Abstract Rheumatoid arthritis (RA) is one of the most common autoimmune diseases worldwide. Due to high heterogeneity in disease manifestation, accurate and fast diagnosis of RA is difficult. This study analyzed the potential relationship between the infrared (IR) spectra obtained by attenuated total reflectance Fourier transform infrared spectroscopy (ATR-FTIR) and the presence of autoantibodies and antibodies against urease in sera. Additionally, the wave number of the IR spectrum that enabled the best differentiation between patients and healthy blood donors was investigated. Using a mathematical model involving principal component analysis and discriminant analysis, it was shown that the presence of anti-citrullinated protein antibody, rheumatoid factor, anti-neutrophil cytoplasmic antibodies, and anti-nuclear antibodies correlated significantly with the wave numbers in the IR spectra of the tested sera. The most interesting findings derived from determination of the best predictors for distinguishing RA. Characteristic features included an increased reaction with urease mimicking peptides and a correspondence with particular nucleic acid bands. Taken together, the results demonstrated the potential application of ATR-FTIR in the study of RA and identified potential novel markers of the disease.

Abstract Peripheral blood mononuclear cells (PBMC) are widely used as a biomarker source in nutrition/obesity studies because they reflect gene expression profiles of internal tissues. In this pilot proof-of-concept study we analysed in humans if, as we previously suggested in rodents, PBMC could be a surrogate tissue to study overweight/obesity impact on lipid metabolism. Pre-selected key lipid metabolism genes based in our previous preclinical studies were analysed in PBMC of normoglycemic normal-weight (NW), and overweight-obese (OW-OB) subjects before and after a 6-month weight-loss plan. PBMC mRNA levels of CPT1A, FASN and SREBP-1c increased in the OW-OB group, according with what described in liver and adipose tissue of humans with obesity. This altered expression pattern was related to increased adiposity and early signs of metabolic impairment. Greater weight loss and/or metabolic improvement as result of the intervention was related to lower CPT1A, FASN and SREBP-1c gene expression in an adjusted linear mixed-effects regression analysis, although no gene expression recovery was observed when considering mean comparisons. Thus, human PBMC reflect lipid metabolism expression profile of energy homeostatic tissues, and early obesity-related alterations in metabolic at-risk subjects. Further studies are needed to understand PBMC usefulness for analysis of metabolic recovery in weigh management programs.

Lung cancer is one of the most common cancers worldwide, causing nearly one million deaths each year. Herein, we present the effect of 2-methoxyestradiol (2-ME), the endogenous metabolite of 17β-estradiol (E2), on non-small cell lung cancer (NSCLC) cells. We observed that 2-ME reduced the viability of lung adenocarcinoma in two-dimensional (2D) and three-dimensional (3D) spheroidal A549 cell culture models. Molecular modeling was carried out aiming to visualize amino acid residues within binding pockets of the acyl-protein thioesterases, namely 1 (APT1) and 2 (APT2), and thus to identify which ones were more likely involved in the interaction with 2-ME.Our findings suggest that 2-ME acts as an APT1 inhibitor enhancing protein palmitoylation and oxidative stress phenomena in the lung cancer cell. In order to support our data, metabolomics of blood serum from NSCLC patients was also performed. Moreover, computational analysis suggests that 2-ME as compared to other estrogen metabolism intermediates is relatively safe in terms of its possible non-receptor bioactivity within healthy human cells due to a very low electrophilic potential and hence no substantial risk of spontaneous covalent modification of biologically protective nucleophiles.We propose that 2-ME can be used as a selective tumor biomarker in the course of certain types of lung cancers and possibly as a therapeutic adjuvant or neoadjuvant.

