Abstract

Antimicrobial peptides (AMPs) constitute promising alternatives to classical antibiotics for the treatment of drug-resistant infections, which are a rapidly emerging global health challenge. However, our understanding of the structure-function relationships of AMPs is limited, and we are just beginning to rationally engineer peptides in order to develop them as therapeutics. Here, we leverage a physicochemical-guided peptide design strategy to identify specific functional hotspots in the wasp-derived AMP polybia-CP and turn this toxic peptide into a viable antimicrobial. Helical fraction, hydrophobicity, and hydrophobic moment are identified as key structural and physicochemical determinants of antimicrobial activity, utilized in combination with rational engineering to generate synthetic AMPs with therapeutic activity in a mouse model. We demonstrate that, by tuning these physicochemical parameters, it is possible to design nontoxic synthetic peptides with enhanced sub-micromolar antimicrobial potency in vitro and anti-infective activity in vivo. We present a physicochemical-guided rational design strategy to generate peptide antibiotics.

Inclusion body nephropathy (IBN) and kidney fibrosis in aged immunodeficient mice and, to lesser extent, in immunocompetent mice have been recently linked to infection of mouse kidney parvovirus (MKPV), also known as murine chapparvovirus (MuCPV). Knowledge about its prevalence and the complete genome sequence of more MKPV strains is essential for understanding phylogenetic relationships and pathogenicity among MKPV strains. In the present study using PCR and genome walking, we determined the complete 4440-nucleotide genome of a new MKPV strain, namely MIT-WI1, which was identified in IBN-affected Il2rg-/-Rag2-/- c-Kit W-sh/W-sh mice housed in the vivarium at Whitehead Institute for Biomedical Research (WI). The overall nucleotide (>94%) and deduced amino acid sequences (>98%) of p10, p15, NS1 (replicase), NS2 and VP1 (capsid protein) within the MIT-WI1 genome, are closely related to MKPV/MuCPV strains described in laboratory and wild Mus musculus mice. In addition, PCR and qPCR assays using newly designed primers conserved among the known MKPV/MuCPV genomes were developed and utilized to assess MKPV status in selected mice. MKPV was also detected in immunodeficient (NSG) and immunocompetent (Crl:CD1(ICR), UTXflox) mouse strains/stocks. The abundance of the MKPV genome copies was significantly correlated with the severity of IBN. Our data indicate that MKPV is present in selected mouse strains/stocks, and provides new insights into the genome evolution of MKPV.

Abstract

This article presents findings from a study on the impact of a child's death on parents. We explored the prominence and adaptiveness of parents' continuing bonds expressions, psychological adjustment, and grief reactions. A qualitative case study methodology was used to describe six cases. Participants were classified into two groups based on scores on the Inventory of Complicated Grief. Commonalities in themes on the Continuing Bonds Interview and projective drawings were assessed. Those in the Non-Complicated Grief Group reported internalization of positive qualities and identification with the deceased child as a role model, whereas participants in the Complicated Grief Group did not report these experiences. In addition, the drawings of those in the Non-Complicated Grief Group were evaluated as more adaptive than those in the Complicated Grief Group. doi: 10.2190/OM.60.1.a

Abstract

Historically, from a Freudian and medical model perspective, emotional disengagement from the deceased was seen as essential to the successful adaptation of bereavement. A major shift in the bereavement literature has occurred and it is now generally accepted that despite the permanence of physical separation, the bereaved remains involved and connected to the deceased and can be emotionally sustained through continuing bonds. The majority of literature has focused on adults and on the nature of continuing bonds following the death of a spouse. In this article, the authors demonstrate how the continuing bonds concept applies to the sibling relationship. We describe the unique continued relationship formed by bereaved children and adolescents following a sibling loss, highlight the factors that influence the siblings continuing bonds expressions, and offer clinical interventions. In our view, mental health professionals can play an important role in helping parents encourage activities that may facilitate the creation and maintenance of continuing bonds in their children.

123. Social evaluation of faces across gender and familiarity

Abstract

Models of social evaluation aim to capture the information people use to form first impressions of unfamiliar others. However, little is currently known about the relationship between perceived traits across gender. In Study 1, we asked viewers to provide ratings of key social dimensions (dominance, trustworthiness etc.) for multiple images of 40 unfamiliar identities. We observed clear sex differences in the perception of dominance – with negative evaluations of high dominance in unfamiliar females, but not males. In Study 2, we used the social evaluation context to investigate key predictions about the importance of pictorial information in familiar and unfamiliar face processing. We compared the consistency of ratings attributed to different images of the same identities and demonstrated that ratings of images depicting the same familiar identity are more tightly clustered than those of unfamiliar identities. Such results imply a shift from image rating to person rating with increased familiarity, a finding which generalises results previously observed in studies of identification.

