Aims Prescription of weight loss to individuals is often characterized by weight fluctuations. However, current body weight management metrics may have difficulty characterizing the changes in body weight over time. We aims to characterize the long-term changes using body weight time in target range (TTR) and test its independent association with cardiovascular outcomes. Methods We included 4,468 adults from the Look AHEAD (Action for Health in Diabetes) trial. Body weight TTR was defined as the percentage of time during which body weight was within the Look AHEAD weight loss goal range. The associations of body weight TTR with cardiovascular outcomes were analyzed using multivariable Cox modeling and restricted cubic spline function. Results Among the participants (mean age 58.9 years, 58.5% women, 66.5% White), there were 721 incident primary outcomes (cumulative incidence: 17.5%, 95% confidence interval [CI]: 16.3%-18.8%) during a median of 9.5 years of follow-up. Each 1-SD (standard deviation) increase in body weight TTR was significantly associated with a decreased risk of the primary outcome (hazard ratio [HR]: 0.84, 95% CI: 0.75-0.94) after adjusting for mean and variability of body weight and traditional cardiovascular risk factors. Further analyses using restricted cubic spline indicated the inverse association between body weight TTR and primary outcome in a dose-dependent manner. Similar associations remained significant among the participants with a lower baseline or mean body weight. Conclusion In adults with overweight/obesity and type 2 diabetes, higher body weight TTR was independently associated with lower risks of cardiovascular adverse events in a dose-response manner.

Studying the mechanism of drought stress in potato leaves at tuber expansion period by exogenous uniconazole revealed new insights into potato stress physiology. Therefore, a completely random pot experiments with time gradients of 5, 10 and 15 d of drought treatment. were carried out with uniconazole (40 mg L− 1) with respect to photosynthesis, active oxygen metabolism, antioxidant enzyme activity and the ascorbate-glutathione (AsA-GSH) cycle on varieties kexin No 1 (drought-tolerant, KX 1) and Atlantic (drought-sensitive, Atlantic). The results showed that uniconazole can effectively alleviate the degradation of chlorophyll under drought stress, Compared with CK, drought significantly inhibited the Pn, Tr, Gs and Ci of the leaves of the two potato varieties. Exogenous uniconazole effectively alleviated the inhibition of drought on the photosynthetic parameters of potato leaves, compared with D treatment, uniconazole reduced malondialdehyde (MDA), the rate of superoxide radical (O2−) and hydrogen peroxide (H2O2) content in leaves of KX 1 and Atlantic, and increased the activity of antioxidant enzymes in antioxidant defense system to remove excessive accumulation of reactive oxygen species (ROS) and reduce membrane lipid peroxidation, effectively alleviated the loss of yield factors caused by drought stress, and the drought-tolerant variety KX 1 was less damaged by drought than the drought-sensitive variety Atlantic, in addition, under drought stress, the antioxidant enzyme activity and non-enzymatic antioxidant content of the two varieties increased to adapt to the stress response caused by drought stress, D + S treatment could further increase the contents of osmotic adjustment substances (soluble protein and proline) and AsA-GSH cycle products and substrates, including ascorbic acid (AsA), dehydroascorbate (DHA), glutathione (GSH) and oxidized glutathione (GSSG), and increase the ratio of AsA/DHA and GSH/GSSG under drought stress in two potato varieties, so as to improve the level of reducing power and antioxidant capacity in leaves, thus reducing the damage of reactive oxygen species to cell membrane. The drought-tolerant variety KX 1 recovered better than the drought-sensitive variety Atlantic. These results provided a reference for the mechanism of drought resistance in potato and the effect of uniconazole on alleviating drought stress.

3D action recognition is referred to as the classification of action sequences which consist of 3D skeleton joints. While many research work are devoted to 3D action recognition, it mainly suffers from three problems: highly complicated articulation, a great amount of noise, and a low implementation efficiency. To tackle all these problems, we propose a real-time 3D action recognition framework by integrating the locally aggregated kinematic-guided skeletonlet (LAKS) with a supervised hashing-by-analysis (SHA) model. We first define the skeletonlet as a few combinations of joint offsets grouped in terms of kinematic principle, and then represent an action sequence using LAKS, which consists of a denoising phase and a locally aggregating phase. The denoising phase detects the noisy action data and adjust it by replacing all the features within it with the features of the corresponding previous frame, while the locally aggregating phase sums the difference between an offset feature of the skeletonlet and its cluster center together over all the offset features of the sequence. Finally, the SHA model which combines sparse representation with a hashing model, aiming at promoting the recognition accuracy while maintaining a high efficiency. Disruption of the lung endothelial barrier is a hallmark of acute respiratory distress syndrome (ARDS), for which no effective pharmacologic treatments exist. Prior work has demonstrated that FTY720 S-phosphonate (Tys), an analog of sphingosine-1-phosphate (S1P) and FTY720, exhibits potent endothelial cell (EC) barrier protective properties. In this study, we investigated the in vitro and in vivo efficacy of Tys against methicillin-resistant Staphylococcus aureus (MRSA), a frequent bacterial cause of ARDS. Tys-protected human lung EC from barrier disruption induced by heat-killed MRSA (HK-MRSA) or staphylococcal α-toxin and attenuated MRSA-induced cytoskeletal changes associated with barrier disruption, including actin stress fiber formation and loss of peripheral VE-cadherin and cortactin. Tys-inhibited Rho and myosin light chain (MLC) activation after MRSA and blocked MRSA-induced NF-κB activation and release of the proinflammatory cytokines, IL-6 and IL-8. In vivo, intratracheal administration of live MRSA in mice caused significant vascular leakage and leukocyte infiltration into the alveolar space. Pre- or posttreatment with Tys attenuated MRSA-induced lung permeability and levels of alveolar neutrophils. Posttreatment with Tys significantly reduced levels of bronchoalveolar lavage (BAL) VCAM-1 and plasma IL-6 and KC induced by MRSA. Dynamic intravital imaging of mouse lungs demonstrated Tys attenuation of HK-MRSA-induced interstitial edema and neutrophil infiltration into lung tissue. Tys did not directly inhibit MRSA growth or viability in vitro. In conclusion, Tys inhibits lung EC barrier disruption and proinflammatory signaling induced by MRSA in vitro and attenuates acute lung injury induced by MRSA in vivo. These results support the potential utility of Tys as a novel ARDS therapeutic strategy.
