In de teelt van aardbeien vormt vruchtrot, veroorzaakt door Botrytis cinerea, een belangrijk probleem. Van jaar tot jaar verschilt de mate van aantasting, en daarmee het opbrengstverlies, zeer sterk. Chemische bestrijding van vruchtrot behoort tot de mogelijkheden. Daarnaast vormen trips en meeldauw ook bedreigingen voor de teelt. Het beleid in Nederland is erop gericht de afhankelijkheid van chemische gewasbescherming te verminderen. Eén van de mogelijkheden is de ontwikkeling van een strategie ter bevordering van de inzet van antagonisten of Gewasbeschermingsmiddelen van Natuurlijke Oorsprong (GNO’s). Daarnaast is het van belang alleen dan een chemische bestrijding uit te voeren als dit noodzakelijk is. Beslissing Ondersteunende Systemen (BOS) kunnen daarbij helpen. Eén van de best practises is het toepassen van fungicidenbestrijding op basis van een waarschuwingsysteem. In de praktijk wordt vruchtrot bestrijding op basis van een BOS nog onvoldoende overgenomen. In de periode 2006 tot en met 2008 werd onderzoek gedaan naar gebruik van BOS-varianten bij de bestrijding van vruchtrot en meeldauw in aardbei. Doel van deze proeven is het verbeteren en testen van BOS-varianten om gebruik hiervan in de praktijk te stimuleren. Naast het gebruik van waarschuwingssystemen werd ook gekeken naar de toepassing van een GNO tegen meeldauw, met mogelijk een nevenwerking op Botrytis en naar een antagonist tegen Botrytis

Wageningen UR heeft een schimmel geïsoleerd, waaruit een isolaat wordt gehaald (H39) dat de schurftschimmel in appelbomen zeer goed blijkt te bestrijden. Met dit isolaat, aanwezig in vinasse, zijn al enkele jaren veldproeven gedaan. Toch is nog een lange weg te gaan voordat het product de commerciële markt op kan, zegt onderzoeker Jürgen Köhl van Plant Research International in Wageningen. Appelschurft, veroorzaakt door de schimmel Venturia inaequalis, is een groot probleem in de appelteelt. Schurft wordt in de biologische appelteelt wereldwijd veelal bestreden met koperhoudende fungiciden. Europese milieuregelgeving wil het gebruik van dit soort fungiciden terugdringen. In Nederland is de toepassing van deze middelen al verboden. De ziekte geeft schade aan het blad en de vruchten. Voor een rendabele biologische appelteelt zijn alternatieve bestrijdingsmiddelen gewenst. Appelschurft Wageningen UR heeft een antagonistische schimmel geïsoleerd die de schurftschimmel in appelbomen zeer goed blijkt te bestrijden. Het onderzoek is uitgevoerd in het kader van het EU-project REPCO (=Replacement of Copper Fungicides in Organic Production of Grapevine and Apple in Europe.) Het gevonden isolaat voldoet aan alle gestelde criteria en wordt nu door een commercieel bedrijf verwerkt tot een pilotproduct. Toepassingen van het alternatieve middel op de bladeren in de zomer bleek een vermindering van 50 tot 80% van de de schimmelsporenproductie te geven. Final Activity Report: The objective of REPCO was to contribute to the replacement of copper fungicides in organic agriculture by new measures for control of downy mildew (Plasmopara viticola) in grapevine and scab (Venturia inaequalis) in apple. Both major European organic crops strongly depend on copper fungicides. Permitted amounts will be reduced stepwise in the years ahead (Council Regulation (EEC) 2092/91, Annex II) to avoid environmental risks. In European countries where copper fungicides are already out of use, production of organic apples suffers severe economic problems because of insufficient scab control. Potentiators of resistance, organically based fungicides and biocontrol agents have been screened and evaluated in grapevine and apple. The risk of pathogen evolution during use of novel control measures has been estimated to enable the development of sustainable strategies. Effects of crop management practices in organic agriculture on overwintering of Venturia inaequalis were assessed. Novel disease control measures and knowledge have been integrated into organic management systems. 'Pipeline' products already under development elsewhere have been included and where necessary optimised in their use. Implementation by end-users and industries qualified for commercialisation of project findings has strongly been emphasised. Small-medium enterprises (SME) as project partners ensured a strong link between end-users and research. At the end of the project several compounds and biocontrol agents can be delivered to qualified industries for development of products for use in organic agriculture. Additionally, knowledge of integrated use of control measures can be delivered to organic growers. The project results thus strongly support EU policies to replace the use of copper fungicides in organic agriculture in the near future.

