Evidence regarding screen use and outdoor activity during very early childhood (i. e., from aged 1 to 3 years) and their potential combined links to the later preschool myopia is limited. This information is needed to release effective public health messages and propose intervention strategies against preschool myopia. We collected information regarding very early childhood screen use, outdoor activity and the kindergartens vision screenings of 26,611 preschoolers from Longhua Child Cohort Study by questionnaires. Logistic regression models were used to examine the associations between reported outdoor activity, screen use from 1 to 3 years of age, and preschool myopia. Throughout very early childhood, from 1 to 3 years, the proportion of children exposed to screens increased (from 35.8 to 68.4%, p < 0.001), whereas the proportion of children who went outdoors ≥7 times/week (67.4–62.1%, p < 0.001) and who went outdoors for ≥60 min/time (53.3–38.0%, p < 0.001) declined. Exposure to fixed screen devices [adjusted odds ratio (AOR) = 2.66, 95% confidence interval (CI) = 2.09–3.44], mobile screen devices (AOR = 2.76, 95% CI = 2.15–3.58), and limited outdoor activity (AOR = 1.87, 95% CI = 1.42–2.51) during early childhood were associated with preschool myopia. Among children whose parents were myopic, the interactions between outdoor activity and fixed or mobile screen use on later preschool myopia were significant; the ORs and 95% CI were 3.34 (1.19–9.98) and 3.04 (1.06–9.21), respectively. Our findings suggest the possibility that the impact of screen exposure during early childhood on preschool myopia could be diminished by outdoor activity for children whose parents have myopia.

Background: αVβ3 integrin has been implicated in the physiological processes and pathophysiology of important angiogenesis-related disorders; however, the preclinical and clinical data on integrin αVβ3 antagonists have not demonstrated improved outcomes. Our goal was to test the hypothesis that inhibition of αVβ3 integrin improves blood flow in a mouse hindlimb ischemia model.Methods: In this study, we examined the effect of cilengitide, an αVβ3/αVβ5 integrin-specific RGD-mimetic cyclic peptide, on blood perfusion and angiogenesis after hindlimb ischemia. Blood flow was measured using Laser Doppler Scanner. Vascular density, and macrophages infiltration were examined by immunofluorescence. Macrophage polarization was measured by quantitative real time PCR.Results: We found that low-dose, not high-dose, cilengitide increased blood flow perfusion, capillary formation, and pericyte coverage, accompanied by an accumulation of macrophages and increased expression of the chemokine (C-C motif) ligand 2 (CCL2) in ischemic muscles. Macrophage depletion using clodronate liposomes resulted in a reduction in low-dose cilengitide-induced blood flow perfusion, macrophage accumulation, pericyte coverage, and CCL2 expression. Finally, in vitro assays showed that low-dose, not high-dose, cilengitide increased macrophage migration.Conclusion: These studies identified a novel role of the inhibition of αVβ3 integrin in modulating ischemia-induced angiogenesis, possibly through effects on macrophage infiltration and polarization, and revealed αVβ3 integrin inhibition to be a promising therapeutic strategy for peripheral artery disease.

Background: The pharmacological inhibition of dipeptidyl peptidase-4 (DPP-4) potentiates incretin action, and DPP-4 is a drug target for type 2 diabetes and reducing cardiovascular risk. However, little is known about the non-enteroendocrine pathways by which DPP-4 might contribute to ischaemic cardiovascular events.Methods: We tested the hypothesis that inhibition of DPP-4 can inhibit platelet activation and arterial thrombosis by preventing platelet mitochondrial dysfunction and release. The effects of pharmacological DPP-4 inhibition on carotid artery thrombosis, platelet aggregation, and platelet mitochondrial respiration signaling pathways were studied in mice.Results: Platelet-dependent arterial thrombosis was significantly delayed in mice treated with high dose of linagliptin, a potent DPP-4 inhibitor, and fed normal chow diet compared to vehicle-treated mice. Thrombin induced DPP-4 expression and activity, and platelets pretreated with linagliptin exhibited reduced thrombin-induced aggregation. Linagliptin blocked phosphodiesterase activity and contrained cyclic AMP reduction when thrombin stimulates platelets. Linagliptin increases the inhibition of platelet aggregation by nitric oxide. The bioenergetics profile revealed that platelets pretreated with linagliptin exhibited decreased oxygen consumption rates in response to thrombin. In transmission electron microscopy, platelets pretreated with linagliptin showed markedly reversed morphological changes in thrombin-activated platelets, including the secretion of α-granules and fewer mitochondria.Conclusion: Collectively, these findings identify distinct roles for DPP-4 in platelet function and arterial thrombosis.