Industrial hemp (Cannabis sativa L.) is a plant species cultivated as a raw material for fiber extraction. Alternatively, hemp biomass can be used for feeding or energy purposes. This study was conducted to investigate the effect of inoculation with a lactic acid bacteria starter culture on the fermentation and chemical compositions of hemp silages. Hemp shoots (HS) and hemp flowers (HF) were ensiled in mini laboratory silos without or with the inoculation of the commercial starter culture Lactosil Biogaz (Lentilactobacillus buchnerii KKP 907 p; L. buchneri A KKP 2047 p; Pediococcus acidilactici KKP 2065 p). After 7 and 42 days of ensiling, the fermentation quality and chemical compositions of the silages were assessed. The use of Lactosil Biogas for hemp resulted in a decrease in pH, increase in lactic acid (LA), and reduction in fungal abundance in the HS silage. In the case of the HF silage, the bacterial inoculation was less effective; however, an increase in LA and a decrease in butyric acid (BA) were observed. As a result of the ensilage process, decreases in crude fiber and hemicellulose were observed in the HS and HF silages. Thus, hemp ensiling with biological additives is an effective pre-treatment of hemp plants for subsequent biofuel production that can preserve the biomass and provide the year-round availability of feedstock.

The main objective of this work was to explore the association of dietary phytate intake with bone mineral density (BMD) in a Mediterranean population of postmenopausal women. For this purpose, a cross-sectional analysis of 561 women aged 55–75 years with overweight/obesity and metabolic syndrome from a Mediterranean area and with data on dual-energy X-ray absorptiometry (DXA) scans in femur and lumbar spine was performed. Estimated phytate intake was calculated using a validated food frequency questionnaire. Our results indicated that phytate intake was associated with BMD [β(95%CI) per each 25 mg/100 kcal] in femoral neck [0.023(0.060–0.040) g/cm2], femoral Ward’s triangle [0.033(0.013–0.054) g/cm2], total femur [0.018(0.001–0.035) g/cm2], and all the analyzed lumbar spine sites [L1–L4: 0.033(0.007–0.059) g/cm2] after adjusting for potential confounders. The sensitivity analysis showed that phytate intake was directly associated with lumbar spine BMD in women younger than 66 years, with a body mass index higher than 32.6 kg/cm2 and without type 2 diabetes (all p-for interactions < 0.05). The overall results indicated that phytate, a substance present in food as cereals, legumes and nuts, was positively associated with BMD in Mediterranean postmenopausal women. Phytate may have a protective effect on bone resorption by adsorbing on the surfaces of HAP. Nevertheless, large, long-term, and randomized prospective clinical studies must be performed to assess the possible benefits of phytate consumption on BMD in postmenopausal women.

ObjectiveTo evaluate the effect of Non-alcoholic fatty liver disease (NAFLD) status in the impact of lifestyle over Health-related quality of life (HRQoL) in patients with metabolic syndrome (MetS).MethodsBaseline and 1 year follow up data from the PREDIMED-plus cohort (men and women, 55-75 years old with overweight/obesity and MetS) were studied. Adherence to an energy-restricted Mediterranean Diet (er-MeDiet) and Physical Activity (PA) were assessed with a validated screeners. Hepatic steatosis index (HSI) was implemented to evaluate NAFLD while the SF-36 questionnaire provided HRQoL evaluation. Statistical analyses were performed to evaluate the influence of baseline NAFLD on HRQoL as affected by lifestyle during 1 year of follow up.ResultsData from 5205 patients with mean age of 65 years and a 48% of female participants. Adjusted linear multivariate mixed regression models showed that patients with lower probability of NAFLD (HSI < 36 points) were more responsive to er-MeDiet (β 0.64 vs β 0.05 per er-MeDiet adherence point, p< 0.01) and PA (β 0.05 vs β 0.01 per MET-h/week, p = 0.001) than those with high probability for NAFLD in terms Physical SF-36 summary in the 1 year follow up. 10 points of er-MeDiet adherence and 50 MET-h/week were thresholds for a beneficial effect of lifestyle on HRQoL physical domain in patients with lower probability of NAFLD.ConclusionThe evaluation of NAFLD by the HSI index in patients with MetS might identify subjects with different prospective sensitivity to lifestyle changes in terms of physical HRQoL (http://www.isrctn.com/ISRCTN89898870).

