This study aimed to evaluate the hydrolysates from orange peel waste (OPW) as the low-cost carbon source for lycopene production. Initially, the dilute acid pretreatment combined with enzymatic hydrolysis of OPW resulted in a total sugar concentration of 62.18 g/L. Meanwhile, a four-month adaptive laboratory evolution (ALE) experiment using a d-galacturonic acid minimal medium resulted in an improvement in the growth rate of our previously engineered Escherichia coli strain for lycopene production. After evolutionary adaptation, response surface methodology (RSM) was adapted to optimize the medium composition in fermentation. The results obtained from RSM analysis revealed that the 5.53 % carbon source of orange peel hydrolysate (OPH), 6.57 g/L nitrogen source, and 30 °C temperature boosted lycopene production in the final strain. Subsequently, the optimized treatment for lycopene fermentation was then conducted in a 5 L batch fermenter under the surveillance of a kinetic model that uses the Logistic equation for strain growth (μm = 0.441 h−1), and Luedeking-Piret equations for lycopene production (Pm = 1043 mgL−1) with growth rate constant (α = 0.1491). At last, lycopene biosynthesized from OPH was extracted and analyzed for qualitative validation. Likewise, its data on phytic acid (between 1.01 % and 0.86 %) and DPPH radical scavenging (between 38.06 % and 29.08 %) highlighted the better antioxidant capacity of lycopene. In conclusion, the OPH can be used as a fermentation feedstock which opens new possibilities of exploiting fruit crop residues for food and pharmaceutical applications.

Background Tomato, a fruit with a high vitamin content, is popular for consumption and economically important in Thailand. However, in the past year, the extensive usage of chemicals has significantly decreased tomato yields. Plant Growth-Promoting Rhizobacteria (PGPR) is an alternative that can help improve tomato production system growth and yield quality while using fewer chemicals. The present study aimed to determine whether endophytic actinomycetes promote growth and fruit quality of tomato (Solanum lycopersicum). Methods The experiment was conducted in a net-houses at the Center for Agricultural Resource System Research, Faculty of Agriculture, Chiang Mai University, Chiang Mai province, Thailand. The randomized completely block design (RCBD) was carried out for four treatments with three replications, which was control, inoculation with TGsR-03-04, TGsL-02-05 and TGsR-03-04 with TGsL-02-05 in tomato plant. Isolated Actinomycetes spp. of each treatment was then inoculated into the root zone of tomato seedlings and analyzed by Scanning Electron Microscopy (SEM). The height of tomato plants was measured at 14, 28, 56, and 112 days after transplanting. Final yield and yield quality of tomato was assessed at the maturity phase. Results The SEM result illustrated that the roots of tomato seedlings from all treatments were colonized by endophytic actinomycetes species. This contributed to a significant increase in plant height at 14 days after transplanting (DAT), as found in the TGsR-03-04 treatment (19.40 cm) compared to the control. Besides, all inoculated treatments enhanced tomato yield and yield quality. The highest weight per fruit (47.38 g), fruit length (52.37 mm), vitamin C content (23.30 mg 100 g−1), and lycopene content (145.92 µg g−1) were obtained by inoculation with TGsR-03-04. Moreover, the highest yield (1.47 kg plant−1) was obtained by inoculation with TGsL-02-05. There was no statistically significant difference in the number of fruits per plant, fruit width, brix, and antioxidant activity when various inoculations of endophytic actinomycetes were applied. Therefore, the use of endophytic actinomycetes in tomato cultivation may be an alternative to increase tomato yield and yield quality.

The present study sought to investigate the protective effect of Lycopene (LYC) on Bisphenol A (BPA)-induced lipid accumulation and elucidate the potential molecular mechanism. In vitro, the PK15 cells with or without LYC were exposed to BPA for 6 h to detect the lipid accumulation using Nile red’s and Oil Red O’ staining. Meanwhile, redox status, mitochondrial function, gene expression and protein levels relating to lipid accumulation were evaluated by cellular biochemical assays, immunofluorescence staining, quantitative real-time PCR (qRT-PCR) and Western blot, respectively. In addition, ROS scavenger N-acetylcysteine (NAC), SIRT1 inhibitor EX527, SIRT1 siRNA and PGC-1α siRNA were used to verify the target signaling pathways responsible for the protective effect of LYC against BPA-indcued lipid accumulation. In vivo, the twenty four male Wistar rats were randomly divided into three groups, received either BPA (50 mg/kg/day BW) or LYC (10 mg/kg/day BW) orally for 60 days. The kidney tissues were evaluated by HE’s and Oil Red O’s staining, lipid-associated genes and proteins expression were detected through qRT-PCR and Western blot. The results showed that BPA-exposed PK15 cells and rats had increased lipid droplets with elevated transcription of lipogenic factors SREBP1 and PPARγ, and down-regulated PPARα expression, and which is accompanied by redox imbalance and abnormal mitochondrial function. Conversely, LYC prevented BPA-induced lipid metabolism disorder and lipid accumulation through impeding ROS overproduction. Moreover, PK15 cells and rats exposed to BPA showed decrease in SIRT1 and PGC-1α levels, whereas LYC blocked the decrease of SIRT1 and PGC-1α protein levels. Furthermore, inhibition or silence of SIRT1 and PGC-1α abrogated the improvements of LYC on ROS generation, mitochondrial mass impairement and lipid accumulation in BPA-treated PK15 cells. Collectively, these findings indicate that SIRT1/PGC-1α signaling pathway is involved in the protective effect of LYC on BPA-induced lipid accumulation. These results also provide a potential promising therapeutic of LYC against BPA-induced lipid accumulation in kidney cells and organs.

Temporomandibular joint disorder is a common condition characterized by jaw joint and muscle pain and its treatment is often multifaceted. While conventional treatment modalities exist for the same, interest has grown in exploring alternative therapies such as antioxidants for their potential analgesic properties. The prospective therapeutic use of lycopene, a naturally occurring antioxidant present in tomatoes and other fruits, as an anti‑inflammatory therapeutic modality in the management of temporomandibular joint problems is examined in this narrative review. Lycopene is a carotenoid with high antioxidant activity. Numerous studies show its positive effects in the prevention and amelioration of many diseases. However, due to its lack of water solubility, its use is very limited. Developing a formulation with lycopene with favorable therapeutic parameters will allow for a more effective use of this ingredient. The aim of this study was therefore to use supercritical phase extraction to obtain lycopene-containing preparations, and to obtain complexes of the extract with cyclodextrins to improve its solubility and increase its antioxidant potential. Lycopene-containing extracts were obtained by ultrasound-assisted acetone extraction and supercritical phase extraction. The supercritical extract was combined with g-cyclodextrin, b-cyclodextrin and 2-hydroxypropyl-b-cyclodextrin. Attenuated total reflectance Fourier transform infrared spectroscopy (ATR-FTIR) and X-ray diffraction (XRD) analyses were performed for the obtained systems and extracts. A paddle apparatus was used to evaluate the in vitro dissolution, and the samples collected were analysed by HPLC. The antioxidant activity was assessed using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical method. The results show that cyclodextrins increase the dissolution of lycopene into an acidic environment and enhance the antioxidant potential of the compound. We conclude that the development of a formulation containing a combination of lycopene obtained by supercritical extraction and cyclodextrin will allow for a wider and more effective use of this ingredient.

