Objective: Programmed cell death protein-1 (PD-1) inhibitors are widely u sed for the treatment of hepatocellular carcinoma (HCC). Thyroid dysfunction is common in patients treated with this therapy, although the dynamic changes in thyr oid function and sonographic features remain unclear. Methods: We analyzed 38 patients with HCC who received anti-PD-1 therap y at our hospital. Demographic, clinical, laboratory, and ultrasound data were extracted from electronic medical records. The grading of thyroid nodules was based on the American College of Radiology Thyroid Imaging Reporting and Data System classification. Statistical analyses were performed using GraphPad Prism 5.0. Results: Fifteen patients (40%) had hypothyroidism, among which six had hypothyroidism at baseline, three had overt hypothyroidism, and six had subcli nical hypothyroidism after anti-PD1 therapy. The proportion of patients with euthyro id function and thyroid antibody positivity was significantly lower than that of patient s with thyroid dysfunction (10% vs 39%, P < 0.05). Nine patients (24%) had irregular echo patterns on s onographic imaging, six of whom had irregular echo patterns present during the treatment, but only one had them persist until the end of treatment. At baseline, t he classification of most thyroid nodules was grade 3, with a significant increase in grade 4A and 4B classifications during treatment, though most nodules remained grade 3 at the e nd of treatment. There were no significant differences in survival rates between t he euthyroid and thyroid dysfunction groups. Conclusion: Anti-PD-1 therapy-induced thyroid dysfunction was accompanied by changes in thyroid function, antibodies, and ultrasonography. Therefore, in patients receiving anti-PD-1 therapy, close, dynamic monitoring of thyroid function, antibodies, and ultrasonographic characteristics is necessary.

Objective To analyze the safety of apheresis granulocyte(AG) collection from blood donors mobilized by G-CSF and apheresis granulocyte transfusion efficacy in patients. Methods The blood routine results, collection process and follow-up of blood donors mobilized by G-CSF before and after AG collection were collected to analyze the safety of AG collection, and the blood routine results, clinical symptom improvement and treatment outcome of patients before and after AG transfusion were collected to analyze the transfusion efficacy. Results A total of 27 blood donors donated 29 U AG, with collection time at (229±20)min, circulating blood volume at (9 890±1 107)mL, and the dosage of anticoagulant at (1 002±97)mL.Two blood donors had adverse reactions to blood donation, and the AG collection was carried out after treatment.After G-CSF mobilization, WBC increased significantly from (5.61±1.06) ×109/L to (22.85±5.23) ×109/L, while RBC, Hb, Hct and Plt showed no significant change.The blood routine returned to the level before G-CSF mobilization 1-2 days after blood donation.No physical discomfort occurred during the one week after blood donation.Four patients with granulocyte deficiency complicated with multidrug-resistant bacterial infection, who failed to respond to antibiotic treatment, were transfused with 29 U AG, with no adverse reactions and no obvious change in blood routine, but the infection symptoms were improved significantly judged from clinical manifestation, bacterial culture results, temperature monitoring and CT examination, suggesting that the AG infusion was effective.Among the 4 patients, 1 was cured and discharged, 1 gave up treatment, 1 died of sepsis, and 1 died of multiple organ failure. Conclusion It is safe to collect AG from blood donors mobilized by G-CSF through blood cell separator, and the AG products basically meet the national quality requirements and the treatment needs.Sustained high-dose AG transfusion has a significant effect on infection control in patients with agranulocytosis combined with refractory multidrug-resistant bacterial or fungal infection.

Abstract Background Benign prostate hyperplasia (BPH) is the most common urological problem in elderly males. Recent studies have reported polymorphism in various metabolic genes in BPH. However, their association with the susceptibility of BPH is still inconsistent. Here, we systematically reviewed and performed a meta-analysis of CYP17, VDR, and ACE genes to determine their precise association with the risk of BPH. Methods A comprehensive literature search for published studies on candidate gene associations involving vitamin D receptor (VDR), angiotensin-converting enzyme (ACE), and CYP17 genes with the risk of BPH was done up to April 2020 in PubMed, Scopus, Cochrane Central Register of Controlled Trials (CENTRAL), and Google Scholar databases. Fixed/random effects models were used to estimate the odd’s ratio (OR) and 95% confidence intervals (CIs). Begg’s funnel plot was used to assess the potential for publication bias. Results We found a total of 23 studies containing 3461 cases and 3833 controls for these gene polymorphisms. A significant association of ACE gene polymorphism was observed under the recessive (II vs. ID + DD) model for BPH susceptibility compared to control subjects (overall OR = 1.67, 95% CI = 1.03–2.73). Similar trends were observed for ACE gene polymorphism in Caucasian (OR = 6.18, 95% CI = 1.38–27.68) and Asian (OR = 1.42, 95% CI = 0.99–2.03) populations under study. No significant association was observed in VDR and CYP17 gene polymorphisms in any dominant or recessive models. Conclusion Significant OR demonstrated the implication of ACE gene polymorphism in the proliferation of prostate tissue, which in turn is associated with BPH susceptibility. However, prospective studies at large scale and sample size are needed to confirm the current findings.

Abstract Backgroud and purpose Triglyceride-glucose (TyG) index has been considered a surrogate marker of insulin resistance. We investigated the association between TyG index and stroke recurrence and compared the effectiveness of TyG index with homeostasis model assessment of insulin resistance (HOMA-IR) in predicting stroke recurrence and death in nondiabetic acute ischemic stroke patients. Methods Nondiabetic acute ischemic stroke patients from the ACROSS-China (Abnormal Glucose Regulation in Patients with Acute Stroke across China) registry were included. TyG index was performed and classified into four groups by quartiles. The outcomes were stroke recurrence and death within 1 year. The association between TyG index and the risk of stroke recurrence and death were analyzed by Cox regression models. Receiver operating characteristic (ROC) curve analysis was performed to evaluate the prediction of TyG index and HOMA-IR for stroke recurrence and death. Delong test was used for comparing the differences between area under the curve (AUC) of TyG index and HOMA-IR. Results Among the 1226 patients included, the median (interquartile range) of TyG index was 5.8 (5.5–6.2). Both the third and fourth quartiles of TyG index were associated with an increased risk of stroke recurrence (adjusted hazard ratio 2.04, 95% confidence interval 1.26–3.31; adjusted hazard ratio 1.86, 95% confidence interval 1.13–3.06). Patients with fourth quartiles of TyG index were associated with a higher mortality risk (adjusted hazard ratio, 2.91; 95% confidence interval, 1.62–2.53). Regarding stroke recurrence within 1 year, the AUC (95% confidence interval) of the ROC curve for the TyG index was similar to that of the HOMA-IR[0.56 (0.52–0.61) vs 0.57 (0.52–0.61); P = 0.93]. Regarding death within 1 year, the AUCs (95% confidence interval) of the ROC curve for the TyG index and HOMA-IR were 0.55 (0.50–0.61) and 0.59 (0.53–0.64), respectively (P = 0.32). Conclusions Elevated TyG index was associated with an increased risk of stroke recurrence and death. However, neither of TyG nor HOMA-IR can be a qualified predictor of stroke recurrence and death in nondiabetic acute ischemic stroke patients.

