Abstract Background This paper describes the design of an event ontology being developed for application in the machine understanding of infectious disease-related events reported in natural language text. This event ontology is designed to support timely detection of disease outbreaks and rapid judgment of their alerting status by 1) bridging a gap between layman's language used in disease outbreak reports and public health experts' deep knowledge, and 2) making multi-lingual information available. Construction and content This event ontology integrates a model of experts' knowledge for disease surveillance, and at the same time sets of linguistic expressions which denote disease-related events, and formal definitions of events. In this ontology, rather general event classes, which are suitable for application to language-oriented tasks such as recognition of event expressions, are placed on the upper-level, and more specific events of the experts' interest are in the lower level. Each class is related to other classes which represent participants of events, and linked with multi-lingual synonym sets and axioms. Conclusions We consider that the design of the event ontology and the methodology introduced in this paper are applicable to other domains which require integration of natural language information and machine support for experts to assess them. The first version of the ontology, with about 40 concepts, will be available in March 2008.

Pancreatic islet transplantation is one of the clinical options for certain types of diabetes. However, difficulty in maintaining islets prior to transplantation limits the clinical expansion of islet transplantations. Our study introduces a dynamic culture platform developed specifically for primary human islets by mimicking the physiological microenvironment, including tissue fluidics and extracellular matrix support. We engineered the dynamic culture system by incorporating our distinctive microwell-patterned porous collagen scaffolds for loading isolated human islets, enabling vertical medium flow through the scaffolds. The dynamic culture system featured four 12 mm diameter islet culture chambers, each capable of accommodating 500 islet equivalents (IEQ) per chamber. This configuration calculates > five-fold higher seeding density than the conventional islet culture in flasks prior to the clinical transplantations (442 vs 86 IEQ/cm 2 ). We tested our culture platform with three separate batches of human islets isolated from deceased donors for an extended period of 2 weeks, exceeding the limits of conventional culture methods for preserving islet quality. Static cultures served as controls. The computational simulation revealed that the dynamic culture reduced the islet volume exposed to the lethal hypoxia (< 10 mmHg) to ~1/3 of the static culture. Dynamic culture ameliorated the morphological islet degradation in long-term culture and maintained islet viability, with reduced expressions of hypoxia markers. Furthermore, dynamic culture maintained the islet metabolism and insulin-secreting function over static culture in a long-term culture. Collectively, the physiological microenvironment-mimetic culture platform supported the viability and quality of isolated human islets at high-seeding density. Such a platform has a high potential for broad applications in cell therapies and tissue engineering, including extended islet culture prior to clinical islet transplantations and extended culture of stem cell-derived islets for maturation.

Zr-based metal-organic frameworks (Zr-MOFs) are emerging as potential adsorbents for removing nitrogen contaminants in fuel and wastewater. Functionalized Zr-MOFs exhibited superior indole (IND) adsorption capacity than the original MOF, but the activity of the multiple adsorption sites has not been discussed. Through experiment measurement and Density Functional Theory (DFT) calculations, this research investigated the IND adsorption mechanism at various sites on Zr-BDC-NH2 and Zr-BDC-NO2. Based on the adsorption isotherm, the adhesion of IND with Zr-MOFs was determined via the Freundlich model. DFT results suggest that the IND adsorption occurred at highly active sites such as O-linker, μ3-O-cluster, N-amine, and O-nitro via hydrogen and π-hydrogen bonds. Furthermore, the binding energy at each adsorption site in Zr-BDC-NH2 is higher than in Zr-BDC-NO2. In addition to the hydrogen acceptor bond, the π-hydrogen bond has also been analyzed through the electron density difference.

Abstract Tornadoes are responsible for several high-impact weather disasters in Japan. However, little is known about how these events have changed over the last several decades or how they may change in future climates. This study examines environmental conditions associated with tornados in Japan using pseudo-soundings from the high-resolution fifth-generation ECMWF reanalysis. We first determine appropriate discriminators of F2+ tornadoes using thermodynamic (convective available potential energy, convective inhibition, lifting condensation level, and the K-index), kinematic (bulk wind difference and storm-relative helicity), and multivariate tornado parameters (energy helicity index, K-helicity index, and the significant tornado parameter), and confirm that F2+ tornadoes occur in environments with higher instability and helicity, but are better distinguished using multivariate parameters. Recent trends indicate that F2+ environments have increased significantly in some regions over the last four decades. We also examined future changes for each parameter using a large ensemble 2-K warming experiment. Robust increases in strong tornado environments are depicted in many regions in Japan, particularly on the Sea of Japan side and the Kanto region. This indicates that despite projected decreases in bulk wind difference and higher convective inhibition, significant increases in atmospheric instability compensate, leading to more days with F2+ tornado potential.

Abstract Background Female sex and younger age are reported risk factors for chemotherapy‐induced nausea and vomiting (CINV) in highly emetogenic chemotherapy, but the underlying mechanism has not been elucidated. The purpose of this study was to clarify the impact of menopause on CINV. Methods This retrospective observational study analyzed data from consecutive patients who received their first cycle of perioperative anthracycline‐based chemotherapy for breast cancer between January 2018 and June 2020. The endpoints were association between CINV (vomiting, ≥Grade 2 nausea, complete response [CR] failure) and menopause as well as the association between CINV and follicle‐stimulating hormone [FSH]/estradiol [E2]. Results Data for 639 patients were analyzed. Among these patients, 109 (17.1%) received olanzapine (four antiemetic combinations) and 530 (82.9%) did not (three antiemetic combinations). Premenopausal state (amenorrhea lasting ≥12 months) was significantly associated with ≥Grade 2 nausea and CR failure in univariate analysis but not in multivariate analysis.