Background: Lycopene is a dietary antioxidant which provides defense against oxidation. Oxidative stress may result in periodontal tissue damage. There is a positive relationship between lycopene consumption and reduction in the risk of development of degenerative diseases caused by free radicals, such as periodontitis and other diseases like cancer, cardiovascular diseases, asthma, arthritis, stroke, hepatitis. This systematic review discusses the studies conducted to assess the effect of lycopene in periodontal health. Aim: To evaluate the effect of lycopene in maintaining periodontal health. Search Methods: An electronic database search was conducted from the following databases: PubMed, EBSCO Host, Science Direct, EMBASE and Google scholar. Selection Criteria: Randomized controlled trials, experimental studies, case-control studies, and cohort studies published in peer reviewed journals in English language were included. Case reports, case series, animal model studies, in-vitro studies, studies where lycopene was combined with other antioxidants or with additional therapeutic methods and unpublished research were excluded. Data Collection and Analysis: Total 36451 results were obtained after database search. It included case- control studies, experimental studies, reviews and randomized control trials. Out of which, only six studies satisfied the selection criteria. Data extraction sheet was prepared & the studies were analyzed. Results: Out of the six studies analyzed, all were randomized controlled trial. Conclusion: Lycopene has been used widely as an antioxidant in the treatment of chronic diseases. However, Literature is deficient in the studies regarding the effect of lycopene on periodontal health.

Abstract Ionizing radiation (IR) is one of the key contributors that cause male infertility by disturbing spermatogenesis. Lycopene, a carotenoid with strong antioxidant properties, was shown to protect against oxidative damage induced by IR in several experimental models. The present study was designed to explore the possible protective effects of lycopene against IR‐induced testicular damage in C57BL/6 mice. Mice were administered lycopene (20 mg/kg) by oral gavage for seven consecutive days prior to a single dose of whole‐body X‐ray irradiation (4 Gy, 1 Gy/min). We observed that lycopene remarkably augmented sperm motility and reduced sperm abnormalities in mice following IR exposure. Histopathological analyses also revealed that lycopene ameliorated the structural damage of seminiferous tubules and enhanced the regeneration of seminiferous epithelium following IR stress. Moreover, lycopene attenuated IR‐induced oxidative stress, as evidenced by a decreasing lipid peroxidation level and an increase in the antioxidant enzyme superoxide dismutase activity. In addition, lycopene reduced the γH2AX expression and the number of TUNEL‐positive cells in the germinal epithelium, as well as restoring the imbalance of Bax/Bcl‐2 expression induced by IR exposure. Furthermore, lycopene prevented mitochondrial membrane potential depolarization and ATP reduction and preserved the activities of mitochondrial complexes I‐IV in the testes of mice after exposure to IR. Lycopene also improved mitochondrial biogenesis in testes of mice exposed to IR, presenting as restored expressions of PGC‐1α, Nrf1, and Tfam. Taken together, our results suggest that lycopene alleviates IR‐induced testicular damage, and the underlying mechanism involves at least in part the inhibition of the mitochondrial apoptotic pathway and the maintenance of mitochondrial respiration and biogenesis. The beneficial effect of lycopene highlights the therapeutic potential of this plant‐derived antioxidant against impaired spermatogenesis and male infertility induced by IR.

Lycopene, a carotenoid with numerous physiological benefits, particularly in its Z-isomer form, faces challenges in its application due to low chemical stability. To address this limitation, high internal phase emulsion was successfully synthesized using ovalbumin-chitosan complexes. The aim was to enhance the stability of lycopene including Z-lycopene. The solubility, particle size, ζ-potential and uniformity of the mixture were dependent on pH value and biopolymer proportion. Notably, optimal ovalbumin-chitosan complex formation occurred at pH 2.5 with a ratio of 4:1 resulting in the highest solubility and optimal uniformity which contributed to its superior emulsification properties. Evaluation of encapsulating efficiency and loading amount revealed 98.19% and 1.7661 mg/g respectively for lycopene in ovalbumin-chitosan stabilized emulsions, inhibiting the transformation from Z-lycopene to (all-E)-lycopene. The encapsulated lycopene possessed UV stability where retention rate remained high at 81.86%. The retention rate was up to 65.37% and 41.82% at 45 °C and 80 °C, respectively.

Yarrowia lipolytica is a metabolic engineering host of growing industrial interest due to its ability to metabolize hydrocarbons, fatty acids, glycerol, and other renewable carbon sources. This dimorphic yeast undergoes a stress-induced transition to a multicellular hyphal state, which can negatively impact biosynthetic activity, reduce oxygen and nutrient mass transfer in cell cultures, and increase culture viscosity. Identifying mutations that prevent the formation of hyphae would help alleviate the bioprocess challenges that they create. To this end, we conducted a genome-wide CRISPR screen to identify genetic knockouts that prevent the transition to hyphal morphology. The screen identified five mutants with a null-hyphal phenotype-ΔRAS2, ΔRHO5, ΔSFL1, ΔSNF2, and ΔPAXIP1. Of these hits, only ΔRAS2 suppressed hyphal formation in an engineered lycopene production strain over a multiday culture. The RAS2 knockout was also the only genetic disruption characterized that did not affect lycopene production, producing more than 5 mg L-1 OD-1 from a heterologous pathway with enhanced carbon flux through the mevalonate pathway. These data suggest that a ΔRAS2 mutant of Y. lipolytica could prove useful in engineering a metabolic engineering host of the production of carotenoids and other biochemicals.

