The period of Normalization in Czechoslovakia is often perceived as a grey “Eastern iceberg” where life stood still and uniformity governed. My analysis of sexological discourse, particularly of texts focused on perversity, juxtaposes the normalized ethos of the period with deviant sexual subjectivities. I analyze papers and debates presented at annual sexological conferences in the 1970s and 1980s. Sexuality, especially in its non-normal/deviant forms, was revealed as unstable, a quality sought to be “rectified” through gender which was perceived as binary. The family was interrogated as a source of deviance and also as a place of redress. While sexological writings in general tend to biologize sexuality, my analysis shows that sexologists attributed social genealogy to deviance, a finding that attests to rigid social conditions during Normalization.

The gut microbiota provides a wide range of beneficial functions for the host, and has an immense effect on the host's health status. The presence of microbiome in the gut may often influence the effect of an orally administered drug. Molecular mechanisms of this process are however mostly unclear. We investigated how the effect of a nonsteroidal drug nabumetone on expression of drug metabolizing enzymes (DMEs) in mice intestine and liver is changed by the presence of microbiota, here, using the germ free (GF) and specific pathogen free (SPF) BALB/c mice. First, we have found in a preliminary experiment that in the GF mice there is a tendency to increase bioavailability of the active form of nabumetone, which we have found now to be possibly influenced by differences in expression of DMEs in the GF and SPF mice. Indeed, we have observed that the expression of the most of selected cytochromes P450 (CYPs) was significantly changed in the small intestine of GF mice compared to the SPF ones. Moreover, orally administered nabumetone itself altered the expression of some CYPs and above all, in different ways in the GF and SPF mice. In the GF mice, the expression of the DMEs (CYP1A) responsible for the formation of active form of the drug are significantly increased in the small intestine and liver after nabumetone application. These results highlight the importance of gut microbiome in processes involved in drug metabolism in the both gastrointestinal tract and in the liver with possible clinical relevance. Preimplantation genetic diagnosis (PGD) is an established application of genetic testing in the context of in vitro fertilization. PGD is an alternative method to prenatal diagnosis which aims to prevent the transmission of an inherited disorder to the progeny by implanting only embryos that do not carry genetic predisposition for a particular disease. The aim of this study is to provide an overview of eye disorders for which PGD has been carried out.The European literature search focused on best practices, ethical issues, risks and results of PGD for inherited eye disorders.PGD is performed for a number of ocular disorders; a prerequisite for its application is however, the knowledge of a disease-causing mutation(s). The main advantage of this method is that the couple is not exposed to a decision of whether or not to undergo an abortion. Qualified counselling must be provided prior to the PGD in order to completely understand the risk of disability in any child conceived, consequences of disease manifestation, and advantages as well as limitations of this method. In the group of non-syndromic eye diseases and diseases in which ocular findings dominate, PGD has been performed in European countries for aniridia, choroideremia, congenital fibrosis of extraocular muscles, Leber congenital amaurosis, ocular albinism, retinitis pigmentosa, X-linked retinoschisis, Stargardt disease, blepharophimosis-ptosis-inverse epicanthus syndrome and retinoblastoma. Sexing for X-linked or mitochondrial diseases has been carried out for blue cone monochromatism, choroideremia, familial exudative vitreoretinopathy, Leber hereditary optic neuropathy, macular dystrophy (not further specified), Norrie disease, X-linked congenital stationary night blindness, X-linked retinoschisis and nystagmus (not further specified).In recent years, there has been an increase in potential to use PGD. The spectrum of diseases for this method has widened to include severe inherited eye diseases.Key words: preimplantation genetic diagnosis; monogenic eye diseases; in vitro fertilization.

Abstract The aim of this article was to evaluate the viability of maize protoplasts, cell wall regeneration, Cd uptake by protoplasts, and the impact of silicon under cadmium cations (Cd) stress in two maize hybrids with contrasting tolerances to Cd toxicity. The differences in protoplast viability between the sensitive (Novania) and tolerant (Almansa) hybrids were noticeable even at the beginning of culture. The percentage of living protoplasts in the presence of Cd was higher in the tolerant hybrid. In both hybrids, Si supplementation significantly increased the viability of protoplasts exposed to Cd. The percentage of protoplasts with regenerated cell walls gradually increased in both hybrids and by the end of the culture it had reached almost identical values. Differences were observed during the first four days, when a lag phase occurred in the protoplasts of the sensitive hybrid accompanied by a rapid decrease in protoplast viability in all the variants tested. The addition of Si increased the cell wall regeneration compared with the Cd variant in both hybrids. The Cd content was higher in the tolerant hybrid than in the sensitive one during the first four days and declined on the seventh day. This may be connected with the increasing intensity of cell wall formation from the fourth up to the seventh day. The addition of Si decreased the Cd uptake into protoplasts of both hybrids. Despite the higher content of Cd, the protoplasts of the tolerant hybrid showed higher viability, obviously indicating unequal mechanisms of Cd processing in studied hybrids. Capsule Protoplasts of two maize hybrids were tested for their viability, regeneration, Cd-uptake and the mitigation of cadmium stress by silicon. 119. Saving newborns, defining livebirth: The struggle to reduce infant mortality in East-Central Europe in comparative and transnational perspectives, 1945–1965