Telers en handel vrezen de zwarte-vlekkenziekte in bewaarpeen, vooral omdat onvoorspelbaar is wanneer een aantasting ontstaat. Op peen die blakend van gezondheid de koelcel in gaat en na enkele maanden nog gezond is, kunnen zich zwarte vlekken manifesteren nadat de peen uit de bewaring is gehaald

In deze studie wordt beschreven hoe en onder welke omgevingscondities een schimmelaantasting zich vestigt en verspreidt in de kas. De schimmel die in glastuinbouwgewassen de meeste schade veroorzaakt is Botrytis cinera. Botrytis heeft beschadighd of zwak weefsel nodig om in te groeien, in tegenstelling tot bijvoorde meeldauw die op levend weefsel kan parasiteren. Dit betekent dat wonden aan de plant en de organen met weinig weerstand, zoals rijpe bloemen en vruchten, sterk vatbaar zijn. Botrytis heeft geen specifieke voorkeur voor een bepaalde plantensoort. Naast Botrytis zorgen ook Mycosphaerella in komkommer en Fusarium in paprika voor een belangrijk deel van de schimmelschade in de glastuinbouw. Net als bij Botrytis geldt dat beide schaimmeltypen gerelateerd zijn aan de lokale klimaatomstandigheden en ze vermoedelijk via een gerichte klimaatsturing onder controle gehouden kunnen worden.

Zwarte vlekkenziekte in koud bewaarde peen treedt altijd onverwacht op en pas als de peen uit de bewaring komt. Oorzaak zijn verschillende schimmels die op verschillende tijdstippen toeslaan. Bij de zaadproductie, teelt, bewaring en veredeling vergt iedere schimmel zijn eigen aanpak. Maar daarover is nog weinig bekend. Economische schade is deels te voorkomen door tijdens de bewaring via een test uit te zoeken welke partijen het meest besmet zijn en die eerder te verkopen.

Integrated disease management in organic farming combines the use of various measures. The usefulness of certain measures depends on the specific crop-pathogen combination. In many crops, preventative measures can control diseases without the need of plant protection products. However, for certain disease problems, preventative measures are not sufficient. For example, organic apple production strongly depends on the multiple use plant protection products.

Novel antagonists for biological control of Venturia inaequalis causing apples scab were screened. Several hundred fungal isolates were obtained from sporulating colonies of V. inaequalis on apple leaves collected at various locations. Candidate antagonists were pre-screened to exclude those isolates with potential risks or low economical feasibility. Remaining isolates were tested on apple seedlings for their ability to reduce conidiation of V. inaequalis. Several promising antagonists could be selected and will be tested under field conditions in summer 2006.

The use of organic propagation material is obligatory according to the current EU regulations for organic production. However, frequently difficulties are en-countered regarding the availability, the costs or the quality. In the Netherlands a national research program aims at developing solutions, needed for improving the production of high quality organic propagation material. The main emphasis in the pro-gram is on the model crops cabbage, onion, wheat and potato. Individual projects within the program include epidemiological studies of seed borne dis-eases to develop disease prevention strategies, meth-ods to improve resistance of seeds and seedlings towards pathogens and methods to control silver scurf in seed potatoes, development of multi-spectral analysis and sorting techniques and methods for analysis and improvement of seed vigour. Active involvement of producers and users of the seeds or seed potatoes ensures that the results will be imple-mented in practice. The program also aims at strengthening the international collaboration, amongst others through involvement in international research projects.

