Cyrtomium adenotrichum Y. Nong & R.H. Jiang (Dryopteridaceae), a new species from Guangxi, China, is described and illustrated. This new species is similar to C. nephrolepioides (Christ) Copel., C. obliquum Ching & K. H. Shing ex K. H. Shing, C. sinningense Ching & K. H. Shing ex K. H. Shing and C. calcis Liang Zhang, N.T.Lu & Li Bing Zhang in having erect rhizomes, dense, leathery lamina and rounded sori, but it can be easily distinguishable by its stipe sparsely glandular, base obvious oblique, basiscopic base truncate, acroscopic base auriculate or ovate.

By dispersing the fuel particles in an inert metal matrix, the dispersion nuclear fuel element has the advantages of high burnup, high thermal conductivity, great safety and strong designability, etc. It has been successfully applied in the test reactors. Within a dispersion nuclear fuel element, the failure of fuel meat is significantly influenced by the particle agglomeration behavior, including the agglomeration extent, the location of agglomeration region, fuel particle spacing, etc. The present study investigates the crucial factors influencing the failure behavior in the dispersion nuclear fuel meat, focusing on the particle agglomeration effect on the failure of fuel meat. By developing a script for parametric modeling and numerical calculations of the representative volume element in the dispersion nuclear fuel element, the Mises stress field of dispersion nuclear fuel meat incorporating with different particle agglomeration characteristics was simulated in this paper. Numerical modeling was performed by considering the damage layer at the fuel-matrix interface, irradiation-induced fuel particles swelling and the environmental pressure outside the cladding. The effects of particle agglomeration volume fraction, location of the agglomerate fuel particles, ambient hydro-static pressure and matrix material properties on the failure of dispersion nuclear fuel meat were analyzed systematically. The fuel particles in agglomeration region were geometrically modeled through dense stacking, while the local particle agglomeration volume fraction was defined by the minimum particle spacing. The results show that the failure behavior in dispersion nuclear fuel meat is enhanced with the increasing of particle agglomeration volume fraction. The environmental pressure outside the cladding can inhibit the stress concentration in the dispersion fuel meat, but this influence declines with the extent of local fuel agglomeration increasing. Furthermore, the particle agglomeration location between the upper and lower cladding layers of the thin plate indicates little effect on the maximum Mises stress in the dispersion nuclear fuel meat. However, compared to the dispersion meat adjacent to the surrounding cladding, fuel particles agglomerating in central meat (i.e. only constrained by the upper and lower cladding) are more sensitive to environmental pressure. The local fuel particle agglomeration has a shortest stave effect on the failure of dispersion nuclear fuel meat. The volume fraction of agglomeration will seriously exacerbate the stress concentration, and the agglomeration location in the interior of dispersion fuel meat will fail first. Therefore, it is concluded that the extent of fuel particle agglomeration, the location of agglomeration region within the dispersion nuclear fuel meat, and the environmental pressure are crucial factors influencing the failure behavior of fuel meat. The fuel particle agglomeration region is the onset location for the local macroscopic failure, and the environmental pressure outside the cladding can decrease the plastic stress concentration in the dispersion fuel meat to improve the safety of reactor core. This work provides an analytical approach and numerical reference for the failure condition prediction, the reliability evaluation, and the structural optimization design of the plate dispersion nuclear fuel element.

Abstract Natural killer (NK) cells play essential roles in the tumor development, diagnosis, and prognosis of tumors. In this study, we aimed to establish a reliable signature based on marker genes in NK cells, thus providing a new perspective for assessing immunotherapy and the prognosis of patients with gastric cancer (GC). We analyzed a total of 1560 samples retrieved from the public database. We performed a comprehensive analysis of single-cell RNA-sequencing (scRNA-seq) data of gastric cancer and identified 377 marker genes for NK cells. By performing Cox regression analysis, we established a 12-gene NK cell-associated signature (NKCAS) for the Cancer Genome Atlas (TCGA) cohort, that assigned GC patients into a low-risk group (LRG) or a high-risk group (HRG). In the TCGA cohort, the areas under curve (AUC) value were 0.73, 0.81, and 0.80 at 1, 3, and 5 years. External validation of the predictive ability for the signature was then validated in the Gene Expression Omnibus (GEO) cohorts (GSE84437). The expression levels of signature genes were measured and validated in GC cell lines by real-time PCR. Moreover, NKCAS was identified as an independent prognostic factor by multivariate analysis. We combined this with a variety of clinicopathological characteristics (age, M stage, and tumor grade) to construct a nomogram to predict the survival outcomes of patients. Moreover, the LRG showed higher immune cell infiltration, especially CD8+ T cells and NK cells. The risk score was negatively associated with inflammatory activities. Importantly, analysis of the independent immunotherapy cohort showed that the LRG had a better prognosis and immunotherapy response when compared with the HRG. The identification of NK cell marker genes in this study suggests potential therapeutic targets. Additionally, the developed predictive signatures and nomograms may aid in the clinical management of GC.

