Abstract

We evaluated real-world effectiveness and safety of CT-P10 (Truxima®) compared with originator rituximab in diffuse large B-cell lymphoma (DLBCL) treatment. Before and after the introduction of CT-P10 to our institute (November 2017), 221 newly-diagnosed DLBCL patients received rituximab with standard cyclophosphamide, vincristine, doxorubicin and prednisone. Patients received originator rituximab throughout (n = 95), switched from originator rituximab to CT-P10 (n = 36), or received CT-P10 throughout (n = 90). There were no significant differences between groups in overall response rate (91.6% vs 94.4% vs 96.7%, respectively; p = 0.403) or complete response rate (84.2% vs 77.8% vs 86.7%, respectively; p = 0.467). Kaplan–Meier survival curves also showed no significant differences in progression-free survival and overall survival between groups (log-rank p = 0.794 and p = 0.955, respectively). Safety profiles were comparable between treatment groups. These data support the ability of CT-P10 to successfully replace originator rituximab in DLBCL treatment and, given the lowered financial barrier, to improve the overall prognosis for DLBCL patients. Abstract

Despite years of research into its pathobiology and continuing clinical trials for novel therapies, the prognosis for patients with glioblastoma (GBM) remains dismal. An important obstacle against treatment efficacy may be a high degree of intra- and inter-tumoral heterogeneity within GBMs, which may be caused by the presence of self-renewing GBM stem cells (GSCs). Recent advances in multi-omics technology introduce new possibilities for applying personalized strategies to GBM therapy. As drug discovery is accelerating with the transition from non-selective, cytotoxic therapy to a precision, targeted approach, the appropriate in vivo platform for GBM is critical for validating drug targets and prioritizing candidates for clinical studies, for co-development of companion diagnostics and, ultimately, for drug approval. Here we will describe GBM orthotopic patient-derived xenografts (PDXs) as more useful, clinically relevant resources for individually tailored strategies for GBM.

120. Shot Boundary Detection with Graph Theory using Keypoint Features and Color Histograms

Abstract

The TRECVID report of 2010 [14] evaluated video shot boundary detectors as achieving "excellent performance on [hard] cuts and gradual transitions." Unfortunately, while re-evaluating the state of the art of the shot boundary detection, we found that they need to be improved because the characteristics of consumer-produced videos have changed significantly since the introduction of mobile gadgets, such as smartphones, tablets and outdoor activity purposed cameras, and video editing software has been evolving rapidly. In this paper, we evaluate the best-known approach on a contemporary, publicly accessible corpus, and present a method that achieves better performance, particularly on soft transitions. Our method combines color histograms with key point feature matching to extract comprehensive frame information. Two similarity metrics, one for individual frames and one for sets of frames, are defined based on graph cuts. These metrics are formed into temporal feature vectors on which a SVM is trained to perform the final segmentation. The evaluation on said "modern" corpus of relatively short videos yields a performance of 92% recall (at 89% precision) overall, compared to 69% (91%) of the best-known method.

Abstract

Introduction Germline BRCA mutations may have therapeutic implications as surrogate markers of DNA-damage repair status in pancreatic ductal adenocarcinoma (PDAC). We performed a prospective study to evaluate the efficiency of risk criteria based on personal or family history of breast and ovarian cancer for determining germline BRCA mutations in PDAC patients with Asian ethnicity. Methods Between November 2015 and May 2016, we screened consecutive PDAC patients with locally advanced unresectable or metastatic disease who were referred for systemic chemotherapy. Analyses for germline BRCA mutations were performed if patients had one or more first-degree or second-degree relatives with breast or ovarian cancers or had a personal medical history of these diseases. DNA was extracted from whole blood, and all coding exons and their flanking intron regions of BRCA1 and BRCA2 were sequenced. Results A total of 175 patients were screened for personal and family history and 10 (5.7%) met the inclusion criteria for genetic sequencing. Pathogenic germline BRCA2 mutation [c.7480C>T (p.Arg2494*)] was identified in one male patient, resulting in a frequency of 10% for the risk-stratified patients and 0.6% for the unselected PDAC population. Two patients had germline BRCA2 variants of uncertain significance [c.1744A>C (p.Thr582Pro) and c.68-7T>A]. Conclusion Personal or family history of breast or ovarian cancers is a feasible, cost-effective risk categorization for screening germline BRCA mutations in Asian PDAC patients as 10% of this population had the pathogenic mutation herein. Abstract

