104. Comparison of NACA 0012 Laminar Flow Solutions: Structured and Unstructured Grid Methods

Abstract

In this paper we consider the solution of the compressible Navier-Stokes equations for a class of laminar airfoil flows. The principal objective of this paper is to demonstrate that members of this class of laminar flows have steady-state solutions. These laminar airfoil flow cases are often used to evaluate accuracy, stability and convergence of numerical solution algorithms for the Navier-Stokes equations. In recent years, such flows have also been used as test cases for high-order numerical schemes. While generally consistent steady-state solutions have been obtained for these flows using higher order schemes, a number of results have been published with various solutions, including unsteady ones. We demonstrate with two different numerical methods and a range of meshes with a maximum density that exceeds 8 × 106 grid points that steady-state solutions are obtained. Furthermore, numerical evidence is presented that even when solving the equations with an unsteady algorithm, one obtains steady-state solutions.

Abstract

Breast cancer is the leading cause of cancer-related death in women worldwide. Development of novel noninvasive diagnostic and predictive pathophysiological biomarkers would represent a significant clinical improvement. Here, we explored the utility of non-invasive 23Na MRI to profile tumour physiology using preclinical mouse models of breast cancer. We establish that tissue Na+ concentration ([Na+]) is elevated vs non-tumour regions across multiple different tumour models. Ex vivo SBFI fluorescence imaging corroborated that this elevation in tumour [Na+] is due to increased intracellular [Na+]. Effective treatment with cytotoxic chemotherapy reduced tumour tissue [Na+], but was not detected by 1H diffusion-weighted imaging (DWI). Moreover, combining 23Na MRI and DWI measurements enabled superior classification accuracy of tumour vs non-tumour regions compared to either parameter alone. Quantification of breast tumour tissue [Na+] using 23Na MRI thus represents a novel, accurate, non-invasive diagnostic and predictive imaging biomarker.

112. The interpersonal processes of non-suicidal self-injury: a systematic review and meta-synthesis

Abstract

Background: Understanding the processes underlying non-suicidal self-injury (NSSI) is important given the negative consequences of this behaviour. Qualitative research has the potential to provide an in-depth exploration of this. There has been limited research regarding the interpersonal processes associated with NSSI; therefore, a meta-synthesis was conducted to investigate this. Methods: A search of PsycINFO, MEDLINE, Web of Science, and CINAHL electronic databases from date of inception to November 2020 was conducted. In total, 30 papers were included in the final review. A meta-ethnographic approach was utilized to synthesize the data. Results: Two overarching themes were found. Within ‘Powerful relational dynamics’, NSSI was cited as a response to participants becoming stuck in aversive or disempowering relational positions with others. Within the ‘Taking matters into their own hands’ subtheme, NSSI was reported as a way for participants to get interpersonal and emotional needs met. Limitations: Several included papers did not comment on the researcher–participant relationship, which may have affected qualitative results. A small number of potentially eligible papers were unavailable for synthesizing. Conclusion: Findings provide a more nuanced investigation of the interpersonal processes underlying NSSI. Consistent with relevant theories, NSSI appears to be a way of mitigating difficult interpersonal experiences or getting interpersonal needs met. NSSI may be engaged in as a substitute to other, less damaging ways to cope. An argument is made for a more empathetic understanding of NSSI and the use of relational interventions. Practitioner points: Self-injury may occur in response to interpersonal stressors Self-injury can be a means to get interpersonal needs met Self-injury may replace other means of coping that become blocked or thwarted Emotional distress can be closely linked with interpersonal factors for this group Relational therapies may be beneficial

