Abstract

Eugenia jambolana belonging to family Myrtaceae is a large evergreen tree of about 50 ft height and possesses a large crown. The present study was designed to evaluate the analgesic activity of the petroleum ether, chloroform, methanol and aqueous extract of seeds of Eugenia jambolana Linn. and isolation of bioactive compound. The shade dried, coarsely powdered dried seeds of E. jambolana were successively extracted with petroleum ether, chloroform and methanol for 48 hours each using soxhlet apparatus and finally boiled with distilled water for 4 hours. The extracts were filtered, concentrated in vacuo. The crude dried extracts were evaluated for analgesic activity using hot plate test and tail immersion test. Maximum bioactive extract was subjected to column chromatography. The effect of various extracts of E. jambolana (intraperitonially) elicited a significant analgesic activity in hot plate test and tail immersion test as shown by the increase in latency time in seconds as compared with the control. The increase in nociceptive latency time was dose-dependent. The methanol extract of E. jambolana showed significant increase in latency time. The other plant extracts were not as potent as the methanol extract as compared to the latency time of the standard drug (Pentazocine). As the methanol extract showed maximum analgesic activity therefore, it was subjected to column chromatography and an anthocyanin glycoside was isolated. The results clearly indicate that the seeds of E. jambolana showed good analgesic activity in both the experimental models which could be due to the presence of anthocyanin compound and exert its effect predominantly through µ-opioid receptors. 113. Anti-inflammatory effect of methanolic extract of Gmelina arborea bark and its fractions against carrageenan induced paw oedema in rats.

Abstract

Gmelina arborea has been traditionally used for treatment of abdominal pain, burning sensation and as stomachic. The anti-inflammatory effect of aqueous and methanol extract of this plant has been studied but detailed investigations on anti-inflammatory activity of G. arborea stem bark are still not available. Therefore, the present study was designed to evaluate the anti-inflammatory activity of methanol extract and its fractions using carrageenan induced paw oedema model. Methanol extract at the dose of 500 mg/kg and its ethyl acetate fraction at 50 mg/kg showed significant reduction in paw oedema in comparison with standard drug. Ethyl acetate fraction was subjected to column chromatography which resulted in isolation of a new flavonoid (GM-01). Anti-inflammatory effect of methanol extract and its fractions can be attributed to the presence of the flavonoid GM-01. Further studies are underway for evaluation of anti-inflammatory effect of GM-01 and identification of active constituents present in ethyl acetate fraction. [ABSTRACT FROM AUTHOR]

Abstract

Rodent pups use vocalizations to communicate with one or both parents in biparental species, such as California mice (Peromyscus californicus). Previous studies have shown California mice developmentally exposed to endocrine disrupting chemicals, bisphenol A (BPA) or ethinyl estradiol (EE), demonstrate later compromised parental behaviors. Reductions in F1 parental behaviors might also be due to decreased emissions of F2 pup vocalizations. Thus, vocalizations of F2 male and female California mice pups born to F1 parents developmentally exposed to BPA, EE, or controls were examined. Postnatal days (PND) 2–4 were considered early postnatal period, PND 7 and 14 were defined as mid-postnatal period, and PND 21 and 28 were classified as late postnatal period. EE pups showed increased latency to emit the first syllable compared to controls. BPA female pups had decreased syllable duration compared to control and EE female pups during the early postnatal period but enhanced responses compared to controls at late postnatal period; whereas, male BPA and EE pups showed greater syllable duration compared to controls during early postnatal period. In mid-postnatal period, F2 BPA and EE pups emitted greater number of phrases than F2 control pups. Results indicate aspects of vocalizations were disrupted in F2 pups born to F1 parents developmentally exposed to BPA or EE, but their responses were not always identical, suggesting BPA might not activate estrogen receptors to the same extent as EE. Abstract

AbstractNumerous plants used in traditional system of medicine exhibit pharmacological properties with great potential for therapeutic applications in the treatment of anxiety disorders. The aerial parts of Melilotus officinalis(Fabaceae) have been used traditionally for the treatment of convulsions, insomnia and as a nervine tonic. Despite a long tradition of its use in nervous system disorders, no systematic phytochemical and pharmacological work has been carried out on this potential plant. The aim of the present study was to isolate the antianxiety constituent(s) from M. officinalisaerial parts following bioactivity guided fractionation approach. Ethanol extract was partitioned successively with ethyl acetate and butanol and all fractions were evaluated for antianxiety activity. The bioactive ethyl acetate fraction was further subjected to column chromatography to get seven fractions which were evaluated for antianxiety effect in elevated plus maze (EPM). The bioactive EAF6when subjected to column chromatography resulted in isolation of a flavonoid, which was characterized by standard spectral techniques and its anxiolytic activity was evaluated using EPM and mirror chamber model. TLC densitometric method was developed to quantify the isolated compound. The present study resulted in isolation of kaempferol, which exhibits potent anxiolytic activity at 2 mg/kg which was comparable to that of diazepam. The study validates the traditional use of the M. officinalisaerial parts as an anxiolytic. Kaempferol was found to be responsible for anxiolytic effect of M. officinalisaerial parts.