BACKGROUND: Lycopene (LYC), a carotenoid found in abundance in ripe red fruits, exhibits higher singlet oxygen quenching activity than other carotenoids. However, the stability of LYC is extremely poor due to its high double-bond content. In this paper, a nano-encapsulation strategy based on highly stable marine-derived ferritin GF1 nanocages was used to improve the thermal stability and oxidation resistance of LYC, thereby boosting its functional effectiveness and industrial applicability. RESULTS: The preparation of GF1-LYC nanoparticles benefited from the pH-responsive reversible self-assembly of GF1 to capture LYC molecules into GF1 cavities with a LYC-to-protein ratio of 51 to 1. After the encapsulation of the LYC, the reassembled GF1 nanocages maintained intact morphology and good monodispersity. The GF1-LYC nanoparticles incorporated the characteristic LYC peaks in spectrograms, and their powder form contained the crystalline form of LYC. Molecular docking revealed that LYC bound with the inner triple-axis channel areas of GF1, interacting with VAL139, LYS72, LYS65, TYR69, PHE129, HIS133, HIS62, and TYR134 amino acids through hydrophobic bonds. Fourier transform infrared spectroscopy also demonstrated the bonding of GF1 and LYC. In comparison with free LYC, GF1 reduced the thermal degradation of encapsulated LYC at 37 °C significantly and maintained the 2,2-Diphenyl-1-picrylhydrazyl (DPPH)-scavenging ability of LYC. CONCLUSION: As expected, the water solubility, thermal stability, and antioxidant capacity of encapsulated LYC from GF1-LYC nanoparticles was notably improved in comparison with free LYC, indicating that the shell-like marine ferritin nanoplatform might enhance the stable delivery of LYC and promote its utilization in the field of food nutrition and in other industries. © 2023 Society of Chemical Industry. BACKGROUND: Cherry tomatoes are nutritious and favored by consumers. Processing them into dried cherry tomatoes can prolong their storage life and improve their flavor. The pretreatment of tomato pericarp is crucial for the subsequent processing. However, the traditional physical and chemical treatments of tomato pericarp generally cause nutrient loss and environmental pollution. RESULTS: In this study, a novel enzymatic method for cherry tomatoes was performed using mixed enzymes containing cutinase, cellulase and pectinase. Results showed that the pericarp permeability of cherry tomatoes was effectively improved due to enzymatic treatment. Changes in the microscopic structure and composition of the cuticle were revealed. After treatment with different concentrations of enzymes, cherry tomatoes exhibited higher pericarp permeability and sensory quality to varying degrees. The lycopene content and total polyphenol content significantly increased 2.4- and 1.45-fold, respectively. In addition, the satisfactory effect of the six-time reuse of enzymes on cherry tomatoes could still reach the same level as the initial effect, which effectively reduced the cost of production. CONCLUSIONS: This study revealed for the first time that a mixed enzymatic treatment consisting of cutinase, pectinase and cellulase could effectively degrade the cuticle, enhance the pericarp permeability and improve the quality of cherry tomatoes, with the advantages of being mildly controllable and environmentally friendly, providing a new strategy for the processing of dried cherry tomatoes. Carotenoids are important bioactive substances in breast milk, the profile of which is seldom studied. This study aimed to explore the profile of carotenoids in breast milk and maternal/cord plasma of healthy mother–neonate pairs in Shanghai, China, and their correlation with dietary intake. Maternal blood, umbilical cord blood and breast milk samples from five lactation stages (colostrum, transitional milk and early-, mid- and late-term mature milk) were collected. Carotenoid levels were analysed by HPLC. Carotenoid levels in breast milk changed as lactation progressed (P < 0·001). β -Carotene was the primary carotenoid in colostrum. Lutein accounted for approximately 50 % of total carotenoids in transitional milk, mature milk and cord blood. Positive correlations were observed between five carotenoids in umbilical cord blood and maternal blood (P all < 0·001). β -Carotene levels were also correlated between maternal plasma and three stages of breast milk (r = 0·605, P < 0·001; r = 0·456, P = 0·011, r = 0·446; P = 0·013, respectively). Dietary carotenoid intakes of lactating mothers also differed across lactation stages, although no correlation with breast milk concentrations was found. These findings suggest the importance of exploring the transport mechanism of carotenoids between mothers and infants and help guide the development of formulas for Chinese infants as well as the nutritional diets of lactating mothers. Carotenes have been identified as the most essential carotenoids in the human body, together with lycopene, lutein, and β-cryptoxanthin due to their beneficial antioxidant activities. Because of these properties, we have designed and conducted in 2018--2021 an experiment where various tomato accessions were metabolically profiled for their antioxidant content and other fruit parameters for further usage in crop improvement programs. Same genotypes were phenotyped and profiled with DNA markers to characterize their relative genetic distance. Aggregated data from metabolic and genetic profiling were complemented by the phenotypic data to select for the most promising cross (Pl. karotina x L1116) that was performed and developed into F1 and F2 generations. After phenotyping and genetic profiling of the 104 F2 individuals obtained from that cross, 24 individuals were selected for metabolic profiling for antioxidant contents. In relation to carotenoids accumulated in fully matured fruits the highest lycopene concentration detected in our study was 587.03 µg/g DW, while β-carotene reached 440.29 µg/g DW. Fruit color of tomato is determined by the accumulation of carotenoids which are biochemically represented as C 40 isoprenoid molecules. Tomato is a major dietary source of carotenoids like lycopene and β-carotene. In the present study, we have characterized High β-carotene content (Hbc) mutant which was identified from EMS mutagenized M 2 population of Solanum lycopersicum cv. Arka Vikas (wild-type). Wild-type and mutant fruit phenotype segregation analysis in F 2 population had shown a 3:1 (χ2 ≤ 3.84) ratio of orange and red fruit color and these results suggested that monogenic dominant mutation is responsible for orange colour fruit pigmentation in mutant. We evaluated the contents of phytoene, β-carotene and lycopene at mature green, breaker, turning, and fully (or) red ripe fruit stages in Hbc mutant and wild-type. It was found to have high β-carotene content in Hbc mutant (42.1 ± 8.82 µg/g FW) compared to wild-type (5.28 ± 2.17 µg/g FW). Effect of hbc mutation on whole transcriptome data was analyzed by microarray. This analysis has led to the identification of thousands of differentially expressed genes during fruit ripening. Gene expression pattern affected by hbc mutation, display a tendency for down-regulation rather than up-regulation. The Hbc mutant exhibits four folds up-regulation of the CYC-B gene expression and eight folds high β-carotene content at fully ripen stage. A total of three SNPs at -77, -204 and -1972 positions were identified in upstream region of the CYC-B gene, and on further segregation analysis confirms that -77 SNP was associated with orange fruit color of Hbc mutant. These results concluded that there is a close relationship between the CYC-B gene expression and orange color fruit. Characterization of Hbc mutant gives valuable information to breeders for the development of new varieties, having high β-carotene using molecular markers. The aim of this review is to enumerate medicinal plants and their bioactive compounds that may become potential leads in the mitigation of oral submucous fibrosis (OSMF) in the forthcoming future. It is focused on pathophysiology, risk factors, current treatment regimen, potential plant leads, and future therapies for OSMF. Data were