Abstract

Objective: Diabetic patients suffer more frequently from biofilm-associated infections than normoglycemic patients. Well described in the literature is a relationship between elevated blood glucose levels in patients and the occurrence of biofilm-associated wound infections. Nevertheless, the underlying pathophysiological pathways leading to this increased infection vulnerability and its effects on biofilm development still need to be elucidated. We developed in our laboratory a model to allow the investigation of a biofilm-associated wound infection in diabetic mice under controlled insulin treatment. Methods: A dorsal skinfold chamber was used on 16 weeks old BKS.Cg-Dock7m +/+ Leprdb/J mice and a wound within the observation field of the dorsal skinfold chamber was created. These wounds were infected with Staphylococcus aureus ATCC 49230 (106 cells/mL). Simultaneously, we implanted implants for sustained insulin release into the ventral subcutaneous tissue (N=5 mice). Mice of the control group (N=5) were treated with sham implants. Serum glucose levels were registered before intervention and daily after the operation. Densitometrical analysis of the wound size was performed at day 0, 3, and 6 after intervention. Mice were sacrificed on day 6 and wound tissue was submitted to fluorescence in situ hybridization (FISH) and colony forming unit (CFU) analysis in addition to immunohistochemical staining to observe wound healing. Experiments were carried out in accordance with the National Institute of Health Guidelines for the Care and Use of Laboratory Animals (protocol number 05/19).Results: The insulin implants were able to reduce blood glucose levels in the mice. Hence, the diabetic mice in the intervention group were normoglycemic after the implantation. The combination with the dorsal skinfold chamber allowed for continuous, in vivo measurements of the infection development. Implantation of the insulin implant and the dorsal skinfold chamber was a tolerable condition for the diabetic mice. We succeeded to realize reproducible biofilm infections in the animals. Discussion: We developed a novel model to assess interactions between blood glucose level and S. aureus -induced biofilm-associated wound infections. The combination of the dorsal skinfold chamber model with a sustained insulin treatment has not been described so far. It allows a broad field of glucose and insulin dependent studies of infection. Einleitung: Patienten mit Diabetes mellitus leiden häufiger unter Biofilm-assoziierten Infekten als normoglykämische Patienten. Das vermehrte zeitgleiche Auftreten erhöhter Blutzuckerwerte und Biofilm-assoziierter Wundinfekte ist gut beschrieben; der dieser erhöhten Infektvulnerabilität zugrundeliegende Pathomechanismus ist allerdings bisher nicht abschließend erforscht. Um dieses Forschungsfeld künftig tiefergehend zu studieren, haben wir in unserem Labor ein neues Tiermodel zur Untersuchung der Biofilmbildung in diabetischen Mäusen unter einer kontrollierten Insulinbehandlung etabliert. Material und Methoden: 16 Wochen alten BKS.Cg-Dock7m +/+ Leprdb/J Mäusen wurde in einem Rückenhautkammermodell eine dermale Wunde gesetzt. Diese Wunden wurden mit Staphylococcus aureus ATCC 49230 (106 Zellen/mL) infiziert. Zeitgleich wurden den Tieren je nach Versuchsgruppe (je N=5) entweder Insulin verabreichende subkutane Implantate (LinShin Canada Inc., Toronto, Canada) oder wirkstofffreie Implantate appliziert. Die Tiere lebten insgesamt 6 Tage. Täglich wurden das Gewicht, die Temperatur und Blutzuckerwerte der Tiere bestimmt. Nach 6 Tagen wurden Gewebeproben jeweils für Fluoreszenz-in-situ -Hybridisierung, CFU und immunhistochemische Färbungen gewonnen. Die Versuche sind entsprechend der Tierversuchsordnung des Landes Sachsen durchgeführt worden (TVV05/19).Ergebnisse: Wir konnten ein neuartiges zuverlässiges Modell zur Untersuchung der Insulintherapie auf die Ausbildung Biofilm-assoziierter Wunden in diabetischen Mäusen etablieren. Die Insulinpumpentherapie konnte eine Reduktion der Blutzuckerwerte erreichen. Die Kombination des Rückenhautkammermodells, welches kontinuierliche In-vivo-Untersuchungen der Infektion erlaubt, mit Insulinimplantaten ist ein für die diabetischen Mäuse gut tolerierbares Modell.Diskussion: Wir haben ein neuartiges Modell zur Untersuchung des Zusammenhangs zwischen Insulin- und Blutzuckerwerten auf die Bildung von Biofilm-assoziierten Wundinfekten am diabetischen Tiermodell entwickelt. Dieses Modell eröffnet ein weites Spektrum an künftigen Forschungsfeldern.

