104. Identification of turbulence structures above a forest canopy using a wavelet transform

Abstract

The wavelet transform is used to identify scales of large coherent structures present in atmospheric turbulence above the subarctic forest at Schefferville. Individual coherent structures contributing to much of the exchange between the forest and the atmosphere are depicted in terms of both scale and location using contour diagrams of wavelet transform coefficients. Three typical case studies of turbulence and flux observations were selected to examine the physical characteristics of these flux-filled events and their evolution with distance away from the forest canopy. A wavelet transform spectral technique is applied to vertical velocity, temperature, and turbulent heat flux data observed over the sparse coniferous forest to extract the relative importance of each scale present in those data series. The scale of turbulence structures in relation with their characteristic spacing is discussed.

113. Footprint prediction of scalar fluxes - Reliability and implications for airborne flux measurements over the FIFE site

Abstract

Estimates of the location and extension of the upwind ground area that affects flux observations most directly are examined to determine the reliability of airborne versus near-ground flux measurements. The theoretical issues regarding the 'footprint' are examined, and specific observations are analyzed by studying the data over a grid regarding sensible heat, latent heat, CO2, and greenness. The grid is footprint-corrected to correlate better with independently observed surface characteristics, and an optimized footprint is developed that satisfies the relationships between the observed variables. Optimized mapping of the surface flux is given which demonstrates the importance of considering local advection to correlate airborne and ground-based flux observations. The technique is particularly applicable to situations in which significant variations in the surface flux density exist.

Abstract

International audience; Sandy islets (" Caye ") of New Caledonia have a major role in the lagoonal ecosystems and a high importance in the Caledonian culture and way of life. One of the recurrent issues for people and governmental agencies concerns the islets's future in the perspective of Climate Change and Sea Level Rise. In this context, the Coastal Observatory of New Caledonia (OBLIC) has initiated research about the recent and present evolution of islets in order to predict their " behavior " and their future. The past evolution trends at pluri-decadal scale as well as present behaviors are highly variable from one islet to another from slight accretion to huge erosion and land loss (fig. 1) which endanger the islets. Figure 1: Coastal erosion (Petit Ténia islet) Analysis and observations show that six main stages constitute the life-cycle of Caledonian islets, namely: nucleation, growing, maturity, decay, relic and endangered. Changes of environmental parameters act as forcing factors which can drive the islets evolution from one stage to another. The becoming of each islet is linked to its geomorphology, its location within the lagoon, its past evolution, its present state and future evolution of environmental parameters. Abstract

Objective. Individuals with obesity and type 2 diabetes differ from lean and healthy individuals in their abundance of certain gut microbial species and microbial gene richness. Abundance of Akkermansia muciniphila, a mucin-degrading bacterium, has been inversely associated with body fat mass and glucose intolerance in mice, but more evidence is needed in humans. The impact of diet and weight loss on this bacterial species is unknown. Our objective was to evaluate the association between faecal A. muciniphila abundance, faecal microbiome gene richness, diet, host characteristics, and their changes after calorie restriction (CR). Design. The intervention consisted of a 6-week CR period followed by a 6-week weight stabilisation diet in overweight and obese adults (N=49, including 41 women). Faecal A. muciniphila abundance, faecal microbial gene richness, diet and bioclinical parameters were measured at baseline and after CR and weight stabilisation. Results. At baseline A. muciniphila was inversely related to fasting glucose, waist-to-hip ratio and subcutaneous adipocyte diameter. Subjects with higher gene richness and A. muciniphila abundance exhibited the healthiest metabolic status, particularly in fasting plasma glucose, plasma triglycerides and body fat distribution. Individuals with higher baseline A. muciniphila displayed greater improvement in insulin sensitivity markers and other clinical parameters after CR. These participants also experienced a reduction in A. muciniphila abundance, but it remained significantly higher than in individuals with lower baseline abundance. A. muciniphila was associated with microbial species known to be related to health. Conclusions. A. muciniphila is associated with a healthier metabolic status and better clinical outcomes after CR in overweight/obese adults. The interaction between gut microbiota ecology and A. muciniphila warrants further investigation.