Objective: Both primary thyroid lymphoma (PTL) and diffuse sclerosing vari ant of papillary thyroid carcinoma (DSVPTC) are two rare malignant tum ours with different therapies and prognoses. This study compared their clinical fea tures. Methods: From a retrospective review of the pathologic database at our institute between January 2015 and August 2020, 52 PTL patients and 40 DSVPTC pat ients were included. Demographic, clinical, laboratory and ultrasound data were extr acted from electronic medical records. Statistical analyses were performed using Grap hPad Prism 5.0. Results: Both PTL and DSVPTC were more likely to occur in women (83.7 and 67.5%, respectively), but DSVPTC patients were younger (median age: 36 vs 64.5), had fewer compressive symptoms, and more frequently had neck lymph node metastasis than PTL patients. The prevalence of Hashimoto’s thyroiditis (HT) and hypothyroidism was significantly higher in PTL patients than in DSVPTC patients (31% vs 17.5%). Hyperthyroidism could only be found in DSVPTC patients, which accounted for 7.5%. Heterogeneous echogenicity and irregular edges were frequently observed in both PTL and DSVPTC. However, compared with PTL, DSVPTC exhibited smaller lesion sizes, higher frequencies of diffuse sonographic patterns and calcification and lower frequencies of hypoechoic features and internal blood flow signal. The overall survival rate with PTL was 77.23%, which was lower than that with DSVPTC (90.91%), but this difference was not significant (P = 0.096). Conclusion: Clinical characteristics such as age, compression symptoms, and sonographic features such as a large mass with heterogeneous echogenicity, hypoechoic, irregular edges, and calcification are helpful for impression diagnosis of PTL and DSVPTC before surgery.

Chemodynamic Therapy (CDT) based on Fenton reaction that can produce highly toxic reactive oxygen species to kill cancer cells has attracted considered attention. However, the effect of the CDT is limited in specific conditions (e.g. pH and H2O2 content) in tumor microenvironment (TME). In this paper, a glucose oxidase (GOx) and Fe3+-loaded metal organic framework system (GOx@MOF@Fe3+), to enhance the cascade reaction of the therapy, was developed for combined starvation therapy and CDT with the assistance of electrolysis. The GOx@MOF@Fe3+ in the tumor microenvironment was decomposed and thus released the Fe3+ and GOx. The electrolysis procedure, can not only trigger the Fe3+ and Fe2+ cyclic reactions, also provided O2 for the oxidation and decomposition of glucose to generate H2O2. This resulted in the improvement of the pH in TME for Fenton reaction, to enhance the CDT treatment effect. Meanwhile, glucose and O2 were consumed by GOx to achieve the effect of blocking nutrients. Compared with other control groups, the GOx@MOF@Fe3+ under electrolysis showed the considerable inhibitory effect of tumor cells, and the apoptosis rate even reached 80%. Our work presents an ingenious nanoenzyme for Fenton reactions that achieves a self-supplying strategy under electrolysis to improve anti-tumor efficacy.

Sepsis is a life-threatening organ dysfunction due to dysregulated host responses induced by infection. The presence of immune disturbance is key to the onset and development of sepsis but has remarkably limited therapeutic options. Advances in biomedical nanotechnology have provided innovative approaches to rebalancing the host immunity. In particular, the technique of membrane-coating has demonstrated remarkable improvements to therapeutic nanoparticles (NPs) in terms of tolerance and stability while also improving their biomimetic performance for immunomodulatory purposes. This development has led to the emergence of using cell-membrane-based biomimetic NPs in treating sepsis-associated immunologic derangements. In this minireview, we present an overview of the recent advances in membrane-camouflaged biomimetic NPs, highlighting their multifaceted immunomodulatory effects in sepsis such as anti-infection, vaccination, inflammation control, reversing of immunosuppression, and targeted delivery of immunomodulatory agents.

BackgroundAcute thrombus in atrial septal defect occluders is a rare complication that requires aggressive, effective, and safe management. Tirofiban, a platelet glycoprotein IIb/IIIa receptor antagonist, is widely used for the management of thromboembolic diseases, such as coronary heart disease and stroke. To date, there is no report using the GPIIb/IIIa receptor antagonist tirofiban for the management of ASD closure-related thrombosis in children.Case presentationHerein, we reported a case of a 5-year-old girl with ASD who presented with acute thrombus on the left disc of the occluder device immediately after transcatheter closure of ASD. The thrombus was successfully dissolved 24 h after a combined infusion of heparin and tirofiban, followed by 1 months of aspirin and clopidogrel and 5 months of aspirin alone. No thromboembolism or hemorrhage events occurred during follow-up for more than 2 years.ConclusionThe continuous infusion of GPIIb/IIIa receptor antagonist tirofiban combined with heparin may have beneficial effects for the management of thrombosis during ASD closure procedure.

ObjectiveTo understand the varieties, evaluation, treatment, and prognosis of severe neurological diseases using the third NCU survey in China.DesignA cross-sectional questionnaire study. The study was completed in three main steps: filling in the questionnaire, sorting out the survey data, and analyzing the survey data.ResultsOf 206 NCUs, 165 (80%) provided relatively complete information. It was estimated that 96,201 patients with severe neurological diseases were diagnosed and treated throughout the year, with an average fatality rate of 4.1%. The most prevalent severe neurological disease was cerebrovascular disease (55.2%). The most prevalent comorbidity was hypertension (56.7%). The most prevalent complication was hypoproteinemia (24.2%). The most common nosocomial infection was hospital-acquired pneumonia (10.6%). The GCS, APACHE II, EEG, and TCD were the most commonly used (62.4–95.2%). The implementation rate of the five nursing evaluation techniques reached 55.8–90.9%. Routinely raising the head of the bed by 30°, endotracheal intubation and central venous catheterization were the mostprevalent treatment strategies (97.6, 94.5, and 90.3%, respectively). Traditional tracheotomy, invasive mechanical ventilation and nasogastric tube feeding (75.8, 95.8, and 95.8%, respectively) were more common than percutaneous tracheotomy, non-invasive mechanical ventilation and nasogastric tube insertion (57.6, 57.6, and 66.7%, respectively). Body surface hypothermia brain protection technology was more commonly used than intravascular hypothermia technology (67.3 > 6.1%). The rates of minimally invasive hematoma removal and ventricular puncture were only 40.0 and 45.5%, respectively.ConclusionIn addition to traditional recognized basic life assessment and support technology, it is necessary to the use of promote specialized technology for neurological diseases, according to the characteristics of critical neurological diseases.

