Abstract

A genome-wide survey of mosaic genomic variation obtained by analyzing illumina 1M SNP array data from blood or buccal DNA samples of 1991 adult individuals from the Spanish Bladder Cancer/EPICURO genome-wide association study is presented. It was recommended to consider somatic mosaicism in the expanding repertoire of inter- and intraindividual genetic variation, some of which might cause somatic human disease and might contribute to modifying inherited disorders and/or late-onset multifactorial traits.

Abstract

Power is an important component of general health, fitness, and athletic performance. Traditional overload techniques require considerable time, intensity, and volume of training. Whole-body vibration (WBV) is a potentially less time-consuming method for increasing power performance than traditional training. However, the exact protocols that can maximize power output have not yet been identified. Eleven healthy men, aged 32.3 [+ or -] 4.1 years, and 9 healthy women, aged 29.1 [+ or -] 3.5 years, performed countermovement jumps (CMJs) of maximal volition to assess peak power pre and post (immediately and at 1, 5, and 10 minutes) randomized WBV stimuli set at different frequency (30, 35, 40, and 50 Hz), displacement (2-4 vs. 4-6 mm), and duration (30, 45, and 60 seconds) combinations. Repeated-measures analysis of variance on peak power normalized to initial power (nPP) revealed no significant effects attributable to duration of stimulus. However, high frequencies were more effective when combined with high displacements, and low frequencies were more effective in conjunction with low displacements (p < 0.05). Additionally, the greatest improvements in nPP occurred at 1 minute posttreatment, with significant improvements lasting through 5 minutes posttreatment (p < 0.05). Optimal acute effects can be attained using as little as 30 seconds of WBV, and they are highest from 1 to 5 minutes posttreatment. Additionally, high frequencies were most effective when applied in conjunction with high displacements, whereas low frequencies were most effective when applied in conjunction with low displacements. Abstract

Ischemic preconditioning (IPC) involves brief, repeated bouts of limb occlusion and reperfusion capable of improving exercise performance at least partially by enhancing local skeletal muscle oxygenation. This study sought to investigate the effect of a lower limb IPC protocol, with either a 5-min or 45-min post-application delay, on vastus lateralis tissue saturation index (TSI) and systemic cardiac hemodynamics at rest and during short-duration intense cycling. Twelve young adults randomly completed four interventions: IPC (at 220 mmHg) with 5-min delay (IPC5), IPC with 45-min delay (IPC45), SHAM (at 20 mmHg) with 5-min delay (SHAM5), and SHAM with 45-min delay (SHAM45). Following IPC intervention and recovery delay, participants completed 5, 60-s high-intensity (100% Wpeak) cycle sprints separated by 120-sec of active recovery (30% Wpeak). Compared to baseline, TSI immediately following IPC5, but pre-exercise, remained lower than the equivalent for IPC45 (−5.9 ± 1.5%, p =.002). IPC, imposed at least 45-min before the completion of five 60-s sprint cycling efforts, significantly enhanced TSI during active recovery between sprint intervals compared to a 5-min delay (6.6 ± 2.4%, p =.021), and identical SHAM conditions (SHAM5: 5.8 ± 2.2%, p =.024; SHAM45: 6.2 ± 2.5%, p =.029). A 45-min delay following IPC appears to provide heightened skeletal muscle metabolic rebound prior to intense sprint cycling as compared to a 5-min delay. Furthermore, IPC followed by a 45-min delay enhanced recovery of skeletal muscle oxygenation during low intensity active sprint recovery, despite an unchanged decline in skeletal muscle oxygenation during near-maximal sprinting efforts. We combined tracer and arteriovenous (a-v) balance techniques to evaluate the effects of exercise and endurance training on leg triacylglyceride turnover as assessed by glycerol exchange. Measurements on an exercising leg were taken to be a surrogate for working skeletal muscle. Eight men completed 9 wk of endurance training [5 days/wk, 1 h/day, 75% peak oxygen consumption ([[??].sub.2peak])], with leg glycerol turnover determined during two pretraining trials [45 and 65% [[??].sub.2peak] (45% Pre and 65% Pre, respectively)] and two posttraining trials [65% of pretraining [[??].sub.2peak] (ABT) and 65% of posttraining [[??].sub.2peak] (RLT)] using [[sup.2][H.sub.5]]glycerol infusion, femoral a-v sampling, and measurement of leg blood flow. Endurance training increased [[??].sub.2peak] by 15% (45.2 [+ or -] 1.2 to 52.0 [+ or -] 1.8 ml x [kg.sup.-1] x [min.sup.-1], P < 0.05). At rest, there was tracer-measured leg glycerol uptake (41 [+ or -] 8 and 52 [+ or -] 15 [micro]mol/min for pre- and posttraining, respectively) even in the presence of small, but significant, net leg glycerol release (-68 [+ or -] 19 and -50 [+ or -] 13 [micro]mol/min, respectively; P < 0.05 vs. zero). Furthermore, while there was no significant net leg glycerol exchange during any of the exercise bouts, there was substantial tracer-measured leg glycerol turnover during exercise (i.e., simultaneous leg muscle uptake and leg release) (uptake, release: 45% Pre, 194 [+ or -] 41, 214 [+ or -] 33; 65% Pre, 217 [+ or -] 79, 201 [+ or -] 84; ABT, 275 [+ or -] 76, 312 [+ or -] 87; RLT, 282 [+ or -] 83, 424 [+ or -] 75 [micro]mol/min; all P < 0.05 vs. corresponding rest). Leg glycerol turnover was unaffected by exercise intensity or endurance training. Abstract

