituents on the photophysical properties to the parent compounds. For the Znb₂ complexes, absorption and emission spectra as well as the HOMO energy levels are effectively modified by the aryl-substitution while the LUMO energy level remains largely unaffected. Whereas, for the PBI derivatives, the spectroscopic profiles as well as the HOMO and LUMO energy levels were tuned by the attached aryl groups. These results indicate that inductive effects are more dominant in both Znb₂ and PBI derivatives. All the involved reactions, especially borylation of both the HBT and PBI were studied in detail to achieve maximum yield and simple purification. The separation of a series of 1,7- and 1,6-diaryl-PBIs by normal column chromatography was an important step in perylene chemistry. The individual regioisomers of diaryl-PBIs were unambiguously characterized by observing the chemical shifts of α- methylene protons using 300 MHz ¹H NMR spectroscopy. Obtained results indicate that both 1,7- and 1,6-regioisomers of same diaryl-PBI possess slightly different opto-electronic properties, which might be important while designing a system for optical devices. Moreover, the synthesized Znb₂ complexes were used as anode buffer layers in thin film bulk-hetero junction (BHJ) based inverted organic solar cells. The presence of vacuum evaporated Znb₂ buffer layers was found to enhance the photovoltaic performance by up to 40 % when compared to a similar device containing tris(8-hydroxyquinlinato)aluminum(III) (Alq₃) buffer layer. The device stability also improved considerably when compared to the corresponding device with the Alq₃ buffer layer. Finally, benzothiazole-perylene bisimide (HBT-PBI) dyads, in which two chromophore units were linked in close proximity to each other, were synthesized and studied by optical spectroscopy to explore a new donor-acceptor light-harvesting photovoltaic antennae system.

The number of people affected by degenerative diseases of the retina increases notably due to the longer life expectancy. The weakening of the sight concerns especially older people, who suffer from degeneration of the retina and its retinal pigment epithelium (RPE) monolayer. Currently, these degenerative diseases of the retina are incurable. Cell transplantation of healthy RPE cells is considered to be a potential treatment for these diseases. Due to the unlimited self-renewal capacity and pluripotency human pluripotent stem cells (hPSCs) are considered as possible cell sources for transplantation purposes. However, the current differentiation protocols for RPE cells from hPSCs are inefficient and result in low yields. The aim of this thesis was to study whether biodegradable poly(L-lactide) (PLLA) microparticles and commercial biostable cellulose anion exchange diethylaminoethyl (DE) 53 microparticles could be exploited in increasing the efficacy of differentiation of hPSCs into RPE cells. In addition, the potential of the microparticle culture system for stem cell maintenance in feeder-free conditions was evaluated. The attachment and proliferation of hPSCs on microparticles were analyzed by immunofluorescence analysis and microscopy. The success of differentiation of RPE from stem cells in microparticle cultures was evaluated by inspecting the appearance of pigmentation in cultures as well as by investigating the expression of RPE maturation markers on gene expression levels by RT-PCR. The PLLA microparticles did not support the attachment and proliferation of hPSCs in feeder-free conditions. The poor performance of PLLA particles in cell culture studies might be due to the hydrophobic nature of the particle surfaces. In contrast, hPSCs attached, proliferated and maintained their pluripotency on DE 53 particles. Furthermore, the hPSCs were successfully differentiated into RPE cells on these DE 53 particles. However, the differentiation efficacy did not outperform the conventional differentiation protocol. This was the first study in which the hPSCs were successfully differentiated into RPE cells on microparticles. Microparticles are far from being routinely used in hPSC maintenance and differentiation to mature cell types. More long-term and large-scale studies need to be conducted in the future. Nevertheless, at the moment they seem appealing and promising with a possibility to grow stem cells in large-scale and cost effectively. The microparticle culture systems set challenges for the cell analysis due to their 3-dimensional structure and autofluorescence property. Therefore, novel methods for characterization, analysis and imaging of microparticle culture systems should be developed. /Kir12