Traditional hub location problems are usually based on deterministic circumstances. However, many uncertain factors can cause demand to vary in the long run, which increases the difficulty of strategic hub location planning. The hub location problem for fresh agricultural products is studied considering the perishability of the products and the uncertainty of customer demands. An uncertain demand variable is described by an affine function of the nominal mean and several independent uncertainty sources and is further adjusted by the deterioration rate. An uncapacitated robust hub location model for fresh agricultural products with uncertain demand is first established and solved using the Lagrangian relaxation approach. Then, a robust optimization model for the corresponding capacitated hub location problem is given. A numerical study based on the Australian Post data set (AP20) shows that the deterioration rate of fresh agricultural products, the uncertainty of demand and the degree of conservatism of decision makers all have significant impacts on the total transportation profit. Furthermore, the capacitated model yields more profit than the uncapacitated model because it allows the effects of the deterioration rate and uncertainty to be moderated through flow reallocation. The proposed models are useful for helping decision makers determine the locations and capacities of hubs for fresh agricultural products in accordance with different risk preferences.

Abstract This work employed an inductively coupled plasma wind tunnel to study the dynamic oxidation mechanisms of carbon fiber reinforced SiC matrix composite (Cf/SiC) in high-enthalpy and high-speed plasmas. The results highlighted a transition of passive/active oxidations of SiC at 800–1600 °C and 1–5 kPa. Specially, the active oxidation led to the corrosion of the SiC coating and interruption of the SiO2 growth. The transition borders of active/passive oxidations were thus defined with respect to oxidation temperature and partial pressure of atomic O in the high-enthalpy and high-speed plasmas. In the transition and passive domains, the SiC dissipation was negligible. By multiple dynamic oxidations of Cf/SiC in the domains, the SiO2 thickness was not monotonously increased due to the competing mechanisms of passive oxidation of SiC and dissipation of SiO2. In addition, the mechanical properties of the SiC coating/matrix and the Cf/SiC were maintained after long-term dynamic oxidations, which suggested an excellent thermal stability of Cf/SiC serving in thermal protection systems (TPSs) of reusable hypersonic vehicles.

Nitric oxide (NO)—the product of arginine metabolism catalyzed by nitric oxide synthases (NOS)—is a well-known neurotransmitter which plays an important role in metabolism and amino acid transportation in the nervous system. In particular, it can inhibit monoamine neurotransmitter transportation which affects animal behavior, especially social behavior. Shoaling—is a one kind of social behavior. It is a behavior that individual fish choose to join with their group within two factors; food and predation risk. Shoaling fish has quickly responded to predator and increased the change in feeding competition. In addition, shoaling also effect to stress response on stock density of aquaculture system. The effect of NO molecular signaling on the dopamine pathway was investigated using zebrafish (Danio rerio) as a model organism. Our aim was to understand the role of NOS and NO in shoaling behavior, which is typical of zebrafish. The concentration of NO in the zebrafish brain was modulated using a knockout for the neuronal NOS gene, and NO production was induced through treatment with L-arginine. The existence of NO in the zebrafish brain was confirmed by using a fluorescent probe. Dopamine concentration in the brain was measured by UPLC tandem mass spectrometer. We measured shoaling cohesion of all individual fish of D. rerio, using average distance between all pairs of fish (nearest neighbor distance) and analyzed tracking by Zebralab ViewPoint software. Collectively, our results suggest that a lower level of NO was associated with a higher level of dopamine, which in turn leads to the shoaling behavior.

ObjectiveThis article aims to explore the potential categories of college students’ sports behavior motivation and the differences between different family social classes on potential categories.ParticipantsIn total, 1,092 college students were investigated in this study.MethodsThis article used the “College Students’ Sports Behavior Motivation Questionnaire” to survey 1,092 college students using the whole group sampling method. The potential profile analysis method was applied to classify the types of college students’ sports behavior motivation and further analyze the characteristics of different family social classes and sports behavior motivation.ResultsCollege students’ sports behavior motivation types can be divided into the following four categories: “low motivation type” (0.82%), “healthy appearance type” (11.45%), “healthy fun type” (37.36%), and “high motivation type” (50.37%). The higher the family’s social class, the more the sports behavior motivation of college students tends to be healthy appearance, health, fun, and high motivation. The lower the family social class, the more motivational sports behavior of college students tends to be low motivation. The better the perception of health, the higher the probability that college students belong to the high-motivation type. The fewer chronic diseases that college students have are associated with a healthy appearance and high motivation.ConclusionThere is a certain relationship between the type of college students’ sports behavior motivation and their family social class. Therefore, in school physical education, we should open all kinds of sports activities according to college students’ sports behavior motivation needs and focus on college students with lower family social class in order to intervene precisely on their motivation for sports behaviors, improve participation, and promote the overall health of college students.

In this study, we explored the relationship between the platelet endothelial aggregation receptor 1 (PEAR1) polymorphisms, platelet reactivity, and clinical outcomes in patients with minor stroke or transient ischemic attack (TIA). Randomized controlled trial subgroups were assessed, wherein patients received dual antiplatelet therapy for at least 21 days. Platelet reactivity was measured at different time intervals. Genotypes were categorized as wild-type, mutant heterozygous, and mutant homozygous. Clinical outcomes were evaluated after 90 days. The rs12041331 polymorphism predominantly influenced adenosine diphosphate channel platelet activity, with the AA genotype displaying significantly lower residual platelet activity to the P2Y12 response unit (p < 0.01). This effect was more evident after 7 days of dual antiplatelet treatment (p = 0.016). Mutant A allele carriers had decreased rates of recurrent stroke and complex endpoint events but were more prone to bleeding (p = 0.015). The rs2768759 polymorphism majorly impacted arachidonic acid (AA) channel platelet activity, which was particularly noticeable in the C allele carriers. Our regression analysis demonstrated that rs12041331 AA + GA and rs2768759 CA predicted 90-day post-stroke bleeding. In conclusion, the PEAR1 polymorphisms rs12041331 and rs2768759 interfere with platelet aggregation and the performance of antiplatelet drugs. These genetic variations may contribute to bleeding events associated with minor stroke and TIA.

Detecting infrared (IR) small targets effectively and robustly is crucial for the tasks such as infrared searching and guarding. While methods based on the human vision system (HVS) have achieved great success in this field, detecting dim targets in complex backgrounds remains a challenge due to the multi-scale framework and over-simplified disparity calculations. In this paper, infrared small targets are detected with a novel local contrast measurement named double-layer patch-based contrast (DLPC). Firstly, we crafted an elaborated double-layer local contrast measure, to suppress the background, which can accurately measure the gray difference between the target and its surrounding complex background. Secondly, we calculated the absolute value of the grayscale difference between the target and the background in the diagonal directions as a weighting factor to further enhance the target. Then, an adaptive threshold on the DLPC was employed to extract the target from the IR image. The proposed method can detect small targets effectively with a fixed-scaled mask template while being computationally efficient. Experimental results in terms of background suppression factor (BSF), signal-to-clutter ratio gain (SCRG) and receiver operating characteristic (ROC) curve on five IR image datasets demonstrated that the proposed method has better detection performance compared to six state-of-the-art methods and is more robust in addressing complex backgrounds.