To evaluate the contribution of working muscle to whole body lipid oxidation, we examined the effects of exercise intensity and endurance training (9 wk, 5 days/wk, 1 h, 75% [Vo.sub.2peak]) on whole body and leg free fatty acid (FFA) kinetics in eight male subjects (26 [+ or -] 1 yr, means [+ or -] SE). Two pretraining trials [45 and 65% [Vo.sub.2max] (45UT, 65UT)] and two posttraining trials [65% of pretraining [Vo.sub.2peak] (ABT), and 65% of posttraining [Vo.sub.2peak] (RLT)] were performed using [1-[sup.13]C] palmitate infusion and femoral arteriovenous sampling. Training increased [Vo.sub.2peak] ak by 15% (45.2 [+ or -] 1.2 to 52.0 [+ or ] 1.8 ml * [kg.sup.-1] * [min.sup.-1], P < 0.05). Muscle FFA fractional extraction was lower during exercise (EX) compared with rest regardless of workload or training status [approximately equal to] 20 vs. 48%, P < 0.05). Two-leg net FFA balance increased from net release at rest ([approximately equal to]-36 [micro]mol/min) to net uptake during EX for 45UT (179 [+ or -] 75), ABT (236 [+ or -] 63), and RLT (136 [+ or -] 110) (P < 0.05), but not 65UT (51 [+ or -] 127). Leg FFA tracer measured uptake was higher during EX than rest for all trials and greater during posttraining in RLT (716 [+ or -] 173 [micro]mol/min) compared with pretraining (45UT 450 [+ or -] 80, 65UT 461 [+ or -] 72, P < 0.05). Leg muscle lipid oxidation increased with training in ABT (730 [+ or -] 163 [micro]mol/min) vs. 65UT (187 [+ or -] 94, P < 0.05). Leg muscle lipid oxidation represented ~62 and 30% of whole body lipid oxidation at lower and higher relative intensities, respectively. Abstract

The Human Genome Variation Society (HGVS) nomenclature guidelines encourage the accurate and standard description of DNA, RNA, and protein sequence variants in public variant databases and the scientific literature. Inconsistent application of the HGVS guidelines can lead to misinterpretation of variants in clinical settings. Reliable software tools are essential to ensure consistent application of the HGVS guidelines when reporting and interpreting variants. We present the hgvs Python package, a comprehensive tool for manipulating sequence variants according to the HGVS nomenclature guidelines. Distinguishing features of the hgvs package include: (1) parsing, formatting, validating, and normalizing variants on genome, transcript, and protein sequences; (2) projecting variants between aligned sequences, including those with gapped alignments; (3) flexible installation using remote or local data (fully local installations eliminate network dependencies); (4) extensive automated tests; and (5) open source development by a community from eight organizations worldwide. This report summarizes recent and significant updates to the hgvs package since its original release in 2014, and presents results of extensive validation using clinical relevant variants from ClinVar and HGMD.

Abstract

We evaluated the hypothesis that net leg total FFA, LDL-C, and TG uptake and HDL-C release during moderate-intensity cycling exercise would be increased following endurance training. Eight sedentary men (26 [+ or -] 1 yr, 77.4 [+ or -] 3.7 kg) were studied in the postprandial state during. 90 min of rest and 60 min of exercise twice before (45% and 65% [Vo.sub.2 peak]) and twice after 9 wk of endurance training (55% and 65% posttraining [Vo.sub.2 peak]). Measurements across an exercising leg were taken to be a surrogate for active skeletal muscle. To determine limb lipid exchange, femoral arterial and venous blood samples drawn simultaneously at rest and during exercise were analyzed for total and individual FFA (e.g., palmitate, oleate), LDL-C, HDL-C, and TG concentrations, and limb blood flow was determined by thermodilution. The transition from rest to exercise resulted in a shift from net leg total FFA release (-44 [+ or -] 16 [micro]mol/min) to uptake (193 [+ or -] 49 [micro]mol/min) that was unaffected by either exercise intensity or endurance training. The relative net leg release and uptake of individual FFA closely resembled their relative abundances in the plasma with ~21 and 41% of net leg total FFA uptake during exercise accounted for by palmitate and oleate, respectively. Endurance training resulted in significant changes in arterial concentrations of HDL-C (49 [+ or -] 5 vs. 52 [+ or -] 5 mg/dl, pre vs. post) and LDL-C (82 [+ or -] 9 vs. 76 [+ or -] 9 mg/dl, pre vs. post), but there was no net TG or LDL-C uptake or HDL-C release across the resting or active leg before or after endurance training. In conclusion, endurance training favorably affects blood lipoprotein profiles, even in young, healthy normolipidemic men, but muscle contractions per se have little effect on net leg LDL-C, or TG uptake or HDL-C release during moderate-intensity cycling exercise. Abstract