Abstract Background The optimal distribution between physical activity (PA) levels and sedentary behaviour (SB) for the greatest benefits for body composition among older adults with overweight/obesity and chronic health conditions remains unclear. We aimed to determine the prospective association between changes in PA and in SB with concurrent changes in body composition and to examine whether reallocating inactive time into different physical activity levels was associated with 12-month change to body composition in older adults. Methods Longitudinal assessment nested in the PREDIMED-Plus trial. A subsample (n = 1564) of men and women (age 55–75 years) with overweight/obesity and metabolic syndrome from both arms of the PREDIMED-Plus trial was included in the present analysis. Participants were followed up at 6 and 12 months. Physical activity and SB were assessed using validated questionnaires. Out of 1564 participants, 388 wore an accelerometer to objectively measure inactive time and PA over a 7-day period. At each time point, participants’ body composition was measured using dual-energy X-ray absorptiometry (DXA). Standard covariate-adjusted and isotemporal substitution modelling were applied to linear mixed-effects models. This paper presents the results of an analysis of the changes in the stiffness of asphalt binders modified with a bio-flux additive and subjected to the processes of foaming and short-term ageing. The purpose of the analyses was to determine changes in the rheological properties of asphalt binder as a result of technological processes characteristic of hot and warm mix asphalt technology. Three asphalt binders with similar penetration but varying degrees of polymer modification were tested: 50/70, 45/80–55 polymer-modified bitumen, and 45/80–80 highly modified bitumen. Tests were carried out on four groups of binders: original binders, foamed binders after 14 days of storage, non-foamed binders after Rolling Thin Film Oven Test (RTFOT) ageing, and foamed binders after 14 days of storage subjected to RTFOT ageing. The master curves of the complex shear modulus G* were analysed, and three indexes of binder stiffening were determined, characterising the investigated effects. The tests showed that some of the stiffening indices significantly depended on the degree of polymer modification and the content of the bio-flux additive. Moreover, it was found that the foaming process in the case of paving-grade bitumen and polymer-modified bitumen did not contribute to the additional stiffening of the binders.

The presented work shows the antibacterial activity of TiO2 photocatalysts modified by 3-aminopropyltriethoxysilane (APTES). The APTES-functionalized TiO2 samples were obtained by the solvothermal process followed by calcination. The antibacterial activity of APTES/TiO2 samples was evaluated with two species of bacteria, Escherichia coli and Staphylococcus epidermidis, under artificial solar light (ASL) irradiation. The used bacteria are model organisms characterized by negative zeta potential (approx. −44.2 mV for E. coli and −42.3 mV for S. epidermidis). For the first time, the antibacterial properties of APTES-functionalized TiO2 were evaluated against mono- and co-cultured bacteria. The high antibacterial properties characterized the obtained APTES-modified nanomaterials. The best antibacterial properties were presented in the TiO2-4 h-120 °C-300 mM-Ar-300 °C sample (modified with 300 mM of APTES and calcined at 300 °C). The improvement of the antibacterial properties was attributed to a positive value of zeta potential, high surface area, and porous volume.

The article presents the findings of a study that examines the perception of the educational paradigm in the context of educating a child with a chronic illness in either a school or medical facility by special education teacher candidates. It was assumed that the beliefs of educators largely reflect the educational paradigm, which reveals the thought process regarding children with chronic illnesses in two cognitive categories: (1) the actions of teachers; and (2) the objectives of educational activities aimed at pupils with chronic illnesses and their circumstances. The undertaken study was qualitative. The research was carried out in Poland on a group of 102 sophomore special education teacher candidates. The results indicated that the approach to educating a child with a chronic illness in a medical institution is predominantly influenced by the humanistic paradigm. In the revealed pattern of thinking about educating a child with a chronic illness in a medical facility, amongst the subjects, the key difference lies in the concept of preparing the child with the illness for life. Special education teacher candidates are primarily exposed to person-centred interventions that aim to facilitate optimal adaptation of children with disabilities to the typical demands of their healthy peers. This work presents an overview of the reports on the bacterial cell photocatalytic destruction and mineralization process in the presence of TiO2-based photocatalysts. The presented research included experiments conducted in air and water. Numerous works confirmed that a photocatalytic process with TiO2 led to bacteria and their organic residues' mineralization. Additionally, based on the obtained results, a possible two-stage mechanism of photocatalytic mineralization in the presence of TiO2-based materials was proposed. To help future studies, challenges of photocatalytic microorganism mineralization are also proposed. There are some aspects that need to be addressed, such as the lack of standardization of conducted research or relatively small amount of research on photocatalytic microorganism mineralization. According to our best knowledge, in the available literature, no work regarding a summary of previous research on photocatalytic bacterial mineralization process was found. [ABSTRACT FROM AUTHOR]