Summary: Background: Co-existence of efficient transportation networks and geographic imbalance of medical resources greatly facilitated inter-city migration of patients of infectious diseases in China. Methods: To characterize the migration patterns of major notifiable infectious diseases (NIDs) during 2016–2020 in China, we collected migratory cases, who had illness onset in one city but were diagnosed and reported in another, from the National Notifiable Infectious Disease Reporting System, and conducted a nationwide network analysis of migratory cases of major NIDs at the city (prefecture) level. Findings: In total, 2,674,892 migratory cases of NIDs were reported in China during 2016–2020. The top five diseases with the most migratory cases were hepatitis B, tuberculosis, hand, foot and mouth disease (HFMD), syphilis, and influenza, accounting for 79% of all migratory cases. The top five diseases with the highest proportions of migratory cases were all zoonotic or vector-borne (37.89%‒99.98%). The network analysis on 14 major diseases identified three distinct migration patterns, where provincial capitals acted as key node cities: short distance (e.g., pertussis), long distance (e.g., HIV/AIDS), and mixed (e.g., HFMD). Strong drivers for patient migration include population mobility and labor flow intensities between cities as well as the economic development level of the destination city. Interpretation: Collaborative prevention and control strategies should target cities experiencing frequent patient migration and cater to unique migration patterns of each disease. Addressing disparity in healthcare accessibility can also help alleviate case migration and thereby reduce cross-regional transmission. Epilepsy, a chronic neurological disorder affecting millions globally, is often exacerbated by neuroinflammation and oxidative stress. Existing antiepileptic drugs primarily manage symptoms, leaving the disease’s progression largely unaddressed. Flavonoids, ubiquitous plant metabolites with potent anti-inflammatory and antioxidant properties, show promise in epilepsy treatment. Unlike conventional therapies, they target multiple pathophysiological processes simultaneously, offering a comprehensive approach to this complex neurological disorder. This review explores the dual role of flavonoids in mitigating neuroinflammation and reducing oxidative stress through various molecular pathways. By inhibiting key inflammatory mediators and pathways such as NF-κB, MAPK, JNK, and JAK, flavonoids offer neuronal protection. They enhance the body’s natural antioxidant defenses by modulating enzyme activities, including superoxide dismutase, catalase, and glutathione peroxidase. Moreover, flavonoids influence crucial antioxidant response pathways like PI3K/AKT, Nrf2, JNK, and PKA. Despite their therapeutic promise, the low bioavailability of flavonoids poses a considerable challenge. However, cutting-edge strategies, including nanotechnology and chemical modifications, are underway to improve their bioavailability and therapeutic efficacy. These advancements support the potential of flavonoids as a valuable addition to epilepsy treatment strategies.