The sluggish kinetics of the oxygen reduction reaction (ORR) on electrocatalysts represents a major obstacle in the development of fuel cell technology. A tremendous amount of work has reported the increasing ORR activity for catalysts. Nevertheless, when applied to practical Membrane Electrode Assembly (MEA, an assembled stack of a proton exchange membrane fuel cell) configuration, the high-performance catalysts on the rotating disk electrode (RDE) may not display the same high activity as in the lab-scale tests. This led us to reexamine the ORR evaluation based on the RDE technique. With the development of high active electrocatalysts, it may become significant to determine the reasonable kinetic current at a conventional fixed potential approaching the limited current by using the Koutecky-Levich (K-L) technique on RDE for the evaluation of ORR activity. Here we describe such a challenging situation and systematically discuss the proper kinetic region when comparing the ORR activity with the unsuitable potential or Pt loading based on the K-L technique. Furthermore, the rational benchmarking guidelines are given for the evaluation of the ORR electrocatalysts.

MoS2–TiL/MoS2–TiH coating (L and H are low and high power of the sputtered Ti target) possesses excellent tribological properties owing to its nano-multilayer structure. In this study, the tribological properties of MoS2–TiL/MoS2–TiH coating were extensively analyzed at different loads. Relative to the MoS2–Ti monolayer coating, the nano-multilayer structure not only improved the wear resistance but also increased the critical load at which the coating began to peel off. The MoS2–Ti coating maintained its lubrication only in the early stage of the test even at a load of 2 N. In contrast, the friction coefficient and wear rate of the multilayer coating were small and stable until the load reached 20 N. The critical load of the optimized MoS2–TiL/MoS2–TiH coating was 10 times that of the MoS2–Ti coating. At excessive loads, the multilayer coating lost its lubrication in the early stage of the test because the coating was completely worn off under such loading.

Factor V (FV) is a critical component in the blood coagulation cascade. In patients, FV inhibitors have been reported to be associated with malignancy. FV is present in plasma and platelets, which exhibit physical and functional differences. However, the functions of FV in cancer progression remain poorly understood. We evaluated the impact of different levels of FV in plasma and platelets on the haematogenous mouse pulmonary metastasis model to determine whether FV determines the metastatic potential of circulating tumor cells. The role of platelet-derived FV was evaluated using a murine B16F10 pulmonary metastasis model, an assay of tumor cell adhesion to endothelial cells, and western blotting. By combining genetic models and FV inhibitory antibody, the transgenic mice with lower platelet FV expression showed significant increases in metastases compared with mice with higher platelet FV expression. In vitro, labeled B16F10 melanoma cells appeared to exhibit increased adhesion to endothelial cells that were treated with lower levels of platelet FV, but not platelet-poor plasma. Furthermore, platelets from mice with lower platelet FV levels expressed TFPIα at lower levels than with mice with higher platelet FV expression. Based on these findings, platelet-derived FV contributes to haematogenous pulmonary metastasis and is associated with the regulation of tumor cell adhesion to the vessel wall.

Previous studies have dealt with Cr and its alloy films that exhibit promising characteristics as surface modification layers for antiwear, anticorrosive, and decorative applications. However, the effect of Ti alloying on the structure and mechanical properties of Cr films has not been studied. This work aimed to the structure and mechanical properties of Cr-Ti alloy films in the Cr-rich side. To this end, pure Cr, Cr-6 at.% Ti, Cr-11 at.% Ti, Cr-16 at.% Ti, and Cr-21 at.% Ti alloy films were prepared by magnetron sputtering, and the structure and mechanical properties of the films were evaluated. The results indicated that all the films exhibited a Cr-based growth with body-centered cubic structure, and increasing the Ti content decreased the (110) orientation growth of Cr basis. Ti alloying increased the hardness of the films, while leaded to a monotonic decrease in the modulus of the films. The first-principles method was employed to demonstrate that the reduced modulus was determined by the Ti alloying degree, rather than the orientation evolution of the films. The analysis of H/E value suggested that the wear resistance of the films was improved by Ti alloying. The mechanical properties of present Cr-Ti alloy films, and other Cr-based alloy films or metallic glasses in publications were compared and discussed. We proposed that Ti alloying is a considerable way to explore advanced mechanical properties of Cr-based alloy films.