extracted from a vast literature survey by using SciFinder, Web of Science, Google Scholar, and PubMed search engines with relevant keywords. Upon literature survey, we found that the phytochemical 'arecoline' present in the areca nut is the main causative agent of OSMF condition. Currently, OSMF is treated by immunomodulatory and anti‐inflammatory agents such as corticosteroids, enzymes (hyaluronidase, chymotrypsin, and collagenase), anti‐inflammatory mediators (isoxsuprine and pentoxifylline), dietary supplements (vitamins, antioxidants, and micronutrients), and anti‐fibrotic cytokines like interferon‐gamma that provides short‐term symptomatic relief to OSMF patients. However, some plant leads have been proven effective in alleviating symptoms and mitigating OSMF, which ultimately improves the quality of OSMF patients' life. We concluded that plant drugs like lycopene, curcumin, Aloe vera, colchicine, and Glycyrrhiza glabra are effective against OSMF in various in vitro and/or clinical studies and are being used by modern and traditional practitioners. S-nitrosoglutathione reductase (GSNOR) is a well-known regulator in controlling protein S-nitrosylation modification and nitric oxide (NO) homeostasis. Here, a GSNOR inhibitor N6022 and SlGSNOR silencing were applied to investigate the roles of SlGSNOR in tomato fruit postharvest ripening. We found that the application of N6022 and S-nitrosoglutathione (GSNO, a NO donor), and SlGSNOR silencing delayed the transition of fruit skin color by improving total chlorophyll level by 88.57%, 44.78%, and 91.03%, respectively. Meanwhile, total carotenoid and lycopene contents were reduced by these treatments. Concurrently, the activity of chlorophyll biosynthesis enzymes and the expression of related genes were upregulated, and the transcript abundances of total carotenoid bioproduction genes were downregulated, by N6022 and GSNO treatments and SlGSNOR silencing. In addition, fruit softening was postponed by N6022, GSNO, and SlGSNOR silencing, through delaying the decrease of firmness and declining cell wall composition; structure-related enzyme activity; and gene expression levels. Furthermore, N6022, GSNO, and SlGSNOR silencing enhanced the accumulation of titratable acid; ascorbic acid; total phenol; and total flavonoid, but repressed the content of soluble sugar and soluble protein accompanied with the expression pattern changes of nutrition-related genes. In addition, the endogenous NO contents were elevated by 197.55%; 404.59%; and 713.46%, and the endogenous SNOs contents were enhanced by 74.65%; 93.49%; and 94.85%; by N6022 and GSNO treatments and SlGSNOR silencing, respectively. Altogether, these results indicate that SlGSNOR positively promotes tomato postharvest fruit ripening, which may be largely on account of its negative roles in the endogenous NO level. The aim of this work was to give as much information as possible on Rosa canina dried fruit that is commercially available in Serbia. In order to provide the chemical composition, the UHPLC-DAD-ESI-MS method was employed for both polar and non-polar extracts of samples obtained with a solvent mixture consisting of hexane, acetone, and ethanol in a volume ratio of 2:1:1, respectively, and 0.05% (w/v) butylated hydroxytoluene. In addition, the total content levels of lycopene, β-carotene, total polyphenols, and flavonoids were determined by means of UV-vis spectrophotometry. The antioxidant activity was tested by applying four different methods: ABTS, DPPH, FRAP, and CUPRAC. Overall, nine compounds were identified. The results of chemical composition analysis were used as the basis for the interpretation of the calculated results for the antioxidant and antimicrobial activity. The obtained results for R. canina dried fruit extract are as follows: β-carotene—7.25 [mg/100 g fruit weight]; lycopene—2.34 (mg/100 g FW); total polyphenol content (TPC)—2980 [mg GAE/kg FW]; total flavonoid content (TFC)—1454 [mg CE/kg FW]; antioxidant activity—ABTS 12.3 [μmol/100 g FW], DPPH 6.84, FRAP 52.04, and CUPRAC 15,425; and antimicrobial activity—Staphylococcus aureus MIC/MMC 4/0 [mg∙mL−1], Enterococcus faecalis 4/0, Bacillus cereus 4/0, Escherichia coli 4/0, Salmonella enteritidis 4/4, Enteroabacter aerogenes 4/0, Pseudomonas aeruginosa 2/0, and Candida albicans 2/0. Objective: In this study, we analyzed the correlation between the preoperative plasma lycopene levels, postoperative adverse complications of chemoradiotherapy, and nutritional risk scores in patients with laryngeal carcinoma. Methods: A total of 114 patients with laryngeal carcinoma and 114 healthy respondents were enrolled in this study. The patients with laryngeal carcinoma were divided into two groups: 62 patients with laryngeal carcinoma, with an NRS2002 score higher than 3 points and whose diet contained lycopene, were enrolled in the observation group, and 52 patients with laryngeal carcinoma during the corresponding time period, whose diet did not contain lycopene, were enrolled in the reference group. The immune indexes (CD4 + , CD8 + , IGA, IGM, IGG), nutritional indexes (albumin, prealbumin, transferrin), and postoperative adverse complications of chemo-radiotherapy in the two groups were recorded. Results: The lycopene levels were lower in patients with advanced tumor stage (III and IV). The diagnosis threshold of the plasma lycopene level for laryngeal carcinoma was 0.503 μmol/L. The area under the curve for plasma lycopene levels in cancer diagnosis was 0.96, with a clinical specificity of 0.943 and a sensitivity of 0.859. There was a significant negative correlation between the plasma lycopene levels and Nutrition Risk Screening (NRS) 2002 score (R2 = − 0.523, P < 0.001), which was related to the increase in NRS-2002 scores and nutritional hazards in patients with laryngeal carcinoma. The observation group showed a significant increase in nutritional and immune indices, as compared to the reference group, as well as a lower incidence of severe and serious adverse reactions to chemo-radiotherapy. Lycopene supplementation, tumor stage, NRS-2002 scores, nutritional and immune indices were all significant predictors of postoperative severe and serious adverse complications of chemoradiotherapy. Context. The varicocele is the leading cause of male infertility and can impair sperm quality and testicular function through various mechanisms. In our previous study, we found that lycopene could attenuate hypoxia-induced testicular injury. Aims. To illustrate the detailed mechanism of lycopene on spermatocytes. Methods. The effect of lycopene on GC-2 cells under hypoxia were detected by flow cytometry and western blot assay. miR-seq was used to determine miRNA expression in varicocele rat model testes. The function of miR-23a/b were determined by flow cytometry and western blot assay. Key results. We demonstrate that lycopene could alleviate hypoxia-induced GC-2 cell apoptosis and could elevate miR-23a/b expression of the hypoxia model in vivo and in vitro. The miR-23a and -23b mimics could reduce the hypoxia-induced GC-2 cell apoptosis. Both miR-23a and -23b could directly bind with prokineticin 2 (PROK2) mRNA and downregulate its expression. Conclusions. Lycopene could attenuate hypoxia-induced spermatocyte injury through the miR-23a/b–PROK2 pathway. Implications. Background and Aims: The polycystic ovarian syndrome is a menacing metabolic condition with high prevalence rate in Asian countries involving women of reproductive age. This study was commenced to assess favorable effects of Lycopene--a dietary component in letrozole-induced PCOS rat model. Materials and Methods: Fifty adult female Sprague Dawley rats were divided into five groups, ten rats each for a study of 5weeks. Control group A was given standard rat diet. Rats of experimental group B were given 1mg/kg of Letrozole to induce PCO model. Group C was given 1mg/kg of letrozole+15mg/kg/day of lycopene. Group D was given 1mg/kg of letrozole+ 500 mg/kg/day metformin. Group E was given 1mg/kg of letrozole +combination of Lycopene and metformin together. Relative body weight was taken on the day1, day21 and end of experiment. Antioxidant activity was tested using CAT, SOD and