Premise: Developing an effective and easy‐to‐use high‐molecular‐weight (HMW) DNA extraction method is essential for genomic research, especially in the era of third‐generation sequencing. To efficiently use technologies capable of generating long‐read sequences, it is important to maximize both the length and purity of the extracted DNA; however, this is frequently difficult to achieve with plant samples. Methods and Results: We present a HMW DNA extraction method that combines (1) a nuclei extraction method followed by (2) a traditional cetyltrimethylammonium bromide (CTAB) DNA extraction method for plants with optimized extraction conditions that influence HMW DNA recovery. Our protocol produced DNA fragments (percentage of fragments >20 kbp) that were, on average, ca. five times longer than those obtained using a commercial kit, and contaminants were removed more effectively. Conclusions: This effective HMW DNA extraction protocol can be used as a standard protocol for a diverse array of taxa, which will enhance plant genomic research. [ABSTRACT FROM AUTHOR]

Young adults endorse more individualistic and internal adulthood milestones compared to prior generations. Arnett (1994) introduced the Markers of Adulthood (MoA) scale to capture this shift in the transition to adulthood using 38 markers associated with becoming an adult, including marriage, having children, and living independently. These items were based on psychological, anthropological, and sociological determinations concerning adulthood, and were arranged into subscales based on their theoretical association rather than statistical analysis. Since the scale was introduced, researchers have addressed crucial questions about the validity of the MoA scale's milestones. A recurring theme was identifying items that could be sorted into reliable subscales. We examined a collection of original items and included new ones, such as "have a professional social media account" and "recognize personal capabilities and shortcomings" to configure a revised MoA model. A total of 861 participants in seven national locations responded to a demographic survey, the Inventory of the Dimensions of Emerging Adulthood (IDEA; Reifman, et al., 2007), and a collection of MoA items. We conducted a principal component analysis to identify 22 items and four factors (role transitions, independence, legality markers, and relative maturity) which represented 55% of the total variance in the dataset. All factors except legality markers were identified by prior researchers. While four factors demonstrated the best fit for subscale configurations, the revised MoA was considered most reliable when used in its entirety. The gut microbiome of Antarctic fish species has rarely been studied due to difficulties in obtaining samples. The mackerel icefish, Champsocephalus gunnari, primary feeds on krill and is one of the key species in the food web of the Southern Ocean. In this study, we characterized the gut microbiota of C. gunnari by sequencing the V3–V4 region of the bacterial 16S rRNA gene based on the Illumina MiSeq sequencing platform. We collected three types of samples: (1) whole intestine, (2) intestinal wall, and (3) intestinal content. The results showed no significant difference in the alpha diversity between different sample types. However, the microbial community composition of intestinal wall samples was distinct from other sample types. The relative abundance of Photobacterium was higher in intestinal content compared with the walls, which could be due to their chitinolytic activity. In contrast, potential pathogens such as Escherichia, Shigella, and Pseudomonas relatively more abundant in the intestinal wall compared with the intestinal contents. Unlike the gut microbiome of other marine fish species, Vibrio and Lactobacillus were nearly absent in the gut microbiome of C. gunnari. Functional gene profile of the gut microbiome predicted by PICRUSt2 showed higher relative abundance of genes related to biodegradation of nutrients in intestinal content. In contrast, the relative abundance of genes related to biosynthesis of important metabolites, such as menaquinols, was higher in intestinal wall. The difference in the microbial community structure of intestinal wall and intestinal content found in our study supports niche separation in the gut environment and emphasizes the importance of collecting intestinal wall samples in addition to intestinal content samples to understand the full picture of gut microbiome. Metal-catalysed reactions are often hypothesized to proceed on bifunctional active sites, whereby colocalized reactive species facilitate distinct elementary steps in a catalytic cycle1–8. Bifunctional active sites have been established on homogeneous binuclear organometallic catalysts9–11. Empirical evidence exists for bifunctional active sites on supported metal catalysts, for example, at metal–oxide support interfaces2,6,7,12. However, elucidating bifunctional reaction mechanisms on supported metal catalysts is challenging due to the distribution of potential active-site structures, their dynamic reconstruction and required non-mean-field kinetic descriptions7,12,13. We overcome these limitations by synthesizing supported, atomically dispersed rhodium–tungsten oxide (Rh-WOx) pair site catalysts. The relative simplicity of the pair site structure and sufficient description by mean-field modelling enable correlation of the experimental kinetics with first principles-based microkinetic simulations. The Rh-WOx pair sites catalyse ethylene hydroformylation through a bifunctional mechanism involving Rh-assisted WOx reduction, transfer of ethylene from WOx to Rh and H2 dissociation at the Rh-WOx interface. The pair sites exhibited >95% selectivity at a product formation rate of 0.1 gpropanal cm−3 h−1 in gas-phase ethylene hydroformylation. Our results demonstrate that oxide-supported pair sites can enable bifunctional reaction mechanisms with high activity and selectivity for reactions that are performed in industry using homogeneous catalysts.Catalysts consisting of oxide-supported pair sites can enable bifunctional reaction mechanisms with high activity and selectivity for reactions and so overcome the limitations in industry imposed by the use of homogeneous catalysts.