18F-fluoro-2-dexoy-D-glucose-positron emission tomography (PET)/computed tomography (CT) is a useful imaging technique for monitoring the treatment response in lymphoma cases. We investigated the value of interim brain PET/CT (I-PET/CT) for monitoring the response to intensive methotrexate-based chemotherapy in primary central nervous system lymphoma (PCNSL) patients with diffuse large B cell lymphoma (DLBCL). Of the 76 PCNSL patients treated with intensive methotrexate and cytarabine chemotherapy between September 2006 and December 2012, 66 patients with DLBCL were included in this study. The patient cohort of 66 individuals comprised 43 men and 23 women with a median age of 59 years (range, 17-75 years). During chemotherapy, 36 patients (54.5%) showed a negative metabolism on I-PET/CT, and 47 (71.2%) were negative on final (F) PET/CT. The baseline characteristics were similar between I-PET/CT-negative (n = 36) and I-PET/CT-positive patients (n = 30) except ECOG performance status. After a median follow-up of 27.5 months, there was no difference in the progression-free survival (PFS; P = 0.701) or overall survival (OS; P = 0.620) between the I-PET/CT-negative and I-PET/CT-positive groups. However, PFS in the F-PET/CT-negative group was significantly longer than that in the F-PET/CT-positive group (P < 0.001) without a significant difference in OS (P = 0.892). I-PET/CT may not predict the survival outcome of PCNSL patients with DLBCL treated with intensive methotrexate and cytarabine chemotherapy. Abstract

脳外傷者の就業状況や脳外傷者の就業を促進する要因と妨げとなる要因を明らかにし,今後の就業援助のための示唆を得ることを目的に,韓国の地域職業リハビリテーションセンターに過去約5年間(1995～2000)求職相談に訪れた障害者,1542名のうち65名を脳外傷と同定し,その就業状況を公式記録から得て分析した.65名のうち,初回求職相談後2000年8月の調査時点までに一度でも就業を経験した者は36名であった.調査時点に就業中であった者は25名で,そのうち事業所に就業して1年以上勤続していた者は6名のみであった.求職相談後の就業にプラスに働いた要因は,居住地域がセンターの近隣または都市部であったこと,受傷後求職相談に訪れるまでの間に何らかの教育・職業訓練経験,就業経験があったこと,及び,求職相談時に就業斡旋と判定され,実際にセンターの就業斡旋を受けたことであった.また,勤続にプラスに働いた要因には障害が軽度で,障害受容ができたこと,家族,会社,職業リハビリテーション機関からのサポートがあったことで,マイナス要因には,本人の身体的・精神的能力の問題,作業の不適合性,会社内の対人関係の問題や物理的・制度的環境の不備,経済状況などがあった., The purpose of this research was to investigate the employment status of people with traumatic brain injury and factors related to their employability in Korea. From the official documents of a local vocational rehabilitation center, 65 people were identified as victims of head trauma among 1542 people with disabilities over 4 years and 8 months (1995-2000). Information about their demographical factors, disabilities, educational/vocational experience after disability, the vocational rehabilitation services of the center and vocational experience after registration was gathered and analyzed descriptively. The factors which influenced employment were also analyzed by logistic regression. Educational and/or job experience after the injury, accessibility to the vocational rehabilitation service, and job introductions from the center were related to their employment after registration to the center. The results were further analyzed with reference to the Canadian Model of Occupational Therapy (CMOP). Finally, this article suggests the necessity of a client-centered vocational rehabilitation program for people with traumatic brain injury in the community.

Abstract

In this paper, we address the problem of building a grid map as accurately as possible using inexpensive and error-prone sonar sensors. In this research area, incorrect sonar measurements, which fail to detect the nearest obstacle in their beamwidth, generally have been dealt with in the same manner as correct measurements or have been excluded from the mapping. In the former case, the map quality may be severely degraded. In the latter case, the resulting map may have insufficient information after the incorrect measurements are removed because only correct measurements are frequently insufficient to cover the whole environment. We propose an efficient grid-mapping approach that incorporates incorrect measurements in a specialized manner to build a better map; we call this the enhanced maximum likelihood (eML) approach. The eML approach fuses the correct and incorrect measurements into a map based on sub-maps generated from each set of measurements. We also propose the maximal sound pressure (mSP) method to detect incorrect sonar readings using the sound pressure of the waves from sonar sensors. In several indoor experiments, integrating the eML approach with the mSP method achieved the best results in terms of map quality among various mapping approaches. We call this the maximum likelihood based on sub-maps (MLS) approach. The MLS map created using only two sonar sensors exhibited similar accuracy to the reference map, which was an accurate representation of the environment.

Abstract

An advanced sonar morphology-based posterior approach (SMP) to building an occupancy grid map for a mobile robot is proposed in this study. It is very important for a mobile robot to find its surrounding saliencies and to localize itself for indoor navigation. Ultrasonic sensors are of great practical value in building environmental maps and for autonomous operation. However, grid maps constructed by ultrasonic sensors cannot typically form a realistic representation of a given environment due to incorrect sonar measurements caused by specular reflection and wide beam width. The sonar sensor model proposed in this study, in which the negative effect of incorrect sonar measurements is minimized by geometric association with sonar footprints, is adopted to build a high-quality grid map. Experimental results and evaluations in home and corridor environments demonstrate the validity of the proposed methods. [ABSTRACT FROM AUTHOR]