MDA. Ovaries were removed for H&E staining and studied under light microscopy. Statical analysis was carried out for result interpretations. Results: The results showed, reduced rat weight by use of Lycopene. Histological architecture of ovary showed better count of secondary and graafian follicles towards normal values which was quite reduced in group B proved by significant p-value<0.001. While raised number of atretic follicles in group B 6±1.6 was reduced to 3±0.9, 4±0.6, 4±0.7 in group C, D &E respectively. Overall improvement in distorted/cystic ovaries was noted by treatment therapy, evident by significant p-value. Group B showed mean count of cyst 7±1.0, which was brought to 2±0.5, 3±0.8, 2±0.7 in groups C, D &E. Elevated Oxidative markers (ROS) were also reduced. Guava (Psidium guajava L.), a member of the family Myrtaceae, is the third richest source of vitamin C after Barbados cherry and aonla. Guava has many medicinal properties associated with every plant part. Coloured guava has additional nutritive value due to its lycopene, anthocyanin, and carotenoid content, which impart colour to its pulp and serve as potential antioxidants. In this review, the various categories of pulp colour in guava, genetics of pulp colour, pigments responsible for various pulp colours, and the biosynthetic pathway of those pigments are presented in detail. The recent advances in guava show that both carotenoids and anthocyanins contribute significantly to its pulp colour. Poultry farmers in the tropics suffer great losses in broiler production especially during the post brooding phase as a result of heat stress occasioned by high ambient temperature. This study therefore, investigated the ameliorative effects of synthetic lycopene dietary supplement on growth performance, slaughter traits and serum corticosterone (CORT) levels of COBB500 broiler chickens reared during heat period. A total of 120 birds were selected at 4th week of rearing and allotted into a control group (C) and three experimental groups designated as T1, T2 and T3. Each group had 3 replicates. The four groups were fed the same starter diet for 4 weeks and then differently on finisher diet supplemented with lycopene at 0mg/kg feed (control), 5mg/kg (T1), 10mg/kg (T2), and 15mg/kg (T3) for 3 weeks. Data obtained were analyzed for statistical differences using ANOVA. The results showed that lycopene supplementation increased feed intake significantly (p<0.05) in the birds under groups T1 and T3. Final body weights and weight gain were statistically similar across the groups but numerically improved in the lycopene-supplemented broilers. While the thigh muscle weight increased significantly (p<0.05) across the groups with increasing supplementation, feed conversion ratio (FCR) decreased optimally (2.03) at 15mg/kg supplementation level leading to improved general performance. The supplement also decreased serum CORT concentration significantly (p<0.05) at 5mg/kg level. The broiler industry supplies high-quality animal protein to the world. The ban of antibiotics as growth promoters has opened the way for plenty of phytochemicals and antioxidants to be explored. This study summarizes the use of natural antioxidants in a broiler diet as a way through which to deal with stressors, as well as their effects on the expression of various genes. The transcriptional factors and genes involved in the regulation of redox homeostasis are described and emphasis is placed on nuclear factor erythroid 2-related factor 2 and nuclear factor kappa B. Sources such as fruits, vegetables, spices, mushrooms, and algae contain numerous natural antioxidant compounds. The antioxidant activity of these compounds has also been confirmed at the genome level. This study focuses on the regulation of oxidative stress-related genes, as well as on genes that regulate the inflammatory response, apoptosis, response to heat stress, lipid metabolism, and the intestinal barrier status. The natural compounds presented include, but are not limited to, the following: rutin, lycopene, magnolol, genistein, hesperidin, naringin, quercetin, curcumin, bisdemethoxycurcumin, resveratrol, astaxanthin, squalene, pterostilbene, protocatechuic acid, taraxasterol, myricetin, and proanthocyanidins. Several studies have revealed a dose-dependent action. The tomato as a raw material for processing is globally important and is pivotal in dietary and agronomic research due to its nutritional, economic, and health significance. This study explored the potential of machine learning (ML) for predicting tomato quality, utilizing data from 48 cultivars and 28 locations in Hungary over 5 seasons. It focused on °Brix, lycopene content, and colour (a/b ratio) using extreme gradient boosting (XGBoost) and artificial neural network (ANN) models. The results revealed that XGBoost consistently outperformed ANN, achieving high accuracy in predicting °Brix (R² = 0.98, RMSE = 0.07) and lycopene content (R² = 0.87, RMSE = 0.61), and excelling in colour prediction (a/b ratio) with a R² of 0.93 and RMSE of 0.03. ANN lagged behind particularly in colour prediction, showing a negative R² value of −0.35. Shapley additive explanation's (SHAP) summary plot analysis indicated that both models are effective in predicting °Brix and lycopene content in tomatoes, highlighting different aspects of the data. SHAP analysis highlighted the models' efficiency (especially in °Brix and lycopene predictions) and underscored the significant influence of cultivar choice and environmental factors like climate and soil. These findings emphasize the importance of selecting and fine-tuning the appropriate ML model for enhancing precision agriculture, underlining XGBoost's superiority in handling complex agronomic data for quality assessment. The surface cleaning of metals plays a pivotal role in ensuring their overall performance and functionality. Dielectric barrier discharge (DBD) plasma, due to its unique properties, has been considered to be a good alternative to traditional cleaning methods. The confinement of DBD plasma in microreactors brings additional benefits, including excellent stability at high pressures, enhanced density of reactive species, reduced safety risks, and less gas and energy consumption. In the present work, we demonstrated a DBD plasma-based method for the degradation of stains from metal surfaces in a microreactor. Aluminum plates with capsanthin stains were used to investigate the influence of operational parameters on the decolorization efficiency, including plasma discharge power, plasma processing time, and O2 content in the atmosphere. The results revealed that an increase in plasma discharge power and plasma processing time together with an appropriate amount of O2 in the atmosphere promote the degradation of capsanthin stains. The optimum processing condition was determined to be the following: plasma power of 11.3 W, processing time of 3 min, and Ar/O2 flow rate of 48/2 sccm. The evolution of composition, morphology, bonding configuration, and wettability of aluminum plates with capsanthin and lycopene stains before and after plasma treatment were systematically investigated, indicating DBD plasma can efficiently degrade stains from the surface of metals without damage. On this basis, the DBD plasma cleaning approach was extended to degrade rhodamine B and malachite green stains from different metals, suggesting it has good versatility. In the present investigation watermelon juice was spray-dried by varying the concentration of maltodextrin (3 and 5%) and inlet temperature (130, 140 and 150 °C). The physicochemical properties of the spray-dried powder of watermelon juice were studied (SWP). It was observed that moisture content, water activity, lycopene content and particle density decreased with increase in maltodextrin concentration and inlet temperature, whereas dissolution increased with increase in maltodextrin concentration and inlet temperature. The colour parameters were also affected by the maltodextrin concentration and inlet temperature. The powder (SWP with 3% maltodextrin and inlet temperature of 140 °C) with desired characteristics was enriched with freeze-dried papaya peel extract (FPP) at