The emissivity of typical SiC-based thermal protection materials was measured in-situ at a wide temperature range (800 ~ 2300°C) inside a plasma wind tunnel that was capable of simulating hypersonic environments on-ground. Based on it, the evolution mechanism dominated by dynamic oxidation was discussed. The results suggest an emissivity of Cf/SiC 0.84 ~ 0.88 at 858 ~ 1502°C, prior to “temperature jump”. If “temperature jump” emerged, the emissivity was decreased rapidly to ≈0.76. The emissivity drop was explained by the microstructural transition of the oxidized surfaces that were triggered by the dissipation of SiO2 oxide scale at 1600 ~ 1900°C. Similar emissivity evolution was observed in SiCf/SiC after “temperature jump”. The effect of temperature on the emissivity of ZrB2-SiC was more pronounced. It was increased from ≈0.73 to ≈0.98 at 1009 ~ 1297°C, and was plateaued at 1298 ~ 1497°C, ≈0.98. This was a consequence of the formation of higher percentage SiO2-rich layers. However, due to the dissipation of SiO2 and B2O3, the emissivity of ZrB2-SiC was declined at higher temperatures, from ≈0.98 (≈1497°C) to ≈0.85 (≈1768°C).

The TEA domain (TEAD) family proteins (TEAD1‒4) are essential transcription factors that control cell differentiation and organ size in the Hippo pathway. Although the sequences and structures of TEAD family proteins are highly conserved, each TEAD isoform has unique physiological and pathological functions. Therefore, the development and discovery of subtype selective inhibitors for TEAD protein will provide important chemical probes for the TEAD-related function studies in development and diseases. Here, we identified a novel TEAD1/3 covalent inhibitor (DC-TEADin1072) with biochemical IC50 values of 0.61 ± 0.02 and 0.58 ± 0.12 μmol/L against TEAD1 and TEAD3, respectively. Further chemical optimization based on DC-TEAD in 1072 yielded a selective TEAD3 inhibitor DC-TEAD3in03 with the IC50 value of 0.16 ± 0.03 μmol/L, which shows 100-fold selectivity over other TEAD isoforms in activity-based protein profiling (ABPP) assays. In cells, DC-TEAD3in03 showed selective inhibitory effect on TEAD3 in GAL4-TEAD (1–4) reporter assays with the IC50 value of 1.15 μmol/L. When administered to zebrafish juveniles, experiments showed that DC-TEAD3in03 reduced the growth rate of zebrafish caudal fins, indicating the importance of TEAD3 activity in controlling proportional growth of vertebrate appendages.

Zhimin Hou, Yanhui Liu et al. used single cell RNA-seq to analyze the model organism, Arabidopsis thaliana, at three stages during female germline differentiation. They reconstructed the continuous trajectory of female germline differentiation, providing a valuable reference for future investigation of germline cell fate determination in A. thaliana.

Berberine ursodeoxycholate has been studied for its serum lipid and lipoprotein lowering effects. Here the authors report an 18-week phase 2, randomised, double-blind, placebo-controlled clinical trial that tested the effect of berberine ursodeoxycholate in patients with fatty liver disease and diabetes, and showed that the group taking the higher dose of the drug had reduced liver fat content.

Abstract Background One of the most cost-effective treatments for secondary prevention of stroke and other non-communicable diseases is a long-term medication regimen. However, the complexities of medication adherence extend far beyond individual behavior change, particularly in low- and middle-income countries. The purpose of this study was to examine stakeholder perspectives on barriers to medication adherence for stroke patients in Beijing, China, identifying opportunities to improve care and policy in resource-constrained settings. Methods We conducted a qualitative, phenomenological analysis of data obtained from 36 individuals. Participants were patients; caregivers; healthcare providers; and representatives from industry and government, purposively selected to synthesize multiple perspectives on medication management and adherence for stroke secondary prevention in Beijing, China. Data was analyzed by thematic analysis across iterative coding cycles. Results Four major themes characterized barriers on medication adherence, across stakeholders and geographies: limitations driven by individual patient knowledge / attitudes; lack of patient-provider interaction time; lack of coordination across the stratified health system; and lack of affordability driven by high overall costs and limited insurance policies. Conclusions These barriers to medication management and adherence suggest opportunities for policy reform and local practice changes, particularly for multi-tiered health systems. Findings from this study in Beijing, China could be explored for applicability in other low- and middle-income countries with urban centers serving large geographic regions.

Background Reperfusion therapy is the most effective treatment for acute ischaemic stroke (AIS) but remains underutilised in China. There is an urgent need to develop tailored strategies to increase adherence to intravenous thrombolysis (IVT) and endovascular thrombectomy (EVT) within the guideline-recommended time window for eligible patients.Aims This study aims to investigate the efficacy of a comprehensive quality improvement intervention on adherence to guideline-recommended reperfusion therapy for patients with AIS in China.Design The Improve Acute Reperfusion Treatment Quality for Stroke in China (IMPROVE Stroke Care in China) trial is designed as a stepped wedge cluster randomised trial within 51 hospitals. We developed the comprehensive intervention ‘STEP’ (Strategies, Toolkit, Exploration, Paradigm) to promote the reconstruction of workflow in stroke centres and shorten in-hospital delay of reperfusion treatment for patients with AIS. The participating hospitals (clusters) were randomised to three groups (cohorts) for different predefined steps to intervention implementation. The primary outcome was the adherent rate of IVT or EVT for eligible patients within the time window. The sample size was estimated to be 7644, and was determined by the number of cases to be enrolled in five study periods to detect a relative increase of 30% (from 19% to 25%) with 90% power and intraclass correlation coefficient of 0.03. All efficacy analyses will be conducted based on the intention-to-treat principle. The primary outcome will be analysed using a mixed-effects logistic regression with a random effect for the cluster (hospital), and a fixed effect for the strategy and period.Conclusions If the efficacy is well established, this targeted comprehensive intervention STEP will inform national strategies to increase adherence to guideline-recommended performance on reperfusion therapy.Trial registration number clinicaltrials.gov Identifier: NCT003578107