Abstract

Transcriptomic imputation approaches combine eQTL reference panels with large-scale genotype data in order to test associations between disease and gene expression. These genic associations could elucidate signals in complex genome-wide association study (GWAS) loci and may disentangle the role of different tissues in disease development. We used the largest eQTL reference panel for the dorso-lateral prefrontal cortex (DLPFC) to create a set of gene expression predictors and demonstrate their utility. We applied DLPFC and 12 GTEx-brain predictors to 40,299 schizophrenia cases and 65,264 matched controls for a large transcriptomic imputation study of schizophrenia. We identified 413 genic associations across 13 brain regions. Stepwise conditioning identified 67 non-MHC genes, of which 14 did not fall within previous GWAS loci. We identified 36 significantly enriched pathways, including hexosaminidase-A deficiency, and multiple porphyric disorder pathways. We investigated developmental expression patterns among the 67 non-MHC genes and identified specific groups of pre- and postnatal expression.

Abstract

Transcriptomic imputation approaches combine eQTL reference panels with large-scale genotype data in order to test associations between disease and gene expression. These genic associations could elucidate signals in complex genome-wide association study (GWAS) loci and may disentangle the role of different tissues in disease development. We used the largest eQTL reference panel for the dorso-lateral prefrontal cortex (DLPFC) to create a set of gene expression predictors and demonstrate their utility. We applied DLPFC and 12 GTEx-brain predictors to 40,299 schizophrenia cases and 65,264 matched controls for a large transcriptomic imputation study of schizophrenia. We identified 413 genic associations across 13 brain regions. Stepwise conditioning identified 67 non-MHC genes, of which 14 did not fall within previous GWAS loci. We identified 36 significantly enriched pathways, including hexosaminidase-A deficiency, and multiple porphyric disorder pathways. We investigated developmental expression patterns among the 67 non-MHC genes and identified specific groups of pre- and postnatal expression.

Abstract

Research into the effects of mindfulness meditation on behavioral outcomes has received much interest in recent years, with benefits for both short-term memory and working memory identified. However, little research has considered the potential effects of brief mindfulness meditation interventions or the nature of any benefits for visual short-term memory. Here, we investigate the effect of a single, 8-minute mindfulness meditation intervention, presented via audio recording, on a short-term memory task for faces. In comparison with two control groups (listening to an audiobook or simply passing the time however they wished), our mindfulness meditation participants showed greater increases in visual short-term memory capacity from pre- to post-intervention. In addition, only mindfulness meditation resulted in significant increases in performance. In conclusion, a single, brief mindfulness meditation intervention led to improvements in visual short-term memory capacity for faces, with important implications regarding the minimum intervention necessary to produce measurable changes in short-term memory tasks. Abstract

Abstract Background Alfalfa, a perennial, outcrossing species, is a widely planted forage legume producing highly nutritious biomass. Currently, improvement of cultivated alfalfa mainly relies on recurrent phenotypic selection. Marker assisted breeding strategies can enhance alfalfa improvement efforts, particularly if many genome-wide markers are available. Transcriptome sequencing enables efficient high-throughput discovery of single nucleotide polymorphism (SNP) markers for a complex polyploid species. Result The transcriptomes of 27 alfalfa genotypes, including elite breeding genotypes, parents of mapping populations, and unimproved wild genotypes, were sequenced using an Illumina Genome Analyzer IIx. De novo assembly of quality-filtered 72-bp reads generated 25,183 contigs with a total length of 26.8 Mbp and an average length of 1,065 bp, with an average read depth of 55.9-fold for each genotype. Overall, 21,954 (87.2%) of the 25,183 contigs represented 14,878 unique protein accessions. Gene ontology (GO) analysis suggested that a broad diversity of genes was represented in the resulting sequences. The realignment of individual reads to the contigs enabled the detection of 872,384 SNPs and 31,760 InDels. High resolution melting (HRM) analysis was used to validate 91% of 192 putative SNPs identified by sequencing. Both allelic variants at about 95% of SNP sites identified among five wild, unimproved genotypes are still present in cultivated alfalfa, and all four US breeding programs also contain a high proportion of these SNPs. Thus, little evidence exists among this dataset for loss of significant DNA sequence diversity from either domestication or breeding of alfalfa. Structure analysis indicated that individuals from the subspecies falcata, the diploid subspecies caerulea, and the tetraploid subspecies sativa (cultivated tetraploid alfalfa) were clearly separated. Conclusion We used transcriptome sequencing to discover large numbers of SNPs segregating in elite breeding populations of alfalfa. Little loss of SNP diversity was evident between unimproved and elite alfalfa germplasm. The EST and SNP markers generated from this study are publicly available at the Legume Information System (http://medsa.comparative-legumes.org/) and can contribute to future alfalfa research and breeding applications.

