Abstract

Ovarian cancer (OC) is a particularly lethal disease due to intratumoral heterogeneity, resistance to traditional chemotherapy, and poor response to targeted therapy and immunotherapy. Interferon-γ (IFN-γ) is an attractive therapeutic cytokine, with positive responses achieved in multiple OC clinical trials. However, clinical application of IFN-γ in OC is still hindered, due to the severe toxicity when used at higher levels, as well as the considerable pro-metastatic adverse effect when used at lower levels. Thus, an effective combined intervention is needed to enhance the anti-tumor efficacy of IFN-γ and to suppress the IFN-γ-induced metastasis. Here, we uncovered that OC cells develop an adaptive strategy by upregulating midkine (MDK) to counteract the IFN-γ-induced anti-tumor activity and to fuel IFN-γ-induced metastasis. We showed that MDK is a critical downstream target of IFN-γ in OC, and that this regulation acts in a dose-dependent manner and is mediated by STAT1. Gain-of-function studies showed that MDK overexpression promotes cell proliferation and metastasis in OC, indicating that IFN-γ-activated MDK may antagonize IFN-γ in inhibiting OC proliferation but synergize IFN-γ in promoting OC metastasis. Subsequently, we assessed the influence of MDK inhibition on IFN-γ-induced anti-proliferation and pro-metastasis effects using an MDK inhibitor (iMDK), and we found that MDK inhibition robustly enhanced IFN-γ-induced growth inhibition (all CIs < 0.1) and reversed IFN-γ-driven epithelial-to-mesenchymal transition (EMT) and metastasis in OC in vitro. Collectively, these data identify an IFN-γ responsive protein, MDK, in counteracting anti-proliferation while endowing the pro-metastatic role of IFN-γ in cancer treatment, and we therefore propose the combined utilization of the MDK inhibitor in IFN-γ-based therapies in future OC treatment. Abstract

At present, the space network is booming. In the face of an increasingly large space network, how to monitor the network status with high real-time performance becomes increasingly important. This paper is directed to the space network, and proposes a highly real-time technology scheme for space constellation network status monitoring, which supports real-time acquisition of various types of space network status information. Based on the idea of the integration of the satellite and the ground, the design and implementation of both the satellite and the ground are carried out.

Abstract

The bacterium, Vibrio coralliilyticus, is well known as one of the main pathogens involved in coral bleaching; however, there have been no previous reports of this bacterium in sea urchins. In the current study, a dominant bacterium, Rb102, was isolated from the coelomic fluid of a sea urchin (Strongylocentrotus intermedius) with red spotting disease, at a farm in Dalian, Liaoning Province. The isolates were identified based on Gram staining, morphological observations, 16S rDNA phylogenetic tree analysis, identification of physiological and biochemical characteristics, drug sensitivity tests, and artificial infection experiments. The identification and isolation results showed Rb102 to be V. coralliilyticus. Artificial infection experiments showed that Rb102 could infect S. intermedius by injection, causing the typical red spotting symptoms of this disease; the strains isolated from the diseased sea urchins were of the same strain type. The results of the bacterial susceptibility tests showed that Rb102 was highly susceptible to gentamicin, neomycin, tetracycline, cefradine, and ofloxacin, but showed strong resistance to vancomycin, ceftriaxone, kanamycin, rifampicin, midecamycin, cefoperazone, ceftazidime, cotrimoxazole, and penicillin. The results of this study provide a reference for further study of the pathogenesis of V. coralliilyticus-induced erythema in sea urchins (S. intermedius) and its prevention and control. Abstract

With the rapid development of Space technology, on-board software plays a more and more important role in the spacecraft. Stack is an important storage resource for on-board software. If the allocation space of stack is not enough, it may cause stack overflow and software crash. Based on SPARC V8 architecture, this paper introduces a static method for detection of stack overflow. This method does not need to run the on-board software dynamically or design complex test cases. By directly analyzing assembly file generated by the compiler, the stack usage space and the call relationship of functions can be obtained. Taking the entry function of each task as the starting point of stack depth analysis, the function call path is traversed by the stack data structure, and the maximum stack depth of each task is finally calculated. An instance of a task stack detection shows that by analyzing the static assembly file, the maximum depth of stack can be obtained directly, the risk of stack overflow can be avoided, and the reliability and security of on-board software can be improved.

Abstract

An easy-to-use, fast and highly selective method, which used unmodified gold nanorods (GNRs) as a chiral nanosensor for the chiral recognition of cysteine (Cys) enantiomers, has been developed in this work. Under the optimized conditions, L-Cys caused GNRs aggregation, and the absorbance decrease of GNRs at a wavelength of 650 nm was 0.601. However, the GNRs were almost monodispersed and the absorbance decrease was only 0.289 for D-Cys. The combined assay process with the optical rotation of GNRs, and the chiral recognition phenomenon could be assigned to the fact that L-Cys induced the end-to-end assembly of GNRs through crosslinking agent Cu2+ ions. The detection limit of this method for L-Cys was 0.325 μM. The relative standard deviation (RSD) was 1.5% for trace L-Cys, which indicated that the method presented good precision for the analysis of trace L-Cys in solution samples. Futhermore, we demonstrated that the nanosensor possessed high selectivity for the chiral recognition of Cys enantiomers. The response of GNRs (ΔALD) to Cys enantiomers was 7.8 times to tryptophan (Try) enantiomers, and 9.2 times to phenylalanine (Phe) enantiomers. In our perception, the assay showed new opportunities for efficient enantiosensing strategies.