The IL-33/ST2 axis is reported to be controversially associated with coronary artery disease (CAD). A systematic review of the association between the IL-33/ST2 axis and CAD revealed that IL-33/ST2 plays a protective role in CAD and serum sST2 and IL-33 levels are increased in patients with cardiovascular disease. Therefore, the association of IL-33/ST2 single nucleotide polymorphisms (SNPs) with CAD prevalence, prognosis, and risk factors was assessed by performing a meta-analysis. Through a literature search of relevant articles in various databases using the relevant keywords, seven studies were included in the analysis. The meta-analysis showed that the IL-33/ST2 axis was associated with increased CAD risk [pooled odds ratio (OR) = 1.17, 95% confidence interval (CI): 1.13–1.20]. Gene subgroup analysis showed a close association of IL1RL1 (OR = 1.25, 95% CI: 1.20–1.30; I2 = 85.9%; p = 0.000) and IL1RAcP (OR = 1.42, 95% CI: 1.26–1.60; I2 = 27.1%; p = 0.203) with increased CAD risk. However, the association for the IL-33 gene was not statistically significant. SNPs rs7044343 (T), rs10435816 (G), rs11792633 (C) in IL-33 gene were associated with a protective effect in CAD. However, rs7025417 (T) in IL-33, rs11685424 (G) in IL1RL1, rs950880 (A) in sST2, and rs4624606 (A) in IL1RAcP were related to increased CAD risk. Overall, polymorphisms in IL-33/ST2 axis components were associated with increased CAD risk. These results may help identify key features of IL-33/ST2 immunobiology in CAD along with potential treatment strategies to lower disease burden.

Abstract Background Little is known about the distribution of the intracranial arteries that are responsible for noncardiogenic posterior circulation stroke (PCS) in the Chinese population. Furthermore, few studies have compared the imaging manifestations and outcomes across PCS due to the disease of different intracranial arteries. Therefore, our aim was to demonstrate the distribution of the intracranial arteries that were responsible for noncardiogenic PCS and to compare the imaging manifestations and outcome across PCS due to the disease of different intracranial arteries. Methods We prospectively enrolled 690 patients from 22 Chinese centers with noncardiogenic PCS due to intracranial artery disease. Intracranial artery disease was classified as intracranial vertebral artery disease (IVAD) and intracranial nonvertebral artery disease (INVAD). Clinical‐radiologic patterns and outcomes were compared between IVAD and INVAD. Results INVAD was more frequent than IVAD. Compared to the INVAD group, the IVAD group more frequently had hypertension, coronary heart disease, multiple infarcts, border zone infarcts, territorial infarcts, small cortical infarcts, multilevel infarcts, multisector infarcts, and more often had severe stenosis of the responsible artery, more often presented with decreased alertness, gaze palsy, bilateral limb weakness, ataxia, dysphagia, diplopia, vertigo, and headache. In addition, the IVAD group had a higher National Institutes of Health stroke scale score and modified Rankin Scale score at discharge and more deaths and recurrent ischemic cerebrovascular disease within 1 year of onset. Multivariable Cox regression identified IVAD as an independent predictor of recurrent ischemic cerebrovascular disease. Conclusions PCS due to INVAD was more common in the Chinese population. However, PCS due to IVAD had more serious clinical‐radiologic patterns and worse outcomes.

Summary: Background: After multiple lines of therapies, no guideline or consensus is currently available for the treatment of patients with metastatic breast cancer. This study aims to evaluate the efficacy of a novel re-subtyping and treatment strategy based on ctDNA alterations. Methods: This observational, multicentre study recruited 223 patients with metastatic breast cancer intending to receive late-line therapy from Dec 1, 2016, to June 31, 2019. This study took place in Hunan Cancer Hospital, the Forth Hospital of Changsha and Zhuzhou Central Hospital in China. ctDNA alterations were assessed by next-generation sequencing (NGS). Patients with druggable ctDNA alterations were treated with corresponding targeted drugs which are clinically available. Other patients received physician-chosen treatment. To investigate the influence of molecular weight (Mw) on the anti-inflammatory activity of β-D-glucan from Ganoderma lucidum, ultrasonic irradiation was applied to treat the β-D-glucan (GLP, 2.42 × 106 g/mol) solution to obtain two degraded fractions with molecular weight of 6.53 × 105 g/mol (GLPC) and 3.49 × 104 g/mol (GLPN). Structural analysis proved that the degraded fractions possessed similar repeated units with the original β-D-glucan. The in vitro anti-inflammatory activity studies showed that all fractions could significantly inhibit LPS-induced expression of cytokines including TNF-α, IL-8, MIF and MCP-1 in Caco-2 cells at certain concentrations. Moreover, GLPC and GLPN exhibited better anti-inflammatory activity than GLPC. The intestinal anti-inflammatory activity evaluated by dextran sulfate sodium (DSS)—induced colitis mice model showed that intragastric administration of GLPN (lower Mw fraction) could significantly recover inflamed tissues of mice. Compared with GLP and GLPC, GLPN exhibited stronger ability to inhibit the secretion of pro-inflammatory cytokines (TNF-α, IL-1β, and IL-6). The results revealed that Mw of β-D-glucan influenced its anti-inflammatory activity and decreasing of Mw would improve the activity, which provided evidence for the potential use of β-D-glucan from G. lucidum as anti-colitis ingredients.

ObjectiveIn the context of Healthy China, the effect of external motivation on sports anomie behavior from the perspective of gender among Chinese college students was investigated.ParticipantsIn total, 2,340 college students were involved in this study.MethodsThe self-made scales were used, which were about anomie behavior and external motivation in sports. The independent sample T-test was used to compare the sports anomie behavior between male and female students. Then, multiple linear regression analysis was adopted to examine the effects of external motivation on sports anomie behavior.ResultsGirls’ sports anomie behavior was lower than boys. There were significant gender differences in honor motivation, obedience motivation, and economic motivation; girls’ motivations were lower than boys. The academic motivation was negatively correlated with the sports anomie behavior of college students of both genders. The economic motivation was positively correlated with their sports anomie behavior. The honor motivation was negatively correlated with the sports anomie behavior of female college students.ConclusionFemale college students’ sports anomie behavior and external motivation are lower than that of male college students. External motivation had a significant influence on college students’ sports anomie behavior.

BackgroundDifferentiating multiple pulmonary lesions as multiple primary lung cancer (MLC) or intra-pulmonary metastasis (IPM) is critical. Lung cancer also has a high genetic heterogeneity, which influenced the treatment strategy. Genetic information may aid in tracing lineage information on multiple lung lesions. This study applied comprehensive genomic profiling to decipher the intrinsic genetics of multiple lung lesions.MethodsSixty-six lung adenocarcinomas (LUAD) tumor lesions (FFEP) archived from 30 patients were included in this study. The 508 cancer-related genes were evaluated by targeted next-generation sequencing (MGI-seq 2000).ResultsThe study included a total of 30 LUADs (66 samples). The majority of tumors demonstrated intra-tumoral heterogeneity. Two hundred twenty-four mutations were detected by sequencing the 66 samples. We investigated the driver gene mutations of NSCLC patients with multiple lesions. EGFR was the most frequently (48/198) mutated driver gene. The codons in EGFR mainly affected by mutations were p.L858R (18/66 [27.3%]) and exon 19del (8/66 [12.1%]). In addition, additional driver genes were found, including TP53, BRAF, ERBB2, MET, and PIK3CA. We also found that the inter-component heterogeneity of different lesions and more than two different mutation types of EGFR were detected in seven patients with two lesions (P3, P10, P24, P25, P28, P29, and P30). The TMB values of different lesions in each patient were different in 26 patients (except P4, P5, P14, and P30).ConclusionsComprehensive genomic profiling should be applied to distinguishing the nature of multiple lung lesions irrespective of radiologic and histologic diagnoses.