Ampelopsis grossedentata (AG) is an ancient medicinal plant that is mainly distributed and used in southwest China. It exerts therapeutic effects, such as antioxidant, anti-diabetic, and anti-inflammatory activities, reductions in blood pressure and cholesterol and hepatoprotective effects. Researchers in China recently reported the anti-obesity effects of AG extract in diet-induced obese mice and rats. To verify these findings, we herein investigated the effects of AG extract and its principal compound, ampelopsin, in high-fat diet (HFD)- and alcohol diet-fed mice, olive oil-loaded mice, and differentiated 3T3-L1 cells. The results obtained showed that AG extract and ampelopsin significantly suppressed increases in the weights of body, livers and abdominal fat and also up-regulated the expression of carnitine palmitoyltransferase 1A in HFD-fed mice. In olive oil-loaded mice, AG extract and ampelopsin significantly attenuated increases in serum triglyceride (TG) levels. In differentiated 3T3-L1 cells, AG extract and ampelopsin promoted TG decomposition, which appeared to be attributed to the expression of hormone-sensitive lipase. In alcohol diet-fed mice, AG extract and ampelopsin reduced serum levels of ethanol, glutamic oxaloacetic transaminase (GOT), and glutamic pyruvic transaminase (GPT) and liver TG. An examination of metabolic enzyme expression patterns revealed that AG extract and ampelopsin mainly enhanced the expression of aldehyde dehydrogenase and suppressed that of cytochrome P450, family 2, subfamily e1. In conclusion, AG extract and ampelopsin suppressed diet-induced intestinal fat accumulation and reduced the risk of fatty liver associated with HFD and alcohol consumption.

Volumetric muscle defect, caused by trauma or combat injuries, is a major health concern leading to severe morbidity. It is characterized by partial or full thickness loss of muscle and its bio-scaffold, resulting in extensive fibrosis and scar formation. Therefore, the ideal therapeutic option is to use stem cells combined with bio-scaffolds to restore muscle. For this purpose, muscle-derived stem cells (MDSCs) are a great candidate due to their unique multi-lineage differentiation potential.In this study, we evaluated the regeneration potential of MDSCs for muscle loss repair using a novel in situ fibrin gel casting. Muscle defect was created by a partial thickness wedge resection in the tibialis anterior (TA) muscles of NSG mice which created an average of 25% mass loss. If untreated, this defect leads to severe muscle fibrosis. Next, MDSCs were delivered using a novel in situ fibrin gel casting method.Our results demonstrated MDSCs are able to engraft and form new myofibers in the defect when casted along with fibrin gel. LacZ labeled MDSCs were able to differentiate efficiently into new myofibers and significantly increase muscle mass. This was also accompanied by significant reduction of fibrotic tissue in the engrafted muscles. Furthermore, transplanted cells also contributed to new vessel formation and satellite cell seeding.These results confirmed the therapeutic potential of MDSCs and feasibility of direct in situ casting of fibrin/MDSC mixture to repair muscle mass defects. Developing cost-effective advanced carbon anode is critical for innovation of sodium ion batteries. Herein, we develop a powerful combined method for rational synthesis of free-standing binder-free carbon nanospheres arrays via chemical bath plus hydrothermal process. Impressively, carbon spheres with diameters of 150–250 nm are randomly interconnected with each other forming highly porous arrays. Positive advantages including large porosity, high surface and strong mechanical stability are combined in the carbon nanospheres arrays. The obtained carbon nanospheres arrays are tested as anode material for sodium ion batteries (SIBs) and deliver a high reversible capacity of 102 mAh g−1 and keep a capacity retention of 95% after 100 cycles at a current density of 0.25 A g−1 and good rate performance (65 mAh g−1 at a high current density of 2 A g−1). The good electrochemical performance is attributed to the stable porous nanosphere structure with fast ion/electron transfer characteristics.

