Abstract

Heat shock protein family A member 5 (HSPA5) plays a pivotal role in the endoplasmic reticulum (ER) stress response and unfolded protein response (UPR), both of which are crucial for protein folding, assembly, and quality control within cells. In response to ER stress, HSPA5 becomes overexpressed to preserve cellular homeostasis. A previous study revealed a robust association between HSPA5 expression and various cancers. However, the prognostic function of HSPA5 and its involvement in tumor formation remain largely unknown. In this study, we integrated HSPA5 expression data from databases such as the Clinical Proteomic Tumor Analysis Consortium (CPTAC) and The Cancer Genome Atlas (TCGA) to conduct a comprehensive pan-cancer analysis of HSPA5. Our findings revealed that HSPA5 is overexpressed in various tumor types and is significantly associated with poor prognosis. Additionally, HSPA5 expression is significantly correlated with immune checkpoints, stromal infiltration, and consequent alterations in the immune landscape. Verification was conducted on samples from patients with various tumor types, including breast and liver cancers. Additionally, we also performed verification in vitro. In conclusion, HSPA5 may offer a potential target for cancer treatment.

Abstract

A classic inferential problem in statistics is the two-sample hypothesis test, where we test whether two samples of observations are either drawn from the same distribution or two distinct distributions. However, standard methods for performing this test require strong distributional assumptions on the two samples of data. We propose a semi-Bayesian nonparametric (semi-BNP) procedure for the two-sample hypothesis testing problem. First, we will derive a novel BNP maximum mean discrepancy (MMD) measure-based hypothesis test. Next, we will show that our proposed test will outperform frequentist MMD-based methods by yielding a smaller false rejection and acceptance rate of the null. Finally, we will show that we can embed our proposed hypothesis testing procedure within a generative adversarial network (GAN) framework as an application of our method. Using our novel BNP hypothesis test, this new GAN approach can help to mitigate the lack of diversity in the generated samples and produce a more accurate inferential algorithm compared to traditional techniques.

Abstract

The protein expression levels of NSD2, MDM2, P-p53, Bcl-2, Bax, Caspase 3, Cleaved Caspase 3, and Caspase 8 were detected in 6b treated HepG2 cells. To further promote the apoptosis of HepG2 cells after 6b administration, we used flow cytometry to detect its apoptosis. ACTB is abnormally expressed in a number of malignancies, changing the cytoskeleton and influencing tumor formation. To further test this speculation, we assessed the major structural changes in the HepG2 cell line after treatment with compound 6b by staining actin filaments and nuclei with phalloidin/FITC and DAPI, respectively. Qualitative results from treatment with 5, 10, and 20 μM showed that compound 6b promoted nuclear and cytoplasmic changes.

Abstract

It has become an increasingly common practice in modern science and engineering to collect samples of multiple network data in which a network serves as a basic data object. The increasing prevalence of multiple network data calls for developments of models and theories that can deal with inference problems for populations of networks. In this work, we propose a general procedure for hypothesis testing of networks and in particular, for differentiating distributions of two samples of networks. We consider a very general framework which allows us to perform test on large and sparse networks. Our contribution is two-fold: (1) We propose a test statistics based on the singular value of a generalized Wigner matrix. The asymptotic null distribution of the statistics is shown to follow the Tracy–Widom distribution as the number of nodes tends to infinity. The test also yields asymptotic power guarantee with the power tending to one under the alternative; (2) The test procedure is adapted for change-point detection in dynamic networks which is proven to be consistent in detecting the change-points. In addition to theoretical guarantees, another appealing feature of this adapted procedure is that it provides a principled and simple method for selecting the threshold that is also allowed to vary with time.