- Abstract
After World War II, infant mortality rates started dropping steeply. We show how this was accomplished in socialist countries in East-Central Europe. Focusing on the two postwar decades, we explore comparatively how medical experts in Poland, Hungary, Czechoslovakia, and East Germany saved fragile newborns. Based on an analysis of medical journals, we argue that the Soviet Union and its medical practices had only a marginal influence; the four countries followed the recommendations of the World Health Organization instead, despite not being members. Importantly, we analyze the expert clashes over definitions of livebirth, which impact infant mortality statistics. We analyze the divergent practices and negotiations between countries: since the infant mortality rate came to represent the level of socioeconomic advancement, its political significance was paramount. Analyzing the struggle to reduce infant mortality thus helps us understand how socialist countries positioned themselves within the transnational framework while being members of the “socialist bloc.”

- Abstract
Carbonic anhydrase IX (CAIX) is a transmembrane enzyme that regulates pH in hypoxic tumors and promotes tumor cell survival. Its expression is associated with the occurrence of metastases and poor prognosis. Here, we present nine derivatives of the cobalt bis(dicarbollide)(1−) anion substituted at the boron or carbon sites by alkysulfamide group(s) as highly specific and selective inhibitors of CAIX. Interactions of these compounds with the active site of CAIX were explored on the atomic level using protein crystallography. Two selected derivatives display subnanomolar or picomolar inhibition constants and high selectivity for the tumor-specific CAIX over cytosolic isoform CAII. Both derivatives had a time-dependent effect on the growth of multicellular spheroids of HT-29 and HCT116 colorectal cancer cells, facilitated penetration and/or accumulation of doxorubicin into spheroids, and displayed low toxicity and showed promising pharmacokinetics and a significant inhibitory effect on tumor growth in syngenic breast 4T1 and colorectal HT-29 cancer xenotransplants.

- Abstract
Acute methanol poisoning leads to optic neuropathy and necrotic lesions of basal ganglia (BG) and subcortical white matter. Survivors of methanol poisoning exhibit long‐term executive and memory deficits. Associations between brain volumetry parameters and cognitive sequelae of methanol poisoning are not known. The aim of our study was to identify long‐term associations between the cognitive performance of survivors of methanol poisoning and the volume of the brain structures that are selectively vulnerable to methanol. We conducted a cross‐sectional follow‐up study on a sample of patients (n= 33, age 50 ± 14 years, 82% males) who survived acute methanol poisoning during methanol mass poisoning outbreak from September 2012 till January 2013 in the Czech Republic. A battery of neuropsychological tests and brain magnetic resonance imaging were included in the clinical examination protocol. Specific brain structures (putamen, globus pallidus, nucleus caudatus, and frontal white matter) were selected as regions of interest, and their volumes were estimated using the MorphoBox prototype software. In robust multiple regression models, sustained visual attention performance (as assessed by Trail Making Test and Prague Stroop Test) was positively associated with BG structures and frontal white matter volumes (Wald = 9.03 to 85.50, p < 0.01), sensitivity to interference (as assessed by Frontal Battery Assessment) was negatively associated with frontal white matter volume (Wald = 35.44 to 42.25, p < 0.001), and motor performance (as assessed by Finger Tapping Test) was positively associated with globus pallidus and frontal white matter volumes (Wald = 9.66 to 13.29, p < 0.01). Our results demonstrate that smaller volumes of elements of BG–thalamocortical circuitry, namely the BG and frontal white matter, relate to attention and motor performance in methanol poisoning from a long‐term perspective. Disruption of those functional circuits may underlie specific cognitive deficits observed in methanol poisoning. Cognitive sequelae of methanol poisoning involve executive dysfunction and memory impairment possibly mediated by disruption of basal ganglia (BG) circuitry. Here we show that in survivors of methanol poisoning smaller BGstructures volumes are associated with worse performance in sustained visual attention, and psychomotor speed tasks. We propose the results can be used to build a neuropsychological model of acute toxicological BGdamage which can be extrapolated to effects of other poisons that were shown to selectively affect BG.