Xanthomonas campestris pv. campestris (Xcc) is a seed borne pathogen that causes black rot, a destructive disease of cabbage. Exclusion of infec-tions is the most effective way to control black rot in organic seed production. Within this frame, the role of insects in transmission of Xcc to seed was deter-mined. It was found that Xcc survived for three days on flies (Calliphora vomitoria), used for pollination of cabbage plants in tunnels and greenhouses. In tunnel experiments, both the use of Xcc-contaminated flies and brush inoculation of flowers with Xcc resulted in internal seed infections. The implications for seed production are discussed.

Improved prevention and control of apple scab caused by Venturia inaequalis is aimed at without the use of copper containing products in the Repco-project. Substantial progress is made in selection of potential products against summer epidemics. A patent application is made for E73. New effective biocontrol agents are selected to reduce inoculum during winter. The product potassium bicarbonate has shown good efficacy and Repco contributes to the registration of this product in Europe. Earthworms tended to be stimulated to consume apple leaves treated with amino acids or beetpulp, especially when applied fresh under controlled environmental condi-tons.

Venturia inaequalis causes apple scab. The control of the disease in organic apple production depends on the use of copper which will be restricted in future for environmental reasons. In the search for environmental friendly microbial biocontrol agents and stimulation of antagonistic populations in situ the molecular technique of DGGE fingerprinting can is applied. This fingerprinting technique allows to bypass culturing techniques and even can identify not yet culturable organisms. Comparison of microbial fingerprints and pathogen development in the leaf samples during winter and spring has been performed to identify populations with antagonistic potential. Such potential antagonistic organisms can be further identified by their specific DNA sequence. Preliminary results identify possible antagonists. The identification based on the sequence can help to focus on specific isolation or stimulation of a possible antagonist or a consortium of antagonists.

An important problem in organic carrot production in the Netherlands is the occurrence of black spots during storage. Several different fungal pathogens cause similar symptoms, which are collectively known as black spot diseases. We identified Alternaria radicina, A. dauci and Rhexocercosporidium carotae (syn. Acrothecium carotae) as the most prevalent black spot pathogens. We developed laboratory test methods for resistance to A. radicina and R. carotae, and assessed resistance in a collection of cultivated varieties and more exotic material

Production of high quality organic vegetable seeds encounters several challenges. Research is performed to support seed companies in producing vigorous and healthy organic vegetable seeds. Examples are provided with respect to research on seed vigour, determining critical control points to avoid disease transmission to the seeds, seed treatments with natu-ral compounds, new seed sorting techniques and enhancement of the natural plant defence. It is noticed that the restrictions in EU regulation 2092/91 on organic production are at present blocking the use of promising natural and sustainable components for seed treatments.

Dark leaf spot caused by Alternaria brassicicola is a seed-borne disease of Brassicae. Production of healthy seed is essential for the organic vegetable production. Literature on the epidemiology of the disease in organic seed production of Brassica was reviewed and an epidemiological field experiments was carried out. External and internal contamination of seeds with A. brassicicola increased steadily during their development. Colonisation of pod tissues as quantified by TaqMan-PCR increased exponentially. The developed knowledge can be used for optimizing cropping systems for organic seed production with lower risks for seed contamination by Alternaria spp. and to develop critical control points for disease management.