Abstract

Background: Germline mutation screening of BRCA1 and BRCA2 genes is performed in suspected familial breast cancer cases, but a causative mutation is found in only 30% of patients. The development of additional methods to identify good candidates for BRCA1 and BRCA2 analysis would therefore increase the efficacy of diagnostic mutation screening. With this in mind, we developed a study to determine molecular signatures of BRCA1—or BRCA2—mutated breast cancers. Materials and Methods: Array-cgh (comparative genomic hybridization) and transcriptomic analysis were performed on a series of 103 familial breast cancers. The series included 7 breast cancers with a BRCA1 mutation and 5 breast cancers with a BRCA2 mutation. The remaining 91 cases were obtained from 73 families selected on the basis of at least 3 affected first-degree relatives or at least 2 affected first-degree relatives with breast cancer at an average age of 45 years. Array-cgh analyses were performed on a 4407 BAC-array (CIT-V8) manufactured by IntegraGen. Transcriptomic analyses were performed using an Affymetrix Human Genome U133 Plus 2.0 chip. Results: Using supervised clustering analyses we identified two transcriptomic signatures: one for BRCA1-mutated breast cancers consisting of 600 probe sets and another for BRCA2-mutated breast cancers also consisting of 600 probes sets. We also defined cgh-array signatures, based on the presence of specific genomic rearrangements, one for BRCA1-mutated breast cancers and one for BRCA2-mutated breast cancers. Conclusions: This study identified molecular signatures of breast cancers with BRCA1 or BRCA2 germline mutations. Genes present in these signatures could be exploited to find new markers for such breast cancers. We also identified specific genomic rearrangements in these breast cancers, which could be screened for in a diagnostic setting using fluorescence in situ hybridization, thus improving patient selection for BRCA1 and BRCA2 molecular genetic analysis.

Abstract

International audience; Context and problematic: On July 7th 2008, UNESCO included parts of Lagoons of New Caledonia in the list of the Reef Diversity and Associated Ecosystems. Sandy islets from New Caledonian lagoons lie on lagoonal reef platforms or on the reef barrier.They have a major role in these specific and rich ecosystems, being the seat of nesting for turtles, seabirds, sea kraits etc. In addition, islets have a high importance in the Caledonian culture and way of life; economy of the islands has been developed around tourism and services with specific activities such as sports and relaxation. One of the recurrent issues for people and governmental agencies concerns the islets's future in the perspective of Climate Change and Sea Level Rise. In this context, the Coastal Observatory of New Caledonia (OBLIC) has initiated research about the recent and present evolution of islets in order to predict their " behavior ". Method Around twenty islets from the southwestern and eastern lagoons of Grande-Terre (New Caledonia main island) and one from Nokanhui atoll (Ile des Pins) have been studied. Such studies integrate field work and observations (erosion scars, accretion area, sedimentology, etc.) and analysis of historical photographs and satellites images. Geomorphological and sedimentological data have been collected during 2013 and 2014 field surveys. Past extensions of each islet have been interpreted using available aerial views and satellite sensing. Old aerial photographs are rather rare because islets being sometimes quite away from the shore, they were not covered by aerial survey of the main island. All available data have been integrated in a GIS. Thus, islets's reconstitution extends from one decade to 70 years. Time evolutions of shape and surface of each islet have been mapped in order to compute surface changes and the present percentage of coast's lengths in erosion, in accretion or stable. Moreover, the forcing factors such as winds, wave, tropical storms or cyclone, and the ENSO have been analyzed, in order to understand potential links with the islet behavior. Results In terms of size and shape, a high diversity and a variety of evolution trends of islets during the past decades can be noticed. Actually, processes affecting the coast of each islet (erosion, accretion, stability) are highly variable. All islets have at least 50% of their coasts affected by erosion and for four this rate is close to 100%. Islets showing increasing surface during the past years are very rare. Analysis and observations show that five main stages constitute the life-cycle of Caledonian islets, namely: nucleation, growing, maturity, decay, relic or endangered. Changes of environmental parameters and forcing factors as well as the inherited geomorphology lead the islets from one stage to another. The becoming of each islet is linked to its past evolution, its present state and future evolution of environmental parameters. Parameters are linked to the climate variability like ENSO or IPO which control the intensity and direction of trade winds and the average sea level in the SW Pacific. Forcing factors include also extreme events like cyclones, storms and austral swells which can trigger at very short term powerful erosion or accretion with high impact on the islet. Sea level rise induced by the anthropogenic climate change has also to be taken into account. Abstract

In the rat and the rabbit, a number of studies have reported the effects of angiotensin II (ANG II) on [Na.sup.+] reabsorption by the proximal (PT) and distal (DT) convoluted tubules of the kidney. The aim of the present study was to examine the effect of ANG II on [Ca.sup.2+] uptake by the luminal membranes of the PT and DT of the rabbit. Incubation of PT and DT with [10.sup.-12] M ANG II enhanced the initial [Ca.sup.2+] uptake in the two segments. Dose-response experiments revealed, for [Ca.sup.2+] as well as for [Na.sup.+] transport, a biphasic action with a maximal effect at [10.sup.-12] M. [Ca.sup.2+] transport by the DT luminal membrane presents a dual kinetic. ANG II action influenced the high-affinity [Ca.sup.2+] channel, increasing maximal velocity from 0.72 [+ or -] 0.03 to 0.90 [+ or -] 0.05 pmol [multiplied by] [micro][g.sup.-1] [multiplied by] 10 [s.sup.-1] (P [is less than] 0.05, n = 3) and leaving the Michaelis-Menten constant unchanged. The effect of ANG II was abolished by losartan, suggesting that the hormone is acting through [AT.sub.1] receptors. In the PT, calphostin C inhibited the effect of the hormone. It is therefore probable that protein kinase C is involved as a messenger. In the DT, however, neither Rp cAMP, calphostin C, nor econazole (a phospholipase A inhibitor) influenced the hormone action. Therefore, the mechanisms involved in the hormone action remain undetermined. Finally, we questioned whether ANG II acts in the same DT segment as does parathyroid hormone on [Ca.sup.2+] transport. The two hormones increased [Ca.sup.2+] transport, but their actions were not additive, suggesting that they both influence the same channels in the same segment of the distal nephron, i.e., the segment responsible for the high-affinity calcium channel. renal calcium transport