This research, focusing on the environmental impact of tire and brake disc pad wear, constitutes a significant area of transport-related studies. These two key vehicle components are not only the most frequently worn but also generate micro- and nano-pollutants (i.e., rubber, metal oxides) that potentially harm the environment. Over half of the globally produced natural and synthetic rubbers, which amounted to about 30 million tons in 2022, are used for tire production. This work focuses on the study of roadside snow, sand, and standing water deposits from various locations in the urban agglomeration (Poznań, Poland) during the winter season, determining their qualitative composition and the quantitative content of pollutants originating from tire abrasion. In addition, the method of washing nano- and micro-rubber particles and their full characteristics was also presented. Fourier-transform infrared (FT-IR) and nuclear magnetic resonance (NMR) spectroscopic studies, optical and scanning electron microscopy (SEM-EDS), particle size studies using a dynamic light scattering (DLS) particle analyzer, and thermogravimetric analysis (TGA) were conducted for a detailed characterization of the pollutants in the environment. The conducted particle separation methods allowed for the extraction of a fraction mainly containing gum residues with particle sizes less than 2 µm. The results of these tests make it possible to estimate the level of contamination with rubber and metal residues during the abrasion of tires, pads, and brake discs while driving, which is crucial for understanding the impact of vehicle part exploitation on the environment. Background. Tokophobia is a type of specific phobia related to the pathological fear of natural childbirth and occurs in about 6–10% of expectant mothers. Tokophobia is multifaceted, and its underlying factors are biological, psychological, and social. There are primary and secondary tokophobia and tokophobia coexisting with depression. The crucial symptom of tokophobia is the fear of pain. Tokophobia is associated with low pain tolerance, inhibiting the endogenous mechanisms of analgesia caused by pregnancy. The basic form of treatment for tokophobia is psychotherapy. Objectives. The purpose of this article is to review the latest publications which state the methods of therapy for tokophobia. Material and methods. To assess available cognitive behavioral therapy techniques in the treatment of tokophobia, a systematic review of four electronic databases (EBSCOhost, PEDro, PubMed, and Scopus) were searched until the end of May 2023. Results and conclusions. The use of cognitive-behavioral therapy techniques for this type of specific phobia can minimize negative beliefs about childbirth and experienced pain, which can result in a reduction of negative experiences and thereby reduce the number of caesarean sections on demand. The inclusion of alternative interventions, such as the practice of mindfulness and compassion, seems to be a supportive treatment option in the case of a tendency to worry in pregnant women, as well as the experience of discomfort during pregnancy [ABSTRACT FROM AUTHOR]

Nickel alloys, despite their good strength properties at high temperature, are characterized by limited weldability due to their susceptibility to hot cracking. So far, theories describing the causes of hot cracking have focused on the presence of impurities in the form of sulphur and phosphorus. These elements form low-melting eutectic mixtures that cause discontinuities, most frequently along solid solution grain boundaries, under the influence of welding deformations. Progress in metallurgy has effectively reduced the presence of sulphur and phosphorus compounds in the material, however, the phenomenon of hot cracking continues to be the main problem during the welding of nickel-based alloys. It was determined that nickel-based alloys, including Inconel 617, show a tendency towards hot cracking within the high-temperature brittleness range (HTBR). There is no information on any structural changes occurring in the HTBR. Moreover, the literature indicates no correlations between material-related factors connected with structural changes and the amount of energy delivered into the material during welding. This article presents identification of correlations between these factors contributes to the exploration of the mechanism of hot cracking in solid-solution strengthened alloys with an addition of cobalt (e.g. Inconel 617). The article was ended with development of hot cracking model for Ni-Cr-Mo-Co alloys.