In the Netherlands winter carrot for the fresh market is a cash crop for many organic growers of field vegetables. In the last few years blackish spots have been observed during storage of carrot roots. An inventory was carried out in 2001/2002 and 2002/2003 including in total 42 carrot lots from 22 growers. Statistical analysis was performed by multiple regression analysis to identify factors and variables that best explained the occurrence of black spots on stored carrots. Rhexocercosporidium carotae was the dominating pathogen in black spots of carrots harvested in 2001. From carrots harvested in 2002, Alternaria radicina was isolated most frequently, followed by A. dauci and R. carotae. Multiple regression analysis indicated that the occurrence of R. carotae was associated with wild umbelliferous plants or carrot production during the previous year in the neighbourhood of the investigated carrot fields. A high occurrence of the symptoms of black spots was associated with increasing damage caused during harvest or with higher temperatures during harvest.

PRI heeft in 2001 en 2002 onderzocht welke teeltfactoren van invloed kunnen zijn op het optreden van zwarte vlekken in bewaarpeen, een verschijnsel dat de laatste jaren steeds vaker voorkomt bij biologische telers in de IJsselmeerpolders. Studies were conducted in annual crops of strawberry (Fragaria x ananassa) (cv. Elsanta to assess the relative importance of petals as an inoculum source of grey mould, caused by Botrytis cinerea and to identify during which period of flower and fruit development the presence of petals has a significant effect on development of grey mould on fruits. In 1998, the incidence of B. cinerea on flower parts was assessed, and the symptoms of grey mould on fruits were characterised with regards to their starting point. The incidence of B. cinerea on petals was 65-85% of those flowers that harboured B. cinerea. The starting point of symptoms was located underneath the sepals in 65-85% of fruits with grey mould, and petals were present at this site in about 50% of the cases. In 1999, four field experiments were conducted to assess the effect of petal removal at different stages of flower and fruit development on incidence of grey mould. The incidence of grey mould on fruits with petals retained till harvest was circa 55% more than on fruits where petals were removed or had dropped naturally by the end of flowering, regardless of planting date or inoculum level of B. cinerea. The incidence of grey mould was hardly different between treatments where petals were removed at young flower stage, old flower stage, or green fruit stage. It is concluded that petals are an effective and stable source of inoculum for fruit infection by B. cinerea. Considering the high probability that retention of petals during fruit expansion and ripening results in fruit infection, the elimination of petals as an inoculum source appears worthwhile even when the incidence of petal retention on fruits is relatively low

De gele bladvlekkenziekte (DTR) is in 1999 en 2000 respectievelijk op 75% en 25% van de geïnventariseerde percelen aangetroffen. De aantasting was gering; toch wijzen de ernstige aantastingen die op enkele percelen zijn aangetroffen op een potentieel gevaar. Het onderzoek heeft aangetoond dat het risico van een zware DTR-aantasting, naast ruime vruchtwisseling, sterk vermindert door intensieve grondbewerking waardoor de eerste infecties door ascosporen van besmette tarwestoppels onmogelijk worden

The re-emergence of fusarium head blight throughout the world and especially in Western Europe prompted a survey of the situation in the Netherlands. To allow for a high throughput screening of large numbers of samples, a diagnostic PCR method was developed to detect the most common species of Fusarium occurring on wheat. Seven primer pairs were tested for their ability to identify isolates of Fusarium avenaceum, F. culmorum, F. graminearum, F. poae, F. proliferatum and Microdochium nivale var. majus and M. nivale var. nivale. Each primer pair only generated a PCR product with the corresponding Fusarium species and all PCR fragments had different molecular sizes. This allowed the generation of these amplicons using a mixture of all seven primer pairs. The robustness of this multiplex PCR encouraged us to screen a large series of isolates collected in 2000 and 2001. In both years 40 fields were sampled leading to a collection of 209 isolates from 2000 and 145 isolates from 2001. The results of the multiplex PCR demonstrated that F. graminearum was the most abundant species in the Fusarium complex on wheat in both years. This is in sharp contrast to reports from the 1980s and early 1990s, which found F. culmorum as the predominant species. Primers derived from the tri7 and tri13 genes, which are implicated in the acetylation and oxygenation of the C-4 atom of the backbone of the trichothecene molecule, were used to discriminate between deoxynivalenol and nivalenol (NIV) producers.