Abstract

Les activités décrites dans cet article ont été menées dans le cadre des projets Interreg successifs intitulés « ECOLIRI » et « ECOLIRIMED »*. Elles avaient pour but de contribuer à l’application du génie végétal pour résoudre des problèmes purement hydrauliques rencontrés avec les cours d’eau non navigables de la région transfrontalière comportant la Wallonie, la Lorraine française et le Grand-Duché de Luxembourg, en ciblant la stabilisation des berges, la sauvegarde de la biodiversité et le maintien de la qualité des eaux de surface. Des écotypes d’aulne glutineux, de saules et de frênes ont été rassemblés à partir de prospections menées sur les berges de rivières faisant partie des bassins du Rhin, de la Meuse et, dans une moindre mesure, de l’Escaut, constituant ainsi des variétés multiclonales d’écotypes ligneux. Si le frêne a été rapidement abandonné suite à l’émergence de la chalarose, une filière de production d’aulnes et de saules a été mise en œuvre. Elle repose sur l’existence de parcs à bois, la mise au point de conditions de bouturage, d’élevage et de conservation optimisées des plants ainsi que sur des règles de gestion et de diffusion du matériel végétal; l’objectif étant de permettre un approvisionnement régulier et fiable, en plants destinés aux programmes de renaturation des ripisylves et de stabilisation durable des berges de cours d'eau. La multiplication des écotypes est conduite à partir de parcs à bois gérés par des institutions publiques et entretenus selon les pratiques horticoles qui maintiennent la capacité rhizogène du matériel végétal. Traités lorsque nécessaire en vue d’une bonne reprise, les plants sont délivrés en fonction des conditions d’accessibilité aux sites de végétalisation. Bien que la qualité génétique reste inexplorée, la valeur phénotypique (port, vigueur) est connue. Outre l’origine certifiée de tous les clones, certains aulnes ont été repérés pour leur tolérance au dépérissement causé par Phytophthora alni alors que d’autres aulnes et saules étaient notés pour leur tolérance aux éléments-traces métalliques à la suite d’évaluations spécifiques réalisées en complément. Plusieurs aulnes issus de graines collectées sur des arbres sans symptôme en site infecté par P. alni ou produits in vitro pour induire des propriétés de phytoremédiation par variation somaclonale, ont été intégrés aux collections représentatives de la diversité naturelle. Dans ces conditions, le matériel végétal proposé doit pouvoir s’adapter naturellement aux risques biologiques prévisibles pour une durabilité satisfaisante des travaux impliquant les techniques végétales non seulement pour la renaturation de berges, mais aussi, la végétalisation de zones humides ou écologiquement équivalentes. Il contribuera à valoriser les paysages et à préserver, voire même à améliorer l’équilibre des écosystèmes de la région. La réimplantation de ces essences complémentaires doit permettre, à moyen terme, de mieux raisonner la gestion des phénomènes d’érosion et d’inondation.

Abstract

International audience; Our knowledge of species and functional composition of the human gut microbiome is rapidly increasing, but it is still based on very few cohorts and little is known about variation across the world. By combining 22 newly sequenced faecal metagenomes of individuals from four countries with previously published data sets, here we identify three robust clusters (referred to as enterotypes hereafter) that are not nation or continent specific. We also confirmed the enterotypes in two published, larger cohorts, indicating that intestinal microbiota variation is generally stratified, not continuous. This indicates further the existence of a limited number of well-balanced host-microbial symbiotic states that might respond differently to diet and drug intake. The enterotypes are mostly driven by species composition, but abundant molecular functions are not necessarily provided by abundant species, highlighting the importance of a functional analysis to understand microbial communities. Although individual host properties such as body mass index, age, or gender cannot explain the observed enterotypes, data-driven marker genes or functional modules can be identified for each of these host properties. For example, twelve genes significantly correlate with age and three functional modules with the body mass index, hinting at a diagnostic potential of microbial markers.