Abstract Colorectal cancer (CRC) is a frequently occurring digestive system cancer and postoperative tumor metastasis and recurrence are the main reasons for the failure of CRC treatment. The aim of this study was to identifying and validating key genes associated with metastatic recurrence of CRC. RNA expression of three datasets (GSE17538, GSE32323, and GSE29623) was used for biomarker discovery. We identified integrin‐binding sialoprotein (IBSP) as a candidate biomarker which was validated in three clinical cohorts (GSE41258, GSE21510, and GSE39582) and our clinical specimens. The results suggested that IBSP expression significantly increased at mRNA and protein levels among CRC cases, which was associated with metastatic recurrence, metastasis, high risk of recurrence, and poor survival in CRC. Consistent results were obtained in CRC cells. The relative level of serum IBSP evidently increased among CRC patients relative to normal controls, and downregulated after operation. As suggested by gene set enrichment analysis (GSEA), the IBSP level was associated with cell‐matrix adhesion in CRC. Functional experiments in vitro showed that IBSP promoted the growth and aggressiveness of CRC, and the potential mechanism by which IBSP promoted carcinogenesis of CRC was the abnormal activation of Fyn/β‐catenin signaling pathway. To sum up, findings in the present work indicate that IBSP can serve as the candidate biomarker for the diagnosis, treatment, and prognosis of CRC.

Abstract To study the pathogenicity of new duck reovirus (NDRV) to chickens, eighty 3-day-old SPF chickens were equally divided into two groups. The experimental group was inoculated with a NDRV challenge strain of 100 μL (10–5.00 ELD50/0.1 mL) by the subcutaneous (s.c.) route, and the control group was inoculated with 100 μL of sterile phosphate-buffered saline (PBS) by the same route. In the experimental group, chickens exhibited introflexion of claws, performing of splits, stunting syndrome, weight loss and death. Gross lesions such as enlargement and yellowish-white focal necroses were observed in the liver and spleen. Microscopic changes were typical including varying degrees of hepatocyte steatosis and necrosis, splenic lymphocyte necrosis, interstitial pneumonia. Viral loads were detected in lung, liver, heart, spleen, duodenum, burse and kidney. The liver and spleen viral loads remained a much higher level and maintained for a longer time, suggesting that these tissues might be the target organs. In summary, NDRV can cause systemic infections and death in chickens, which indicated that chickens may be infected by NDRV in poultry production.

Cloud radar is an important tool to observe and retrieve the microphysical and dynamic profiles in cloud and precipitation systems. The paper reviewed the cloud radar work modes, data quality control and merging algorithms, retrieval methods based on the relationship between reflectivity and raindrop terminal fall velocity, tracers of clear-air motion algorithm with single wavelength cloud radar, reflectivity density data ratio between dual wavelength cloud radar for vertical air motions and raindrop size distributions. The special characteristics of these methods were discussed. Coherent integration, incoherent integration and pulse compression techniques are used to improve radar sensitivity. This paper provided references for cloud radar observation and data analysis.

In this paper, the observation data measured by the dual-frequency precipitation radar DPR on board the GPM satellite and the ground-based Ka/Ku dual-band radar CR of the Guangdong Longmen Observatory from April to September 2019 are matched in time and space. Combining with the raindrop spectrum data of the micro rain radar MRR for the CR attenuation correction, we compare the difference of echo intensity detected by DPR and CR, and analyze the systematic differences between the two. The main result are as follows:(1) The distributions of precipitation echoes detected by DPR and CR have similar trends with height. The height of the brightband is basically the same. The echo intensity detected by DPR at the same altitude is significantly greater than that of CR.(2) For non-precipitating clouds, the systematic difference between Ku band and Ka band in CR is about 3.1 dBz.(3) The stronger the rainfall intensity, the greater the thickness of the water film of the CR's radome, and the greater the attenuation of the CR echo.(4) The raindrop spectrum data are used to calculate the attenuation coefficient in the process of bin by bin library correction, and the correction amount of the reflectivity factor of CR in the Ka and Ku bands does not exceed 4 dBz.(5) Integrating all the collocated data, the average systematic difference between the reflectivity factors of CR and DPR after the attenuation correction for the Ka band is 15.7 dBz, and that for the Ku band is 14.4 dBz. Although the systematic difference between the two radars after attenuation correction is still large, these results have significantly improved the data quality of the ground-based Ka/Ku dual-band radar.

YAP is known to play a role both in organ size via the Hippo signalling pathway and in inflammation, though the precise mechanism are unclear. Here, the authors report that YAP promotes activation of the NLRP3 inflammosome through binding interactions.

In recent years, tragedies caused by forest fires have been frequently reported. Forest fires not only result in significant economic losses but also cause environmental damage. The utilization of computer vision techniques and unmanned aerial vehicles (UAVs) for forest fire monitoring has become a primary approach to accurately locate and extinguish fires during their early stages. However, traditional computer-based methods for UAV forest fire image segmentation require a large amount of pixel-level labeled data to train the networks, which can be time-consuming and costly to acquire. To address this challenge, we propose a novel weakly supervised approach for semantic segmentation of fire images in this study. Our method utilizes self-supervised attention foreground-aware pooling (SAP) and context-aware loss (CAL) to generate high-quality pseudo-labels, serving as substitutes for manual annotation. SAP collaborates with bounding box and class activation mapping (CAM) to generate a background attention map, which aids in the generation of accurate pseudo-labels. CAL further improves the quality of the pseudo-labels by incorporating contextual information related to the target objects, effectively reducing environmental noise. We conducted experiments on two publicly available UAV forest fire datasets: the Corsican dataset and the Flame dataset. Our proposed method achieved impressive results, with IoU values of 81.23% and 76.43% for the Corsican dataset and the Flame dataset, respectively. These results significantly outperform the latest weakly supervised semantic segmentation (WSSS) networks on forest fire datasets.