The term contraception can be used to define a set of methods that control fertility as well as help conscious family planning. The growing interest in the subject of contraception methods, which are used by majority of women today, cause that there are more and more ways of fertility regulation. In this paper are presented the most common contraceptive methods, taking into account the effectiveness of a given method, contraindications, side effects and the mechanism of action. The Pearl Index indicates failure rate of the use of a given contraceptive method, i.e. it allows to determine the number of pregnancies in a group of 100 women using a given contraceptive method within one year. It is still a frequently used way of comparing the effectiveness of different contraceptive methods. Most of the available methods are based on the use of synthetic hormones from the group of progestins and estradiol derivatives in the form of combinened oral contraceptive pills, mini-pills, patches, intramuscular injections, vaginal rings, implants, intrauterine devices (IUDs with hormones - levonorgestrel and without hormones, copper). Combined contraceptive pills are very popular, especially among sexually active young women. Despite their lower effectiveness, mini-pills tablets are often used by women who, for various reasons, cannot use ethinylestradiol derivatives. The undoubted advantage of vaginal rings, contraceptive patches and intramuscular injections is the lack of a daily regimen of their use. IUDs are most often chosen by women who do not plan pregnancy in the next few years. Natural methods were also examined, the greatest effectiveness of which, confirmed by the Pearl Index, is based on the use of cycle computers. The biggest limitation of these cycle computers is their high price. Mechanical methods and less frequently used spermicides are also discussed. Contraceptive methods should characterize a broad safety profile, be low-priced, easily accessible and their effects should be fully reversible. Despite of the various available forms of contraception nowadays, there still is no method which would match all of the above given criteria.

Local currencies represent an alternative to fiat money. They contribute to the development of local communities, primarily by supporting small and medium-sized enterprises, which remain fundamental for the economy of every European country. The purpose of the study is to identify both ways and methods focused on supporting the implementation of the concept of sustainable development by the selected local currencies in the area of Europe, analysed in the years 1932–2020. The paper takes the form a descriptive analysis based on the studies covering Polish, Czech, Dutch, German and English source literature as well as the Community Currencies in Action (CCIA) publications, websites of the analysed local currencies and other studies in the field of financial law. The method of comparative analysis was used to examine the similarities and differences in supporting the implementation of the concept of sustainable development. The basic conclusion formulated after conducting the research is the fact that each local currency does support sustainable development in an indirect way. In addition, certain local currencies can be identified, in the case of which the support of sustainable development is the essence of their functioning. The effectiveness of the impact exerted by local currencies on sustainable development depends on the involvement of the creators of these currency systems and the specificity of the existing conditions. The ability to establish partnerships between non-governmental organisations, business entities and public administration may also play a significant role.

BackgroundRate of nicotine metabolism is an important factor influencing cigarette smoking behavior, dependence, and efficacy of nicotine replacement therapy. The current study examined the hypothesis that chronic alcohol abuse can accelerate the rate of nicotine metabolism. Nicotine metabolite ratio (NMR, a biomarker for rate of nicotine metabolism) and patterns of nicotine metabolites were assessed at three time points after alcohol cessation.MethodsParticipants were 22 Caucasian men randomly selected from a sample of 165 smokers entering a 7-week alcohol dependence treatment program in Poland. Data were collected at three time points: baseline (week 1, after acute alcohol detoxification), week 4, and week 7. There is preliminary evidence that music therapy plays a beneficial role for preterm infants and their primary caregivers during the neonatal period; however, available research considers mostly cohorts from North and South Americas, Australia, Israel and certain western European countries, excluding Poland. Remembering that music, and therefore music therapy, is highly culturally dependent, there is no “perfect program package” that can be directly applied across all communities to achieve desired effects everywhere. The commencement of the multi-center international randomized controlled trial, Longitudinal Study of music Therapy’s Effective-ness for Premature (LongSTEP) infants and their caregivers, has provided the impetus for Polish music thera-pists to begin offering music therapy services in neonatal intensive care units as a part of clinical research. Since research on music therapy dedicated to premature babies and their families is only beginning to emerge in Poland, there is a call to develop a culturally and context-based approach that can be implemented in Polish neonatal settings. The objective of this perspective article is to present recommendations for clinical practice with premature babies and their families based on available international research and practical guidelines, as well as national experiences from the LongSTEP trial that make it culturally appropriate. This paper might be of great interest to readers interested in implementing music therapy interventions with local conditions and opportunities to improve outcomes for a “real-world” impact.

