Selenium (Se) is an important micronutrient but also a strong toxin with a narrow tolerance range for many organisms. As such, a globally heterogeneous Se distribution in soils is responsible for various disease patterns (i.e. Se excess and deficiency) and environmental problems, whereby plants play a key role for the Se entrance into the biosphere. Selenium isotope variations were proved to be a powerful tracer for redox processes and are therefore promising for the exploration of the species dependent Se metabolism in plants and the Se cycling within the Critical Zone. Plant cultivation setups enable systematic controlled investigations, but samples derived from them-plant tissue and phytoagar-are particularly challenging and require specific preparation and purification steps to ensure precise and valid Se isotope analytics performed with HG-MC-ICP-MS. In this study, different methods for the entire process from solid tissue preparation to Se isotope measurements were tested, optimized and validated. A particular microwave digestion procedure for plant tissue and a vacuum filtration method for phytoagar led to full Se recoveries, whereby unfavorable organic residues were reduced to a minimum. Three purification methods predominantly described in the literature were systematically tested with pure Se solution, high concentrated multi-element standard solution as well as plant and phytoagar as target matrices. All these methods efficiently remove critical matrix elements, but differ in Se recovery and organic residues. Validation tests doping Se-free plant material and phytoagar with a reference material of known Se isotope composition revealed the high impact of organic residues on the accuracy of MC-ICP-MS measurements. Only the purification method with no detectable organic residues, hydride generation and trapping, results in valid mass bias correction for plant samples with an average deviation to true δ82/76Se values of 0.2 ‰ and a reproducibility (2 SD) of ± 0.2 ‰. For phytoagar this test yields a higher deviation of 1.1 ‰ from the true value and a 2 SD of ± 0.1 ‰. The application of the developed methods to cultivated plants shows sufficient accuracy and precision and is a promising approach to resolve plant internal Se isotope fractionations, for which respective δ82/76Se values of +2.3 to +3.5 ‰ for selenate and +1.2 to +1.9 ‰ for selenite were obtained.

Im Jahr 2007 wurde der Vertrag von Malta in das niederländische Denkmalschutzgesetz integriert. Als Folge dieser Gesetzgebung ist in den Niederlanden der Ausgrabungssektor weitgehend privatisiert. Ausgrabungen werden seitdem von spezialisierten Firmen durchgeführt. Eigentümer der archäologischen Funde und der Dokumentation sind die Provinzen, auf deren Gebiet die Untersuchungen stattgefunden haben. Sie sind verpflichtet, ein Magazin für deren Aufbewahrung zu unterhalten. Die Grabungsfirmen müssen die Funde und die Dokumentation nach der Auswertung bei den provinzialen Depots abgeben. In der Provinz Gelderland ist dieses Depot beim Museum Het Valkhof in Nimwegen angesiedelt. Durch die Einführung des neuen Gesetzes ist in der Provinz Gelderland ein starker Anstieg an archäologischen Untersuchungen zu verzeichnen, wodurch das Depot mit akuter Raumnot zu kämpfen hat. In der Regel werden Funde, die einmal beim Depot angeliefert wurden, nicht mehr abgestoßen. Daher findet eine Selektion vor allem vor der Übergabe statt. Ziel ist es, die Fundmasse auf ein vertretbares Minimum zu reduzieren, das eine erneute Bewertung des Fundplatzes ermöglicht. Für diesen Selektionsprozess sind Richtlinien in der so genannten Kwaliteitsnorm Nederlandse Archeologie (KNA) aufgestellt. Auch die Provinz ist als Eigentümer der Funde an diesem Prozess beteiligt. Vor der Übergabe an das Depot müssen die Funde (digital) erfasst, konserviert und adäquat verpackt werden. Hierdurch ist das Material gut erschlossen und zugänglich. In der Praxis arbeitet dieses System gut, bedarf aber bezüglich der Selektionskriterien und des Umgangs mit den abgestoßenen Funden einer feineren Ausarbeitung., In 2007 the Malta Treaty was integrated in the Dutch laws on the protection and preservation of monuments and buildings. As a result of this legislation the Dutch excavation sector is largely privatised and excavations are carried out by specialised contractors. The county in which an excavation-site is located is owner of the archaeological finds and documentation and obliged to keep an (archaeological) depot in which finds and documentation are stored (for future reference). The excavation firms have to hand in the archaeological material and documentation after analysis and interpretation. In the county Gelderland the depot is located in the Museum Het Valkhof in Nijmegen. The introduction of the new Malta Treaty has led to an increase in archaeological excavations, which resulted in an acute shortage of storage space. According to the rule, that finds, which are delivered at the depot will not be refused, the selection has to take place before the transfer of the material into the depot. The goal is to reduce the quantity of excavated material to an acceptable minimum, which should still allow a new evaluation of the site if necessary. The criteria for the selection process are laid down in the so-called Kwaliteitsnorm Nederlandse Archeologie (KNA). The county, being the owner of the finds, is taking part in this process. Before the transfer, the finds must be (digitally) described, preserved and adequately packed for storage, so that the material is evaluated and accessible. In practice, this system works well, but for the definition of the selection criteria and the handling of the de-selected finds further elaboration is required. (Translation: M. Magnée-Nentjes)