Mêdog and Nagqu are two typical climate regions of the Tibetan Plateau, with different atmospheric conditions and local orography. This may lead to different diurnal variation patterns of clouds and precipitation. This paper investigates the diurnal variations of clouds and precipitation in Mêdog and Nagqu, using ground-based measurements from Ka-band cloud radar and a Particle Size and Velocity (PARSIVEL) disdrometer. High frequencies of cloud cover and precipitation occur from 23:00 local solar time (LST) to 05:00 LST in Mêdog, while low frequencies appear from 11:00 LST to 17:00 LST. The occurrence frequencies in Nagqu maintain high values from 13:00 LST to 21:00 LST. In terms of mean rain rate, heavier rainfall appears in the evening and at night in Mêdog, with peaks at 00:00 LST and 18:00 LST, respectively. In Nagqu, the heaviest rainfall occurs at 12:00 LST. In addition, the afternoon convective rainfall in Nagqu is characterized by a much higher concentration of large drops, which can be classified as continental-like. The morning rainfall has the lowest concentration of large drops and can be classified as maritime-like. Finally, the mechanisms of diurnal variations in the two regions are discussed. The diurnal cycle of clouds and precipitation in Mêdog may be associated with the nocturnal convergence of moisture flux and mountain–valley wind circulation. Diurnal variations in Nagqu have a high correlation with the diurnal cycle of solar radiation. The high nocturnal frequency of clouds and precipitation in the two regions at night is closely related to the convergence of moisture flux.

Objective High-performance liquid chromatography inductively coupled plasma mass spectrometry(HPLC-ICP/MS) was used to analyze five arsenic species in oyster sauce seasonings, including arsenobetaine(AsB), arsenite As(Ⅲ), dimethyl arsenic(DMA), monomethyl arsenic(MMA) and arsenate As(V). Methods Oyster sauce samples were extracted with 1% nitric acid at 90 ℃ for 3 h, the extract was centrifuged at 8 000 r/min for 10 min, and the supernatant was filtered through 0.22 μm water filtration membrane. A Hamilton PRP-X100 anion exchange column(250 mm×4.1 mm, 10 μm) was used with 10 and 20 mmol/L diammonium hydrogen phosphate methanol solution(containing 2% methanol) as mobile phase for gradient elution. Results The linear range of five arsenic species were good in the range of 0.5-100 μg/L, and the correlation coefficient(r) was above 0.999. The detection limits were 0.1, 0.1, 0.1, 0.2 and 0.2 μg/L, respectively. The recoveries were 83.2%-103.4%, and the relative standard deviations(RSD) were less than 5%. Conclusion The method is simple with a wide linear range and high sensitivity. It is suitable for the determination of oyster sauce samples.

Purpose – Legislation plays a vital role in solving carbon emissions reduction and climate change issues. China began to implement a low-carbon economic policy in 2010, but the effect of the policy needs to be evaluated. Accordingly, this paper aims to discuss China’s low-carbon policy through exploring the following two questions, namely, whether the policy effect reaches the expected goal and whether the policy effects will balance economic development and emission reduction. Then, the paper puts forward suggestions for the improvement of China’s low-carbon policy. Design/methodology/approach – This paper is organized around three distinct aspects of policy effect evaluation. This paper uses the synthetic control method to construct a policy effect evaluation model and conducts a quasi-natural experiment. The paper selects annual panel data from 2003 to 2015, which is selected from 33 provinces. A comparative analysis of carbon dioxide emissions, energy consumption and economic development between Hubei Province and Liaoning Province. Findings – The results reveal that the implementation of the low-carbon pilot province policy in 2010 has a significant impact on the emission reduction effect of Liaoning Province, but the impact on the emission reduction effect of Hubei Province is not significant. The carbon emission trading system implemented in 2012 has reduced the emission reductions in Hubei Province and Liaoning Province has achieved better emission reduction effects after the implementation of this policy. After the implementation of the policy, the economic development of Hubei Province has been improved, but it has not brought help to the economic development of Liaoning Province. These findings provide new insights into the use of an emissions trading system for improving economic development and ultimately facilitate the attainment of the broader goal of sustainability. Originality/value – This paper proposes an innovative policy effect evaluation method by considering the status of unit gross domestic product, fixed asset investment in the energy industry, energy consumption, emission reduction technology innovation and other evaluation indicators. This paper contributes to broadening current methods of policy effect evaluation in China.

To improve the dispersity and antioxidant properties of curcumin, curcumin emulsions covered with cellulose particles (CP) with different structures were successfully prepared, and the structural characteristics, stability, and antioxidant properties of emulsions were investigated. The results showed that the CP obtained by increasing the hydrolysis time had smaller particle size, better water dispersion, and interfacial adsorption capacity. The encapsulation efficiency of curcumin in emulsion stabilized by cellulose particle hydrolyzed for 10 h can reach about 80%. After 9 days, all emulsions showed good stability, and no obvious creamed layer was observed. Compared to cellulose particles hydrolyzed for 2 h (CP2), emulsions stabilized by cellulose particles hydrolyzed for 6 h (CP6) and 10 h (CP10) exhibited better stability and free fatty acid (FFA) release. Meanwhile, the DPPH scavenging activity of curcumin emulsion stabilized by CP significantly increased with increasing the hydrolysis time and was much higher than that of pure emulsion and curcumin/water due to the higher solubility (1,455 times compared with curcumin/water solution) of curcumin, and these results could provide useful data for the stability and encapsulation of curcumin.

Stress granules (SGs) are dynamic cytoplasmic protein-RNA structures that form in response to various stress conditions, including viral infection. Porcine epidemic diarrhea virus (PEDV) variant-related diarrhea has caused devastating economic losses to the swine industry worldwide. In this study, we found that the percentage of PEDV-infected cells containing SGs is nearly 20%; meanwhile, PEDV-infected cells were resistant to sodium arsenite (SA)-induced SGs formation, as demonstrated by the recruitment of SGs marker proteins, including G3BP1 and TIA1. Moreover, the formation of SGs induced by SA treatment was suppressed by PEDV papain-like protease confirmed by confocal microscopy. Further study showed that PEDV infection disrupted SGs formation by downregulating G3BP1 expression. Additionally, PEDV replication was significantly enhanced when SGs' assembly was impaired by silencing G3BP1. Taken together, our findings attempt to illuminate the specific interaction mechanism between SGs and PEDV, which will help us to elucidate the pathogenesis of PEDV infection in the near future.

Introduction: The angiotensin-converting enzyme 2 (ACE2) as well as the transmembrane protease serine type 2 (TMPRSS2) have been found to play roles in cell entry for severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), the virus causing Coronavirus disease 2019 (COVID-19). SARS-CoV-2 infection risk and severity of COVID-19 might be indicated by the expression of ACE2 and TMPRSS2 in the lung. Methods: A high salt diet rat model and RAAS blockade were used to test whether these factors affect ACE2 and TMPRSS2 expression of the lung. A normal (0.3% NaCl), a medium (2% NaCl), or a high (8% NaCl) salt diet was fed to rats for 12 weeks, along with enalapril or telmisartan, before examining the lung for histopathological alteration. Using immunofluorescence and qRT-PCR, the localization as well as mRNA expression of ACE2 and TMPRSS2 were investigated. Results: The findings provide evidence that both TMPRSS2 and ACE2 are highly expressed in bronchial epithelial cells as well as ACE2 was also expressed in alveolar type2 (AT2) cells. High salt diet exposure in rats leads to elevated ACE2 expression on protein level. Treatment with RAAS blockers had no effect on lung tissue expression of ACE2 and TMPRSS2. Conclusions: These findings offer biological support regarding the safety of these drugs that are often prescribed to COVID-19 patients with cardiovascular co-morbidity. High salt intake on the other hand might adversely affect COVID-19 outcome. Our preclinical data should stimulate clinical studies addressing this point of concern.