Advancements in reprogramming somatic cells into induced pluripotent stem cells (iPSCs) have provided a strong framework for in vitro disease modeling, gene correction and stem cell-based regenerative medicine. In cases of skeletal muscle disorders, iPSCs can be used for the generation of skeletal muscle progenitors to study disease mechanisms, or implementation for the treatment of muscle disorders. We have recently developed an improved directed differentiation method for the derivation of skeletal myogenic progenitors from hiPSCs. This method allows for a short-term (2 weeks) and efficient skeletal myogenic induction (45–65% of the cells) in human pluripotent stem cells (ESCs/iPSCs) using small molecules to induce mesoderm and subsequently myotomal progenitors, without the need for any gene integration or modification. After initial differentiation, skeletal myogenic progenitors can be purified from unwanted cells using surface markers (CD10+CD24−). These myogenic progenitors have been extensively characterized using in vitro gene expression/differentiation profiling as well as in vivo engraftment studies in dystrophic (mdx) and muscle injury (VML) rodent models and have been proven to be able to engraft and form mature myofibers as well as seeding muscle stem cells. The current protocol describes a detailed, step-by-step guide for this method and outlines important experimental details and troubleshooting points for its application in any human pluripotent stem cells.

Real-time measurements of carbonate ion concentrations in the ocean are critical to advancing marine environmental monitoring and research into deep-sea hydrothermal activity. Herein, we report the first example of deep-sea hydrothermal field exploration using a carbonate ion-selective electrode (ISE). The novel carbonate ISE was composed of a Ni wire as substrate, carbon film as transducers and carbonate-selective membrane layers. This paper describes the preparation process of the electrode and characterises its performance via scanning electron microscopy (SEM) and electrochemical analysis. The detection limit of the electrode for CO32− is 2.821 × 10−6 mol/L, the linear response range is 1.0 × 10−5–1.0 × 10−1 mol/L and the Nernst slope was −30.4 mV/decade. In April 2021, the carbonate ISE was mounted on multi-parameter sensors with pH and Eh (redox) electrodes for the search of hydrothermal activity at the Southwest Indian Ridge. The simultaneous potential anomalies appeared at this carbonate electrode with the pH and Eh electrodes when passing through the hydrothermal field. The study of the hydrothermal field was supported by the in situ camera video and the sulphide samples. Additionally, the carbonate electrode provides enhanced information of water chemistry for the study of the hydrothermal field.

Magnetic flux leakage (MFL) testing has been widely used as a non-destructive testing method for various materials. However, it is difficult to separate the influences of the defect geometrical parameters such as depth, width, and length on the received leakage signals. In this paper, a “near-field” MFL method is proposed to quantify defect widths. Both the finite element modelling (FEM) and experimental studies are carried out to investigate the performance of the proposed method. It is found that that the distance between two peaks of the “near-field” MFL is strongly related to the defect width and lift-off value, whereas it is slightly affected by the defect depth. Based on this phenomenon, a defect width assessment relying on the “near-field” MFL method is proposed. Results show that relative judging errors are less than 5%. In addition, the analytical expression of the “near-field” MFL is also developed.

Solar-thermal energy storage based on phase-change materials suffers from slow thermal-diffusion-based charging. Here the authors alleviate this issue by introducing optical absorbers and controlling their distribution to accelerate charging process and thus improve solar-thermal energy conversion.

Recently, a new type of limb-girdle muscular dystrophy (LGMD type 2Z) has been identified due to a missense mutation in POGLUT1 (protein O-glucosyltransferase-Rumi), an enzyme capable of adding glucose to a distinct serine residue of epidermal growth factor-like repeats containing a C-X-S-X-(P/A)-C consensus sequence such as Notch receptors. Affected patients demonstrate reduced Notch signaling, decreased muscle stem cell pool and hypoglycosylation of α-dystroglycan, leading to LGMD phenotype. Here we report the generation and characterization of an iPSC line (CSCRMi001-A) from a LGMD-2Z patient with missense mutation in POGLUT1 which can be used for in vitro disease modeling.

Recent reports have documented the differentiation of human pluripotent stem cells toward the skeletal myogenic lineage using transgene- and cell purification-free approaches. Although these protocols generate myocytes, they have not demonstrated scalability, safety, and in vivo engraftment, which are key aspects for their future clinical application. Here we recapitulate one prominent protocol, and show that it gives rise to a heterogeneous cell population containing myocytes and other cell types. Upon transplantation, the majority of human donor cells could not contribute to myofiber formation. As a proof-of-principle, we incorporated the inducible PAX7 lentiviral system into this protocol, which then enabled scalable expansion of a homogeneous population of skeletal myogenic progenitors capable of forming myofibers in vivo. Our findings demonstrate the methods for scalable expansion of PAX7+ myogenic progenitors and their purification are critical for practical application to cell replacement treatment of muscle degenerative diseases.