Catecholamine release is known to be regulated by feedforward and feedback mechanisms. Norepinephrine (NE) and epinephrine (Epi) concentrations rise in response to stresses, such as exercise, that challenge blood glucose homeostasis. The purpose of this study was to assess the hypothesis that the lactate anion is involved in feedback control of catecholamine concentration. Six healthy active men (26 [+ or -] 2 yr, 82 [+ or -] 2 kg, 50.7 [+ or -] 2.1 ml x [kg.sup.-1] x [min.sup.-1]) were studied on five occasions after an overnight fast. Plasma concentrations of NE and Epi were determined during 90 min of rest and 90 min of exercise at 55% of peak [O.sub.2] consumption (V[O.sub.2 peak]) two times with exogenous lactate infusion (lactate clamp, LC) and two times without LC (CON). The blood lactate profile (~4 mM) of a preliminary trial at 65% V[O.sub.2 peak] peak (65%) was matched during the subsequent LC trials. In resting men, plasma NE concentration was not different between trials, but during exercise all conditions were different with 65% > CON > LC (65%: 2,115 [+ or -] 166 pg/ml, CON: 1,573 [+ or -] 153 pg/ml, LC: 930 [+ or -] 174 pg/ml, P < 0.05). Plasma Epi concentrations at rest were different between conditions, with LC less than 65% and CON (65%: 68 [+ or -] 9 pg/ml, CON: 59 [+ or -] 7 pg/ml, LC: 38 [+ or -] 10 pg/ml, P < 0.05). During exercise, Epi concentration showed the same trend (65%: 262 [+ or -] 37 pg/ml, CON: 190 [+ or -] 34 pg/ml, LC: 113.2 [+ or -] 23 pg/ml, P < 0.05). Abstract

This paper describes the Cyber Situational Understanding (Cyber SU) Proof of Concept (CySUP) software system for exploring advanced Cyber SU capabilities. CySUP distills complex interrelationships among cyberspace entities to provide the “so what” of cyber events for tactical operations. It combines a variety of software components to build an end-to-end pipeline for live data ingest that populates a graph knowledge base, with query-driven exploratory analysis and interactive visualizations. CySUP integrates with the core infrastructure environment supporting command posts to provide a cyber overlay onto a common operating picture oriented to tactical commanders. It also supports detailed analysis of cyberspace entities and relationships driven by ad hoc graph queries, including the conversion of natural language inquiries to formal query language. To help assess its Cyber SU capabilities, CySUP leverages automated cyber adversary emulation to carry out controlled cyberattack campaigns that impact elements of tactical missions.

Abstract

To evaluate the hypothesis that precursor supply limits gluconeogenesis (GNG) during exercise, we examined training-induced changes in glucose kinetics [rates of appearance ([R.sub.a]) and disappearance ([R.sub.d])], oxidation ([R.sub.ox]), and recycling ([R.sub.r]) with an exogenous lactate infusion to 3.5-4.0 mM during rest and to pretraining 65% peak [O.sub.2] consumption ([Vo.sub.2 peak]) levels during exercise. Control and clamped trials (LC) were performed at rest pre- ([P.sub.R]R, [P.sub.R]R-LC) and posttraining ([P.sub.O]R, [P.sub.O]R-LC) and during exercise pre- ([P.sub.R][E.sub.X]) and posttraining at absolute ([P.sub.O][A.sub.B], [P.sub.O][A.sub.B]-LC) and relative ([P.sub.O][R.sub.L], [P.sub.O][R.sub.L]-LC) intensities. Glucose [R.sub.r] was not different in any rest or exercise condition. Glucose [R.sub.a] did not differ as a result of LC. Glucose [R.sub.ox] was significantly decreased with LC at [P.sub.O]R (0.38 [+ or -] 0.03 vs. 0.56 [+ or -] 0.04 mg*[kg.sup.-1]*[min.sup.-1]) and [P.sub.O][A. sub.B] (3.82 [+ or -] 0.51 vs. 5.0 [+ or -] 0.62 mg*[kg.sup.-1]*[min.sup.-1]). Percent glucose [R.sub.d] oxidized decreased with all LC except [P.sub.O][R.sub.L]-LC ([P.sub.R]R, 32%; [P.sub.R]R-LC, 22%; [P.sub.O]R, 27%; [P.sub.O]R-LC, 20%; [P.sub.O][A.sub.B], 95%; [P.sub.O][A. sub.B]-LC, 77%), which resulted in a significant increase in oxidation from alternative carbohydrate (CHO) sources at rest and [P.sub.O][A.sub.B].
