117. Effect of Bacillus subtilis and Lactobacillus plantarum on solid‐state fermentation of soybean meal.

Abstract

BACKGROUND: As feeding probiotics, Bacillus subtilis FJAT‐4842 and Lactobacillus plantarum FJAT‐13737 were employed to improve the nutritional value and reduce the risk of contamination in the solid‐state fermentation of soybean meal (SBM). RESULTS: After fermentation with bacterium starters, the crude protein, free amino acid and lactic acid increased, with higher protease and cellulose activity. The addition of L. plantarum could increase the crude protein and lactic acid content by 50.1% and 94.9%, respectively. After fermentation, the contents of crude fiber and phytic acid decreased by 45.9% and 48.1%. Compared with control treatment, the addition of both B. subtilis FJAT‐4842 and L. plantarum FJAT‐13737 could effectively elevate the production of free amino acid and esters. Moreover, the addition of bacterium starter could prevent the production of mycotoxin and promote bacterial diversity of the fermented SBM. Especially, the addition of B. subtilis could reduce the relative content of Staphylococcus. After fermentation for 7 days, lactic acid bacteria including Pediococcus, Weissella and Lactobacillus became the main bacteria in the fermented SBM. 119. Re-evaluating the Contribution of a Fe-Based Current Collector to Bioelectrochemical Methanogenesis: Role and Mechanisms

Abstract

The metal-based current collector has been adopted as an essential component of cathodes for electron delivery in microbial electrosynthesis (MES) cells, while the effect of its corrosion on biofilm development and electromethanogenesis activity was overlooked. In this study, the corrosion of the Fe-based current collector was identified to in situ decorate cathode naturally which substantially boosted the performance of CO2electromethanogenesis in terms of taking over two-thirds less time starting up MES and increasing the CH4production rate by 3.5 times. Despite the low concentration of Fe (0.13 at%), the electrochemical analysis indicated that it was possible for these Fe deposits to act as electron shuttles and catalysts for H2production to benefit methanogenesis. The Fe aggregates weakened the dependence of methanogens on electroactive bacteria (EABs) to conduct methanogenesis via interspecies electron transfer as the proportion of EABs on Bio FeCF (with Fe current collector, where CF is carbon felt) was only 25.5% of that on Bio CF (without Fe current collector). On the contrary, the abundance of genes encoding the proteins to uptake extracellular electrons of methanogens on Bio FeCF was 2.3 times higher than that on Bio CF. The enhanced energy transfer maintained high amounts of methanogens and live microorganisms. This study comprehensively explored the multiple roles of Fe-based current collectors in enhancing CO2electromethanogenesis.

122. Effects of a Mixture of Brevibacillus brevis with Other Bacillus sp. Strains against Gray Mold and on Enzyme Activities of Grape.

Abstract

Gray mold disease, caused by Botrytis cinerea, has reduced grape's output and market quality globally. In this study, the antifungal activity of a mixed microbial combination of Brevibacillus brevis FJAT-0809-GLX and Bacillus sp. strains was investigated. The results showed that the inhibition rate against B. cinerea was 85.10% when B. brevis FJAT-0809-GLX was mixed with a mixture of B. brevis FJAT-10623 and Bacillus velezensis FJAT-55034 at a proportion of 80%:20%, which was significantly higher than those of other combination proportions. The inhibitory rates of the mixed microbial combination diluted 0 times, 10 times, and 100 times were 89.14%, 88.10%, and 86.33%, respectively, with no significant differences between each other. The mixed microbial combination appeared to be temperature-insensitive and significantly stable from pH 3 to pH 7. Furthermore, it was discovered that its antifungal activity was significantly stable with UV radiation for 30 min, 60 min, and 90 min, with values of 84.82%, 83.89%, and 82.22%, respectively. An amount of 0.025 mol mL−1 of KCl, CuSO4, and MgCl2 had no effects on the antifungal activity of the mixed microbial combination, while 0.025 mol mL−1 of NaCl, ZnSO4, FeSO4, CaCl2, and MnSO4 reduced the inhibition rate. The mixed microbial combination demonstrated antifungal activities against a variety of fungi, with inhibition rates ranging from 68.78% to 85.10%. The grape fruits and grape leaves treated with the mixed microbial combination decayed at 27.27% and 48.34%, respectively. Additionally, the mixed microbial combination improved grape fruit resistance by increasing the activity of defense enzymes polyphenol oxidase (PPO) and catalase (CAT). Abstract

This article develops a computationally efficient fault-detection method for power systems, by exploiting phasor measurement unit (PMU) data and Hessian locally linear embedding (HLLE) technique. First, via HLLE technique, high-dimensional PMU data are transformed into low-dimensional embedding coordinates, which effectively captures the fluctuations of PMU data. Next, based on the feature space of embedding coordinates, T-squared statistic is employed for online fault detection. The method is evaluated on IEEE 39-bus system and a real-world system, exhibiting decent fault-detection performance as well as a considerably lower computational complexity when compared with existent methods.

134. Identification of lncRNA Signature of Tumor-Infiltrating T Lymphocytes With Potential Implications for Prognosis and Chemotherapy of Head and Neck Squamous Cell Carcinoma

Abstract

Purpose: We systematically analyzed HNSCC-infiltrating T lymphocytes lncRNAs (HILTlncRNAs) to assess their predictive value for the survival outcome and immunotherapy response of patients with anti-programmed death-1 (PD-1) therapy and to evaluate their predictive power to chemotherapeutic agents.Methods: HNSCC transcriptome and clinical information was obtained from The Cancer Genome Atlas (TCGA) database. Immunocell microarray data were obtained from the Gene Expression Omnibus (GEO) database. T-cell-specific lncRNAs were identified by differential expression analysis. Prognostic paired HILTlncRNAs (PHILTlncRNAs) were filtered and modeled by univariate cox, lasso and multivariate cox regression analysis. To construct lncRNA-miRNA-mRNA competitive endogenous RNA (ceRNA) regulatory networks, differentially expressed mRNAs in HNSCC patients were incorporated, microRNAs and differentially expressed mRNAs interacting with T-cell-specific lncRNAs were filtered out based on miRcode, miRDB, miRTarBase, and TargetScan databases.Results: 75 T-cell-specific lncRNAs and 9 prognostic PHILTlncRNAs were identified. Low-risk HNSCC patients had a better prognosis and significant immune cell infiltration, driving the immune response. Differential expression of RNA-binding proteins (RBPs), PD-1 and programmed cell death 1 ligand 1 (PD-L1) was demonstrated in the high and low risk groups of HNSCC patients. In the high risk group, high expression of PD-1 improved patient prognosis, whereas the opposite was observed in the low-risk group. The promoter methylation levels of two RBPs (DNMT1 and ZC3H12D) were decreased in HNSCC patients compared with normal samples, their expression levels were positively correlated with PD-1 and PD-L1 levels and T-cell infiltration. Finally, we screened the sensitivity of HNSCC patients to chemotherapeutic agents and found it differed between high and low risk groups.Conclusion: HILTlncRNAs provided a theoretical basis for immune targeted therapy and drug development.