Rice blast caused by Magnaporthe oryzae is one of the most destructive plant diseases. The secondary metabolites of Streptomyces have potential as biological control agents against M. oryzae. However, no commercial secondary antimicrobial products of Streptomyces have been found by gene prediction, and, particularly relevant for this study, a biocontrol agent obtained from Streptomyces bikiniensis has yet to be found. In this research, genomic analysis was used to predict the secondary metabolites of Streptomyces, and the ability to develop biocontrol pharmaceuticals rapidly was demonstrated. The complete genome of the S. bikiniensis HD-087 strain was sequenced and revealed a number of key functional gene clusters that contribute to the biosynthesis of active secondary metabolites. The crude extract of lipopeptides (CEL) predicted by NRPS gene clusters was extracted from the fermentation liquid of S. bikiniensis HD-087 by acid precipitation followed by methanol extraction, and surfactins, iturins, and fengycins were identified by liquid chromatography-mass spectrometry (LC–MS). In vitro, the CEL of this strain inhibited spore germination and appressorial formation of M. oryzae by destroying membrane integrity and through the leakage of cellular components. In vivo, this CEL reduced the disease index of rice blast by approximately 76.9% on detached leaves, whereas its control effect on leaf blast during pot experiments was approximately 60%. Thus, the S. bikiniensis CEL appears to be a highly suitable alternative to synthetic chemical fungicides for controlling M. oryzae.

Highly virulent Klebsiella pneumoniae often causes invasive infections with high morbidity and mortality rates, posing an immense clinical challenge. Rapid and accurate detection of pathogenic bacteria is of great significance for treatment and preventive control. Conventional detection by polymerase chain reaction (PCR) is limited by a dependence on laboratory equipment and professional staff. Recombinase polymerase amplification (RPA) combined with a lateral flow strip (LFS) can rapidly amplify and visualize target genes in a short period of time. The aim of this study was to develop an RPA-LFS technique for detection of the K. pneumoniae virulence gene rmpA2. Primers were designed against conserved sequences specific to the virulence gene, and primer probe design was optimized by introducing base substitution to obtain a specific and sensitive primer-probe combination for clinical detection. We tested 65 actual samples collected from clinics to evaluate the performance of the newly established RPA-LFS system in comparison with conventional PCR methods and qPCR methods. The RPA-LFS assay was performed at for 25 min a constant temperature of 37°C, and results could be observed without instrumentation. The system could specifically identify highly virulent K. pneumoniae carrying the virulence gene rmpA2 with a minimum detection limit of 10−1 ng/μL and 10 copies/μL. For the 65 clinical samples tested, The RPA-LFS assay results were in complete agreement with the qPCR results and PCR results. The RPA-LFS assay provides a rapid, accurate, and simple method for identification of highly virulent K. pneumoniae carrying rmpA2.

Vitellogenin (VTG) plays a very important role in the development of oocytes. This study aims to explore the effect of swimming on the VTG synthesis of conger eel (Conger myriaster). Circulating water flow and hormone injection regimen were two factors set in the following trials: A1 (no injection, no water flow), A2 (no injection, water flow), B1 (injection, no water flow), and B2 (injection, water flow). The flow velocity of the flowing water was 0.4 m/s (12 h, 60 days). We examined VTG, estrogen receptor (Er) gene expression, and VTG content in the liver and serum on the 30th and 60th days. VTG gene expression in A1 and A2 gradually decreased and was not significantly different between the two groups. The expression of Er gene in A1 was significantly higher than that in A2. The expression of VTG gene in group B1 was significantly higher than that in group B2, whereas the expression level of Er gene in group B1 was slightly higher than that in group B1. The VTG content in the liver in group B1 reached 1,396.93 μg/L, which was significantly higher than that in group B2 (1,302.06 μg/L). Results showed that the flowing water factor can inhibit the expression of VTG and Er genes in the liver and reduce the synthesis of VTG in the liver. Stimulation of flowing water can inhibit the yolk accumulation during the ovarian development of conger eel.

This paper deals with the problem of large number of parameters and complex calculation in video abstract generation of Fully Connected Network and Convolutional Neural Network. At the same time, the training and testing of such model need a lot of time and computer resources. We came up with a deep learning network parameter compression method based on Singular Value Decomposition(SVD) and Trucker Decomposition (TD) is proposed to generate the video summaries. The experiment was compared with other methods on TVSum and SumMe dataset, and the F1 value was 55.3% in TVSum dataset and 46.8% in SumMe dataset. At the same time, the degree of test time shortening under the same data volume is taken as the evaluation basis. The experimental results show that the proposed method achieves 1.04 times of acceleration in the SVD forward calculation, and 1.29 times of acceleration in the TD forward calculation. In a conclusion, the neural network model based on low-rank decomposition can effectively save computer resources and the time consumed by running programs.

Abstract Background Reduction in elevated serum cholesterol concentrations is important in the management of individuals at risk of atherosclerotic cardiovascular disease (ASCVD), such as myocardial infarction and thrombotic stroke. Although HMGCoA reductase inhibitors (“statins”) are frequently used for this purpose, a significant proportion of patients remain at increased residual risk of ASCVD as they do not adequately address some of the associated co-morbidities such as diabetes and fatty liver disease. Methods A double-blind, randomized, placebo-controlled, dose ranging study was carried out that compared three doses of berberine ursodeoxycholate (BUDCA) to placebo in a cohort of subjects with a history of hypercholesterolemia and serum LDL cholesterol levels above 2.59 mmol/L (> 99.9 mg/dL). BUDCA was administered in two divided doses each day for 28 days. The primary endpoints of the study were safety and tolerability of this new compound, as well as its effect in lowering serum lipid and lipoprotein concentrations. Results A total of 50 subjects were enrolled into three dose cohorts in this study. BUDCA was generally well tolerated, even at doses of 2000 mg per day (the highest dose group); there were no significant adverse effects reported and this highest dose was associated with significant reductions in LDL cholesterol. By day 28 and with the highest dose of BUDCA, there were significant reductions in the serum concentrations of total cholesterol by 8.2% (P = 0.0004) and LDL cholesterol by 10.4% (P = 0.0006), but no significant changes in triglyceride and HDL cholesterol concentrations. Conclusions BUDCA is a new single molecular entity that has a significant but modest effect in safely lowering serum LDL-cholesterol concentrations in individuals with a history of hypercholesterolemia. It has a potential use for treating hypercholesterolemia in individuals who cannot take statins, and possibly as adjunctive to other agents, such as ezetimibe or bempedoic acid.
