101. Variation at the insulin gene VNTR (variable number tandem repeat) polymorphism and early growth: studies in a large Finnish birth cohort

Abstract

Variation at the insulin gene (INS-)VNTR (variable number of tandem repeats) minisatellite polymorphism has been reported to be associated with both early growth and adult metabolic phenotypes. However, the samples studied have been small and the relationship between INS-VNTR variation and parameters of early growth inconsistent, with four previous studies producing conflicting results. We have studied the relationship between INS-VNTR class (measured by genotyping the nearby -23HphI variant with which it is in tight linkage disequilibrium) and early growth in 5,646 members of the Northern Finnish Birth Cohort of 1966. Comparing class III homozygotes with other genotypes using multivariate linear regression analysis, we found no significant associations with any early growth measure (birth weight, birth length, ponderal index, and head circumference at 1 year), even after stratifying subjects by growth trajectory during infancy and/or birth order. For example, among infants with limited postnatal growth realignment (n = 2,470), class III/III infants were no heavier at birth (difference [+/-SE] in the means [fully adjusted], 58 +/- 51 g; P = 0.26) than class I/- infants. No significant associations were detected following reanalysis with an additive model (for example, for birth weight, beta = 20 g [95% CI -3 to 44], P = 0.09). Studies of this large population-based cohort have failed to generate convincing evidence that INS-VNTR variation influences early growth.

Abstract

Three reproductive management systems designed to use prostaglandin F2α (PGF) to save labor needed for estrous detection and artificial insemination (AI) were compared to conventional dairy herd reproductive management to evaluate their potential to maintain herd reproductive performance. The four experimental groups were: (1) CONTROL = daily estrous detection for AI with no PGF intervention, (2) Appointment (APPT) AI = double PGF injection at a 12-day interval followed by single appointment AI at 72 hrs for first AI, with repeat AI based on estrous detection, (3) 7-DAY = weekly (Monday) PGF injection for all cows ready for first or repeat AI with AI based on estrous detection which occurred only four days (Wednesday through Saturday) of each week, and (4) 21-DAY = repeating 21-day blocks of 13 days of AI based on estrous detection with PGF given on day 8 to cows yet to be inseminated and 8 days with no estrous detection or AI. 109. Neurochemical characterization of neuronal populations expressing protein kinase C gamma isoform in the spinal cord and gracile nucleus of the rat

Abstract

Protein kinase C gamma (PKCgamma) is widely distributed throughout the CNS and is thought to play a role in long term hyper-excitability in nociceptive neurones. Here, we provide the first report of PKCgamma cells in the dorsal column nuclei of the adult rat. Retrograde labeling of PKCgamma cells from the thalamus with choleragenoid revealed that 25% of the PKCgamma positive gracile cells projected to the thalamus. Further, we have characterized the distribution of PKCgamma within gracile nucleus in terms of colocalization with various neurotransmitter receptors or enzymes and calcium binding proteins, and compared this with PKCgamma colocalization in cells of laminae I-III of the spinal cord. We show that approximately 90% of the PKCgamma cells in the gracile nucleus and 60% in the dorsal horn were immuno-positive for the AMPA receptor subunit glutamate 2/3 (GluR2/3). Little coexpression was seen with neurokinin 1 receptor, nitric oxide synthase (NOS) and the AMPA receptor subunit GluR1, markers of distinct neuronal subpopulations. In the spinal cord, a quarter of PKCgamma cells expressed calbindin, but very few cells did so in the gracile nucleus. Electrical stimulation at c-fiber strength of the normal or injured sciatic nerve was used to induce c-fos as a marker of postsynaptic activation in the spinal cord and gracile nucleus. Quantitative analysis of the number of PKCgamma positive gracile cells that expressed also c-fos increased from none to 24% after injury, indicating an alteration in the sensory activation pattern in these neurones after injury. C-fos was not induced in inner lamina II following c-fiber electrical stimulation of the intact or axotomized sciatic nerve, indicating no such plasticity at the spinal cord level. As dorsal column nuclei cells may contribute to allodynia after peripheral nerve injury, pharmacological modulation of PKCgamma activity may therefore be a possible way to ameliorate neuropathic pain after peripheral nerve injury.

Abstract

The aim of the present work is to grow compressive strained InGaAs/GaAs quantum well structures and study energy levels in them. Multiple layers of 20 InGaAs/GaAs quantum wells were grown by MBE. The energy for the C1-HH1 transition of the grown structure was measured using Photoluminescence and Electroluminescence. Low energy ion scattering was carried out on the samples to study the depth profile of the multiple quantum wells.

121. A Cross-regional exploration of barriers to the adoption and use of electronic meeting systems

Abstract

Electronic Meeting Systems (EMS) are intended to support group collaboration in completing tasks.While there have been many case studies and laboratory experiments on how EMS can support group tasks, large scale macro investigations exploring EMS adoption and use have been practically non-existent. Furthermore, while several barriers to EMS adoption and use have been suggested, their validation across organizations remains unexplored. We undertook a global initiative to explore information technology support for task-oriented collaboration in the US, Australia, Hong Kong, and Norway. In this paper, we focus specifically on assessing the adoption and use of EMS, and barriers to their adoption and use in organizations across the four regions. Our results suggest that EMS currently have limited adoption and are used infrequently across all the four regions. A further investigation into barriers to EMS adoption and use suggests that significant numbers of respondents do agree with a list of fourteen suggested barriers. However, while there is significant agreement between two countries (US & Australia) over how these barriers are ranked, there is no significant agreement between the remaining pairs of countries. Implications of our findings are discussed for practitioners and researchers.