In this paper, a set of online measurement devices of multi-electrochemical sensor was investigated. Combined with industrial distributed control system, it was first applied in extracting bromine from seawater to realize the real-time adjustment of production process parameters. In the process of extracting bromine from seawater, the pH value of acidified raw brine, the addition amount of Cl2 in the oxidation stage and the addition amount of SO2 in the absorption stage are key parameters to control the whole production process. The multi-electrochemical sensor realized a rapid and high-throughput detection of the above parameters by integrating an all-solid-stage bromide ion selective electrode (Br-ISE), Eh electrode and pH electrode. The Br-ISE and the pH electrode were self-developed electrodes and the Pt electrode was Eh electrode. The pH electrode was used to control the addition amount of H2SO4 during the acidification of the brine. The Eh electrode was used to control the addition amount of Cl2 during the oxidation stage and the addition amount of SO2 during the absorption stage. The Br-ISE was used to monitor the Br− concentration change in the raw brine. Results showed the optimum range of Eh in the oxidation stage and absorption stage of brine were 950–1000 mV and 580–610 mV, respectively. The application of multi-electrochemical sensor in industrial bromine production can realize real-time control of material addition and save the cost of production.

It is known that eddy current effect has a great influence on magnetic flux leakage testing (MFL). Usually, contact-type encoder wheels are used to measure MFL testing speed to evaluate the effect and further compensate testing signals. This speed measurement method is complicated, and inevitable abrasion and occasional slippage will reduce the measurement accuracy. In order to solve this problem, based on eddy current effect due to the relative movement, a speed measurement method is proposed, which is contactless and simple. In the high-speed MFL testing, eddy current induced in the specimen will cause an obvious modification to the applied field. This modified field, which is measured by Hall sensor, can be utilized to reflect the moving speed. Firstly, the measurement principle is illustrated based on Faraday's law. Then, dynamic finite element simulations are conducted to investigate the modified magnetic field distribution. Finally, laboratory experiments are performed to validate the feasibility of the proposed method. The results show that Bmz(r1) and Bmx(r2) have a linear relation with moving speed, which could be used as an alternative measurement parameter. [ABSTRACT FROM AUTHOR]

Abstract Background The metastatic cascade is a complex and multistep process with many potential barriers. Recently, miR-193a has been reported to be a suppressive miRNA in multiple types of cancers, but its underlying anti-oncogenic activity in non-small cell lung cancers (NSCLC) is not fully elucidated. Methods The expressions of miR-193a (miR-193a-5p) in human lung cancer tissues and cell lines were detected by real-time PCR. Dual-luciferase reporter assay was used to identify the direct target of miR-193a. Cell proliferation, apoptosis, and metastasis were assessed by CCK-8, flow cytometry, and Transwell assay, respectively. Results The expression of miR-193a in lung cancer tissues was decreased comparing to adjacent non-tumor tissues due to DNA hypermethylation in lung cancer tissues. Ectopic expression of miR-193a inhibited cell proliferation, colony formation, migration, and invasion in A549 and H1299 cells. Moreover, overexpression of miR-193a partially reversed tumor growth factor-β1 (TGF-β1)-induced epithelial-to-mesenchymal transition (EMT) in NSCLC cells. Mechanistically, miR-193a reduced the expression of WT1, which negatively regulated the protein level of E-cadherin, suggesting that miR-193a might prevent EMT via modulating WT1-E-cadherin axis. Importantly, knockdown of WT1 resembled the anti-cancer activity by miR-193a and overexpression of WT1 partially reversed miR-193a-induced anti-cancer activity, indicating that WT1 plays an important role in miR-193a-induced anti-cancer activity. Finally, overexpression of miR-193a decreased the growth of tumor xenografts in mice. Conclusion Collectively, our results have revealed an important role of miR-193a-WT1-E-cadherin axis in metastasis, demonstrated an important molecular cue for EMT, and suggested a therapeutic strategy of restoring miR-193a expression in NSCLC.