six different concentrations (0–2.5% of SWP). The results demonstrated that the total phenolic content increased and antioxidant activity increased when the concentration of FPP was increased. Measurement of photosensitized luminescence of singlet oxygen has been applied to studies of singlet oxygen generation and quenching by C40 carotenoids (neurosporene, lycopene, rhodopin, and spirilloxanthin) with long chain of conjugated double bonds (CDB) using hexafluorobenzene as a solvent. It has been found that neurosporene, lycopene, and rhodopin are capable of the low efficient singlet oxygen generation in aerated solutions upon photoexcitation in the spectral region of their main absorption maxima. The quantum yield of this process was estimated to be (1.5–3.0) × 10−2. This value is near the singlet oxygen yields in solutions of ζ-carotene (7 CDB) and phytoene (3 CDB) and many-fold smaller than in solutions of phytofluene (5 CDB) (Ashikhmin et al. Photogeneration of singlet oxygen was not observed in spirilloxanthin solutions. A correlation was found between the singlet oxygen yields and the quantum yields and lifetimes of the fluorescence of the carotenoid molecules. All carotenoids were shown to be strong physical quenchers of singlet oxygen. The rate constants of 1O2 quenching by the carotenoids with long chain of CDB (9–13) were close to the rate constant of the diffusion-limited reactions ≈1010 M−1 s−1, being many-fold greater than the rate constants of 1O2 quenching by the carotenoids with the short chain of CDB (3–7) phytoene, phytofluene, and ζ-carotene studied in prior papers of our group (Ashikhmin et al. 2020, 2022). To our knowledge, the quenching rate constants of rhodopin and spirilloxanthin have been obtained in this paper for the first time. This study aims to investigate the effects of lycopene on apoptotic, autophagic, and necrotic pathways, oxidative status, and DNA damage in diabetic nephropathy at the molecular level. The sample of the study includes seven groups: lycopene (L), high glucose (G), high glucose + lycopene (GL), and control (C) groups tested at 12 and 24 h. The expression levels of genes in oxidative, apoptotic, autophagic, and necrotic cell death pathways are determined by reverse transcription‐quantitative polymerase chain reaction analysis. The comet assay method is used for the analysis of DNA damage. It is observed that adding lycopene to high glucose for protective purposes reduces the expression of genes related to apoptosis, autophagy, and necrosis, as well as the DNA damage index, compared to cells given high glucose alone. Cardiovascular toxicity includes damage to the heart resulting from inflammation, oxidative stress, and toxin-induced abnormalities in electrophysiology, and muscle damage. The current study set out to investigate the potential cardioprotective properties of lycopene (LCP) and grape seed extract (GSE) as potential novel therapies to counteract the cardiotoxicity caused by salbutamol. Histopathological and immunohistochemical analysis were used to achieve this. Male albino rats weighing between (150_180g b.wt) were randomly divided into six sets, each containing seven rats. Group I received saline and kept as a control group. Group II was given oral administration of salbutamol (65 mg/kg b.wt.) for 2 consecutive days. Group lll received oral dose of LCP (1 mg/kg b.wt.) for three weeks. Group IV was given oral dose of GSE (100 mg/kg b.wt.) for three weeks. Group V was pretreated orally with LCP for three weeks followed by salbutamol oral administration (65 mg/kg b.wt.) for two consecutive doses. Group VI was pretreated orally with GSE (100 mg/kg b.wt.) for three weeks followed by salbutamol oral administration (65 mg/kg b.wt.) for two consecutive doses. All treatments were administered once daily by oral route using gastric tube. Results revealed that salbutamol induced cardiac damage manifested by congestion of myocardial blood vessels, intermuscular hemorrhages, endocardial degenerative changes, microcalcification and subendocardial congestion and hemorrhage, focal cardiomyocytes hyalinization with nuclear peripheralization and pyknosis and individual cellular apoptosis. In comparison with salbutamol group, pre-treatment of rats with GSE demonstrated a moderate ameliorative effect comparable to that of LCP pretreated group; however the residual tissue changes are little bit worse. Immunohistochemistry results supported the histopathological investigations. In conclusion, pre-administration of GSE and LCP provide potential cardioprotective effects by reducing salbutamol-induced cardiac damage linked to alterations in histopathology and immunohistochemistry. Background: Liver diseases are constantly increasing throughout the world and are often associated with other diseases, but above all they are caused by improper diet. Adherence to a diet with abundant vegetables has now been widely demonstrated to be important in combating this pathological condition. The aim of this study was to explore the protective role of lycopene (LYC) extracts from cooked and fresh tomato. Methods: The study cohort included 969 participants assessed in the NUTRIHEP cohort (2005–2006) and the associated follow-up (2014–2016), divided into two groups, based on liver condition: NAFLD, or AFLD and FLD. Results: The results indicated a statistical significance of LYC consumption, showing a protective role against liver disease, the best concentration being 9.50 mg/die, with an RR value of 0.59, p = 0.01, 0.39 to 0.90 at 95% C.I., and RRR = 0.40, p = 0.002, 0.22 to 0.71 at 95% C.I. Conclusions: The protective role of LYC extracts from tomato has not been amply demonstrated in humans. We conclude that this is one of the few papers in the literature to evaluate the protective effect of LYC against liver disease, as well as how this molecule could be used in future possible treatments. Komagataella phaffii, a nonconventional yeast, is increasingly attractive to researchers owing to its posttranslational modification ability, strict methanol regulatory mechanism, and lack of Crabtree effect. Although CRISPR-based gene editing systems have been established in K. phaffii, there are still some inadequacies compared to the model organism Saccharomyces cerevisiae. In this study, a redesigned gRNA plasmid carrying red and green fluorescent proteins facilitated plasmid construction and marker recycling, respectively, making marker recycling more convenient and reliable. Subsequently, based on the knockdown of Ku70 and DNA ligase IV, we experimented with integrating multiple DNA fragments at a single locus. A 26.5-kb-long DNA fragment divided into 11 expression cassettes for lycopene synthesis could be successfully integrated into a single locus at one time with a success rate of 57%. A 27-kb-long DNA fragment could also be precisely knocked out with a 50% positive rate in K. phaffii by introducing two DSBs simultaneously. Finally, to explore the feasibility of rapidly balancing the expression intensity of multiple genes in a metabolic pathway, a yeast combinatorial library was successfully constructed in K. phaffii using lycopene as an indicator, and an optimal combination of the metabolic pathway was identified by screening, with a yield titer of up to 182.73 mg/L in shake flask fermentation. Lycopene is an essential antioxidant found in tomatoes (Solanum lycopersicum L.) that can inhibit and prevent cell damage in the body. Thus, enhancing lycopene content is one of the crucial objectives in tomato breeding. Efforts should focus on assembling the tomato with high fruit yield and good quality by conducting transgressive segregant selection through plant breeding programs. The study aimed to select tomato lines with the potential for transgressive segregation based on high fruit yield and lycopene content. The research began in an augmented design combined with factors comprising 54 non-repeated tomato lines, compared with four repeated standard cultivars (Mawar, Chung, Karina, and Tymoti). The tomato lines' planting into seven blocks had no repeats, while the check cultivars for comparison engaged in repeated planting in each block. The observations underwent the