123. ‘We Won’t Ban Castrating Pervs Despite What Europe Might Think!’: Czech Medical Sexology and the Practice of Therapeutic Castration

The Czech Republic holds one of the highest numbers of men labelled as sexual delinquents worldwide who have undergone the irreversible process of surgical castration – a policy that has elicited strong international criticism. Nevertheless, Czech sexology has not changed its attitude towards ‘therapeutic castration’, which remains widely accepted and practised. In this paper, we analyse the negotiation of expertise supporting castration and demonstrate how the changes in institutional matrices and networks of experts (Eyal 2013) have impacted the categorisation of patients and the methods of treatment. Our research shows the great importance of historical development that tied Czech sexology with the state. Indeed, Czech sexology has been profoundly institutionalised since the early 1970s. In accordance with the state politics of that era, officially named Normalisation, sexology focused on sexual deviants and began creating a treatment programme that included therapeutic castration. This practice, the aim of which is to protect society from sex offenders, has changed little since. We argue that it is the expert–state alliance that enables Czech sexologists to preserve the status quo in the treatment of sexual delinquents despite international pressure. Our research underscores the continuity in medical practice despite the regime change in 1989. With regard to previous scholarship on state-socialist Czechoslovakia, we argue that it was the medical mainstream that developed and sustained disciplining and punitive features.

Results from retrospective studies suggest that use of neuromuscular blocking agents during general anaesthesia might be linked to postoperative pulmonary complications. We therefore aimed to assess whether the use of neuromuscular blocking agents is associated with postoperative pulmonary complications.

This paper deals high carbon pearlitic steel wires (patented wires), which used as reinforcement in vehicle tires. The aim is to achieve the production of fine-grained pearlite microstructure. The theoretical part deals with a patent in the production structure as well as the patented brass plated wires. In the experimental focused on the microstructure and macroscopic analysis, followed by degradation of the wire, from an evaluation of the mechanical properties, determination of the depth of decarburization and the detection of corrosion in operation. Idiopathic pes equinovarus (clubfoot) is a congenital deformity of the foot and lower leg defined as a fixation of the foot in plantar flexion, adduction, supination and varus. The deformity does not affect only the foot position, which is usually investigated by radiography, CT, micro-CT, MRI or ultrasound but logically influence the whole gait biomechanics. It is supposed, that clubfoot belongs to a group of fibroproliferative disorders whose origin and multi-hierarchical effect remain unknown. It has been suggested that fibroblasts and growth factors may be involved. To gain a more global view, direct analysis of the protein composition of extracellular matrix, a proteomic approach was used. At present two principle methods are mostly used for the treatment of clubfoot: physiotherapy and the Ponseti method. The determination of the general biological and biomechanical parameters for various regions of the clubfoot can potentially help in the understanding of the mechanisms participating on this serious anomaly and thus contribute to the development of the more efficient therapeutic approach. This review summarizes the present knowledge on the possible pathogenetic mechanisms participating in the development of the clubfoot and their possible relation to the new therapeutic approaches. Background: Several reports describe MRI signs of CNS demyelination in infl ammatory bowel diseases (IBD) patients treated with monoclonal antibody against tumor necrosis factor alpha (anti- TNF-α) and even indicate that anti-TNF-α can trigger multiple sclerosis (MS). The fi rst aim of the study was to evaluate the neurological status of Crohn's disease (CD) patients before anti-TNF-α therapy initiation in order to avoid possible complications in patients with latent demyelinating diseases. Our second aim was to evaluate, by prospectively following their neurological and MRI status, the risk of developing CNS demyelination in patients with CD treated with anti-TNF-α. Methods: Fifty patients, followed for more than 2 years, were included in the prospective phase of the study. Thirty of these patients were treated with anti-TNF-α. Twenty patients without biological therapy were used as controls. Neurostatus and brain MRI were performed in all patients at baseline and after 1.5 years of treatment. CSF examination was performed if MRI raised suspicion of MS. Results: 54% of patients had abnormalities in neurostatus. MRI changes suggestive of demyelination were found in seven cases. In one patient, CSF-specifi c oligoclonal bands were found and anti-TNF-α treatment was contraindicated. No changes in neurological or MRI status were observed after 1.5 years of treatment in either group. Conclusions: Preselection of patients at high risk of developing MS can increase the safety of anti-TNF-alpha treatment. [ABSTRACT FROM AUTHOR]

Within the Solve-RD project (https://solve-rd.eu/), the European Reference Network for Intellectual disability, TeleHealth, Autism, and Congenital Anomalies aimed to investigate whether a reanalysis of exomes from unsolved cases based on ClinVar annotations could establish additional diagnoses. We present the results of the “ClinVar low-hanging fruit” reanalysis, reasons for the failure of previous analyses, and lessons learned.