Background: Glycoprotein non-metastatic melanoma B (GPNMB)/osteoactivin was first identified in the human melanoma cell lines. GPNMB plays a key role in the anti-inflammatory and antioxidative functions as well as osteoblast differentiation, cancer progression, and tissue regeneration. Recently, GPNMB was used as an anti-aging vaccine for mice. The present study aimed to investigate the potential of biofluid GPNMB as an aging biomarker in humans using serum and urine samples from an aging Chinese population. Methods: We analyzed RNA-sequencing data (GSE132040) from 17 murine organs across different ages to assess the gene expression of potential ageing biomarkers. Spearman's correlation coefficients were used to evaluate the relationship between gene expression and age. Meanwhile, a cross-sectional population study was conducted, which included 473 participants (aged 25–91 years), a representative subset of participants from the Peng Zu Study on Healthy Ageing in China (Peng Zu Cohort). Biofluid GPNMB levels were measured by ELISA. The associations of serum and urine GPNMB levels with various clinical and anthropometrical indices were assessed using ANOVA, Kruskal-Wallis H test, and univariate and multivariate linear regression analyses. Herein, we proposed novel three-in-one DNA nanowheels with simultaneous chemo and gene therapy to treat tumor, especially to prevent simultaneous drug resistance, which could be disassembled via a cascaded hybridization reactions triggered by the highly expressed microRNA in cancer cells for smart and efficient cancer therapy. Typically, with breast cancer as a model, microRNA 21 could trigger the self-disassembly of DNA nanowheel 1 via hybridization with a specially designed oligonucleotide (anti-microRNA 21) in DNA nanowheel 1, releasing another special oligonucleotide (Contact sequence) to trigger the self-disassembly of DNA nanowheel 2 with releasing of a special oligonucleotide (anti-Contact sequence) to trigger the self-disassembly of DNA nanowheel 1 cyclically, and thus the cascaded hybridization reactions with three-in-one anti-cancer functions could be generated based on three main therapeutic effects via releasing doxorubicin to inhibit macromolecular biosynthesis, antisense oligonucleotide of microRNA 21 to activate the apoptotic cell pathway and antisense oligonucleotide of MDR1 to prevent the drug resistance respectively. As expected, the proposed method showed improved therapeutic efficacy on the cancer cells with about 80% apoptosis ratio, especially on the drug resistant cancer cells with about 75% apoptosis ratio, compared with that in the conventional anti-cancer systems of about 70% on cancer cells and below 40% on drug resistant cancer cells, respectively. Most importantly, this strategy opened the door for generation of complex functional DNA-based structures for target triggering drugs releasing system combining with chemo- and gene-therapy to generate tumor regression and prevent drug resistance with an optimized therapeutic efficacy, providing a new avenue for efficient cancer treatment, especially drug resistant cancers.

BackgroundUnderstanding and mapping the distribution of sandflies and sandfly-associated pathogens (SAPs) is crucial for guiding the surveillance and control effort. However, their distribution and the related risk burden in China remain poorly understood.MethodsWe mapped the distribution of sandflies and SAPs using literature data from 1940 to 2022. We also mapped the human visceral leishmaniasis (VL) cases using surveillance data from 2014 to 2018. The ecological drivers of 12 main sandfly species and VL were identified by applying machine learning, and their distribution and risk were predicted in three time periods (2021-2040, 2041-2060, and 2061-2080) under three scenarios of climate and socioeconomic changes.ResultsIn the mainland of China, a total of 47 sandfly species have been reported, with the main 12 species classified into three clusters according to their ecological niches. Additionally, 6 SAPs have been identified, which include two protozoa, two bacteria, and two viruses. The incidence risk of different VL subtypes was closely associated with the distribution risk of specific vectors. The model predictions also revealed a substantial underestimation of the current sandfly distribution and VL risk. The predicted areas affected by the 12 major species of sandflies and the high-risk areas for VL were found to be 37.9-1121.0% and 136.6% larger, respectively, than the observed range in the areas. The future global changes were projected to decrease the risk of mountain-type zoonotic VL (MT-ZVL), but anthroponotic VL (AVL) and desert-type zoonotic VL (DT-ZVL) could remain stable or slightly increase.ConclusionsCurrent field observations underestimate the spatial distributions of main sandfly species and VL in China. More active surveillance and field investigations are needed where high risks are predicted, especially in areas where the future risk of VL is projected to remain high or increase.

Summary: The emergence of novel Omicron subvariants has raised concerns regarding the efficacy of immunity induced by prior Omicron subvariants breakthrough infection (BTI) or reinfection against current circulating Omicron subvariants. Here, we prospectively investigated the durability of antibody and T cell responses in individuals post Omicron BA.2.2 BTI, with or without subsequent Omicron BA.5 reinfection. Our findings reveal that the emerging Omicron subvariants, including CH.1.1, XBB, and JN.1, exhibit extensive immune evasion induced by previous infections. Notably, the level of IgG and neutralizing antibodies were found to correlate with subsequent Omicron BA.5 reinfection. Fortunately, T cell responses recognizing both Omicron BA.2 and CH.1.1 peptides were observed. Furthermore, Omicron BA.5 reinfection may alleviate immune imprinting induced by WT-vaccination, bolster virus-specific ICS+ T cell responses, and promote the phenotypic differentiation of virus-specific memory CD8+ T cells. Antigen-updated or T cell-conserved vaccines are needed to control the transmission of diverse emerging SARS-CoV-2 variants.