analysis of variance (ANOVA), path analysis, correlation, heritability (narrow sense), and transgressive segregants. Based on transgressive segregation, the high-yielding tomato lines selected bore analysis for lycopene content. Based on the plant height and productive bunches per plant (directly supporting the fruit yield), the 14 promising transgressive segregant families' selection comprised 10 MC crosses, MC10.10, MC10.4, MC11.4, MC12.3, MC27.12, MC27.7, MC29.4, MC35.7, MC74.12, and MC8.3 and four KM crosses, KM23.2, KM26.1, KM30.5, and KM.5.5. For increased lycopene content, the five F4 families MC10.10, MC27.12, MC35.7, KM23.2, and KM30.5 showed better performance wherein selecting 10 transgressive segregant lines resulted with high lycopene content and fruit yield. Fruit yield and quality of greenhouse tomatoes are strongly influenced by light conditions and nitrogen (N) availability, however, the interaction between these factors is still unclear. We evaluated the effects on cherry tomatoes of two tunnel plastic covers with different optical properties and three N doses, also in combination with a biostimulant treatment. We compared a diffuse light film (Film1) and a conventional clear film (Film2), and three N levels, corresponding to 50% (N50), 75% (N75) and 100% (N100) of the optimal dose, with and without a microbial plus a protein hydrolysed biostimulant, compared to a non-treated control. The three experimental treatments significantly interacted on several yield and quality parameters. In control plants (untreated with biostimulants), the early yield was higher at reduced N doses compared to N100, with greater increments under the diffusive Film1 compared to the clear Film2 (+57.7% and +37.0% vs. +31.7% and +16.0%, in N50 and N75 respectively). Film1 boosted the total fruit production at all the N rates and with or without biostimulants, compared to Film2, with stronger effects under sub-optimal N (+29.4% in N50, +21.2% in N75, and +7.8% in N100, in plants untreated with biostimulant). Total yield decreased with decreasing N levels, while it always increased with the application of biostimulants, which counterbalanced the detrimental effects of N shortage. Quality traits were mainly affected by the cover film and the biostimulant treatment. The diffusive film increased the content of carotenoids, lycopene and total phenols compared to the clear one, and the biostimulants increased texture, soluble solids, phenols and ascorbic acid compared to the untreated control. It is worth noting that in plants fertilized at 75% of the reference N dose, the biostimulants determined higher yield than the N100 untreated control, under both the covers (+48% in Film1 and +20% in Film2). In conclusion, the diffusive film improved the fruit yield and quality of greenhouse tomatoes in the spring–summer period, presumably avoiding plant stress due to high-intensity direct light. Reduced N rates limited the plant productivity, however, the biostimulant application was effective in compensating for the detrimental effects of sub-optimal supply of N synthetic fertilizers. Over-accumulating salts in soil are hazardous materials that interfere with the biochemical pathways in growing plants drastically affecting their physiological attributes, growth, and productivity. Soil salinization poses severe threats to highly-demanded and important crops directly challenging food security and sustainable productivity. Recently, there has been a great demand to exploit natural sources for the development of nontoxic nanoformulations of growth enhancers and stress emulators. The chitosan (CS) has growth-stimulating properties and widespread use as nanocarriers, while curcumin (CUR) has a well-established high ROS scavenging potential. Herein, we use CS and CUR for the preparation of CSNPs encapsulating CUR as an ecofriendly nanopriming agent. The hydroprimed, nanoprimed (0.02 and 0.04%), and unprimed (control) wheat seeds were germinated under salt stress (150 mM NaCl) and normal conditions. The seedlings established from the aforementioned seeds were employed for germination studies and biochemical analyses. Priming imprints mitigated the ionic toxicity by upregulating the machinery of antioxidants (CAT, POD, APX, and SOD), photosynthetic pigments (Chl a, Chl b, total Chl, and lycopene), tannins, flavonoids, and protein contents in wheat seedlings under salt stress. It controlled ROS production and avoided structural injuries, thus reducing MDA contents and regulating osmoregulation. The nanopriming-induced readjustments in biochemical attributes counteracted the ionic toxicity and positively influenced the growth parameters including final germination, vigor, and germination index. It also reduced the mean germination time, significantly validating the growth-stimulating and stress-emulating role of the prepared nanosystem. The primary objective is to facilitate the transfer of carotenoids into the juice fraction, yielding carotenoid-enriched juice seamlessly integrable into aqueous-based food matrices. The untreated guava puree exhibited a modest release of carotenoids, with only 66.26% of β-carotene and 57.08% of lycopene reaching the juice. Contrastly, both high-pressure homogenization (HPH) at 25 MPa and enzyme (EM) treatment significantly enhanced carotenoid release efficiency (p < 0.05), while high hydrostatic pressure (HHP) at 400 MPa and pulsed electric field (PEF) of 4 kV/cm did not (p > 0.05). Notably, HPH demonstrated the most substantial release effect, with β-carotene and lycopene reaching 90.78% and 73.85%, respectively. However, the stability of EM-treated samples was relatively poor, evident in a zeta-potential value of −6.51 mV observed in the juice. Correlation analysis highlighted the interactions between pectin and carotenoids likely a key factor influencing the stable dissolution or dispersion of carotenoids in the aqueous phase. Simple Summary: The Food and Agriculture Organization of the United Nations, the World Health Organization, and the Commission on Food Additives recognize lycopene as a Class A nutrient, but relatively few systematic studies have been conducted on lycopene regulation of antioxidant and meat quality improvement and intestine microbiota in poultry. This study investigated the dietary lycopene from the effects on growth performance, antioxidant capacity, meat quality, intestine histomorphology, and cecal microbiota in broiler chickens. Compared with the control group, the dietary lycopene increased the average daily gain and decreased the feed conversion ratio in the experimental groups; the drip loss24h were decreased in breast muscles in the group treated with 20 mg/kg dietary lycopene; the jejunum villous height was increased in the groups treated with 20 mg/kg dietary lycopene; 13,223 OTUs were identified, each representing a microbial species, and the control group; the Unclassified-f-Ruminococcaceae relative abundance was increased in the groups treated with 20 mg/kg dietary lycopene. In this study, the 20 mg/kg dietary lycopene improved the growth performance, antioxidant capacity, meat quality, intestine histomorphology, and cecal microbiota in broiler chickens. We randomly divided five hundred and seventy-six one-day-old male broilers into four groups each with six replicates and 24 chickens in each replicate. The control group (CG) was fed the basal diet, and the other groups were given powder lycopene of 10, 20, and 30 mg/kg lycopene (LP10, LP20, and LP30, respectively). Salmorejo, a Mediterranean tomato-oil puree, is considered a dietary source of antioxidant vitamins C and E and carotenoids lycopene and β-carotene, the latter endowed with provitamin A activity. However, these antioxidants can be degraded in oxidation reactions catalysed or not by enzymes during pasteurisation and storage treatments used to stabilise the salmorejo before consumption. Due to its better penetration, the use of dielectric heating by radiofrequency (RF) may improve results of pasteurisation in this product. The objective was to assess the effects of pasteurisation temperature (70–100 °C, at 5 °C intervals) and storage time (0–5 months, at one-month intervals) on levels of ascorbic