Aspergillus flavus is a filamentous fungus which is widespread on agricultural products and also able to cause various human diseases. This species is frequently isolated from indoor air as well, furthermore, it is known as a common causal agent of keratomycosis, particularly in subtropical and tropical areas. It is also able to produce aflatoxins, one of the most carcinogenic mycotoxins which are harmful to animals and humans. In this study, 59 A. flavus isolates from four different habitats and 1 A. minisclerotigenes isolate were investigated. The isolates were identified and confirmed at the species level by the sequence analysis of a part of their calmodulin gene. Applying a combined analysis of UP-PCR, microsatellite, and calmodulin sequence data, the four group of isolates formed separate clusters on the phylogenetic tree. Examining the distribution of mating type genes MAT1-1 and MAT1-2, a ratio of approximately 3:1 was determined, and no correlation was found between the carried mating type gene and the aflatoxin production capability. HPLC analysis revealed that none of the examined isolates collected from indoor air or maize in Central Europe were able to produce aflatoxins, while about half of the isolates from India produced these mycotoxins under the test conditions. Influence of edaphic factors and metal content on diversity of Trichoderma species at 14 different soil sampling locations, on two depths, was examined. Forty-one Trichoderma isolates from 14 sampling sites were determined as nine species based on their internal transcribed spacer (ITS) sequences. Our results indicate that weakly alkaline soils are rich sources of Trichoderma strains. Also, higher contents of available K and P are connected with higher Trichoderma diversity. Increased metal content in soil was not inhibiting factor for Trichoderma species occurrence. Relationship between these factors was confirmed by locally weighted sequential smoothing (LOESS) nonparametric smoothing analysis. Trichoderma strain (Szeged Microbiology Collection (SZMC) 22669) from soil with concentrations of Cr and Ni above remediation values should be tested for its potential for bioremediation of these metals in polluted soils. Introduction: Human infections caused by Aspergillus brasiliensis have not yet been reported. We describe the first two known cases of fungal keratitis caused by Aspergillus brasiliensis.Case Presentations: A 49-year-old Indian Tamil woman agricultural worker came with pain and defective vision in the right eye for one month. Meanwhile, a 35-year-old Indian Tamil woman presented with a history of a corneal ulcer involving the left eye for 15 days. The fungal strains isolated from these two cases were originally suspected to belong to Aspergillus section Nigri based on macro- and micromorphological characteristics. Molecular identification revealed that both isolates represent A. brasiliensis.Conclusion: The two A. brasiliensis strains examined in this study were part of six keratitis isolates from Aspergillus section Nigri, suggesting that this recently described species may be responsible for a significant proportion of corneal infections caused by black Aspergilli. The species composition of Trichodernia isolates from the rhizosphere of different vegetables collected at different locations in Hungary was examined during this study. Trichoderma strains were isolated from the rhizosphere samples on dichloran-rose bengal medium. After puriflcalion of genomic DNA, the PCR amplification of the internal transcribed spacer (ITS1-5.8S rDNA-ITS2) region and its sequence analysis were used for the identification of the isolates at the species level. Altogether, 45 Trichoderma isolates were identified from the examined samples. The detected Trichoderma species were T. asperellum, T. atroviride, T. citrinoviride, T. gamsii, T. hamatum, T. harzianum, T. koningiopsis/T. ovalisporum, T. longibrachiatum/H. onentalis, T. pleuroticola and T. virens. Besides species known as opportunistic pathogens of humans (T. longibrachiatum / H. orientalis, T. citrinoviride) or as causative agents of mushroom green mould disease (T. pleuroticola), beneficial taxa (T. harzianum, T. virens, T. atroviride) widely used for the biological control of plant pathogenic fungi could also be identified in the examined samples, suggesting that the rhizosphere of vegetables may be a rich source of potential biocontrol agents. In vitro antagonism was examined in dual culture tests and the Biocontrol Index (BCI) values were determined for the particular isolates. Certain T. asperellum, T. virens and T. atroviride isolates proved to possess good in vitro antagonistic activities against plant pathogenic Fusarium solani and F. oxyspo rum strains, suggesting that they might be promising for the development of Trichoderma-based biocontrol strategies for the suppression of