122. Barriers to EMS assimilation in the US, Australia, Hong Kong, Norway and Switzerland: A Cross regional comparison

Abstract

Electronic Meeting Systems (EMS) have the capabilities to support group members in task collaboration. While there have been many case studies and lab experiments on how EMS can support group tasks, large scale macro investigations exploring EMS adoption and use have been practically non-existent. Furthermore, while several EMS assimilation barriers have been suggested, their validation across organizations remains unexplored. In this paper, we describe a global initiative to explore information technology support for task-oriented collaboration in the US, Australia, Hong Kong, Norway and Switzerland. We focus specifically on EMS and investigate their adoption, use and assimilation barriers in organizations across the five regions. Our results suggest that EMS adoption and use is limited across all the regions. A further investigation in EMS assimilation barriers suggests that these barriers differ across the regions and there is little agreement over how they are ranked. Implications of our findings are discussed for practitioners and researchers.

133. Sequence variation in the androgen receptor gene is not a common determinant of male sexual orientation

Abstract

To test the hypothesis that DNA sequence variation in the androgen receptor gene plays a causal role in the development of male sexual orientation, we have (1) measured the degree of concordance of androgen receptor alleles in 36 pairs of homosexual brothers, (2) compared the lengths of polyglutamine and polyglycine tracts in the amino-terminal domain of the androgen receptor in a sample of 197 homosexual males and 213 unselected subjects, and (3) screened the the entire androgen receptor coding region for sequence variation by PCR and denaturing gradient-gel electrophoresis (DGGE) and/or single-strand conformation polymorphism analysis in 20 homosexual males with homosexual or bisexual brothers and one homosexual male with no homosexual brothers, and screened the amino-terminal domain of the receptor for sequence variation in an additional 44 homosexual males, 37 of whom had one or more first- or second-degree male relatives who were either homosexual or bisexual. These analyses show that (1) homosexual brothers are as likely to be discordant as concordant for androgen receptor alleles; (2) there are no large-scale differences between the distributions of polyglycine or polyglutamine tract lengths in the homosexual and control groups; and (3) coding region sequence variation is not commonly found within the androgen receptor gene of homosexual men. The DGGE screen identified two rare amino acid substitutions, ser205-to-arg and glu793-to-asp, the biological significance of which is unknown.

Abstract

Weapons Directors (WDs) are cognitively complex, decision making jobs involving air traffic control of friendly assets and tracking hostiles from an airborne platform. Three cognitive task analyses (CTAs) were reviewed for WD jobs on the Airborne Warning and Control System aircraft. 230 tasks were derived from this "meta-CTA." Nine performance categories emerged, representing a cognitive-behavioral model of the WD job domain. To test the model, 38 WDs were observed participating in a computer simulation exercise during which they verbalized aloud their thoughts. These verbal protocols were recorded, transcribed, and reduced to task statements. Using a checklist derived from the meta-CTA, subject matter experts classified each of the statements as either primarily cognitive in nature, behavioral in nature, or a combination of the two. Classifications were analyzed to determine job performance differences between experienced and inexperienced WDs. Verbal protocol analysis presents the opportunity to integrate cognitive task analyses into a job model, yielding a new classification system based on a cognitive-behavior approach. This typology for describing jobs transcends traditional job analysis and has applicability to other complex management jobs involving decision-making, problem-solving, and resource management. 138. Alignment of fluid molecules in an EHD contact

Abstract

Dichroic infrared emission spectra have been obtained from an operating elastohydrodynamic contact under conditions showing unequivocally that the molecules of the fluid are aligned under shear flow in the bulk of the fluid and that the extent of the alignment is increased by the presence of an additive in a small concentration. This work applied particularly to polyphenyl ether and to 1,1,2-trichloroethane as the additive, but similar results have been obtained with polycyclohexyls and the same additive. The observation of two separate glass transitions for the polycyclohexyl solution is an indication of the occurrence of a phase separation in the Hertzian inlet zone. A model has been developed to explain the effect of these phenomena and their relation to traction.

139. Temperature gradients through EHD films and molecular alignment evidenced by infrared spectroscopy

Abstract

Partial and complete emission band reversals have been observed in the infrared emission spectra from portions of operating sliding contacts. An elementary analysis has been carried out to show that partial reversals are due to temperature gradients in the fluid film - the film acts both as a radiation-emitter and absorber, and that total reversals - an emission spectrum appears as an absorption spectrum are likely to be due to a continuum source, such as hot solid asperities. The total energy radiated under the latter conditions exceeds that under the others. A decrease in gap width with increased load was accompanied by a dramatic spectral change in the case of 5P4E polyphenyl ether, which is indicative of molecular alignment.

Abstract

Addition of trichloroethane to a traction fluid was studied in a ball/diamond plate EHD apparatus by infrared emission spectroscopy of the radiation transmitted through the plate. Small concentrations of an organic chloride at first increased infrared radiance, then decreased it. Large concentrations caused a rapid decrease. During running, the infrared spectrum showed an increase of aromatics and olefins. A polymeric deposit formed on the plate. X-ray examination of the ball surface showed the presence of chlorine. Reuse of the same ball with fresh fluid resulted in similar chain of events, indicating that adsorbed chloride could promote fluid deterioration.

Abstract

A far-infrared interferometer was converted into an emission microspectrophotometer for surface analysis. To cover the mid-infrared as well as the far-infrared the Mylar beamsplitter was made replaceable by a germanium-coated salt plate, and the Moire fringe counting system used to locate the moveable Michelson mirror was improved to read 0.5 micron of mirror displacement. Digital electronics and a dedicated minicomputer were installed for data collection and processing. The most critical element for the recording of weak emission spectra from small areas was, however, a reflecting microscope objective and phase-locked signal detection with simultaneous referencing to a blackbody source. An application of the technique to lubrication problems is shown.