acid, α-tocopherol and carotenoids and antioxidant capacity (AC) in salmorejo pasteurised (over 10 s) by conventional (CH) or RF continuous heating. Two successive experiments were conducted to select an adequate pasteurisation temperature for use in the shelf-life study. Pasteurisation upon tested conditions allows a good retention of salmorejo antioxidants. Either CH or RF pasteurisers can be used with similar results. Vitamin C (L-ascorbic + dehydroascorbic acids) was more abundant (15–19 mg 100 g−1) than carotenoids (0.9–2.6 mg 100 g−1) (all-trans + cis lycopene and β-carotene) and α-tocopherol (0.8–1.2 mg 100 g−1) in the pasteurised product. Using excessively low temperatures (70 °C) resulted in partial losses of the three antioxidants, possibly due to oxidase residual activities. Intensifying thermal treatment improved this issue with minor losses of the thermolabile vitamin C and increased carotenoid content. Using a suitable temperature (80 °C) did not prevent most vitamin C from being degraded by the first month, while α-tocopherol, and, to a lesser extent, carotenoids, showed good retention levels during shelf life under refrigeration. Vitamins C and E and carotenoids, either by degradation, regeneration or releasing, likely contribute to the AC in salmorejo. Phenolic antioxidant response, radical-scavenging activities and redox potential values confirmed this finding. The pasteurised-chilled salmorejo shows good antioxidant properties with potential health implications, a positive nutritional aspect for consumers of this tomato-oil homogenate. Background: Erectile dysfunction is a condition with a high incidence among adult men. Lycopene has been shown to lower blood glucose and reduce weight in diabetic patients because of its antioxidant and anti‐inflammatory properties. However, the association between lycopene and the incidence of erectile dysfunction is unclear. Objective: The aim of this study was to examine the dietary lycopene intake and its association with erectile dysfunction risk in the US population. Materials and methods: We investigated the lycopene intake of adult participants with complete information on clinical variables from the National Health and Nutrition Examination Survey between 2001 and 2004. Dose–response curve analysis was applied to explore the association between lycopene intake and erectile dysfunction. Logistic regression models were used to adjust for confounders. Different ethnicities, body mass index level, hypertension status, diabetes status, and smoking status were analyzed as subgroups. Propensity score matching was employed to eliminate the effects of potential confounders to confirm the reliability of the results. Results: A total of 3265 participants with lycopene consumption data were included in our study, including 931 individuals with erectile dysfunction and 2334 without erectile dysfunction during National Health and Nutrition Examination Survey 2001–2004. We found more consumption of lycopene in the non‐erectile dysfunction group than in the erectile dysfunction group. Dose–response curve analysis revealed a significant negative association between lycopene intake and erectile dysfunction prevalence. After adjusting for age, race, cigarette smoking, body mass index, annual family income, education, physical activity, hypertension, diabetes, depression, and testosterone level, we found that increased lycopene intake reduced the odds ratio of erectile dysfunction. Low lycopene intake was positively related to erectile dysfunction in almost all subgroups, especially in Mexican American, non‐Hispanic white, body mass index <25, hypertension positive, diabetes mellitus negative, and smoke negative. Furthermore, the results were confirmed in the 1:1 matched group. Conclusion: Our national data suggest that lower dietary lycopene intake is positively associated with an increased risk of erectile dysfunction in US men. [ABSTRACT FROM AUTHOR]

Summary: The antioxidant and anticancer properties of lycopene make it an excellent ingredient for daily food supplements. The present study developed a novel protocol based on crystallisation to produce lycopene of high purity from a complex extract obtained by n‐hexane from tomato peel. The crystallisation process aimed to achieve a higher recovery and purity of lycopene. The crystallisation system was optimised using ethanol and n‐hexane at a volume ratio of 0.8:1 after evaporating the extract to a solid concentration of 66%. The recovery of lycopene reached 78.45% with a purity of 96.12%. The combination of the two crystallisation methods used, antisolvent and evaporation crystallisation, removed most impurities from the lycopene crystals. Background: Sortilin1 (SORT1) is a ubiquitously expressed transporter involved in sorting or clearing proteins and is pathologically linked to tissue fibrosis and calcification. Targeting SORT1 may have potential clinical efficacy in controlling or reversing cardiovascular fibrosis and/or calcification. Hence, this study assessed the protein–protein network of human SORT1 and its targetability using known nutra-/pharmaceuticals. Material and methods: Network proteins of human SORT1 were identified using the String database, and the affinity of the protein–protein interaction of this network was analysed using Chimera software (Chimera-1.17.3-mac64). The tissue-specific expression profile of SORT1 was evaluated and assessed for enrichment in different cell types, including immune cells. A library of in-house small molecules and currently used therapeutics for cardiovascular diseases were screened using AutoDock Vina to assess the targetability of human SORT1. The concentration affinity (CA) ratio of the small molecules was estimated to assess the clinical feasibility of targeting SORT1. Results: IGF2R, NTRK2, GRN and GGA1 were identified as high-affinity interaction networks of SORT1. Of these high-affinity interactions, IGF2R and GRN can be considered relevant networks in regulating tissue fibrosis or the microcalcification process due to their influence on T-cell activation, inflammation, wound repair, and the tissue remodelling process. The tissue cell-type enrichment indicated major expression of SORT1 in adipocytes, specialised epithelial cells, monocytes, cardiomyocytes, and thyroid glandular cells. The binding pocket analysis of human SORT1 showed twelve potential drug interaction sites with varying binding scores (0.86 to 5.83) and probability of interaction (0.004 to 0.304). Five of the drug interaction sites were observed to be targetable at the therapeutically feasible concentration of the small molecules evaluated. Empagliflozin, sitagliptin and lycopene showed a superior affinity and CA ratio compared to established inhibitors of SORT1. Conclusion: IGF2R and GRN are relevant networks of SORT1, regulating tissue fibrosis or the microcalcification process. SORT1 can be targeted using currently approved small-molecule therapeutics (empagliflozin and sitagliptin) or widely used nutraceuticals (lycopene), which should be evaluated in a randomised clinical trial to assess their efficacy in reducing the cardiac/vascular microcalcification process. Predictability and robustness are challenges for bioproduction because of the unstable intracellular synthetic activities. With the deeper understanding of the gene expression process, fine-tuning has become a meaningful tool for biosynthesis optimization. This study characterized several gene expression elements and constructed a multiple inducible system that responds to ten different small chemical inducers in halophile bacterium Halomonas bluephagenesis. Genome insertion of regulators was conducted for the purpose of gene cluster stabilization and regulatory plasmid simplification. Additionally, dynamic ranges of the multiple inducible systems were tuned by promoter sequence mutations to achieve diverse scopes for high-resolution gene expression control. The multiple inducible system was successfully employed to precisely control chromoprotein expression, lycopene and poly-3-hydroxybutyrate (PHB) biosynthesis, resulting in colorful bacterial pictures, optimized cell growth, lycopene and PHB accumulation.