Abstract

Researchers have long been interested in how social evaluations are made based upon first impressions of faces. It is also important to consider the level of agreement we see in such evaluations across raters and what this may tell us. Typically, high levels of inter-rater agreement for facial judgements are reported, but the measures used may be misleading. At present, studies commonly report Cronbach’s α as a way to quantify agreement, although problematically, there are various issues with the use of this measure. Most importantly, because researchers treat raters as items, Cronbach’s α is inflated by larger sample sizes even when agreement between raters is fixed. Here, we considered several alternative measures and investigated whether these better discriminate between traits that were predicted to show low (parental resemblance), intermediate (attractiveness, dominance, trustworthiness), and high (age, gender) levels of agreement. Importantly, the level of inter-rater agreement has not previously been studied for many of these traits. In addition, we investigated whether familiar faces resulted in differing levels of agreement in comparison with unfamiliar faces. Our results suggest that alternative measures may prove more informative than Cronbach’s α when determining how well raters agree in their judgements. Further, we found no apparent influence of familiarity on levels of agreement. Finally, we show that, like attractiveness, both trustworthiness and dominance show significant levels of private taste (personal or idiosyncratic rater perceptions), although shared taste (perceptions shared with other raters) explains similar levels of variance in people’s perceptions. In conclusion, we recommend that researchers investigating social judgements of faces consider alternatives to Cronbach’s α but should also be prepared to examine both the potential value and origin of private taste as these might prove informative.

And this is a key aspect of why this method of deception often works: when the person checking the ID is unfamiliar to you, they are substantially less accurate in determining whether the photograph often present on these types of documents does, in fact, depict your face (Young & Burton, 2018). Similarly, providing several images of a face with the aim of conveying information regarding that face's variability also doesn't consistently help with the task (Ritchie et al., 2020, 2021; White et al., 2014). Some researchers have hypothesised that the use of averages - computer-generated images created from several photos of the same face - may improve face matching, because these "wash away" any extraneous information while maintaining the features that are identifiable "you". Face matching, as it's referred to by professionals, is difficult when the person is unfamiliar because the checker doesn't have any knowledge of how that person's face can look across different situations. Summary: Deciding whether two different face photographs or voice samples are from the same person represent fundamental challenges within applied settings. To date, most research has focussed on average performance in these tests, failing to consider individual differences and within‐person consistency in responses. Here, participants completed the same face (Experiment 1) or voice matching test (Experiment 2) on two separate occasions, allowing comparison of overall accuracy across the two timepoints as well as consistency in trial‐level responses. In both experiments, participants were highly consistent in their performances. In addition, we demonstrated a large association between consistency and accuracy, with the most accurate participants also tending to be the most consistent. This is an important result for applied settings in which organisational groups of super‐matchers are deployed in real‐world contexts. Abstract

As a mild-mannered reporter, Clark Kent is able to blend into human society without drawing much attention to himself. Although he utilises several methods of disguise (clothing, posture, hair style), perhaps his most famous is a simple pair of glasses (see Figure 1). We know that wearing glasses can make you look more educated and intelligent (e.g., Hellström & Tekle, 1994), but for Superman, the goal is primarily to hide his true identity. Of course, one of the cornerstones of enjoying superhero fiction is that we suspend our disbelief and try to ignore the obvious questions (for example, how useful or plausible is it that Squirrel Girl can communicate with and understand squirrels?!). However, the scientist inside us sometimes breaks through and we are given the opportunity to investigate. Here, we tackle the question that comic book fans have been asking for decades – could Superman really hide his identity using a pair of glasses?