142. Impact of Hydrophobic Additives on Properties of Clay Plaster

- Abstract
Use of clay plasters in the exterior forces owners to conduct frequent maintenance of the plaster’s surface, which may be damaged by the weather. This paper describes partial results of the research, which strives to develop cost-efficient hydrophobic additives to increase service life of exterior clay plasters. We have chosen 7 hydrophobic materials and added them to the reference prefabricated plaster mixture. Samples of the enhanced plaster were tested to evaluate the additives’ impact on the overall mechanical, thermal and moisture characteristics of the plaster. Long-term testing of the best hydrophobic additives will follow to verify their properties in real buildings. Clay plasters play a significant role for the healthy indoor climate due to the influence on the stability of the indoor temperature and humidity.

In the past, due to a harmonization of standards of the Slovak Republic to European Union standards, there was the amendment of Act no. 455/1991 Coll. on small business. Based on adjustments related regulations was a canceled competence requirement of persons performing regular cleaning and inspection of the chimney. According to statistics of Ministry of Interior of the Slovak Republic is a chimney still one of the most common reasons of fire in residential buildings. The article deals with the analysis of fires caused by chimneys and flues. The main goal is to point out the necessity to solve this problem by stricter regulations on testing and approval of the chimney.

Green tea represents a favourite beverage and green tea extracts are popular components of dietary supplements. The aim of present in vivo study was to report on the effect of defined green tea extract (Polyphenon) in various dosage schemes on drug-metabolizing enzymes in mice. The specific activities and expressions of a panel of drug-metabolizing enzymes (totally 16) were tested in liver and small intestine. Nine enzymes were significantly altered by Polyphenon treatment. The intestinal enzymes were more affected than the hepatic ones. The effects were mostly dose-dependent but short-term treatment had more pronounced impact than the long-term administration. Based on the results, normal consumption of green tea seems to be safe but extremely high doses of green tea extracts in dietary supplements could influence drugs metabolism and efficacy. The effect of microstructure on the mechanical properties of cord steel wires was investigated. Material properties and damage behaviours were identified. Metal cord, for reinforcing articles of an elastomeric material, such as tires, conveyor belts and so on of the single strand type, in particular made up of a plurality of 3, 4 or 6 wires, where in the said wires are twisted together loosely. The metal cord characterized by the fact that the diameter of the individual constituent wires varies between 0.12 and 0.30 mm. R
m tested cord wire was max. 2 946 MPa. Fracture surfaces cords steels were observed.- Published

- Abstract
The paper presents the evaluation of corrosion resistance of aluminium alloy AlSi1MgMn. This alloy is used above all in any atmospheric conditions. The corrosion resistance of the alloy was evaluated by determining the open circuit potential (OCP) in solution SARS (this solution simulates the industrial atmosphere) after the 10 months of exposure time. Currently, the automotive industry used sheets of different qualities. The most common include IF (inter Interstitial Free) steel and alloyed steel. Use the sheet quality depends on the point of application in the production car. Testing and product testing is a standard part of the process of innovation and production itself. Testing of automotive steels under dynamic conditions is increasingly important. Changing the hardness HV 1 was performed on the fractured bars on the static and dynamic loading conditions. Tests were made on steel IF and S 460.

In 2010 sex education became a surprising target of criticism in the secular Czech Republic. The voices of social conservatives were raised and were answered by the Minister of Education, who launched a reform of educational curricula to exclude sex education. This article analyses the discursive strategies employed by conservative opponents of sex education and highlights the interpretive repertoires of sexuality and gender. The authors argue that Czech conservatives deploy both a moral panic strategy and a discursive strategy of empowerment that uses positive sanctions to support 'good sex', defined exclusively as marital, procreative heterosex. This interpretive repertoire of 'good and healthy sexuality' is universally intelligible and thus has the potential to resonate not only with social conservatives but throughout society. When combined with other socially conservative agendas it has the capacity to regiment the public space and diminish the role of public institutions. [ABSTRACT FROM AUTHOR]