plant pathogenic fungi in the rhizosphere of vegetables produced in organic farmland soils. Certain species of the filamentous fungal genus Trichoderma (e.g. Trichoderma longibrachiatum and Trichoderma citrinoviride) are among the emerging clinical pathogens and also the most common species in the indoor space of mould-damaged buildings. The molecules involved in its pathology are not known. In the present study, we report that 0.5-2.6 wt% of the T. longibrachiatum mycelial biomass consisted of thermostable secondary metabolites mitochondriotoxic to mammalian cells. These were identified by LC/ MS as one 11-residue and eight 20-residue peptaibols, AcAib-Asn-Leu/Ile-Leu/Ile-Aib-Pro-Leu/Ile-Leu/Ile-Aib-Pro-Leuol/Ileol (1175 Da) and AcAib-Ala-Aib-Ala-Aib-Ala/Aib-Gln-Aib-Val/Iva-Aib-Gly-Leu/Ile-Aib-Pro-Val/Iva-Aib-Val/Iva/Aib-Gln/Glu-Gln-Pheol(1936-1965 Da) (Aib, α-aminoisobutyric acid; Ac, acetyl; Ileol, isoleucinol; Iva, isovaline; Leuol, leucinol; Pheol, phenylalaninol). The toxic effects on boar sperm cells depended on these peptaibols, named trilongins. The trilongins formed voltage dependent, Na+/ K+ permeable channels in biomembranes. The permeability ratios for Na+ ions, relative to K+, of the 11-residue trilongin channel (0.95 : 1) and the 20-residue trilongin channel (0.8 : 1) were higher than those of alamethicin. The combined 11-residue and 20-residue trilongins generated channels that remained in an open state for a longer time than those formed by either one of the peptaibols alone. Corresponding synergy was observed in toxicokinetics. With 11-residue and 20-residue trilongins combined 1 : 2 w/w, an effective median concentration ( EC50) of 0.6 μg· mL−1 was reached within 30 min, and the EC50 shifted down to 0.2 μg· mL−1 upon extended exposure. By contrast, with 11-residue or 20-residue trilonging separately in 30 min of exposure, the EC50 values were 15 and 3 μg· mL−1, respectively, and shifted down to 1.5 and 0.4 μg· mL−1 upon extended exposure. Sheath blight caused by Rhizoctonia solani is one of the most serious rice diseases worldwide. The disease is currently managed only by the excessive application of chemical fungicides which are toxic and not environmentally friendly. Therefore, greater emphasis should be given to biological control as being both safe and effective. Trichoderma species are ubiquitous fungi in the soil and have an antagonistic activity against several soil-borne plant pathogens including R. solani. The present study was undertaken to evaluate the potential of indigenous Trichoderma strains from Mazandaran province, Northern Iran (a Mediterranean region on the southern coast of the Caspian Sea) against R. solani AG1-IA in vitro, and against sheath blight disease in the glasshouse, in order to find biocontrol isolates for application in the field. More than 200 Trichoderma strains were isolated from the soil, plant debris and the phyllosphere in rice fields. Strains were first screened for their antagonism to R. solani by in vitro antagonism tests including dual culture, antibiosis, the effect of Trichoderma strains on the production and viability of R. solani sclerotia, and hyperparasitism on microscopic slides. According to the in vitro experiments, several strains belonging to T. harzianum, T. virens and T. atroviride showed excellent biocontrol. These potential antagonist strains were further evaluated for their effectiveness in controlling sheath blight under glasshouse conditions. Among the 55 selected strains, seven significantly controlled the disease. T. harzianum AS12-2 was the most effective strain in controlling rice sheath blight, better even than propiconazole, the most commonly used fungicide in Iran. [ABSTRACT FROM AUTHOR]

Green mold of Pleurotus ostreatus, caused by Trichoderma species, has recently resulted in crop losses worldwide. Therefore, there is an emerging need for rapid means of diagnosing the causal agents. A PCR assay was developed for rapid detection of Trichoderma pleurotum and Trichoderma pleuroticola, the two pathogens causing green mold of P. ostreatus. Three oligonucleotide primers were designed for identifying these species in a multiplex PCR assay based on DNA sequences within the fourth and fifth introns in the translation elongation factor 1α gene. The primers detected the presence of T. pleurotum and/or T. pleuroticola directly in the growing substrates of oyster mushrooms, without the need for isolating the pathogens. The assay was used to assess the presence of the two species in natural environments in which P. ostreatus can be found in Hungary, and demonstrated that T. pleuroticola was present in the growing substrates and on the surface of the basidiomes of wild oyster mushrooms. Other Trichoderma species detected in these substrates and habitats were Trichoderma harzianum, Trichoderma longibrachiatum and Trichoderma atroviride.