Abstract

BACKGROUND: Asthma is one of the most common chronic diseases in both children and adults. Asthma first occurring in adulthood (adult-onset asthma, AOA) is associated with poorer prognosis compared to childhood-onset asthma (COA), which urgently calls for more research in this area. The aim of this work was to analyze the data on asthma collected in the German National Cohort and compare it with the German Health Interview and Examination Survey for Adults (DEGS), in particular regarding AOA. MATERIAL AND METHODS: Our analysis was based on the dataset of the main questionnaire at mid-term of the German National Cohort baseline examination, comprising 101,723 participants. Variables considered in the analyses were self-reported diagnosis of asthma, age at first diagnosis, asthma treatment in the past 12 months, age, and sex. RESULTS: In the midterm dataset, 8.7% of women and 7.0% of men in the German National Cohort reported that they had ever been diagnosed with asthma. Approximately one third of participants with asthma received their initial diagnosis before their 18th birthday. COA affected 2.2% of women and 2.8% of men, whereas AOA affected 6.5% of women and 4.2% of men. During the previous 12 months, 33% of COA cases and 60% of AOA cases were medically treated. CONCLUSION: The proportion of persons affected by asthma in the German National Cohort, as well as observed patterns regarding age and gender, corresponds to other data sources such as DEGS. However, in our analysis, the proportion of individuals with AOA was higher than described in the literature. The increase in cumulative asthma diagnoses with age is markedly steeper in younger participants, indicating a rising trend over time.

Abstract

BACKGROUND: Infectious diseases continue to play an important role for disease perception, health-economic considerations and public health in Germany. In recent years, infectious diseases have been linked to the development of non-communicable diseases. Analyses of the German National Cohort (GNC) may provide deeper insights into this issue and pave the way for new targeted approaches in disease prevention. OBJECTIVES: The aim was to describe the tools used to assess infectious diseases and to present initial data on infectious disease frequencies, as well as to relate the GNC assessment tools to data collection methods in other studies in Germany. METHODS: As part of the baseline examination, questions regarding infectious diseases were administered using both an interview and a self-administered touchscreen questionnaire. Data from the initial 101,787 GNC participants were analysed. RESULTS: In the interview, 0.2% (HIV/AIDS) to 8.6% (shingles) of respondents reported ever having a medical diagnosis of shingles, postherpetic neuralgia (in cases where shingles was reported), hepatitis B/C, HIV/AIDS, tuberculosis or sepsis if treated in hospital. In the questionnaire, 12% (cystitis) to 81% (upper respiratory tract infections) of respondents reported having experienced at least one occurrence of upper or lower respiratory tract infections, gastrointestinal infections, cystitis or fever within the past 12 months. OUTLOOK: The cross-sectional analyses of data and tools presented here - for example on determinants of susceptibility to self-reported infections - can be anticipated from the year 2021 onward. Beyond that, more extensive research into infectious disease epidemiology will follow, particularly once analyses of GNC biological materials have been performed.

Abstract

BACKGROUND: A nationwide assessment of the respiratory status on the basis of standardized lung function measurements has so far not been available in Germany. The present work describes the lung function tests in the German National Cohort (GNC) and presents initial results based on the GNC Midterm Baseline Dataset. MATERIAL AND METHODS: The assessment of lung function in the GNC comprised spirometry (level 1) and the determination of exhaled nitric oxide (FeNO, level 2). Our quality assurance concept included regular training of lung function test procedures at various GNC sites, interim evaluations of test quality, as well as regular calibration/measurement checks of test equipment. For spirometry, we established a stepwise procedure for offline quality control based on raw flow volume curves. RESULTS: In the present dataset (n = 101,734), spirometry was available for 86,893 study participants and FeNO was available for 15,228 participants. The average (±SD) FEV(1) Z score (according to GLI 2012) was -0.321 ± 1.047, the FVC Z score was -0.153 ± 0.941, and the FEV(1)/FVC Z score was -0.337 ± 0.901. The difference in FEV(1)/FVC between current smokers and never-smokers increased with age. The average FeNO was 14.2 ÷ 2.0 ppb. Current smoking reduced FeNO levels by 43%, whereas respiratory allergy increased FeNO levels by 16% in nonsmokers. DISCUSSION: The results of spirometry and the FeNO measurements are in the expected range with regard to their distributions and correlates. The GNC provides a valuable basis for future investigations of respiratory health and its determinants as well as research into the prevention of respiratory diseases in Germany.