Vitiligo is a chronic depigmentation disease. It is characterized by damage or destruction of melanocytes (melanin-producing cells). The desease affects about 2% of population. The pathogenesis is complex and not fully understood. Recent insights into vitiligo pathognenesis highlight impact of various factors like environmental factors, genetic factors, cellular stress and autoimmune factors. In vitiligo patients abnormal response of innate immune system is observed. The latest research shows that the IFN-γ/CXCL10 axis plays a special role in the pathogenesis of vitiligo. Interferon γ (IFN-γ) can be produced by autoreactive CD8 + T cells. Receptors for interferon gamma (IFN-γR) are located on the surface of keratinocytes. After binding IFN-γ to its receptor (IFN-γR) there is an activation of the JAK-STAT pathway. That causes the release of the chemokines like CXCL9 and CXCL10. Patients with vitiligo have a higher serum concentration of CXCL10 chemokine compared to healthy people. CXCR3 receptors for the CXCL10 chemokine are located on autoreactive CD8+ lymphocytes. The CXCL10 chemokine binds to its receptor CXCR3. That leads to the migration of autoreactive T cells to the epidermis and destruction of melanocytes. Furthermore, changes in cytokine secretion by T helper cells Th1, Th2, Th17 and regulatory T cells (Treg) may contribute to disease progression. In vitiligo is observed elevated levels of IL-2, IL-6, IL-17 and decreased level of IL-4. Current treatment methods are not satisfying and mainly based on the use of off-label drugs. Better understandig etiology of vitiligo enable searching for new effective treatment methods. To minimize systemic side effects, topical drug forms are searching. The effectiveness of new combined therapies is also being studied, including topical treatment with phototherapy, surgical methods with phototherapy, general therapy with surgical methods, etc. New potential therapies, especially with JAK/STAT inhibitors, may provide new perspectives for the treatment of vitiligo.

The development of power industry obligates designers, materials engineers to create and implement new, advanced materials, in which Inconel 617 alloy is included. Nowadays, there are a lot of projects which describe microstructure and properties of Inconel 617 alloy. However, the welded joints from mentioned material is not yet fully discussed in the literature. The description of welded joints microstructure is a main knowledge source for designers, constructors and welding engineers in estimating durability process and degradation assessment for elements and devices with welds of Inconel 617 alloy. This paper presents the analysis and assessment of advanced nickel alloy welded joints, which have been done by tungsten inert gas (TIG). Investigations have included analysis made by light microscope and scanning electron microscope. The disclosed precipitates were identified with Energy Dispersive Spectroscopy (EDS) microanalysis, then it were done X-Ray Diffraction (XRD) phases analysis. To confirm the obtained results, a scanning-transmission electron microscope (STEM) analysis was also performed. The purpose of the article was to create a comprehensive procedure for revealing the Inconel 617 alloy structure. The methodology presented in this article will be in future a great help for constructors, material specialists and welding engineers in assessing the structure and durability of the Inconel 617 alloy.

Abstract Background This study explored the association between inactive time and measures of adiposity, clinical parameters, obesity, type 2 diabetes and metabolic syndrome components. It further examined the impact of reallocating inactive time to time in bed, light physical activity (LPA) or moderate-to-vigorous physical activity (MVPA) on cardio-metabolic risk factors, including measures of adiposity and body composition, biochemical parameters and blood pressure in older adults. Methods This is a cross-sectional analysis of baseline data from 2189 Caucasian men and women (age 55–75 years, BMI 27–40 Kg/m2) from the PREDIMED-Plus study (http://www.predimedplus.com/). All participants had ≥3 components of the metabolic syndrome. Inactive time, physical activity and time in bed were objectively determined using triaxial accelerometers GENEActiv during 7 days (ActivInsights Ltd., Kimbolton, United Kingdom). Multiple adjusted linear and logistic regression models were used. Isotemporal substitution regression modelling was performed to assess the relationship of replacing the amount of time spent in one activity for another, on each outcome, including measures of adiposity and body composition, biochemical parameters and blood pressure in older adults. Results Inactive time was associated with indicators of obesity and the metabolic syndrome. Reallocating 30 min per day of inactive time to 30 min per day of time in bed was associated with lower BMI, waist circumference and glycated hemoglobin (HbA1c) (all p-values
