Abstract

The influence of the P–M–P bite angle in diphosphine ligands on selectivity has been observed in various catalytic reactions. A better understanding of the ligand bite angle concept is important for the rational design of efficient catalytic systems. In the present work, the mechanism of cobalt-catalyzed C–H functionalization of aldehydes with enynes and how the diphosphine ligands alter regioselectivity were investigated by density functional theory (DFT) calculations. The catalytic cycle is initiated by the oxidative cyclization of enynes rather than the oxidative addition of aldehydes. Regioselectivity arises from competing σ-bond metathesis and migratory insertion steps, in which the steric effects of diphosphine ligands are the dominant factors influencing the activation barriers. The calculations indicate that σ-bond metathesis is more challenging and its feasibility is highly dependent on the ligand bite angle. The improved mechanistic understanding will enable further design of transition-metal-catalyzed selective cyclization reactions. [ABSTRACT FROM AUTHOR]

Abstract

Debranched Canna edulis Ker starch (DCEKS) was effectively esterified in 1-butyl-3- methylimidazolium chloride (ionic liquid) to shorten the esterification time of phthalic anhydride and enhance the substitution degree. The results indicated that the esterification of phthalic anhydride obviously improved the thermal stability of CEKS, while the debranching slightly increased the thermal stability of CEKS. DCEKS was still a semicrystalline polymer like CEKS, and its crystalline structure was not changed by the debranching, but phthalate debranched Canna edulis Ker starch (PDCEKS) was almost an amorphous polymer due to the high esterification in the ionic liquid. The esterification decreased the average size of DCEKS, whereas the debranching increased the average size. The esterification was unevenly completed on DCEKS particles. Both debranching and esterification could result in a decrease in the freeze–thaw stability, swelling power and viscosity of CEKS. The stretching vibration of benzene rings appeared at the wave numbers of 1598 cm−1, 1077 cm−1 and 640 cm−1.

149. SNP in pre-miR-1666 decreases mature miRNA expression and is associated with chicken performance

Abstract

Polymorphisms in miRNA genes could potentially alter various biological processes by influencing the processing and (or) target selection of miRNAs. The rs14120863 (C > G) mutation, which we characterized in a Gushi-Anka [F.sub.2] resource population, resides in the precursor region of miR-1666. Association analysis with chicken carcass and growth traits showed that the SNP was significantly associated with carcass weight, evisceration weight, breast muscle weight, leg muscle weight, and body weight at 8 weeks of age, as well as some body size indexes including shank girth, chest breadth, breast bone length, and body slanting length, in the Gushi-Anka [F.sub.2] resource population. Quantitative RT-PCR results showed that miR-1666 expression levels in muscle tissues differed within various genotypes. Experiment in DF1 cells further confirmed that the SNP in miR-1666 could significantly alter mature miRNA production. Subsequently, using dual-luciferase report assay, we verified that miR-1666 could perform its function through targeting of the CBFB gene. In conclusion, the SNP in the precursor of miR-1666 could significantly reduce mature miR-1666 production. It may further affect the function of miR-1666 through the target gene CBFB, hence it is associated with chicken growth traits. Key words: chicken, microRNA, biogenesis, SNP, CBFB. Le polymorphisme au sein de genes codant pour des miARN pourrait potentiellement alterer divers processus biologiques en influencant la maturation ou la selection de la cible de ces miARN. La mutation rs14120863 (C > G), que les auteurs ont caracterise au sein de la population [F.sub.2] Gushi-Anka, est situee dans la region precurseur du miR1666. Une analyse d'association a montre que ce SNP etait significativement associe avec le poids de la carcasse, le poids eviscere, le poids de la poitrine desossee, le poids de lacuisse desossee, le poids ahuit semaines ainsique certains indices corporels comme la circonference du jarret, l'angle de poitrine, la longueur du brechet and la longueur du dos au sein de la population [F.sub.2] Gushi-Anka. Les resultats d'une analyse RT-PCR quantitative ont montre que l'expression du miR-1666 dans les muscles etait differente chez differents genotypes. Une experience avec des cellules DF1 a confirme que le SNP au sein du miR-1666 pouvait changer de maniere significative la production de miARN mature. Ensuite, a l'aide d'un rapporteur a luciferase double, les auteurs ont confirme que le miR-1666 pouvait agir en ciblant le gene CBFB. En conclusion, le SNP situe dans le precurseur du miR-1666 pourrait entrainer une reduction significative de la production de miR-1666 mature. Il pourrait egalement affecter la fonction du miR-1666 via le gene-cible CBFB, d'ou son association avec les composantes de croissance chez le poulet.
